Aflatoxin B1 (AFB1) is the most harmful mycotoxin. Aflatoxin occurrence in tea makes this beverage unsuitable for consumption and presented risks to human health. Therefore, researches in aflatoxin microbial degradation are necessary to overcome this problem. Kombucha beverage is associated with health promoting effects. Thus, novel strains (Lactic acid bacteria and yeasts) were isolated from a Kombucha culture and assessed for AFB1 degradation in the liquid medium (Man Rogosa and Sharpe broth, yeast extract peptone dextrose broth and black tea). The main strains involved in AFB1 decontamination were identified based on DNA sequencing and the toxicity of the new products was evaluated on Hep2 cells and on Brine shrimp (Artemia salina). Our results showed that after 7 days of fermentation, kombucha was able to degrade 97% of AFB1 in black tea. Moreover, the effective yeasts present in Kombucha were identified as Pichia occidentalis, Candida sorboxylosa and Hanseniaspora opuntiae and the highest AFB1 degradation capacity was accorded to P. occidentalis (59%) when cultivated in black tea. Data on cytotoxicity tests on Hep2 cells and Brine shrimp (Artemia salina) showed that the biodegraded products were less toxic than pure AFB1. These findings suggest that, kombucha isolated strains could be potential candidates for application in the food and feed industry with a potential aflatoxin B1 detoxification properties.

In the current study, a simple method for the synthesis of 3-methyl-2-propyl-2H-[1,2,4]triazolo[4,3b][1,2,4,6]thiatriazine-1,1-dioxide (2) was carried out. In the presence of pyridine, a reaction between amidine (1) and sulfuryl chloride occurs. FTIR spectroscopy, 1H and 13C NMR, mass spectra, and elemental analysis were utilized in order to verify the structure of a novel synthetic molecule. The antibacterial activities of compound (2) were tested against eight pathogenic bacteria and the minimum inhibitory concentration as well as minimum bactericidal concentration were determined. Moreover, the possible antibiofilm effect of compound (2) was evaluated. Molecular docking was investigated to determine the interaction between compound (2) and eight crystal structures of bacterial and yeast proteins associated with virulence activity and antimicrobial resistance. Our results showed that the new 3-methyl-2-propyl-2H-[1,2,4]triazolo[4,3b][1,2,4,6]thiatriazine-1,1-dioxide (2) compound has a moderate antibacterial activity toward the selected pathogenic bacteria. The obtained MICs varied from 32 to 512 µg/mL being the lowest values attributed to Staphylococcus epidermidis ATCC 14990 and Streptococcus mutans ATCC 25175 (MIC = 32 µg/mL).We noted also that heterocyclic compound (2) may inhibit bacterial biofilm formation at concentration depend manner with a lowest value obtained against S.mutans ATCC 25175 (BIC50= 490 µg/mL). Molecular docking showed a promising inhibitory activity of compound (2) on TetM-mediated tetracycline resistance (3J25) and Staphylococcus aureus gyrase (3G7B) with lower binding energy compared to the other target proteins. Commiphora gileadensis, locally known as becham, is a plant used in traditional Arabian medicine for treating headache, constipation, stomach, joint pain, and inflammatory disorders. Several studies have reported its antibacterial properties; however, no study has demonstrated its antiviral activity. This study aimed to evaluate the antiviral activity of C. gileadensis as well as to isolate its active compound and investigate its mode of action. This activity was evaluated using 4 viruses, herpes simplex virus type 2 (HSV-2), respiratory syncytial virus type B (RSV-B), coxsackie virus B type 3, and adenovirus type 5 by performing the plaque reduction assay and the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays for enveloped and nonenveloped viruses, respectively. The methanol extract of C. gileadensis leaves only showed antiviral activity against enveloped viruses with a selectivity index of 11.19 and 10.25 for HSV-2 and RSV-B, respectively. The study of the mechanism underlying antiviral activity demonstrated a virucidal effect by direct contact with these target viruses. The active compound, isolated using bio-guided assays involving TLC, was identified as guggulsterone by HPLC-diode array detection coupled with electrospray ionization mass spectrometry. Guggulsterone is an antagonist of the bile acid receptor and a modulator of cholesterol metabolism; however, its antimicrobial properties have been reported for the first time in this study.

Mycotoxins are secondary metabolites produced by various toxigenic fungi belonging to the genera Aspergillus, Fusarium and Penicillium. They have mutagenic, teratogenic, carcinogenic, immunosuppressive and endocrine disrupting effects on both humans and animals. The ubiquitous occurrence and heat stability of these mycotoxins during food and feed processing pose serious health problems in human and animals. Therefore successful biotechnological detoxification tools are in great demand. Recently, the use of bacteria, yeast, moulds and enzymes to mitigate/bind or biodegrade various mycotoxins is a novel strategy used for mycotoxins removal. In this review, recent development in biological control of mycotoxins removal and detoxification will be discussed.

