Benthic cyanobacterial mats (BCMs) are natural phenomena in marine environments. Reports of BCMs occurring across coastal marine environments have increased, partly driven by nutrient loading and climate change; thus, there is a need to understand the diversity involved in the proliferations and potential toxicity of the BCMs. Furthermore, marine cyanobacterial mats are observed growing on and affecting the health of corals with one specific cyanobacterial genus, Roseofilum, dominating the microbial mats associated with black band disease (BBD), a destructive polymicrobial disease that affects corals. To explore the diversity of Roseofilum, cyanobacterial mats from various marine habitats were sampled, and individual isolates were identified based on morphology, 16S rRNA gene phylogenies, 16S–23S ITS rRNA region sequence dissimilarities, and phylogenomics. Four novel species of Roseofilum were isolated from benthic marine mats, three from the coasts of Florida, United States (R. capinflatum sp. nov., R. casamattae sp. nov., and R. acuticapitatum sp. nov.) and one from the coast of France (R. halophilum sp. nov.). Our analyses revealed that Roseofilum associated with coral BBD and those not associated with corals but rather from coastal benthic mats are systematically distinct based on both phylogenetic and phylogenomic analyses. We report first observations of three coral diseases including black band disease on Acropora, a syndrome resembling yellow band disease and red band disease on Porites colonies from depth 5–6 m at Abu-Musa Island after widespread coral bleaching in the northern part of the Persian Gulf in October 2012.

Pervasive and sustained coral diseases contribute to the systemic degradation of reef ecosystems, however, to date an understanding of the physicochemical controls on a coral disease event is still largely lacking. Water circulation and residence times and submarine groundwater discharge (SGD) all determine the degree to which reef organisms are exposed to the variable chemistry of overlying waters; understanding these physical controls is thus necessary to interpret spatial patterns in coral health. The recent discovery of coral Black Band Disease at Mākua Reef on Kaua‘i, Hawai‘i prompted an investigation into the physicochemical drivers and geomorphic controls of reef water circulation, and the temporally variable nutrient fluxes derived from SGD. Results reveal localized stagnant water parcels at Mākua Reef where groundwater-derived high nutrient loading and low salinities act in concert as stressors to coralline health – and where Black Band Disease was uniquely identified. The observed high nutrient levels during low tide conditions are likely associated with nearby upstream cesspools and drain fields. Information obtained using such a multidisciplinary approach has direct value for successful management of coastal aquifers and the health and sustainability of adjacent nearshore coral reef ecosystems.

Roseofilum reptotaenium is a gliding, filamentous, phycoerythrin-rich cyanobacterium that has been found only in the horizontally migrating, pathogenic microbial mat, black band disease (BBD) on Caribbean corals. R. reptotaenium dominates the BBD mat in terms of biomass and motility, and the filaments form the mat fabric. This cyanobacterium produces the cyanotoxin microcystin, predominately MC-LR, and can tolerate high levels of sulfide produced by sulfate reducing bacteria (SRB) that are also associated with BBD. Laboratory cultures of R. reptotaenium infect coral fragments, suggesting that the cyanobacterium is the primary pathogen of BBD, but since this species cannot grow axenically and Koch’s Postulates cannot be fulfilled, it cannot be proposed as a primary pathogen. However, R. reptotaenium does play several major pathogenic roles in this polymicrobial disease. Here, we provide an overview of the ecology of this coral pathogen and present new information on R. reptotaenium ecophysiology, including roles in the infection process, chemotactic and other motility responses, and the effect of pH on growth and motility. Additionally, we show, using metabolomics, that exposure of the BBD microbial community to the cyanotoxin MC-LR affects community metabolite profiles, in particular those associated with nucleic acid biosynthesis.

Seawater temperature anomalies associated with warming climate have been linked to increases in coral disease outbreaks that have contributed to coral reef declines globally. However, little is known about how seasonal scale variations in environmental factors influence disease dynamics at the level of individual coral colonies. In this study, we applied a multi-state Markov model (MSM) to investigate the dynamics of black band disease (BBD) developing from apparently healthy corals and/or a precursor-stage, termed ‘cyanobacterial patches’ (CP), in relation to seasonal variation in light and seawater temperature at two reef sites around Pelorus Island in the central sector of the Great Barrier Reef. The model predicted that the proportion of colonies transitioning from BBD to Healthy states within three months was approximately 57%, but 5.6% of BBD cases resulted in whole colony mortality. According to our modelling, healthy coral colonies were more susceptible to BBD during summer months when light levels were at their maxima and seawater temperatures were either rising or at their maxima. In contrast, CP mostly occurred during spring, when both light and seawater temperatures were rising. This suggests that environmental drivers for healthy coral colonies transitioning into a CP state are different from those driving transitions into BBD. Our model predicts that (1) the transition from healthy to CP state is best explained by increasing light, (2) the transition between Healthy to BBD occurs more frequently from early to late summer, (3) 20% of CP infected corals developed BBD, although light and temperature appeared to have limited impact on this state transition, and (4) the number of transitions from Healthy to BBD differed significantly between the two study sites, potentially reflecting differences in localised wave action regimes.

