The objective of this work was to evaluate the properties of beverages formulated with Brazil nuts (Bertholletia excelsa Bonpl) and bocaiuva almonds (Acrocomia aculeata (Jacq.) Lodd. Ex Mart.). Five beverages were developed with Brazil nut, bocaiuva almond, and water (m/m/v), as follows: (i) NB, nut:water, 1:10; (ii) AB, almond:water, 1:10; (iii) NAB1, 1:0.1:10, nut:almond:water; (iv) NAB5, nut:almond:water, 1:0.5:10; and (v) NAB10, nut:almond:water, 1:1:10. The physicochemical, chemical, technological, and microbiological parameters were evaluated. After heat treatment (HT) and enrichment with minerals, the beverages that demonstrated stability in these characteristics were tested in a biological assay. The physical and biochemical parameters of male Wistar rats were evaluated after administering beverages for 28 days. HT decreased the total phenolic content and the antioxidant activity; however, it guaranteed microbiological safety. Mineral supplementation changed the colors and increased the pH values of the beverages. After the beverages were administered, the Wistar rats in the (i) NB group showed decreases in retroperitoneal adipose tissue, total cholesterol, and triglycerides; (ii) those in the AB group exhibited decreased triglycerides contents; and (iii) those in the NAB10-group presented decreased liver weights. The beverages evaluated in this study demonstrate a protective effect against risk factors such as fat accumulation in the liver, retroperitoneal adipose tissue, and hypercholesterolemia.

Enquadramento: A formação em saúde tem sido debate em diferentes cenários especialmente pelas mudanças nos modelos de atenção à saúde, sendo a Educação Interprofissional em Saúde uma estratégia afinada para este fim. Objetivo: Analisar as visões sobre educação interprofissional e os desafios na mudança do modelo de formação profissional, conforme percebido por docentes e precetores de cursos de saúde numa universidade pública do sul do Brasil, através da participação no Programa de Educação pelo Trabalho (PET) - Saúde Interprofissionalidade. Metodologia: Pesquisa qualitativa, do tipo descritiva-exploratória, utilizando entrevistas semiestruturadas e análise temática. Resultados: As experiências no programa provocaram novas perceções sobre o trabalho em equipa, colaborativo e uni profissional destacando a Educação Interprofissional (EIP) como fundamental na formação e trabalho. Conclusão: Integrar a colaboração interprofissional na educação é crucial para mudanças curriculares significativas, como visto no Programa de Educação pelo Trabalho (PET). Isso impulsiona o trabalho colaborativo no Sistema Único de Saúde (SUS) através da Educação Interprofissional (EIP), impactando o perfil profissional na saúde.

This study assessed the safety and efficacy of OncoTherad® (MRB-CFI-1) nanoimmunotherapy for non-muscle invasive bladder cancer (NMIBC) patients unresponsive to Bacillus Calmette-Guérin (BCG) and explored its mechanisms of action in a bladder cancer microenvironment. A single-arm phase I/II study was conducted with 44 patients with NMIBC who were unresponsive to BCG treatment. Primary outcomes were pathological complete response (pCR) and relapse-free survival (RFS). Secondary outcomes comprised response duration and therapy safety. Patients’ mean age was 65 years; 59.1% of them were refractory, 31.8% relapsed, and 9.1% were intolerant to BCG. Moreover, the pCR rate after 24 months reached 72.7% (95% CI), whereas the mean RFS reached 21.4 months. Mean response duration in the pCR group was 14.3 months. No patient developed muscle-invasive or metastatic disease during treatment. Treatment-related adverse events occurred in 77.3% of patients, mostly grade 1–2 events. OncoTherad® activated the innate immune system through toll-like receptor 4, leading to increased interferon signaling. This activation played a crucial role in activating CX3CR1+ CD8 T cells, decreasing immune checkpoint molecules, and reversing immunosuppression in the bladder microenvironment. OncoTherad® has proved to be a safe and effective therapeutic option for patients with BCG-unresponsive NMIBC, besides showing likely advantages in tumor relapse prevention processes.

