In order to gain insight into the hydrochemical characteristics and formation mechanism of high fluoride groundwater in Zhuanlongwan Coal Mine, we conducted a comprehensive analysis of 48 samples including quaternary groundwater (QGW), sandstone groundwater (SGW), and mine water (MGW) using statistical analysis and hydrochemical methods. The findings demonstrate that the groundwater in Zhuanlongwan Coal Mine generally exhibits weak alkalinity, with predominant cations comprising of Ca2+ and Na+, and anions consisting of HCO3– and Cl–. The mean mass concentration of SO4 2–, Cl–, TDS, pH, and CO3 2– in groundwater follows the order of MGW>SGW>QGW, whereas HCO3– and F– exhibit the trend of SGW>MGW>QGW. Specifically, the average mass concentration of F– in QGW, SGW, and MGW is 0.76 mg/L, 4.55 mg/L, and 4.35 mg/L, respectively. Notably, most samples from SGW and MGW significantly exceed the permissible F−concentration threshold of 1.0 mg/L. The hydrochemical type of low fluoride (\begin{document}$\rho_{{\mathrm{F}}^{-}} $\end{document}1 mg/L) are mainly HCO3−Na (35.42%) and SO4·Cl−Na (45.83%). The dissolution of fluorinated minerals serves as the main source of F– in the high fluoride groundwater of Zhuanlongwan Coal Mine. Moreover, cation exchange, competitive adsorption, as well as the dissolution and precipitation of dolomite and calcite, are significant factors influencing the occurrence of high fluoride groundwater.

Abstract The origin of energetic charged particles in universe remains an unresolved issue. Astronomical observations combined with simulations have provided insights into particle acceleration mechanisms, including magnetic reconnection acceleration, shock acceleration, and stochastic acceleration. Recent experiments have also confirmed that electrons can be accelerated through processes such as magnetic reconnection and collisionless shock formation. However, laboratory identifying stochastic acceleration as a feasible mechanism is still a challenge, particularly in the creation of collision-free turbulent plasmas. Here, we present experimental results demonstrating kinetic turbulence with a typical spectrum k −2.9 originating from Weibel instability. Energetic electrons exhibiting a power-law distribution are clearly observed. Simulations further reveal that thermal electrons undergo stochastic acceleration through collisions with multiple magnetic islands-like structures within the turbulent region. This study sheds light on a critical transition period during supernova explosion, where kinetic turbulences originating from Weibel instability emerge prior to collisionless shock formation. Our results suggest that electrons undergo stochastic acceleration during this transition phase.

Abstract Photothermal therapy (PTT) is a promising cancer treatment method due to its ability to induce tumor-specific T cell responses and enhance therapeutic outcomes. However, incomplete PTT can leave residual tumors that often lead to new metastases and decreased patient survival in clinical scenarios. This is primarily due to the release of ATP, a damage-associated molecular pattern that quickly transforms into the immunosuppressive metabolite adenosine by CD39, prevalent in the tumor microenvironment, thus promoting tumor immune evasion. This study presents a photothermal nanomedicine fabricated by electrostatic adsorption among the Fe-doped polydiaminopyridine (Fe-PDAP), indocyanine green (ICG), and CD39 inhibitor sodium polyoxotungstate (POM-1). The constructed Fe-PDAP@ICG@POM-1 (FIP) can induce tumor PTT and immunogenic cell death when exposed to a near-infrared laser. Significantly, it can inhibit the ATP-adenosine pathway by dual-directional immunometabolic regulation, resulting in increased ATP levels and decreased adenosine synthesis, which ultimately reverses the immunosuppressive microenvironment and increases the susceptibility of immune checkpoint blockade (aPD-1) therapy. With the aid of aPD-1, the dual-directional immunometabolic regulation strategy mediated by FIP can effectively suppress/eradicate primary and distant tumors and evoke long-term solid immunological memory. This study presents an immunometabolic control strategy to offer a salvage option for treating residual tumors following incomplete PTT.

Abstract Betatron x-ray sources from laser-plasma interaction are characterized by compactness, ultrashort duration, broadband spectrum and micron source size. However, high-quality measurements with good statistics, especially in a single shot, require fluxes and energies beyond the current capabilities. Here, we propose a method to enhance the flux and brightness of the betatron sources without increasing the laser energy. By irradiating an edge of a microtape target with a femtosecond laser, a strong surface plasma wave (SPW) is excited at the edge and travels along the lateral plasma-vacuum interfaces. Tens of nC of electrons are peeled off and accelerated to superponderomotive energies by the longitudinal field of the SPW, whilst undergoing transverse betatron oscillations, leading to emission of hard x-rays. Via three-dimensional particle-in-cell simulations, we demonstrate that a tabletop 100 TW class femtosecond laser can produce an ultrabright hard x-ray pulse with flux up to 107 photons eV−1 and brilliance about 1023 photons s−1 mm−2 mrad−2 0.1%BW−1, paving the way for single-shot x-ray measurements in ultrafast science and high-energy-density physics.

Abstract Background We tested the hypothesis that radiofrequency ablation (RFA) for hepatocellular carcinoma (HCC) promotes tumor cell release and explored a method for reducing these effects. Methods A green fluorescent protein-transfected orthotopic HCC model was established in 99 nude mice. In vivo flow cytometry was used to monitor circulating tumor cell (CTC) dynamics. Pulmonary fluorescence imaging and pathology were performed to investigate lung metastases. First, the kinetics of CTCs during the periablation period and the survival rate of CTCs released during RFA were investigated. Next, mice were allocated to controls, sham ablation, or RFA with/without hepatic vessel blocking (ligation of the portal triads) for evaluating the postablation CTC level, lung metastases, and survival over time. Moreover, the kinetics of CTCs, lung metastases, and mice survival were evaluated for RFA with/without ethanol injection. Pathological changes in tumors and surrounding parenchyma after ethanol injection were noted. Statistical analysis included t-test, ANOVA, and Kaplan-Meier survival curves. Results CTC counts were 12.3-fold increased during RFA, and 73.7% of RFA-induced CTCs were viable. Pre-RFA hepatic vessel blocking prevented the increase of peripheral CTCs, reduced the number of lung metastases, and prolonged survival (all p ≤ 0.05). Similarly, pre-RFA ethanol injection remarkably decreased CTC release during RFA and further decreased lung metastases with extended survival (all p ≤ 0.05). Histopathology revealed thrombus formation in blood vessels after ethanol injection, which may clog tumor cell dissemination during RFA. Conclusion RFA induces viable tumor cell dissemination, and pre-RFA ethanol injection may provide a prophylactic strategy to reduce this underestimated effect. Relevance statement RFA for HCC promotes viable tumor cell release during ablation, while ethanol injection can prevent RFA induced tumor cell release. Key points • RFA induced the release of viable tumor cells during the ablation procedure in an animal model. • Hepatic vessel blocking can suppress tumor cells dissemination during RFA. • Ethanol injection can prevent RFA-induced tumor cell release, presumably because of the formation of thrombosis. Abstract In laser-driven inertial confinement fusion, driving pressure boosting and smoothing are major challenges. A proposed hybrid-drive (HD) scheme can offer such ideal HD pressure performing stable implosion and nonstagnation ignition. Here we report that in the hemispherical and planar ablator targets installed in the semicylindrical hohlraum scaled down from the spherical hohlraum of the designed ignition target, under indirect-drive (ID) laser energies of ~43–50 kJ, the peak radiation temperature of 200 ± 6 eV is achieved. And using only direct-drive (DD) laser energies of 3.6–4.0 kJ at an intensity of 1.8 × 1015 W/cm2, in the hemispherical and planar targets the boosted HD pressures reach 3.8–4.0 and 3.5–3.6 times the radiation ablation pressure respectively. In all the above experiments, significant HD pressure smoothing and the important phenomenon of how a symmetric strong HD shock suppresses the asymmetric ID shock pre-compressed fuel are demonstrated. The backscattering and hot-electron energy fractions both of which are about one-third of that in the DD scheme are also measured.

