Post-acute sequelae of SARS-CoV-2 (PASC) is a significant public health concern. We describe Patient Reported Outcomes (PROs) on 590 participants prospectively assessed from hospital admission for COVID-19 through one year after discharge. Modeling identified 4 PRO clusters based on reported deficits (minimal, physical, mental/cognitive, and multidomain), supporting heterogenous clinical presentations in PASC, with sub-phenotypes associated with female sex and distinctive comorbidities. During the acute phase of disease, a higher respiratory SARS-CoV-2 viral burden and lower Receptor Binding Domain and Spike antibody titers were associated with both the physical predominant and the multidomain deficit clusters. A lower frequency of circulating B lymphocytes by mass cytometry (CyTOF) was observed in the multidomain deficit cluster. Circulating fibroblast growth factor 21 (FGF21) was significantly elevated in the mental/cognitive predominant and the multidomain clusters. Future efforts to link PASC to acute anti-viral host responses may help to better target treatment and prevention of PASC.

Modern criminal investigations heavily rely on trace bodily fluid evidence as a rich source of DNA. DNA profiling of such evidence can result in the identification of an individual if a matching DNA profile is available. Alternatively, phenotypic profiling based on the analysis of body fluid traces can significantly narrow down the pool of suspects in a criminal investigation. Urine stain is a frequently encountered specimen at the scene of crime. Raman spectroscopy offers great potential as a universal confirmatory method for the identification of all main body fluids, including urine. In this proof-of-concept study, Raman spectroscopy combined with advanced statistics was used for race differentiation based on the analysis of urine stains. Specifically, a Random Forest (RF) model was built, which allowed for differentiating Caucasian (CA) and African American (AA) descent donors with 90% accuracy based on Raman spectra of dried urine samples. Raman spectra were collected from samples of 28 donors varying in age and sex. This novel technology offers great potential as a universal forensic tool for phenotypic profiling of a potential suspect immediately at the scene of a crime, providing invaluable information for a criminal investigation. Blood glucose concentration is an important index for the diagnosis of diabetes, its self-monitoring technology is the method for scientific diabetes management. Currently, the typical household blood glucose meters have achieved great success in diabetes management, but they are discrete detection methods, and involve invasive blood sampling procedures. Optical detection technologies, which use the physical properties of light to detect the glucose concentration in body fluids non-invasively, have shown great potential in non-invasive blood glucose detection. This article summarized and analyzed the basic principles, research status, existing problems, and application prospects of different optical glucose detection technologies. In addition, this article also discusses the problems of optical detection technology in wearable sensors and perspectives on the future of non-invasive blood glucose detection technology to improve blood glucose monitoring in diabetic patients. In other words, the HIV (Human Immunodeficiency Virus) continues to exist nowadays, and a lot of individuals who are living with HIV have had difficulties all throughout their lives. And it can be acquired by getting into someone who has been infected by having fluids from the body, exchanging a syringe with blood, breastfeeding, or having sexual intercourse are all possible sources. This research study aims to explore and observe the specific situations and problems that encountered of the people living with HIV including the lived experiences particularly; (1) to discuss the lived experiences of People Living with HIV (2) to discover the challenges faced by People Living with HIV (3) to identify the coping mechanisms of people living with HIV. With the use of Thematic Analysis (TA), the study findings are the following; (1) Some of the people in the Philippines easily jumps into conclusions wherein in their perspectives when a gay (Bakla) they easily addressed and tend to assumed that this person are associated and normalizing that they contracted on the HIV diseases resulting that this is a discrimination and misconception for them especially to LGBT which this can be also have a big impact on their overall well-being as a person. (2) People living with HIV are facing and experiencing anxiety and depression regarding their HIV status wherein these individuals tend to overthink and cry because of the situations that they had. Other than that they also struggle when it comes to services, and finances including; the laboratory-based diagnosis, transit for care access and limited availability in healthcare. (3) Lastly, despite the situation and experiences that they had, people living with HIV had coping strategies at which point the advocacy really helps them to understand and to cope up their HIV status to understand their situations as a HIV individual. This study presents a new density filter for a pipe-shaped structure and its application to fluid topology optimization. A simple and straight pipe-shaped structure for fluid is preferred for many engineering purposes rather than the perplex manifold structure provided by the topology optimization method. To determine an optimal pipe structure for fluid, we develop a new density filter and apply it to fluid topology optimization. Hence, the original spatially varying design variables of the fluid topology optimization are modified based on the pipe density filter. Subsequently, the filter design variables, including a uniform pipe wall thickness and adjusted cross-section, are used for artificial pseudo-rigid bodies in fluid topology optimization. An additional constraint is imposed to maintain a nearly uniform pipe thickness. Several numerical examples are solved to demonstrate the validity of the present pipe density filter for fluid topology optimization problems minimizing the energy dissipation of the fluid and controlling the particles suspended in the fluid. Background: Clinical biomarkers, allow better classification of patients according to their disease risk, prognosis, and/or response to treatment. Although affordable omics-based approaches have paved the way for quicker identification of putative biomarkers, validation of biomarkers is necessary for translation of discoveries into clinical application. Objective: Accordingly, in this study, we emphasize the potential of in silico approaches and have proposed and applied 3 novel sequential in silico pre-clinical validation steps to better identify the biomarkers that are truly desirable for clinical investment. Design: As protein biomarkers are becoming increasingly important in the clinic alongside other molecular biomarkers and lung cancer is the most common cause of cancer-related deaths, we used protein biomarkers for lung cancer as an illustrative example to apply our in silico pre-clinical validation approach. Methods: We collected the reported protein biomarkers for 3 cases (lung adenocarcinoma-LUAD, squamous cell carcinoma-LUSC, and unspecified lung cancer) and evaluated whether the protein biomarkers have cancer altering properties (i.e., act as tumor suppressors or oncoproteins and represent cancer hallmarks), are expressed in body fluids, and can be targeted by FDA-approved drugs. Results: We collected 3008 protein biomarkers for lung cancer, 1189 for LUAD, and 182 for LUSC. Of these protein biomarkers for lung cancer, LUAD, and LUSC, only 28, 25, and 6 protein biomarkers passed the 3 in silico pre-clinical validation steps examined, and of these, only 5 and 2 biomarkers were specific for lung cancer and