Background and aim: Carvone (CV) is a monoterpene in some essential oils that has therapeutic uses. The current study was undertaken to assay the antioxidant and antidepressant activities of CV and its possible mode of action using tail suspension test (TST) and open-field test (OFT). Methods: The acute toxicity of the CV was assessed by Lorke's method. Several doses of CV (25–100 mg kg−1) were injected 1 hour prior to TST. To evaluate the possible mode of action, receptor antagonists were administered 1 hour prior to the CV treatment (100 mg kg −1), and after 30 min, the TST was conducted. Moreover, the animal's serum catalase (CAT), glutathione (GSH), and nitric oxide(NO) were measured. Results: The LD50 of CV was 3.807 g kg−1. Moreover, the CV increased serum CAT and GSH and decreased NO levels. The 50 and 100 mg kg−1 of CA have potential antidepressant activity and decresed the numbers of crossings and rearings in OFT. Our results indicate that haloperidol, sulpiride, SCH23390, ketanserin WAY100135, yohimbine, prazosin, p-chlorophenylalanine (pCPA), and reserpine inhibited the antidepressant potential of CV (100 mg kg−1). The simultaneous use of a sub-dose of CV (25 mg kg−1) with fluoxetine (5 mg kg−1) enhances the antidepressant-like response. Conclusion: The CV presumably exerts antidepressant potential by acting as an antioxidant and enhancing dopamine, noradrenaline, and serotonin in synaptic clefts. Besides, the reduction in locomotor activity is indicative of the sedative effects of CV. These results promote the use of CV in commercial combinations as an adjuvant agent with traditional antidepressant agents.

Background: The prevalence of obesity is rising worldwide. Due to the side effects associated with modern drugs, there is a need for an alternative treatment for obesity management. Triphala (TP), a polyherbal formulation from Ayurveda, is used by Ayurvedic physicians for weight loss. Purpose: This study was designed to evaluate the effects of TP on obesity at the biochemical, histological, and molecular levels. Materials and Methods: Male Wistar rats (125–150 g, 6–7 weeks old) were fed with a high-fat diet (HFD) for total of 48 days. From day 24 to day 48, along with HFD, rats were given an aqueous extract of TP (50, 100, and 200 mg/kg/day) or atorvastatin (1.2 mg/kg/day). Biochemical parameters were estimated in serum. Histopathology was done for the liver and adipose tissue. Levels of the genes involved in lipid metabolism were determined by quantitative real-time polymerase chain reaction (qRT-PCR). Monoamine levels were estimated from the brain tissue. Results: At the end of the study (day 48), compared to only HFD receiving group (DC), all TP-treated groups showed a significant decrease in body weight, serum glucose, total cholesterol, triglycerides, leptin, interleukin-6, C-reactive protein, malondialdehyde, and noradrenaline levels. High-density lipoprotein, adiponectin, superoxide dismutase, serotonin, and dopamine levels were found to be increased compared to DC rats. Expression of various genes involved in lipid metabolism was found to be down regulated only in TP 100 group compared with DC. Additionally, all TP groups showed a reduction in adipocyte size and restored monoamine levels. Conclusion: The aqueous extract of TP had shown an anti-obesity effect as demonstrated by lowering inflammation and oxidative stress, adipocyte size, and modulation of expression of genes involved in lipid metabolism. Monoamine transporters (MATs) are responsible for the reuptake of dopamine, serotonin, and norepinephrine, modulating concentrations of these essential brain neurotransmitters and thus regulating behavior, mood, and cognitive functions. Studying the role of various genes in complex physiological processes is a promising area of neurobiology and sport physiology. Here, we summarize mounting evidence linking specific genetic variants of MAT genes to various aspects of sport performance. For example, the 10-repeat allele of the dopamine transporter gene (DAT), the L-allele of the serotonin transporter gene (SERT), and the single nucleotide polymorphism rs1805065 of the norepinephrine transporter gene (NET) appear to correlate with a higher athletic performance due to stress resistance, as well as the maintenance of motivation and cognitive behavioral competencies, i.e. qualities required to achieve sporting success. Objective: Depression is a serious psychiatric disorder affects over 350 million peoples worldwide. The present study aimed to address the protective effect of vinpocetine in reserpine induced-depressed rats Methods: The experimental protocol comprised of 4 groups (6 rats each), where rats received normal saline in group 1 and reserpine (0.5mg/kg, i.p) in group 2 for 14 consecutive days. Group 3 and 4 received the test drug, vinpocetine (6mg/kg) and the standard drug, fluoxetine (5mg/kg), before 30 minutes of reserpine administration for 14 days respectively. Then the effect on all groups was assessed on behavioral parameters by TST, open field test (OFT) and forced swim test (FST). While oxidative stress parameters (TBARS, GSH, SOD and CAT) were estimated in brain and Histopathological examination carried out by Hematoxylin & eosin (H&E) staining of hippocampus and cortex of brain. Result: Result indicates that Vinpocetine significantly reversed the depression induced by reserpine in rats by increasing locomotor activity in OFT, reducing immobility time in TST, and elevating swimming time in forced swim task. Vinpocetine also reduce the oxidative stress by elevating the content of GSH and antioxidant enzymes (SOD & catalase) and decrease malondialdehyde