Members of the genus Paradileptus are apex predators in microbial food webs. They are often encountered in freshwater biotopes and have been used in research on water quality monitoring and ecology. Nevertheless, our understanding of the biodiversity of Paradileptus, especially its ecological and genetic diversities, is very poor which hinders our ability to understand the ecosystem services it provides. The present study gives a detailed account of two Chinese populations of Paradileptus elephantinus and P. conicus including their living morphology, infraciliature, and molecular phylogenies based on 18S, 5.8S, and ITS ribosomal DNA sequences. The phylogenetic relationships between these two species and other rhynchostomatians are investigated. We also explore the potential contribution of differentiation of the proboscis (e.g., extrusomes, dorsal brush, and differentiated kineties) to niche partitioning and speciation in Paradileptus. The global distribution of Paradileptus is summarized based on published data. Finally, a key to the identification of the valid species of Paradileptus is provided.

The morphology and phylogeny of two new sessilid species, Zoothamnium weishanicum n. sp. and Epicarchesium sinense n. sp., two insufficiently known species, Zoothamnium arbusculaEhrenberg, 1831 and Zoothamnium hentscheliKahl, 1935, and a well-known species, Carchesium polypinum (Linnaeus, 1767) Ehrenberg, 1838, collected from freshwater habitats of China, were investigated. Zoothamnium weishanicum n. sp. is characterized by its inverted bell-shaped zooids, double-layered peristomial lip, alternately branched stalk, and two different-length rows in infundibular polykinety 3 (P3). Epicarchesium sinense n. sp. is recognized by its asymmetric-pyriform zooids, single-layered peristomial lip, conspicuous cortical blisters on the pellicle, dichotomously branched stalk, and P3 containing one short inner row and two long outer rows. Based on previous and newly obtained data of the three known species, improved diagnoses and redescriptions are provided including, for the first time, data on the infraciliature of Z. arbuscula and Z. hentscheli. In addition, we analyzed the phylogeny of each species based on SSU rDNA sequence data.

4. Integrative Studies on a New Ciliate Campanella sinica n. sp. (Protista, Ciliophora, Peritrichia) Based on the Morphological and Molecular Data, With Notes on the Phylogeny and Systematics of the Family Epistylididae

During an investigation on freshwater peritrichs, a new colonial sessilid ciliate, Campanella sinica n. sp., was isolated from aquatic plants in an artificial freshwater pond in Qingdao, China. Specimen observations of this species were performed both in vivo and using silver staining. C. sinica n. sp. is characterized by the appearance of the mature colony, which is up to 2 cm high and contains more than 1,000 zooids, the asymmetric horn-shaped zooids, strongly everted and multi-layered peristomial lip, the slightly convex peristomial disc, and the well-developed haplokinety and polykinety, which make more than four circuits of the peristome before descending into the infundibulum. The small subunit ribosomal DNA (SSU rDNA), 5.8s rDNA and its flank internal transcribed spacers (ITS1-5.8s rDNA-ITS2), and large subunit ribosomal DNA (LSU rDNA) are sequenced and used for phylogenetic analyses which reveal that the family Epistylididae Kahl, 1933 is non-monophyletic whereas the genus Campanella is monophyletic and nests within the basal clade of the sessilids. The integrative results support the assertion that the genus Campanella represents a separate lineage from other epistylidids, suggesting a further revision of the family Epistylididae is needed. We revise Campanella including the transfer into this genus of a taxon formerly assigned to Epistylis, which we raise to species rank, i.e., Campanella ovata (Nenninger, 1948) n. grad. & n. comb. (original combination Epistylis purneri f. ovataNenninger, 1948). In addition, we provide a key to the identification of the species of Campanella.