LUAD, respectively. Conclusion: In this study, we applied our in silico pre-clinical validation approach the protein biomarkers for lung cancer cases. However, this approach can be applied and adapted to all cancer biomarkers. We believe that this approach will greatly facilitate the transition of cancer biomarkers into the clinical phase and offers great potential for future biomarker research. The early diversification of tetrapods into terrestrial environments involved adaptations of their locomotor apparatus that allowed for weight support and propulsion on heterogeneous surfaces. Many lineages subsequently returned to the water, while others conquered the aerial environment, further diversifying under the physical constraints of locomoting through continuous fluid media. While many studies have explored the relationship between locomotion in continuous fluids and body mass, none have focused on how continuous fluid media have impacted the macroevolutionary patterns of limb shape diversity.We investigated whether mammals that left terrestrial environments to use air and water as their main locomotor environment experienced constraints on the morphological evolution of their forelimb, assessing their degree of morphological disparity and convergence. We gathered a comprehensive sample of more than 800 species that cover the extant family‐level diversity of mammals, using linear measurements of the forelimb skeleton to determine its shape and size.Among mammals, fully aquatic groups have the most disparate forelimb shapes, possibly due to the many different functional roles performed by flippers or the relaxation of constraints on within‐flipper bone proportions. Air‐based locomotion, in contrast, is linked to restricted forelimb shape diversity. Bats and gliding mammals exhibit similar morphological patterns that have resulted in partial phenotypic convergence, mostly involving the elongation of the proximal forelimb segments.Thus, whereas aquatic locomotion drives forelimb shape diversification, aerial locomotion constrains forelimb diversity. These results demonstrate that locomotion in continuous fluid media can either facilitate or limit morphological diversity and more broadly that locomotor environments have fostered the morphological and functional evolution of mammalian forelimbs. The main purpose of this review was to examine the evidence of the relationship between active smoking or passive smoking during pregnancy and atopic dermatitis in offspring. The protocol was written following the PRISMA Checklist and was registered in the PROSPERO database (registration number CRD42022381136). We implemented a comprehensive search in PubMed, Embase and Web of Science databases to identify all potentially related articles from inception through 1 December 2022. We assessed cohort studies and case–control studies using the Newcastle–Ottawa Scale (NOS), and the Joanna Briggs Institute (JBI) critical appraisal tool to assess the quality of cross‐sectional studies. Heterogeneity was investigated by using Cochrane Q tests and I2 statistics. In addition, according to the research design, population source and population size, the reasons for the heterogeneity were analysed. A total of 15 observational studies were included in this analysis. Our meta‐analysis suggests that atopic dermatitis in offspring is not associated with active smoking during pregnancy (pooled OR, 0.96 [95% CI 0.86–1.07]); however, it is related to passive smoking (OR, 1.52 [95% CI 1.36–1.70]). Passive smoking during pregnancy is associated with an increased risk of eczema development in offspring. More research is needed to explore the risk of active smoking and eczema development in offspring, especially the association between measurements of pregnancy cotinine levels in maternal body fluids and AD in offspring. Many fatal intoxications have been reported in connection with the consumption of newer, highly potent synthetic cannabinoids. Yet, a possible postmortem redistribution (PMR) might complicate reliable interpretation of analytical results. Thus, it is necessary to investigate the PMR-potential of new synthetic cannabinoids. The pig model has already proven to be suitable for this purpose. Hence, the aim of this study was to study the PMR of the synthetic cannabinoid 5F-MDMB-P7AICA and its main metabolite 5F-MDMB-P7AICA-dimethylbutanoic acid (DBA). 5F-MDMB-P7AICA (200 µg/kg body weight) was administered by inhalation to anesthetized and ventilated pigs. At the end of the experiment, the animals were euthanized and stored at room temperature for 3 days. Tissue and body fluid samples were taken daily. Specimens were analyzed after solid phase extraction using a standard addition method and LC–MS/MS, blood was quantified after protein precipitation using a validated method. In perimortem samples, 5F-MDMB-P7AICA was found mainly in adipose tissue, bile fluid, and duodenum contents. Small amounts of 5F-MDMB-P7AICA were found in blood, muscle, brain, liver, and lung. High concentrations of DBA were found primarily in bile fluid, duodenum contents, urine, and kidney/perirenal fat tissue. In the remaining tissues, rather low amounts could be found. In comparison to older synthetic cannabinoids, PMR of 5F-MDMB-P7AICA was less pronounced. Concentrations in blood also appear to remain relatively stable at a low level postmortem. Muscle, kidney, fat, and duodenum content are suitable alternative matrices for the detection of 5F-MDMB-P7AICA and DBA, if blood specimens are not available. In the bloodstream or other physiological fluids, "circulating cells and sub-cellular bio-particles" include many microscopic biological elements such as circulating tumor cells (CTCs), cell-free DNA (cfDNA), exosomes, microRNAs, platelets, immune cells, and proteins are the most well-known and investigated. These structures are crucial biomarkers in healthcare and medical research for the early detection of cancer and other disorders, enabling treatment to commence before the onset of clinical symptoms and enhancing the efficacy of treatments. As the size of these biomarkers to be detected decreases and their numbers in body fluids diminishes, the detection materials, ranging from visual inspection to advanced microscopy techniques, begin to become smaller, more sensitive, faster, and more effective, thanks to developing nanotechnology. This review first defines the circulating cells and subcellular bio-particles with their biological, physical, and mechanical properties and second focuses on their diagnostic importance, including their most recent applications as biomarkers, the biosensors that are utilized to detect them, the present obstacles that must be surmounted, and prospective developments in the domain. As technology advances and biomolecular pathways are deepens, diagnostic tests will become more sensitive, specific, and thorough. Finally, integrating recent advances in the diagnostic use of circulating cells and bioparticles into clinical practice is promising for precision medicine and patient outcomes. Composites comprising mechanically stronger ceramic and bioactive alongside resorbable ceramic are a most welcomed choice in load-bearing bone replacement. In this pursuit, the present study aims to investigate the varied levels of cerium additions in the ZrO 2 –CaSiO 3 composite. The influence of cerium concentration on the structural, mechanical, apatite formation, resorbability and in-vitro biological behaviour of the ZrO 2 –CaSiO 3 composite has been undertaken in the current study. Sol-gel synthesis technique has been adopted to yield composite powders and the structural behaviour of the resultant has been performed using various analytical techniques. The results envisaged the crucial role of Ce3+ concentration in accomplishing a phase stable t/c -ZrO 2 - α -CaSiO 3 composite after heat treatment at 1300 °C. XPS analysis revealed the presence of dual oxidation state (+3 