Tissue degradation and mediated mortality have turned into a major threat to coral reef systems around the world. Detailed knowledge on interactions of prime biological factors that mediate tissue loss and mortality is of paramount importance in understanding the prevailing reef health scenario and to trial management actions. In the present study, a series of benthic surveys were conducted in Lakshadweep islands to understand the interactions of plausible biological factors in causing tissue loss and mediated mortality. Interactions of biological scenarios were prioritized using hierarchical regression analysis. The hierarchical regression model analysis revealed black band disease ( β = 0.59; p < 0.001) and algal interactions ( β = 0.48; p < 0.001) as the major factors responsible for tissue-loss-mediated coral mortality in the region. The observations from the hierarchical analysis were used to derive vulnerability maps based on weighted overlay analysis. The vulnerability mapping revealed that lagoon of Kalpeni Island is very highly vulnerable to coral degradation with 683.5 ha followed by Kavaratti (70.2 ha), Bangaram (70 ha), and Amini (47 ha). Chethalath and Agatti lagoons were inferred as low vulnerable sites where coral reefs can sustain. The vulnerability maps derived can be used as a baseline observation to identify areas of very high vulnerability and specific stressor prevalent in those sites. Black band disease (BBD) affects many coral species worldwide and is considered a major contributor to the decline of reef-building coral. On the Florida Reef Tract BBD is most prevalent during summer and early fall when water temperatures exceed 29 °C. BBD is rarely reported in pillar coral ( Dendrogyra cylindrus) throughout the Caribbean, and here we document for the first time the appearance of the disease in this species on Florida reefs. The highest monthly BBD prevalence in the D. cylindrus population were 4.7% in 2014 and 6.8% in 2015. In each year, BBD appeared immediately following a hyperthermal bleaching event, which raises concern as hyperthermal seawater anomalies become more frequent. [ABSTRACT FROM AUTHOR]

Black band disease (BBD) of corals is a horizontally migrating, pathogenic, polymicrobial mat community which is active above a temperature threshold of 27.5°C on the reef. Bacterial isolates from BBD, the surface mucopolysaccharide layer (SML) of healthy corals and SML of healthy areas of BBD-infected corals were tested for production of short- to medium-chain acyl homoserine lactones (AHLs) using the Chromobacterium violaceum CV026 reporter strain. Of 110 bacterial isolates tested, 19 produced AHLs and 15 of these were from BBD. Eight AHLs were identified using LC-MS/MS, with 3OHC4 the most commonly produced, followed by C6. AHL-producing isolates exposed to three temperatures (24°C, 27°C, 30°C) revealed that production of three AHLs (3OHC4, 3OHC5 and 3OHC6) significantly increased at 30°C when compared to 24°C. 16S rRNA gene sequencing revealed that all of the AHL-producing BBD isolates were vibrios. Metagenomic data of BBD communities showed the presence of AHL (and autoinducer-2) genes, many of which are known to be associated with vibrios. These findings suggest that quorum sensing may be involved in BBD pathobiology and community structure due to enhanced production of quorum-sensing signal molecules (AHLs) above the temperature threshold of this globally distributed coral disease. Dark Spot Syndrome (DSS) is one of the most common diseases of boulder corals in the Caribbean. It presents as sunken brown lesions in coral tissue, which can spread quickly over coral colonies. With this study, we tested the hypothesis that similar to other coral diseases, DSS is a dysbiosis characterized by global shifts in the coral microbiome. Because Black Band Disease (BBD) was sometimes found following DSS lesions, we also tested the hypothesis that DSS is a precursor of BBD. To track disease initiation and progression 24 coral colonies were tagged. Of them five Orbicella annularis corals and three O. faveolata corals exhibited DSS lesions at tagging. Microbiota of lesions and apparently healthy tissues from DSS-affected corals over the course of 18 months were collected. Final visual assessment showed that five of eight corals incurred substantial tissue loss while two corals remained stable and one appeared to recover from DSS lesions. Illumina sequencing of the V6 region of bacterial 16S rRNA genes demonstrated no significant differences in bacterial community composition associated with healthy tissue or DSS lesions. The epimicrobiomes of both healthy tissue and DSS lesions contained high relative abundances of Operational Taxonomic Units (OTUs) assigned to Halomonas, an unclassified gammaproteobacterial genus, Moritella, an unclassified Rhodobacteraceae genus, Renibacterium, Pseudomonas, and Acinetobacter. The relative abundance of bacterial taxa was not significantly different between samples when grouped by tissue type (healthy tissue vs. DSS lesion), coral species, collection month, or the overall outcome of DSS-affected corals (substantial tissue loss vs. stable/recovered). Two of the tagged corals with substantial tissue loss also developed BBD during the 18-month sampling period. The bacterial community of the BBD layer was distinct from both healthy tissue and DSS lesions, with high relative abundances of the presumed BBD pathogen Roseofilum reptotaenium and an unclassified Bacteroidales genus, similar to previous results. Roseofilum was detected in all samples from this study, with the highest relative abundance in healthy tissue from DSS-affected corals sampled in August, suggesting that while DSS is not a precursor to BBD, DSS-affected corals are in a weakened state and therefore more susceptible to additional infections.

