The goal of this study is to identify non-invasive optical hemodynamic biomarkers that can index laboratory hematology measurements in sickle cell disease (SCD). We acquired frequency-domain NIRS (FD-NIRS) and diffuse correlation spectroscopy (DCS) data from the forearms and foreheads of 17 participants in a randomized, double-blind, placebo-controlled trial evaluating effects of isoquercetin (IQ) on thromboinflammation in SCD. We observed multiple, significant correlations between optical and hematology biomarkers including cerebral tissue oxygen saturation (StO2) and hematocrit (HCT); oxyhemoglobin ([O2Hb]) recovery rate and intercellular adhesion molecule 1 (ICAM-1); and blood flow index (BFI) reperfusion rate and coagulation index (CI). The potential of these non-invasive optical biomarkers for assessing vascular pathophysiology for the management of SCD warrants further exploration.

At the heart of sustainability is the relationship between humans and the planet. The binary of anthropocentric or ecocentric worldviews appears to be powerful in defining this relationship. Sustainability requires nuanced approaches which go beyond simple binaries, and therefore a dialectic approach which works to synthesise the binaries may be helpful. This paper draws on Maori cultural understandings of fire to trouble the ecocentric versus anthropocentric binary. Maori oral traditions of fire identify the connections between people and the planet and see people as part of fire and fire as part of people. By exploring Maori oral traditions, it is possible to see fire as more than purely an element that contributes to environmental problems and reveals the pedagogical potential of campfires to reignite the relationship between humans and the planet.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is a devastating disease most often associated with tobacco consumption that induces a field of mutations from which a tumor arises. Identification of ways to prevent the emergence of cancer in high-risk patients is an ultimate goal for combatting all types of cancer, including OSCC. METHODS: Our study employs a mouse model of tongue carcinogenesis induced by tobacco carcinogen mimetic, 4-nitroquinoline 1-oxide (4NQO), to establish tongue dysplasia and OSCC. We use conventional histology, immunohistochemistry, multispectral imaging, mass cytometry, novel cell lines, pharmaceutical inhibition of PI3Kγ, T-cell suppression assays and mouse transplant models in our functional experimentation. RESULTS: In our study, we identify Ly6G+ granulocytes as the most abundant immune cell type in a model of tongue carcinogenesis induced by tobacco carcinogen mimetic 4NQO. Targeting Ly6G+ granulocytes with a pharmacologic inhibitor of PI3Kγ, an isoform of PI3K exclusively expressed by myeloid cells, resulted in reduced tongue dysplasia severity, and reduced rates of OSCC. Importantly, we performed functional assays with the Ly6G+ granulocytes induced in cell line models of 4NQO carcinogenesis to demonstrate that these granulocytes have increased polymorphonuclear myeloid-derived suppressor cells (PMN-MDSC) activity against T-cell proliferation and these PMN-MDSCs play a functional role in promoting tumor formation by inhibiting tumor regression in a PI3Kγ-dependent manner. CONCLUSIONS: Overall, our data suggest that recruitment of PMN-MDSCs to sites of dysplasia is critical to immune suppression of CD8 T cells, thereby permitting malignancy, and PI3Kγ inhibitors are one mechanism to reduce PMN-MDSC recruitment, immunosuppression and tumorigenesis in OSCC.

The crown of a successful academic career is often considered an achievement of tenure. The path to tenure may vary based on the type of Carnegie University at which one is employed. Carnegie Doctoral granting R1 Universities place a high priority on conducting research, grant writing and publishing while other Carnegie classified Universities place more emphasis on teaching. Regardless of a University's Carnegie classification the path to tenure requires sound time management skills if an academic is to achieve tenure. To that end, the authors have decided to revisit Covey's Time Management Matrix (CTMM). The authors provide an ethnographic perspective with respect to the Quadrants of the CTMM for the purpose of providing a priority driven time self-management instrument to assist academics navigating towards tenure. Priority driven effective time self-management is key to successfully achieving tenure and we forward our academic interpretation of the CTMM as a potential guide for academics and their administrators. Further, the authors suggest that a priority driven effective time self-management strategy is portable across disciplines and national/cultural borders.

Leave No Trace (LNT) is globally the most widely accepted minimum impact program and has been linked to behavior change and the maintenance of a range of ecological measures. Critiques of LNT have emerged, including that LNT ignores wider impacts that contribute to climate change and diverse world views. Many outdoor education students carefully follow LNT practices. When students encounter these critiques, they often conclude either that LNT is effective and the detractors are misplaced or that LNT should be abandoned, thereby avoiding deep engagement. Education must prepare students for the contested project of creating a sustainable future. We argue that LNT shows considerable promise as a context for learning through dialectical approaches where students must engage with divergent articles and examples. LNT can be used to help students understand that taking informed action is important. LNT can also be used to show that there are no innocent actions.