and + 4) of cerium in the composite powders and a homogeneous mixture of the composite has been determined from morphological analysis. Mechanical properties determined from nanoindentation indicated better strength displayed by the composite than the natural bone. The immersion tests conducted in simulated body fluids indicated the development of apatite crystals on the composite. Resorbability and biocompatibility tests demonstrated the virtuous appropriateness of the 30 wt% Ce3+/Ce4+ doped t/c -ZrO 2 - α -CaSiO 3 composite for load-bearing bone replacement. Leukocyte esterase strips have been widely used to detect the presence of leukocyte in human body fluids. We investigated the correlation between fecal leukocyte esterase (FLE) and fecal calprotectin (FC) levels and compared manual with machine automated interpretation of FLE level. This prospective study enrolled inflammatory bowel disease and colitis patients in National Taiwan University Hospital from Dec 2021 to Feb 2022. FLE and FC measured using the same sample were compared with various FC cutoff values. The correlation between values indicated by the two tests was analyzed. Sensitivity, specificity, positive and negative predictive values, and area under the receiver operating characteristic curve (AUROC) were calculated using SAS. A total of 103 samples were analyzed. The correlation between FLE and FC level was moderate and positive (r = 0.3505, P = 0.0003). With an FLE reading more than 1+ indicating mucosa inflammation, when the FC cutoff was 50, 250, and 500 mg/kg, the sensitivities of FLE readings were 60.3 %, 74.3 %, and 84.6 %, respectively, and the specificities were 62.9 %, 58.8 %, and 58.4 %, respectively. With an FLE reading greater than 1+ indicating mucosa inflammation, FLE reflected FC with AUROC values at the optimal cutoff (500 mg/kg) of 0.72. No difference was noted between manual and machine readings for FLE. Positive FLE can predict FC levels of more than 500 mg/kg. The test is widely available, produces results on the same day, and is low cost; therefore, FLE should be further investigated for use in bowel inflammation monitoring. [ABSTRACT FROM AUTHOR]

Porous Zn scaffold, as a novel biodegradable metallic material, has emerged as a promising candidate for biomedical applications. In this paper, by using carbamide particles as the pore-making agent and Zn and Mg powders as the starting materials, porous Zn-Mg alloy scaffolds were fabricated via the low-temperature sintering route. A so-called one-step hydrothermal method was applied to form a hydrophobic film on surface of the porous Zn alloy scaffolds. The degradability of the porous Zn alloy scaffolds coated with/without hydrophobic film in the simulated body fluid (SBF) was investigated, and the results show that the hydrophobic film, which can adhere well to the metal substrate with thickness around 50 μm, can effectively delay the degradation of porous Zn alloy scaffolds; the degradation rates of porous Zn alloy scaffolds coated by the hydrophobic film are 26–57% lower than those of porous scaffolds without coating; especially the degradation rate of the porous Zn-5 wt.%Mg scaffold with the coating in SBF, which is 0.32 mg/(cm2 day), is at the same level as that of pure Zn. Abstract and ARP Position Statement: Based on the available body of scientific evidence and with the goals of promoting safety of combat sports athletes and striving for the advancement of clean sport, the Association of Ringside Physicians recommends the following regarding cannabis: • Use of marijuana or synthetic cannabinoids by combat sports athletes is discouraged due to unproven benefits and many known adverse effects. Acute use can impair cognition and complex motor function, which likely leads to reduced performance in combat sports. Chronic use can increase risk for heart and lung disease, several cancers, schizophrenia, and can reduce testosterone in men and impair fertility. Benefits from cannabis in most contexts, including athletic performance, have not been proven. • Use of topical purified CBD is neither encouraged nor discouraged. • Since acute cannabis intoxication can impair complex cognitive and motor function, any athlete suspected of acute intoxication at the time of competition – based on clinical judgment – should be banned from that competition. • Wide-scale regulation of cannabis based on quantitative testing has limited usefulness in combat sports, for the following reasons: ∘ Cannabis is not ergogenic and is likely ergolytic. ∘ Concentrations in body fluids correlate poorly with clinical effects and timing of use. ∘ Access to testing resources varies widely across sporting organizations. Point‐of‐care (POC) monitoring of patient condition is crucial for effective cancer treatment and prognosis. This can be achieved non‐invasively by analyzing exosomes in body fluids. However, the heterogeneity of exosomes and non‐standardized quantification methods may interfere with clearly determining the patient's condition. Therefore, there is a need for technology that can precisely analyze both tumor‐derived exosomes and normal exosomes. Herein, this study presents the exosome multiple‐separation for simultaneous detection (EXO‐MUSSID) platform, which simultaneously isolates different exosomes based on their magnetization properties and monitors therapeutic efficacy of drugs. Using immunoaffinity magnetophoresis technology, HER2‐overexpressing and normal exosomes are collected separately, enabling real‐time monitoring of HER2 (also known as ERBB2) expression by analyzing the mRNA of each exosome based on a catalytic hairpin assembly (CHA) reaction. A portable fluorescence reader customized for the EXO‐MUSSID platform is developed for POC monitoring of HER2‐overexpressing breast cancer (HER2+ BC). The performance of the EXO‐MUSSID platform is validated using urine samples from HER2+ BC mouse models, confirming the progression of HER2+ BC and the changes in HER2 expression due to trastuzumab treatment. ABSTRACT Currently, the majority of methods to monitor cancer treatment through the analysis of body fluids are based on a highly selective detection of single molecules or cells. In this study, we are considering the analysis of the aqueous medium of liquid samples, that is, water, itself, using aquaphotomics and near‐infrared spectroscopy (NIR) for spectral data acquisition and processing, within cancer research. Water, as a molecular system, is a rich source of information about the current state of a patient, which can be extracted from near‐infrared spectra of liquid samples via simple algorithms based on multivariate data analysis. Understanding the dynamics of the filling process of a pore body with a nonwetting fluid is important in the context of dynamic pore network models and others. It can justify many of the assumptions behind the different rules that describe how the network behaves during imbibition and drainage processes. It also provides insight into the different regimes pertinent to this system. The filling process starts with the contact line pinning at the pore entrance. Three regimes can be identified during the filling process that is related to how the contact line advances. In the first two regimes, the contact line pins at the pore entrance while the emerging droplet develops, and in the third one, the contact line departs the entrance of the pore and advances along the pore surface. During the first regime, which is brief, the curvature of the meniscus increases, and likewise, the corresponding capillary pressure, while in the other two regimes, the curvature decreases and so does the capillary pressure. Such behavior results in the rate at which the nonwetting fluid invades the pore to change. It initially decreases, then increases as the meniscus advances. The radius of curvature of the meniscus, eventually, increases to infinity for which the interface assumes a flat configuration. A