In many diverse ecosystems, ranging from natural surfaces in aquatic ecosystems to the mammalian gut and medical implants, bacterial populations and communities exist as biofilms. While the process of biofilm development has been well-studied for those produced by unicellular bacteria such Pseudomonas aeruginosa, little is known about biofilm development associated with filamentous microorganisms. Black band disease (BBD) of corals is characterized as a polymicrobial biofilm (mat) community, visually-dominated by filamentous cyanobacteria. The mat migrates across a living coral host, completely lysing coral tissue and leaving behind exposed coral skeleton. It is the only known cyanobacterial biofilm that migrates across a substratum, thus eliciting questions about the mechanisms and unique characteristics of this system. Fragments of the coral Montastraea annularis, five artificially infected with BBD and two collected from a naturally BBD-infected colony, were used to address these questions by detailed examination using scanning and transmission electron microscopy (SEM and TEM). In areas close to the interface of coral tissue and the mature disease band two types of clusters of cyanobacteria were observed, one with random orientation and one with parallel orientation of filaments. The latter exhibited active secretion of extracellular polysaccharide (EPS) while the randomly oriented clusters did not. Within the well developed band cyanobacterial filaments were observed to be embedded in EPS and were present as layers of filaments in parallel orientation. These observations suggest that BBD cyanobacteria orient themselves and produce EPS in a sequential process during migration to form the complex BBD matrix.En muchos ecosistemas diversos, que van desde ecosistemas acuáticos hasta los intestinos de mamíferos e implantes médicos, las poblaciones y comunidades de bacterias existen como biopelículas (biofilms). El proceso de desarrollo de las biopelículas ha sido bien estudiado para aquellos producidos por bacterias unicelulares como Pseudomonas aeruginosa, pero se conoce muy poco acerca del desarrollo de biopelículas asociadas con microorganismos filamentosos. La Enfermedad de Banda Negra (EBN) de coral es caracterizada como una comunidad polimicrobiana que forma una biopelícula (lecho), visualmente-dominada por una cianobacteria filamentosa. El lecho migra a través de un huésped de coral vivo, rompiendo completamente el tejido del coral y dejando atrás el esqueleto de coral expuesto. Es la única biopelícula cianobacteriana que migra a través de un sustrato, por lo tanto esto genera preguntas acerca de los mecanismos y las características únicas de este sistema. Fragmentos del coral Montastraea annularis, cinco artificialmente infectados con EBN y dos colectados de una colonia EBN-infectada, fueron usados para abordar estas preguntas mediante exámenes detallados con microscopía electrónica de barrido y de transmisión (MEB y MET). En zonas cercanas a la interfaz de tejido del coral y la banda de la enfermedad madura, se han observado dos tipos de grupos de cianobacterias, uno con orientación aleatoria y otro con una orientación paralela de los filamentos. Este último exhibe la secreción activa de polisacáridos extra-celulares (PEC), mientras que los grupos orientados al azar no lo hicieron. Dentro de la banda de filamentos cianobacterianas bien desarrollados se observó que estaban integradas en PEC y que se presentaban como capas de cianobacteria con orientación paralela. Estas observaciones sugieren que la cianobacteria de EBN se orienta a sí misma y produce PEC en un proceso secuencial durante la migración para formar la matriz complejo de EBN.