Morphogenesis is an important process that widely occurs in almost all the organisms, including the ciliated protists. Ciliates are a large group of single-celled eukaryotes that can reproduce asexually (e.g., binary fission) and perform sexual process (e.g., conjugation). Morphogenesis happens in both asexual reproduction and sexual process in ciliates and the reorganization during conjugation is more complex. However, studies of morphogenesis focusing on conjugation are very limited. Here we studied the morphogenetic process during conjugation in the marine species Euplotes raikovi Agamaliev, 1966. The results indicate that: (1) the ciliature in the ventral side reorganizes twice during sexual process, i.e., conjugational and postconjugational reorganization; (2) the adoral zone of membranelles (AZM) is generated de novo in a pouch beneath the cortex during both reorganizations, with the anterior part generated during the first reorganization, while the posterior part formed during the second reorganization; (3) the frontoventral-transverse (FVT) cirri anlagen are formed de novo in both processes with the fragmentation pattern of 2:2:3:3:2; (4) one left marginal cirrus is generated de novo during both reorganizations; and (5) the dorsal ciliature remains intact during the whole process, except that the two caudal cirri originate from the end of the right-most two dorsal kineties during both reorganizations. Comparisons of the morphogenetic process during conjugation demonstrate a considerably stable pattern within Euplotes while the patterns vary dramatically among different ciliate groups.

6. Single-Cell Genomic Sequencing of Three Peritrichs (Protista, Ciliophora) Reveals Less Biased Stop Codon Usage and More Prevalent Programmed Ribosomal Frameshifting Than in Other Ciliates

Peritrichs are one of the largest groups of ciliates with over 1,000 species described so far. However, their genomic features are largely unknown. By single-cell genomic sequencing, we acquired the genomic data of three sessilid peritrichs (Cothurnia ceramicola, Vaginicola sp., and Zoothamnium sp. 2). Using genomic data from another 53 ciliates including 14 peritrichs, we reconstructed their evolutionary relationships and confirmed genome skimming as an efficient approach for expanding sampling. In addition, we profiled the stop codon usage and programmed ribosomal frameshifting (PRF) events in peritrichs for the first time. Our analysis reveals no evidence of stop codon reassignment for peritrichs, but they have prevalent +1 or -1 PRF events. These genomic features are distinguishable from other ciliates, and our observations suggest a unique evolutionary strategy for peritrichs.

The ciliate genus Pleuronema comprises approximately 30 nominal species and has been reported in freshwater, brackish water, and marine habitats. Nevertheless, recent studies have indicated that there might be a large undiscovered species diversity. In the present work, four new Pleuronema species, namely P. foissneri sp. nov., P. parasmalli sp. nov., P. parasalmastra sp. nov., and P. paraorientale sp. nov., collected from Shenzhen, southern China, was investigated using taxonomic methods. The diagnosis, description, comparisons with morphologically related species and detailed morphometric data are supplied for each. The small subunit ribosomal RNA (SSU rRNA) gene of the four new species is sequenced and their molecular phylogeny is analyzed. The SSU rRNA gene tree shows that Pleuronema is polyphyletic comprising several separate clades. All four new species cluster consistently with P. orientale KF206429, P. puytoraci KF840520 and P. setigerum FJ848874 within the core Pleuronematidae + Peniculistomatidae clade. Phylogenies of Pleuronematidae-related taxa are also discussed.

Three Diophrys species, D. quadrinucleata n. sp., D. oligothrix Borror, 1965 and D. scutum (Dujardin, 1841) Kahl, 1932, were identified from the subtropical brackish waters in China, their morphology was investigated using live observation and protargol impregnation. D. quadrinucleata n. sp. can be distinguished from the congeners by four ellipsoidal macronuclear nodules. Phylogenetic analyses show that the three species with single marginal cirrus are closely related, namely D. quadrinucleata n. sp., D. apoligothrix Song et al., 2009 and D. japonica Hu, 2008. Our population of D. oligothrix corresponds very well with previous populations in both living morphology and ciliature. However, the SSU rDNA sequences of three well-described populations differ from each other by 11 to 18 nucleotides, indicating that there might be cryptic species in D. oligothrix. Moreover, we provided a detailed description of the symbiotic bacteria/archaeobacteria (?) concentrated on the back surface of D. scutum based on the present population, which was either overlooked or misinterpreted as cortical granules previously.

