Abstract Basal progenitor cells are crucial for maintaining foregut (the esophagus and forestomach) homeostasis. When their function is dysregulated, it can promote inflammation and tumorigenesis. However, the mechanisms underlying these processes remain largely unclear. Here, we employ genetic mouse models to reveal that Jag1/2 regulate esophageal homeostasis and foregut tumorigenesis by modulating the function of basal progenitor cells. Deletion of Jag1/2 in mice disrupts esophageal and forestomach epithelial homeostasis. Mechanistically, Jag1/2 deficiency impairs activation of Notch signaling, leading to reduced squamous epithelial differentiation and expansion of basal progenitor cells. Moreover, Jag1/2 deficiency exacerbates the deoxycholic acid (DCA)-induced squamous epithelial injury and accelerates the initiation of squamous cell carcinoma (SCC) in the forestomach. Importantly, expression levels of JAG1/2 are lower in the early stages of human esophageal squamous cell carcinoma (ESCC) carcinogenesis. Collectively, our study demonstrates that Jag1/2 are important for maintaining esophageal and forestomach homeostasis and the onset of foregut SCC.

Abstract Near-infrared luminophores have many advantages in advanced applications, especially for structures without π-conjugation aromatic rings. However, the fabrication of red clusteroluminogens from nonconjugated polymers is still a big challenge, let alone the near-infrared clusteroluminogens. Here, we develop nonconjugated luminophores with full-spectrum from blue to near-infrared light (470 ~ 780 nm), based on color phenomenon of nonconjugated polyesters synthesized from the amine-initiated copolymerization of epoxides and cyclic anhydrides. We reveal that amines act as initiators attached to polymer chain ends. The formation of various amine-ester complexes in polyesters induces red to near-infrared light, conceptually, amine-ester complexed clusteroluminescence via intra/inter-chain charge transfer. Significantly, emission colors can be easily tuned by the contents and types of amines, microstructures of polyesters, and their concentration. This work provides a low-cost, scalable platform and strategy for the production of high-efficiency, multicolor luminescent materials.

Abstract Recent findings have shown that fatty acid metabolism is profoundly involved in ferroptosis. However, the role of cholesterol in this process remains incompletely understood. In this work, we show that modulating cholesterol levels changes vulnerability of cells to ferroptosis. Cholesterol alters metabolic flux of the mevalonate pathway by promoting Squalene Epoxidase (SQLE) degradation, a rate limiting step in cholesterol biosynthesis, thereby increasing both CoQ10 and squalene levels. Importantly, whereas inactivation of Farnesyl-Diphosphate Farnesyltransferase 1 (FDFT1), the branch point of cholesterol biosynthesis pathway, exhibits minimal effect on ferroptosis, simultaneous inhibition of both CoQ10 and squalene biosynthesis completely abrogates the effect of cholesterol. Mouse models of ischemia-reperfusion and doxorubicin induced hepatoxicity confirm the protective role of cholesterol in ferroptosis. Our study elucidates a potential role of ferroptosis in diseases related to dysregulation of cholesterol metabolism and suggests a possible therapeutic target that involves ferroptotic cell death.

As a migratory invasive pest, Spodoptera frugiperda (fall armyworm, FAW) has recently posed a serious threat to food security in newly invaded areas (especially in Africa and Asia). Understanding its migration (or dispersal) patterns in newly invaded areas is crucial for regional forecasting and management efforts. By screening an appropriate marking technique to conduct mark–release–recapture (MRR) experiments, the migration patterns of the FAW can be effectively studied. In this study, we added different concentrations of Calco Oil Red N-1700 (an oil-soluble marker) to a self-made artificial diet and assessed the rearing and marking efficacy. The results indicated that a concentration of 0.2% of Calco Oil Red N-1700 in the diet was optimal for marking adult FAWs. The biological indicators (e.g., developmental duration, reproductive parameters, and flight ability) of FAWs fed this diet were basically consistent with those of FAWs fed a normal diet, with a larval stage of 15.46 days, a pupal stage of 9.81 days, a pupal mass of 278.18 mg, an adult longevity of 15.41 days, and an egg deposition count of 1503.51. Meanwhile, the flight distance, duration, and velocity were 24.91 km, 7.16 h, and 3.40 km/h, respectively (12 h tethered-flight tests), without difference with the control. Females and males exhibited distinctive marking colors (red or pink) that persisted for at least 5 and 9 days, respectively. This study developed an economically effective internal marking method for the adult FAW, laying the foundation for conducting MRR experiments. This will help clarify the migration behavior and routes of the FAW, providing a scientific basis for formulating effective pest management strategies.

