International audience; In the present study, we propose a high-throughput sequencing protocol using aNextera XT Library DNA kit on an Illumina MiSeq instrument. We made major modifications to this library preparation in order to multiplex 384 samples in a single Illumina flow cell. To validate our protocol, we compared the sequences obtained with the modified Illumina protocol to those obtained with the GridION Nanopore protocol. For the modified Illumina protocol, our results showed that 94.9% (357/376) of the sequences were interpretable, with a viral genome coverage between 50.5% and 99.9% and an average depth of 421×. For the GridION Nanopore protocol, 94.6% (356/376) of the sequences were interpretable, with a viral genome coverage between 7.0% and 98.6% and an average depth of 2123×. The modified Illumina protocol allows for gaining EUR 4744 and returning results of 384 samples in 53.5 h versus four times 55.5 h with the standard Illumina protocol. Our modified MiSeq protocol yields similar genome sequence data as the GridION Nanopore protocol and has the advantage of being able to handle four times more samples simultaneously and hence is much less expensive.

International audience; After the end of the first epidemic episode of SARS-CoV-2 infections, as cases began to rise again during the summer of 2020, we at IHU Méditerranée Infection in Marseille, France, intensified the genomic surveillance of SARS-CoV-2, and described the first viral variants. In this study, we compared the incidence curves of SARS-CoV-2-associated deaths in different countries and reported the classification of SARS-CoV-2 variants detected in our institute, as well as the kinetics and sources of the infections. We used mortality collected from a COVID-19 data repository for 221 countries. Viral variants were defined based on ≥5 hallmark mutations along the whole genome shared by ≥30 genomes. SARS-CoV-2 genotype was determined for 24,181 patients using next-generation genome and gene sequencing (in 47 and 11% of cases, respectively) or variant-specific qPCR (in 42% of cases). Sixteen variants were identified by analyzing viral genomes from 9,788 SARS-CoV-2-diagnosed patients. Our data show that since the first SARS-CoV-2 epidemic episode in Marseille, importation through travel from abroad was documented for seven of the new variants. In addition, for the B.1.160 variant of Pangolin classification (a.k.a. Marseille-4), we suspect transmission from farm minks. In conclusion, we observed that the successive epidemic peaks of SARS-CoV-2 infections are not linked to rebounds of viral genotypes that are already present but to newly introduced variants. We thus suggest that border control is the best mean of combating this type of introduction, and that intensive control of mink farms is also necessary to prevent the emergence of new variants generated in this animal reservoir.