Plant–soil feedback (PSF) was initially developed in the field of agricultural practices. In recent years, PSF has been extended to various ecosystems. Root exudates, essential for the exchange of materials, energy, and information at the plant–soil interface, significantly influence PSF. However, how PSF is driven by root secretions and the role of these secretions in different PSF pathways still needs to be further explored, particularly in forest ecosystems. Soil nutrients, microbial communities, and nematodes are important research topics in the process of PSF driven by root exudates. Investigating these aspects driven by root exudates provides valuable insights into the complex interactions both above ground and below the surface. This research can offer theoretical support and guidance for building stable, healthy, and sustainable forest ecosystems in the future.

Background: Actin-related protein 2/3 complex subunit 1B (ARPC1B) is reported to be involved in tumorigenesis and progression. However, its role in kidney renal clear cell carcinoma (KIRC), correlation with tumor-infiltrating immune cells, and prognostic significance remain unclear.Methods: Data sets from the TCGA, GTEx, GEPIA, GEO, UALCAN, and CPTAC databases were extracted and analyzed to investigate the expression difference, prognosis, and clinicopathological features of ARPC1B. Single-sample Gene Set Enrichment Analysis (ssGSEA), CIBERSORT, and TISCH2 analysis were used to examine the relationship between ARPC1B expression and tumor immune infiltration in KIRC. The potential function of ARPC1B in KIRC was explored by GO functional annotation and KEGG pathway analysis. The TIDE algorithm was used to predict and analyze the relationship between ARPC1B expression and response to immune checkpoint blockade (ICB). The expression of ARPC1B was further validated by using quantitative real-time polymerase chain reaction (qRT-PCR) and immunohistochemistry (IHC).Results: The study showed that ARPC1B expression was an independent prognostic factor of KIRC, with high ARPC1B expression being associated with poor overall survival (OS). Enrichment of GO annotation and pathway analysis showed multiple immune-related functional pathways affected by ARPC1B such as regulation of immune effector process, inflammatory response regulation, antigen processing and presentation, asthma, autoimmune thyroid disease, graft versus host disease, intestinal immune network for IgA production, and type I diabetic mellitus. Moreover, ARPC1B expression positively correlated with infiltrating levels of myeloid-derived suppressor cells (MDSCs) and regulatory T cells (Tregs) in KIRC. Importantly, high ARPC1B expression predicted a low response to ICB in KIRC.Conclusion: This study indicates that ARPC1B expression is an independent prognostic biomarker for OS in KIRC patients. High ARPC1B expression is closely associated with MDSCs and Tregs infiltration. These findings suggest that ARPC1B may serve as a biomarker for prognosis and immune infiltration in KIRC, potentially aiding in the development of novel treatment strategies to improve the survival outcomes for KIRC patients.

Abstract Background Combined therapy based on the effects of cascade reactions of nanoplatforms to combat specific solid tumor microenvironments is considered a cancer treatment strategy with transformative clinical value. Unfortunately, an insufficient O2 supply and the lack of a visual indication hinder further applications of most nanoplatforms for solid tumor therapy. Results A visualizable nanoplatform of liposome nanoparticles loaded with GOD, H(Gd), and PFP and grafted with the peptide tLyP-1, named tLyP-1H(Gd)-GOD@PFP, was constructed. The double-domain peptide tLyP-1 was used to specifically target and penetrate the tumor cells; then, US imaging, starvation therapy and sonodynamic therapy (SDT) were then achieved by the ultrasound (US)-activated cavitation effect under the guidance of MR/PA imaging. GOD not only deprived the glucose for starvation therapy but also produced H2O2, which in coordination with 1O2 produced by H(Gd), enable the effects of SDT to achieve a synergistic therapeutic effect. Moreover, the synergistic therapy was enhanced by O2 from PFP and low-intensity focused ultrasound (LIFU)-accelerated redox effects of the GOD. The present study demonstrated that the nanoplatform could generate a 3.3-fold increase in ROS, produce a 1.5-fold increase in the maximum rate of redox reactions and a 2.3-fold increase in the O2 supply in vitro, and achieve significant tumor inhibition in vivo. Conclusion We present a visualizable nanoplatform with tumor-penetrating ability that can be unlocked by US to overcome the current treatment problems by improving the controllability of the O2 supply, which ultimately synergistically enhanced cascade therapy.

Objectives To examine the prognostic value of preoperative alfa-fetoprotein (AFP) density and other clinical factors in patients undergoing percutaneous radiofrequency ablation (RFA) of hepatocellular carcinoma (HCC).Methods From January 2010 to December 2018, a total of 543 patients undergoing RFA for HCC meeting the Milan criteria were included at our institution. AFP density was calculated as absolute AFP pre-ablation divided by the total volume of all HCC lesions. The survival rates according to AFP density were estimated using the Kaplan–Meier method and compared using the log-rank test. Univariate and multivariate Cox proportional-hazards regression analyses were used to assess predictors of overall survival (OS) and progression-free survival (PFS).Results The Kaplan–Meier 1-, 3-, and 5-year OS rates were 98.8%, 88.5%, and 70.4%, respectively, for the low AFP density group, and 98.3%, 74.9%, and 49.4%, respectively, for the high AFP density group. The corresponding PFS rates were 78.9%, 56.7%, and 40.9% (low AFP density group), and 63.6%, 40.8%, and 27.5% (high AFP density group). Abstract Signal‐amplified imaging of microRNAs (miRNAs) is a promising strategy at the single‐cell level because liquid biopsy fails to reflect real‐time dynamic miRNA levels. However, the internalization pathways for available conventional vectors predominantly involve endo‐lysosomes, showing nonideal cytoplasmic delivery efficiency. In this study, size‐controlled 9‐tile nanoarrays are designed and constructed by integrating catalytic hairpin assembly (CHA) with DNA tile self‐assembly technology to achieve caveolae‐mediated endocytosis for the amplified imaging of miRNAs in a complex intracellular environment. Compared with classical CHA, the 9‐tile nanoarrays possess high sensitivity and specificity for miRNAs, achieve excellent internalization efficiency by caveolar endocytosis, bypassing lysosomal traps, and exhibit more powerful signal‐amplified imaging of intracellular miRNAs. Because of their excellent safety, physiological stability, and highly efficient cytoplasmic delivery, the 9‐tile nanoarrays can realize real‐time amplified monitoring of miRNAs in various tumor and identical cells of different periods, and imaging effects are consistent with the actual expression levels of miRNAs, ultimately demonstrating their feasibility and capacity. This strategy provides a high‐potential delivery pathway for cell imaging and targeted delivery, simultaneously offering a meaningful reference for the application of DNA tile self‐assembly technology in relevant fundamental research and medical diagnostics.

