BackgroundThe outbreak of SARS-CoV-2 virus has caused a major international health crisis with serious consequences in terms of public health and economy. In France, two lockdown periods were decided in 2020 to avoid the saturation of intensive care units (ICU) and an increase in mortality. The rapid dissemination of variant SARS-CoV-2 VOC 202012/01 has strongly influenced the course of the epidemic. Vaccines have been rapidly developed. Their efficacy against the severe forms of the disease has been established, and their efficacy against disease transmission is under evaluation. The aim of this paper is to compare the efficacy of several vaccination strategies in the presence of variants in controlling the COVID-19 epidemic through population immunity.MethodsAn agent-based model was designed to simulate with different scenarios the evolution of COVID-19 pandemic in France over 2021 and 2022. The simulations were carried out ignoring the occurrence of variants then taking into account their diffusion over time. The expected effects of three Non-Pharmaceutical Interventions (Relaxed-NPI, Intensive-NPI, and Extended-NPI) to limit the epidemic extension were compared. The expected efficacy of vaccines were the values recently estimated in preventing severe forms of the disease (75% and 94%) for the current used vaccines in France (Pfizer-BioNTech and Moderna since January 11, 2021, and AstraZeneca since February 2, 2021). All vaccination campaigns reproduced an advanced age-based priority advised by the Haute Autorité de Santé. Putative reductions of virus transmission were fixed at 0, 50, 75 and 90%. The effects of four vaccination campaign durations (6-month, 12-month, 18-month and 24-month) were compared.ResultsIn the absence of vaccination, the presence of variants led to reject the Relaxed-NPI because of a high expected number of deaths (170 to 210 thousands) and the significant overload of ICUs from which 35 thousand patients would be deprived. In comparison with the situation without vaccination, the number of deaths was divided by 7 without ICU saturation with a 6-month vaccination campaign. A 12-month campaign would divide the number of death by 3 with Intensive-NPI and by 6 with Extended-NPI (the latter being necessary to avoid ICU saturation). With 18-month and 24-month vaccination campaigns without Extended-NPI, the number of deaths and ICU admissions would explode.ConclusionAmong the four compared strategies the 6-month vaccination campaign seems to be the best response to changes in the dynamics of the epidemic due to the variants. The race against the COVID-19 epidemic is a race of vaccination strategy. Any further vaccination delay would increase the need of strengthened measures such as Extended-NPI to limit the number of deaths and avoid ICU saturation.

BackgroundCompartmental models help making public health decisions. They were used during the COVID-19 outbreak to estimate the reproduction numbers and predict the number of hospital beds required. This study examined the ability of closely related compartmental models to reflect equivalent epidemic dynamics.MethodsThe study considered three independently designed compartmental models that described the COVID-19 outbreak in France. Model compartments and parameters were expressed in a common framework and models were calibrated using the same hospitalization data from two official public databases. The calibration procedure was repeated over three different periods to compare model abilities to: i) fit over the whole lockdown; ii) predict the course of the epidemic during the lockdown; and, iii) provide profiles to predict hospitalization prevalence after lockdown. The study considered national and regional coverages.ResultsThe three models were all flexible enough to match real hospitalization data during the lockdown, but the numbers of cases in the other compartments differed. The three models failed to predict reliably the number of hospitalizations after the fitting periods at national as at regional scales. At the national scale, an improved calibration led to epidemic course profiles that reflected hospitalization dynamics and reproduction numbers that were coherent with official and literature estimates.ConclusionThis study shows that prevalence data are needed to further refine the calibration and make a selection between still divergent models. This underlines strongly the need for repeated prevalence studies on representative population samples.

Multivariate extreme value statistical analysis is concerned with observations on several variables which are thought to possess some degree of tail dependence. The main approaches to inference for multivariate extremes consist in approximating either the distribution of block component-wise maxima or the distribution of the exceedances over a high threshold. Although the expressions of the asymptotic density functions of these distributions may be characterized, they cannot be computed in general. In this paper, we study the case where the spectral random vector of the multivariate max-stable distribution has known conditional distributions. The asymptotic density functions of the multivariate extreme value distributions may then be written through univariate integrals that are easily computed or simulated. The asymptotic properties of two likelihood estimators are presented, and the utility of the method is examined via simulation.

