Abstract Background The potentially fatal attacks experienced by porphyria carriers are triggered by various porphyrinogenic drugs. However, determining the safety of particular drugs is challenging. Methods We retrospectively used the U.S. Food and Drug Administration’s Adverse Event Reporting System (FAERS) to identify drugs associated with porphyria as an adverse event (AE) extracted from data from January 2004 to March 2022. The associated search terms included “Porphyria,” “Porphyria screen,” “Porphyria non-acute,” “Porphyria acute,” “Acquired porphyria,” and “Pseudoporphyria.” Signal mining analysis was performed to identify the association between drugs and AEs by four algorithms, namely the reporting odds ratio, proportional reporting ratio, Bayesian confidence propagation neural network, and multi-item gamma Poisson shrinker. Results FAERS reported 1470 cases of porphyria-related AEs, and 406 drugs were screened after combining trade and generic names. All four algorithms identified 52 drugs with signals. The characteristics of all the reports and signaling drugs were analyzed. Conclusions This is the first report of drug-associated porphyria that provides critical information on drug porphyrogenicity, facilitating rational and evidence-based drug prescription and improving the accuracy of porphyrogenicity prediction based on model algorithms. Moreover, this study serves a reference for clinicians to ensure that porphyrinogenic drugs are not prescribed to carriers of porphyria genetic mutations.

The substitutional structure of cyclotriphosphazene derivatives significantly influences their flame-retardant effectiveness. A cyclotriphosphazene derivative with triazole group, referred to as hexa(1,2,4-triazol-3-ylamine) cyclotriphosphazene (HATA), was utilized to improve the flame retardancy of epoxy resin (EP). Differential scanning calorimetry, thermogravimetric analysis, and dynamic mechanical analysis were employed to characterize the thermal properties of EP/HATA thermosets. HATA facilitated the curing of EP due to its triazole and secondary amine structure. EP/HATA thermosets exhibited improved char-forming ability and storage modulus, attributed to the rigid cyclophosphonitrile structure of HATA. As a result of incorporating 5% HATA with 0.73 wt% phosphorus, EP passed UL-94 V-0 level. Subsequent analysis using a cone calorimeter revealed obvious reductions in the peak heat release rate, fire growth rate, and total smoke production of EP with the addition of HATA. Simultaneously, there was a significant enhancement in the char yield of EP during combustion, indicating notable improvements in fire safety. Additional investigations, including X-ray photoelectron spectroscopy, scanning electron microscopy, TG-FTIR, and pyrolysis gas chromatography/mass spectrometry, were employed to analyze the char residue and gaseous volatiles. HATA promoted the formation of a dense, continuous, and intumescent char layer containing cyclophosphonitrile structure in EP. Moreover, the decomposition of HATA released a notable quantity of nitrogen-containing volatiles, effectively mitigating flammable gases originating from the EP matrix in gaseous phase. A biphasic flame-retardant mode of action was proposed, underscoring cooperative flame-retardant effects arising from the interaction between triazole substituents and cyclophosphonitrile structure in HATA molecular for EP.

Abstract Purpose The purpose of this study is to investigate whether gene mutations can lead to the growth of malignant pulmonary nodules. Methods Retrospective analysis was conducted on patients with pulmonary nodules at Hebei Provincial People’s Hospital, collecting basic clinical information such as gender, age, BMI, and hematological indicators. According to the inclusion and exclusion criteria, 85 patients with malignant pulmonary nodules were selected for screening, and gene mutation testing was performed on all patient tissues to explore the relationship between gene mutations and the growth of malignant pulmonary nodules. Nitrogen dioxide (NO2) is a prevalent air pollutant in urban areas, originating from outdoor sources, household gas consumption, and secondhand smoke. The limited evaluation of the disease burden attributable to NO2, encompassing different health effects and contributions from various sources, impedes our understanding from a public health perspective. Based on modeled NO2 exposure concentrations, their exposure–response relationships with lung cancer, chronic obstructive pulmonary disease, and diabetes mellitus, and baseline disability-adjusted life years (DALYs), we estimated that 1,675 (655–2,624) thousand DALYs were attributable to NO2 in urban China in 2019 [138 (54–216) billion Chinese yuan (CNY) economic losses]. The transition from gas to electricity for household cooking was estimated to reduce the attributable economic losses by 35%. This reduction falls within the range of reductions achieved when outdoor air meets the World Health Organization interim target 3 and air quality guidelines for annual NO2, highlighting the significance of raising awareness of gas as a polluting household energy for cooking. These findings align with global sustainable development initiatives, providing a sustainable solution to promote public health while potentially mitigating climate change.

Abstract RNF214 is an understudied ubiquitin ligase with little knowledge of its biological functions or protein substrates. Here we show that the TEAD transcription factors in the Hippo pathway are substrates of RNF214. RNF214 induces non-proteolytic ubiquitylation at a conserved lysine residue of TEADs, enhances interactions between TEADs and YAP, and promotes transactivation of the downstream genes of the Hippo signaling. Moreover, YAP and TAZ could bind polyubiquitin chains, implying the underlying mechanisms by which RNF214 regulates the Hippo pathway. Furthermore, RNF214 is overexpressed in hepatocellular carcinoma (HCC) and inversely correlates with differentiation status and patient survival. Consistently, RNF214 promotes tumor cell proliferation, migration, and invasion, and HCC tumorigenesis in mice. Collectively, our data reveal RNF214 as a critical component in the Hippo pathway by forming a signaling axis of RNF214-TEAD-YAP and suggest that RNF214 is an oncogene of HCC and could be a potential drug target of HCC therapy.

Abstract Simultaneously modulating the inflammatory microenvironment and promoting local bone regeneration is one of the main challenges in treating bone defects. In recent years, osteoimmunology has revealed that the immune system plays an essential regulatory role in bone regeneration and that macrophages are critical components. In this work, a mussel-inspired immunomodulatory and osteoinductive dual-functional hydroxyapatite nano platform (Gold/hydroxyapatite nanocomposites functionalized with polydopamine - PDA@Au-HA) is developed to accelerate bone tissues regeneration by regulating the immune microenvironment. PDA coating endows nanomaterials with the ability to scavenge reactive oxygen species (ROS) and anti-inflammatory properties, and it also exhibits an immunomodulatory ability to inhibit M1 macrophage polarization and activate M2 macrophage secretion of osteogenesis-related cytokines. Most importantly, this nano platform promotes the polarization of M2 macrophages and regulates the crosstalk between macrophages and pre-osteoblast cells to achieve bone regeneration. Au-HA can synergistically promote vascularized bone regeneration through sustained release of Ca and P particles and gold nanoparticles (NPs). This nano platform has a synergistic effect of good compatibility, scavenging of ROS, and anti-inflammatory and immunomodulatory capability to accelerate the bone repair process. Thus, our research offers a possible therapeutic approach by exploring PDA@Au-HA nanocomposites as a bifunctional platform for tissue regeneration.