Abstract Background The lack of a satisfactory strategy for postoperative pain management significantly impairs the quality of life for many patients. However, existing nanoplatforms cannot provide a longer duration of nerve blockage with intensity-adjustable characteristics under imaging guidance for clinical applications. Results To overcome this challenge, we proposed a biocompatible nanoplatform that enables high-definition ultrasound imaging-guided, intensity-adjustable, and long-lasting analgesia in a postoperative pain management model in awake mice. The nanoplatform was constructed by incorporating perfluoropentane and levobupivacaine with red blood cell membranes decorated liposomes. The fabricated nanoplatform can achieve gas-producing and can finely escape from immune surveillance in vivo to maximize the anesthetic effect. The analgesia effect was assessed from both motor reactions and pain-related histological markers. The findings demonstrated that the duration of intensity-adjustable analgesia in our platform is more than 20 times longer than free levobupivacaine injection with pain relief for around 3 days straight. Moreover, the pain relief was strengthened by repeatable ultrasound irradiation to effectively manage postoperative pain in an intensity-adjustable manner. No apparent systemic and local tissue injury was detected under different treatments. Conclusion Our results suggest that nanoplatform can provide an effective strategy for ultrasound imaging-guided intensity-adjustable pain management with prolonged analgesia duration and show considerable transformation prospects.

Abstract Calamansi or Philippine lime (Citrofortunella macrocarpa) is an important crop for local economic in Hainan Island. There is no study about Calamansi germplasm evaluation and cultivar development. In this study, Calamansi data were collected from 151 of Calamansi seedling trees, and 37 phenotypic traits were analyzed to investigate their genetic diversities. The cluster analysis and principal component analysis were conducted aiming to provide a theoretical basis for the Calamansi genetic improvement. The results of the diversity analysis revealed: (1) the diversity indexes for qualitative traits were ranged from 0.46–1.39, and the traits with the highest genetic diversity level were fruit shaped and pulp colored (H′ > 1.20); and the diversity indexes for quantitative traits ranged from 0.67–2.10, with the exception of a lower in fruit juice rate (1.08) and lower in number of petals (0.67). (2) The clustering analysis of phenotypic traits have arranged the samples into 4 categories: the first group characterized by fewer flesh Segment number per fruit (SNF) and more Oil cell number (OCN); the second group had 7 samples, all characterized with larger Crown breadth (CB), higher Yield per tree (YPT), the lager leaf, the higher Ascorbic acid (AA), and less Seed number per fruit (SNPF); the third group had 25 samples characterized by smaller Tree foot diameter (TFD),smaller Fruit shape index (FSI) and higher Total soluble solids (TSS) contain; the fourth group had 87 samples, they were characterized by shorter Petiole length (PEL), larger fruit, higher Juice ratio (JR), multiple Stamen number (SN) and longer Pistil length (PIL). (3) The principal component analysis showed the values of the first 9 major components characteristic vectors were all greater than 3, the cumulative contribution rate reach 72.20%, including the traits of single fruit weight, fruit diameter, tree height, tree canopy width etc. Finally, based on the comprehensive main component value of all samples, the Calamansi individuals with higher testing scores were selected for further observation. This study concludes that Calamansi seedling populations in the Hainan Island holds great genetic diversity in varies traits, and can be useful for the Calamansi variety improvements.

A photoelectrochemical (PEC) detection platform was built based on the branched rutile/anatase titanium dioxide (RA-TiO2) electrode. Theoretical calculations proved that the type-II band alignment of rutile and anatase could facilitate charge separation in the electrode. The self-generated electric field at the interface of two phases can enhance the electron transfer efficiency of the electrode. Carboxylated CdTe quantum dots (QDs) were applied as signal amplification factors. Without the target DNA presence, the CdTe QDs were riveted to the surface of the electrode by the hairpin probe DNA. The sensitization of CdTe QDs increased the photocurrent of the electrode significantly. When the target DNA was present, the structural changes of the hairpin probe DNA resulted in the failure of the sensitized structure. Benefiting from excellent electrode structure design and CdTe QDs sensitization strategy, the PEC assays could achieve highly sensitive and specific detection of target DNA in the range of 1 fM to 1 nM, with a detection limit of 0.23 fM. The electrode construction method proposed in this article can open a new avenue for the preparation of more efficient PEC sensing devices.

Our research group successfully designed the F. carica and Taxus cuspidata Sieb. et Zucc mixed forest, and confirmed that their interspecific relationship was stable and F. carica has a promoting effect on neighboring T. cuspidata growth. However, the promoting mechanism has not been elucidated. In this study, F. carica was used as the donor plant and T. cuspidata was used as the recipient plant. T. cuspidata seedlings were irrigated with F. carica root extracts of different concentrations (10.0, 20.0, 40.0 g·L−1), and the plant height, base diameter, photosynthetic parameters, photosynthetic pigments, MDA contents, and antioxidant enzyme activities were measured. Soil physical and chemical properties, enzyme activities, and microbial diversity were measured. The results showed that the abundance of growth-promoting bacteria increased and the number of pathogenic bacteria decreased in the rhizosphere of T. cuspidata soil. It was speculated that the chemicals secreted by F. carica roots interacted with soil microorganisms of T. cuspidata soil after enrichment, changed soil microbial diversity, and indirectly promoted the growth of T. cuspidata. UPLC-QTOF-MS/MS was used to analyze F. carica root water extract and F. carica root exudates, respectively, and it was found that the main components were similar. Therefore, the promoting effect of F. carica on T. cuspidata is mainly caused by the accumulation of potential chemicals in F. carica root exudates in the soil through interaction with soil microorganisms. Furthermore, the allelopathic-promoting mechanism of F. carica on T. cuspidata was discussed from various aspects, to provide a theoretical basis for the protection, breeding, and sustainable management of T. cuspidata resources.