one-dimensional modeling approach is developed that accounts for all these regimes. The model also considers the two immiscible fluids over a wide spectrum of contrast in viscosity. Information about the mean velocity of the invading fluid, the location of the contact line, the radius of curvature of the meniscus, the volume of the emerging droplet, and several others are among the details that the model provides. A computational fluid dynamics (CFD) simulation has also been considered to confirm the proposed fates of the interface and to provide a framework for comparisons. Surface enhanced Raman spectroscopy (SERS) is meeting the requirements in biomedical science being a highly sensitive and specific analytical tool. By employing portable Raman systems in combination with customized sample pre-treatment, point-of-care-testing (POCT) becomes feasible. Powerful SERS-active sensing surfaces with high stability and modification layers if required are available for testing and application in complex biological matrices such as body fluids, cells or tissues. This review summarizes the current state in sample collection and pretreatment in SERS detection protocols, SERS detection schemes, i.e. direct and indirect SERS as well as targeted and non-targeted SERS, and SERS-active sensing surfaces. Moreover, the recent developments and advances of SERS in biomedical application scenarios, such as infectious diseases, cancer diagnostics and therapeutic drug monitoring is given, which enables the readers to identify the sample collection and preparation protocols, SERS substrates and detection strategies that are best-suited for their specific applications in biomedicine. In recent years, various approaches have been undertaken to eliminate lipoproteins co‐isolated with extracellular vesicles, as they were initially regarded as contaminating entities. However, novel discoveries are reshaping our perspective. In body fluids, these distinct particles not only co‐exist, but also interactions between them are likely to occur. Extracellular vesicles and lipoproteins can associate with each other, share cargo, influence each other's functions, and jointly have a role in the pathomechanisms of diseases. Additionally, their association carries important implications for therapeutic and pharmacological aspects of lipid‐lowering strategies. Extracellular vesicles and lipoprotein particles may have roles in the elimination of each other from the circulation. The objective of this minireview is to delve into these aspects. Simple Summary: The diagnosis and treatment of ovarian cancer still pose many problems; so, it is important to search for effective biomarkers that will allow for the detection of changes in the early stages of the disease. There is more and more talk about the use of circulating tumor DNA in oncology as a promising biomarker and an aid in tailoring individual therapy to patients. We describe what changes we can observe in ctDNA in patients with ovarian cancer and how we can use this information in diagnosis and therapy. Ovarian cancer is the deadliest of all gynecological diseases because its diagnosis and treatment still pose many problems. Surgical excision, hormone therapy, radiation, chemotherapy, or targeted therapy for eradicating the main tumor and halting the spread of metastases are among the treatment options available to individuals with ovarian cancer, depending on the disease's stage. Tumor DNA that circulates in a patient's bodily fluids has been studied recently as a possible novel biomarker for a number of cancers, as well as a means of quantifying tumor size and evaluating the efficacy of cancer therapy. The most significant alterations that we could find in the ctDNA of ovarian cancer patients—such as chromosomal instability, somatic mutations, and methylation—are discussed in this review. Restoring the structures and functions of tissues along with organs in human bodies is a topic gathering attention nowadays. These issues are widely discussed in the context of regenerative medicine. Excipients/delivery systems play a key role in this topic, guaranteeing a positive impact on the effectiveness of the drugs or therapeutic substances supplied. Advances in materials engineering, particularly in the development of hydrogel biomaterials, have influenced the idea of creating an innovative material that could serve as a carrier for active substances while ensuring biocompatibility and meeting all the stringent requirements imposed on medical materials. This work presents the preparation of a natural polymeric material based on pullulan modified with silymarin, which belongs to the group of flavonoids and derives from a plant called Silybum marianum. Under UV light, matrices with a previously prepared composition were crosslinked. Before proceeding to the next stage of the research, the purity of the composition of the matrices was checked using Fourier-transform infrared (FT-IR) spectroscopy. Incubation tests lasting 19 days were carried out using incubation fluids such as simulated body fluid (SBF), Ringer's solution, and artificial saliva. Changes in pH, electrolytic conductivity, and weight were observed and then used to determine the sorption capacity. During incubation, SBF proved to be the most stable fluid, with a pH level of 7.6–7.8. Sorption tests showed a high sorption capacity of samples incubated in both Ringer's solution and artificial saliva (approximately 350%) and SBF (approximately 300%). After incubation, the surface morphology was analyzed using an optical microscope for samples demonstrating the greatest changes over time. The active substance, silymarin, was released using a water bath, and then the antioxidant capacity was determined using the Folin–Ciocâlteu test. The tests carried out proved that the material produced is active and harmless, which was shown by the incubation analysis. The continuous release of the active ingredient increases the biological value of the biomaterial. The material requires further research, including a more detailed assessment of its balance; however, it demonstrates promising potential for further experiments. [ABSTRACT FROM AUTHOR]

Indole-3-carbinol (I3C) is a major dietary component produced in Brassica vegetables from glucosinolates (GLS) upon herbivores' attack. The compound is gaining increasing interest due to its anticancer activity. However, reports about improving its level in plants or other sources are still rare. Unfortunately, I3C is unstable in acidic media and tends to polymerize rendering its extraction and detection challenging. This review presents a multifaceted overview of I3C regarding its natural occurrence, biosynthesis, isolation, and extraction procedure from dietary sources, and optimization for the best recovery yield. Further, an overview is presented on its metabolism and biotransformation inside the body to account for its health benefits and factors to ensure the best metabolic yield. Compile of the different analytical approaches for I3C analysis in dietary sources is presented for the first time, together with approaches for its detection and its metabolism in body fluids for proof of efficacy. Lastly, the chemopreventive effects of I3C and the underlying action mechanisms are summarized. Optimizing the yield and methods for the detection of I3C will assist for its incorporation as a nutraceutical or adjuvant in cancer treatment programs. Rationale: A general N‐glycoproteomics analysis pipeline has been established for characterization of mutation‐related gain‐of‐glycosylation (GoG) at intact N‐glycopeptide molecular level, generating comprehensive site and structure information of N‐glycosylation. Methods: This study focused on mutation‐originated GoG using a mass spectrometry‐based N‐glycoproteomics analysis workflow. In brief, GoG intact N‐glycopeptide databases were built, consisting of 2701 proteins (potential GoG