Dyes are chemicals found in industrial water discharge, which may harm aquatic life and humans. In this study, halophilic bacteria was isolated from sea water in Jeddah, KSA, and then identified using 16SrDNA sequencing technique. Mathematical modeling has been achieved to predict the number of runs needed to reach the maximum of dye removal. Then the effect of four independent variables (NaCl concentration, Peptone concentration, temperature and agitation) was studied using full Factorial Design (FFD) for the determination of optimal abiotic and biotic condition, which reaches a maximum percentage of colour removal (% of decolourization) in artificial seawater (ASW). A model was proposed for the statistical analysis of colour removal prediction. Analytical techniques (UV–vis and FTIR) was also used to determining the degradation mechanisms. The active strain was then genetically characterized to identify genes related to chemical and dyes biodegradation. The isolated strain was identified as Photobacterium ganghwense. Our results revealed that P. ganghwense (5.106 CFU/ mL) was able to decolourize four tested dyes (Methyl orange, Congo red, Crystal violet and Malachite green) in modified ASW with different degrees and the maximum of decolourization was attributed to malachite green (79.61%). The biodegradation capability of the selected strain was confirmed by UV–Vis spectrum and FTIR analysis. Experimental data and model predictions were found to be in good agreement. Genomic analysis confirm the presence of several enzymes (reductase, dehydrogenase, oxidase, kinase, hydrolase, and transferase) associated with dyes and xenobiotic degradation, indicating its possible use in biotechnology applications.

The Aims of the study was to evaluate the antibacterial susceptibility and the biofilm eradication of three natural compounds carvacrol (CAR), thymol (TH) and eugenol (EUG), alone or in combination with nalidixic acid (NA) against twelve Salmonella Typhimurium strains. The minimum inhibitory concentration (MIC) and the minimum biofilm eradication concentration (BEC50) of the tested compounds (CAR, TH and EUG) and their combinations with NA were evaluated. In order to assess whether these bacteria had active efflux pumps, ethidium bromide (EtBr) accumulation assays was achieved using spectrophotometric accumulation assays. Moreover, scanning electron microscopy was used to visualize the bacterial biofilm formation on stainless steel surfaces after exposed to NA, CAR, TH and EUG alone and in combination. TH was the most effective essential oil, with the lowest MICs values ranging from 32 to 128 μg/mL followed by EUG and CAR. In addition, the combination of NA with the different compounds enhances antibiotic susceptibility of the tested bacterial strains. These results were confirmed by EtBr accumulation assays. A pronounced effect in decreasing biofilm mass was also noticed. Moreover, SEM revealed that bacterial membrane was disrupted and a complete loss of membrane integrity was also evident. The combination of natural compounds with antibiotic enhances bacterial susceptibility to NA. This combination ameliorates eradication of biofilm formed by S. Typhimurium on polystyrene microtitre plates. Additionally, this synergy induces an alteration of the bacterial cell surface visualized by SEM.

Aflatoxin B

Almonds and peanuts are a rich source of proteins, vitamins and unsaturated fatty acids. However, they can be also contaminated by mycotoxigenic fungi; a reason that has enhanced to investigate efficient strategies of management of these fungal contaminations. Some Lactic acid bacteria have been proven capable of inhibiting growth and mycotoxin production in livestock and transform it into nontoxic derivatives. In this work, four lactic acid bacteria (LAB) were tested for their abilities to inhibit the growth and mycotoxin production of Aspergillus flavus and Aspergillus carbonarius. Antifungal activity was evaluated in agar medium as well as in almonds and peanuts. Results showed that LAB significantly inhibited Aspergillus flavus and Aspergillus carbonarius in agar medium but none of the strains were able to completely inhibit fungal growth. The highest fungal growth inhibition was obtained using L. kefiri FR7 (51.67% and 45.56% growth inhibition of A. flavus and A. carbonarius, respectively). The cell-free supernatants (CFS) from LAB reduced fungal growth with average growth inhibitions ranging from 13.33% to 40.56% and 12.78% to 37.78% for A. flavus and A. carbonarius, respectively. We noted also that cell-free supernatants at pH7 (CFS-pH7) from the entire tested LAB did not inhibit fungal growth. L. kefiri FR7 was the most effective strain in mycotoxin suppression with a reduction percentage reaching 97.22%, 95.27% and 75.26% for AFB1, AFB2 and OTA respectively. Moreover, the inoculation of L. kefiri FR7 in almonds artificially contaminated with A. flavus decrease 85.27% of AFB1 and 83.94% of AFB2 content after 7 days of incubation. On the other hand, application of L. kefiri FR7 in peanuts artificially contaminated with A. carbonarius reduced OTA content to 25%. Our study revealed the potential use and application of L. kefiri FR7 in the control of fungi growth and mycotoxins production in almonds and peanuts.