Abstract Background Diabetic sarcopenia is a disease‐related skeletal muscle disorder that causes progressive symptoms. The complete understanding of its pathogenesis is yet to be unravelled, which makes it difficult to develop effective therapeutic strategies. This study investigates how MFG‐E8 affects mitophagy and the protective role of D‐pinitol (DP) in diabetic sarcopenia. Methods In vivo, streptozotocin‐induced diabetic SAM‐R1 (STZ‐R1) and SAM‐P8 (STZ‐P8) mice (16‐week‐old) were used, and STZ‐P8 mice were administrated of DP (150 mg/kg per day) for 6 weeks. Gastrocnemius muscles were harvested for histological analysis including transmission electron microscopy. Proteins were evaluated via immunohistochemistry (IHC), immunofluorescence (IF), and western blotting (WB) assay. In vitro, advanced glycation end products (AGEs) induced diabetic and D‐galactose (DG) induced senescent C2C12 models were established and received DP, MFG‐E8 plasmid (Mover)/siRNA (MsiRNA), or 3‐MA/Torin‐1 intervention. Proteins were evaluated by IF and WB assay. Immunoprecipitation (IP) and co‐immunoprecipitation (CO‐IP) were used for hunting the interacted proteins of MFG‐E8. Results In vivo, sarcopenia, mitophagy deficiency, and up‐regulated MFG‐E8 were confirmed in the STZ‐P8 group. Home appliance manufacturers are increasingly focusing on building smarter home systems by incorporating user feedback to enhance products and services. To support this, we designed a federated learning (FL) system that includes a reputation mechanism to help manufacturers leverage customer data to train machine learning models. First, it downloads the initial model provided by the manufacturer and trains it with local data. Then, it asks customers to sign their models and upload them to the blockchain. To protect customer privacy, we implemented differential privacy and introduced a new normalization technique. In addition, we also attract more customers to participate in crowdsourced FL tasks by rewarding their contributions, thereby ensuring that the datasets for model training are robust and diverse. This system not only promotes collaboration between customers and manufacturers, but also facilitates the development of more responsive and smarter home appliance systems through advanced FL and blockchain technologies.

Osteoporosis (OP)-associated fractures can result in severe morbidity and disability, reduced quality of life, and death. Previous studies have suggested that small noncoding RNAs, for example, small regulatory microRNAs (miRNAs), play a key role in OP by inhibiting target gene expression. Cuproptosis, a recently proposed copper-induced cell death pathway, is linked with OP. Here, we describe the contribution of exosomal miRNAs and cuproptosis to OP. First, we highlight the characteristics of exosomes and roles of exosome-related miRNAs. Next, we discuss the relationship between cuproptosis and OP. Subsequently, we analyze the crosstalk of exosomal miRNAs with cuproptosis in the development of OP. This review aims to investigate a new clinical treatment method for OP.

To reduce agricultural pollutant emission (APE) and postharvest loss of grain (PHLG), the Chinese government enacted a series of subsidy policies; however, the profit-oriented supply chain members are seriously lacking or reducing APE and PHLG efforts. To address this issue, we considered as the research objective a grain supply chain consisting of a producer, a retailer and the government. We proposed the concept and functional expressions of supply chain members' reduction efforts for APE and PHLG. We then proposed two main variables: the environmental innovation subsidy coefficient and the quantity attenuation factor of grain. According to the actual situation, four investment subsidy models were proposed. The results showed the following: i) supply chain members' equilibrium prices and incomes were negatively correlated with the degree of the producer's APE effort regardless of whether the supply chain members were investing in PHLG technology; ii) when the government subsidises APE and PHLG technology for other supply chain members, the government should stop subsidising the retailer's inputs in reduction loss technology to ensure that the government's own interests are not damaged; iii) the government's income was restricted by the degree of its subsidising of other supply chain members. This study provides a theoretical support for the government to formulate appropriate policies to reduce APE and PHLG, which is important for maintaining national food security.

Objectives Scoliosis associated with spondylolisthesis is a common phenomenon. Recent research has reported that scoliosis can spontaneously disappear after lumbar spinal fusion surgery. Researchers have advocated that, for scoliosis associated with vertebral slippage, surgery for the latter may be the only necessary intervention, while unnecessary surgery for scoliosis should be avoided. So we propose that minimally invasive techniques can achieve treatment effects similar to those of open surgery. Therefore, in this study, we aimed to investigate the clinical efficacy of unilateral dual‐channel endoscopic lumbar interbody fusion (ULIF) for treating lumbar spondylolisthesis with spinal scoliosis. Methods This study retrospectively analyzed patients with lumbar spondylolisthesis and spinal scoliosis who underwent ULIF between September 2021 and September 2023. Measurements of the Cobb angle, lumbar lordosis (LL) angle, sacral slope (SS), slip percentage (SP), slip angle (SA), L1 plumb line‐S1 distance (LASD), and average intervertebral height (AIH) were taken preoperatively, immediately following surgery, 3 months after surgery, and at the final follow‐up. The visual analogue scale (VAS), Oswestry disability index (ODI), and Japanese Orthopaedic Association (JOA) scoring systems were used to assess clinical results. The surgical efficacy was evaluated by comparing these parameters before and after surgery. Comparison of indicators within the same group was conducted using one‐way repeated‐measures analysis of variance or paired sample t‐tests, whereas between‐group differences were compared using an independent t‐test. Results This study included 31 individuals who underwent surgery and completed follow‐up. The follow‐up period did not show a significant loss of corrective angles. Furthermore, the Cobb angle, SP, SA, and LASD significantly decreased after surgery, whereas the LL angle, SS, and AIH significantly increased (all p 0.05). However, other parameters significantly improved during the follow‐up period at all time points, except from 3 months to the final follow‐up period (p > 0.05). Objective Medial opening‐wedge high tibial osteotomy (MOWHTO) is a surgical procedure to treat medial compartment osteoarthritis in the knee with varus deformity. However, factors such as patellar height (PH) and the sagittal plane's posterior tibial slope angle (PTSA) are potentially overlooked. This study investigated the impact of alignment correction angle guided by computer‐designed personalized surgical guide plate (PSGP) in MOWHTO on PH and PTSA, offering insights for enhancing surgical techniques. Methods This retrospective study included patients who underwent 3D‐printed PSGP‐assisted MOWHTO at our institution from March to September 2022. The paired t‐tests assessed differences in all preoperative and postoperative measurement parameters. Multivariate linear regression analysis examined correlations between PTSA, CDI (Caton–Deschamps Index), and the alignment correction magnitude. Receiver operating characteristic (ROC) curve analysis determined the threshold of the correction angle, calculating sensitivity, specificity, and area under the curve. Results A total of 107 patients were included in our study. The CDI changed from a preoperative mean of 0.97 ± 0.13 (range 0.70–1.34) to a postoperative mean of 0.82 ± 0.13 (range 0.55–1.20). Abstract A key challenge in millimeter-wave and terahertz wireless networks is blockage of the line-of-sight path between a base station and a user. User and environmental mobility can lead to blockage of highly directional beams by intervening people or objects, yielding link disruptions and poor quality of service. Here, we propose a solution to this problem which leverages the fact that, in such scenarios, users are likely to be located within the electromagnetic near field of the base station, which opens the possibility to engineer wave fronts for link maintenance. We show that curved beams, carrying data at high bit rates, can realize a link by curving around an intervening obstacle. We develop a model to analyze and experimentally evaluate the bandwidth limitations imposed by the use of self accelerating beams. We also demonstrate that such links employ the full aperture of the transmitter, even those portions which have no direct line of sight to the receiver, emphasizing that ray optics fails to capture the behavior of these near-field wave fronts. This approach, which is ideally suited for use at millimeter-wave and terahertz frequencies, opens vast new possibilities for wave front management in directional wireless networks.

