Abstract

Sentiment analysis approaches have offered a useful tool for decision support systems in various fields, including politics, network management, marketing, and healthcare. Owing to the increasing impact of online social networks on these fields and the fact that they are rich sources of information, the current sentiment analysis techniques in this scenario have evolved successfully. WhatsApp is a social network platform that enables users interact with close ties in a particular manner to communicate more meaningful, genuine, tangible, and personal information to the recipient, such as a sentiment. Hence, WhatsApp domain can be defined as a sub-language of the first language. However, only few studies have focused on WhatsApp sentiment analysis. These works usually employ outdated sentiment lexicon techniques and do not assess the most modern techniques based on deep learning. This study aims to evaluate this techniques for sentiment analysis based on deep neural networks and transfer learning, considering the intrinsic features of sub-language in WhatsApp domain. BERT1 and ALBERT1 (transfer learning approaches) achieve the best performance in accuracy and F1 (88% on average for both metrics and classifiers) similarly to other domains (Twitter). Although DCNN and LSTM with static embeddings usually achieve good performance when they are pre-trained on a larger corpus of other domains, these approaches reach the worst performance for WhatsApp domain. Furthermore, ELMo provides a good trade-off between the accuracy and training time complexity, mainly when taking into account the small size of our corpus training of WhatsApp. Hence, it can be inferred that the specific characteristics of the WhatsApp sub-language has an impact on the performance of some traditional SA classifiers

9. On noncompact warped product Ricci solitons.

Abstract

The goal of this paper is to investigate complete noncompact warped product gradient Ricci solitons. Nonexistence results, estimates for the warping function and for its gradient are proven. When the soliton is steady or expanding these nonexistence results generalize to a broader context certain estimates and rigidity obtained when studying warped product Einstein manifolds. When the soliton is shrinking, it is presented as a nonexistence theorem with no counterpart in the Einstein case, which is proved using properties of the first eigenvalue of a weighted Laplacian. Abstract

This work presents a flight path optimization model for drones that serve as a communication gateway or a data mule for IoT (Internet of Things) devices distributed on an Intelligent Campus. The results presented here consider a drone with limited energy autonomy (battery life), which causes the reduction of flight time and the number of covered devices in certain scenarios. This article proposes two approaches that ensure serving the maximum number of possible devices, considering the drones autonomy: the first using the Linear Programming, and the second through the Ant Colony Optimization Algorithm.

16. Establishment of sister chromatid cohesion during DNA replication in Saccharomyces cerevisiae

Abstract

Establishment of sister chromatid cohesion is a process thought to occur as the replication fork passes chromosomal loci bound by the cohesin complex. After fork passage, cohesin holds together pairs of replication products to allow their recognition by the mitotic machinery for segregation into daughter cells. In budding yeast, cohesin is loaded onto chromosomes during the G1 phase of the cell cycle. During S phase, the replication fork-associated acetyltransferase Eco1 acetylates the cohesin subunit Smc3 to promote the establishment of sister chromatid cohesion. At the time of anaphase, Smc3 loses its acetylation again, but the Smc3 deacetylase and the possible importance of Smc3 deacetylation were unknown. I show that the class I histone deacetylase family member Hos1 is responsible for Smc3 deacetylation. Cohesin is protected from deacetylation while bound to chromosomes but is deacetylated as soon as it dissociates from chromosomes at anaphase onset. Nonacetylated Smc3 is required as a substrate for cohesion establishment in the following cell cycle. Smc3 acetylation during DNA replication renders cohesin resistant against the cohesin destabiliser Wapl. However, because in the absence of Wapl cohesin acetylation is dispensable for cohesion establishment, I have turned my attention to additional ‘cohesion establishment factors’ replication fork-associated proteins required for efficient cohesion establishment. These include Chl1, Ctf4, Ctf18, Mrc1, Tof1 and Csm3. I have used genetic and molecular assays to investigate the relationship of these cohesion establishment factors with the cohesin acetylation pathway. This revealed a contribution of all of these factors to efficient cohesin acetylation. However, removal of the cohesin destabiliser Wapl corrected the cohesion defect in all of the cohesion establishment mutants, except Ctf4 and Chl1. Furthermore, my genetic analysis revealed pronounced synthetic interactions of these two factors with Eco1. Ctf4 and Chl1 therefore define a subset of Eco1-independent cohesion establishment factors, whose possible mechanism of action I have started to investigate.