Platelets were used to explore the effect of membrane curvature induced by phospholipid excess on cell shape and on organization of the actin cytoskeleton. We showed that the addition of short chain analogues of phospholipids to the outer leaflet of plasma membrane of resting platelets immediately induced a shape change with long filopodia formation containing newly polymerized actin. Cells recovered rapidly their discoid shape and their initial F-actin content only with the phosphatidylserine analogue, which was transported to the inner leaflet by aminophospholipid translocase. Filopodia formation and actin polymerization were inhibited in platelets pre-incubated with cytochalasin D. Both wortmannin and LY294002, two unrelated inhibitors of phosphoinositide 3-kinase, considerably reduced actin polymerization and filopodia formation. Phospholipid imbalance was accompanied by a reversible translocation of phosphoinositide 3-kinase from cytoplasm to plasma membrane. In agreement with a role for PI 3-kinase, when phospholipids were added to platelets, PtdIns(3,4)P2 increased two-fold and Akt protein was partly phosphorylated. A similar shape change was also observed in nocodazole-treated L929 fibroblasts which were incubated with the similar phospholipid analogues. In those nucleated cells, where the microtubule cytoskeleton was disrupted, a major actin-dependent membrane extension was induced by addition of short chain phospholipids that required the functional integrity of PI 3-kinase. We conclude that any physical constraint acting on plasma membrane and resulting on local changes in membrane curvature is sufficient to initiate transient actin polymerization via phosphoinositide 3-kinase activation.

Nous proposons une classe paramétrée de distorsions de probabilités qui permettent, itérées, de s'approcher aussi finement que voulu d'une fonction de survie cible. Par construction, la classe proposée respecte plusieurs propriétés que nous détaillons, et en particulier l'inversibilité analytique et la stabilité dont nous présentons l'intérêt pour certaines études actuarielles. Nous étudions l'impact asymptotique de ces distorsions sur le taux de hasard. Nous formulons les conditions sous lesquelles distorsions proposées constituent la base d'un indicateur de risque. Nous établissons la forme des compositions de distorsions ainsi que la convergence de la loi initiale déformée vers la loi cible. Une méthodologie d'estimation et des valeurs d'initialisations sont proposées. Des applications à l'analyse de la mortalité fournissent des résultats pour la modélisation d'évènements catastrophiques, ainsi que des représentations paramétrées très fidèles de l'évolution des lois de mortalité au fil des ans. Une analyse de l'impact de la paramétrisation choisie est également conduite. Nous suggérons finalement un modèle de simulation de mortalité prospective qui découle des constructions précédentes.

We have investigated phospholipid redistribution, membrane vesicle shedding, shape change, and granule release following A23187 activation of platelets from a patient with Scott syndrome, characterized by impaired transmembrane migration of phosphatidylserine (PS) accompanied by haemorrhagic complications, and two of her children. Electron spin resonance spectroscopy measurement of phospholipids redistribution showed that the internalization of PS was unaffected by the disorder but, after activation, PS exposure was significantly reduced in platelets from the homozygous-type patient. Vesicle shedding was also reduced in these platelets. However, the slow redistribution of phosphatidylcholine was similar to that observed in normal platelets. When treated with calpeptin, platelets from the homozygous-type patient, unlike normal or heterozygous Scott syndrome platelets, showed a smoothly rounded shape without filopods after activation. Following A23187 activation of normal platelets, filopod formation was consecutive to the re-exposition of aminophospholipids on the outer leaflet of the plasma membrane, and the existence of a floppase (outward aminoPLs translocase) has been suggested. In homozygous Scott syndrome platelets the deficiency in PS re-exposition, the absence of filopod formation, and low vesicle shedding are correlated with each other, and argue in favour of a disruption of the proposed floppase activity.