Objective To explore the influencing factors of restenosis after percutaneous transluminal angioplasty in hemodialysis （HD） patients and provide a basis for predicting the risk of dysfunction and prolonging the service time. Methods Retrospective analysis was performed on the clinical and follow-up data of 335 patients who underwent percutaneous transluminal angioplasty（PTA） in Haidian District, The Third Hospital of Peking University （Haidian Hospital） from January 1 2020 to January 1 2021 as study. Results There were 187 males （55.8%） and 148 females （44.2%）. Kaplan-Meier survival analysis indicated that primary patency rates were 77.31% and 50.15% at Month 6/12 post-PTA. At Month 12 post-PTA, the risk of restenosis was significantly higher in AVG patients than that of AVF （Log-rank χ2 = 719.522, P＜0.001）. The risk of restenosis in patients with old lesions was significantly higher than that of those with new lesions （Log-rank χ2 = 23.106, P＜0.001）. The risk of restenosis in patients with local anesthesia was significantly higher than that of those with brachial plexus anesthesia （Log-rank χ2＝27.415, P＜0.001）. Multivariate Cox proportional risk regression analysis at Month 6 revealed that old/new lesions （HR = 0.393, 95%CI: 0.237-0.650, P＜0.001）, type/mode of anesthesia （HR = 3.223, 95%CI: 1.956-5.311, P＜0.001） and platelet count （HR = 1.005, 95%CI: 1.001-1.008, P = 0.008） were independent risk factors for restenosis post-PTA. Multivariate Cox proportional risk regression analysis at Month 12 indicated that type of fistula AVG （HR = 0.550, 95%CI: 0.387-0.782, P = 0.001）, old/new lesions （HR = 0.587, 95%CI: 0.422-0.815, P = 0.002） and type/mode of anesthesia （HR = 2.359, 95%CI: 1.686-3.302, P＜0.001） were independent risk factors for restenosis post-PTA. Conclusion Multiple factors may affect restenosis post-PTA in HD patients. And type/mode of anesthesia, old/new lesions and type of fistula are independent risk factors for restenosis post-PTA. Preoperative examinations and postoperative comprehensive follow-ups may prevent fistula dysfunction and help to formulate accurate surgical plans and conduct timely postoperative follow-ups.

Abstract Background The impact of sodium intake on cardiovascular disease (CVD) health and mortality has been studied for decades, including the well-established association with blood pressure. However, non-linear patterns, dose–response associations, and sex differences in the relationship between sodium and potassium intakes and overall and cause-specific mortality remain to be elucidated and a comprehensive examination is lacking. Our study objective was to determine whether intake of sodium and potassium and the sodium–potassium ratio are associated with overall and cause-specific mortality in men and women. Methods We conducted a prospective analysis of 237,036 men and 179,068 women in the National Institutes of Health-AARP Diet and Health Study. Multivariable-adjusted Cox proportional hazard regression models were utilized to calculate hazard ratios. A systematic review and meta-analysis of cohort studies was also conducted. Results During 6,009,748 person-years of follow-up, there were 77,614 deaths, 49,297 among men and 28,317 among women. Adjusting for other risk factors, we found a significant positive association between higher sodium intake (≥ 2,000 mg/d) and increased overall and CVD mortality (overall mortality, fifth versus lowest quintile, men and women HRs = 1.06 and 1.10, Pnonlinearity

The ambient air quality standard (AAQS) is a vital policy instrument for protecting the environment and human health. Hainan Province is at the forefront of China's efforts to protect its ecological environment, with an official goal to achieve world-leading air quality by 2035. However, neither the national AAQS nor the World Health Organization guideline offers sufficient guidance for improving air quality in Hainan because Hainan has well met the former while the latter is excessively stringent. Consequently, the establishment of Hainan's local AAQS becomes imperative. Nonetheless, research regarding the development of local AAQS is scarce, especially in comparatively more polluted countries such as China. The relatively high background values and significant interannual fluctuations in air pollutant concentrations in Hainan present challenges in the development of local AAQS. Our research proposes a world-class local AAQS of Hainan Province by reviewing the AAQS in major countries or regions worldwide, analyzing the influence of different statistical forms, and carefully evaluating the attainability of the standard. In the proposed AAQS, the annual mean concentration limit for PM2.5, the annual 95th percentile of daily maximum 8-h mean (MDA8) concentration limit for O3, and the peak season concentration limit for O3 are set at 10, 120, and 85 μg/m3, respectively. Our study indicates that, with effective control policies, Hainan is projected to achieve compliance with the new standard by 2035. The implementation of the local AAQS is estimated to avoid 1,526 (1,253–1,789) and 259 (132–501) premature deaths attributable to long-term exposure to PM2.5 and O3 in Hainan in 2035, respectively.