The peritrichous genus Ophrydium is relatively distinctive since its colonies have a gelatinous lorica. In the present work, one morphologically well-described species, Ophrydium crassicaule, was collected from a freshwater habitat in China. Both morphological characters and sequence data for SSU rDNA, ITS1-5.8S-ITS2 region and LSU rDNA were obtained. Morphologically, O. crassicaule is characterized by its elongated vase-shaped zooid, single-layered peristomial lip, dichotomously branched stalk, colony with gelatinous lorica, and infundibular polykinety 3 (P3) containing one short inner row and two long outer rows. Phylogenetic analyses revealed that the genus Ophrydium is monophyletic and nests within the family Vorticellidae. In addition, we briefly revise the nominal congeners for aiding the species identification in future studies.

Two colonial sessilid peritrichs, Epistylis qingdaoensis n. sp. and Carchesium cyclopidarum Nenninger, 1948, were isolated from a marine crustacean and a freshwater mayfly, respectively. Morphological characters for each species were revealed by in vivo observations and silver staining methods. Epistylis qingdaoensis n. sp. is characterized by the apperance of the colony which is up to 250 μm high and usually contains fewer than eight zooids, the single-layered peristomial lip, the conspicuously conical peristomial disc, and the structure of infundibular polykinety 3 which comprises three isometric ciliary rows. Carchesium cyclopidarum is recognized by the colony height of about 600 μm, the small zooid size of 35-50 × 20-30 μm, and the low number of silverlines. Phylogenetic analyses based on small subunit ribosomal DNA sequences were performed to reveal their evolutionary relationships. Surprisingly, neither species clustered with its congeners. Epistylis qingdaoensis n. sp. nested within a clade of Zoothamnium species that was sister to a clade comprising core Epistylis species and several Zoothamnium species. Carchesium cyclopidarum nested within a clade comprising solitary peritrichs and was distantly related to the type species of Carchesium, C. polypinum. These findings imply for the first time that Carchesium is non-monophyletic.

The morphology and ciliature of a new freshwater ciliate, Frontonia apoacuminata sp. nov., isolated from an artificial pond in Qingdao, PR China, were investigated using live observation and silver staining methods. The main features separating F. apoacuminata sp. nov. from its congeners are as follows: a broad elliptical body that is slightly pointed at the posterior end, four ophryokineties, one or two spherical micronuclei of a ‘compact’ type, a dorsally positioned contractile vacuole, and peniculi 1–3 each with five kinetosome rows though the left-most two rows in peniculus 3 are extremely shortened (with only two or three kinetosomes each). Additionally, an improved diagnosis of F. acuminata is provided. Phylogenetic analyses based on the small subunit ribosomal RNA (SSU rRNA) gene show that F. apoacuminata sp. nov. clusters with F. atra, F. minuta, F. acuminata and F. terricola. These five species group with Disematostoma, Marituja and Stokesia rather than with other Frontonia species, causing polyphyly of the genus Frontonia.

Three epibiotic Epistylis species, i.e., Epistylis weishanensis sp. nov., Epistylis daphniae Fauré-Fremiet, 1905, and Epistylis pygmaeum (Ehrenberg, 1838) Foissner et al., 1999, were investigated based on their living morphology, infraciliature, and small subunit (SSU) rDNA sequence data. Epistylis weishanensis sp. nov. is characterized by its double-layered peristomial lip, contractile vacuole located on the dorsal wall of the infundibulum, infundibular polykinety 3 (P3) composed of three equal-length rows that terminate above infundibular polykinety 1 (P1), 50-62 silverlines between the peristome and the trochal band, and about 30 silverlines between the trochal band and the scopula. Based on previous and newly obtained data for E. daphniae and E. pygmaeum, improved diagnoses and redescriptions are provided including, for the first time, data on their infraciliature. Phylogenetic analyses reveal that all three species do not group within the major clade of Epistylis, supporting the assertion that the genus Epistylis should be an assemblage of morphospecies and therefore needs to be revised.