Helicoverpa armigera (Hübner) is a destructive agricultural pest. Facultative migration usually causes regional catastrophes; therefore, developing a simple and easy new technology for the monitoring and early warning of immigrant populations is urgent. Between 2021 and 2023, we conducted a population-monitoring study on H. armigera immigrants in Xundian County, Yunnan Province, where the migration pathway for pests from southeast Asia extends to southwest China. Based on the differences in the reproductive organ development parameters of H. armigera at different ages, we established an adult age discrimination model. The monitoring results of field populations with semiochemical attractants and sex pheromones between 2021 and 2023 showed that the daily average age of the adult population of H. armigera fluctuated above 6 days, and the trapping dynamics with semiochemical attractants and sex pheromones were the same. Both trapping methods yielded H. armigera males of different ages and had identical age structures, indicating that the H. armigera population primarily migrated from other regions. The trajectory analysis showed that the H. armigera population that migrated to Xundian between May and September was primarily from South Kunming, and after October, it was primarily the southward-migrating population north of Qujing City, Yunnan Province. Based on the relationship between the daily average fecundity and the age of H. armigera, a dynamic prediction model for the fecundity of the immigrant population was established. In this study, the prediction models and methods based on semiochemical attractants could potentially be used in the surveillance and population alerting of H. armigera.

Abstract The uterine epithelium undergoes a dramatic spatiotemporal transformation to enter a receptive state, involving a complex interaction between ovarian hormones and signals from stromal and epithelial cells. Redox homeostasis is critical for cellular physiological steady state; emerging evidence reveals that excessive lipid peroxides derail redox homeostasis, causing various diseases. However, the role of redox homeostasis in early pregnancy remains largely unknown. It is found that uterine deletion of Glutathione peroxidase 4 (GPX4), a key factor in repairing oxidative damage to lipids, confers defective implantation, leading to infertility. To further pinpoint Gpx4’s role in different cell types, uterine epithelial‐specific Gpx4 is deleted by a lactotransferrin (Ltf)‐Cre driver; the resultant females are infertile, suggesting increased lipid peroxidation levels in uterine epithelium compromises receptivity and implantation. Lipid peroxidation inhibitor administration failed to rescue implantation due to carbonylation of major receptive‐related proteins underlying high lipid reactive oxygen species. Intriguingly, superimposition of Acyl‐CoA synthetase long‐chain family member 4 (ACSL4), an enzyme that promotes biosynthesis of phospholipid hydroperoxides, along with uterine epithelial GPX4 deletion, preserves reproductive capacity. This study reveals the pernicious impact of unbalanced redox signaling on embryo implantation and suggests the obliteration of lipid peroxides as a possible therapeutic approach to prevent implantation defects.

Abstract Ferroptosis suppressor protein 1 (FSP1, also known as AIMF2, AMID or PRG3) is a recently identified glutathione-independent ferroptosis suppressor1–3, but its underlying structural mechanism remains unknown. Here we report the crystal structures of Gallus gallus FSP1 in its substrate-free and ubiquinone-bound forms. The structures reveal a FAD-binding domain and a NAD(P)H-binding domain, both of which are shared with AIF and NADH oxidoreductases4–9, and a characteristic carboxy-terminal domain as well. We demonstrate that the carboxy-terminal domain is crucial for the catalytic activity and ferroptosis inhibition of FSP1 by mediating the functional dimerization of FSP1, and the formation of two active sites located on two sides of FAD, which are responsible for ubiquinone reduction and a unique FAD hydroxylation respectively. We also identify that FSP1 can catalyze the production of H2O2 and the conversion of FAD to 6-hydroxy-FAD in the presence of oxygen and NAD(P)H in vitro, and 6-hydroxy-FAD directly inhibits ferroptosis in cells. Together, these findings further our understanding on the catalytic and ferroptosis suppression mechanisms of FSP1 and establish 6-hydroxy-FAD as an active cofactor in FSP1 and a potent radical-trapping antioxidant in ferroptosis inhibition.