Agricultural land use and temperature change can profoundly affect soil organic carbon (SOC), but their combined effects on regulating cropland SOC fractions over broad geographic scales remain poorly understood. Based on 871 topsoil (0–20 cm) samples collected from the Sichuan Basin of China, we used random forest model and partial correlations analysis to explore how the functionally distinct SOC fractions, including particulate and mineral-associated organic C (POC and MAOC), are affected by both agricultural land use and temperature over a large scale. The results show that POC accounts for 32.54 % of the total cropland SOC and soils with high SOC contents have larger proportions of POC. Cropland type is the dominant control of the two SOC fractions and regulates the impacts of other factors. The contents of soil POC and MAOC in dryland are much lower than that in paddy field and dryland-paddy rotation land. Significant correlations between mean annual temperature and the two SOC fractions are only found in dryland and dryland-paddy rotation land, while mean annual temperature has no impact on both POC and MAOC in paddy field. Meanwhile, the correlation coefficients between mean annual temperature and POC and MAOC for dryland decreased by 61.90 % and 69.23 %, respectively, and for dryland-paddy rotation land decreased by 28.00 % and 14.29 %, respectively, after removing correlations with net primary productivity. Additionally, correlation between mean annual temperature and POC is slightly higher than that between mean annual temperature and MAOC in dryland, while the reverse in dryland-paddy rotation land. These results suggest that the temperature effects on POC and MAOC are cropland type-dependent. To accurately predicting future cropland SOC change and reliably evaluate the potential contribution of cropland soil C sequestration, fraction-specific control mechanisms related to cropland type should be sufficiently considered in carbon models.

Cyclosporine is an immunosuppressant used to prevent organ rejection in kidney, liver, and heart allogeneic transplants. This study aimed to assess the safety of cyclosporine through the analysis of adverse events (AEs) related to cyclosporine in the US Food and Drug Administration Adverse Event Reporting System (FAERS). To detect AEs associated with cyclosporine, a pharmacovigilance analysis was conducted using four algorithms on the FAERS database: reporting odds ratio (ROR), proportional reporting ratio (PRR), Bayesian confidence propagation neural network (BCPNN), and empirical Bayes geometric mean (EBGM). A statistical analysis was performed on data extracted from the FAERS database, covering 19,582 case reports spanning from 2013 to 2022. Among these cases, 3,911 AEs were identified, with 476 linked to cyclosporine as the primary suspected drug. Cyclosporin-induced AEs targeted 27 System Organ Classes (SOCs). Notably, the highest case at the SOC level included eye disorders, injury, poisoning, and procedural complications, as well as immune system disorders, all of which are listed on the cyclosporine label. Furthermore, we discovered novel potential AEs associated with hepatobiliary disorders, among others. Moreover, unexpected adverse drug reactions (ADRs), such as biliary anastomosis complication and spermatozoa progressive motility decrease, were identified. Importantly, these newly identified ADRs were not mentioned on the cyclosporine label, which were involved in injury, poisoning, and procedural complications, and investigations at the SOC level. The study used pharmacovigilance analysis of FAERS database to identify new and unexpected potential ADRs relating to cyclosporine, which can provide safety tips for the safe use of cyclosporine.

Fiber reinforced polymer (FRP) bars have become increasingly popular, while the studies on durability of FRP bars are primarily on small-diameter FRP bars. This study investigated the tensile strength retention in glass FRP (GFRP) bars of different diameters (13 mm and 25 mm) after immersion in an alkaline solution (pH=12.6) at various temperatures (20 °C, 40 °C and 60 °C) for 1, 2, 3, and 6 months. The results reveal that the degradation of GFRP bars was slow at 20 °C, accelerated but not pronounced at 40 °C and considerable at 60 °C. Particularly, the 13 mm diameter GFRP bars exhibited a more significant reduction in tensile strength, with a decrease of 20.12 % after 6 months, while the 25 mm diameter bars only decreased by 13.23 %. Results reveal that, importantly, degradation of GFRP bars is primarily attributed to the diffusion of the moisture and alkalis, which disrupts the bond between the fibers and the matrix, causing interface damage. Finally, based on the Arrhenius theory, it is predicted that the tensile strength retention of 13 mm and 25 mm diameter GFRP bars will be 66.4 % and 79.8 %, respectively, after 50 years of exposure at an average annual temperature of 35 °C. The important finding that the small-diameter FRP bars are more vulnerable to the alkaline exposure than larger diameter bars suggests that the current studies on durability of FRP bars are conservative to be referred in practice.

Abstract Intrahepatic cholangiocarcinoma (iCCA) is a highly lethal biliary epithelial cancer in the liver. Here, Laminin subunit gamma‐2 (LAMC2) with important oncogenic roles in iCCA is discovered. In a total of 231 cholangiocarcinoma patients (82% of iCCA patients) across four independent cohorts, LAMC2 is significantly more abundant in iCCA tumor tissue compared to normal bile duct and non‐tumor liver. Among 26.3% of iCCA patients, LAMC2 gene is amplified, contributing to its over‐expression. Functionally, silencing LAMC2 significantly blocks tumor formation in orthotopic iCCA mouse models. Mechanistically, it promotes EGFR protein translation via interacting with nascent unglycosylated EGFR in the endoplasmic reticulum (ER), resulting in activated EGFR signaling. LAMC2‐mediated EGFR translation also depends on its interaction with the ER chaperone BiP via their C‐terminus. Together LAMC2 and BiP generate a binding “pocket” of nascent EGFR and facilitate EGFR translation. Consistently, LAMC2‐high iCCA patients have poor prognosis in two iCCA cohorts. LAMC2‐high iCCA cells are highly sensitive to EGFR tyrosine kinase inhibitors (TKIs) treatment both in vitro and in vivo. Together, these data demonstrate LAMC2 as an oncogenic player in iCCA by promoting EGFR translation and an indicator to identify iCCA patients who may benefit from available EGFR‐targeted TKIs therapies.

Summary: Thymocyte development requires precise control of PI3K-Akt signaling to promote proliferation and prevent leukemia and autoimmune disorders. Here, we show that ablating individual clusters of the miR-17∼92 family has a negligible effect on thymocyte development, while deleting the entire family severely impairs thymocyte proliferation and reduces thymic cellularity, phenocopying genetic deletion of Dicer. Mechanistically, miR-17∼92 expression is induced by Myc-mediated pre-T cell receptor (TCR) signaling, and miR-17∼92 promotes thymocyte proliferation by suppressing the translation of Pten. Retroviral expression of miR-17∼92 restores the proliferation and differentiation of Myc-deficient thymocytes. Conversely, partial deletion of the miR-17∼92 family significantly delays Myc-driven leukemogenesis. Intriguingly, thymocyte-specific transgenic miR-17∼92 expression does not cause leukemia or lymphoma but instead aggravates skin inflammation, while ablation of the miR-17∼92 family ameliorates skin inflammation. This study reveals intricate roles of the miR-17∼92 family in balancing thymocyte development, leukemogenesis, and autoimmunity and identifies those microRNAs (miRNAs) as potential therapeutic targets for leukemia and autoimmune diseases.