Ampullofolliculina Hadži, 1951 is a monotypic genus, the only species being A. lageniformis which was first discovered in estuarine habitats in the U.S. and has never been redescribed. In the present study, we investigated the taxonomy and phylogeny of A. lageniformis Hadži, 1951 based on analyses of a population collected from a brackish wetland in Ningbo, China. The main characteristics of this species are as follows: trophont about 450-700 μm long in vivo with two short, rounded peristomial lobes of equal size; lorica flask-shaped, transparent and smooth with a short, wide neck at the base of which two transparent valves are asymmetrically inserted; cortex with brownish to reddish cortical granules and greenish pigment granules; about 80 somatic kineties evenly arranged; moniliform macronucleus with 4-8 ellipsoidal nodules; swarmer dark green, vermiform in shape, about 200-350 μm long in vivo, with about 60 adoral membranelles and 85 somatic kineties, no mouth nor paroral membrane. Phylogenetic analyses inferred from SSU rDNA sequences show that A. lageniformis is closely related to Folliculina and Eufolliculina which nest within a large clade that comprises five families, i.e. Stentoridae, Blepharismidae, Fabreidae, Maristentoridae, and Folliculinidae.

A novel polyoxometalate-based metal–organic framework templated by twin-Dawson clusters, [{Cu3(μ3-O)}2(trz)6Cu2(H2O)13][H1.73P2As1.73W16.27O62]·8.25H2O (1) (trz = 1,2,4-triazole), has been synthesized under hydrothermal conditions. In 1, there are two crystal distinct motifs: a 3D metal–organic nanotube framework and seven-connected Dawson clusters. It is worth mentioning that the 3D framework possesses nanotube-like channels. The twin-H1.73P2As1.73W16.27O62 clusters (abbreviated as P2(As/W)18 clusters) as templates occupy channels of the nanotube framework. To the best of our knowledge, this represents the first metal–organic nanotube framework templated by twin-Dawson clusters. The electrochemical experiments indicate that the 1-based glassy carbon electrode (1-GCE) possesses high catalytic efficiency and high stability toward reduction of inorganic bromate molecules and oxidation of the biological molecule ascorbic acid. The electrocatalytic efficiency towards the reduction of bromate in 1 M H2SO4 solution and oxidation of AA in N2 purged solution is ca. 848.4% and 896.8% (catalytic substrate: 0.5 mM), respectively. The current signal after 100 cycles exhibits almost no loss for 1-GCE.

Morphogenesis is an important process that widely occurs in almost all the organisms, including the ciliated protists. Ciliates are a large group of single-celled eukaryotes that can reproduce asexually (e.g., binary fission) and perform sexual process (e.g., conjugation). Morphogenesis happens in both asexual reproduction and sexual process in ciliates and the reorganization during conjugation is more complex. However, studies of morphogenesis focusing on conjugation are very limited. Here we studied the morphogenetic process during conjugation in the marine species Euplotes raikovi Agamaliev, 1966. The results indicate that: (1) the ciliature in the ventral side reorganizes twice during sexual process, i.e., conjugational and postconjugational reorganization; (2) the adoral zone of membranelles (AZM) is generated de novo in a pouch beneath the cortex during both reorganizations, with the anterior part generated during the first reorganization, while the posterior part formed during the second reorganization; (3) the frontoventral-transverse (FVT) cirri anlagen are formed de novo in both processes with the fragmentation pattern of 2:2:3:3:2; (4) one left marginal cirrus is generated de novo during both reorganizations; and (5) the dorsal ciliature remains intact during the whole process, except that the two caudal cirri originate from the end of the right-most two dorsal kineties during both reorganizations. Comparisons of the morphogenetic process during conjugation demonstrate a considerably stable pattern within Euplotes while the patterns vary dramatically among different ciliate groups.