Spodoptera frugiperda (Lepidoptera: Noctuidae) is an important migratory agricultural pest worldwide, which has invaded many countries in the Old World since 2016 and now poses a serious threat to world food security. The present monitoring and early warning strategies for the fall army worm (FAW) mainly focus on adult population density, but lack an information technology platform for precisely forecasting the reproductive dynamics of the adults. In this study, to identify the developmental status of the adults, we first utilized female ovarian images to extract and screen five features combined with the support vector machine (SVM) classifier and employed male testes images to obtain the testis circular features. Then, we established models for the relationship between oviposition dynamics and the developmental time of adult reproductive organs using laboratory tests. The results show that the accuracy of female ovary development stage determination reached 91%. The mean standard error (MSE) between the actual and predicted values of the ovarian developmental time was 0.2431, and the mean error rate between the actual and predicted values of the daily oviposition quantity was 12.38%. The error rate for the recognition of testis diameter was 3.25%, and the predicted and actual values of the testis developmental time in males had an MSE of 0.7734. A WeChat applet for identifying the reproductive developmental state and predicting reproduction of S. frugiperda was developed by integrating the above research results, and it is now available for use by anyone involved in plant protection. This study developed an automated method for accurately forecasting the reproductive dynamics of S. frugiperda populations, which can be helpful for the construction of a population monitoring and early warning system for use by both professional experts and local people at the county level.

AbstractDiabetes Mellitus (DM) is a chronic disease characterized by abnormally uncontrolled high blood glucose level. The Risk Assessment and Management Program (RAMP) in Hong Kong has been providing long-term face-to-face follow-up to DM patients in the government out-patient clinics since 2009. However, under the current outbreak of COVID-19, these face-to-face consultations were ceased over and over again to lower the risk of disease transmission. With the advancement in technology, the recent emergence of telecare has provided an alternative to replace the conventional consultations in the clinics. Its clinical effectiveness on DM patients has also been supported by numerous studies. Yet, there is only a paucity of literatures discussing the practicality of such implementation design in the real-world settings. This study aims at studying both the effectiveness and implementation outcomes of telecare in Hong Kong DM patients. It adopts a type 2 hybrid effectiveness-implementation design. It will be conducted in seven government out-patient clinics in Hong Kong. The subjects will be randomly assigned to an intervention group or a control group when they 1) are aged 18 or above, 2) have a confirmed diagnosis of diabetes, and 3) are having regular follow-up appointment in the clinic. Subjects in the intervention group will receive a 84-week Risk Assessment and Management Program (RAMP) in an alternate telecare and face-to-face consultations mode, while the control group will receive the same program but in usual face-to-face consultation mode. RE-AIM is employed as the implementation and effectiveness outcome evaluation framework. The primary outcome measure will be HbA1c. Data will be collected pre-intervention (T1), 42-week (T2), and 84-week (T3). The study will provide effectiveness-implementation assessment of telecare mode for DM patients in Hong Kong, as an alternative or in addition to conventional face-to-face consultations. It also aimed to provide insights for the future adoption in a broader health care setting.

Abstract Ferroptosis is an iron-dependent regulated cell death marked by excessive oxidative phospholipids (PLs). The polyunsaturated fatty acids-containing phospholipids (PUFA-PLs) are highly susceptible to lipid peroxidation under oxidative stress. Numerous pulmonary diseases occurrences and degenerative pathologies are driven by ferroptosis. This review discusses the role of ferroptosis in the pathogenesis of pulmonary diseases including asthma, lung injury, lung cancer, fibrotic lung diseases, and pulmonary infection. Additionally, it is proposed that targeting ferroptosis is a potential treatment for pulmonary diseases, particularly drug-resistant lung cancer or antibiotic-resistant pulmonary infection, and reduces treatment-related adverse events.