This thesis is part of the sustainable development context where the energy challenges rely on designing numerous and lumped power converters with good power density and high efficiency. New power semiconductor devices, namely wide band semiconductors (GaN, SiC) are used in designing the converters. The high frequency control of these converters makes the system more sensitive to parasitic elements. The latter elements disrupt the switching behavior of the transistors and generate additional losses. In this context this work was carried out in a cotutelle partnership between Ampère Laboratory in Villeurbanne and LN2 laboratory at the University of Sherbrooke; the aim being to make a contribution in optimizing the switching conditions of GaN HEMTs. The first work axis consists in managing the voltage and current switching speed through gate control strategies in order to improve the conducted EMI. Firstly, most of the proposed control circuits are developed in open-loop and then secondly in closed-loop in order to compensate the effects of non-linearities (with respect to temperature, load current and operating voltage). Concerning the development of control systems, it can be done first by the use of available discrete components, then by the alternative of the monolithic GaN integration which is considered in order to bring more speed and efficiency. Monolithic integration would also solve the problem of parasitic inductances. To facilitate the design of integrated circuits and control systems, the development of a behavioral model of HEMT GaN will serve as a modeling tool. The second axis of the work consists in experimentally validating well-adapted control system for the gate of the power transistor in order to master the transient behaviors of the power transistors. Namely it is necessary to allow a satisfying management of losses during dead time in a half bridge converter. At the end of this work, the control systems developed in open loop made it possible to slow the switching speeds by at least 30 % but causing an increase in switching losses up to 50% in some cases. Due to the fast switching speed of HEMT GaNs and the limitations of discrete components on the market, the reduction rate of switching speeds obtained with the closed loop (reduction rate less than 20%) is less attractive than that of the open loop. Using a monolithic circuit can be an alternative to increase the rate of reduction of closed loop switching speeds. SPICE simulation toward monolithic circuit are the basis of this hypothesis. Concerning the second axis, the application of multilevel gate voltage control of the transistors of half bridge made it possible to reduce the losses of reverse conduction and the losses due to the phenomena of Cross Talk by at least by 30 %.; Cette thèse s'inscrit dans un contexte de développement durable où les enjeux énergétiques consistent à concevoir des convertisseurs de puissance plus disséminés, donc avec une Spécification ambitieuse en termes de densités massique et volumique. Les composants à semiconducteur dit à grand Gap permettent l’augmentation de la fréquence de commutation et permettent un fonctionnement à plus haute température locale. Les commutations à front raides et à haute fréquence des transistors rendent le système plus sensible aux éléments parasites. Ceci perturbe en retour la commutation des transistors et génère des pertes joules supplémentaires. Dans ce contexte les travaux ont été effectués dans le cadre d’une cotutelle entre les laboratoires Ampère (INSA Lyon) et LN2 (Université de Sherbrooke), le but étant d’apporter des contributions à l’optimisation de la commutation des HEMTs GaN. Le premier axe des travaux consiste à mettre en place des stratégies de contrôle de vitesses de commutation en tension et en courant, par la grille, dans le but d’améliorer la signature CEM. Les circuits de contrôle proposés sont développés dans un premier temps en boucle ouverte puis dans un second temps en boucle fermée afin de compenser des non-linéarités (température, courant de charge et tension de fonctionnement). Les prototypes de contrôle de grille ont été testés à partir de composants discrets du marché. Des limites apparaissent, que l’intégration monolithique GaN doit corriger à terme, en particulier en atténuant fortement le problème des inductances parasites. Les analyses en simulation ont reposé sur l’adoption d’un modèle comportemental de HEMT GaN identifiable. Le deuxième axe des travaux consiste à vérifier de manière systémique différentes stratégies de contrôle de grille notamment pour la gestion du compromis entre pertes joule pendant les temps morts au sein d’un à bras d’onduleur et la performance fréquentielle des commutations. Aux termes de ces travaux, les systèmes de contrôles développés en boucle ouverte ont permis de ralentir les vitesses de commutation d’au moins 30 %, occasionnant une augmentation des pertes de commutation, dans un ordre de grandeur inférieur à 50 %. Due à la rapidité de commutation des HEMT GaN et aux limites des composants discrets du marché, le taux de réduction des vitesses de commutation obtenu avec la boucle fermée (taux de réduction inférieur à 20 %) est moins intéressant qu’avec la boucle ouverte. L’utilisation d’un circuit monolithique peut être une alternative pour augmenter le taux de réduction des vitesses de commutation en boucle fermée. Des résultats de simulation sous SPICE en vue du circuit monolithique sont à la base de cette hypothèse. Concernant le deuxième axe, l’application de commande multiniveaux de grille des transistors du bras d’onduleur a permis de réduire les pertes de conduction inverse et les pertes dues aux phénomènes de Cross Talk d’au moins 30 %.

A cross-reading of the writings of Machiavelli and Schmitt not only allows us to decode certain past events, it also illuminates our understanding of the most recent challenges to the security of the state. Indeed, both authors conceptualize states of emergency – understood as responses to disorders threatening the preservation of the state not adequately covered by positive law – and their analyses demonstrate points of agreement as well as divergence. Comparing and contrasting them makes salient their lines of force. Both authors shed important light on the Anglo- American and French revolutionary movements of the seventeenth and eighteenth centuries as exceptional situations. Moreover, they also provide insight into the more recent emergency situations encountered by these states in confronting the terrorist phenomenon. The interest of such a perspective is twofold. On the one hand, the confrontation of the Machiavelo-Schmittian theory with concrete events demonstrates – despite insurmountable anachronism – their undeniable influence on the operations of law in crisis situations. On the other, from the perspective of their practical application, the theories of Machiavelli and Schmitt both prove inadequate in their apprehension of the exceptional phenomenon, notably with regard to the means that might be deployed to curb the threat. With this in mind, it is imperative to understand both the relevance of the political thought of these two authors, and take the measure of the limits of their theories. A proper understanding of the pertinence and limits of their theories with regard to the present situation should enable us to devise improved legal tools in response to the current crisis.; La lecture croisée de Machiavel et Schmitt ne permet pas seulement de déchiffrer certains événements passés, elle éclaire encore la compréhension des phénomènes les plus récents. En effet, la conceptualisation que chacun de ces deux auteurs offre de la notion de situation exceptionnelle – comprise comme un trouble mettant en péril la préservation de l’État, et face auquel le droit positif peine à s’appliquer – démontre des points d’accord autant que de divergence, mais dont précisément la confrontation tend à faire émerger des lignes de force. Or, d’une part, ces dernières semblent permettre d’envisager le mouvement révolutionnaire anglo-américain et français des XVIIe et XVIIIe siècles sous l’angle d’une situation exceptionnelle. D’autre part, et d’une manière plus contemporaine, elles permettent encore de rendre compte de l’exceptionnalité du phénomène terroriste affectant actuellement ces mêmes États. L’intérêt d’une telle mise en perspective est double. D’un côté, la confrontation de la théorie machiavélo-schmittienne aux événements concrets démontre – malgré un indépassable anachronisme – leur incontestable influence sur le droit des situations de crise. Mais d’un autre côté, et dans le même temps, l’observation de la pratique révèle les insuffisances de Machiavel et Schmitt quant à leur appréhension du phénomène exceptionnel, en particulier à propos des moyens à mettre en oeuvre pour l’endiguer. Face à ce constat, il sera nécessaire de tenir compte de la pertinence des présupposés de ces deux auteurs, autant que de les critiquer, afin de pouvoir finalement s’en émanciper pour proposer des outils juridiques aptes à régir le phénomène exceptionnel actuel.