The class Oligohymenophorea is one of the most diverse assemblage of ciliated protists, which are particularly important in fundamental biological studies including understanding the evolutionary relationships among the lineages. Phylogenetic relationships within the class remain largely elusive, especially within the subclass Peniculia, which contains the long-standing problematic taxa Urocentrum and Paranassula. In the present study, we sequenced the genomes and/or transcriptomes of six non-culturable oligohymenophoreans using single-cell sequencing techniques. Phylogenomic analysis was performed based on expanded taxon sampling of 85 taxa, including 157 nuclear genes encoding 36,953 amino acids. The results indicate that: (1) urocentrids form an independent branch that is sister to the clade formed by Scuticociliatia and Hymenostomatia, which, together with the morphological data, supports the establishment of a new subclass, Urocentria n. subcl., within Oligohymenophorea; (2) phylogenomic analysis and ortholog comparison reveal a close relationship between Paranassula and peniculines, providing corroborative evidence for removing Paranassula from Nassulida and elevating it as an order, Paranassulida, within the subclass Peniculia; (3) based on the phylogenomic analyses and morphological data, we hypothesize that Peritrichia is the earliest diverging clade within Oligohymenophorea while Scuticociliatia and Hymenostomatia share the most common ancestor, followed successively by Urocentria and Peniculia. In addition, stop codon analyses indicate that oligohymenophoreans widely use UGA as the stop codon, while UAR are reassigned to glutamate (peritrichs) or glutamine (others), supporting the evolutionary hypothesis.

The peritrich genus Epistylis is speciose, however many species lack complete morphological description based on modern criteria and/or molecular data. In the present study, one new species, i.e., E. foissneri n. sp., and two morphologically similar species, i.e., E. hentscheli Kahl, 1935 and E. vaginula Stokes, 1884, collected from freshwater habitats in China, were studied. Epistylis foissneri n. sp. is characterized by its extremely slender zooids encased in a gelatinous sheath, symmetrically dichotomously branched stalk, trochal band located at mid-body, contractile vacuole located on dorsal wall of infundibulum, infundibular polykinety 3 (P3) composed of three equal-length rows that terminate above infundibular polykinety 1 (P1), 105-110 silverlines between the peristome and the trochal band, and about 110 silverlines between the trochal band and the scopula. Epistylis hentscheli is characterized by its asymmetric pyriform zooids (average length ca. 160 μm in vivo), dichotomously branched stalk with transverse striations on the surface of the upper portion, P3 three-rowed and terminating slightly above P1, 60-75 silverlines between the peristome and the trochal band, and 55-90 silverlines between the trochal band and the scopula. Epistylis vaginula is characterized by its elongated body shape (about 100 μm in length in vivo), dichotomously branched and smooth stalk, P3 three-rowed and terminating above P1, 80-100 silverlines between the peristome and the trochal band, and 45-80 silverlines between the trochal band and the scopula. The small subunit ribosomal DNA gene (SSU rDNA) of these three species was sequenced and supported the validity of each. Phylogenetic analyses based on SSU rDNA sequence data revealed that all three morphospecies group with other congeners within the major clade of Epistylis.

Zoothamnium is a speciose genus, most species of which have incomplete morphological data based on modern criteria. In the present study, the morphology of three species of Zoothamnium, i.e., Z. apoarbuscula n. sp., Z. apohentscheli n. sp., and Z. alternans, collected in Qingdao, China, was revealed using living observation and silver staining. In addition, the SSU rDNA of each species was sequenced for phylogenetic analyses. Zoothamnium apoarbuscula n. sp. is characterized by its umbellate colony which is up to 900 μm high, dichotomously branched stalk, differentiation of zooids, and infundibular polykinety 3 comprising three equal-length ciliary rows. Zoothamnium apohentscheli n. sp is characterized by its large colony up to 1700 μm high, alternately branched stalk, and infundibular polykinety 3 comprising three equal-length ciliary rows. A population of Z. alternans is described in detail. Phylogenetic analyses based on SSU rDNA sequence data revealed that species with an alternately branched stalk cluster together in gene trees and probably represent an independent lineage within the genus Zoothamnium.