Three bacterial strains (TE1, TD3 and FB2) were isolated from date palm (degla), pistachio and barley. The presence of nitrate reductase (narG) and nitrite reductase (nirS and nirK) genes in the selected strains was detected by PCR technique. Molecular identification based on 16S rDNA sequencing method was applied to identify positive strains. In addition, the D-optimal mixture experimental design was used to optimize the optimal formulation of probiotic bacteria for denitrification process. Strains harboring denitrification genes were identified as: TE1, Agrococcus sp LN828197; TD3, Cronobacter sakazakii LN828198 and FB2, Pedicoccus pentosaceus LN828199. PCR results revealed that all strains carried the nirS gene. However only C. sakazakii LN828198 and Agrococcus sp LN828197 harbored the nirK and the narG genes respectively. Moreover, the studied bacteria were able to form biofilm on abiotic surfaces with different degree. Process optimization showed that the most significant reduction of nitrate was 100% with 14.98% of COD consumption and 5.57 mg/l nitrite accumulation. Meanwhile, the response values were optimized and showed that the most optimal combination was 78.79% of C. sakazakii LN828198 (curve value), 21.21% of P. pentosaceus LN828199 (curve value) and absence (0%) of Agrococcus sp LN828197 (curve value).

The overuse of antibiotics and biofilm formation ability has led to the emergence of bacterial resistant strains. The combined use of several antibiotics has been found as an efficient strategy to overcome this resistance. In this study, two exopolysaccharides (EPS) obtained from Lactobacillus plantarum (EPS-Lp) and Bacillus spp. (EPS-B), isolated from a traditional Tunisian food “ricotta cheese” and hypersaline environment respectively, were used to counteract the biofilm formation and efflux pumps activities in Escherichia coli ATCC35218. The obtained results revealed that the tested EPSs can be effective against E. coli at a concentration > 1 mg/ml and were able to modulate biofilm formation by 50%. Moreover, at a concentration of 512 μg/ml, the tested EPSs inhibit the EtBr efflux in the tested bacteria and no significant difference was shown compared to cells treated with reserpine (P > 0.05). The positive effect of the tested EPSs may be due to the decrease of Indole production level proposed as a signal involved in quorum sensing and through the significant reduction of the hydrophobicity percentage between the treated and untreated cells. Overall, EPS-Lp and EPS-B, when used at appropriate concentration, may inhibit biofilm formation and reduce efflux pumps implicated in bacterial adhesion and antimicrobial resistance. These results make them an interesting candidate in the design of a new strategies to control bacterial biofilm-associated infections.

Synthesized Pyridin-2-yl hexanoate was screened for its antibacterial and antibiofilm activities using the broth microdilution method and XTT assay respectively. The obtained results revealed that synthesized molecule inhibited the growth of pathogenic bacteria (MICs value ranged from 80 to 640 μg ml−1). Moreover, it induced a strong antibiofilm effect against Gram positive cocci (BIC50 was 366, 378 and 412 μg ml−1 for Streptococcus mutans ATCC 25175, Streptococcus salivarius ATCC 13419 and Streptococcus oralis ATCC 6249 respectively). Basis on these results, pyridin-2-yl hexanoate may be considered as effective compound with antibacterial and antibiofilm activities.

Dental caries remains the most prevalent oral infectious disease worldwide. In this study, the antibacterial and the antibiofilm activities of five essential oils (EO's): eugenol (EUG), carvacrol (CAR), thymol (TYH), p-cymene (CYM) and γ-terpinene (TER) were tested (alone or in combinaison with tetracycline) against oral bacteria. In addition, their potential roles to enhance the accumulation of ethidium bromide (EtBr) in bacterial cells were tested. Our results indicated that EO's induced a selective antimicrobial activity. A synergistic effect of EO's and tetracycline (TET) was noticed with a reduction rate ranged from 2 to 8-fold. In addition, the efflux of EtBr was inhibited with a decrease in loss of EtBr from the bacteria. On the other hand a significant anti-biofilm activities of EO's (alone or combined with antibiotics) was noticed. In conclusion the tested EO's may be considered as a potential natural source with a resistance-modifying activity and may be applied to eradicate bacterial biofilm.