Significant heterogeneity has been observed among different ecosystem services (ES). Understanding the trade-offs and synergies among ES and delineating ecological functional zones is crucial for formulating regional management policies that improve human well-being and sustainably develop and maintain ecosystems. In this study, we used the Integrated Valuation of Ecosystem Services and Trade-offs (InVEST) and Carnegie–Ames–Stanford Approach (CASA) models to evaluate the spatial distribution patterns of nine ES (food supply, raw material supply, water resource supply, water connotation, climate regulation, soil conservation, water purification, habitat quality, and entertainment tourism) in the Qilian Mountains from 2000 to 2018. We also investigated the trade-offs and synergistic relationships among ES through Spearman correlation analysis, identified ES hotspots through exploratory spatial data analysis, and identified ES bundles (ESB) using K-means clustering. Our results revealed that water purification and habitat quality remained relatively stable, while food supply, raw material supply, water resource supply, water conservation, climate regulation, soil conservation, and entertainment tourism increased by 1038.83 Yuan·ha−1, 448.21 Yuan·ha−1, 55.45 mm, 7.80 mm, 0.60 tc·ha−1, 40.01 t·ha−1 and 4.82, respectively. High-value areas for water resource supply were mainly concentrated in the high-altitude mountainous area, whereas high-value areas for soil conservation were found in the western and eastern parts of the study area. The low-value areas of water purification were primarily located in the east, while the remaining six services were highly distributed in the east and were less common in the west. Correlation analysis showed that water resource supply, water conservation, and soil conservation exhibited a synergistic relationship in the Qilian Mountains. Moreover, food supply, raw material supply, climate regulation, habitat quality, and entertainment tourism showed synergistic relationships. However, there were trade-offs between food supply and water purification as well as water resource supply, and habitat quality showed a tradeoff with water resource supply, water conservation, and soil conservation. We identified four ESB. The food supply bundle consisted mainly of farmland ecosystems, while the windbreak and sand fixation and ecological coordination bundles were dominant in the Qilian Mountains. Notably, the area of the water conservation bundle increased significantly. Our comprehensive findings on ES and ESB can provide a theoretical foundation for the formulation of ecological management policies and the sustainable development of ecosystems in the Qilian Mountains.

Abstract Background Circular RNAs (circRNAs) could participate in cis-dichlorodiammineplatinum (DDP) resistance of human cancers. However, circRNAs role in DDP resistance of oral squamous cell carcinoma (OSCC) progression remains largely undeveloped. Here, we attempted to explore the role of circ-SCMH1 (ID hsa_circ_0011946) in acquired DDP resistance. Methods Expression of circ-SCMH1, microRNA (miR)-338-3p and Lin-28 homolog B (LIN28B) was detected by real-time quantitative PCR and western blotting, and their interactions were confirmed by dual-luciferase reporter assay, RNA immunoprecipitation and RNA pull-down assay. DDP resistance was assessed by MTT assay, colony formation assay, flow cytometry, transwell assays, western blotting, and xenograft experiment. Transmission electron microscopic analysis, nanoparticle tracking analysis and western blotting confirmed the characterizations of extracellular vesicles (EVs). Results Circ-SCMH1 was upregulated in DDP-resistant OSCC tissues and cells (SCC-15/DDP and CAL-27/DDP). Circ-SCMH1 knockdown suppressed the half-maximal inhibitory concentration of DDP, colony formation, and migration/invasion in SCC-15/DDP and CAL-27/DDP cells, but promoted apoptosis rate and apoptotic proteins (Bax and cleaved-caspase-3) expression. However, silencing miR-338-3p abrogated above effects, and overexpressing miR-338-3p mimicked that. Similarly, miR-338-3p overexpression role could be counteracted by restoring LIN28B. Moreover, interfering circ-SCMH1 retarded tumor growth of SCC-15/DDP cells in vivo with DDP treatment or not. Mechanistically, circ-SCMH1 directly sponged miR-338-3p in regulating LIN28B, a target gene for miR-338-3p. Notably, circ-SCMH1 was an EVs cargo, and DDP-resistant OSCC cells-derived EVs could provoke circ-SCMH1 upregulation in parental cells. Conclusion Circ-SCMH1 contributes to chemoresistance of DDP-resistant OSCC cells partially via EVs secretion and circ-SCMH1/miR-338-3p/LIN28B axis.

Abstract Background 3-Phenylpropanol with a pleasant odor is widely used in foods, beverages and cosmetics as a fragrance ingredient. It also acts as the precursor and reactant in pharmaceutical and chemical industries. Currently, petroleum-based manufacturing processes of 3-phenypropanol is environmentally unfriendly and unsustainable. In this study, we aim to engineer Escherichia coli as microbial cell factory for de novo production of 3-phenypropanol via retrobiosynthesis approach. Results Aided by in silico retrobiosynthesis analysis, we designed a novel 3-phenylpropanol biosynthetic pathway extending from l-phenylalanine and comprising the phenylalanine ammonia lyase (PAL), enoate reductase (ER), aryl carboxylic acid reductase (CAR) and phosphopantetheinyl transferase (PPTase). We screened the enzymes from plants and microorganisms and reconstructed the artificial pathway for conversion of 3-phenylpropanol from l-phenylalanine. Then we conducted chromosome engineering to increase the supply of precursor l-phenylalanine and combined the upstream l-phenylalanine pathway and downstream 3-phenylpropanol pathway. Finally, we regulated the metabolic pathway strength and optimized fermentation conditions. As a consequence, metabolically engineered E. coli strain produced 847.97 mg/L of 3-phenypropanol at 24 h using glucose-glycerol mixture as co-carbon source. Conclusions We successfully developed an artificial 3-phenylpropanol pathway based on retrobiosynthesis approach, and highest titer of 3-phenylpropanol was achieved in E. coli via systems metabolic engineering strategies including enzyme sources variety, chromosome engineering, metabolic strength balancing and fermentation optimization. This work provides an engineered strain with industrial potential for production of 3-phenylpropanol, and the strategies applied here could be practical for bioengineers to design and reconstruct the microbial cell factory for high valuable chemicals.

X-ray generation via synchrotron radiation and bremsstrahlung in the interaction of short laser pulses with a solid target is of much current interest owing to its numerous applications. The efficiency of laser to X-ray energy conversion is thus a crucial factor. We found that the energy conversion efficiency of synchrotron radiation and bremsstrahlung is mainly governed by the ratio of the laser pulse width to the preplasma width, which is in turn governed by the laser profile, intensity, and spot size. Synchrotron radiation dominates when the ratio is less than unity, otherwise bremsstrahlung dominates. The type of radiation can thus be controlled by tailoring the laser parameters.

The Pillars of Creation, one of the most recognized objects in the sky, are believed to be associated with the formation of young stars. However, so far, the formation and maintenance mechanism of the pillars are still not fully understood due to the complexity of the nonlinear radiation magnetohydrodynamics (RMHD). Here, assuming laboratory laser-driven conditions, we studied the self-consistent dynamics of pillar structures in magnetic fields by means of two-dimensional and three-dimensional (3D) RMHD simulations, and the results support our proposed experimental scheme. We find that only when the magnetic pressure and ablation pressure are comparable, the magnetic field can significantly alter the plasma hydrodynamics. For medium-magnetized cases ( β _initial ≈ 3.5), the initial magnetic fields undergo compression and amplification. This amplification results in the magnetic pressure inside the pillar becoming large enough to support the sides of the pillar against radial collapse due to pressure from the surrounding hot plasma. This effect is particularly pronounced for the parallel component ( B _y ), which is consistent with observational results. In contrast, a strong perpendicular ( B _x , B _z ) magnetic field ( β _initial < 1) almost retains its initial distribution and significantly suppresses the expansion of blown-off gas plasma, leading to the inability to form pillar-like structures. The 3D simulations suggest that the bending at the head of “Column I” in the Pillars of Creation may be due to nonparallel magnetic fields. After similarity scaling transformation, our results can be applied to explain the formation and maintenance mechanism of the pillars, and can also provide useful information for future experimental designs.

