The construction of highways leads to several environmental and landscape impacts, including the fragmentation of natural habitats for many animal species. Highway projects are therefore generally accompanied by mobilisations from the inhabitants of the areas concerned and environmental associations. This work aims to model the potential impacts of a highway project in France on ecological networks and to study the reception of the results by the opponents of this project. We have adopted a three-step approach. First, a land-cover map of the study area was produced at a fine scale of 10 m resolution. Second, we developed a multi-species approach by defining fifteen species groups representative of different habitats of our study area. Third, the design of landscape graphs and the resulting calculation of connectivity metrics allowed mapping the impact of the highway on multi-species ecological connectivity. Reflexive feedback from comments on these results by the public during a mobilisation day against the highway project allows assessment of the relevance of such a modelling approach in this context.

In the domain of heavy to extra heavy oil production, viscous polymer may be injected after water injection (tertiary mode), or as an alternative (secondary mode) to improve the sweep efficiency and increase oil recovery. To prepare field implementation, nine polymer injection experiments in heavy oil have been performed at core scale, to assess key modelling parameters in both situations. Among this consistent set of experiments, two have been performed on reconstituted cylindrical sandpacks in field-like conditions, and seven on consolidated Bentheimer sandstone in laboratory conditions. All experiments target the same oil viscosity, between 2000 cP and 7000 cP, and the viscosity of Partially Hydrolyzed Polyacrylamide solutions (HPAM 3630) ranges from 60 cP to 80 cP. Water and polymer front propagation are studied using X-ray and tracer measurements. The new experimental results presented here for water flood and polymer flood experiments are compared with experiments described in previous papers. The effects of geometry, viscosity ratio, injection sequence on recoveries, and history match parameters are investigated. Relative permeabilities of the water flood experiment are in line with previous experiments in linear geometry. Initial water floods led to recoveries of 15–30% after one Pore Volume Injected (PVI), a variation influenced by boundary conditions, viscosity, and velocities. The secondary polymer flood in consolidated sandstone confirms less stable displacement than tertiary floods in same conditions. Comparison of secondary and tertiary polymer floods history matching parameters suggests two mechanisms. First, hysteresis effect during oil bank mobilization stabilizes the tertiary polymer front; secondly, the propagation of polymer at higher oil saturation leads to lower adsorption during secondary experiment, generating a lower Residual Resistance Factor (RRF), close to unity. Finally, this paper discusses the use of the relative permeabilities and polymer properties estimated using Darcy equation for field simulation, depending on water distribution at polymer injection start-up.

In this paper, we consider adaptive numerical simulation of miscible displacement problems in porous media, which are modeled by single phase flow equations. A vertex-centred finite volume method is employed to discretize the coupled system: the Darcy flow equation and the diffusion-convection concentration equation. The convection term is approximated with a Godunov scheme over the dual finite volume mesh, whereas the diffusion-dispersion term is discretized by piecewise linear conforming finite elements. We introduce two kinds of indicators, both of them of residual type. The first one is related to time discretization and is local with respect to the time discretization: thus, at each time, it provides an appropriate information for the choice of the next time step. The second is related to space discretization and is local with respect to both the time and space variable and the idea is that at each time it is an efficient tool for mesh adaptivity. An error estimation procedure evaluates where additional refinement is needed and grid generation procedures dynamically create or remove fine-grid patches as resolution requirements change. The method was implemented in the software MELODIE, developed by the French Institute for Radiological Protection and Nuclear Safety (IRSN, Institut de Radioprotection et de Sûreté Nucléaire). The algorithm is then used to simulate the evolution of radionuclide migration from the waste packages through a heterogeneous disposal, demonstrating its capability to capture complex behavior of the resulting flow.