Disruption of the blood-spinal cord barrier (BSCB) leads to inflammatory cell infiltration and neural cell death, thus, contributing to poor functional recovery after spinal cord injury (SCI). Previous studies have suggested that Sirtuin 1 (SIRT1), an NAD+-dependent class III histone deacetylase, is abundantly expressed in endothelial cells and promotes endothelial homeostasis. However, the role of SIRT1 in BSCB function after SCI remains poorly defined. Here, we report that SIRT1 is highly expressed in spinal cord endothelial cells, and its expression significantly decreases after SCI. Using endothelial cell-specific SIRT1 knockout mice, we observed that endothelial cell-specific knockout of SIRT1 aggravated BSCB disruption, thus, resulting in widespread inflammation, neural cell death and poor functional recovery after SCI. In contrast, activation of SIRT1 by the agonist SRT1720 had beneficial effects. In vitro, knockdown of SIRT1 exacerbated IL-1β-induced endothelial barrier disruption in bEnd.3 cells, whereas overexpression of SIRT1 was protective. Using RNA-seq and IP/MS analysis, we identified p66Shc, a redox protein, as the potential target of SIRT1. Further studies demonstrated that SIRT1 interacts with and deacetylates p66Shc, thereby attenuating oxidative stress and protecting endothelial barrier function. Overall, our results indicate that SIRT1 decreases endothelial ROS production and attenuates BSCB disruption by deacetylating p66Shc after SCI, and suggest that SIRT1 activation has potential as a therapeutic approach to promote functional recovery against BSCB disruption following SCI.

Abstract Emerging evidence reveals that amino acid metabolism plays an important role in ferroptotic cell death. The conversion of methionine to cysteine is well known to protect tumour cells from ferroptosis upon cysteine starvation through transamination. However, whether amino acids‐produced metabolites participate in ferroptosis independent of the cysteine pathway is largely unknown. Here, the authors show that the tryptophan metabolites serotonin (5‐HT) and 3‐hydroxyanthranilic acid (3‐HA) remarkably facilitate tumour cells to escape from ferroptosis distinct from cysteine‐mediated ferroptosis inhibition. Mechanistically, both 5‐HT and 3‐HA act as potent radical trapping antioxidants (RTA) to eliminate lipid peroxidation, thereby inhibiting ferroptotic cell death. Monoamine oxidase A (MAOA) markedly abrogates the protective effect of 5‐HT via degrading 5‐HT. Deficiency of MAOA renders cancer cells resistant to ferroptosis upon 5‐HT treatment. Kynureninase (KYNU), which is essential for 3‐HA production, confers cells resistant to ferroptotic cell death, whereas 3‐hydroxyanthranilate 3,4‐dioxygenase (HAAO) significantly blocks 3‐HA mediated ferroptosis inhibition by consuming 3‐HA. In addition, the expression level of HAAO is positively correlated with lipid peroxidation and clinical outcome. Together, the findings demonstrate that tryptophan metabolism works as a new anti‐ferroptotic pathway to promote tumour growth, and targeting this pathway will be a promising therapeutic approach for cancer treatment.

Abstract Clusteroluminogens (CLgens), nonconjugated structures with visible luminescence at the clustering state, have recently received remarkable attention due to their great theoretical significance and practical values. In carbonyl‐based aliphatic polymers, (n, π*) transition of carbonyl groups and the through‐space interactions have been demonstrated to play an important role in their clusteroluminescence (CL) properties, but it is still a big challenge to manipulate their CL at the molecular level. In this work, six nonconjugated carbonyl‐based polymers with different heteroatoms and steric hindrances were synthesized, and their photophysical properties were systematically studied. These polymers all showed CL but with different emission efficiency and wavelength. Experimental and theoretical studies indicated that the CL properties could be manipulated by changing the electronic structures of carbonyl groups and the rigidity of polymer chains. This work not only gains further insights into the CL mechanism but also provides reliable strategies to design and manipulate non‐conjugated luminescent materials.

Purpose: Spinal cord injury (SCI) has a damaging impact on patients, amid being a worldwide problem with no effective treatment. Herein, we reported a method for functional therapy of SCI in rats, wherein we combined thermo-sensitive hydrogel with Sonic Hedgehog (SHH) expressed in rat bone-marrow derived mesenchymal stem cells (RMSCs).Methods: Bone marrow-derived mesenchymal stem cells (BMSCs) were isolated from Sprague-Dawley (SD) female rats. The SHH was optimized and transferred into RMSCs via cationic liposomes, while thermo-sensitive hydrogel was reformed with hyaluronate (HA) and Pluronic F127. Then, a rat model with SCI was established accordingly by male SD rats and randomized into sham, model, RMSCs with hydrogel and SHH-RMSCs with hydrogel. The evaluation of SCI repair based on Basso, Beattie Bresnahanlocomotor rating scale (BBB scale) and inclined plate score. Immunofluorescence, immunohistochemistry and hematoxylin-eosin were utilized to explore the expression of protein (GFAP, GAP43, NF200 and MBP) and histopathology.Results: It was demonstrated that transfection of SHH with cationic liposomes exhibited more effect in RMSCs than lipofectamine 2000. As shown in SEM, 3.5% HA-F127 demonstrated porous structure. In the MTT and dead/live assay, 3.5% HA-F127 showed good biocompatibility for RMSCs. Both RMSCs and SHH-RMSCs groups could significantly promote BBB and inclined plate scores (p < 0.01) compared with the model. Furthermore, the SHH-RMSC group was significantly improved than RMSC with the expression of related proteins, where NF200, MBP, and GAP43 were principally enhanced with the GFAP expression being virtually down-regulated.Conclusion: All in all, the results suggested that transplantation of RMSCs with SHH could improve the function of SCI and promote nerve regeneration.