Abstract Background The SARS-CoV-2 RNA was detected positive again after discharged from hospital in some COVID-19 patients, with or without clinical symptoms such as fever or dry cough. Methods 1008 severe COVID-19 patients, with SARS-CoV-2 RNA positive detected with the mixed specimen of nasopharyngeal swab and oropharyngeal swab by real-time fluorescence quantitative PCR (RT-qPCR), were selected to monitor SARS-CoV-2 RNA with the 12 types of specimens by RT-qPCR during hospitalization. All of 20 discharged cases with COVID-19 were selected to detect SARS-CoV-2 RNA in isolation period with 7 types of specimens by RT-qPCR before releasing the isolation period. Results Of the enrolled 1008 severe patients, the nasopharyngeal swab specimens showed the highest positive rate of SARS-CoV-2 RNA (71.06%), followed by alveolar lavage fluid (66.67%), oropharyngeal swab (30.77%), sputum (28.53%), urine (16.30%), blood (12.5%), stool (12.21%), anal swab (11.22%) and corneal secretion (2.99%), and SARS-CoV-2 RNA couldn’t be detected in other types of specimen in this study. Of the 20 discharged cases during the isolation period, the positive rate of SARS-CoV-2 RNA was 30% (6/20): 2 cases were positive in sputum at the eighth and ninth day after discharge, respectively, 1 case was positive in nasopharynx swab at the sixth day after discharge, 1 case was positive in anal swab at the eighth day after discharge, and 1 case was positive in 3 specimens (nasopharynx swab, oropharynx swab and sputum) simultaneously at the fourth day after discharge, and no positive SARS-CoV-2 RNA was detected in other specimens including stool, urine and blood at the discharged patients. Conclusions SARS-CoV-2 RNA should be detected in multiple specimens, such as nasopharynx swab, oropharynx swab, sputum, and if necessary, stool and anal swab specimens should be performed simultaneously at discharge when the patients were considered for clinical cure and before releasing the isolation period.

Abstract Background Combining ultrasound imaging with photoacoustic imaging provides tissue imaging with high contrast and resolution, thereby enabling rapid, direct measurements and the tracking of tumour growth and metastasis. Moreover, ultrasound-targeted nanobubble destruction (UTND) provides an effective way to deliver drugs, effectively increasing the content of the drug in the tumour area and reducing potential side effects, thereby successfully contributing to the treatment of tumours. Results In this study, we prepared multifunctional nanobubbles (NBs) carrying indocyanine green (ICG) and paclitaxel (PTX) (ICG-PTX NBs) and studied their applications in ultrasound imaging of prostate cancer as well as their therapeutic effects on prostate cancer when combined with UTND. ICG-PTX NBs were prepared by the mechanical oscillation method. The particle size and zeta potential of the ICG-PTX NBs were 469.5 ± 32.87 nm and − 21.70 ± 1.22 mV, respectively. The encapsulation efficiency and drug loading efficiency of ICG were 68% and 2.52%, respectively. In vitro imaging experiments showed that ICG-PTX NBs were highly amenable to multimodal imaging, including ultrasound, photoacoustic and fluorescence imaging, and the imaging effect was positively correlated with their concentration. The imaging effects of tumour xenografts also indicated that ICG-PTX NBs were of good use for multimodal imaging. Kangfuxiaoyan suppository (KFXYS) is a commonly used traditional Chinese medicine (TCM) preparation for the treatment of chronic pelvic inflammatory disease (CPID) clinically, and its safety and effectiveness have been well verified. However, the potential mechanism remains unclear. The integrated strategy of metabolomics and network pharmacology was employed in the study to reveal the potential mechanism of KFXYS in the treatment of CPID. Our research consists of five steps. First, the effect of KFXYS in reversing uterine inflammation indexes was verified. Second, based on the comprehensive characterization of 123 chemical ingredients of KFXYS, the ingredients of KFXYS absorbed into blood were identified by UPLC-Q-TOF/MS, then ADME research was carried out on the main ingredients. Third, the differential metabolites with significant correlation to inflammatory indexes were discovered by metabolomics and correlation analysis. Fourth, the potential targets and pathways of KFXYS in treating CPID were predicted by network pharmacology based on the ingredients which had good ADME behavior. Fifth, the proteins in common pathways of metabolomics and network pharmacology were used to screen the key targets from the potential targets of network pharmacology, and the potential mechanism of KFXYS in treating CPID was clarified. As a result, KFXYS significantly reversed the uterine inflammation indexes, including IL-1 and IL-6. The ingredients absorbed into blood including matrine, sophocarpine, aloin, esculetin-O-glucuronide, 7,4′-dihydroxyisoflavone-O-glucuronide, and 4′-methoxyisoflavone-7-O-glucuronide had good ADME behavior in vivo. Among the differential metabolites, Leukotriene A4, 5-Hydroxyindoleacetic acid, Ornithine, Arginine, and PC (20:1 (11Z)/20:4 (8Z,11Z,14Z,17Z)) were significant correlation to inflammation indexes. The integration analysis of metabolomics and network pharmacology shows that KFXYS may regulate the key targets including ARG1, NOS2, NOS3, etc. We speculate that ingredients of KFXYS, such as matrine, sophocarpine, aloin etc. act on the key proteins including ARG1, NOS2, and NOS3, to exert anti-inflammatory effect.

Abstract The modification of chimeric antigen receptor (CAR) endowing T cells with tumor‐specific cytotoxicity induces antitumor immunity. However, the structural characteristics of solid tumors, the loss of specific antigens, and the strong immunosuppressive environment are challenges to treat solid tumors with CAR‐T therapy. The purpose of our study was to find and verify the potentials of CAR‐T therapies for patients with head and neck squamous cell carcinoma (HNSCC). First, we selected MUC1 as our research target and verified its differential expression in cancer tissues and adjacent non‐neoplastic tissues (ANNT). Next, we constructed a second‐generation CAR and validated the cytotoxic function in vitro. In our study, we found that exogenous addition human IL22 recombinant protein could increase the MUC1 expression and enhance the function of T cells. In addition, we constructed a fourth‐generation CAR that secretes IL22. Finally, we verified the antitumor function of two different CAR‐T cells in vitro and in vivo, respectively. CAR‐MUC1‐IL22 T cells were found to have a stronger and more effective cytotoxic function against MUC1 + HNSCC cells. Taken together, these results demonstrate the potential effectiveness of CAR‐T in the treatment of patients with HNSCC and provide evidence‐based of MUC1 + CAR‐T therapy.

The results of a commissioning experiment on the SILEX-Ⅱ laser facility (formerly known as CAEP-PW) are reported. SILEX-Ⅱ is a complete optical parametric chirped-pulse amplification laser facility. The peak power reached about 1 PW in a 30 fs pulse duration during the experiment. The laser contrast was better than 1010 at 20 ps ahead of the main pulse. In the basic laser foil target interaction, a set of experimental data were collected, including spatially resolved x-ray emission, the image of the coherent transition radiation, the harmonic spectra in the direction of reflection, the energy spectra and beam profile of accelerated protons, hot-electron spectra, and transmitted laser energy fraction and spatial distribution. The experimental results show that the laser intensity reached 5 × 1020 W/cm2 within a 5.8 µm focus (FWHM). Significant laser transmission did not occur when the thickness of the CH foil was equal to or greater than 50 nm. The maximum energy of the accelerated protons in the target normal direction was roughly unchanged when the target thickness varied between 50 nm and 15 µm. The maximum proton energy via the target normal sheath field acceleration mechanism was about 21 MeV. We expect the on-target laser intensity to reach 1022 W/cm2 in the near future, after optimization of the laser focus and upgrade of the laser power to 3 PW.