A Ru(II) complex of an unsymmetrical macrocycle (containing two tertiary amine nitrogen atoms acting as ligands) exhibits a marked catalytic activity for disproportionation of hydrogen peroxide, with both high rate and turnover number. Although the stoichiometry is the same as for catalase, the reaction does not obey the simple kinetic law reported for this enzyme. The proposed reaction scheme includes a first slow step leading to a catalytic species which undergoes a catalase-like redox cycle. This scheme fits the experimental kinetic data measured in large concentration ranges of hydrogen peroxide and catalyst. The nature of the active species is discussed.

International audience; We introduce a parametric class of composite probability distortions that can be combined to converge to a target survival function. These distortions respect analytic invertibility and stability, which are shown to be relevant in many actuarial fields. We study the asymptotic impact of such distortions on hazard rates. The paper provides an estimation methodology, including hints for initialisation. Some applications to survival data bring results for catastrophic event impact modelling. We also obtain accurate parametric representations of the mortality trend over years. Finally, we suggest a prospective mortality simulation model that comes naturally from the above analysis.

Platelets were incubated in the presence of calpeptin to inhibit calpain-mediated cytoskeleton proteolysis during further activation by Ca2+ ionophore A23187. The appearance of filamin and myosin subfragments (93 kDa and 135 kDa, respectively) was inhibited by low calpeptin doses (1 microgram/ml). Higher doses (10-20 micrograms/ml) were required to completely inhibit talin and filamin degradation. Vesiculation strongly depended on cytoskeleton proteolysis and was reduced by 60% when platelets were preincubated with 10 micrograms/ml calpeptin. Activated platelets bore longer and more filopods when pretreated with calpeptin. Filopods were straight and regular when high calpeptin doses were used, whereas they were shorter and broader with bloated surfaces when calpeptin was omitted. Some bloated areas were also found in straight filopods. These results suggest that the cytoskeleton proteolysis, and more specifically filamin proteolysis, induced bloating of filopod surfaces, thus facilitating fragmentation of filopod into vesicles.

The aim of this study is to investigate the intracellular delivery of polynucleotides by fusogenic immunoliposomes. We have studied the internalization of poly(rI)-poly(rC) (polyIC) by liposomes into murine L929 cells. The liposomes were prepared by incorporating Sendaï virus fusogenic F protein into the lipid bilayer and targeted by a monoclonal antibody (mAb) bound to the liposomes via protein A (Staphylococcus aureus). The immunoliposomes ensured a sufficient yield of polyIC internalization, which was estimated by its ability to induce antiviral activity. In the absence of RNase treatment free and encapsulated polyIC had the same inducing effect, but in the presence of nuclease only the encapsulated polyIC, and not free polyIC, maintained its antiviral effect. The fusion process making possible the internalization of polyIC was confirmed by the fact that the polyIC effect was mainly inhibited by an anti-F protein mAb which inhibited erythrocyte hemolysis by the virus.

We introduce the systemic tail risk distribution of a financial market to characterize the asset return linkages during financial crisis. This distribution provides the probabilities that several assets of a market lose a large part of their nominal value given that the price of at least one of them collapses. It introduces a new way of assessing the stability of a financial market during potential systemic risk events. We propose a new type of multivariate extreme value distribution for high-dimensional vectors to model the extremal dependence between asset prices, and we use efficient likelihood inference methods to estimate the parameters of the systemic tail risk distribution. Our real data show that the empirical static systemic tail risk distribution is U-shaped, while the empirical conditional distribution is L-shaped.

The electron paramagnetic (EPR) properties of a transient species detected during a cytochrome P450 (P450)-mediated peroxidative reaction have been compared with those of peroxidases Compound I and model metalloporphyrins. The reaction which was studied with cholesterol-specific P450scc and (20R)-20-hydroperoxycholesterol, occured without enzyme denaturation. The resulting transient species, which reached its maximum after 50 s reaction, was characterized by a one-line EPR spectrum, g = 2.0035, Δ 1 2 = 1.08 mT . The decay of this radical was concomitant with the production of (20R)-20,21-dihydroxycholesterol. The reaction (almost 100% yield) conserved the stereo-specificity of the natural pathway. We suggest this intermediate is a candidate for the in vivo ultimate oxidant in the P450-mediated hydroxylation process. The comparison of the observed EPR spectrum with those of peroxidase Compound I and related synthetic models allows us to propose a FeIV porphyrin II cation radical structure for the intermediate.