A strategy to reduce the deleterious effects of mycotoxins is to use dietary supplements that contain microorganisms that bind mycotoxins and diminish their gastrointestinal absorption. Novel strains were isolated from a Kefir culture and assessed for their mycotoxin adsorption and biotransformation ability. The most active strains were identified using DNA sequencing, and the stability of microorganism/mycotoxin complexes was evaluated using buffer solutions to simulate the pH conditions in the gastrointestinal tract. Our results showed that the microorganism consortium of Kefir grains adsorbed 82 to 100% of aflatoxin B1 (AFB1), zearalenone (ZEA) and ochratoxin A (OTA) when cultivated in milk. The main strains that were capable of mycotoxin adsorption were identified as Lactobacillus kefiri, Kazachstania servazzii and Acetobacter syzygii. The strain L. kefiri KFLM3 was the most active, adsorbing 80 to 100% of the studied mycotoxins when cultivated in milk. Nonetheless, the strain K. servazzii KFGY7 retained more mycotoxin after the desorption experiments (65, 69 and 67% for AFB1, OTA and ZEA, respectively). These findings suggest that Kefir consumption may help to reduce gastrointestinal absorption of these mycotoxins and consequently reduce their toxic effects. The persistence of pathogenic bacteria in the marine environment has been thoroughly investigated. The potential threat that these microorganisms pose to public health in recreational waters has always been a concern. In this study, the persistence and the response of Pseudomonas aeruginosa ATCC27853 to starvation and osmotic stress were studied after its incubation in sterilized seawater during 12 months. Three different colonial variants were isolated: A7 after one month, and A81 and A82 after 8 months of incubation period. The incubation effect on the bacterial phenotype and genotype were studied by analyzing modifications in morphology, antibiotic and metal resistance, molecular typing (PFGE and MLST), pigment production and virulence factors. The starved variants showed three different colony forms, but an indistinguishable PFGE pattern and belonged to ST155, as P. aeruginosa ATCC27853. The starved variants maintained the susceptibility to the 13 tested antibiotics, with the exception of the imipenem-resistant A82 strain, which also showed a small colony variant phenotype and the highest values of tolerance to the CuSO4 + NaCl combination. Significant differences were detected in the pigment production, the elastase activity and cytotoxic potential of the starved isolates in comparison to P. aeruginosa ATCC27853. Long-term exposure to stress, such as the incubation in seawater, was shown to induce different responses in P. aeruginosa, including virulent and resistant phenotypes.

In view of compliance with increasingly stringent environmental legislation imposed by regional, national and supranational (for example, European Union) authorities, innovative environmental technologies for the treatment of dye-contaminated effluents are necessary in the color industry. In this study, azo (methyl orange (MO) and methyl red (MR)) and triphenylmethanes (malachite green (MG), fushin (F) and crystal violet (CV)) dyes were subjected to distinct treatment strains following an initial qualitative characterization. The effectiveness of their biodegradation using Staphylococcus epidermidis strain was compared with respect to parameters such as residual color, and toxicity on human cells. The Hep-2 cells were exposed to three azo and two triphenylmethane dyes at various concentrations. After 72 h exposure, the protein contents of the samples compared to the protein contents of non-exposed cells were measured. The level of protein content indicates the viability of the cells. The IC20 values show the limiting value of toxicity. The IC50 values before biodegradation show whether samples are clearly toxic. The IC50 value for the azo dyes (MO, MR) were 65 and 10 μg/ml, the IC20 value were 10 and 5 μg/ml. The IC50 for the triphenylmethane dyes (MG, F and CV) were 35, 25 and 30 μg/ml, the IC20 value were 5, 5 and 10 μg/ml. The viability of the cells was good, the protein content of the samples being over 80% compared to the non-exposed cells. The Hep-2 cell test indicated the toxicity of pure dyes before biodegradation; the dyes after decolorization had no adverse effect. The human keratinocyte Hep-2 cells seem to be a useful tool for the study of the purity/toxicity of dyes and other substances applied to textiles. In addition, the initial cytotoxicity of dyes was still present after the biodegradation, while it was completely removed through the bacterial treatment. Our results show that S. epidermidis was a slime-producer developing almost black colonies on Congo red agar plate. A significant increase in azoreductase, lignin peroxidase and laccase activities in the cells were obtained after complete decolorization. Tunisian Juglans regia bark ethanol extract (JREE) was examined for antibacterial, antibiofilm, and antioxidant activity. Its anti-proliferative effect was evaluated on normal (MRC-5) and cancer (HT29 and HEp-2) cell lines. The data revealed that JREE inhibited the growth of pathogenic bacteria (MICs value ranged from 32 to 128 μg.mL-1) and induced a strong antibiofilm effect (BIC50 was 23, 30, and 43 μg.mL-1 for Micrococcus luteus, Staphylococcus epidermidis, and Staphylococcus aureus, respectively). The JREE showed high scavenging property against 2,2-diphenyl-l-1-picrylhydrazil (DPPH) radical (IC50 = 3.5 μg.mL-1) and exhibited anti-proliferative effects with IC50 values ranging from 6.6 to 25.5 μg.mL-1.