For liquid crystals (LCs) and liquid crystalline polymers (LCPs), a chiral smectic C (SmC*) phase has been mandatory for breaking the symmetry and achieving ferroelectricity. However, this SmC* phase leads to rather low spontaneous polarization (Ps, 0.1–5 mC/m2), which has limited their usage in various electronic and electro-optical applications. In this mini-review, we highlight three new types of ferroelectric LCPs with high Ps values reported in the last decade. The first system refers to the ferroelectric nematic LCs and LCPs. The large dipole moment (>9 Debye or D) and oblique molecular shape induce a polar packing of calamitic nematics. The Ps can reach as high as 40 mC/m2. The second example is a ferroelectric supramolecular LCP, in which the highly polar cyano groups in the core lead to a polar structure of the hexagonal columnar phase after electric poling. The Ps can reach ∼ 20 mC/m2. The third system utilizes highly dipolar sulfonyl groups (dipole moment ∼4.5 D) in the side chains of mesogen-free comb-shaped LCPs. By combining finely tuned dipolar interactions and mobile LC order, these mesogen-free comb-like LCPs have shown good potential for ferroelectricity with high Ps. These ferroelectric LCPs with high Ps will enable new electronic and electro-optical applications in the future.

In previous studies of disc degeneration, the structural and mechanical properties of the endplate were often neglected. In this paper, the station legislation was used to construct an animal model of minor trauma disc degeneration, and the mechanism of disc degeneration was further investigated by observing the changes of mesoscopic structure and developing the mechanical properties of endplate bone.Twenty-eight 6-month-old Japanese white rabbits were divided into two groups: control group and experimental group. An animal model of intervertebral disc degeneration was established by upright experiment in the experimental group. The bone mesoscopic structures in different areas of each endplate were observed by histological and imaging methods, and the mechanical properties of the endplates were measured by indentation method. The two groups of data were compared by one-way ANOVA.After the experimental animals stood for 17 weeks, The experimental group showed the characteristics of early disc degeneration. The microstructure of the degenerative group showed that the end plate mineralization degree was higher, the bone mass was larger, and the number and thickness of bone trabeculae were larger. The results of indentation test showed that the mechanical properties of the degeneration group were enhanced, and the lower endplate was obviously enhanced.We successfully established a model of human disc degeneration with non invasive trauma and more consistent with the process of human disc degeneration through the standing experiment. In the experimental group, the internal structure of the endplate was dense and pore distance was reduced. The change of bone mesoscopic structure further affects the endplate, resulting in the enhancement of the mechanical properties of the endplate after intervertebral disc degeneration. The reduction of the pore distance and the narrowing of the internal channel structure of the endplate also hinder the nutrition supply of the intervertebral disc, which may be the key reason affecting the degeneration of the intervertebral disc. A biomechanical method for investigating the mechanism of intervertebral disc degeneration can be provided in this paper.

PURPOSE: Through three neurocritical care unit (NCCU) surveys in China, we tried to understand the development status of neurocritical care and clarify its future development. METHODS: Using a cross-sectional survey method and self-report questionnaires, the number and quality of NCCUs were investigated through three steps: administering the questionnaire, sorting the survey data, and analyzing the survey data. RESULTS: At the second and third surveys, the number of NCCUs (76/112/206) increased by 47% and 84%, respectively. The NCCUs were located in tertiary grade A hospitals or teaching hospitals (65/100/181) in most provinces (24/28/29). The numbers of full-time doctors (359/668/1337) and full-time nurses (904/1623/207) in the NCCUs increased, but the doctor–bed ratio and nurse–bed ratio were still insufficient (0.4:1 and 1.3:1). CONCLUSION: In the past 20 years, the growth rate of NCCUs in China has accelerated, while the allocation of medical staff has been insufficient. Although most NCCU hospital bed facilities and instruments and equipment tend to be adequate, there are obvious defects in some aspects of NCCUs.

The design of LDMOS (Lateral double diffused metal oxide semiconductor) devices with CFP (Contact field plate) has been of great significance in recent years, according to its advantages of low resistance and high switch efficiency. In this paper, this ultra-low $R_{\mathrm{ on,sp}}$ (Specific on-resistance) LDMOS device is simulated, designed, and fabricated. The effects on FOM (Figures-of-merits) characters from physical dimensions, including field plate length ${L}$ , field plate thickness ${H}$ , and slot contact width ${W}$ , have been analyzed and discussed. The best device structure is proposed through simulation, and a related fabrication process is introduced correspondingly. Finally, the electrical measurement results show that $R_{\mathrm{ on,sp}}$ can achieve as low as 6.9m $\Omega \cdot $ mm2 when the source-drain ${BV}$ (Breakdown voltage) arrives at 34.1V, which is improved by 47.1% compared with conventional devices. Furthermore, the TLP (Transmission line pulse) test results indicate that the device owns an ideal SOA (Safe operation area) from both typical devices (W $=\,\,10~\mu \text{m}$ ) and very large devices (W = 2mm).

Abstract For the current stellar spectral simulation can not realize the stellar color temperature information with large dynamic range simulation, this paper proposes a broad spectrum high-resolution subdivision and spatial beam zoning modulation combined with a large dynamic range of stellar radiation information simulation method, designed a kind of imaging and non-imaging stellar radiation information simulation optical system, using an optical system to achieve multi-color temperature spectrum and large dynamic range stellar simulation. The experimental results show that the designed system can simultaneously achieve the spectral simulation accuracy (single point evaluation) better than ± 7% in the range of spectral 450–1000 nm and color temperature 3000–11,000 K; on the premise of ensuring the spectral simulation accuracy, the magnitude simulation range reaches 0 to + 12 Mv, and the magnitude simulation accuracy is better than ± 0.05 Mv; Accurate simulation of stellar spectral information and energy large dynamic range tuning is realized, and the system is extended. The system function has been extended to realize the switching of broadband and narrowband modes, The half-peak width of the narrowband output beam is better than 4.1 nm, which extends the application of the spectral simulation technology and provides the theoretical and technical basis for the ground calibration of the development of the high-precision stellar radiation information ground simulation system.

Highlights The well-conceived Pt–Ni3S2 heteronanocrystals with dual-monodispersed characteristics are synthesized through interfacial electronic modulation. The asymmetrical charge distribution at Pt–Ni3S2 hetero-interface results in the formation of high-valent Ni sites and negatively-charged Pt δ−. It eventually accelerates water dissociation and achieves the steady concurrent generation of value-added formate and hydrogen.

