A systematic list of echinoderms from Malpelo Fauna and Flora Sanctuary (MFFS) was prepared, based on localsampling, literature review and identification of specimens from Museo de Historia Natural Marina de Colombia(MHNMC) and from National Museum of Natural History, Smithsonian Institution, Washington D. C. (USNM).Standard methodology for monitoring the Eastern Tropical Pacific Seascape was used in December 2006 and March2007 to sample shallow water echinoderms (1-20 m deep). Malpelo is the richest locality of the Colombian Pacific with84 species reported (56 % of them deep water organisms), belonging to 22 orders, 42 families and 68 genera, distributedbetween the surface and a depth of 5000 m. Thirty-six new records for MFFS and Colombian Pacific are reported. InColombia the shallow water species are widely distributed in the Pacific, while deep forms only occur at Malpelo. Themajority of species are mostly related to the Panamanian and Galapagos region showing a possible connectivity. Thislist increases knowledge on the echinoderm biodiversity from the Malpelo Island in 68 %.

We provide the first assessment for a coral reef fish population of a potentially threatened endemic species from a remote and isolated oceanic island. Based on a 3-dimensional (3D) numerical field model of the reef habitat and underwater visual censuses conducted by 2 different teams in February and May 2016, we show that the 3.7 km2 of the Clipperton Atoll reefs shallower than 30 m host >50000 ind. (average density of 0.02 ind. m-2) of the endemic Clipperton angelfish Holacanthus limbaughi. Based on limited submersible observations to 150 m, the population likely extends below the 30 m depth stratum, resulting in an even larger population size and providing a potential depth refuge against expected warming ocean temperatures.

Summary: This study proposes length relationship equations to determine total length of four species of sharks (Carcharhinus falciformis, Mustelus lunulatus, Sphyrna lewini and Carcharhinus limbatus) based on trunk lengths (TrL) and interdorsal lengths (IL) caught in Colombia and Central America Pacific Ocean. In Central American Pacific, data were recorded for C. falciformis, C. limbatus and S. lewini between 2006 and 2012. In the Colombian Pacific, data was recorded in 1994–1995 and from 2006 to 2012 for C. falciformis, S. lewini and M. lunulatus. In the Central American Pacific the IL – TL relationship was acceptable for C. falciformis (r2 = .69) and C. limbatus which presented a good relationship (r2 = .81) where most specimens were juveniles; the same trend was found for S. lewini (r2 = .96). The TL‐TrL relationship estimated for species caught in the Colombian Pacific was significant for C. falciformis (r2 = .98) and for M. lunulatus (r2 = .84); however, for S. lewini this relationship was low (r2 = .55). Results indicate this is a useful tool for fishery statistics and fishery management for elasmobranch species of the Eastern Tropical Pacific. [ABSTRACT FROM AUTHOR]

The scalloped hammerhead shark, Sphyrna lewini , is a Critically Endangered, migratory species known for its tendency to form iconic and visually spectacular large aggregations. Herein, we investigated the population genetic dynamics of the scalloped hammerhead across much of its distribution in the Eastern Tropical Pacific (ETP), ranging from Costa Rica to Ecuador, focusing on young-of-year animals from putative coastal nursery areas and adult females from seasonal aggregations that form in the northern Galápagos Islands. Nuclear microsatellites and partial mitochondrial control region sequences showed little evidence of population structure suggesting that scalloped hammerheads in this ETP region comprise a single genetic stock. Galápagos aggregations of adults were not comprised of related individuals, suggesting that kinship does not play a role in the formation of the repeated, annual gatherings at these remote offshore locations. Despite high levels of fisheries exploitation of this species in the ETP, the adult scalloped hammerheads here showed greater genetic diversity compared with adult conspecifics from other parts of the species' global distribution. A phylogeographic analysis of available, globally sourced, mitochondrial control region sequence data ( n  = 1818 sequences) revealed that scalloped hammerheads comprise three distinct matrilines corresponding to the three major world ocean basins, highlighting the need for conservation of these evolutionarily unique lineages. Sharks