Three Lactobacilli were isolated from traditional Tunisian fermented food and then characterized for its ability to inhibit Listeria monocytogenes growth. Antagonistic effect of Lactobacillus plantarum on four L. monocytogenes strains was tested in soft artisanal cheese. Anti-biofilm activity of Lactobacillus extracts was also tested. Our results demonstrate that the selected lactic acid bacteria (LAB) extract exhibited a good antibacterial effect against L. monocytogenes ATCC 19115 with low MICs values: 8.33 2.1, 20 1.2 and 23.33 1, respectively. L. plantarum also exhibited a stronger inhibitory effect against L. monocytogenes when grown in cheese. Moreover, a potential anti-biofilm effect of the three LAB extracts with BIC50 values ranging from 5% to 15% for L. monocytogenes ATCC 19115 was demonstrated. Although LAB extracts were able to eradicate significantly a preformed L. monocytogenes biofilm (P < 0.05). Growth inhibition of preformed biofilm was more difficult to achieve. LAB could be used as a bioprotective culture in cheese ripening to prevent Listeria growth.

In this study the minimal inhibitory concentration (MICs) of tetracycline (Tet), erythromycin (Ery) and benzalkonium chloride (BC) in absence and in presence of a sub-MIC of juglone (Jug) were determined. In addition, the Ethidium bromide (EtBr) efflux assay was performed to assess the effect of Jug on EtBr cells accumulation. Our results showed a selective antimicrobial activity of Jug against the tested strains. A synergistic effect of Jug, drugs (Tet and Ery) and disinfectant (BC) was noticed with a reduction rate varied from 2 to 16-fold. In addition, the efflux of EtBr was inhibited depending on the Jug concentration. In the presence of Jug, a decrease in loss of EtBr from bacteria was observed. The concentration inducing 50 % of EtBr efflux inhibition after 15 min was about 182 μg ml-1 for S. aureus ATCC 25923, 236 μg ml-1 for S. aureus B193 and 195 μg ml-1 for S. aureus B456. It appears from this study that Jug may be used as a natural source for resistance-modifying activity in same bacteria.

In this study, three lactic acid bacteria (LAB), isolated from barley, traditional dried meat and fermented olive were characterized and tested for their anti-bacterial and anti-biofilm activities against oral bacteria. Our results revealed that the tested LAB were γ-hemolytic and were susceptible to four antibiotics. All the strains were resistant to low pH, bile salt, pepsin and pancreatin. Furthermore, FB2 displayed a high aut-oaggregative phenotype (99.54%) while FF2 exhibited the best co-aggregation rate. Concerning the microbial adhesion to solvent, FB2 was the most hydrophobic strain (data obtained with chloroform and n-hexadecane). In addition Pediococcus pentosaceus FB2 and Lactobacillus brevis FF2 displayed a significant inhibitory effect against Streptococcus salivarius B468 (MIC = 10%). Moreover the selected strains were able to inhibit biofilm formation of Bacillus cereus ATCC14579 (MBIC 50 = 28.16%) and S . salivarius B468 (MBIC 50 = 42.28%). The selected LAB could be considered as candidate probiotics for further application in functional food and mainly in the prevention of oral diseases.

Candida is a major Human pathogen causing a variety of infections and can survive for extended period of time in aquatic environment including marine and fresh water. In this study we compared a colorimetric XTT assay to colony forming units (CFU) count to evaluate the survival potential of Candida albicans incubated in water microcosms. Our results showed that cells maintain cultivability within a long period followed by a decline in cultivability and a drop of plate counts to less than 20 cell ml(-1) after 150 days in tap water, 190 days in rain water and 200 days in seawater. In addition we noted that 10% of cells viability was reached after 150 days in seawater, 180 days in rain water and 210 days in tap water. Molecular method confirms the persistence of C. albicans cells in water during long time starvation period.

Staphylococcus aureus is an important pathogen associated to dental infection. Many antiseptic agents are used in hygienic handwash to prevent nosocomial infections by methicillin-resistant staphylococci. In this study, 22 Staphylococcus aureus strains isolated from the oral cavity of Tunisian children were investigated for their susceptibility to benzalkonium chloride (0.5–512 μg/ml) and antibiotics. The β-lactams resistance gene blaZ, the erythromycin resistance methylase genes (ermA, ermB and ermC), the macrolide efflux gene (msrA) and the disinfectant resistance genes (qacH, qacA, qacB and qacC) were also investigated. Determination of the minimal inhibitory concentration values revealed that 54.5, 54.5, 68, and 82% of isolates were resistant to benzalkonium chloride, oxacillin, tetracycline and erythromycin respectively. The frequency of strains positive for the antibiotic resistance genes tested was 100 (blaZ), 50 (ermA), 36.4 (ermB), 22.7 (ermC) and 13.6% (mrsA). The qacH and the qacA genes were found in 22.7% of isolates and qacB and qacC in 13.6%. Two strains harboured three qac (qacH, qacA and qacB or qacC) genes. These data highlight the importance of determining the susceptibility to antibiotics and disinfectants of strains isolated in dental medicine in order to monitor the epidemiology and spread of multi-drug resistant staphylococci.