Patients (N=260) at two tertiary public health centers, who were residents of the São Paulo metropolitan area, completed standardized questionnaires regarding their disease-related expenses. We used simple and multiple generalized linear models to assess the correlations between total cost and patient-related, as well as disease-related variables.Results: The total average annual cost of Parkinson’s disease was estimated at US$5,853.50 per person, including US$3,172.00 in direct costs (medical and nonmedical) and US$2,681.50 in indirect costs. Costs were directly correlated with disease severity (including the degree of motor symptoms), patients’ age, and time since disease onset.Conclusion: In this study, we determined the cost of Parkinson’s disease in Brazil and observed that disease-related motor symptoms are a significant component of the costs incurred on the public health system, patients, and society in general. Abstract

This paper addresses the urgent need to meet increasing energy demand while avoiding greenhouse gas emissions by improving energy efficiency. One significant challenge is the energy losses that occur during gas pressure reduction at city gates in natural gas distribution systems. To tackle this issue, this study proposes installing pressure reduction turbines (PRTs) parallel to existing reduction valves, which can generate electricity and enhance system efficiency. This research mainly focuses on Brazil’s natural gas supply, where the potential for installing PRTs is evaluated. The methodology analyzes the country’s distribution network and estimates the electricity generation capacity achievable from PRTs. The results indicate a potential power generation of 66 MW, capable of avoiding the emission of 235,800 tons of CO2 annually and generating 333 GWh/year of electricity, all at a LCOE of $27/MW. These contribute to the environment using the energy efficiency achieved through the installation of PRTs, consequently, Brazil can help to a cleaner and more sustainable energy future.

Abstract

Background: as gene-targeted therapies are increasingly being developed for Parkinson's disease (PD), identifying and characterizing carriers of specific genetic pathogenic variants is imperative. Only a small fraction of the estimated number of subjects with monogenic PD worldwide are currently represented in the literature and availability of clinical data and clinical trial-ready cohorts is limited. Objective: the objectives are to (1) establish an international cohort of affected and unaffected individuals with PD-linked variants; (2) provide harmonized and quality-controlled clinical characterization data for each included individual; and (3) further promote collaboration of researchers in the field of monogenic PD. Methods: we conducted a worldwide, systematic online survey to collect individual-level data on individuals with PD-linked variants in SNCA, LRRK2, VPS35, PRKN, PINK1, DJ-1, as well as selected pathogenic and risk variants in GBA and corresponding demographic, clinical, and genetic data. All registered cases underwent thorough quality checks, and pathogenicity scoring of the variants and genotype–phenotype relationships were analyzed. Results: we collected 3888 variant carriers for our analyses, reported by 92 centers (42 countries) worldwide. Of the included individuals, 3185 had a diagnosis of PD (ie, 1306 LRRK2, 115 SNCA, 23 VPS35, 429 PRKN, 75 PINK1, 13 DJ-1, and 1224 GBA) and 703 were unaffected (ie, 328 LRRK2, 32 SNCA, 3 VPS35, 1 PRKN, 1 PINK1, and 338 GBA). In total, we identified 269 different pathogenic variants; 1322 individuals in our cohort (34%) were indicated as not previously published. Conclusions: within the MJFF Global Genetic PD Study Group, we (1) established the largest international cohort of affected and unaffected individuals carrying PD-linked variants; (2) provide harmonized and quality-controlled clinical and genetic data for each included individual; (3) promote collaboration in the field of genetic PD with a view toward, Open Access funding enabled and organized by Projekt DEAL. Abstract

Cultural organizations, artists and other professionals, and their publics are today in the centre of the discussions on public investment in culture. The diversity of work profiles of the organizations and the new modes of arts participation encourage the analysis of “multiple sources” to characterize structures, contexts and publics. The article analyzes the support by the central state and evaluates how it operates in the capacity of artistic creation, divulgation and localized-action of structures. Using a case study of theatre, it considers the importance of concerted models of public support using organizations objectives, but also contexts and cultural territories specificities to reduce the overlap of old asymmetries and to amplify the action-axes from “local theatres”., As organizações culturais, os artistas e os seus públicos estão hoje no centro da discussão em torno dos investimentos na cultura. A diversidade de perfis de trabalho das organizações e os novos modos de participação nas artes exigem uma análise de múltiplas fontes para caracterizar estruturas, contextos e públicos. O artigo analisa o apoio público central e avalia como este opera na criação, divulgação e ação-localizada das organizações. Utiliza-se o caso do teatro e considera-se a importância de explorar modelos de apoio que aproveitem os objetivos das organizações, mas também os contextos e territórios culturais específicos para reduzir as velhas assimetrias e ampliar os eixos de atuação dos “teatros do bairro”.