IntroductionPeanut (Arachis hypogaea L.) is a widespread oilseed crop of high agricultural importance in tropical and subtropical areas. It plays a major role in the food supply in the Democratic Republic of Congo (DRC). However, one major constraint in the production of this plant is the stem rot (white mold or southern blight) disease caused by Athelia rolfsii which is so far controlled mainly using chemicals. Considering the harmful effect of chemical pesticides, the implementation of eco-friendly alternatives such as biological control is required for disease management in a more sustainable agriculture in the DRC as in the other developing countries concerned. Bacillus velezensis is among the rhizobacteria best described for its plant protective effect notably due to the production of a wide range of bioactive secondary metabolites. In this work, we wanted to evaluate the potential of B. velezensis strain GA1 at reducing A. rolfsii infection and to unravel the molecular basis of the protective effect.Results and discussionUpon growth under the nutritional conditions dictated by peanut root exudation, the bacterium efficiently produces the three types of lipopeptides surfactin, iturin and fengycin known for their antagonistic activities against a wide range of fungal phytopathogens. By testing a range of GA1 mutants specifically repressed in the production of those metabolites, we point out an important role for iturin and another unidentified compound in the antagonistic activity against the pathogen. Biocontrol experiments performed in greenhouse further revealed the efficacy of B. velezensis to reduce peanut disease caused by A. rolfsii both via direct antagonism against the fungus and by stimulating systemic resistance in the host plant. As treatment with pure surfactin yielded a similar level of protection, we postulate that this lipopeptide acts as main elicitor of peanut resistance against A. rolfsii infection.

Triterpene saponins (TS) are a structurally diverse group of metabolites that are widely distributed in plants. They primarily serve as defense compounds and their production is often triggered by biotic stresses through signaling cascades that are modulated by phytohormones such as the jasmonates (JA). Two JA-modulated basic helix-loop-helix (bHLH) transcription factors (TFs), triterpene saponin biosynthesis activating regulator 1 (TSAR1) and TSAR2, have previously been identified as direct activators of TS biosynthesis in the model legume Medicago truncatula. Here, we report on the involvement of the core endoplasmic reticulum (ER) stress-related basic leucine zipper (bZIP) TFs bZIP17 and bZIP60 in the regulation of TS biosynthesis. Expression and processing of M. truncatula bZIP17 and bZIP60 proteins were altered in roots with perturbed TS biosynthesis or treated with JA. Accordingly, such roots displayed an altered ER network structure. M. truncatula bZIP17 and bZIP60 proteins were shown to localize in the nucleus and appeared to be capable of interfering with the TSAR-mediated transactivation of TS biosynthesis genes. Furthermore, interference between ER stress-related bZIP and JA-modulated bHLH TFs in the regulation of JA-dependent terpene biosynthetic pathways may be widespread in the plant kingdom, as we demonstrate that it also occurs in the regulation of monoterpene indole alkaloid biosynthesis in the medicinal plant Catharanthus roseus.

The necrotrophic fungus Botrytis cinerea is a major threat to strawberry cultivation worldwide. By screening different Fragaria vesca genotypes for susceptibility to B. cinerea, we identified two genotypes with different resistance levels, a susceptible genotype F. vesca ssp. vesca Tenno 3 (T3) and a moderately resistant genotype F. vesca ssp. vesca Kreuzkogel 1 (K1). These two genotypes were used to identify the molecular basis for the increased resistance of K1 compared to T3. Fungal DNA quantification and microscopic observation of fungal growth in woodland strawberry leaves confirmed that the growth of B. cinerea was restricted during early stages of infection in K1 compared to T3. Gene expression analysis in both genotypes upon B. cinerea inoculation suggested that the restricted growth of B. cinerea was rather due to the constitutive resistance mechanisms of K1 instead of the induction of defense responses. Furthermore, we observed that the amount of total phenolics, total flavonoids, glucose, galactose, citric acid and ascorbic acid correlated positively with higher resistance, while H2O2 and sucrose correlated negatively. Therefore, we propose that K1 leaves are more resistant against B. cinerea compared to T3 leaves, prior to B. cinerea inoculation, due to a lower amount of innate H2O2, which is attributed to a higher level of antioxidants and antioxidant enzymes in K1. To conclude, this study provides important insights into the resistance mechanisms against B. cinerea, which highly depend on the innate antioxidative profile and specialized metabolites of woodland strawberry leaves.