The occurrence of dental caries is mainly associated with oral pathogens, especially cariogenic bacteria. Numerous studies have validated the traditional use of medicinal plants by investigating the biological activity of essential oils. The Eugenia caryophyllata (clove) essential oil was tested in vitro against a large number of oral pathogens (114 streptococci and 46 yeast strains) using a disc diffusion method. The cytotoxicity assay of Eugenia caryophyllata essential oil on cancer cells (HT29, A549, Hep2, raw 264.7) and normal cells (MRC-5) was determined by the ability of the cells to metabolically reduce MTT to a formazan dye. Our results revealed that Eugenia essential oil possessed an excellent antibacterial activity against oral streptococci including the cariogenic bacteria as well as an excellent antifungal activity. Furthermore, the Eugenia caryophyllata essential oil showed significant cytotoxic effects against all studied cancer cell lines as judged by IC50 and its value ranges from 15.75 to 200 μg/ml. In conclusion, it is clear that clove oil shows powerful antibacterial and antifungal activity. The cytotoxic activity of the essential oil was dependent on the tested cell lines.

Staphylococcus aureus is an important pathogen that forms biofilm. In this study, 22 S. aureus have been isolated from the oral cavity of Tunisian children and investigated for slime production using Congo red agar method (CRA) and semi quantitative adherence assay. The hydrophobicity of strains was evaluated by the microbial adhesion to solvent (MATS) test. The adherence of S. aureus to Hep2 cells was examined by light microscopy. The genes implicated in adhesion (icaA, icaD, fnbA, cna, clfA) were detected. Polymerase chain reaction was used. The affinity to hexadecane was low proving a hydrophilic character of all the studied strains. Qualitative biofilm production revealed that 50% of strains were slime producers. The result of OD(570) showed that four strains isolated from the caries-active children were highly biofilm positive. In addition, 50% of strains were icaA and icaD positive. The fnbA gene was present in 59.1% of isolated strains. Furthermore, 54.5% of strains harboured the cna gene, 9.1% were clfA positive and 50% were hla positive. Quantitative adherence varied considerably among the tested strains. All strains showed adherence to Hep2 cells. However, the level of adhesion varied between strains as follows. Seven strains were defined as moderately adherent, nine as strongly adherent and six as weakly adherent. The percentage of infected cells ranged from 15+/-0.0376 (B374) to 96+/-0.019 (B295) and the total number of bacteria per 100 cells ranged from 15+/-5.1 (B374) to 1824+/-30.1 (B295).

Oral diseases, such as dental caries and periodontal disease are directly linked with the ability of bacteria to form biofilm. The development of dental caries involves acidogenic and aciduric Gram-positive bacteria colonizing the supragingival biofilm (Streptococcus, Lactobacillus and Actinomycetes). Periodontal diseases have been linked to anaerobic Gram-negative bacteria forming a subgingival plaque (Porphyromonas gingivalis, Actinobacillus, Prevotella and Fusobacterium). Cells embedded in biofilm are up to 1000-fold more resistant to antibiotics compared to their planctonic ones. Several mechanisms have been proposed to explain biofilms drug resistance. Given the increased bacterial resistance to antibiotics currently used in dentistry, a great importance is given to natural compounds for the prevention of oral bacterial growth, adhesion and colonization. Over the past decade, interest in drugs derived from medicinal plants has markedly increased. It has been well documented that medicinal plants and natural compounds confer considerable antibacterial activity against various microorganisms including cariogenic and periodontal pathogens. This paper provides a review of the literature focusing on the studies on (i) biofilm in the oral cavity, (ii) drug resistance of bacterial biofilm and (iii) the potential use of plant extracts, essential oils and natural compounds as biofilm preventive agents in dentistry, involving their origin and their mechanism of biofilm inhibition.

The occurrence of several microbial species in the oral cavity of 4–12-year-old Tunisian children was investigated. Samples were taken from 158 children (81 caries actives and 77 caries free). Genomic DNA was extracted and analyzed for the presence of 17 microbial species using a polymerase chain reaction assay. All samples were positive for at least one of the target microbial strains. Streptococcus mutans was the most prevalent species (76.5%) detected in genomic DNA collected from carious lesions. Other prevalent species were Candida spp (63%), Streptococcus salivarius (59%) and Streptococcus oralis (42%). The frequency of Lactobacillus acidophilus, Lactobacillus plantarum, and Lactobacillus casei-group in caries lesions was 29.5%, 34.5% and 22% respectively. Pathogenic bacteria such as Staphylococcus aureus was found in 28.5% of carious lesion samples compared to 15.5% in the control. Frequency of Porphyromonas endodontali, Actinomyces radicidentis and Treponema denticola recovery did not differ significantly between origins of samples. PCR analysis of genomic DNA detect various oral bacteria that differ between caries actives and caries-free children. In addition, the association of same aciduric bacteria (S. mutans, S. salivarius, L. acidophilus) and caries formation was noticed.