A quality assessment method based on quantitative analysis of multi-components by single marker (QAMS) and fingerprint was constructed from 15 batches of dandelion (Taraxacum mongolicum), using multivariate chemometric methods (MCM). MCM were established by hierarchical cluster analysis (HCA) and factor analysis (FA). HCA was especially performed using the R language and SPSS 22.0 software. The relative correction factors of chlorogenic acid, caffeic acid, p-coumaric acid, luteolin and apigenin were calculated with cichoric acid as a reference, and their contents were determined. The differences between external standard method (ESM) and QAMS were compared. There was no significant difference (t-test, p > 0.05) in quantitative determination, proving the consistency of the two methods (QAMS and ESM). Dandelion material from Yuncheng, Shandong was used as a reference chromatogram. The fingerprints in 15 batches of dandelion were established by HPLC analysis. The similarity of the fingerprints in different batches of dandelion material was greater than or equal to 0.82. A total of 10 common peaks were identified. This strategy is simple, rapid and efficient in multiple component detection of dandelion. It is beneficial in simplifying dandelion's quality control processes and providing references to enhance quality control for other herbal medicines.

Background: Hemophilia A (HA) is an X-linked recessive blood coagulation disorder caused by a variety of abnormalities in F8 gene, resulting in the absence of impaired molecule production of factor VIII (FVIII) in the plasma. The genetic testing of the F8 gene encoding FVIII is used for confirmation of HA diagnosis, which significantly reduced serious complications of this disease and, ultimately, increased life expectancy. Methods: Sanger sequencing was performed in F8 gene exons of the suspected patients with blood coagulation-related indicators. Results: A novel F8 indel variant c.6343delC, p.Leu2115SerfsTer28 in exon 22 of the F8 gene was identified in the suspected families. The infant with this novel variant appeared the symptom of minor bleeding and oral cavity bleeding, and decreased activity of FVIII, which is consistent with that of F8 deleterious variants. The 3’D protein structural analysis of the novel variant shows a change in FVIII protein stability, which may be responsible for the pathogenesis of HA. Conclusions: A novel deleterious variant was identified in our case, which expands the F8 variants spectrum. Our result is helpful for HA diagnosis and benefits carrier detection and prenatal diagnosis. Our study also reveals that mutation screening of the F8 gene should be necessary for HA suspected patients.

The electrodes of two-dimensional (2D) titanium dioxide (TiO2) nanosheet arrays were successfully fabricated for microRNA-155 detection. The (001) highly active crystal face was exposed to catalyze signaling molecules ascorbic acid (AA). Zero-dimensional (0D) titanium carbide quantum dots (Ti3C2Tx QDs) were modified to the electrode as co-catalysts and reduced the recombination rate of the charge carriers. Spectroscopic methods were used to determine the band structure of TiO2 and Ti3C2Tx QDs, showing that a type Ⅱ heterojunction was built between TiO2 and Ti3C2Tx QDs. Benefiting the advantages of materials, the sensing platform achieved excellent detection performance with a wide liner range, from 0.1 pM to 10 nM, and a low limit of detection of 25 fM (S/N = 3).

Macrofungi, which are also known as mushrooms, can produce various bioactive constituents and have become promising resources as lead drugs and foods rich in nutritional value. However, the production of these bioactive constituents under standard laboratory conditions is inefficiency due to the silent expression of their relevant genes. Coculture, as an important activation strategy that simulates the natural living conditions of macrofungi, can activate silent genes or clusters through interspecific interactions. Coculturing not only can trigger the biosynthesis of diverse secondary metabolites and enzymes of macrofungi, but is also useful for uncovering the mechanisms of fungal interspecific interactions and novel gene functions. In this paper, coculturing among macrofungi or between macrofungi and other microorganisms, the triggering and upregulation of secondary metabolites and enzymes, the potential medicinal applications, and the fungal–fungal interaction mechanisms are reviewed. Finally, future challenges and perspectives in further advancing coculture systems are discussed.

By utilizing the FLUENT software, finite element simulation analysis was performed on the two-phase separation of powder injection molding, and the simulation results were verified through experiment. As indicated in the simulation results, the optimal process parameters for the injection molding of 316L stainless steel sample were to inject at 70 MPa and 140 °C, at an injection speed of 3.49 cm3/s. Under these conditions, the maximum average solid volume fraction in observation area of the sample was 57.68 %. Additionally, it was discovered that while preparing 316L stainless steel was prepared by powder injection molding, the smaller the average porosity of the sample, the greater the Vickers hardness. Layer-by-layer (LBL) technique is a well-established method for the formation of nanocapsules, which could encapsulate nanoparticles by polyelectrolyte and has received great interest in some areas of research. In this study, colloidal gold nanoparticles (AuNPs) were used as a vehicle for loading horseradish peroxidase (HRP), and AuNPs-HRP was encapsulated by using LBL technique to form a signal amplification nanocapsule probe. Based on the probe, an immunoassay was developed for the detection of imidacloprid in agricultural food samples (rice, pear and cabbage). The IC50 value of the assay was 0.11 ng mL−1 which was 8-9954 times lower than those of traditional ELISAs and well below the maximum residue limits (MRLs) permitted by the European Union (EU). The recoveries of imidacloprid from three different samples were 80.34–120.84% that had a good correlation (R2 = 0.9739) with those obtained by the commercial kit. The developed assay could provide a sensitive method for detection of imidacloprid.

The velvetleaf (Abutilon theophrasti Medik.) is a strong and competitive weed in fields that inhibits the growth of crops. Reports have suggested that allelopathy is one of the reasons for this inhibition; however, the mechanism of this allelopathy remains unclear. In this study, velvetleaf powder/extracts were shown to inhibit seed germination, growth and yield in maize, wheat and soybean through petri dish, pot and field control experiments. We observed a concentration-dependent inhibition of the seed germination rate for all three crops. The root tip structure changed significantly and the embryo even died when irrigated with a high concentration of the extract (10 mg·mL−1). After adding velvetleaf powder, the malondialdehyde (MDA) content in crop seedlings was dose-dependent, and the superoxide dismutase (SOD) activity of maize, wheat and soybean showed the maximum values under treatment with 1.25, 5 and 5 mg·cm−3, respectively. The activity of peroxidase (POD) showed the highest value under the 5 mg·cm−3 treatment in maize and wheat seedlings and under 10 mg·cm−3 treatment in soybean seedlings. However, sugar, protein content and root activity in all three crops was the lowest under the 10 mg·cm−3 treatment. Therefore, velvetleaf may decrease the productivity of three crops by changing the antioxidant enzyme activities, root system activities and root tip structures.