Resultado da pesquisa de Mestrado em Educação e Contemporaneidade realizada no Colégio Estadual Mário Costa Neto/Salvador-BA, a pesquisa considerou os dez anos da Lei Federal 10.639/03 que prevê a obrigatoriedade da temática "História e Cultura Afro-Brasileira" nas escolas. Analisar o currículo, as práxis pedagógicas depois da aplicação da lei, e as relações étnico-raciais dos discentes foram objetivos do trabalho. O Estudo de Caso, a pesquisa documental, observações do espaço escolar e questionário aos alunos foram adotados. Constatou-se que a aplicação da Lei não está ocorrendo efetivamente.

Resumo: Este artigo é resultado de estágio de pesquisa no exterior realizado na Universidade de Essex - Inglaterra, nos meses de outubro e novembro do ano de 2019. Objetiva apresentar algumas aproximações sobre o debate do Serviço Social inglês, especialmente no que diz respeito às tendências do Serviço Social Radical e Neoliberal, bem como apontar alguns dados levantados em visitas institucionais e em entrevistas realizadas com assistentes sociais desse país.

Este trabalho apresenta parte da pesquisa de análise em torno dos estudos rítmicos de José Eduardo Gramani apresentados em Rítmica (1988) e Rítmica Viva (1996). Nossa hipótese é a de que o uso de ostinatos em seus Sambas se assemelha ao uso das timelines na música africana, explorando princípios da rítmica aditiva que geram motivos assimétricos e dialogam com os padrões polirrítmicos da Costa Oeste da África. Tal abordagem desses estudos dentro das perspectivas abertas pela musicologia africana nos permite traçar desdobramentos ligados à performance e à criação musical, estimulando a dissociação de movimentos, percepção e experimentação do fenômeno rítmico.

Jasmonates (JA) are oxylipin-derived phytohormones that trigger the production of specialized metabolites that often serve in defense against biotic stresses. In Medicago truncatula, a JA-induced endoplasmic reticulum-associated degradation (ERAD)-type machinery manages the production of bioactive triterpenes and thereby secures correct plant metabolism, growth, and development. This machinery involves the conserved RING membrane-anchor (RMA)-type E3 ubiquitin ligase MAKIBISHI1 (MKB1). Here, we discovered two additional members of this protein control apparatus via a yeast-based protein–protein interaction screen and characterized their function. First, a cognate E2 ubiquitin-conjugating enzyme was identified that interacts with MKB1 to deliver activated ubiquitin and to mediate its ubiquitination activity. Second, we identified a heat shock protein 40 (HSP40) that interacts with MKB1 to support its activity and was therefore designated MKB1-supporting HSP40 (MASH). MASH expression was found to be co-regulated with that of MKB1. The presence of MASH is critical for MKB1 and ERAD functioning because the dramatic morphological, transcriptional, and metabolic phenotype of MKB1 knock-down M. truncatula hairy roots was phenocopied by silencing of MASH. Interaction was also observed between the Arabidopsis thaliana (Arabidopsis) homologs of MASH and MKB1, suggesting that MASH represents an essential and plant-specific component of this vital and conserved eukaryotic protein quality control machinery.

O objetivo é que este manuscrito possa servir como mola propulsora para o desenvolvimento de estratégias de cuidado que aliem o controle de sintomas às questões relativas à humanização da assistência, possibilitando que as demandas dessa população sejam atendidas, com consequente qualidade de vida e de morte.