level in the brain. Moreover, In Histopathological examination, vinpocetine significantly reversed the Histopathological alteration of reserpinized rats in hippocampus and cortex region of the brain. The aim of this study is to find out whether maternal methyl-enriched diet affects the content of monoamines and their metabolites in brain structures of adult WAG/Rij offspring. It has been shown for the first time that maternal methyl-enriched diet (choline, betaine, folic acid, vitamin B12, L-methionine, zink) during the perinatal period increases dopaminergic tone of the mesolimbic brain system in adult offspring of WAG/Rij rats, which is accompanied by the suppression of the symptoms of genetic absence epilepsy and comorbid depression. Results suggest that maternal methyl-enriched diet during the perinatal period may be served as a new therapeutic strategy to prevent the development of a hypofunction of the mesolimbic dopaminergic brain system and associated genetic absence epilepsy and comorbid depression in offspring. Alzheimer's disease (AD), classified as neurodegenerative disorder that progresses over a period of time, is characterized by intracellular neurofibrillary tangles and extracellular amyloid plaques. This present research work was designed to develop a polyherbal gel for the treatment of AD. This research study is aimed to confirm the impact and validation of polyherbal gel on tauopathy and neurodegeneration that had been induced by intraperitoneal trimethyltin (TMT) injection to rats. Polyherbal loaded gel was prepared by cold method, and characterized for gel strength, viscosity, permeation and pH. Subsequently, 5 marker based standardized plant materials of Kalyanka ghrita were incorporated in gellan gum and xanthan gum. Finally, an in-vivo investigation employing rats with TMT-induced neurological disease were used to assess the efficacy of the optimized gel. On day 7, the Wistar rats received intraperitoneal injections of TMT. From day 14 to day 35, the corresponding groups received intranasal administration of polyherbal gel. In addition to the molecular parameters such as brain acetyl cholinesterase activity, BDNF (Rat brain derived neurotropic factor), protein phosphatase 2 A, antioxidant parameters, and oxidative stress markers, the behavioral parameters were also determined. Studies were conducted on the brain's monoamine levels and histology. RESULTS: Higher permeation over the nasal mucosa was demonstrated by the optimized In-situ polyherbal gel. Significant improvement in cognition was observed from the reduced escape latency, longer paths, and increased social or novel object recognition tests post polyherbal gel treatment. A documented HPLC technique helped in optimization and standardization of the polyherbal gel. The polyherbal treatment groups exhibited a considerable rise in the levels of monoamines, including norepinephrine, dopamine, and 5-hydroxy tryptamine. This study aimed to prepare defatted ethanol extract of Abelmoschus esculentus leaves, Morus nigra leaves and Punica granatum peel, to identify the chemical composition of these extracts and to explore their efficacy in counteracting diabetic neuropathy. LC-ESI-MS spectrometry was the hyphenated tool for component identification of these extracts. Behavioral, biochemical, and histopathological investigations were carried out after treatments of diabetic rats. The phenolic contents in the extracts are 16.38, 34.75 and 40.57 mg GAE/g extract regarding A. esculentus leaves, M. nigra leaves and P. granatum peel respectively. Chemodiversity of the phenolic contents was observed from the LC/Mass, where A. esculentus extract contained isoflavonoids and flavanones, M. nigra extract consisted of benzofurans, prenylated flavonoids, stilbenes, and xanthones, and P. granatum extract was rich in ellagitanins, condensed tannins, and anthocyanins. The extracts normalize of blood glucose levels, enhance the explorative behavior of the rats and their response time to thermal pain, restore the oxidant/antioxidant balance, attenuate inflammation, augment brain monoamines levels and modulate MAO-A and Ache enzyme activity. Furthermore, they recovered brain histopathological alterations. Exposure to ammonia can cause convulsions, coma, and death. In this study, we investigate the effects of ammonia exposure on immunoregulatory and neuroendocrine changes in Takifugu rubripes. Fish were sampled at 0, 12, 24, 48, and 96 h following exposure to different ammonia concentrations (0, 5, 50, 100, and 150 mg/L). Our results showed that exposure to ammonia significantly reduced the concentrations of C3, C4, IgM, and LZM whereas the heat shock protein 70 and 90 levels significantly increased. In addition, the transcription levels of Mn-SOD, CAT, GRx, and GR in the liver were significantly upregulated following exposure to low ammonia concertation, however, downregulated with increased exposure time. These findings suggest that ammonia poisoning causes oxidative damage and suppresses plasma immunity. Ammonia exposure also resulted in the elevation and depletion of the T3 and T4 levels, respectively. Furthermore, ammonia stress induced an increase in the corticotrophin-releasing hormone, adrenocorticotropic hormone, and cortisol levels, and a decrease in dopamine, noradrenaline, and 5-hydroxytryptamine levels in the brain, illustrating that ammonia poisoning can disrupt the endocrine and neurotransmitter systems. Our results provide insights into the mechanisms underlying the neurotoxic effects of ammonia exposure, which helps to assess the ecological and environmental health risks of this contaminant in marine fish.