Gene expression associated with West Nile virus (WNV) infection was profiled in the central nervous system of horses. Pyrosequencing and library annotation was performed on pooled RNA from the CNS and lymphoid tissues on horses experimentally infected with WNV (vaccinated and naïve) and non-exposed controls. These sequences were used to create a custom microarray enriched for neurological and immunological sequences to quantitate gene expression in the thalamus and cerebrum of three experimentally infected groups of horses (naïve/WNV exposed, vaccinated/WNV exposed, and normal).From the sequenced transcriptome, 41,040 sequences were identified by alignment against five databases. 31,357 good sequence hits (e<10(-4)) were obtained with 3.1% of the sequences novel to the equine genome project. Sequences were compared to human expressed sequence tag database, with 31,473 equine sequences aligning to human sequences (69.27% contigs, 78.13% seed contigs, 80.17% singlets). This indicated a high degree of sequence homology between human and equine transcriptome (average identity 90.17%).Significant differences (p<0.05) in gene expression were seen due to virus exposure (9,020), survival (7,395), and location (7,649). Pathways analysis revealed many genes that mapped to neurological and immunological categories. Involvement of both innate and adaptive components of immunity was seen, with higher levels of expression correlating with survival. This was highlighted by increased expression of suppressor of cytokine signaling 3 in horses exposed to WNV which functions to suppress innate immunity. Pentraxin 3 was most increased in expression for all horses exposed to WNV.Neurological pathways that demonstrated the greatest changes in gene expression included neurotransmitter and signaling pathways. Decreased expression of transcripts in both the glutamate and dopamine signaling pathways was seen in horses exposed to WNV, providing evidence of possible glutamate excitotoxicity and clinical signs associated with decreased dopamine. Many transcripts mapped to non-infectious neurological disease functions, including mental disorders and degenerative neuropathies.

We used a severe challenge model that produces clinical West Nile virus (WNV) disease to test the efficacy of three commercially available equine WNV vaccines in horses. Twenty-four healthy, WNV-seronegative horses of varying ages and genders were placed, in random and blind manner, into three trial groups consisting of eight horses each; two horses in each group received (i) an inactivated WNV vaccine (K-WN), (ii) a modified-live vaccine (CP-WN) containing the WNV prM and E proteins expressed by a canarypox vector, (iii) a live-chimera vaccine (WN-FV) containing WNV prM and E proteins expressed in a YF17D vector, or (iv) a diluent control. Challenge by this model caused grave neurological signs, viremia, moderate to severe histopathologic lesions in the brain and spinal cord, and an outcome of 0% survivorship in all six control horses. In contrast, challenge in horses at between 28 days postvaccination with the chimera vaccine and 56 days postvaccination with the commercial inactivated or modified-live vaccine resulted in 100% survivorship (protection from the onset of WNV encephalitis and viremia). The study was designed to determine whether beta1-integrin plays a role in mediating the acute skeletal response to mechanical unloading. Transgenic (TG) mice were generated to express a dominant negative form of beta1-integrin under the control of the osteocalcin promoter, which targets expression of the transgene to mature osteoblasts. At 63 days of age, wild-type (WT) and TG mice were subjected to hindlimb unloading by tail suspension for 1 wk. Pair-fed, normally loaded WT and TG mice served as age-matched controls. Bone samples from each mouse were processed for quantitative bone histomorphometry and biomechanical testing. The skeletal phenotype of TG mice was characterized by lower cancellous bone mass in the distal femoral metaphysis (-52%) and lumbar vertebral body (-20%), reduced curvature of the proximal tibia (-20%), and decreased bone strength (-20%) and stiffness (-23%) of the femoral diaphysis with relatively normal indexes of cancellous bone turnover. Hindlimb unloading for only 1 wk induced a 10% decline in tibial curvature and a 30% loss of cancellous bone in the distal femur due to a combination of increased bone resorption and decreased bone formation in both WT and TG mice. However, the strength and stiffness of the femoral diaphysis were unaffected by short-term hindlimb unloading in both genotypes. The observed increase in osteoclast surface was greater in unloaded TG mice (92%) than in unloaded WT mice (52%). Cancellous bone formation rate was decreased in unloaded WT (-29%) and TG (-15%) mice, but, in contrast to osteoclast surface, the genotype by loading interaction was not statistically significant. The results indicate that altered integrin function in mature osteoblasts may enhance the osteoclastic response to mechanical unloading but that it does not have a major effect on the development of cancellous osteopenia in mice during the early stages of hindlimb unloading.

A radomized controlled trial was conducted in a metropolitan teaching hospital to determine whether improving follow-up of emergency room patients who had hypertension led to improvements in their medical care and blood pressure control. One hundred fourty four patients were randomly assigned into an intervention group and a control group. In the former, a follow-up clerk assigned patients in returning for follow-up care. Eighty-four percent of patients in this group and 63% of control patients returned to the clinic (P less than 0.1). However, five months after the patients' emergency room visits, 51% of patients in the intervention group and 53% of control patients were normotensive. There were more diagnostic and therapeutic measures in the intervention group, but long-term management was similar in both groups. Improvement in follow-up may not be by itself lead to blood pressure control among hypertensive patients.