N‐glycosites and amino acids derived from MUTAGEN, VARIANT and VAR_SEQ in UniProt) and 6709 human N‐glycans (≤50 sequence isomers per monosaccharide composition). We employed the site‐ and structure‐specific N‐glycoproteomics workflow utilizing intact N‐glycopeptides search engine GPSeeker to identify GoG intact N‐glycopeptides from parental breast cancer stem cells (MCF‐7 CSCs) and adriamycin‐resistant breast cancer stem cells (MCF‐7/ADR CSCs). Results: With the criteria of spectrum‐level false discovery rate control of ≤1%, we identified 87 and 94 GoG intact N‐glycopeptides corresponding to 37 and 35 intact N‐glycoproteins from MCF‐7 CSCs and MCF‐7/ADR CSCs, respectively. Micro‐heterogeneity and macro‐heterogeneity of N‐glycosylation from GoG intact N‐glycoproteins with VAR_SEQ and VARIANT were found in both MCF‐7 CSCs and MCF‐7/ADR CSCs systems. Conclusions: The integration of site‐ and structure‐specific N‐glycoproteomics approach, conjugating with GoG characterization, provides a universal workflow for revealing comprehensive N‐glycosite and N‐glycan structure information of GoG. Introduction: The interstitial space harbours two fluid compartments linked serially to the plasma. This study explores conditions that lead to fluid accumulation in the most secluded compartment, termed the "third space". Methods: Retrospective data was collected from 326 experiments in which intravenous crystalloid fluid was administered to conscious volunteers as well as a small group of anaesthetized patients. The urinary excretion and plasma dilution derived from haemoglobin served as input variables in nine population volume kinetic analyses representing subtly different settings. Results: An infusion of 250-500 mL of Ringer's solution expanded only the central fluid space (plasma), whereas the infusion of 500-1,000 mL extended into a rapidly exchanging interstitial fluid space. When more than 1 L was infused over 30 min, it was distributed across plasma and both interstitial fluid compartments. The remote space, characterized by slow turnover, abruptly accommodated fluid upon accumulation of 700-800 mL in the rapidly exchanging space, equivalent to an 11%-13% volume increase. However, larger expansion was necessary to trigger this event in a perioperative setting. The plasma half-life of crystalloid fluid was 25 times longer when 2,000-2,700 mL expanded all three fluid compartments compared to when only 250-500 mL expanded the central space (14 h versus 30 min). Conclusion: As the volume of crystalloid fluid increases, it apparently occupies a larger proportion of the interstitial space. When more than 1 L is administered at a high rate, there is expansion of a remote "third space", which considerably extends the intravascular half-life. [ABSTRACT FROM AUTHOR]

Canine reproduction differs from that of many other domestic animals, and increased knowledge on biochemical changes during canine pregnancy is important for investigations of infertility or subfertility. The total glycosylation pattern, i.e., the glycome, of body fluids reflects cellular status in health and disease. The aim of the present pilot study was to investigate pregnancy-related changes of the serum N-glycome in bitches. A method based on Rapifluor HILIC-UPLC-FLR-MS was optimized and applied for analysis and quantification of N-glycans in canine serum. Serum samples from six pregnant and five non-pregnant bitches, collected at four well-defined time points, were included. The levels of sialylated and galactosylated complex glycans were significantly elevated in serum from pregnant bitches, consistent with previous reports on human pregnancy. The levels of fucosylated and agalactosylated glycans decreased significantly in pregnant dogs. In non-pregnant dogs, the glycosylation pattern did not change during the cycle. Pregnancy is an inflammatory state, but our findings during canine pregnancy are quite the opposite to changes that have previously been described for dogs with a known parasitic infection. Evaluation of the canine glycome may thus be valuable in studies of canine pregnancy, possibly differing inflammatory changes related to pregnancy to those caused by an infection. Astronauts experience combined exposure to a cephalad fluid shift and mild hypercapnia during space missions, potentially contributing to health problems. Such combined exposure may weaken dynamic cerebral autoregulation. The magnitude of cephalad fluid shift varies between individuals, and dynamic cerebral autoregulation may be affected more by greater cephalad fluid shift during combined exposure. We evaluated the dose‐dependent effects of head‐down tilt (HDT) on dynamic cerebral autoregulation during acute combined exposure to HDT and 3% CO2 inhalation. Twenty healthy participants were randomly exposed to three angles of HDT (−5°HDT+CO2, −15°HDT+CO2 and −30°HDT+CO2). After 15 min of rest, participants inhaled room air for 10 min in a horizontal body position, then inhaled 3% CO2 for 10 min under HDT. The last 6 min of data were used for analysis in each stage. Arterial pressure waveforms were obtained using finger blood pressure, and blood velocity waveforms in the middle cerebral artery were obtained using transcranial Doppler ultrasonography. Dynamic cerebral autoregulation was evaluated by transfer function analysis between waveforms. Statistical analysis was performed by two‐way repeated‐measures analysis of variance. The index of transfer function gain in the low‐frequency range increased significantly with −15°HDT+CO2 and −30°HDT+CO2, but no changes were seen with −5°HDT+CO2. Phase in the low‐frequency range decreased significantly with all three protocols. These results of significant changes in indexes of both gain and phase during combined exposure to steep HDT (−15° to −30°) and 3% CO2 inhalation suggest weakened dynamic cerebral autoregulation with the combination of moderate cephalad fluid shift and mild hypercapnia. African swine fever (ASF) is one of the deadliest swine diseases, causing significant economic losses, threatening food security, and limiting pig production in affected countries. In the absence of an effective ASF vaccine, prevention and control of ASF depend mainly on effective biosecurity measures. In this study, the efficacy of SAFER®, a powdered disinfectant containing clay, an acid complex, and the active ingredient thyme essential oil, was tested against the ASF virus. The results showed that ASFV isolate (VNUA/HY/ASF1/Vietnam/2019) was inactivated by 3.5 and 5 Log10HAD50/ml after 20 and 120  min of treatment with SAFER®, respectively. When body fluids contaminated with ASFV, such as blood, saliva, urine, and feces, were treated with SAFER® for 20  min, the ASFV titer was reduced by 1.6, 2.2, 2.0, and 2.2 Log10HAD50/ml, respectively. The drag force exerted on a body moving in a fluid is empirically known to follow a linear drag law at low Reynolds numbers and a quadratic drag law at very high Reynolds numbers. The major challenge with the empirical description of the drag lies in the uncertainty of the description of the drag coefficient as a function of velocity. If the function is precisely known, it may be possible to find a general formula for the drag, which would, in principle, be applicable for all Reynolds numbers. The general law can consequently be tested using the linear and quadratic drag laws within their validity regimes to confirm its reliability and validity. The current absence of such a general law implies that the prediction of the dynamics of a body moving under drag forces in the intermediate regime of the Reynolds number can only be approximate and inaccurate since it is not guided by any valid law. In this study, we derive a general relation for the drag force, which may be applicable in all the Reynolds number regimes. We achieve this through a simple transformation of Newton's second equation for uniformly decelerated motion. It turns out that our general relation is a modification of the linear drag law. The effect of the introduced correction term in the dynamics of a projectile is evidently promising, as can be seen in the more realistic results generated. [ABSTRACT FROM AUTHOR]