PurposeThe aim is to investigate the impacts of using multiplex immunochemistry (mIHC) staining to analyses the co-expression of programmed death ligand-1 (PD-L1) and tumor infiltrating lymphocytes (TILs) [CD8+ T cells and Forkhead Box Protein 3 (FOXP3)+ regulatory T cells (Tregs)] in different oral diseases, and oral squamous cell carcinoma (OSCC).MethodsFormalin fixed paraffin-embedded tissue sections from different oral diseases were stained with PD-L1 and TILs (CD8+ T cells and FOXP3+ Tregs) by mIHC staining simultaneously. The whole slide was scanned digitally to observe the cell phenotypes stained in the microenvironment. The contents of each slice were read using a computer-aided method to analyze and the cell densities were calculated using statistical software.ResultsWe were able to characterize the tumor microenvironment (TME) of different oral diseases including oral leukoplakia (OLK), inflammatory gingiva (IG), oral lichen planus (OLP), and squamous cell carcinoma (SCC), with accurate visualization of various immune cells harboring complex immune phenotypes by mIHC staining. The results showed that PD-L1 was up-regulated in SCC tissues at different pathological stages, while CD8 and FOXP3 had no significant changes. The ratio of PD-L1/CD8 was also significantly up-regulated in SCC tissues compared with that of other oral diseases. In advanced stages of OSCC, the FOXP3/CD8 ratio increased, and immunosuppressive TME was more pronounced. In addition, we also found different immune phenotypes: the inflamed phenotype, immune-excluded phenotypes, and immune-desert phenotypes. By locating tumor epithelial cells, we found that PD-L1 expression is in both tumor cells and stromal cells.ConclusionsmIHC is useful for the visualization and evaluation of tumor microenvironment in immuno-oncology research. It allows single-cell imaging in situ and could effectively and quickly determine the immune phenotype of different oral diseases.

Divergence gating, a novel method to generate far-field isolated attosecond pulses (IAPs) through controlling divergences of different pulses, is proposed and realized by relativistic chirped laser–plasma interactions. Utilizing various wavefronts for different cycles of incident chirped lasers, reflected harmonics with minimum divergences are obtained only at the peak cycle when plasma targets are adjusted to proper distances from foci of lasers. Therefore, the corresponding attosecond pulse is isolated in far field due to much slower decay during propagation than others. Confirmed by three-dimensional numerical simulations, millijoule-level sub-50 as IAPs with intensity approaching 10 ^16 W cm ^−2 (10 ^17 –10 ^18 W sr ^−1 ) are obtained by our scheme, where low-order harmonics can be preserved.

The vibration signal of gearboxes contains abundant fault information, which can be used for condition monitoring. However, vibration signal is ineffective for some non-structural failures. In order to resolve this dilemma, infrared thermal images are introduced to combine with vibration signals via fusion domain-adaptation convolutional neural network (FDACNN), which can diagnose both structural and non-structural failures under various working conditions. First, the measured raw signals are converted into frequency and squared envelope spectrum to characterize the health states of the gearbox. Second, the sequences of the frequency and squared envelope spectrum are arranged into two-dimensional format, which are combined with infrared thermal images to form fusion data. Finally, the adversarial network is introduced to realize the state recognition of structural and non-structural faults in the unlabeled target domain. An experiment of gearbox test rigs was used for effectiveness validation by measuring both vibration and infrared thermal images. The results suggest that the proposed FDACNN method performs best in cross-domain fault diagnosis of gearboxes via multi-source heterogeneous data compared with the other four methods.

Medicinal-agroforestry systems are one of the multi-functional medicinal plant production systems, gaining attention as a sustainable alternative to traditional monoculture systems. In this study, three planting patterns were established which included: (1) monoculture F. carica (MF); (2) monoculture T. cuspidata (MT); and (3) interplanting F. carica with T. cuspidata (IFT). The differences of growth biomass, photosynthesis, soil nutrients, soil enzyme activities, soil microorganisms, and main secondary metabolites of F. carica and T. cuspidata under the above three models were investigated. Compared with the MF and MT patterns, IFT pattern for 5 months significantly increased the plant growth biomass, photosynthesis, soil organic carbon, total nitrogen, and secondary metabolites content. The activities of acid phosphatase, sucrase, protease, polyphenol oxidase, urease, dehydrogenase, and catalase in soil of IFT were significantly higher than MF and MT patterns. Results showed that IFT pattern is preferred compared to the MF and MT patterns. Our result will help to provide a feasible theoretical basis for the large-scale establishment of F. carica and T. cuspidata mixed forests and obtain high-quality medicine sources for extracting important active ingredients, psoralen and paclitaxel, which are crucial to the long-term sustainable development and production of medicinal plants.

Fig tree cultivation land resources are not fully utilized and introducing them into sustainable medicinal agroforestry systems can effectively achieve resource protection and reuse. Laboratory and pot experiments were applied to study the allelopathic effects of fig tree (Ficus carica L.) leaf aqueous extract at five mass concentrations of 8.3, 10.0, 12.5, 16.7, and 25.0 g/L on the morphological and physiological indexes of mint (Mentha haplocalyx Briq.), dandelion (Taraxacum mongolicum Hand.-Mazz.), and woad (Isatis indigotica Fort.). The results showed that mint had the best seed germination rate. The leaf aqueous extract at lower concentrations had a strong promoting effect on the biomass and photosynthetic parameters of mint, dandelion, and woad. With the increase in leaf aqueous extract concentration, the superoxide dismutase, peroxidase, and catalase activity of mint, dandelion, and woad increased initially and then decreased, but the malondialdehyde content increased. The synthetic allelopathic indexes of the three medicinal plants were in the following order: mint > woad > dandelion. Both the low and medium concentration extracts (8.3 g/L–12.5 g/L) showed an obvious promoting effect, while high concentrations exhibited distinct inhibiting effects. In conclusion, mint is the most suitable medicinal plant to be interplanted with fig trees for introduction into medicinal agroforestry systems.

Equipment manufacturing industry is one of the major industries of the Chinese economy. Previous researches have revealed that the industry has dilemmas of unreasonable industrial structure and high pollution. Using the data of 30 provinces in 2006-2015 in China, this study calculated a comprehensive pollution indicator when estimating the possible pollution reduction brought by the optimization of industrial structure and then evaluated the reasonable level of capital allocation of provinces and industries by using the methods of nonlinear programming and stochastic frontier method. Under the target of collaborative emission reduction, the results show that the optimized output of China's equipment manufacturing industry could be increased by 5.42%, the energy intensity could be reduced by about 10.4%, and the comprehensive emission intensity could be reduced by about 7.47%. Due to the industry heterogeneity and regional heterogeneity, industrial capacity should be transferred between industries and regions. Since the capital investment in the equipment manufacturing industry is significantly mismatched between industries and regions, the capital allocation of provincial industries in China needs to be adjusted properly. This study provides theoretically and practically reference for collaborative pollution reduction, industry restructure, spatial layout and capital investment, which contributes to achieving the stereoscopic optimization of equipment manufacturing industry.

