2. Isolation of laccase gene-specific sequences from white rot and brown rot fungi by PCR

Abstract

The genetic basis of fungal degradation of lignin by the enzyme laccase from white rot and brown rot fungi was investigated by polymerase chain reaction. Analysis of PCR-amplified products showed a DNA band of approximately 200 base pairs in six genera. A significant homology in amino acid sequences was seen in the PCR products of the white rot fungi Ganoderma lucidum, Phlebia brevispora and Trametes versicolor, but not in Lentinula edodes and Lentinus tigrinus. The nucleotide sequence similarity for the three genera of white rot fungi was 65 to 74% while that for the brown rot fungi Gloeophyllum trabeum was 60 to 71%.

Abstract

Laccase plate assays show that cultures of the Phanerochaete chrysosporium BKM-F1767 placed in a medium containing cellulose and ammonium tartrate produce extracellular laccase. Spectrophotometric assay and sodium dodecyl sulfate-polyacrylamide gel electrophoresis also show laccase activity in the concentrated extracellular culture medium. Induced laccase activity is observed in cultures containing cellulose but not in those containing glucose. Laccase is involved in lignin biodegradation.

Abstract

The construction of DNA probes specific for genes encoding vital lignin peroxide (LIP) isozymes H2, H8 and H10 of Phanerochaete chrysosporium, was conducted. These probes were employed to examine the temporal expression of the three lip genes in determined low-nitrogen mediums. H2 gene transcripts were synthesized in high levels on days 4, 5 and 7. Low levels of H2 gene transcripts were synthesized on day 6. No relationship was discerned between lip transcript and isozyme levels.

Abstract

Conventional continuous-time ?? modulators that use non-return-to-zero (NRZ) feedback DACs suffer from distortion due to intersymbol interference (ISI) and are sensitive to clock jitter. Using a return-to-zero (RZ) DAC solves the problem of ISI, but exacerbates clock jitter sensitivity. The clock jitter sensitivity of an NRZ DAC can be reduced using a switched-capacitor (SC) DAC, but the large peak-to-average ratio of the DAC waveform degrades modulator linearity. In this work, we introduce the switched-capacitor return-to-zero (SCRZ) DAC, which combines the low clock jitter sensitivity of the SC DAC with the low distortion of an RZ DAC. The efficacy of the SCRZ principle is borne out by measurement results from a modulator that achieves a DR/SNR/SNDR of 87.1/84.5/82.3 dB in a 2 MHz bandwidth while dissipating 16.5 mW from a 1.8-V supply. The converter, designed in a 0.18-? CMOS technology, reduces clock jitter sensitivity by 28 dB when compared with a traditional RZ DAC. � 1966-2012 IEEE.

14. Nitrogen regulation of lignin peroxidase and manganese-dependent peroxidase production is independent of carbon and manganese regulation in Phanerochaete chrysosporium.

Abstract

In this study, a N-deregulated mutant ( der8-5) of Phanerochaete chrysosporium was used as a tool to investigate the interrelationships between N, C, and Mn(II) regulation of LIP and MNP production in this organism. The results showed that LIP and MNP production by der8-5 was blocked in excess C medium but not in excess N medium. Furthermore, LIP and MNP production in this organism was subject to Mn(II) regulation regardless of the fact whether it is grown in low N medium or in high N medium. These and other results indicate that N regulation of LIP and MNP production in P. chrysosporium is independent of C and Mn(II) regulation. [ABSTRACT FROM AUTHOR]