To achieve stable and continuous supercavitating flow over underwater vehicles, artificial ventilation is implemented, particularly effective at lower speeds. Previous research on supercavitation primarily focused on analyzing ventilated supercavitating flow with various cavitator types and/or ventilation rates. In this investigation, we examine the behavior of ventilated supercavitating flow over an axisymmetric model featuring both a disk cavitator and a Postdam propeller placed at the bow. Utilizing the Large Eddy Simulation turbulence model, Volume of Fluid method, and Kunz cavitation model, our simulation aims to capture the cavitating flow around the propeller and the ventilated supercavitating flow over the model. Validation of our numerical methods is achieved by comparing our results with experimental data of a ventilated model by Chung and Cho ["Ventilated supercavitation around a moving body in a still fluid: Observation and drag measurement," J. Fluid Mech. 854, 367–419 (2018)] and the cavitating Postdam propeller by SVA Postdam [S. Potsdam, "PPTC smp'11 Workshop," in Proceedings of the Workshop on Cavitation and Propeller Performance (2011)]. The results show that with a rotating propeller at the bow of the supercavitating model, the cavitating flow extends and stabilizes compared to configurations utilizing a traditional disk cavitator. The presence of the propeller accelerates the formation of supercavitating flow at a consistent incoming flow speed. Additionally, coupling the propeller with the disk cavitator results in significant increases in propeller thrust, torque, and efficiency. Extracellular vesicles (EVs) contain cell‐derived lipids, proteins and RNAs; however, determining the tissue‐ and cell‐type‐specific EV abundances in body fluids remains a significant hurdle for our understanding of EV biology. While tissue‐ and cell‐type‐specific EV abundances can be estimated by matching the EV's transcriptome to a tissue's/cell type's expression signature using deconvolutional methods, a comparative assessment of deconvolution methods' performance on EV transcriptome data is currently lacking. We benchmarked 11 deconvolution methods using data from four cell lines and their EVs, in silico mixtures, 118 human plasma and 88 urine EVs. We identified deconvolution methods that estimated cell type‐specific abundances of pure and in silico mixed cell line‐derived EV samples with high accuracy. Using data from two urine EV cohorts with different EV isolation procedures, four deconvolution methods produced highly similar results. The three methods were also concordant in their tissue‐ and cell‐type‐specific plasma EV abundance estimates. We identified driving factors for deconvolution accuracy and highlighted the importance of implementing biological knowledge in creating the tissue/cell type signature. Overall, our analyses demonstrate that the deconvolution algorithms DWLS and CIBERSORTx produce highly similar and accurate estimates of tissue‐ and cell‐type‐specific EV abundances in biological fluids. Key Clinical Message: With no family history, and an atypical phenotype, the clinical diagnosing of Noonan syndrome (NS) can be very difficult. The present case emphasized that generalized edema in neonates may be the potential first symptom of NS. Severe generalized edema is a rare pathological condition with high mortality in newborns, in particular the premature infants. It is characterized by the extensive subcutaneous tissue edema and the accumulation of fluid in neonatal body fluid compartments. The etiology and pathogenesis of hydrops in neonates are quite complex. Generally speaking, hydrops can be divided into immune hydrops and non‐immune hydrops according to the etiology. It is still challenging in treating severe neonatal edema. In this study, we presented a preterm newborn with severe generalized edema after birth, which was finally diagnosed with Noonan syndrome (NS). The infant clinically manifested as severe generalized edema alone, without the involvement of multiple organ malformation. Generalized edema in neonates was probably the first symptom of NS. Proteins, saccharides, and low molecular organic compounds in the blood, urine, and saliva could potentially serve as biomarkers for diseases related to diet, lifestyle, and the use of illegal drugs. Lifestyle-related diseases (LSRDs) such as diabetes mellitus (DM), non-alcoholic steatohepatitis, cardiovascular disease, hypertension, kidney disease, and osteoporosis could develop into life-threatening conditions. Therefore, there is an urgent need to develop biomarkers for their early diagnosis. Advanced glycation end-products (AGEs) are associated with LSRDs and may induce/promote LSRDs. The presence of AGEs in body fluids could represent a biomarker of LSRDs. Urine samples could potentially be used for detecting AGEs, as urine collection is convenient and non-invasive. However, the detection and identification of AGE-modified proteins in the urine could be challenging, as their concentrations in the urine might be extremely low. To address this issue, we propose a new analytical approach. This strategy employs a method previously introduced by us, which combines slot blotting, our unique lysis buffer named Takata's lysis buffer, and a polyvinylidene difluoride membrane, in conjunction with electrospray ionization-mass spectrometry (ESI)/matrix-assisted laser desorption/ionization-mass spectrometry (MALDI-MS). Introduction: Investigations of responses of animals and humans to changes of plasma volume are usually reported as average responses of groups of individuals. This ignores considerable quantitative variation between individuals. We examined the hypothesis that individual responses follow a common temporal pattern with variations reflecting different parameters describing that pattern. Methods: We illustrate this approach using data of Hahn, Lindahl and Drobin (Acta Anaesthesiol Scand.2011, 55:987‐94) who measured urine volume and haemoglobin dilution of 10 female subjects during intravenous Ringer infusions for 30 min and subsequent 3.5 h. The published time courses were digitised and analysed to determine if a family of mathematical functions accounted for the variation in individual responses. Results: Urine excretion was characterised by a time delay (Td) before urine flow increased and a time course of cumulative urine excretion described by a logarithmic function. This logarithmic relation forms the theoretical basis of a family of linear relations describing urine excretion as a function of Td. Measurement of Td enables estimation of subsequent values of urine excretion and thereby the fraction of infused fluid retained in the body. Conclusion: The approach might be useful for physiologists and clinical investigators to compare the response to infusion protocols when both test and control responses can be described by linear relations between cumulative urine volume at specific times and Td. The approach may also be useful for clinicians by complementing strategies to guide fluid therapy by enabling the later responses of an individual to be predicted from their earlier response. Introduction: Alexander disease (AxD) is a rare leukodystrophy caused by dominant gain-of-function mutations in the gene encoding the astrocyte intermediate filament, glial fibrillary acidic protein (GFAP). However, there is an urgent need for biomarkers to assist in monitoring not only the progression of disease but also the response to treatment. GFAP is the obvious candidate for such a biomarker, as it is measurable in body fluids that are readily accessible for biopsy, namely cerebrospinal fluid and blood. However, in the case of ASOs, the treatment that is furthest in development, GFAP is the target of therapy and presumably would go down independent of disease status. Hence, there is a critical need for biomarkers that are not directly affected by the treatment strategy. Methods: We explored the potential utility of biomarkers currently being studied in other neurodegenerative diseases and injuries, specifically neurofilament light protein (NfL), phosphorylated forms of tau, and amyloid-β peptides (Aβ42/40). Results and Conclusions: Here, we report that GFAP is elevated in plasma of all age groups afflicted by AxD, including those with adult onset. NfL and p-tau are also elevated, but to a much lesser extent than GFAP. In contrast, the levels of Aß40 and Aß42 are not altered in AxD. [ABSTRACT FROM AUTHOR]