Due to the limitations of small targets in remote sensing images, such as background noise, poor information, and so on, the results of commonly used detection algorithms in small target detection is not satisfactory. To improve the accuracy of detection results, we develop an improved algorithm based on YOLOv8, called LAR-YOLOv8. First, in the feature extraction network, the local module is enhanced by using the dual-branch architecture attention mechanism, while the vision transformer block is used to maximize the representation of the feature map. Second, an attention-guided bidirectional feature pyramid network is designed to generate more discriminative information by efficiently extracting feature from the shallow network through a dynamic sparse attention mechanism, and adding top–down paths to guide the subsequent network modules for feature fusion. Finally, the RIOU loss function is proposed to avoid the failure of the loss function and improve the shape consistency between the predicted and ground-truth box. Experimental results on NWPU VHR-10, RSOD, and CARPK datasets verify that LAR-YOLOv8 achieves satisfactory results in terms of mAP (small), mAP, model parameters, and FPS, and can prove that our modifications made to the original YOLOv8 model are effective.

Abstract Light is a powerful and sustainable resource, but it can be detrimental to the performance and longevity of optical devices. Materials with near-zero light reflectance, i.e. superblack materials, are sought to improve the performance of several light-centered technologies. Here we report a simple top-down strategy, guided by computational methods, to develop robust superblack materials following metal-free wood delignification and carbonization (1500 °C). Subwavelength severed cells evolve under shrinkage stresses, yielding vertically aligned carbon microfiber arrays with a thickness of ~100 µm and light reflectance as low as 0.36% and independent of the incidence angle. The formation of such structures is rationalized based on delignification method, lignin content, carbonization temperature and wood density. Moreover, our measurements indicate a laser beam reflectivity lower than commercial light stoppers in current use. Overall, the wood-based superblack material is introduced as a mechanically robust surrogate for microfabricated carbon nanotube arrays.

Abstract Hollow multi-shell mesoporous metal–organic framework (MOF) particles with accessible compartmentalization environments, plentiful heterogeneous interfaces, and abundant framework diversity are expected to hold great potential for catalysis, energy conversion, and biotechnology. However, their synthetic methodology has not yet been established. In this work, a facile dual-template-directed successive assembly approach has been developed for the preparation of monodisperse hollow multi-shell mesoporous MOF (UiO-66-NH2) particles through one-step selective etching of successively grown multi-layer MOFs with alternating two types of mesostructured layers. This strategy enables the preparation of hollow multi-shell mesoporous UiO-66-NH2 nanostructures with controllable shell numbers, accessible mesochannels, large pore volume, tunable shell thickness and chamber sizes. The methodology relies on creating multiple alternating layers of two different mesostructured MOFs via dual-template-directed successive assembly and their difference in framework stability upon chemical etching. Benefiting from the highly accessible Lewis acidic sites and the accumulation of reactants within the multi-compartment architecture, the resultant hollow multi-shell mesoporous UiO-66-NH2 particles exhibit enhanced catalytic activity for CO2 cycloaddition reaction. The dual-template-directed successive assembly strategy paves the way toward the rational construction of elaborate hierarchical MOF nanoarchitectures with specific physical and chemical features for different applications.

IntroductionHIV molecular network based on genetic distance (GD) has been extensively utilized. However, the GD threshold for the non-B subtype differs from that of subtype B. This study aimed to optimize the GD threshold for inferring the CRF01_AE molecular network.MethodsNext-generation sequencing data of partial CRF01_AE pol sequences were obtained for 59 samples from 12 transmission pairs enrolled from a high-risk cohort during 2009 and 2014. The paired GD was calculated using the Tamura-Nei 93 model to infer a GD threshold range for HIV molecular networks.Results2,019 CRF01_AE pol sequences and information on recent HIV infection (RHI) from newly diagnosed individuals in Shenyang from 2016 to 2019 were collected to construct molecular networks to assess the ability of the inferred GD thresholds to predict recent transmission events. When HIV transmission occurs within a span of 1-4 years, the mean paired GD between the sequences of the donor and recipient within the same transmission pair were as follow: 0.008, 0.011, 0.013, and 0.023 substitutions/site. Using these four GD thresholds, it was found that 98.9%, 96.0%, 88.2%, and 40.4% of all randomly paired GD values from 12 transmission pairs were correctly identified as originating from the same transmission pairs. In the real world, as the GD threshold increased from 0.001 to 0.02 substitutions/site, the proportion of RHI within the molecular network gradually increased from 16.6% to 92.3%. Meanwhile, the proportion of links with RHI gradually decreased from 87.0% to 48.2%. The two curves intersected at a GD of 0.008 substitutions/site.DiscussionA suitable range of GD thresholds, 0.008-0.013 substitutions/site, was identified to infer the CRF01_AE molecular transmission network and identify HIV transmission events that occurred within the past three years. This finding provides valuable data for selecting an appropriate GD thresholds in constructing molecular networks for non-B subtypes.

Objective This study aimed to investigate the knowledge, attitude and practice (KAP) of poststroke depression (PSD) among patients with PSD and their family members.Design Web-based cross-sectional study.Setting This study was conducted in Heilongjiang Province between October 2022 and April 2023.Participants Patients with PSD and their family members.Primary and secondary outcome measures KAP scores.Method The self-administered questionnaire comprised demographic characteristics, knowledge dimension, attitude dimension and practice dimensions.Results A total of 489 valid questionnaires were collected, with the average age of the subjects was 54.68±13.80 years, and including 258 (53.09%) patients who had a stroke. The mean scores for KAP were 6.36±2.66 (possible range: 0–10), 29.07±5.18 (possible range: 8–40) and 37.50±8.49 (possible range: 10–50), respectively. Concerning KAP scores, no differences were found between patients with PSD and their family members. Multivariate logistic regression analysis showed that retirement (OR=0.29, 95% CI 0.11 to 0.77, p=0.012) and monthly income less than ¥2000 (OR=0.46, 95% CI 0.27, 0.79, p=0.005) were independently associated with adequate knowledge. ObjectiveTo investigate the clinical manifestations, imaging and pathological features, treatment methods and prognosis of primary leiomyosarcoma of kidney, and the choice of treatment with tumor thrombus.MethodsThe clinical data of a patient with primary renal leiomyosarcoma complicated with inferior vena cava and right atrial tumor thrombus were retrospectively analyzed. Radical resection of right kidney without cardiopulmonary bypass and removal of inferior vena cava and right atrial tumor thrombus were performed. Adjuvant intravenous chemotherapy was given according to the results, and follow-up observation was made.ResultsPostoperative pathological findings were: leiomyosarcoma (right renal tumor), the size of the mass was about 12.1 cm, and no cancer was found at the incision end of the right ureter.ConclusionPrimary leiomyosarcoma of kidney is rare in clinical practice, and complication of right atrial tumor embolus is even rarer. The disease has high malignant degree and poor prognosis. The clinical manifestations and imaging examinations were non-specific, and pathological diagnosis was the gold standard. Radical surgical resection is the main treatment method at present, and it provides experience for the treatment of grade IV tumor thrombus without cardiopulmonary bypass.