International audience; We demonstrated that benzyl alcohol, a neutral local anesthetic drug, inhibits the uptake and degradation of lowdensity lipoprotein and endocytosis of transferrin receptors of guinea pig leukemic B lymphocytes (L2C). This inhibition is very rapid, concentration dependant and reversible by simple washing. Membrane fluidity of the living cells is also modified.

We study the asymptotic behavior of a mutation--selection genetic algorithm on the integers with finite population of size $p\ge 1$. The mutation is defined by the steps of a simple random walk and the fitness function is linear. We prove that the normalized population satisfies an invariance principle, that a large-deviations principle holds and that the relative positions converge in law. After $n$ steps, the population is asymptotically around $\sqrt{n}$ times the position at time $1$ of a Bessel process of dimension $2p-1$.

International audience; Nonlinear multimodal filtering problems are usually addressed via Monte Carlo algorithms. These algorithms involve sampling procedures that are similar to proportional selection in genetic algorithms, and that are prone to failure due to genetic drift. This work investigates the feasibility and the relevance of niching strategies in this context. Sharing methods are evaluated experimentally, and prove to be efficient in such issues.

International audience; Regulatory features of protein-induced membrane fusion are largely unclear, particularly at the level of the fusion peptide. Fusion peptides being part of larger protein complexes, such investigations are met with technical limitations. Here, we show that the fusion activity of influenza virus or Golgi membranes is strongly inhibited by minor amounts of (lyso)lipids when present in the target membrane but not when inserted into the viral or Golgi membrane itself. To investigate the underlying mechanism, we employ a membrane-anchored peptide system and show that fusion is similarly regulated by these lipids when inserted into the target but not when present in the peptide-containing membrane. Peptide-induced fusion is regulated by a reversible switch of secondary structure from a fusion-permissive alpha-helix to a nonfusogenic beta-sheet. The "on/off" activation of this switch is governed by minor amounts of (lyso)-phospholipids in targets, causing a drop in alpha-helix and a dramatic increase in beta-sheet contents. Concomitantly, fusion is inhibited, due to impaired peptide insertion into the target membrane. Our observations in biological fusion systems together with the model studies suggest that distinct lipids in target membranes provide a means for regulating membrane fusion by causing a reversible secondary structure switch of the fusion peptides.Regulatory features of protein-induced membrane fusion are largely unclear, particularly at the level of the fusion peptide. Interestingly, the susceptibility of the various PC species toward WAE-induced fusion matched a similar order of increase in intrinsic lipid headgroup spacing of the target membrane. The degree of spacing, in turn, was found to be related to the extent by which the fluorescence quantum yield of the Trp residue increased, which occurred upon the interaction of WAE with target membranes. Therefore, these results demonstrate an enhanced ability for WAE to engage in hydrophobic interactions when headgroup spacing increases. Thus, this latter parameter most likely regulates the degree of penetration of WAE into the target membrane. To simulate protein-mediated fusion, many studies on peptide-induced membrane fusion have been conducted on model membranes such as liposomes and have employed synthetic peptides corresponding to the putative fusion sequences of viral proteins, or de novo synthesized peptides. Here, the application of peptides as a model system to understand the molecular details of membrane fusion will be discussed in detail. Data obtained from these studies will be correlated to biological studies, in particular those that involve viral and sperm-egg systems. Structure-function relationships will be revealed, particularly in the context of protein-induced membrane perturbations and bilayer-to-nonbilayer transition underlying the mechanism of fusion. International audience; We recently demonstrated that an amphipathic net-negatively charged peptide consisting of 11 amino acids (WAE 11) strongly promotes fusion of large unilamellar liposomes (LUV) when anchored to a liposomal membrane [Pecheur, E. I., Hoekstra, D., Sainte-Marie, J., Maurin, L., Bienvenue, A., and Philippot, J. R. (1997) Biochemistry 36, 3773-3781]. To elucidate a potential relationship between peptide structure and its fusogenic properties and to test the hypothesis that specific structural motifs are a prerequisite for WAE-induced fusion, three 11-mer WAE-peptide analogues (WAK, WAEPro, and WAS) were synthesized and investigated for their structure and fusion activity. Structural analysis of the synthetic peptides by infrared attenuated total reflection spectroscopy reveals a distinct propensity of each peptide toward a helical structure after their anchorage to a liposomal surface, emphasizing the importance of anchorage on conveying a secondary structure, thereby conferring fusogenicity to these peptides. However, whereas WAE and WAK peptides displayed an essentially nonleaky fusion process, WAS- and WAEPro-induced fusion was accompanied by substantial leakage. It appears that peptide helicity as such is not a sufficient