Nanoparticles are now widely used in various aspects of life, especially zinc oxide nanoparticles (ZnNPs) that used in mouth washing, cosmetics, sunscreens, toothpaste and root canal flings. This research aims to determine the impact of ZnNPs on healthy mice's brain tissue. ZnNPs have caused major changes in the brain monoamines (dopamine, norepinephrine and serotonin) and ions such as Ca2+, Na+, K+ and Zn2+. Concerning the histological picture, administration of ZnNPs caused some histopathological impairment in brain tissue. In addition, ZnNPs reduced the level of glutathione and catalase in brain tissue, although an increase in the level of nitrite / nitrate and ROS was observed, while the level of malondialdhyde was not significantly altered. Moreover, ZnNPs induced DNA fragmentation in brain of mice. Collectively, the obtained results revealed that ZnNPs affected the brain levels of investigated monamines, ions, enzymatic and non-enzymatic antioxidants thus they may have potential influence on central nervous system. Enriching rearing environment is the strategy suggested for improving the post release survivorship of captive-reared animals. Here, an attempt has been made to evaluate the impact of early rearing enrichment on the hypothalamic–pituitary–interrenal axis (HPI axis), blood glucose, and brain dopaminergic and serotonergic systems of Tor putitora. Fifteen-day-old hatchlings of T. putitora were reared up to advanced fry stage in barren, semi-natural, and physically enriched environments and compared them with regard to pre-stress and post-stress levels of whole-body cortisol, blood glucose, brain serotonergic activity (5HIAA/5HT ratio), dopaminergic activity (DOPAC/DA and HVA/DA ratios) and norepinephrine (NE) levels. Significantly low basal whole-body cortisol, glucose and brain NE levels were observed in a physically enriched group of fish as compared to the other two groups. However, after acute stress, all rearing groups showed elevated levels of cortisol, blood glucose, brain 5HIAA/5HT, DOPAC/DA and HVA/DA ratios and NE levels but the magnitude of response was different among different rearing groups. The barren reared group showed a higher magnitude of response as compared to semi-natural and physically enriched groups. Similarly, the recovery rate of whole-body cortisol, blood glucose, and whole-brain monoamines were long-lasting in barren-reared mahseer. We illustrate that increased structural complexity (physical enrichment) during the early rearing significantly modulates various physiological and stress-coping mechanisms of mahseer. In teleosts, brain monoamines (dopamine and serotonin) participate in the early response to different acute stressors. However, little is known regarding their role during chronic stress. In a 2 × 2 factorial design, the influence of a high stocking density (HSD) and/or food deprivation (FD) on the brain monoaminergic activity in gilthead sea bream (Sparus aurata) was evaluated. Following a 21-day experimental design, samples from the plasma and brain regions (telencephalon, hypothalamus, and optic tectum) were collected. The dopamine (DA), serotonin (5HT), and their main metabolites, 3,4-dihydroxyphenylacetic acid (DOPAC) and 5 hydroxyindoleacetic acid (5HIAA), contents were HPLC-assessed in brain tissues, and the ratios DOPAC/DA and 5HIAA/5HT were calculated as indicators of enhanced monoaminergic activity. The plasma levels of cortisol and catecholamine were also evaluated. The cortisol levels increased in fish exposed to HSD and normally fed but, also, in all FD groups, whereas the NA levels decreased in LSD-FD animals. Within the brain, the dopaminergic and serotonergic activities in telencephalon and hypothalamus increased in fish subjected to HSD and in the telencephalon of LSD-FD fish. While DA (hypothalamus) and 5HT (telencephalon) increased in the animals submitted to a HSD, food-deprived fish did not show such an increase. Taken together, our results supported the hypothesis of brain monoaminergic activity participating in maintaining and orchestrating the endocrine response to chronic stress in fish.

Abstract—The effects of space radiation on the central nervous system, which are the most relevant to the assessment of radiation risk in interplanetary flights, can only be addressed in model ground-based experiments on animals. The article substantiates the methodology and presents the results of such experiments: long-term exposure to radiation was simulated using fractionated gamma irradiation of animals combined with exposure to hypogravity (antiorthostatic suspension of rats), and the specific effects of irradiation with 12C carbon ions and protons were simulated in accelerator experiments. Exposure to 12C ions has been shown to evoke significant changes in the brain monoamine metabolism, with the prefrontal cortex, the nucleus accumbens, and the hippocampus characterized as the most sensitive structures. However, the effects of exposure to high-energy protons in similar doses were almost similar to the effects of gamma radiation and could be characterized as minor. [ABSTRACT FROM AUTHOR]

Pain-associated depression is encountered clinically in some cases such as cancer, chronic neuropathy, and after operations. Tramadol is an opioid analgesic drug that may modulate monoaminergic neurotransmission by inhibition of noradrenaline and serotonin reuptake that may contribute to its antidepressant-like effects. Clinically, tramadol is used either alone or in combination with other NSAIDs in the treatment of cases associated with pain and depression, e.g. low back pain, spinal cord injury, and post-operative pain management. However, tramadol monotherapy as an antidepressant is impeded by severe adverse effects including seizures and serotonin syndrome. Interestingly, phosphodiesterase-III inhibitors demonstrated novel promising antidepressant effects. Among which, cilostazol was reported to attenuate depression in post-stroke cases, geriatrics and patients undergoing carotid artery stenting. Therefore, this study was carried out to investigate the possible antidepressant-like effects of tramadol and/or cilostazol on the behavioral level in experimental animals, and to examine the neurochemical and biochemical effects of tramadol, cilostazol and their combination in rats, in order to explore the probable mechanisms of action underlying their effects. To achieve our target, male albino mice and rats were randomly allocated into five groups and administered either vehicle for control, fluoxetine (20 mg/kg, p.o.), tramadol HCl (20 mg/kg, p.o.), cilostazol (100 mg/kg, p.o.), or combination of both tramadol and cilostazol. At day 14, mice and rats were challenged in the tail suspension test and forced swim test, respectively. Rats were sacrificed and brains were isolated for determination of brain monoamines, MDA, NO, SOD, and TNF-α. The current results showed that concurrent administration of cilostazol to tramadol-treated animals modulated depression on the behavioral level, and showed ameliorative neurochemical and biochemical effects in rats exposed to FST.