Four epibiotic sessilid peritrichs, i.e., Zoothamnium wilberti n. sp., Baikalonis microdiscus n. sp., Epistylis anastatica (Linnaeus, 1767) Ehrenberg, 1830, and Rhabdostyla commensalisMöbius, 1888, were isolated from one syllid polychaete and three crustacean hosts in Qingdao, China. For each species, specimens were observed both in vivo and following silver staining. Their SSU rDNA was also sequenced for phylogenetic analyses. Zoothamnium wilberti n. sp. is characterized by the appearance of its colony, which is up to 350 μm high, and usually has fewer than 16 zooids, and the dichotomously branched stalk with transverse wrinkles, the conspicuously conical peristomial disc, and infundibular polykinety 3 comprising three isometric ciliary rows. Baikalonis microdiscus n. sp. can be recognized by its barrel-shaped zooid, small peristomial disc, smooth and short stalk, and its unusual infundibular polykinety 3 comprising a long inner row and a short outer row. Two poorly known species, i.e., Epistylis anastatica and Rhabdostyla commensalis, are redescribed and redefined. Phylogenetic analyses reveal that: (i) R. commensalis is closely related to the family Astylozoidae rather than to the morphologically similar Epistylididae; (ii) B. microdiscus n. sp. is sister to the family Scyphidiidae; (iii) E. anastatica groups with vorticellids and ophrydiids, which further supports the polyphyly of the genus Epistylis; and (iv) Z. wilberti n. sp. is nested within the Zoothamniidae, as expected.

During an investigation of parasitic ciliates in northern China, two Trichodina species, T. acuta Lom, 1970 and T. nigra Lom, 1960, were isolated from the freshwater fish Cyprinus carpio Linnaeus, 1758. The morphology of each species was investigated based on dry silver nitrate-stained specimens. In addition, the molecular phylogeny of each was analyzed based on small subunit ribosomal DNA (SSU rDNA) sequence data. Trichodina acuta can be distinguished from its congeners by the undefined periphery of the central circle, the distinct gap between the rays and the central circle, and the distinctly sickle-shaped blades. Trichodina nigra is a cosmopolitan ciliate and is characterized by its densely linked denticles, broad, rounded spatula-shaped blades, robust central parts, and well developed rays. Phylogenetic analyses revealed that T. acuta and T. nigra nest within different clades, supporting the assertion that the GC content of SSU rDNA sequences could reflect evolutionary relationships among Trichodina species.

Miamiensis avidus Thompson & Moewus, 1964, is a cosmopolitan and well-known marine pathogenic ciliated protist. However, the taxonomy of this species up to now has remained controversial, especially with respect to the validity of the morphologically similar species, Philasterides dicentrarchi, which was considered as a junior synonym of M. avidus. In this study, a population of M. avidus was collected from the skin of pharaoh cuttlefish (Sepia pharaonis) cultured near the East China Sea, Ningbo, China and its morphology and phylogeny were investigated in detail based on living characters, infraciliature, small subunit (SSU) rDNA and ITS1-5.8S-ITS2 region sequences. In addition, the morphometrics of a previously reported free-living population, collected from the Bohai Sea, were rechecked and analyzed. We compared the present two isolates with all historic populations of M. avidus and P. dicentrarchi, and found that their morphological characters were either highly similar or exactly identical, indicating that they are the same morphospecies. However, the phylogenetic analyses based on SSU rDNA or ITS1-5.8S-ITS2 region sequences revealed that most M. avidus and P. dicentrarchi populations formed one clade, and the two isolates of M. avidus from Weifang and American Type Culture Collection clustered in another clade, which indicated that there might be cryptic species in Miamiensis avidus.