The nonunion following a fracture is associated with severe patient morbidity and economic consequences. Currently, accumulating studies are focusing on the importance of macrophages during fracture repair. However, details regarding the process by which macrophages facilitate endochondral ossification (EO) are largely unknown. In this study, we present evidence that apoptotic chondrocytes (ACs) are not inert corpses awaiting removal, but positively modulate the osteoinductive ability of macrophages. In vivo experiments revealed that fatty acid (FA) metabolic processes up-regulated following EO. In vitro studies further uncovered that FAs derived from ACs are taken up by macrophages mainly through macrophage scavenger receptor 1 (MSR1). Then, our functional experiments confirmed that these exogenous FAs subsequently activate peroxisome proliferator-activated receptor α (PPARα), which further facilitates lipid droplets generation and fatty acid oxidation (FAO). Mechanistically, elevated FAO is involved in up-regulating the osteoinductive effect by generating BMP7 and NAD+/SIRT1/EZH2 axis epigenetically controls BMP7 expression in macrophages cultured with ACs culture medium. Our findings advanced the concept that ACs could promote bone regeneration by regulating metabolic and function reprogram in macrophages and identified macrophage MSR1 represents a valuable target for fracture treatments.

p53 is able to induce ferroptosis in response to reactive oxygen species (ROS)-induced stress and suppresses tumour growth. Here, the authors show that iPLA2β suppresses p53-medated ferroptosis by cleaving and detoxifying peroxidized lipids and that this is independent of canonical ferroptosis regulator GPX4.

Spodoptera frugiperda Smith (fall armyworm (FAW)) has invaded many countries in Africa and Asia in recent years, considerably restricting global agricultural production. In this study, we assessed the rearing performance of four artificial diets (D1: an artificial FAW diet based on wheat bran and soybean, maize, and yeast powders; D2: an artificial diet developed for Helicoverpa armigera (Hübner), based on wheat bran and soybean and yeast powders; D3: an artificial diet based on soybean powder; D4: an artificial diet based on wheat bran) for FAWs. We designed D4 based on a traditional diet (D2) but substituted the wheat bran for soybean and yeast powders. At 25 ± 1 °C, 75% ± 5% RH, and a 16:8 h L:D photoperiod, the larval stage of FAWs fed on D4 lasted 15.88 d, the pupal stage lasted 9.48 d, the pupal mass was 270.45 mg, the number of eggs deposited was 1364.78, and the mating rate was 89.53%. Most biological indicators of the larvae that were fed D4 were basically consistent with those of the larvae fed on the traditional diet (D2), but the intrinsic rate of increase (r), finite rate of increase (λ), and net reproduction rate (R0) of the D4 FAWs were lower than those of the D2 FAWs. The flight capacity (flight distance, duration, and velocity were 19.73 km, 6.91 h, and 2.90 km/h, respectively) of the D4 FAWs was comparable to that of the FAWs fed a traditional diet and maize leaves. Compared with the three other formulas, the cost of using D4 was lower by 26.42% on average. These results show that using cheap wheat bran instead of soybean flour and yeast powder as the basic material for an artificial diet for FAWs is feasible, which will substantially reduce rearing costs and promote the development of new controlling measures for FAWs. In addition, this study also has a reference value for reducing the cost of artificial diets for other insects.