Due to its high prevalence, obesity is considered an epidemic, which stimulated research on non-invasive methods to reduce excess body fat. Transcranial direct current stimulation (tDCS) is a non-invasive technique used to modulate the activity of cerebral cortex, which has already found increasing interest in medicine as a promising methodology. The aim of this study was to analyze the impact of tDCS on feeding behavior, metabolic abnormalities and neurotransmitters in certain brain areas involved in appetite control of obese rats. The male Wistar rats were divided into five subgroups depending on consumed diet effect (lean, obese) and tDCS type (anodal, cathodal, sham, and no stimulation). Two 10-min daily sessions of tDCS for 8 consecutive days of the study were applied. Rats subjected to active tDCS (anodal right or cathodal left of the prefrontal cortex) had reduced appetite and showed lesser body weight gain than the animals subjected to sham procedure or those receiving no stimulation at all. Furthermore, tDCS contributed to reduction of epididymal fat pads and to a decrease in blood concentration of leptin. Neurochemical examination revealed that tDCS modulated serotonin pathways of the reward-related brain areas and contributed to a significant decrease in the density of D2 but not D1 dopamine receptors in the dorsal striatum, recorded 5 h after the last stimulation. No significant effect of tDCS on dopamine and it's metabolites in examined brain regions was observed. It seems that the hypothalamus was not affected by tDCS application as no changes in measured neurotransmitters were detected at any examined time point. However, these results do not exclude the possibility of the delayed response of the monoamines in the examined brain areas to tDCS application. Altogether, these findings imply that repeated tDCS of the prefrontal cortex may change feeding behavior of obese rats. Either right anodal or left cathodal tDCS were sufficient to decrease food intake, to reduce body adiposity and to normalize other metabolic anomalies. These beneficial effects can be at least partially explained by changes in serotoninergic and in lesser extent dopaminergic system activity within some brain areas belonging to reward system.

Background and Objectives: Previously we have shown that synthetic lunasin, a 43 amino acid residue-containing peptide, after its central (intracisternal) administration in mice demonstrated antagonism against dopaminergic drug behavioural effects, indicating a putative antipsychotic/anti-schizophrenic profile of lunasin. The aims of the present studies were: to test whether lunasin would show an influence on the dopaminergic system after intranasal administration, and to examine the effect(s) of lunasin on serotonin and glutamatergic systems, which could play an essential role in antipsychotic action. Materials and Methods: Lunasin was administered intra-nasally at doses 0.1 and 1 nmol/mouse in ICR mice (n = 7−8) and tested in an open field on hyperlocomotion caused by amphetamine; serotonin 5-HT 2A/2C receptor agonist 1-(2,5-dimethoxy-4-iodophenyl)- 2-aminopropane (DOI); and glutamate NMDA receptor antagonist phencyclidine. Following behavioural testing, the contents of neurotransmitters and their metabolites in brain hemispheres (n = 6−8) were assessed by ultra-high-performance liquid chromatography-time of flight mas-spectrometry (UHPLC-TOF-MS) method. Also, lunasin binding to serotonin receptors was assessed. Results: Lunasin intra-nasally fully normalized hyper-locomotion and brain monoamine levels in amphetamine- and DOI-treated mice brains. Phencyclidine behavioural effects were not influenced. In vitro receptor binding data demonstrated a low affinity of lunasin (at µM concentrations) compared with DOI (nM concentrations) for the 5-HT2A and 5-HT2C receptors. Conclusions: These results demonstrated, for the first time, that the intranasal administration of oligopeptide lunasin normalized mice behaviour and brain monoamine levels in experimental psychosis mice models. Its neuro-regulatory effects indicated a usefulness of this peptide molecule for the design of novel psychotropic agents.