A new polyoxovanadate-based metal–organic framework, [Cu(Pz)](V4O10) (1) (Pz = Pyrazine) has been synthesized under hydrothermal conditions. Single crystal X-ray diffraction analysis reveals that compound 1 displays a 3D framework with a rare reduced V4O102− layer. The electrochemical study shows that 1 exhibits bifunctional electrocatalytic activities toward reduction of iodate and oxidation of ascorbic acid. Surface photovoltage spectroscopy (SPS) and electric field induced surface photovoltage spectroscopy (EFISPS) measurements reveal that 1 has the behavior of an n-type semiconductor. In addition, 1 shows excellent photocatalytic activity for the decomposition of Rhodamine-B dye under visible light.

31. Recovery of ciliated protozoan communities in response to environmental change in a shrimp-farming pond in southern China

The temporal dynamics of ciliate community structure in a southern Chinese shrimp aquaculture facility were investigated during the period June–September 2012. A total of 53 species belonging to 37 genera and 17 orders were recorded based on analyses of eight samples. Ciliate abundance peaked between 16 August and 14 September 2012, while the maximum number of species occurred on 26 June 2012. Clear temporal patterns were observed in the ciliate community structure. The patterns of succession of the 10 most abundant species were consistent with the results of a Canonical Analysis of Principal coordinates (CAP) analysis. Correlation analyses showed that these patterns of succession were related to temporal changes in environmental variables. In summary, the results demonstrate that the ciliate community responds predictably to environmental variations and recovers from shrimp cultivation.

Ciliates have a long history of being central in evolutionary and ecological studies on eukaryotic microorganisms. Although thousands of species have been discovered, their total diversity still remains unknown. Here, we will discuss two unsolved problems that hinder the further exploration of ciliate diversity at the species level, and potential solutions to these problems are proposed. First, ciliate morphospecies are difficult to identify because the different silver stains are not scalable (they do not represent high-throughput methods) and basic supplies are lacking (e.g., protargol); a solution may be the development of fluorescent staining techniques. Second, ciliate phylogenetic species are difficult to identify because of extensive paralogy in nuclear-protein-coding genes; a solution may be to concentrate on sequencing mitochondrial genomes. These two approaches could be integrated into a high-throughput fluorescent-single-cell sorting and mitochondrial genomes sequencing process that would enable the observation and better understanding of ciliate species on a massive scale. Two new polyoxometalate-based coordination polymers have been synthesized at different pHs under identical hydrothermal conditions, (Hbib)[Cu2(bib)2(PMo12O40)] (1) and (Hbib)2[Cu(bib) (PMo12O40)]·2H2O (2) (bib = 4-bis(imidazol-1-yl) benzene). Their structures were determined by single crystal X-ray diffraction analyses and characterized by routine methods. When the pH was adjusted to 2.0 − 2.5, 1 was obtained, which exhibits a 1-D rail-like chain. Compound 2 was obtained at pH of 2.5 − 3.2 and exhibits a 2-D layer with (44) sql net. The pH of the reaction plays a key role in the assembly of the polyoxometalate-based coordination polymer. The electrochemical experiments indicate that 2-based carbon paste electrode possesses high catalytic efficiency and high stability towards reduction of iodate and oxidation of ascorbic acid.