AimsThis study explored the association of coronary artery calcium (CAC) with incident cancer subtypes in the Multi-Ethnic Study of Atherosclerosis (MESA). CAC is an established predictor of cardiovascular disease (CVD), with emerging data also supporting independent predictive value for cancer. The association of CAC with risk for individual cancer subtypes is unknown.Methods and resultsWe included 6271 MESA participants, aged 45-84 and without known CVD or self-reported history of cancer. There were 777 incident cancer cases during mean follow-up of 12.9 ± 3.1 years. Lung and colorectal cancer (186 cases) were grouped based on their strong overlap with CVD risk profile; prostate (men) and ovarian, uterine, and breast cancer (women) were considered as sex-specific cancers (in total 250 cases). Incidence rates and Fine and Gray competing risks models were used to assess relative risk of cancer-specific outcomes stratified by CAC groups or Log(CAC+1). The mean age was 61.7 ± 10.2 years, 52.7% were women, and 36.5% were White. Overall, all-cause cancer incidence increased with CAC scores, with rates per 1000 person-years of 13.1 [95% confidence interval (CI): 11.7-14.7] for CAC = 0 and 35.8 (95% CI: 30.2-42.4) for CAC ≥400. Compared with CAC = 0, hazards for those with CAC ≥400 were increased for lung and colorectal cancer in men [subdistribution hazard ratio (SHR): 2.2 (95% CI: 1.1-4.7)] and women [SHR: 2.2 (95% CI: 1.0-4.6)], but not significantly for sex-specific cancers across sexes.ConclusionCAC scores were associated with cancer risk in both sexes; however, this was stronger for lung and colorectal when compared with sex-specific cancers. Our data support potential synergistic use of CAC scores in the identification of both CVD and lung and colorectal cancer risk.

Background and aimsCoronary artery calcium (CAC) scores have been shown to be associated with CVD and cancer mortality. The use of CAC scores for overall and lung cancer mortality risk prediction for patients in the Coronary Artery Calcium Consortium was analyzed.MethodsWe included 55,943 patients aged 44-84 years without known heart disease from the CAC Consortium. There were 1,088 cancer deaths, among which 231 were lung cancer, identified by death certificates with a mean follow-up of 12.2 ± 3.9 years. Fine-and-Gray competing-risk regression was used for overall and lung cancer-specific mortality, accounting for the competing risk of CVD death and after adjustment for CVD risk factors. Subdistribution hazard ratios (SHR) were reported.ResultsThe mean age of all patients was 57.1 ± 8.6 years, 34.9% were women, and 89.6% were white. Overall, CAC was strongly associated with cancer mortality. Lung cancer mortality increased with increasing CAC scores, with rates per 1000-person years of 0.2 (95% CI: 0.1-0.3) for CAC = 0 and 0.8 (95% CI: 0.6-1.0) for CAC ≥400. Compared with CAC = 0, hazards were increased for those with CAC ≥400 for lung cancer mortality [SHR: 1.7 (95% CI: 1.2-2.6)], which was driven by women [SHR: 2.3 (95% CI: 1.1-4.8)], but not significantly increased for men. Risks were higher in those with positive smoking history [SHR: 2.2 (95% CI: 1.2-4.2)], with associations driven by women [SHR: 4.0 (95% CI: 1.4-11.5)].ConclusionsCAC scores were associated with increased risks for lung cancer mortality, with strongest associations for current and former smokers, especially in women. Used in conjunction with other clinical variables, our data pinpoint a potential synergistic use of CAC scanning beyond CVD risk assessment for identification of high-risk lung cancer screening candidates.

The diversity of issue interests and party options in multiparty systems makes individual electoral decisions increasingly complex. Voters are challenged to find a political party that represents their own political views in this more complex political space. This research offers a new methodological approach to studying voting choice in a multidimensional party space. We integrate the issue preferences of European voters and the issue preferences of party elites in a two-dimensional model of electoral choice. A common space of political competition for citizens and party elites is defined by the economic and cultural cleavages using data from the 2009 European Elections Studies (EES). Our innovation is to employ multilevel structural equation modeling to address the unique statistical challenges of a multi-dimensional party space, mass-elite comparisons, and cross-national analysis. This new approach generates results that are distinctly different from previous studies—even those using the same dataset. By factoring in the measurement of issue dimensions, economic issues have a stronger impact than recognized using previous methodologies, with more modest cultural influences on voting. Moreover, there are significant cross-over effects of the two cleavages in voters’ choices. The results reveal the complexity of realignment between voters and political parties in Western Europe.

Impaired facial emotion recognition in children with Autism Spectrum Disorder (ASD) is in contrast to their intact emotional music recognition. This study tested whether emotion congruent music enhances facial emotion recognition. Accuracy and reaction times were assessed for 19 children with ASD and 31 controls in a recognition task with angry, happy, or sad faces. Stimuli were shown with either emotionally congruent or incongruent music or no music. Although children with ASD had higher reaction times than controls, accuracy only differed when incongruent or no music was played, indicating that congruent emotional music can boost facial emotion recognition in children with ASD. Emotion congruent music may support emotion recognition in children with ASD, and thus may improve their social skills.

Dropout remains a persistent challenge within high school education. In this paper, we present a case study on automatically detecting whether a student is at-risk of dropout within a diverse school district in Texas. We predict whether a student will drop out in a future school year from data on students' discipline, attendance, course-taking, and grades, using a logistic regression framework. We discuss the predictive properties of the model, and the features that are predictive of dropout in this context.