condition to convey optimal fusion properties to these 11-mer peptides. Studies of changes in the intrinsic Trp fluorescence and iodide quenching experiments were carried out and revealed the absence of migration of the Trp residue of WAS and WAEPro to a hydrophobic environment, upon their interaction with the target membranes. These results do not support the penetration of both peptides as their mode of membrane interaction and destabilization but rather suggest their folding along the vesicle surface, posing them as surface-seeking helixes. This is in striking contrast to the behavior observed for WAE and WAK, for which at least partial penetration of the Trp residue was demonstrated. These results indicate that subtle differences in the primary sequence of a fusogenic peptide could induce dramatic changes in the way the peptide interacts with a bilayer, culminating in equally drastic changes in their functional properties. The data also reveal a certain degree of sequence specificity in WAE-induced fusion.We recently demonstrated that an amphipathic net-negatively charged peptide consisting of 11 amino acids (WAE 11) strongly promotes fusion of large unilamellar liposomes (LUV) when anchored to a liposomal membrane [Pecheur, E. I., Hoekstra, D., Sainte-Marie, J., Maurin, L., Bienvenue, A., and Philippot, J. R. (1997) Biochemistry 36, 3773-3781]. To elucidate a potential relationship between peptide structure and its fusogenic properties and to test the hypothesis that specific structural motifs are a prerequisite for WAE-induced fusion, three 11-mer WAE-peptide analogues (WAK, WAEPro, and WAS) were synthesized and investigated for their structure and fusion activity. Structural analysis of the synthetic peptides by infrared attenuated total reflection spectroscopy reveals a distinct propensity of each peptide toward a helical structure after their anchorage to a liposomal surface, emphasizing the importance of anchorage on conveying a secondary structure, thereby conferring fusogenicity to these peptides. However, whereas WAE and WAK peptides displayed an essentially nonleaky fusion process, WAS- and WAEPro-induced fusion was accompanied by substantial leakage. It appears that peptide helicity as such is not a sufficient condition to convey optimal fusion properties to these 11-mer peptides. Studies of changes in the intrinsic Trp fluorescence and iodide quenching experiments were carried out and revealed the absence of migration of the Trp residue of WAS and WAEPro to a hydrophobic environment, upon their interaction with the target membranes. These results do not support the penetration of both peptides as their mode of membrane interaction and destabilization but rather suggest their folding along the vesicle surface, posing them as surface-seeking helixes. This is in striking contrast to the behavior observed for WAE and WAK, for which at least partial penetration of the Trp residue was demonstrated. These results indicate that subtle differences in the primary sequence of a fusogenic peptide could induce dramatic changes in the way the peptide interacts with a bilayer, culminating in equally drastic changes in their functional properties. Pseudomonas exotoxin (PE) is a cytotoxin which, after endocytosis, is delivered to the cytosol where it inactivates protein synthesis. Using diaminobenzidine cytochemistry, we found over 94% of internalized PE in transferrin (Tf) -positive endosomes of lymphocytes. When PE translocation was examined in a cell-free assay using purified endocytic vesicles, more than 40% of endosomal125I-labeled PE was transported after 2 h at 37°C, whereas a toxin inactivated by point mutation in its translocation domain was not translocated. Sorting of endosomes did not allow cell-free PE translocation, whereas active PE transmembrane transport was observed after > 10 min of endocytosis when PE and fluorescent-Tf were localized by confocal immunofluorescence microscopy within a rab5-positive and rab4- and rab7-negative recycling compartment in the pericentriolar region of the cell. Accordingly, when PE delivery to this structure was inhibited using a 20°C endocytosis temperature, subsequent translocation from purified endosomes was impaired. Translocation was also inhibited when endosomes were obtained from cells labeled with PE in the presence of brefeldin A, which caused fusion of translocation-competent recycling endosomes with translocation-incompetent sorting elements. No PE processing was observed in lymphocyte endosomes, the full-sized toxin was translocated and recovered in an enzymatically active form. ATP hydrolysis was found to directly provide the energy required for PE translocation. Inhibitors of endosome acidification (weak bases, protonophores, or bafilomycin A1) when added to the assay did not significantly affect125I-labeled PE translocation, demonstrating that this transport is independent of the endosome-cytosol pH gradient. Nevertheless, when125I-labeled PE endocytosis was performed in the presence of one of these molecules, translocation from endosomes was strongly inhibited, indicating that exposure to acidic pH is a prerequisite for PE membrane traversal. When applied during endocytosis, treatments that protect cells against PE intoxication (low temperatures, inhibitors of endosome acidification, and brefeldin A) impaired125I-labeled PE translocation from purified endosomes. We conclude that PE translocation from a late receptor recycling compartment is implicated in the lymphocyte intoxication procedure.