Abstract

The pineal melatonin (N-acetyl-5-methoxytryptamine) is a molecule associated in a way or another with probably all physiological systems, aiming to fulfil its functional integrative roles in central nervous system activity, sleep and wakefulness cycles, energy metabolism and thermoregulation, immune, reproductive, endocrine, cardiovascular, respiratory and excretory systems. Within this context, the present study aimed to assess in silico the formation of complexes between ligand melatonin and other potential receptor proteins by molecular docking analyses. The main steps established in this experimental procedure were: a) search and selection of the 3D structure of the melatonin from DrugBank; b) search and selection of 3D structures of other target receptor proteins using STRING, protein BLAST and database PDB; and c) formation of the complexes between melatonin and receptors selected using AutoDock4.0 server by molecular docking analyses. High reliability score and significant similarity were only identified between type 1B melatonin and alpha-2A adrenergic receptor. Thus, molecular docking assays were carried out using ligand melatonin and crystallographic structures of the alpha-2A adrenergic receptor coupled to an antagonist (ID PDB 6kux) and a partial agonist (ID PDB 6kuy) available in the database PDB. Binding energy values of -6.79 and -6.98 kcal/mol and structural stability by non-covalent intermolecular interactions were predicted during the formation of complexes between melatonin and alpha-2A adrenergic receptor 6kux and 6kuy, respectively. In this way, the findings described in current study may indicate strong interactions between melatonin and adrenoceptors, suggesting its possible partial agonist effect on the activation of the alfa-2A adrenergic receptor. Resumo A melatonina pineal (N-acetil-5-metoxitriptamina) é uma molécula associada de um modo ou outro com provavelmente todos os sistemas fisiológicos, visando cumprir seus papéis funcionais integradores na atividade do sistema nervoso central, ciclos de sono e vigília, metabolismo energético e termorregulação, sistemas imunológico, reprodutivo, endócrino, cardiovascular, respiratório e excretor. Assim, o presente estudo objetivou avaliar in silico a formação de complexos entre o ligante melatonina e outras proteínas potenciais receptoras por meio de análises de docagem molecular. As principais etapas estabelecidas neste procedimento experimental foram: a) busca e seleção da estrutura 3D da melatonina a partir do banco de dados DrugBank; b) busca e seleção de estruturas 3D de outras proteínas receptoras-alvo utilizando STRING, proteína BLAST e o banco de dados PDB; e c) avaliação da formação dos complexos entre melatonina e receptores selecionados a partir do servidor AutoDock4.0 para análises de docagem molecular. Alto escore de confiabilidade e similaridade significativa foram identificados apenas entre a melatonina do tipo 1B e o receptor alfa-2A adrenérgico. Valores de energia de ligação de -6,79 e -6,98 kcal/mol e estabilidade estrutural pela presença de interações intermoleculares não covalentes foram preditos durante a formação de complexos entre o ligante melatonina e os receptores adrenérgico alfa-2A 6kux e 6kuy, respectivamente. Dessa forma, os achados descritos no presente estudo podem indicar fortes interações entre melatonina e adrenoceptores, sugerindo seu possível efeito agonista parcial na ativação do receptor alfa-2A adrenérgico.

Abstract

BACKGROUND: In the primary analysis of the HER2CLIMB trial, tucatinib added to trastuzumab and capecitabine significantly improved overall survival (OS) and progression-free survival (PFS) in patients with human epidermal growth factor receptor 2 positive (HER2+) metastatic breast cancer. We report efficacy and safety outcomes, including the final OS and safety outcomes from follow-up in HER2CLIMB. PATIENTS AND METHODS: HER2CLIMB is a randomized, double-blind, placebo-controlled trial in patients with locally advanced or metastatic HER2+ breast cancer, including patients with brain metastases. Patients were randomized 2 : 1 to receive tucatinib or placebo, in combination with trastuzumab and capecitabine. After the primary analysis (median follow-up of 14 months), the protocol was amended to allow for unblinding sites to treatment assignment and cross-over from the placebo combination to the tucatinib combination. Protocol prespecified descriptive analyses of OS, PFS (by investigator assessment), and safety were carried out at ∼2 years from the last patient randomized. RESULTS: Six hundred and twelve patients enrolled in the HER2CLIMB trial. At a median OS follow-up of 29.6 months, median duration of OS was 24.7 months for the tucatinib combination group versus 19.2 months for the placebo combination group [hazard ratio (HR) for death: 0.73, 95% confidence interval (CI): 0.59-0.90, P = 0.004] and OS at 2 years was 51% and 40%, respectively. HRs for OS across prespecified subgroups were consistent with the HR for the overall study population. Median duration of PFS was 7.6 months for the tucatinib combination group versus 4.9 months for the placebo combination group (HR for progression or death: 0.57, 95% CI: 0.47-0.70, P < 0.00001) and PFS at 1 year was 29% and 14%, respectively. Abstract