Background Thymoquinone is an active principle of Nigella sativa seed known as "Habbah Al-Sauda" in Arabic countries and "Sinouj" in Tunisia. Bacterial biofilms tend to exhibit significant tolerance to antimicrobials drugs during infections. Methods The antibacterial activity of Thymoquinone (TQ) and its biofilm inhibition potencies were investigated on 11 human pathogenic bacteria. The growth and development of the biofilm were assessed using the crystal violet (CV) and the 2, 3-bis [2-methyloxy-4-nitro-5-sulfophenyl]-2H-tetrazolium-5-carboxanilide (XTT) reduction assay. Results TQ exhibited a significant bactericidal activity against the majority of the tested bacteria (MICs values ranged from 8 to 32 μg/ml) especially Gram positive cocci (Staphylococcus aureus ATCC 25923 and Staphylococcus epidermidis CIP 106510). Crystal violet assay demonstrated that the minimum biofilm inhibition concentration (BIC50) was reached with 22 and 60 μg/ml for Staphylococcus aureus ATCC 25923 and Staphylococcus epidermidis CIP 106510 respectively. In addition our data revealed that cells oxidative activity was influenced by TQ supplementation. In the same way, TQ prevented cell adhesion to glass slides surface. Conclusion The ability of TQ to prevent biofilm formation warrants further investigation to explore its use as bioactive substances with antibiofilm potential.

We investigate in this work the chemical composition by GC–EIMS, the antibacterial and the cytotoxic activities of Tunisian Nigella sativa essential oil and its bioactive compound, thymoquinone, were tested against various clinical cariogenic bacteria ( n = 30). Eighty-four compounds were identified in the essential oil. The major one was p-cymene (49.48%) whereas thymoquinone represented only 0.79%. The essential oil (2.43 mg/disc) containing only 3.35 μg of thymoquinone showed pronounced dose dependant antibacterial activity against Streptococcus mitis , Streptococcus mutans , Streptococcus constellatus and Gemella haemolysans (15.5 ± 0.707 mm). However, pure thymoquinone compound (150 μg/disk) was active against all the studied strains especially S. mutans and S. mitis (24.5 ± 0.71 and 22 ± 1.41 mm inhibition zones, respectively). Their MIC values are representatives of a good effect. So, the essential oil strongest activity was seen against S. mitis , S. mutans , S. constellatus and G. haemolysans (MIC 2.13 mg/ml). However, thymoquinone was active against all the studied strains. The most important antibacterial activity was seen against S. constellatus (MIC 4 μg/ml). The growth inhibition effects of thymoquinone on human epithelial cell lines (Hep-2) were significantly stronger than the essential oil. The IC50 of thymoquinone and essential oil were 19.25 ± 1.6 and 55.2 ± 2.1 μg/ml, respectively. Our results demonstrate an important anticariogenic activity of the Tunisian N. sativa essential oil and thymoquinone. These effects further validate the traditional use of N. sativa in the folk medicine against various bacterial infections.

Biofilm formation is the leading cause of the pathogenesis of S. aureus associated with biomaterial infections. In S. aureus polysaccharide intercellular adhesin (PIA) was encoded by icaA and icaD genes. Production of PIA is currently responsible for staphylococcal biofilm development. In this study, S. aureus strains isolated from auricular infection (n = 46) and S. aureus ATCC 25923 were phenotyped and genotyped. Slime production was assessed using Congo red agar plate assay. In order to determine the biofilm formation capacity at various pH levels of the studied S. aureus strains, microtiter plate assay was performed. The strains were grown in medium adjusted at various pH levels (3, 5, 7, 9 and 12) and medium supplemented with hydrogen peroxide 3% (v/v). Qualitative biofilm production of S. aureus revealed that 56.5% of strains were slime producers. In addition 78.26% of strains were icaA and icaD positive. Quantitative biofilm showed that biofilm production depended on the pH value of the medium. At highly acidic (pH 3) and alkaline (pH 12) levels, biofilm formation was lower, while at pH 7 the adhesion was moderate. In addition, the cells adhered weakly after 3% hydrogen peroxide supplementation. Our results suggest that pH was a stress factor that led some S. aureus strains to produce the biofilm.