• Accelerant facilitated fire significantly effects the detection of blood on carpet and tile surfaces. • Successful soot removal can increase detection of heat damaged blood. • Wiping is the superior soot removal method on carpet and tile samples. • There is evidential worth in recovering heat damaged blood evidence at arson scenes. Previous literature has established that recovering heat damaged body fluids is possible, however with little investigation into the effect of accelerants used in initiating arson fires. This study therefore aimed to determine whether presumptive blood detection was affected by heat damage resulting from accelerant facilitated fires. Another objective was to examine various techniques for removing soot, which is a noted barrier to blood detection. The study focused on blood deposited on household flooring materials, one porous and one nonporous surface: carpet and tile respectively. Samples were burned with butane, petrol, and kerosene then presumptively tested using the Kastle Meyer colourimetric blood detection test. Testing was then repeated following soot removal by either wiping, scraping, or using liquid latex. The "strength" of positive detections was evaluated using a scale based on reaction speed and colour intensity. Results demonstrated that accelerants weakened detection strength, although nearly all samples tested positive overall, and the impact of each accelerant on both surface types was largely similar. It was also discovered that soot removal improved the strength of blood detection results in approximately 69% of carpet and 47% of tile samples, with wiping being the superior method on both surface types. Consequently, introducing this investigative step may be critical to maximizing blood evidence recovery in arson casework. The widespread use of Unmanned Underwater Vehicles (UUVs) in seafloor observatory networks highlights the need for docking stations to facilitate rapid recharging and effective data transfer. Floating docks are promising due to their flexibility, ease of deployment, and recoverability. To enhance understanding and optimize UUV docking with floating docks, we employ dynamic fluid body interaction (DFBI) to construct a seabed moored suspended dock (SMSD) model that features a guiding funnel, a suspended body, and a catenary of a mooring chain. This model simulates SMSD equilibrium stabilization in various ocean currents. Then, a UUV docking model with contact coupling is developed from the SMSD model to simulate the dynamic contact response during docking. The accuracy of the docking model was validated using previous experimental data. Through investigation of the UUV docking response results, sensitivity studies relating to volume, moment of inertia, mass, and catenary stiffness were conducted, thereby guiding SMSD optimization. Finally, sea tests demonstrated that the SMSD maintained stability before docking. During docking, the SMSD's rotation facilitated smooth UUV entry. After the UUV docked, the SMSD was restored to its original azimuth, confirming its adaptability, stability, and reliability. [ABSTRACT FROM AUTHOR]

Calcium silicate is the basic component of bioactive materials that directly bond to living bone (bone-bonding property) in the body environment because calcium silicate can easily form a hydroxyapatite layer on its surface after exposure to body fluids. The organic modification of calcium silicate ceramics is expected to improve their physicochemical properties. Organic modifications of layered inorganic substances have been studied to allow drug loading and sustained release through structural changes. However, few studies have investigated the effect of organic modifications on drug-release carriers. In this study, layered calcium silicate hydrate (C-S-H) prepared via a hydrothermal process was chemically modified with alkyl groups using alkoxysilane. In addition, the loading behavior of the antibiotic levofloxacin was quantitatively analyzed. Structural analysis of the synthesized samples indicated that the interlayer distance and amount of organic content in C-S-H increased with increasing carbon chain length and concentration of alkoxysilane. Levofloxacin was successfully incorporated into the modified C-S-H. It has been suggested that drug incorporation occurs mainly via surface adsorption rather than intercalation into the interlayer. Levels of anticancer agents in cancer patients' body fluids are typically measured to adjust drug dosages or improve treatment results. The goal of this research is to present a new method for extracting bicalutamide (BCT) from biological samples using a responsive polymeric nanoadsorbent that reacts to temperature and near‐infrared radiation (NIR). To achieve this, the surface layers of tungsten disulfide nanosheets are modified using poly (N‐vinylcaprolactam) and three generations of polymeric dendrimers. The adsorbent product is then characterized using thermogravimetric analysis, Fourier transform infrared spectroscopy, field emission scanning electron microscopy, and X‐ray diffraction techniques. The drug loading operation on the proposed adsorbent is studied through central composite design and response surface strategy, with optimization for temperature (25–45°C), pH (5–9), and contact time (2–18 min). Nonlinear kinetic and adsorption isotherm analysis results show the best fit with Langmuir and pseudo‐second‐order models. The drug release process from the BCT‐loaded adsorbent is investigated via an HPLC‐UV system under both NIR‐irradiated and non‐irradiated conditions. The suggested method demonstrates remarkable recovery rates for BCT spikes from urine (95.23%) and plasma (93.33%), respectively. Background: The diagnosis of Alzheimer's disease (AD) relies on core cerebrospinal fluid (CSF) biomarkers, amyloid beta (Aβ) and tau. As the brain is then already damaged, researchers still strive to discover earlier biomarkers of disease onset and the progression of AD. Glycation, advanced glycation end products (AGEs) and oxidative modifications on proteins in CSF mirror the underlying biological mechanisms that contribute to early AD pathology. However, analyzing free AGEs in the body fluids of AD patients has led to controversial results. Thus, this pilot study aimed to test the feasibility of detecting, identifying and quantifying differentially glycated, AGE or oxidatively modified peptides in CSF proteins of AD patients (n = 5) compared to a control group (n = 5). Methods: To this end, we utilized a data-dependent (DDA) nano liquid chromatography (LC) linear ion trap-Orbitrap tandem mass spectrometry (MS/MS)) approach and database search that included over 30 glycative and oxidative modifications in four search nodes to analyze endogenous modifications on individual peptides. Furthermore, we quantified candidate peptide abundance using LC Quan. Results: We identified 299 sites of early and advanced glycation and 53 sites of oxidatively modified tryptophan. From those, we identified 17 promising candidates as putative biomarkers (receiver operating curve-area under the curve (ROC-AUC) > 0.8), albeit without