Since their proposal in 2016, the FAIR principles have been largely discussed by different communities and initiatives involved in the development of infrastructures to enhance support for data findability, accessibility, interoperability, and reuse. One of the challenges in implementing these principles lies in defining a well-delimited process with organized and detailed actions. This paper presents a workflow of actions that is being adopted in the VODAN BR pilot for generating FAIR (meta)data for COVID-19 research. It provides the understanding of each step of the process, establishing their contribution. In this work, we also evaluate potential tools to (semi)automatize (meta)data treatment whenever possible. Although defined for a particular use case, it is expected that this workflow can be applied for other epidemical research and in other domains, benefiting the entire scientific community., Preprint. Abstract

COVID study groups - PORTUGAL: Portuguese Laboratory Network for the Diagnosis of COVID-19 and Public Health Department of the Health Administrative Regions, Physicians that provided data and samples from suspected cases and SARS-CoV-2 genetic characterization. INSA laboratory team for the diagnosis of SARS-CoV-2. Algarve Biomedical Center and Unilabs. We compared 19,207 cases of SARS-CoV-2 variant B.1.1.7/S gene target failure (SGTF), 436 B.1.351 and 352 P.1 to non-variant cases reported by seven European countries. COVID-19 cases with these variants had significantly higher adjusted odds ratios for hospitalisation (B.1.1.7/SGTF: 1.7, 95% confidence interval (CI): 1.0-2.9; B.1.351: 3.6, 95% CI: 2.1-6.2; P.1: 2.6, 95% CI: 1.4-4.8) and B.1.1.7/SGTF and P.1 cases also for intensive care admission (B.1.1.7/SGTF: 2.3, 95% CI: 1.4-3.5; P.1: 2.2, 95% CI: 1.7-2.8). 48. Intraocular pressure fluctuation during resistance exercise

Abstract

OBJECTIVE: To evaluate the effect of weightlifting (leg press) on intraocular pressure (IOP). DESIGN: Prospective cohort study. SUBJECTS: A total of 24 participants met the inclusion criteria and completed the study procedures. Participants had an average age of 22.7±2.7 years and included nine women. The mean baseline IOP was 13.9 mm Hg (SD=2.4) with an average body mass index of 24.5 (SD= 3.1). METHODS: The maximum load for a single lift was found for each participant. Participants then performed three leg press regimens: one repetition using 95% of maximal load (1RM), six repetitions using 75% of maximal load (6RM) and isometric push against a weight much heavier than maximal load (ISO). MAIN OUTCOME MEASURE: IOP was measured pre-exercise, during and immediately following the exercise using an iCare TA01i rebound tonometer. Blood pressure and HR were being monitored continuously during the lift. Optical coherence tomography images were obtained pre and postexercise session. RESULTS: The average maximum weight lifted by our participants was 331.9 Kg (SD=97.3). Transient increased IOP was observed across the 1RM, 6RM and ISO exercises with an average increase in 26.4 mm Hg (23.7 mm Hg to 28.7 mm Hg) to reach an average max IOP of 40.7 mm Hg (27.8 mm Hg to 54.2 mm Hg), with an absolute maximum of 70 mm Hg in one participant. CONCLUSIONS: There is a transient and dramatic fluctuation in IOP with resistance training. This coupled with regular exposure to resistance training is potentially a significant risk factor for glaucoma. It should be noted that this study has been carried out in a healthy young population, and, thus, the external validity of these results in glaucoma participants requires further investigation.

Abstract

We compared 19,207 cases of SARS-CoV-2 variant B.1.1.7/S gene target failure (SGTF), 436 B.1.351 and 352 P.1 to non-variant cases reported by seven European countries. COVID-19 cases with these variants had significantly higher adjusted odds ratios for hospitalisation (B.1.1.7/SGTF: 1.7, 95% confidence interval (CI): 1.0-2.9; B.1.351: 3.6, 95% CI: 2.1-6.2; P.1: 2.6, 95% CI: 1.4-4.8) and B.1.1.7/SGTF and P.1 cases also for intensive care admission (B.1.1.7/SGTF: 2.3, 95% CI: 1.4-3.5; P.1: 2.2, 95% CI: 1.7-2.8).

Abstract

Late in 2020, two genetically-distinct clusters of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) with mutations of biological concern were reported, one in the United Kingdom and one in South Africa. Using a combination of data from routine surveillance, genomic sequencing and international travel we track the international dispersal of lineages B.1.1.7 and B.1.351 (variant 501Y-V2). We account for potential biases in genomic surveillance efforts by including passenger volumes from location of where the lineage was first reported, London and South Africa respectively. Using the software tool grinch (global report investigating novel coronavirus haplotypes), we track the international spread of lineages of concern with automated daily reports, Further, we have built a custom tracking website (cov-lineages.org/global_report.html) which hosts this daily report and will continue to include novel SARS-CoV-2 lineages of concern as they are detected.