Abstract The reductions in aerosols often exacerbate climate warming. It remains unclear how to effectively alleviate PM2.5 pollution while minimizing the penalty on climate warming. Here we identify the clean air measures in China that are associated with low aerosol climate penalty efficiency (ACPE), which is defined as aerosol radiative forcing per unit PM2.5 concentration reduction. The measures in transportation, residential combustion, and open burning sectors generally caused lower ACPE [0.07, 0.24, and 0.10 (W m−2)/(μg m−3)] than those from other sectors [0.34–0.46 (W m−2)/(μg m−3)]. This is ascribed to relatively small decreases in cloud concentration nuclei per unit PM2.5 reduction in these sectors, which is further attributed to either relatively low aerosol hygroscopicity or relatively small decrease in aerosol number. BackgroundFront-line therapy with an EGFR tyrosine kinase inhibitor (TKI) is the standard of care for treating patients with advanced nonsquamous NSCLC with the common sensitizing EGFR exon 19 deletion and exon 21 L858R point mutations. However, EGFR TKI resistance inevitably develops. The optimal subsequent therapy remains to be identified, although platinum-containing chemotherapy regimens are often administered. Our objectives were to describe baseline characteristics, survival, and subsequent treatment patterns for patients with advanced nonsquamous NSCLC with EGFR exon 19 deletion or L858R mutation who received a platinum-based combination regimen after front-line EGFR TKI therapy.MethodsThis retrospective study used a nationwide electronic health record-derived deidentified database to select adult patients with advanced nonsquamous NSCLC, evidence of EGFR exon 19 deletion or L858R mutation, and ECOG performance status of 0-2 who initiated platinum-containing chemotherapy, with or without concomitant immunotherapy, from 1-January-2011 to 30-June-2020 following receipt of any EGFR TKI as first-line therapy or, alternatively, a first- or second-generation EGFR TKI (erlotinib, afatinib, gefitinib, dacomitinib) as first-line therapy followed by the third-generation EGFR TKI osimertinib as second-line therapy. Data cut-off was 30-June-2022. The Kaplan-Meier method was used to estimate overall survival (OS) after initiation of pemetrexed-platinum (n=119) or any platinum-based combination regimen (platinum cohort; n=311).ResultsThe two cohorts included two-thirds women (65%-66%) and 57%-58% nonsmokers; median ages were 66 and 65 years in pemetrexed-platinum and platinum cohorts, respectively. Median OS was 10.3 months (95% CI, 8.1-13.9) from pemetrexed-platinum initiation and 12.4 months (95% CI, 10.2-15.2) from platinum initiation; 12-month survival rates were 48% and 51%, respectively; 260 patients (84%) had died by the end of the study.ConclusionThe suboptimal survival outcomes recorded in this study demonstrate the unmet need to identify more effective subsequent treatment regimens for patients with EGFR-mutated advanced nonsquamous NSCLC after EGFR TKI resistance develops.

AimTo investigate whether age at first sexual intercourse could lead to any changes in the risk of oral cavity cancer.MethodsA two-sample mendelian randomization was conducted using genetic variants associated with age at first sexual intercourse in UK biobank as instrumental variables. Summary data of Northern American from a previous genome-wide association study aimed at oral cavity cancer was served as outcome. Three analytical methods: inverse variance-weighted, mendelian randomization Egger, and weighted median were used to perform the analysis, among which inverse variance-weighted was set as the primary method. Robustness of the results was assessed through Cochran Q test, mendelian randomization Egger intercept tests, MR PRESSO, leave one out analysis and funnel plot.ResultsThe primary analysis provided substantial evidence of a positive causal relationship age at first sexual intercourse and the risk of oral cavity cancer (p = 0.0002), while a delayed age at first sexual intercourse would lead to a decreased risk of suffering oral cavity cancer (β = -1.013). The secondary outcomes confirmed the results (all β < 0) and all assessments supported the robustness, too (all p > 0.05).ConclusionThe study demonstrates that a delayed sexual debut would provide protection against OCC, thus education on delaying sexual intercourse should be recommended.

Background: Everolimus is an inhibitor of the mammalian target of rapamycin and is used to treat various tumors. The presented study aimed to evaluate the Everolimus-associated adverse events (AEs) through data mining of the US Food and Drug Administration Adverse Event Reporting System (FAERS).Methods: The AE records were selected by searching the FDA Adverse Event Reporting System database from the first quarter of 2009 to the first quarter of 2022. Potential adverse event signals were mined using the disproportionality analysis, including reporting odds ratio the proportional reporting ratio the Bayesian confidence propagation neural network and the empirical Bayes geometric mean and MedDRA was used to systematically classify the results.Results: A total of 24,575 AE reports of Everolimus were obtained using data from the FAERS database, and Everolimus-induced AEs occurrence targeted 24 system organ classes after conforming to the four algorithms simultaneously. The common significant SOCs were identified, included benign, malignant and unspecified neoplasms, reproductive system and breast disorders, etc. The significant AEs were then mapped to preferred terms such as stomatitis, pneumonitis and impaired insulin secretion, which have emerged in the study usually reported in patients with Everolimus. Of note, unexpected significant AEs, including biliary ischaemia, angiofibroma, and tuberous sclerosis complex were uncovered in the label.Conclusion: This study provided novel insights into the monitoring, surveillance, and management of adverse drug reaction associated with Everolimus. The outcome of serious adverse events and the corresponding detection signals, as well as the unexpected significant adverse events signals are worthy of attention in order to improving clinical medication safety during treatment of Everolimus.