statistical significance. Conclusions: The potential candidates with higher discrimination power showed correlations with established diagnostic markers, thus hinting toward the potential of those peptides as biomarkers. Here, we report a rapid and accurate optical method for detecting cells from liquid samples in a label-free manner. The working principle of the method is based on the interference of parts of a conical laser beam, coming from a single-mode optical fiber directly, and reflected from a flat glass surface. The glass is functionalized by antibodies against the cells to be detected from the liquid sample. Cells bound to that surface modify the reflected beam, and hence, change the resulting interference pattern, too. By registering and interpreting the variation in the image, the presence of cells from the sample can be detected. As for a demonstration, cell suspensions from a U937 cell line were used in glass chambers functionalized by antibodies (TMG6-5 (mIgG1)) to which the cells specifically bind. The limit of detection (LOD) of the method was also estimated. This proof-of-concept setup offers a cost-effective and easy-to-use way of rapid and specific detection of any type of cells (including pathogens) from suspensions (e.g., body fluids). Confirmatory identification of dyes in the physical pieces of evidence, such as hair and fabric, is critically important in forensics. This information can be used to demonstrate the link between a person of interest and a crime scene. High performance liquid chromatography is broadly used for dye analysis. However, this technique is destructive and laborious. This problem can be overcome by near-Infrared excitation Raman spectroscopy (NIeRS), non-invasive and non-destructive technique that can be used to determine chemical structure of highly fluorescent dyes. Analyzed fabric materials often possess body fluid stains, which may obscure the accuracy of NIeRS-based identification of dyes. In this study, we investigate the extent to which fabric contamination with body fluids can alter the accuracy of NIeRS. Our results showed that NIeRS coupled with partial-least squared discriminant analysis (PLS-DA) enabled on average 97.6% accurate identification of dyes on fabric contaminated with dry blood, urine and semen. We also found that NIeRS could be used to identify blood, urine and semen on such fabric with 99.4% accuracy. Furthermore, NIeRS could be used to differentiate between wet and dry blood, as well as reveal the presence of blood on washed fabric. These results indicate that NIeRS coupled with PLS-DA could be used as a robust and reliable analytical approach in forensic analysis of fabric. [ABSTRACT FROM AUTHOR]

As well known in bioactives delivery and cancer/inflammation theranostics, polycations have recently extended their biomedical potentials in reducing visceral fat, embolizing body fluid, modulating blood coagulation, and ameliorating hyperammonemia. However, positive charges that dominate various action mechanisms, often raise risks in biosafety concerns. Also, the lack of intrinsic imaging properties of polycations makes it challenging to uncover their pharmacokinetic profiles in living objects. Herein, dopamine conjugation is demonstrated controllable and highly efficient in boosting the fluorescence or even white color emission of various polycations including hyperbranched poly(amido amine) (HPAA), polyethyleneimine and poly‐L‐lysine. By shielding partial surface positive charges, increasing hydrophobicity, and inducing aqueous self‐assembly, dopamine conjugation can significantly enhance the biocompatibility and decrease the toxicity and immunogenicity of polycations. Furthermore, the catechol‐metal binding interaction facilitates the accessibility of polycation‐based positron emission tomography (PET) probes. Different tissue uptake and biodistributions of gallium‐68‐radiolabeled HPAA before and after dopamine conjugation indicate that PET imaging can be a useful supplement to evaluate the dynamic toxicities of polycations. A challenge remains in the development of anti‐infectious coatings for the inert surfaces of biomedical devices that are prone to bacterial colonization and biofilm formation. Here, a facile photocuring method to construct functionalized polymeric coatings on inert polydimethylsiloxane (PDMS) surfaces, is developed. Using atom transfer radical polymerization (ATRP) initiator bearing thymol group, hydrophilic DMAEMA and benzophenone (BP)‐containing monomers are copolymerized to form polymers with end functional groups. An end‐functionalized biocidal coating is then constructed on the inert PDMS surface in one step using a photocuring reaction. The functionalized PDMS surfaces show excellent antibacterial and antifouling properties, are capable of completely eradiating MRSA within ≈6 h, and effectively inhibit the growth of biofilms. In addition, they have good stability and long‐lasting antibacterial activity in body fluid environments such as 0.9% saline and urine. According to bladder model experiments, the catheter's lifespan can be extended from ≈7 to 35 days by inhibiting the growth and migration of bacteria along its inner surface. The photocuring technique is therefore very promising in terms of surface functionalization of inert biomedical devices in order to minimize the spread of infection. [ABSTRACT FROM AUTHOR]

Species of springtails (Collembola) have a stratified distribution along the soil/air profile and present an opportunity to study adaptive traits relating to a gradient of fluctuating humidity. The habitat of eu-edaphic species (found in deeper litter and soil layers) secures a relatively stable humidity, whereas hemi-edaphic (found in upper litter layers) and epigeic springtails (found on soil surface or vegetation) can experience an increased range of moisture fluctuation with much more rapid shifts between extremes. We hypothesized that the capacity for acclimation to drought conditions is better developed in species associated with the soil surface than in soil-dwelling species. To test this hypothesis, we measured the osmotic response of two epigeic, two hemi-edaphic and three eu-edaphic species to mild drought acclimation (3 d at -1.5 MPa; the average wilting point of plants) and compared the drought tolerance of drought acclimated animals with controls. Epigeic species had no or very little osmotic regulation during mild drought acclimation and unexpectedly became less tolerant to subsequent acute drought stress than controls. The hemi-edaphic and eu-edaphic species significantly increased body fluid osmolality in response to mild drought acclimation. However, none of these species became more drought-tolerant after exposure to mild drought. In conclusion, we found that the osmotic response to drought acclimation was indeed related to the vertical position of a species in the soil/air profile, however, the relationship was opposite to our hypothesis. This study investigated the simulated body fluid-assisted stress corrosion cracking (SCC) of an Al-free magnesium alloy (RS66) and a common Al-containing magnesium alloy (AZ91), the former being more suitable for temporary implant applications (however, we used AZ91 for comparison since there are considerable reports on SCC in this alloy). The investigation includes SCC tests under simultaneous conditions of mechanical loading and imposed electrochemical potential that established a combined effect of hydrogen and anodic dissolution as the embrittlement mechanism. Though the RS66 alloy possesses impressive mechanical properties in non-corrosive environments (as a result of its fine grain size), both alloys suffered significant embrittlement when tested in simulated body fluid. The susceptibility of the RS66 alloy to SCC was ~25% greater than that of AZ91, which is attributed to the greater resistance of AZ91 to corrosion/localised corrosion because of its Al content.