Scrotal masse due to polyorchidism is not frequent in children. We report 1 case of polyorchidism associated an hydrocele in an eight years old boy. The diagnostic of the scrotal masses was done by physical examination. Cure of hydrocele was done with a conservative management of the polyorchidisme based on clinical and sonography surveillance of supernumerary testicle.

International audience; The human gut is composed of a large diversity of microorganisms, which have been poorly described. Here, using culturomics, a new concept based on the variation in culture conditions and MALDI-TOF MS identification, we proceed to explore the microbial diversity of the complex ecosystem of the human gut. Using this approach, we isolated strain AT8(T) (=CSUR P2118 = DSM 101782) from stool specimens collected from a 51-year-old obese French woman. Strain AT8(T) is a strictly anaerobic, nonmotile, nonspore-forming gram-positive coccus that do not exhibit catalase and oxidase activities. 16S rDNA-based identification of strain AT8(T) demonstrated 92% gene sequence similarity with Eggerthella lenta DSM 2243, the phylogenetically closed validly named type species. Here, we present a set of features for the strain AT8(T) and the description of its complete genome sequence and annotation. The 2,091,845bp long genome has a G+C content of 63.46% and encodes1,849 predicted genes; 1,781 were protein-coding genes, and 68 were RNAs. On the basis of the characteristics reported here, we propose the creation of a new bacterial genus Hugonella gen. nov., belonging to the Eggerthellaceae family and including Hugonella massiliensis gen. nov., sp. nov., strain AT8(T) as the type strain.

Ce travail a pour but d’évaluer la durabilité du projet de siège et centre de recherche de l’entreprise de biotechnologie Serono à Sécheron, une ancienne friche industrielle. Cette évaluation a été effectuée avec un outil que nous avons créé en nous basant sur des outils existants. Notre outil d’évaluation tient compte des impacts environnementaux, sociaux et économiques du projet, sa flexibilité et son intégration ainsi que le comportement éthique de l’entreprise. Ce projet de Serono va permettre une amélioration de la qualité de l’environnement dans l’entreprise et à Sécheron. La gestion des déchets, de l’énergie et de l’eau sera améliorée. Les émissions seront réduites d’un facteur 1,6 de CO2 ; 3,5 de CO ; 6 de NOx et 200 de SO2. Il n’induira pas de nuisances sonores significatives et les impacts sur le sol sont limités. Les activités industrielles prévues ne présentent pas de risque particulier pour le voisinage. Le projet a permis le recyclage de terrains urbains et de bâtiments. Le regroupement du personnel et des activités et la création d’équipements sociaux à Sécheron, qui bénéficiera de nouveaux emplois, vont améliorer les conditions de travail des employés de l’entreprise et pourraient renforcer le lien social. Les opposants au projet, les problèmes de congestion qu’il pourrait créer, les mésententes entre l’entreprise et certains voisins et quelques lacunes dans son comportement éthique ne vont pas dans le sens de la durabilité. Ils ne constitueraient pas une menace assez sérieuse pour contrecarrer le projet. L’impact économique du projet pour l’entreprise et le quartier de Sécheron reste globalement positif. Le projet, assez flexible, a été assez bien intégré dans le quartier. L’impact globalement positif du projet de Serono à Sécheron nous amène à penser qu’il est durable même si quelques efforts devraient être faits pour combler quelques points faibles.
