We report on three clutches with 13, 28 and 33 eggs, respectively, obtained from three Pristimantis rosadoi couples, caught in the wet Ecuadorian Chocó region. Embryos developed from two clutches and are illustrated herein. One clutch matured and 25 froglets hatched simultaneously after 21 days. The hatching of froglets, with different amounts of yolk remaining, indicates that this was a response to perceived predation risk and thus indicates plasticity in hatching time. We also observed a distinct change from rough to smooth dorsal skin in two of the three females, after disengaging from amplexus. Sibling relationships are conceptualized as one of the longest lasting relationships, but empirical information about young adults' sibling relationships is limited. The current study aimed to understand how young adults (n = 239) conceptualized sibling closeness and how interactive technology influenced their self-identified closest sibling relationship. Through qualitative content analysis, three themes were identified regarding experiences with sibling closeness: (1) determinate and steady features, (2) congruence in relationship expectations, and (3) communication. In a second qualitative content analysis, participants' perceptions of how interactive technology influenced their closest sibling relationship were represented in three themes: (1) functionality, (2) technology that promotes quality communication, and (3) technology is not a benefit for sibling relationship. AbstractAn increasing focus of recent research is on the role of technology in support of teaching, learning and renewal or revitalization of endangered languages. The potential roles of a given piece of technology logically depend, at least to some extent, on the features built into the technology at design stage. Since most language revitalization and renewal efforts are local, community-based initiatives, the preferences and insights of community members are critical to build into the design, to maximize the relevance and appeal of the technology to those who might use it to maintain and increase knowledge and skills in their language. Recognizing this, the project reported on here incorporated co-design into the development of a mobile app, Warrma Mangarrayi (“Listen to Mangarrayi”). The app design also drew on a model of language revitalization, which offers a conceptualization of how technology can be a resource for a language revitalization team whose goals include everyday phrase-based communication in the endangered language. In this article we discuss the iterative user-centered co-design process adopted for the research and the improvements resulting from community user feedback on different versions and how the resulting resource, Warrma Mangarrayi, responded to community needs and aspirations. Testicular germ cell tumors (TGCTs) are the most common cancers of young males. A substantial portion of TGCT patients are refractory to cisplatin. There are no effective therapies for these patients, many of whom die from progressive disease. Embryonal carcinoma (EC) are the stem cells of TGCTs. In prior in vitro studies we found that EC cells were highly sensitive to the DNA methyltransferase inhibitor, 5-aza deoxycytidine (5-aza). Here, as an initial step in bringing demethylation therapy to the clinic for TGCT patients, we evaluated the effects of the clinically optimized, second generation demethylating agent guadecitabine (SGI-110) on EC cells in an animal model of cisplatin refractory testicular cancer. EC cells were exquisitely sensitive to guadecitabine and the hypersensitivity was dependent on high levels of DNA methyltransferase 3B. Guadecitabine mediated transcriptional reprogramming of EC cells included induction of p53 targets and repression of pluripotency genes. As a single agent, guadecitabine completely abolished progression and induced complete regression of cisplatin resistant EC xenografts even at doses well below those required to impact somatic solid tumors. Low dose guadecitabine also sensitized refractory EC cells to cisplatin in vivo. Genome-wide analysis indicated that in vivo antitumor activity was associated with activation of p53 and immune-related pathways and the antitumor effects of guadecitabine were dependent on p53, a gene rarely mutated in TGCTs. These preclinical findings suggest that guadecitabine alone or in combination with cisplatin is a promising strategy to treat refractory TGCT patients.