The toxicity of 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is generally believed to be mediated by aryl hydrocarbon receptor (AhR), but some evidence suggests that the effects of TCDD can also be produced through AhR-independent mechanisms. In previous experiments, we found that mainly AhR-dependent mechanism was involved in the migration inhibition of glioblastoma U87 cells by TCDD. Due to the heterogeneity of glioblastomas, not all tumor cells have significant AhR expression. The effects and mechanisms of TCDD on the migration of glioblastomas with low AhR expression are still unclear. We employed a glioblastoma cell line A172 with low AhR expression as a model, using wound healing and Transwell® assay to detect the effect of TCDD on cell migration. We found that TCDD can inhibit the migration of A172 cells without activating AhR signaling pathway. Further, after being pre-treated with AhR antagonist CH223191, the inhibition of TCDD on A172 cells migration was not changed, indicating that the effect of TCDD on A172 cells is not dependent on AhR activation. By transcriptome sequencing analysis, we propose dysregulation of the expression of certain migration-related genes, such as IL6, IL1B, CXCL8, FOS, SYK, and PTGS2 involved in cytokines, MAPK, NF-κB, and IL-17 signaling pathways, as potential AhR-independent mechanisms that mediate the inhibition of TCDD migration in A172 cells.

Abstract Background Several factors can influence the risk of dental caries, among which dietary factors have a significance impact on the occurrence of dental caries. The limitation of current studies is that they only focus on the influence of individual foods on the risk of dental caries. This study use cluster analysis to examine the relationship between dietary patterns and dental caries experience among adolescents aged 12–15. Methods Based on data from the first oral epidemic survey in Shanxi Province, a cross-sectional study was conducted among 11,351 adolescents aged 12–15 in Shanxi Province through oral examination and questionnaires. The questionnaire included the intake frequency of seven types of food. Descriptive statistics, cluster analysis, and multinomial logistic regression were used to analyze the association between dietary patterns and dental caries experience. Abstract Immunotherapies have revolutionized the treatment paradigms of various types of cancers. However, most of these immunomodulatory strategies focus on harnessing adaptive immunity, mainly by inhibiting immunosuppressive signaling with immune checkpoint blockade, or enhancing immunostimulatory signaling with bispecific T cell engager and chimeric antigen receptor (CAR)-T cell. Although these agents have already achieved great success, only a tiny percentage of patients could benefit from immunotherapies. Actually, immunotherapy efficacy is determined by multiple components in the tumor microenvironment beyond adaptive immunity. Cells from the innate arm of the immune system, such as macrophages, dendritic cells, myeloid-derived suppressor cells, neutrophils, natural killer cells, and unconventional T cells, also participate in cancer immune evasion and surveillance. Considering that the innate arm is the cornerstone of the antitumor immune response, utilizing innate immunity provides potential therapeutic options for cancer control. Up to now, strategies exploiting innate immunity, such as agonists of stimulator of interferon genes, CAR-macrophage or -natural killer cell therapies, metabolic regulators, and novel immune checkpoint blockade, have exhibited potent antitumor activities in preclinical and clinical studies. Here, we summarize the latest insights into the potential roles of innate cells in antitumor immunity and discuss the advances in innate arm-targeted therapeutic strategies.

Abstract As current trend for physician tools, ChatGPT can sift through massive amounts of information and solve problems through easy-to-understand conversations, ultimately improving efficiency. Mycosis is currently facing great challenges, including high fungal burdens, high mortality, limited choice of antifungal drugs and increasing drug resistance. To address these challenges, We asked ChatGPT for fungal infection scenario-based questions and assessed its appropriateness, consistency, and potential pitfalls. We concluded ChatGPT can provide compelling responses to most prompts, including diagnosis, recommendations for examination, treatment and rational drug use. Moreover, we summarized exciting future applications in mycosis, such as clinical work, scientific research, education and healthcare. However, the largest barriers to implementation are deficits in indiviudal advice, timely literature updates, consistency, accuracy and data safety. To fully embrace the opportunity, we need to address these barriers and manage the risks. We expect that ChatGPT will become a new weapon in in the battlefield of mycosis.

Abstract Protecting human health from fine particulate matter (PM) pollution is the ambitious goal of clean air actions, but current control strategies largely ignore the role of source-specific PM toxicity. Here, we proposed health-oriented control strategies by integrating the unequal toxic potencies of the most polluting industrial PMs. Iron and steel industry (ISI)-emitted PM2.5 exhibit about one order of magnitude higher toxic potency than those of cement and power industries. Compared with the current mass-based control strategy (prioritizing implementation of ultralow emission standards in the power sector), the proposed health-oriented control strategy (priority control of the ISI sector) could generate 5.4 times higher reduction in population-weighted toxic potency-adjusted PM2.5 exposure among polluting industries in China. Furthermore, the marginal abatement cost per unit of toxic potency-adjusted mass of ISI-emitted PM2.5 is only a quarter of that of the other two sectors under ultralow emission scenarios. We highlight that a health-oriented air pollution control strategy is urgently required to achieve cost-effective reductions in particulate exposure risks.

Abstract Due to redundancy in the spatiotemporal neighborhood and the global dependency between video frames, video recognition remains a challenge. Some prior works have been mainly driven by 3D convolutional neural networks (CNNs) or 2D CNNs with a well‐designed module for temporal information. However, convolution‐based networks lack the capability to capture the global dependency due to the limited receptive field. Alternatively, transformer for video recognition is proposed to build long‐range dependency between frame patches. Nevertheless, most transformer‐based networks have significant computational costs because attention is calculated among all the tokens. Based on these observations, we propose an efficient network which we dub LGANet. Unlike conventional CNNs and transformers for video recognition, the LGANet can tackle both spatiotemporal redundancy and dependency by learning local and global token affinity in shallow and deep layers, respectively . Specifically, local attention is implemented in the shallow layers to reduce parameters and eliminate redundancy. In the deep layers, spatial‐wise and channel‐wise self‐attention are embedded to realize the global dependency of high‐level features. Moreover, several key designs are made in the multi‐head self‐attention (MSA) and feed‐forward network (FFN). Extensive experiments are conducted on the popular video benchmarks, such as Kinetics‐400, Something‐Something V1&V2. Without any bells and whistles, the LGANet achieves state‐of‐the‐art performance. This paper proposed a positioning and mapping algorithm of sliding window optimization for substation monitoring robots to solve the problems of low precision location and poor robustness of the existing laser odometer in power inspection outdoor scene mapping. A tightly coupled simultaneous localization and mapping (SLAM) algorithm was proposed based on 16-wire LiDAR and inertial measurement unit (IMU). Firstly, the paper estimated the IMU and corrected the motion distortion of the laser point cloud by linear interpolation. Secondly, scene features were extracted by curvature and classified according to different feature properties. The local map was constructed in the sliding window using the inter-frame matching module. Finally, the joint optimization function was built using the distance and IMU data obtained by matching the frame with the local map. The paper used the KITTI and self-recorded datasets to conduct the experiments. The results show that the improved method’s accuracy outperforms the lightweight and ground-optimized LiDAR odometry and Mapping (Lego-LOAM) and LiDAR inertial odometry and mapping (LIO-Mapping) and draws a broad application prospect in power inspection field.