Abstract Background High-altitude headache (HAH) is the most common sickness occurred in healthy people after rapid ascending to high altitude, and its risk factors were still not well understood. To investigate physiological, hematological and biochemical risk factors associated with high-altitude headache (HAH) after acute exposure to 3700 m, we conducted a two-stage, perspective observational study. In 72 h, total 318 young Han Chinese males ascended from sea level (altitude of 50 m) to altitude of 3700 m by train. Demographic data, physiological, hematological and biochemical parameters of all participants were collected within one week prior to the departure, and within 24 h after arrival. Fig (Ficus carica L.) leaves are produced each year and often disposed, resulting in a waste of resources. Fig waste leaves are rich in flavonoids, which have strong antioxidant activity; however, the variety and chemical structure of antioxidants in fig leaves have not been reported in detail. To take full advantage of fig waste leaves, antioxidant capacity of different extracts (petroleum ether, ethyl acetate, and water) was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic) acid (ABTS), and ferric-ion-reducing antioxidant power (FRAP) methods. The results showed that flavonoids in ethyl acetate extraction had the highest content (83.92 ± 0.01 mg/g), maximum DPPH scavenging activity (IC50 0.54 mg/mL), highest ABTS scavenging rate (80.28%), and FRAP (3.46 mmol/g). Furthermore, an HPLC-QTOF-MS/MS-DPPH method was developed to identify 11 flavonoids in fig waste leaves. This rapid and efficient method can not only be used for screening the antioxidant components in fig waste leaves, but also can be combined with mass spectrometry to identify the compounds with antioxidant capacity. There are three flavonoids with significant antioxidant capacity, which are 3-O-(rhamnopyranosyl-glucopyranosyl)-7-O-(glucopyranosyl)-quercetin, isoschaftoside, and rutin. The results confirmed that fig waste leaves contain a variety of antioxidant components, which contributed to increase the value of fig waste leaves as antioxidants.

The radiation reaction effects on electron dynamics in counter-propagating circularly polarized laser beams are investigated through the linearization theorem and the results are in great agreement with numeric solutions. For the first time, the properties of fixed points in electron phase-space were analyzed with linear stability theory, showing that center nodes will become attractors if the classical radiation reaction is considered. Electron dynamics are significantly affected by the properties of the fixed points and the electron phase-space densities are found to be increasing exponentially near the attractors. The density growth rates are derived theoretically and further verified by particle-in-cell simulations, which can be detected in experiments to explore the effects of radiation reaction qualitatively. The attractor can also facilitate realizing a series of nanometer-scaled flying electron slices via adjusting the colliding laser frequencies.

We demonstrate theoretically and numerically that intense isolated circularly polarized (CP) attosecond pulses can be generated from ultrathin foil targets irradiated by two relativistic lasers from opposite sides, where their polarizations are orthogonal to each other. With a proper matching condition, the compressed oscillating plasma mirrors on both sides of the foil are pushed inside by laser radiation pressures, eventually merging together to form a dense electron nanobunch under the effect of orthogonal laser fields. This nanobunch reaches both high density and high energy in only half a laser cycle and smears out in others, resulting in coherent synchrotron emission of a single attosecond pulse with circular polarization. Two-dimensional particle-in-cell simulations show that an intense isolated CP attosecond XUV pulse with an intensity of 1.2 × 10 ^19 W cm ^−2 and a duration of ∼75 as can be obtained by two lasers with the same intensity of 2.1 × 10 ^20 W cm ^−2 .

Three-dimensional particle-in-cell simulations show that relativistic Gaussian laser light can be transformed into relativistic vortex laser light with axial orbital angular momentum (OAM) as it propagates through a homogeneous spiral-profiled low-density plasma slab. In the process, the plane equiphase surfaces of the Gaussian laser undergo azimuthal modulation and become a continuous helical surface. The intensity profile of the laser changes from maximum-on-axis to donut-shaped. Because of the azimuthally varying slab thickness, the laser ponderomotive and the charge-separation forces exert a torque on the plasma when it passes through it, resulting in the creation of oppositely directed OAM in the plasma ions and the laser light, with the electrons remaining nearly OAM free. The proposed scheme can be used to generate both single- and multi-mode relativistic vortex lasers propagating along the direction of the input laser, which is especially convenient for many applications.

The co-culture of Trametes versicolor and Ganoderma applanatum is a model of intense basidiomycete interaction, which induces many newly synthesized or highly produced features. Currently, one of the major challenges is an identification of the origin of induced features during the co-culture. Herein, we report a 13C-dynamic labeling analysis used to determine an association of induced features and corresponding fungus even if the identities of metabolites were not available or almost nothing was known of biochemical aspects. After the co-culture of T. versicolor and G. applanatum for 10 days, the mycelium pellets of T. versicolor and G. applanatum were sterilely harvested and then mono-cultured in the liquid medium containing half fresh medium with 13C-labeled glucose as carbon source and half co-cultured supernatants collected on day 10. 13C-labeled metabolome analyzed by LC-MS revealed that 31 induced features including 3-phenyllactic acid and orsellinic acid were isotopically labeled in the mono-culture after the co-culture stimulation. Twenty features were derived from T. versicolor, 6 from G. applanatum, and 5 features were synthesized by both T. versicolor and G. applanatum. 13C-labeling further suggested that 12 features such as previously identified novel xyloside [N-(4-methoxyphenyl)formamide 2-O-beta-D-xyloside] were likely induced through the direct physical interaction of mycelia. Use of molecular network analysis combined with 13C-labeling provided an insight into the link between the generation of structural analogs and producing fungus. Compound 1 with m/z 309.0757, increased 15.4-fold in the co-culture and observed 13C incorporation in the mono-culture of both T. versicolor and G. applanatum, was purified and identified as a phenyl polyketide, 2,5,6-trihydroxy-4, 6-diphenylcyclohex-4-ene-1,3-dione. The biological activity study indicated that this compound has a potential to inhibit cell viability of leukemic cell line U937. The current work sets an important basis for further investigations including novel metabolites discovery and biosynthetic capacity improvement.

Cytotoxin-associated-gene A (CagA) of Helicobacter pylori (H. pylori) is a virulence factor that plays critical roles in H. pylori-induced gastric inflammation. In the present study, gastric biopsies were used for genotyping cagA and vacA genes, determining the autophagic activity, and the severity of gastric inflammation response. It was revealed that autophagy in gastric mucosal tissues infected with cagA+H. pylori strains was lower than the levels produced by cagA−H. pylori strains, accompanied with accumulation of SQSTM1 and decreased LAMP1 expression. In vitro, deletion mutant of cagA gene resulted in increased autophagic activity, and decreased expression of SQSTM1 and cytokines, whereas over-expression of CagA down-regulated the starvation-induced autophagy, and induced more production of the cytokines. Moreover, the production of the cytokines was increased by inhibition of autophagy, but decreased by enhancement of autophagy. Deletion of CagA decreased the ability to activate Akt kinase at Ser-473 site and increased autophagy. c-Met siRNA significantly affected CagA-mediated autophagy, and decreased the level of p-Akt, p-mTOR, and p-S6. Both c-Met siRNA and MK-2206 could reverse inflammatory response. H. pylori CagA protein negatively regulates autophagy and promotes the inflammation in H. pylori infection, which is regulated by c-Met-PI3K/Akt-mTOR signaling pathway activation.