Yeast two-hybrid (Y2H) is a well-established genetics-based system that uses yeast to selectively display binary protein-protein interactions (PPIs). To meet the current need to unravel complex PPI networks, several adaptations have been made to establish medium- to high-throughput Y2H screening platforms, with several having successfully incorporated the use of the next-generation sequencing (NGS) technology to increase the scale and sensitivity of the method. However, these have been to date mainly restricted to the use of fully annotated custom-made open reading frame (ORF) libraries and subject to complex downstream data processing. Here, a streamlined Y2H library screening strategy, based on integration of Y2H with NGS, called Y2H-seq, was developed, which allows efficient and reliable screening of Y2H cDNA libraries. To generate proof of concept, the method was applied to screen for interaction partners of two key components of the jasmonate signaling machinery in the model plant Arabidopsis thaliana, resulting in the identification of several previously reported as well as hitherto unknown interactors. Our Y2H-seq method offers a user-friendly, specific and sensitive screening method that allows identification of PPIs without prior knowledge of the organism's ORFs, thereby extending the method to organisms of which the genome has not entirely been annotated yet. The quantitative NGS readout allows to increase genome coverage, thereby overcoming some of the bottlenecks of current Y2H technologies, which will further strengthen the value of the Y2H technology as a discovery platform.

Quais discursos sobre surdez, surdos e próteses auditivas estão presentes nas políticas de governo da atualidade e como esses discursos produzem processos de normalização do sujeito surdo? Para responder essa pergunta, assumimos a perspectiva de Michel Foucault sobre discurso e processos de normalização e analisamos um manual de perguntas e respostas sobre o implante coclear, além de três outros documentos do Ministério da Educação e do Ministério da Saúde. Os excertos dos materiais foram agrupados a partir de recorrências, as quais nos permitiram construir as seguintes unidades temáticas, apresentadas como questionamentos: 1) Cumprindo legalmente? 2) Liberdade de escolha? e 3) Possibilidades de sucesso para quem? Na construção dessas unidades, buscamos mostrar que os discursos analisados constituem sujeitos novos, assim como contribuem para a condução e normalização dos sujeitos surdos na Contemporaneidade por meio de estratégias disciplinares e biopolíticas.

The presence of ants in hospitals is health risk for the hospital community, since presents a strong capability to carry pathogenic organisms throughout this environment. This study was carried out from May 2007 to April 2008, aiming the identification of ant species that infest the hospitals in Porto Alegre, Rio Grande do Sul State, Brazil. A total of 989 ants from 19 species were sampled: Acromyrmex niger (Fr. Smith, 1858), Brachymyrmex sp., Camponotus rufipes (Fabricius, 1775), Camponotus sericeiventris (Guerin-Meneville, 1838), Camponotus sp1, Camponotus sp2, Gnamptogenys sp., Hypoponerasp., Labidus coecus (Latreille, 1802), Monomorium pharaonis (Linnaeus, 1758), Pachycondyla sp., Paratrechina fulva (Mayr, 1862), Paratrechina longicornis Latreille, 1802, Pheidole sp1, Pheidole sp2, Pheidole sp3, Pheidole sp4, Pheidole sp5, and Tapinoma melanocephalum Fabricius, 1793. Among them, Brachymyrmex sp. was the most frequent one (47.0%) followed by Monomorium pharaonis (18.7%) and Labidus coecus (17.2%).A presença de formigas em hospitais constitui risco à saúde da comunidade hospitalar, uma vez que elas possuem o poder de carrearmicrorganismos patogênicos por este ambiente. Este estudo foi realizado no período de maio 2007 a abril de 2008, com o objetivo de identificar as espécies de formigas que infestam hospitais em Porto Alegre, Brasil. Este ensaio pretende comparar alguns aspectos da peça A moratória, de Jorge Andrade, com o romance Pedaços da fome, de Carolina Maria de Jesus. A partir de um mesmo painel social - a passagem do campo para a cidade, tema central em boa parte da literatura brasileira do século XX - procuramos delinear como os conteúdos específicos de cada obra são formalizados. Assim, temos a família latifundiária que perde a fazenda de café e vê a cidade como espaço de degradação, em A moratória; em Pedaços da fome, o ponto de vista do migrante que vê em São Paulo a cidade das oportunidades se mistura com uma trama folhetinesca da qual não está excluída a vingança social. Jorge Andrade, membro da elite paulista, formalizou com distanciamento e de maneira moderna a decadência; já Carolina de Jesus, escritora favelada, se apropriou do romance de maneira reveladora de uma situação social que continua atual.