We studied the relationship between neurochemical and functional brain disorders depending on changes in cerebral hemodynamics at different stages of its recovery after acute blood loss. Similar changes in the metabolism of monoamines (MAs) and an increase in seizure susceptibility (SS), which were found at different states of cerebral hemodynamics at 1 hour and 24 hours after hemorrhage, had different causes. In the first case, the changes were mainly due to insufficient cerebral circulation and hypoxia occurring after blood loss, and in the second case, they were due to the prolongation of previous disorders, which continued after the recovery of cerebral hemodynamics. A deficiency of the MA and MA-synthesizing capacity of the brain after acute blood loss extended the duration of functional disorders in the central nervous system, even after the restoration of cerebral circulation. Compensated and decompensated heart failure are characterized by different associations of disorders in the brain and heart. In compensated heart failure, the blood flow in the common carotid and basilar arteries does not change. Exacerbation of heart failure leads to severe decompensation and is accompanied by a decrease in blood flow in the carotid and basilar arteries. Changes in monoamine content occurring in the brain at different stages of heart failure are determined by various factors. The functional exercise test showed unequal monoamine-synthesizing capacities of the brain in compensated and decompensated heart failure. Reduced capacity of the monoaminergic systems in decompensated heart failure probably leads to overstrain of the central regulatory mechanisms, their gradual exhaustion, and failure of the compensatory mechanisms, which contributes to progression of heart failure. Severe feather pecking (SFP) in chickens is a detrimental behaviour with possibly neurochemical deficits at its base. Recent neurological studies depicted conflicting results on the role of serotonin (5-hydroxytryptamine, 5-HT) and dopamine (DA) in the development and display of feather pecking. We studied brain monoamine levels and behaviour in domestic chickens divergently genetically selected on feather pecking behaviour, the Low Feather Pecking (LFP) and High Feather Pecking (HFP) lines, both at a young age and when adult, to elucidate the role of 5-HT and DA in feather pecking. Also pecking behaviour and the behavioural response to challenging test situations was determined. At 8 weeks of age, HFP had lower 5-HT and DA turnover in several brain areas than LFP, whereas these differences had disappeared or were even reversed at 25 weeks of age. Line differences in central monoamine activity were found both in emotion-regulating and motor-regulating areas. As expected from previous generations, HFP exceeded LFP in most types of pecking at other birds, including severe feather pecking. Furthermore, HFP responded more actively in most behavioural tests conducted, and seem more impulsive or (hyper)active in their way of coping with challenges. This paper shows different developmental trajectories of the neurochemical systems (5-HT and DA) for chickens divergently selected on feather pecking behaviour, and a remarkable reversion of differences in monoamine activity at a later stage of life. Gut microbiota colonization is a key event for host physiology that occurs early in life. Disruption of this process leads to altered brain development which ultimately manifests as changes in brain function and behaviour in adulthood. Studies using germ-free (GF) mice highlight the extreme impact on brain health that results from life without commensal microbes. However, the impact of microbiota disturbances occurring in adulthood is less studied. To this end, we depleted the gut microbiota of 10-week-old male SpragueDawley rats via chronic antibiotic treatment. Following this marked, sustained depletion of the gut bacteria, we investigated behavioural and molecular hallmarks of gut-brain communication. Our results reveal that depletion of the gut microbiota during adulthood results in deficits in spatial memory as tested by Morris water maze, increased visceral sensitivity and a greater display of depressive-like behaviours in the forced swim test. In tandem with these clear behavioural alterations we found changes in altered CNS serotonin concentration along with changes in the mRNA levels of corticotrophin releasing hormone receptor 1 and glucocorticoid receptor. Additionally, we found changes in the expression of brain derived neurotrophic factor (BDNF), a hallmark of altered microbiota-gut-brain axis signalling. In summary, this model of antibiotic-induced depletion of the gut microbiota can be used for future studies interested in the impact of the gut microbiota on host health without the confounding developmental influence of early-life microbial alterations. The protective role of Spinacia oleracea (SO) has been evaluated against the development of Amygdala kindled (AMK) experimental epileptogenesis. Thirty six Holtzman strain adult male albino rats (200-250 g) were equally divided into 1) control, 2) SO, 3) AMK, 4) SO+AMK, 5) DZ+AMK group. After discharge duration (ADD) were used as indices of kindled seizures. In AMK group, seizure stages reached upto stage 4–5 within the second week. EEG tracings showed that pretreatment with SO in AMK group decreased the ADD and seizure stages of SO pretreated rats were limited within stage 1–2 from 1st to 4th week of kindling. Brain monoamine content of Serotonin (5-HT) was decreased in cerebral cortex (CC), cerebellum (CB), caudate nucleus (CN), midbrain (MB) and pons-medulla (PM) of AMK group which was increased by SO pre-treatment. Alteration of Dopamine (DA) and Norepinephrine (NE) in different brain regions of AMK group was also modulated by SO pre-treatment. The oxidative stress (OS)-induced prostaglandin (PG) release, in hypobaric hypoxic (HHc) condition, may be linked with the changes of brain monoamines. The present study intends to explore the changes of monoamines in hypothalamus (H), cerebral cortex (CC), and cerebellum (CB) along with the motor activity in rats after exposing them to simulated hypobaric condition and the role of PGs on the daily hypobaric hypoxia (DHH)-induced alteration of