Despite the advancements in synthetic methods, it is still challenging to prepare white light‐emitting carbon dots for solid‐state white light‐emitting diodes (LEDs). Herein, the synthesis and characterization of a carbonized polymer dot (CPD) that emits white light in the solid state are presented. The reaction of an oxygen‐rich precursor, trimethylolpropane tri(cyclic carbonate) ether, with nitrogen‐rich melamine in an alcohol via a single‐step solvothermal method affords CPDs with hydroxyurethane and CN groups. The morphology of the CPDs can be regulated by changing the reaction time. Transmission electron microscopy reveals structural evolution from flocculent polymers to dandelion‐like and spherical particles with an increase in the carbonization time from 6 to 12 and 18 h, respectively. The dandelion‐like CPDs exhibit a relatively high quantum yield of 7.5% in the solid state, which is ascribed to the abundant surface poly(hydroxyurethane) chains that restrict the aggregation‐caused quenching of luminescence. It is proposed that multiple coexistent clusters generate different emission sites, thereby leading to white light emission. Loose clusters formed from hydroxyurethane and CN bonds, which have a low degree of conjugation, emit blue light, whereas compact clusters generated through interactions between the hydroxyurethane and CN bonds of poly(hydroxyurethane) and carbon cores emit yellow light.

Insects’ olfactory receptor plays a central role in detecting chemosensory information from the environment. Odorant receptors (ORs) and ionotropic receptors (IRs) are two types of olfactory receptors, and they are essential for the recognition of ligands at peripheral neurons. Apriona germari (Hope) (Coleoptera: Cerambycidae) is one of the most serious insect pests that cause damage to economic trees and landscaping trees, resulting in massive environmental damages and economic losses. Olfactory-based management strategy has been suggested as a promising strategy to control this wood-boring beetle. However, the olfactory perception mechanism in A. germari is now almost unknown. In the present study, RNA sequencing analysis was used to determine the transcriptomes of adult A. germari antennae. Among 36,834 unigenes derived from the antennal assembly, we identified 42 AgerORs and three AgerIRs. Based on the tissue expression pattern analysis, 27 AgerORs displayed a female-biased expression. Notably, AgerOR3, 5, 13, 33, and 40 showed a significant female-biased expression and were clustered with the pheromone receptors of Megacyllene caryae in the phylogenetic tree, suggesting that these AgerORs could be potential pheromone receptors for sensing male-produced sex pheromones in A. germari. The AgerIRs expression profile demonstrated that AgerIR2 had high expression levels in male labial palps, suggesting that this receptor may function to detect female-deposited trail-sex pheromone blend of A. germari. In addition, the phylogenetic tree showed that the Orco gene of five cerambycidae species was highly conservative. These results provide a foundation for further studies on the molecular mechanisms of olfactory chemoreception in A. germari apart from suggesting novel targets for the control of this pest in the future.

The sibling species Ectropis grisescens and E. obliqua are the major chewing tea pests in China. A difference in sex pheromone components plays a central role in premating isolation in these two species. To investigate the mechanism of premating isolation in these two Ectropis species, we sequenced the transcriptomes of the antennae of female and male E. obliqua individuals and performed phylogenetic analyses, abundance analyses, and tissue expression profile analyses to compare the olfactory genes involved in the detection of sex pheromones. A total of 36 odorant-binding proteins (OBPs) and 52 olfactory receptors (ORs) were identified in E. obliqua. Phylogenetic analyses showed that EoblOBP2, 3, and 25 were grouped in the pheromone-binding protein clade with EgriOBP2, 3, 25, and another lepidopteran PBP. EoblOR25 and 28 were grouped with EgriOR25, 28, and pheromone receptors for the detection of Type-I sex pheromone components. EoblOR24, 31, 37, and 44 were grouped with EgriOR24, 31, 37, and 44. All of these 4 EoblORs and 4 EgriORs showed higher abundance in male antennae than in female ones. Therefore, OBP2, 3, 25 and OR24, 31, 37, 44 of E. grisescens and E. obliqua might be responsible for sex pheromone component detection. However, the sequences of these genes in E. grisescens and E. obliqua were more than 90% identical. This indicates that these orthologous genes might play similar roles in the detection of sex pheromones. In contrast, the observed OBPs and ORs differed in abundance between the antennae of the two Ectropis species. Therefore, we speculate that these two Ectropis species use the different transcript levels of PRs to differentiate sex pheromone components. The results of the present study might contribute in deciphering the mechanism for premating isolation in these species and may be of use in devising strategies for their management.

