6. The importance of vector abundance and seasonality: results from an expert consultation

Abstract

Knowing where vector species occur is crucial for the assessment of vector-borne disease risk. This was recognised by EFSA and ECDC, who, over the period 2014–2018, funded VectorNet, a European network for gathering and sharing data on the geographic distribution of arthropod vectors of disease agents affecting humans and livestock. The VectorNet database contains distribution data on four vector groups: mosquitoes, sandflies, Culicoides midges and ticks. The majority of these data are on vector presence or absence, but since 2015, count data from a number of targeted field surveys, which used standardised sampling methods and that were funded through the network, have been added. The objective of this document is to describe an assessment, for each vector group, of whether vector count data (abundance) and the way these change throughout the year (seasonality) can provide useful information about epidemiological processes of interest, and therefore, whether resources should be devoted to collecting such data. The document also summarises what measures of abundance and seasonality can be collected for each group, what gaps remain in the sampling coverage and what can be done to fill these gaps. Furthermore, it provides guidance for prioritising the acquisition of information.

Abstract

There is a need for standardizing sand fly sampling methodology and guidance on trap positioning for quantitative sand fly studies. We investigated differences in sand fly density with 'sticky' interception and CO2‐light attraction traps, in relation to trap distance to the ground and the presence or absence of a continuous or discontinuous (wire mesh) vertical surface adjacent to the trap. The study, conducted in a dog kennel in southeast Spain, lasted 48 days and collected 692 Phlebotomus papatasi, P. perniciosus, P. ariasi and Sergentomyia minuta specimens. There were no significant differences between species with respect to trap position. Overall, density in sticky traps was highest closest to the ground and next to the continuous vertical surface, followed sequentially by traps similarly placed adjacent to the wire mesh and those hanging from a rope across the kennel yard. In contrast, density in CO2‐light traps was highest in traps hanging from the rope near the ground, followed by those next to the continuous vertical surface. The overall negative relationship between sand fly density and ground distance was not significant for CO2‐light traps next to the continuous vertical surface. Abstract

A 2‐year longitudinal study of enzyme‐linked immunosorbent assay (ELISA) antibodies against Phlebotomus perniciosus and Phlebotomus papatasi (Diptera: Psychodidae) sandfly saliva was performed in 32 Beagle dogs treated preventively with an imidacloprid–permethrin topical insecticide in an endemic area in Spain. Dogs were grouped into three sandfly exposure groups according to the time of inclusion in the study. Assays analysed immunoglobulin G (IgG) against salivary gland homogenates (SGH) of both species and recombinant P. papatasi rSP32 and P. perniciosus rSP03B proteins in serum. The dogs were participating in a Leishmania infantum (Kinetoplastida: Trypanosomatidae) vaccine trial and were experimentally infected with the parasite in the second year. No dog acquired natural L. infantum infections during the first year, but most developed anti‐saliva antibodies, and median log‐transformed optical densities (LODs) were seasonal, mimicking those of local sandflies. This indicates that the repellent efficacy of the insecticide used is below 100%. Multi‐level modelling of LODs revealed variability among dogs, autocorrelation and differences according to the salivary antigen and the dog's age. However, dog seroprevalence, estimated using pre‐exposure LODs as cut‐offs, was relatively low. This, and the fact that dogs did not become naturally infected with L. infantum, would support the efficacy and usefulness of this imidacloprid–permethrin topical insecticide in canine leishmaniasis control. Dogs naturally exposed to Leishmania vectors, the sandflies Phlebotomus perniciosus and Phlebotomus papatasi, and treated preventively with sandfly repellents developed antibodies against the insects' salivary proteins.Anti‐P. perniciosus and ‐P. papatasi saliva antibody concentrations were low, which suggests the repellent limited sandfly bites and is useful in canine leishmaniasis control.Longitudinal monitoring of anti‐saliva antibodies is a good method of marking insect repellent efficacy against P. perniciosus and P. papatasi in dogs. Abstract

Abstract: The spatial and temporal distribution of Phlebotomus perniciosus (Diptera: Psychodidae) (Newstead, 1911), the sand fly vector of pathogens of public and animal health importance, was investigated in a high sand fly density rural area in Spain using light‐attraction and sticky‐interception traps. Traps were placed inside animal buildings and outside at increasing distance from animals. A total of 8506 sand flies were collected, 87% with light traps. Species frequency differed between trap types. The abundance of P. perniciosus decreased exponentially with increasing distance to animals and, while females were most common in the animal enclosure, males predominated in adjoining storage places. Increasing CO2 concentration had an additional positive effect on female abundance only. Both male and female density increased with rising temperature, and there was some indication that females were more active than males at higher relative humidity. The study confirms that P. perniciosus aggregates around animal premises, although male and female distributions differ and should be analysed separately to account for biological and behavioural differences. This provides further evidence that light traps offer an accurate estimation of the relative spatial and temporal abundance of P. perniciosus, conferring an added value for the study of this species and the risk of pathogen transmission. Abstract

Maedi-Visna (MV) is a slow, progressive and fatal lymphoproliferative disease of sheep associated to infection with a lentivirus of the Retroviridae family. The virus has affinity for the monocyte-macrophage cell lineage of the lung, brain, mammary tissue and joints and animals become infected by inhalation of virus-containing aerosols and ingestion of milk containing the virus. The disease is distributed worldwide and present in over 95% of the flocks in the Basque Country in Spain. Since neither antiviral treatment or vaccination are available, disease control relies a good understanding of the pathogenesis and epidemiology of the disease and in particular of the risk factors associated with infection. Recent studies in sheep separated from their dams at birth and reared artificially, have shown a strong association between infection and the life-time infection status of the dam, suggesting there may be an important genetic component to disease susceptibility/resistance (Berriatua et al, in progress). Polymorphism at the major histocompatibility complex (MHC) has been related with disease susceptibility in other retroviral infections of animals such as Enzootic Bovine Leukosis in cattle or Caprine Arthritis-Encephalitis Virus induced arthritis in goats. As part of a large study of genetic susceptibility to viral diseases in sheep, the genetic polymorphism in the second exon of the MHC Class II DRB1 locus is being analyzed in a sample of Basque Latxa breed sheep comprising 13 half-sib families with 3 to 15 offspring. A typing assay based in Single-Strand Conformational Polymorphism (SSCP) previously developed by us is being applied. After the SSCP typing the different patterns will be sequenced, if shown to be different from previously identified. The Latxa breed may be a good model for studies of genetic disease resistance as recent studies on the DRB1 gene second exon (Jugo & Vicario, 2000) have revealed a very high frequency of an allele with the SR motif in positions 70-71 which may be of great importance in the immune response to parasites. In addition to the work currently in progress, we will extend our analysis to a larger number of families and to other MHC genes located in the Class I and Class III regions. (Texte intégral)

Abstract

La infección por el virus del maedi-visna se fué observando a lo largo del siglo pasado pero fue en los años treinta, al entrar el virus en la cabaña ovina islandesa, cuando se empezó su estudio sistemático que condujo a su aislamiento e identificación. La distribución actual del virus es universal, con la notable excepción de Australia y Nueva Zelanda. En España, su distribución es generalizada y su prevalencia media-alta. Para éste virus no existen ni vacunas ni tratamientos y, por lo tanto, tan sólo se pueden llevar a cabo medidas de control. En éste capítulo revisaremos la historia de la enfermedad, su distribución geográfica, contagio, control e importancia económica.