brain monoamines by administering, an inhibitor of PG synthesis, naproxen. The rats were exposed to a decompression chamber at 18,000 ft for 8 hours per day for 6 days after administration of vehicle or naproxen (18 mg/kg body wt.). The monoamine levels (epinephrine, E; norepinephrine, NE; dopamine, DA; and 5-hydroxytryptamine, 5-HT) in CC, CB, and H were assayed by high-performance liquid chromatography (HPLC) with electrochemical detection, and the locomotor behavior was measured by open field test. The NE and DA levels were decreased in CC, CB, and H of the rat brain in HHc condition. The E and 5-HT levels were decreased in CC, but in H and CB, they remained unaltered in HHc condition. These DHH-induced changes of monoamines in brain areas were prevented after administration of naproxen in HHc condition. The locomotor behavior remained unaltered in HHc condition and after administration of naproxen in HHc condition. This study was performed to investigate the biochemical effect of flaxseed oil on oxidative stress and brain monoamines release in streptozotocin-induced diabetic rats. Sixty male albino rats were divided into following four groups (15 for each group): control group, flaxseed oil group, diabetic group, and flaxseed oil-treated diabetic group. Serum glucose, insulin, pentosidine, plasma advanced oxidation protein products (AOPPs), and plasma total antioxidant capacity were estimated. Brain neurotransmitters, malondialdehyde (MDA), and nitric oxide (NO) were also determined. The mean values of serum pentosidine and plasma AOPP showed a significant decrease in treated diabetic group as compared to their values in the diabetic group. Also, brain neurotransmitters levels were improved after treatment with flaxseed. Brain MDA and NO were increased significantly in the diabetic group, while they were significantly decreased after treatment. Brain NO and brain MDA had a significant positive correlation with pentosidine, AOPP, and neurotransmitters. We concluded that flaxseed oil supplementation may be useful in the treatment of brain dysfunction in diabetes. [ABSTRACT FROM AUTHOR]

Exposure to ammonia can cause convulsions, coma, and death. In this study, we investigate the effects of ammonia exposure on immunoregulatory and neuroendocrine changes in Takifugu rubripes. Fish were sampled at 0, 12, 24, 48, and 96 h following exposure to different ammonia concentrations (0, 5, 50, 100, and 150 mg/L). Our results showed that exposure to ammonia significantly reduced the concentrations of C3, C4, IgM, and LZM whereas the heat shock protein 70 and 90 levels significantly increased. In addition, the transcription levels of Mn-SOD, CAT, GRx, and GR in the liver were significantly upregulated following exposure to low ammonia concertation, however, downregulated with increased exposure time. These findings suggest that ammonia poisoning causes oxidative damage and suppresses plasma immunity. Ammonia exposure also resulted in the elevation and depletion of the T3 and T4 levels, respectively. Furthermore, ammonia stress induced an increase in the corticotrophin-releasing hormone, adrenocorticotropic hormone, and cortisol levels, and a decrease in dopamine, noradrenaline, and 5-hydroxytryptamine levels in the brain, illustrating that ammonia poisoning can disrupt the endocrine and neurotransmitter systems. Our results provide insights into the mechanisms underlying the neurotoxic effects of ammonia exposure, which helps to assess the ecological and environmental health risks of this contaminant in marine fish. Effects of chronic overcrowding on the development of depressive-like behavior and changes in monoamine contents in the brain structures involved in the development of depression were studied in rats. It was shown that overcrowding increased grooming duration and did not change the number of crossed squares and postures in the open-field test. In the forced swimming test, overcrowding increased immobility time and deceased the duration of the first episode of active swimming, which attested to depressive-like behavior. Overcrowding reduced the content of dopamine in the striatum, its metabolites (homovanilic and dioxyphenylacetic acids) and dopamine metabolism rate in the hypothalamus, and increased the concentrations of norepinephrine, homovanilic acid, and dioxyphenylacetic acid in the olfactory bulb and homovanilic acid in the prefrontal cortex. No changes were found in the nucleus accumbens. Serotonin content did not change in all analyzed structures. It was hypothesized that the observed changes in the content of monoamines and their metabolites in certain brain structures could underlie the depressive-like behavior induced by overcrowding in rats. Abstract: Background and aims: Establishment of the gut microbiota is one of the most important events in early life and emerging evidence indicates that the gut microbiota influences several aspects of brain functioning, including reactivity to stress. To better understand how the gut microbiota contributes to a vulnerability to the stress-related psychiatric disorders, we investigated the relationship between the gut microbiota, anxiety-like behavior and HPA axis activity in stress-sensitive rodents. We also analyzed the monoamine neurotransmitters in the brain upper structures involved in the regulation of stress and anxiety. Methods: Germfree (GF) and specific pathogen free (SPF) F344 male rats were first subjected to neurological tests to rule out sensorimotor impairments as confounding factors. Then, we examined the behavior responses of rats to social interaction and open-field tests. Serum corticosterone concentrations, CRF mRNA expression levels in the hypothalamus, glucocorticoid receptor (GR) mRNA expression levels in the hippocampus, and monoamine concentrations in the frontal cortex, hippocampus and striatum were compared in rats that were either exposed to the open-field stress or not. Results: GF rats spent less time sniffing an unknown partner than SPF rats in the social interaction test, and displayed a lower number of visits to the aversive central area, and an increase in latency time, time spent in the corners and number of defecations in the open-field test. In response to the open-field stress, serum corticosterone concentrations were 