O presente artigo pontua as principais reflexões apreendidas a partir de pesquisa qualitativa realizada com assistentes sociais inseridos nas Supervisões de Vigilância em Saúde (SUVIS’s) de uma dada região da cidade de São Paulo. Tal estudo aborda como se deu a inserção dos profissionais neste espaço, quais são os trabalhos desenvolvidos, bem como discute a construção histórica da identidade profissional do Serviço Social e como esta se delineia no campo de trabalho específico da SUVIS.

O objetivo deste artigo é a análise de dois romances de Maria José Dupré, escritora da década de 1940. Em Éramos seis e Gina, procuramos articular o contexto social da época à forma das narrativas, preenchendo lacunas deixadas pela crítica nacional, ciosa dos grandes nomes do panteão modernista – o que não constitui propriamente um problema, mas por vezes deixa passar aspectos da relação entre literatura e sociedade que somente os escritores ditos “medianos” são capazes de revelar.

ABSTRACT Objective: To analyze the repercussions of the training of nurses working in the Intensive Care Unit for the management of continuous hemodialysis regarding the safety of critical patients with acute renal failure. Method: Qualitative research developed considering James Reason's reference in the Intensive Care Unit of a private hospital. The data was collected with 23 nurses who worked for more than three months in the management of continuous hemodialysis through a semi-structured interview, and analyzed with the thematic content analysis technique. Results: There are weaknesses in the training of intensive care nurses for the management of continuous hemodialysis that become a latent failure. Such a failure results in difficulties in handling hemodialysis, with risks for these professionals to commit active failures. Final considerations: The training program in service for the management of continuous hemodialysis must be perfected, to develop skills and competencies in nurses and improve their performance.

ABSTRACT Objectives: To identify equipment failures during handling by nurses and analyze the conduct of the professionals when these failures occur. Methods: Descriptive, exploratory and qualitative study, whose field was the intensive care unit of a public institution, and the participants were day nurses that worked providing direct care to patients. Data were produced in 2014 through systematic observation and interviews and were examined with thick description and content analysis. Results: The outcomes evinced the inadequate functioning of infusion bombs, users' errors related to the design of equipment and problems with batteries of artificial fans. These failures related to the management of equipment in the unit. Final considerations: It is necessary to strengthen the monitoring systems of safety conditions of equipment in intensive nursing care to prevent incidents.

ABSTRACT Objective: To describe the Intensive Care Unit nurse’s role in the management of continuous hemodialysis within the scope of the collaborative model, analyzing it regarding the links with patient safety. Method: A descriptive, qualitative, exploratory study based on the Reason safety model performed at the Intensive Care Unit of a specialized hospital, with nurses working in the direct management of continuous hemodialysis, who were interviewed using a script with its contents being thematically analyzed. Results: 23 nurses participated. The role of the intensive care nurse in continuous hemodialysis involves performing preparation/planning and monitoring/follow-up activities, based on interaction with technology and the application of specialized knowledge. The adopted collaborative model reflects on its qualification and availability in relation to the activities that need to be performed, with repercussions on patient safety. Conclusion: There are weaknesses in the participation by intensive care nurses in this continuous hemodialysis model which require elaboration of defensive barriers for the safety of the system.