The study of coral diseases, coral pathogens, and the effects of diseases on tropical and subtropical coral reefs are all current, high-profile research areas. This interest has grown steadily since the first report of a coral disease in 1973. The author of this report was Arnfried Antonius and the publication was an abstract in the proceedings of a scientific meeting of the Association of Marine Laboratories of the Caribbean, or AMLC (then known as the Association of Island Marine Laboratories of the Caribbean). Since Antonius’ pioneering communication he continued working on coral diseases on reefs throughout the world, often documenting the first observation of a novel pathology in a novel location. Each of the coral diseases Antonius first described, in particular black band disease, is the subject of current and ongoing investigations addressing pathogens, etiology, and their effects on coral reefs. Many of the points and observations he made in his early papers are highly relevant to research today. This paper reviews aspects of Antonius’ early work, highlighting contributions he made that include the first in situ experimental studies aimed at discerning coral epizootiology and the first quantitative assessments of the role of environmental factors in coral disease. We announce the draft genome sequence of three Gram-negative bacteria isolated from coral tissues affected with the black band disease (BBD), identified with the NCBI's Assembly Database accession numbers: MBQF, MAYB and MBQE. These genome drafts constitute an useful tool for the characterisation of these bacteria and for the understanding of the relationship between the microbial consortia associated with the disease and the onset and progression of the pathology.

Infectious diseases such as white plague syndrome ( WPS) and black band disease ( BBD) have caused massive coral loss worldwide. We performed a metaproteomic study on the Abrolhos coral Mussismilia braziliensis to define the types of proteins expressed in healthy corals compared to WPS- and BBD-affected corals. A total of 6363 MS/ MS spectra were identified as 361 different proteins. Healthy corals had a set of proteins that may be considered markers of holobiont homoeostasis, including tubulin, histone, Rab family, ribosomal, peridinin-chlorophyll a-binding protein, F0F1-type ATP synthase, alpha- iG protein, calmodulin and ADP-ribosylation factor. Cnidaria proteins found in healthy M. braziliensis were associated with Cnidaria- Symbiodinium endosymbiosis and included chaperones (hsp70, hsp90 and calreticulin), structural and membrane modelling proteins (actin) and proteins with functions related to intracellular vesicular traffic (Rab7 and ADP-ribosylation factor 1) and signal transduction (14-3-3 protein and calmodulin). WPS resulted in a clear shift in the predominance of proteins, from those related to aerobic nitrogen-fixing bacteria (i.e. Rhizobiales, Sphingomonadales and Actinomycetales) in healthy corals to those produced by facultative/anaerobic sulphate-reducing bacteria (i.e. Enterobacteriales, Alteromonadales, Clostridiales and Bacteroidetes) in WPS corals. BBD corals developed a diverse community dominated by cyanobacteria and sulphur cycle bacteria. Hsp60, hsp90 and adenosylhomocysteinase proteins were produced mainly by cyanobacteria in BBD corals, which is consistent with elevated oxidative stress in hydrogen sulphide- and cyanotoxin-rich environments. This study demonstrates the usefulness of metaproteomics for gaining better comprehension of coral metabolic status in health and disease, especially in reef systems such as the Abrolhos that are suffering from the increase in global and local threatening events. Black band disease (BBD) is the oldest recognised disease associated with scleractinian corals. However, despite this, few BBD surveys have been conducted in the Indonesian archipelago, one of the world's hot spots for coral diversity. In this study, we show that BBD was recorded in the reefs of Kepulauan Seribu, Indonesia, at the time of surveying. The disease was found to mainly infect corals of the genus Montipora. In some instances, upwards of 177 colonies (31.64%) were found to be infected at specific sites. Prevalence of the disease ranged from 0.31% to 31.64% of Montipora sp. colonies throughout the archipelago. Although BBD was found at all sites, lower frequencies were associated with sites closest to the mainland (17.99 km), as well as those that were furthest away (63.65 km). Despite there being no linear relationship between distance from major population centers and BBD incidence, high incidences of this disease were associated with sites characterized by higher levels of light intensity. Furthermore, surveys revealed that outbreaks peaked during the transitional period between the dry and rainy seasons. Therefore, we suggest that future surveys for disease prevalence in this region of Indonesia should focus on these transitory periods. [ABSTRACT FROM AUTHOR]

Coral diseases have been little investigated in the Indian Ocean and especially in the Republic of Maldives, where they were firstly observed in 2010. Through repeated monitoring, we observed a peculiar slow progression of Black Band Disease in several large colonies of the reef-building coral Goniopora cf. columna in the Maldivian reefs of Magoodhoo Island (Faafu Atoll). We argue that this may ensure the local persistence of the disease, possibly promoting long-term mortality of other, more susceptible coral species. [ABSTRACT FROM AUTHOR]