Members of the genus Thuricola are a highly specialized group of peritrichous ciliates possessing a protective barrel-shaped lorica. The genus presents many difficulties in terms of species separation and definition, and in this context the present study investigates three species by protargol staining and analyses of SSU rDNA sequences. Based on their morphologic characteristics and biotope, they were identified as three poorly known forms in Thuricola, namely T. obconica Kahl, 1933, T. kellicottiana (Stokes, 1887) Kahl 1935 and T. folliculata Kent, 1881, respectively. T. obconica is characterized by possessing curved lorica and a single valve in vivo. T. kellicottiana is distinguished by two valves with a spine on the main valve, and a generally long internal stalk upon which the zooids sit. T. folliculata also has two valves but lacks a spine. The ciliature of the three species are basically similar. A novel polyoxometalate-encapsulating polycatenated metal-organic framework, (bib)0.5[Cu2(bib)3.5(GeW12O40)]·H2O (1) (bib = 1,4-bis(1-imidazol-yl)-2,5-dimethyl benzene), has been synthesized under hydrothermal conditions. In compound 1, 1D molecular ladders and 2D square grid layers is further catenated to the two adjacent such motifs in inclined fashion to give an 1D+2D→3D polycatenaned metal-organic framework with nanotube channels, in which nanosized Keggin anions as guests are encapsulated for the first time. Compound 1 represents a first case of Keggin-type polyoxometalate-encapsulating 1D+2D→3D polycatenated metal-organic framework. The electrochemical studies show that modified electrode of 1 exhibits highly efficient electrocatalytic activity toward reduction of inorganic molecule bromate (BrO3-). The electrocatalytic reaction mechanism of 1-GCE toward reduction of BrO3- has been illuminated. The photoluminescence property of 1 was also investigated.

The morphology and phylogeny of two hypotrichous ciliates, Metaurostylopsis parastruederkypkeae n. sp. and Neourostylopsis flavicana (Wang et al., 2011) Chen et al., 2013 were investigated based on morphology, infraciliature and the small subunit (SSU) ribosomal RNA gene (rRNA) sequence. The new species, M. parastruederkypkeae n. sp. was identified according to its characteristics: body shape ellipsoidal, size about (165–200) × (45–60) μm in vivo, cell color reddish; two types of cortical granules including wheat grain-like and yellow-greenish larger ones along the marginal cirri rows and dorsal kineties and dot-like and reddish smaller ones, grouped around marginal cirri on ventral side and arranged in short lines on dorsal side; 26–41 adoral membranelles; three frontal and one parabuccal, five to seven frontoterminal, one buccal, and three to six transverse cirri; seven to thirteen midventral pairs; five to nine unpaired ventral cirri, five to seven left and three to five right marginal rows; and three complete dorsal kineties. Phylogenetic analysis based on SSU rDNA sequences showed that both Metaurostylopsis and Neourostylopsis are monophyletic. As the internal relationship between and within both genera are not clear, further studies on the species in these two genera are necessary. The key characteristics of all known twelve Metaurostylopsis-Apourostylopsis-Neourostylopsis species complex were updated.

Members of the genus Thuricola are a highly specialized group of peritrichous ciliates possessing a protective barrel-shaped lorica. The genus presents many difficulties in terms of species separation and definition, and in this context the present study investigates three species by protargol staining and analyses of SSU rDNA sequences. Based on their morphologic characteristics and biotope, they were identified as three poorly known forms in Thuricola, namely T. obconica Kahl, 1933, T. kellicottiana (Stokes, 1887) Kahl 1935 and T. folliculata Kent, 1881, respectively. T. obconica is characterized by possessing curved lorica and a single valve in vivo. T. kellicottiana is distinguished by two valves with a spine on the main valve, and a generally long internal stalk upon which the zooids sit. T. folliculata also has two valves but lacks a spine. The ciliature of the three species are basically similar. The morphology and infraciliature of a poorly-known colepid ciliate, Pinacocoleps tesselatus (Kahl, 1930) Foissner et al ., 2008, collected from brackish-water biotope (salinity 12 ‰) in Hangzhou Bay, China, were investigated using live obser-vations and silver impregnations. This species is characterized by its length of 60–85 μm in vivo , ovular shape, two ante-rior and three posterior spines, 21–25 longitudinal and 11 transverse ciliary rows on average, a macro and micro nucleus, and one terminal contractile vacuole. The key to all known seven Pinacocoleps species is updated. Additionally, we char-acterized the taxon P. tesselatus via small subunit rRNA gene data. Our phylogenetic analyses performed using both max-imum-likelihood and Bayesian methods indicate that P. tesselatus falls into the core assemblage of the family Colepidae.