International audience; The carboxylic antibiotic lasalocid was shown to facilitate the transport of Cd(II) cations through supported liquid membranes, the driving force being the proton gradient between the two aqueous phases. With hydrocarbons as solvents inside the porous support, the initial metal fluxes were linear versus the inverses of the viscosity values, in accordance with a diffusion limited mechanism. With the polar solvent nitrophenyl octyl ether, the initial flux was six times higher than expected from the viscosity value, and up to 95% of the metal ions were transported. In competitive experiments with equal initial Cd(II) and Zn(II) concentrations, after 94% of Cd(II) had been transported the ratio of Cd(II) to Zn(II) in the receiving phase was 3.8 (pH 8.2 in the source phase, pH 2 in the receiving phase, membrane impregnated with lasalocid 5×10−3 M in nitrophenyl octyl ether).

The coupled peptide, present in one population, could thus cause vesicle aggregation via nonspecific electrostatic interaction with PElys. However, the free peptide failed to induce aggregation of PElys vesicles, suggesting that the spatial orientation of the coupled peptide codetermined its ability to bring about vesicle aggregation and fusion. With the monitoring of changes in the intrinsic Trp fluorescence, in conjunction with KI-quenching studies, it would appear that hydrophobic interactions facilitate the fusion event, possibly involving (partial) peptide penetration. Ca2+ chelators, such as EGTA or 1,2-bis(2-aminophenoxy)ethane-N,N,N,N'-tetraacetic acid, and a Ca2+-channel inhibitor (Ni2+) decreased drastically the recycling rate of Tf-R. Tf-R recycling was shown to be slowed by a calmodulin antagonist. Conversely, artificial elevation of free internal Ca2+ in L2C cells, using lectin, accelerated the recycling rate. These results suggest that the intracellular Ca2+ concentration plays an important role in the outward flow of transferrin receptors. The internalization and recycling kinetics of transferrin receptors in leukemic lymphocytes (L2C) differed in the absence or presence of ligand. At 37 degrees C in the absence of ligand, transferrin receptors were mainly distributed internaly. We demonstrated, using a sepharose-bead-Tf complex, the rapid recycling of unoccupied internal transferrin receptors was correlated with ligand binding to surface receptors. The recycling amplitude was related to the occupancy of iron ligated transferrin to plasma membrane surface receptors. Contrary to the results obtained with other cells, redistribution of Tf receptors was not triggered by binding of other ligands to their receptors.