2.8-fold higher in GF than in SPF rats. Compared to that of SPF rats, GF rats showed elevated CRF mRNA expression in the hypothalamus and reduced GR mRNA expression in the hippocampus. GF rats also had a lower dopaminergic turnover rate in the frontal cortex, hippocampus and striatum than SPF rats. Conclusions: In stress-sensitive F344 rats, absence of the gut microbiota exacerbates the neuroendocrine and behavioral responses to acute stress and the results coexist with alterations of the dopaminergic turnover rate in brain upper structures that are known to regulate reactivity to stress and anxiety-like behavior. Abstract: The presence of blue or red-brown substrate on the tank bottom has been previously reported as an efficient means of environmental enrichment for gilthead seabream. The present study aimed to investigate whether this enrichment is still beneficial when gilthead seabream is reared under different social conditions (i.e. a lower 4.9kgm−3 and a higher 9.7kgm−3 density). Water exchange was adjusted according to fish biomass to exclude density effects on water quality. In the enriched tanks single-colour glass gravel was used as substrate (blue and red-brown substrate, or BS and RBS respectively), while control tanks had no gravel. Growth, aggressive behaviour and size distribution results indicated that the lower density created a less favourable social environment. In both densities studied, BS enhanced growth, suppressed aggression and reduced brain serotonergic activity. In the condition of intense social interactions (i.e. the lower density) BS also reduced brain dopaminergic activity. These results along with the negative correlations observed between brain monoamines and fish body mass, indicated that substrate and density effects are socially-induced. However, there may be several biotic and/or abiotic factors interfering with substrate effects that should be investigated before the practical use of a substrate in land-based intensive aquaculture. Diazinon is an organophosphorous pesticide with a prominent toxicity on many body organs. Multiple mechanisms contribute to diazinon-induced deleterious effects. Inhibition of acetyl-cholinesterase, cholinergic hyperstimulation, and formation of reactive oxygen species may play a role. On the other hand, melatonin is a pineal hormone with a well-known potent antioxidant activity and a remarkable modulatory effect on many behavioral processes. The present study revealed that oral diazinon administration (25 mg/kg) increased anxiety behavior in rats subjected to elevated plus maze and open-field tests possibly via the induction of changes in brain monoamines levels (dopamine, norepinephrine, and serotonin). Additionally, brain lipid peroxides measured as malondialdehyde (MDA) and tumor necrosis factor alpha (TNF-α) levels were elevated, while the activity of brain glutathione peroxidase enzyme was reduced by diazinon. Co-administration of oral melatonin (10 mg/kg) significantly attenuated the anxiogenic activity of diazinon, rebalanced brain monoamines levels, decreased brain MDA and TNF-α levels, and increased the activity of brain glutathione peroxidase enzyme. Blood immunological parameters (cytokine profile and interferon status) and the level of monoamines and their metabolites in various brain structures (amygdala, hippocampus, septum, and hypothalamus) were studied in rats kept under standard conditions or in overpopulated cages. Long-term overcrowding was associated with reduced expression of IL-4 gene, increased transcription of IL-17, and decreased production of IFN-γ, which attested to impaired humoral and cell-mediated immunity and disturbances in IFN-γ synthesis at the post-transcriptional level. Under these conditions, the levels of norepinephrine and dopamine decreased in the septum, but increased in the hypothalamus. The amount of dopamine metabolite dihydroxyphenylacetic acid decreased in both these structures, and the index of dopamine metabolism (dihydroxyphenylacetic acid/dopamine ratio, DOPAC/dopamine) decreased only in the hypothalamus. Overcrowding was not followed by changes in the parameters of noradrenergic and dopaminergic systems in the amygdala and hippocampus and serotoninergic system in all study structures. Abstract: Senegalese sole is a highly valued marine teleost introduced in aquaculture since the early 1980s, but growth and survival from juvenile to market-size fish still reveal deficiencies, with stress being one of the possible explanations for this issue. High circulating levels of catecholamines and cortisol are involved in the integrated response to stress in fish and, thus triggering tissue alterations in order to counter the impact of the stressor and satisfy the increased energy demand. Chronic exposure to stressors can lead to an allostatic overload (or distress) affecting negatively food intake, reproduction, growth and immune functions, leading to increased incidence of diseases and reduced animal welfare. Melatonin is mainly synthesized in pineal organ and plays a critical role in the regulation of seasonal and circadian rhythms of physiology and behavior. However, a role as an anti-stress molecule at both central and peripheral levels has been also proposed for the hormone. Based on that, the aim of the present study was to evaluate the putative role of melatonin in reducing stress-related neuroendocrine and metabolic effects in Senegalese sole exposed to different and representative chronic stress conditions such as high stocking density (HSD) and low water replacement (LWR). Our results show that stressing Senegalese sole by high stocking density or low water replacement induce typical changes in parameters related to stress response in teleosts (increased plasma cortisol levels, enhanced liver glycogenolytic potential, and increased dopaminergic and serotonergic activities in the hypothalamus). Whereas melatonin treatment in non-stressed fish increases the dopaminergic activity in the hypothalamus, the opposite effect is observed after the addition of the hormone to tank water before and during stress exposure, thus attenuating the effect of stress on plasma cortisol and liver glycogen levels. The dopaminergic and serotonergic metabolism was also affected by melatonin treatment only in