10. Medical Mycology and the Chemistry Classroom: Germinating Student Interest in Organic Chemistry

Abstract

Efforts to provide active research context to introductory courses in basic sciences are likely to better engage learners and provide a framework for relevant concepts. A simple teaching and learning experiment was conducted to use concepts in organic chemistry to solve problems in the life sciences. Bryant University is a liberal arts university with new programs in biological and environmental sciences. This laboratory exercise was developed for introductory organic chemistry and designed to highlight its applicability to the biological sciences. Students participated in an ongoing project to identify antigens on the surface of "Candida albicans" hyphae by testing a variety of lipid extraction procedures and characterizing the fatty acid and carbohydrate composition of their extractions by thin-layer chromatography and gas chromatography-mass spectrometry. Student extracts were also tested in an immunoblot assay with a specific antibody fragment (scFv) that recognizes an unknown component of the cell surface of "C. albicans" hyphae. In order to build communication skills, students were required to prepare and present posters detailing the project and their findings.

18. Bi-fluorescent Staphylococcus aureus infection enables singlecell analysis of intracellular killing in vivo.

Abstract

Techniques for studying the clearance of bacterial infections are critical for advances in understanding disease states, immune cell effector functions, and novel antimicrobial therapeutics. Intracellular killing of Staphylococcus aureus by neutrophils can be monitored using a S. aureus strain stably expressing GFP, a fluorophore that is quenched when exposed to the reactive oxygen species (ROS) present in the phagolysosome. Here, we expand upon this method by developing a bi-fluorescent S. aureus killing assay for use in vivo. Conjugating S. aureus with a stable secondary fluorescent marker enables the separation of infected cell samples into three populations: cells that have not engaged in phagocytosis, cells that have engulfed and killed S. aureus, and cells that have viable internalized S. aureus. We identified ATTO647N-NHS Ester as a favorable dye conjugate for generating bi-fluorescent S. aureus due to its stability over time and invariant signal within the neutrophil phagolysosome. To resolve the in vivo utility of ATTO647N/ GFP bi-fluorescent S. aureus, we evaluated neutrophil function in a murine model of chronic granulomatous disease (CGD) known to have impaired clearance of S. aureus infection. Analysis of bronchoalveolar lavage (BAL) from animals subjected to pulmonary infection with bi-fluorescent S. aureus demonstrated differences in neutrophil antimicrobial function consistent with the established phenotype of CGD. Abstract

The biochemical function of KpsT adenosine triphosphate (ATP)-binding component in the translocation of the polysialic acid capsule of Escherichia coli was analyzed via selective mutagenesis. Glutamic acid-to-glycine substitution of the KpsT protein impaired the hydrolysis of ATP due to alterations in the protein's nucleotide binding domain. Furthermore, additional mutations in the kpsT gene induced the intragenic suppression of the dominant negative phenotype of the KpsT protein.

Abstract

Analysis of the mutants in Kps gene cluster of E. coli K1 reveals that transport defects affect synthetic activities, as seen by the expression of polysialic acid (PSA), due to the accumulation of protein K in the outer membrane. Kps gene products have the ability to transport a range of acidic polysaccharides due to protein-polysaccharide interaction. KpsD is the periplasmic binding component of the PSA transport system that transport the polymer to the outer membrane.

48. Pathology of Neonatal Non-albicansCandidiasis: Autopsy Study and Literature Review

Abstract

Introduction/Objectives Non-albicans Candidaspecies such as Candida parapsilosisand Candida glabratahave emerged as prevalent pathogens in premature infants. The aim of this study was to systematically delineate the histopathologic findings in neonatal non-albicanscandidiasis.Methods We performed a retrospective clinicopathologic analysis of extremely premature (23–28 weeks’ gestation) infants diagnosed with invasive candidiasis. Archival autopsy tissues were subjected to periodic acid-Schiff, methenamine-silver and anti-Candida(immuno)histochemical stains, as well as dual anti-Candidaand anti-cytokeratin or anti-CD31 immunofluorescence assays. In addition, we studied the prevalence of intestinal Candidacolonization in a consecutive autopsy series of extremely premature infants.Results Based on positive postmortem blood and/or lung cultures, invasive candidiasis (3 non-albicansand 11 Candida albicans) was diagnosed in 14 of the 187 extremely premature infants examined between 1995 and 2017. In contrast to the well-known inflammatory and tissue-destructive phenotype of congenital C. albicansinfection, invasive non-albicanscandidiasis/candidemia caused by C. parapsilosisand C. glabratawas inconspicuous by routine hematoxylin-eosin-based histopathologic analysis despite a heavy fungal presence detected in intestines, lungs, and blood by targeted (immuno)histochemical assays. Intestinal colonization by Candidaspecies was identified in 16 of the 26 (61%) extremely premature neonates who had lived for at least 1 week, as assessed by anti-Candidaimmunostaining.Conclusion Invasive neonatal non-albicanscandidiasis/candidemia appears to have no distinct histopathologic signature. Based on the notoriously low sensitivity of fungal blood cultures and the observed high frequency of Candidaintestinal colonization (>50%), it is likely that non-albicanscandidiasis/candidemia may be underdiagnosed in (deceased) preterm infants. Routine inclusion of targeted (immuno)histochemical fungal detection strategies in the perinatal autopsy may lead to deeper insight into the prevalence and clinical relevance of neonatal non-albicanscandidiasis.