Objective This study aims to investigate the antibiotic susceptibility of strains isolated from the oral cavity of Tunisian children. Design Strains were isolated from the oral cavity of Tunisian children (60 caries-actives and 30 caries-free). Molecular characterization was assessed by PCR assay to detect erythromycin methylase gene (ermB), macrolide efflux (mefI) and tetracycline resistance genes (tetM and tetO). Results A total of 21 species were isolated and identified. Antimicrobial susceptibility revealed that the resistance rate to antibiotics was as follow: erythromycin (22%), tetracycline (15.6%), cefotaxim, (7.3%), trimethoprim-sulfamethoxazol (37.6%), nitrofurantoine (2.8%), pristinamycin (17.4%), quinupristin-dalfopristin (15.6%), and rifampicin (3.7%). The majority of mefI positive strains (31.2%) were isolated from the carious children ( n = 34) in comparison with 8.25% from the control group ( n = 9). In addition, frequency of strains caring resistance genes were as follow: 12.84% for ermB, 9.17% for tetM and 27.52% for tetO from the carious children in comparison to 0.092%, 3.67% and 3.67% from the caries free group respectively. Conclusion Multi-resistance strains towards macrolides and tetracycline were recorded. The majority of strains carrying antibiotics resistance genes were isolated from the caries active children. The presence of multi-resistant bacteria in the oral cavity can be the major cause of antibiotic prophylaxis failure in dental practise.

Polyarylsulfone group is one of the most important polymeric materials used in the biomedical field, due to its excellent properties, such as good thermal, chemical, and mechanical stability. There are three important polyarylsulfone polymers, all of which have excellent electrical properties: polysulfone (PSu), polyarylsulfone (PAS) and polyarylethersulfone (PAES). All these polymers have excellent creep, radiation and high temperature resistance. In this study, we aimed to determine the effect of three sterilization processes (steam, ethylene oxide and gamma rays) on cytotoxicity of polyarylsulfone dialysis membranes. Ten long-term dialysis patients and ten age-matched healthy controls were enrolled in our study. We analysed (1) serum effect on cultured endothelial cell viability using MTT assay and (2) lipid peroxidation assessed by serum malondialdehyde (MDA) formation at the beginning (T0), the middle (T2) and the end (T4) of haemodialysis (HD) session. Our results clearly showed that steam-sterilized membranes improve endothelial cell viability when compared to ethylene oxide or gamma rays-sterilized ones. Moreover, there is a increased generation of MDA in patients sera during HD session. The serum MDA concentration was about 3, 6 and 10 times higher, respectively, for steam, ethylene oxide and gamma rays sterilization procedures when compared to the MDA amount in healthy subject sera. We concluded that using steam instead of ethylene oxide or gamma rays for sterilization may improve the biocompatibility of polyarylsulfone membranes.

Background Enterococci are increasingly associated with opportunistic infections in Humans but the role of the oral cavity as a reservoir for this species is unclear. This study aimed to explore the carriage rate of Enterococci in the oral cavity of Tunisian children and their antimicrobial susceptibility to a broad range of antibiotics together with their adherence ability to abiotic and biotic surfaces. Results In this study, 17 E. faecalis (27.5%) and 4 E. faecium (6.5%) were detected. The identified strains showed resistance to commonly used antibiotics. Among the 17 isolated E. faecalis, 12 strains (71%) were slime producers and 5 strains were non-producers. Among the 4 E. faecium, 2 strains were slime producers. All the tested strains were able to adhere to at least one of the two tested cell lines. Our result showed that 11 E. faecalis and 2 E. faecium strains adhered strongly to Hep-2 as well as to A549 cells. Conclusions Drugs resistance and strong biofilm production abilities together with a high phenotypic adhesion to host cells are important equipment in E. faecalis and E. faecium which lead to their oral cavity colonization and focal infections.

Background Staphylococcus aureus and Coagulase-negative staphylococci (CoNS) are a major source of infections associated with indwelling medical devices. Many antiseptic agents are used in hygienic handwash to prevent nosocomial infections by Staphylococci. Our aim was to determine the antibiotic susceptibility and resistance to quaternary ammonium compound of 46 S. aureus strains and 71 CoNS. Methods S. aureus (n = 46) isolated from auricular infection and CoNS (n = 71), 22 of the strains isolated from dialysis fluids and 49 of the strains isolated from needles cultures were investigated. Erythromycin resistance genes (ermA, ermB, ermC, msrA and mef) were analysed by multiplex PCR and disinfectant-resistant genes (qacA, qacB, and qacC) were studied by PCR-RFLP. Results The frequency of erythromycin resistance genes in S. aureus was: ermA+ 7.7%, ermB+ 13.7%, ermC+ 6% and msrA+ 10.2%. In addition, the number of positive isolates in CoNS was respectively ermA+ (9.4%), ermB+ (11.1%), ermC+ (27.4%), and msrA+ (41%). The MIC analyses revealed that 88 isolates (74%) were resistant to quaternary ammonium compound-based disinfectant benzalkonium chloride (BC). 56% of the BC-resistant staphylococcus isolates have at least one of the three resistant disinfectants genes (qacA, qacB and qacC). Nine strains (7.7%) among the CoNS species and two S. aureus strains (2%) harboured the three-qac genes. In addition, the qacC were detected in 41 strains. Conclusions Multi-resistant strains towards macrolide and disinfectant were recorded. The investigation of antibiotics and antiseptic-resistant CoNS may provide crucial information on the control of nosocomial infections.
