7. Force and velocity of impact during upper limb strikes in combat sports: a systematic review and meta-analysis.

Abstract

Upper limb strikes are frequent movements in combat sports and self-defence systems, in which numerous types of strikes can be applied. Therefore, the aim of this systematic review and meta-analysis was to summarise and compare the mechanical consequences of different types of upper limb strikes among various sports disciplines. A literature search was conducted in Scopus and Web of Science, with the following search formula: (impact force) AND (strike) AND (taekwondo) OR (karate) OR (self-defence) OR (combat sport) OR (boxing). The search resulted in 28 studies describing 9 kinds of strikes, where straight punches and reverse punches have larger mean impact forces than the kung fu punch (p < 0.001) and that a palm strike had a lower strike velocity (p < 0.001) than a reverse punch, straight punch, or junzuki punch. The highest recorded mean force was found for a straight punch (3427 N). Athletes in mixed martial arts, trainers of self-defence or tactical coaches can expect that straight punches and reverse punches should be performed at high speeds (over 10 m/s) and provide similar or larger impacts than other upper limb strikes; therefore, those punches should favour a combat athlete to win a competition or succeed in self-defence. Sportovci smíšených bojových umění používají strategii GaP s údery v dominantním postavení na zemi. Cílem této studie bylo porovnat průměrnou vrcholovou sílu (Fpeak) mezi třemi údery a pro odhad pravděpodobnosti dosažení síly zlomeniny lebky 5,1 kN pro každý typ stávky u sportovců a atletek. Abstract

The Thesis deals with construction of log furniture made of logs in diameter no more than 10 centimetres which are connected to a round joint and a mortise. This furniture is much popular and required not only for its growing approval rating of the log cabins in this country. The work contains drawing documentation of two chairs and two tables inclusive of a technical description, bill of materials, costing, and their practical production which is given evidence of taking photographs. Afterwards, the copious photographic work section is completed by information about practicable producing procedures, instruments and machines. There are summarized information from several branches and experience of most Czech makers of this furniture as a whole which is unique for his content.

Abstract

The main aim of this bachelor's thesis is the material use of branches with the diameter up to 7cm. The aim was to create a list of products, which are already being manufactured or could possibly be manufactured from this sort of material. Information about the tree species which can be used, about prices and also about the processes of manufacturing for selected types of products is included. The market research was carried out mainly via the internet, partially at the village fairs and markets, eventually by the consultations with experts in the field. The best products in terms of utilization, evaluation and practical processing of the material, are the furniture, pencils made of twigs, and partially also the wooden toys.

We develop an approach to tag proteomes synthesized by specific cell types in dissociated cortex, brain slices, and the brains of live mice. By viral-mediated expression of an orthogonal pyrrolysyl-tRNA synthetase-tRNAXXX pair in a cell type of interest and providing a non-canonical amino acid with a chemical handle, we selectively label neuronal or glial proteomes. Abstract

Nature uses 64 codons to encode the synthesis of proteins from the genome, and chooses 1 sense codon—out of up to 6 synonyms—to encode each amino acid. Synonymous codon choice has diverse and important roles, and many synonymous substitutions are detrimental. Here we demonstrate that the number of codons used to encode the canonical amino acids can be reduced, through the genome-wide substitution of target codons by defined synonyms. We create a variant of Escherichia coliwith a four-megabase synthetic genome through a high-fidelity convergent total synthesis. Our synthetic genome implements a defined recoding and refactoring scheme—with simple corrections at just seven positions—to replace every known occurrence of two sense codons and a stop codon in the genome. Thus, we recode 18,214 codons to create an organism with a 61-codon genome; this organism uses 59 codons to encode the 20 amino acids, and enables the deletion of a previously essential transfer RNA.

29. An Evolved Methanomethylophilus alvusPyrrolysyl-tRNA Synthetase/tRNA Pair Is Highly Active and Orthogonal in Mammalian Cells

Abstract

We recently characterized a new class of pyrrolysyl-tRNA synthetase (PylRS)/PyltRNA pairs from Methanomassiliicocalesthat are active and orthogonal in Escherichia coli. The aminoacyl-tRNA synthetases (aaRSs) of these pairs lack the N-terminal domain that is essential for tRNA recognition and in vivoactivity in the Methanosarcina mazei(Mm) PylRS but share a homologous active site with MmPylRS; this facilitates the transplantation of mutations discovered with existing PylRS systems into the new PylRS systems to reprogram their substrate specificity for the incorporation of noncanonical amino acids (ncAAs). Several of the new PylRS/PyltRNA pairs, or their evolved variants [including Methanomethylophilus alvus(Ma) PylRS/MaPyltRNA(6)CUA], are mutually orthogonal to the MmPylRS/MmPyltRNA pair, and the active sites of the Mmpair and Mapair can be diverged to enable the incorporation of distinct ncAAs in response to distinct codons via orthogonal translation in E. coli. Here we demonstrate that MaPylRS/MaPyltRNA(6)CUAis orthogonal to the aaRSs and tRNAs in mammalian cells and directs efficient incorporation of ncAAs into proteins. Moreover, we confirm that the MaPylRS/MaPyltRNA(6) and MmPylRS/MmPyltRNA pairs are mutually orthogonal in mammalian cells and demonstrates that these pairs can be used to encode distinct ncAAs into a protein in mammalian cells. Thus, the MaPylRS/MaPyltRNA(6)CUApair provides an additional pair that is orthogonal in both E. coliand mammalian systems and is mutually orthogonal to the most widely used system for genetic code expansion. Our results provide a foundation for expanding the scope of genetic code expansion and may also facilitate strategies for proteome-wide ncAA tagging with mutually orthogonal systems.