In the last few decades there has been a surge in research focusing on coral disease. While climate change, specifically rising sea surface temperature, has been proposed as a major and growing driver of the emergence of marine diseases, to date a solid connection between disease epizootics and elevated sea surface temperature has not been established. However, a wealth of data now exists, compiled from many different perspectives, that may support such a connection. In this work we provide a comprehensive review targeting one coral disease, black band disease, that spans the infection process, pathobiology, and epizootiology, and links specific mechanisms of the disease process to increasing temperatures. This temperature-driven pattern of infection can be expanded to include similar processes associated with other temperature-related coral diseases. The conclusions presented here are based upon the results of many studies using a diverse suite of approaches that have been synthesized to argue that the emergence and continuing spread of black band disease is linked to warming sea surface temperatures. In summary, as global ocean temperatures increase seasonally and over decades, the environment shifts to become more favorable for the growth of potentially pathogenic microorganisms endemic to the immediate environment of the reef. The increase in the relative number of potential pathogens in the microbial community produces microenvironments conducive to the growth of other potential pathogens, leading to infection by a polymicrobial consortium. This consortium is easily perturbed by a (seasonal) temperature decrease, but remains associated with the coral host and can be reactivated with a subsequent seasonal increase in temperature, resulting in a cycle of temperature-dependent disease emergence. The prevalence of coral disease was previously unreported on the reefs of the Padang Shelf Reef System, West Sumatra and is relatively uncommon. The objective of this study was to get coral disease prevalence of each site which located at inshore, mid-shelf reef and off-shore reefs. The research was carried out by using belt transect method with 1 m left and right of tape as long 30 m with 3 replications. Two kinds of coral disease and two indicators of stressed coral were observed. While relatively infrequent, Black Band Disease was most common (1.3%) occurring on Pandan Island at 5m. White Syndrome occurred at Air patch reef (0.3%). A few bleached colonies of Montipora sp. and Pocillopora verrucosa were observed on Pieh Island reef (2.1%). Focal bleaching was observed on Galaxea sp. and Goniastrea sp. at Gabuo patch reefs (2.5%) and on Pocillopora verrucosa at Sipakal patch reef (3.9%). BBD infected both Montipora sp. and Porites sp. WS only infected on Montipora sp. Montipora, Pocillopora and Porites were the most common genera observed on the reefs. It suggests that coral disease infection was classified as a minor and it will not hamper coral recovery processes and coral health status in Padang Waters, West Sumatera. [ABSTRACT FROM AUTHOR]

Roseofilum reptotaenium is a gliding, filamentous, phycoerythrin-rich cyanobacterium that has been found only in the horizontally migrating, pathogenic microbial mat, black band disease (BBD) on Caribbean corals. R. reptotaenium dominates the BBD mat in terms of biomass and motility, and the filaments form the mat fabric. This cyanobacterium produces the cyanotoxin microcystin, predominately MC-LR, and can tolerate high levels of sulfide produced by sulfate reducing bacteria (SRB) that are also associated with BBD. Laboratory cultures of R. reptotaenium infect coral fragments, suggesting that the cyanobacterium is the primary pathogen of BBD, but since this species cannot grow axenically and Koch's Postulates cannot be fulfilled, it cannot be proposed as a primary pathogen. However, R. reptotaenium does play several major pathogenic roles in this polymicrobial disease. Here, we provide an overview of the ecology of this coral pathogen and present new information on R. reptotaenium ecophysiology, including roles in the infection process, chemotactic and other motility responses, and the effect of pH on growth and motility. Additionally, we show, using metabolomics, that exposure of the BBD microbial community to the cyanotoxin MC-LR affects community metabolite profiles, in particular those associated with nucleic acid biosynthesis. Black band disease (BBD) is the oldest recognised disease associated with scleractinian corals. However, despite this, few BBD surveys have been conducted in the Indonesian archipelago, one of the world’s hot spots for coral diversity. In this study, we show that BBD was recorded in the reefs of Kepulauan Seribu, Indonesia, at the time of surveying. The disease was found to mainly infect corals of the genus Montipora. In some instances, upwards of 177 colonies (31.64%) were found to be infected at specific sites. Prevalence of the disease ranged from 0.31% to 31.64% of Montipora sp. colonies throughout the archipelago. Although BBD was found at all sites, lower frequencies were associated with sites closest to the mainland (17.99 km), as well as those that were furthest away (63.65 km). Despite there being no linear relationship between distance from major population centers and BBD incidence, high incidences of this disease were associated with sites characterized by higher levels of light intensity. Furthermore, surveys revealed that outbreaks peaked during the transitional period between the dry and rainy seasons. Therefore, we suggest that future surveys for disease prevalence in this region of Indonesia should focus on these transitory periods.