are fished for human consumption in Colombia, and fins are exported illegally to international markets. The goal was to identify differences in total mercury (THg) concentrations in fins and muscles of shark species seized in the Buenaventura port (Colombian Pacific), and to assess potential human health risks related to shark consumption. Seven species were considered in this study: Pelagic Thresher (Alopias pelagicus), Pacific Smalltail Shark (Carcharhinus cerdale), Brown Smoothhound (Mustelus henlei), Sicklefin Smoothhound (Mustelus lunulatus), Scalloped Bonnethead (Sphyrna corona), Scalloped Hammerhead (Sphyrna lewini), and Bonnethead Shark (Sphyrna tiburo), and THg was analyzed in shark tissues. 24% muscle samples concentrations were above international recommended limits for human consumption, especially for A. pelagicus and S. lewini. Stable isotopes (δ 13 C and δ 15 N) analysis showed a foraging overlapping in coastal habitats, where overfishing usually occurs. Assessing the impact of global changes and protection effectiveness is a key step in monitoring marine fishes. Most traditional census methods are demanding or destructive. Nondisturbing and nonlethal approaches based on video and environmental DNA are alternatives to underwater visual census or fishing. However, their ability to detect multiple biodiversity factors beyond traditional taxonomic diversity is still unknown. For bony fishes and elasmobranchs, we compared the performance of eDNA metabarcoding and long-term remote video to assess species' phylogenetic and functional diversity. We used 10 eDNA samples from 30 L of water each and 25 hr of underwater videos over 4 days on Malpelo Island (pacific coast of Colombia), a remote marine protected area. Metabarcoding of eDNA detected 66% more molecular operational taxonomic units (MOTUs) than species on video. We found 66 and 43 functional entities with a single eDNA marker and videos, respectively, and higher functional richness for eDNA than videos. Despite gaps in genetic reference databases, eDNA also detected a higher fish phylogenetic diversity than videos; accumulation curves showed how 1 eDNA transect detected as much phylogenetic diversity as 25 hr of video. Environmental DNA metabarcoding can be used to affordably, efficiently, and accurately census biodiversity factors in marine systems. Management and conservation actions in marine-protected areas require baselines for monitoring threatened marine fauna such as elasmobranchs. This article provides evidence of the occurrence of 34 species of elasmobranchs (21 sharks and 13 batoids) in the Malpelo Flora and Fauna Sanctuary, Colombia, including five new records of sharks and three of rays. From 1987 to 2021, new records were obtained by underwater visual census using SCUBA, manned submersibles and deep-ocean cameras to depths of up to 2211 m. Quantifying fish species diversity in rich tropical marine environments remains challenging. Environmental DNA (eDNA) metabarcoding is a promising tool to face this challenge through the filtering, amplification, and sequencing of DNA traces from water samples. However, because eDNA concentration is low in marine environments, the reliability of eDNA to detect species diversity can be limited. Using an eDNA metabarcoding approach to identify fish Molecular Taxonomic Units (MOTUs) with a single 12S marker, we aimed to assess how the number of sampling replicates and filtered water volume affect biodiversity estimates. We used a paired sampling design of 30 L per replicate on 68 reef transects from 8 sites in 3 tropical regions. We quantified local and regional sampling variability by comparing MOTU richness, compositional turnover, and compositional nestedness. We found strong turnover of MOTUs between replicated pairs of samples undertaken in the same location, time, and conditions. Paired samples contained non-overlapping assemblages rather than subsets of one another. As a result, non-saturated localized diversity accumulation curves suggest that even 6 replicates (180 L) in the same location can underestimate local diversity (for an area <1 km). However, sampling regional diversity using ~25 replicates in variable locations (often covering 10 s of km) often saturated biodiversity accumulation curves. Our results demonstrate variability of diversity estimates possibly arising from heterogeneous distribution of eDNA in seawater, highly skewed frequencies of eDNA traces per MOTU, in addition to variability in eDNA processing. This high compositional