Tea grey geometrid (Ectropis grisescens), a devastating chewing pest in tea plantations throughout China, produces Type-II pheromone components. Little is known about the genes encoding proteins involved in the perception of Type-II sex pheromone components. To investigate the olfaction genes involved in E. grisescens sex pheromones and plant volatiles perception, we sequenced female and male antennae transcriptomes of E. grisescens. After assembly and annotation, we identified 153 candidate chemoreception genes in E. grisescens, including 40 odorant-binding proteins (OBPs), 30 chemosensory proteins (CSPs), 59 odorant receptors (ORs), and 24 ionotropic receptors (IRs). The results of phylogenetic, qPCR, and mRNA abundance analyses suggested that three candidate pheromone-binding proteins (EgriOBP2, 3, and 25), two candidate general odorant-binding proteins (EgriOBP1 and 29), six pheromone receptors (EgriOR24, 25, 28, 31, 37, and 44), and EgriCSP8 may be involved in the detection of Type-II sex pheromone components. Functional investigation by heterologous expression in Xenopus oocytes revealed that EgriOR31 was robustly tuned to the E. grisescens sex pheromone component (Z,Z,Z)-3,6,9-octadecatriene and weakly to the other sex pheromone component (Z,Z)-3,9-6,7-epoxyoctadecadiene. Our results represent a systematic functional analysis of the molecular mechanism of olfaction perception in E. grisescens with an emphasis on gene encoding proteins involved in perception of Type-II sex pheromones, and provide information that will be relevant to other Lepidoptera species.

In the process of oil exploitation, the water level of an oil well can be predicted and the position of reservoir can be estimated by measuring the water content of crude oil, with reference for the automatic production of high efficiency in the oil field. In this paper, a visual measuring method for water content of crude oil is proposed. The oil and water in crude oil samples were separated by centrifugation, distillation or electric dehydration, and a water−oil layered mixture was formed according to the unequal densities. Then the volume ratio of water and oil was analyzed by digital image processing, and the water content and oil content was able to be calculated. A new method for measuring water content of crude oil based on IGAVD (image grayscale accumulated value difference) is proposed, which overcomes the uncertainty caused by environmental illumination and improves the measurement accuracy. In order to verify the effectiveness of the algorithm, a miniaturization and low-cost system prototype was developed. The experimental results show that the average power consumption is about 165 mW and the measuring error is less than 1.0%. At the same time, the real-time and remote transmission about measurement results can be realized.

Anthocyanins from highbush blueberry (Vaccinium corymbosum L.) have tremendous potential as natural colorants and functional food with pharmaceutical purposes in food applications. To exploit the potential for food applications, the stability and antioxidant activity of anthocyanins present in blueberries have been studied. The results indicate that anthocyanins from blueberry were stable against the low pH (≤5.0), NaCl (0.125–0.500 mol/L), sucrose (0.584–2.336 mol/L) and preservative (sodium benzoate, 0.035–0.140 mol/L), but were sensitive to alkaline conditions (≥7.0), high temperature (≥80 °C), light (natural light), oxidizing agent (H2O2, 0.5–2.0 %) and reducing agent (Na2SO3, 0.005–0.040 mol/L). At concentrations of 25 and 50 mg/mL, anthocyanins from blueberry could protect ECV-304 cells against oxidative damage induced by H2O2. These results suggest that anthocyanins from blueberry can be regarded as a potential colorant for some acidic (pH≤5.0) food products and could be used as health food to prevent diseases arising from oxidative processes.

Macrophorins H (4) and L (5), two rare HMG-conjugate macrophorins along with three known macrophorins (1-3), three DMOA-derived meroterpenoids (6-8) and two ergosterol derivates (9-10) were isolated from sterilized rice medium cultured Penicillium sp. NX-05-G-3. Their structures were elucidated by 1D and 2D NMR. The cytotoxicities of all compounds were evaluated, and compounds 1 and 2 showed extensive cytotoxicity against human cancer cell lines Hela, SCC15, MDA-MB-453 and A549, with IC50 values ranging from 17.6 to 32.8 µM. A facile and efficient visible-light-mediated method for directly converting 1,4-naphthoquinones into dihydrocyclo-buta[b]naphthalene-3,8-diones (DHCBNDOs) under mild and clean conditions without using any photocatalysts is reported. This approach exhibited favorable compatibility with functional groups and afforded a series of DHCBNDOs with excellent regioselectivity and high yields. Moreover, detailed mechanism studies were carried out both experimentally and theoretically.