variability has consequences for using eDNA to monitor temporal and spatial biodiversity changes in local assemblages. Monitoring large marine mammals is challenging due to their low abundances in general, an ability to move over large distances and wide geographical range sizes.The distribution of the pygmy ( Kogia breviceps ) and dwarf ( Kogia sima ) sperm whales is informed by relatively rare sightings, which does not permit accurate estimates of their distribution ranges. Many species of sharks form aggregations around oceanic islands, yet their levels of residency and their site specificity around these islands may vary. In some cases, the waters around oceanic islands have been designated as marine protected areas, yet the conservation value for threatened shark species will depend greatly on how much time they spend within these protected waters. Eighty-four scalloped hammerhead sharks (Sphyrna lewini Griffith & Smith), were tagged with acoustic transmitters at Cocos Island between 2005-2013. The average residence index, expressed as a proportion of days present in our receiver array at the island over the entire monitoring period, was 0.52±0.31, implying that overall the sharks are strongly associated with the island. Residency was significantly greater at Alcyone, a shallow seamount located 3.6 km offshore from the main island, than at the other sites. Timing of presence at the receiver locations was mostly during daytime hours. Although only a single individual from Cocos was detected on a region-wide array, nine hammerheads tagged at Galapagos and Malpelo travelled to Cocos. The hammerheads tagged at Cocos were more resident than those visiting from elsewhere, suggesting that the Galapagos and Malpelo populations may use Cocos as a navigational waypoint or stopover during seasonal migrations to the coastal Central and South America. Previous investigations of the population genetics of the scalloped hammerhead sharks (Sphyrna lewini) in the Eastern Tropical Pacific have lacked information about nursery areas. Such areas are key to promoting conservation initiatives that can protect young sharks from threats such as overfishing. Here, we investigated the genetic diversity, phylogeography, and connectivity of S. lewini found in 3 areas of Colombia's Pacific coast: around Malpelo Island and in 2 National Natural Parks on the Colombian Pacific mainland (Sanquianga and Ensenada de Utría). We analyzed mtDNA control region (CR) sequences and genotyped 15 microsatellite loci in 137 samples of adults and juveniles. The mtDNA analyses showed haplotypes shared between the Colombian Pacific individuals sampled in this investigation and other areas in the Eastern Tropical Pacific, the Indo-Pacific, and with sequences previously reported in Colombia (Buenaventura Port), as well as 4 unique haplotypes. Population assignment and paternity analyses detected 3 parent-offspring pairs between Malpelo and Sanquianga and 1 between Malpelo and Utría. These results indicate high genetic connectivity between Malpelo Island and the Colombian Pacific coast, suggesting that these 2 areas are nurseries for S. lewini. This is, to our knowledge, the first evidence of nursery areas identified for the scalloped hammerhead shark anywhere in the world. Difficulties in scaling up theoretical and experimental results have raised controversy over the consequences of biodiversity loss for the functioning of natural ecosystems. Using a global survey of reef fish assemblages, we show that in contrast to previous theoretical and experimental studies, ecosystem functioning (as measured by standing biomass) scales in a non-saturating manner with biodiversity (as measured by species and functional richness) in this ecosystem. Our field study also shows a significant and negative interaction between human population density and biodiversity on ecosystem functioning (i.e., for the same human density there were larger reductions in standing biomass at more diverse reefs). Human effects were found to be related to fishing, coastal development, and land use stressors, and currently affect over 75% of the world's coral reefs. Our results indicate that the consequences of biodiversity loss in coral reefs have been considerably underestimated based on existing knowledge and that reef fish assemblages, particularly the most diverse, are greatly vulnerable to the expansion and intensity of anthropogenic stressors in coastal areas., Competing Interests: The authors have declared that no competing interests exist.