the hypothalamus on stressed fish. Thus, the presence of melatonin at night in tank water was able to counteract several of the effects induced by stress especially those related to cortisol synthesis and release, which is in support of an anti-stress role for melatonin in Senegalese sole. Thyroid hormones are recognized as the key metabolic hormones that play a critical role in the development of central nervous system (CNS) throughout life. The present study was designed to determine the changes in brain monoamine concentrations in 6-n-propyl thiouracil (PTU)-induced hypothyroid rats, in addition to the ameliorating role of folic acid treatment. Fifty male albino rats were equally divided into five groups; first and second groups were the control and folic acid groups, respectively, while the third group was the hypothyroid group in which the rats received PTU in drinking water for 6 weeks. The fourth and fifth groups were co- and post-treated folic acid groups with hypothyroid rats, respectively. Our results revealed that serotonin and norepinephrine concentrations were significantly decreased in the hypothalamus and cortex, while it significantly increased in the hippocampus of hypothyroid rats when compared with control group. Serotonin and norepinephrine concentrations were decreased in hypothalamus and cortex in co- and post-treated folic acid groups with hypothyroid rats, while the concentration of dopamine were significantly increased in the hypothalamus and hippocampus of the hypothyroid rats and co-treated folic acid group with hypothyroid rats. In cortex, the dopamine concentration was significantly increased in hypothyroid rats and post-treated folic acid group with hypothyroid rats, while it significantly decreased in co-treated folic acid group with hypothyroid rats when compared with the control group. Also, our results revealed that, folic acid treatment was better if it is administered as an adjuvant after returning to the euthyroid state by withdrawing PTU from the drinking water. The present study explored the potential role of brain catecholaminergic and serotoninergic systems as neuronal targets for the toxicological effects of acute ammonia exposure (0.28 mg l of un-ionized ammonia for 12 and 24 h) in juvenile sole ( Solea senegalensis). In addition, plasma cortisol levels were measured. The results showed significant increases in their concentrations that were similar after 12 and 24 h of exposure. These data indicate that acute exposure (12 and 24 h) to ammonia initiates a typical stress response in the Senegalese sole, with stimulation of the hypothalamus-pituitary-interrenal axis. The concentrations of dopamine (DA), serotonin (5-hydroxytryptamine; 5-HT) and noradrenaline (NA), and their metabolites, 3, 4-dihydroxyphenylacetic acid (DOPAC) and 5-hydroxy-3-indoleacetic acid (5HIAA), were measured in the hypothalamus, telencephalon and optic tectum. The main changes induced by acute exposure to ammonia were decreases in the concentrations of 5-HT and DA, which were significant in most of the brain regions studied. The ratios of 5-HIAA/5-HT and DOPAC/DA increased in all regions and at all times studied, although in the case of the DOPAC/DA ratio, the increases were only significant in the hypothalamus (24 h exposure) and in the optic tectum (12 and 24 h exposure). These changes indicated that exposure to ammonia elicited time-dependent increases in serotoninergic and dopaminergic activity in the hypothalamus, telencephalon and optic tectum. Stress is the leading psychopathological cause for several mental disorders. Physiological and psychological responses to stress are mediated by the hypothalamic--pituitary--adrenal (HPA), sympathoadrenal system (SAS), and brain monoaminergic systems (BMS). Eugenol is reported to substantially modulate brain functions by regulating voltage-gated cation channels and release of neurotransmitters. This study was designed to evaluate the anti-stress effect of eugenol in the 4-h restraint model using rats. Ulcer index was measured as a parameter of the stress response. HPA axis and the SAS were monitored by estimating plasma corticosterone and norepinephrine (NE), respectively. Analysis of NE, serotonin (5-HT), dopamine, and their metabolites in discrete brain regions was performed to understand the role of BMS in the anti-stress effect of eugenol. Stress exposure increased the ulcer index as well as plasma corticosterone and NE levels. Eugenol pretreatment for 7 days decreased the stress-induced increase in ulcer index and plasma corticosterone but not NE levels, indicating a preferential effect on the HPA axis. Furthermore, eugenol showed a ''U''-shaped dose--response curve in decreasing ulcer index and plasma corticosterone levels. Eugenol also reversed the stress-induced changes in 5-HT levels in all brain regions, whereas NE levels were reversed in all brain regions except hippocampus. Abstract: The study investigated whether dietary reduced glutathione (GSH) could be useful in alleviating arsenic-induced female gonadal injury in Wistar rats. Ingestion of sodium arsenite contaminated water (0.4ppm) to Wister rats for seven estrous cycles (28 days) significantly diminished ovarian key steroidogenic enzymes activities and gonadotrophins and estradiol signaling along with disrupted ovarian and uterine growth. Oxidative stress as well as histological distortion of ovarian and uterine tissue was prominent along with prolonged diestrus in arsenic-exposed rats. Decreased levels of dopamine (DA) and norepinephrine (NE) and elevated level of 5-hydroxytryptamine (5-HT) in midbrain and diencephalons were also evident following arsenic exposure. All these deleterious effects of arsenic ingestion were mitigated by administration of GSH (8.0mg/100g body weight/day). Only tissue deposition of arsenic appeared to be unaffected by dietary GSH. Abstract: Maternal stress in pregnant sows may induce long-lasting alterations in the behavior, physiology, and immunity of their offspring. The aim of the present study was to investigate the consequences of repeated social stress during late gestation on determinants of the hypothalamic–pituitary–adrenal axis and on hippocampal neurotransmitter profiles in pig offspring. All pregnant gilts were housed in pairs. Each Stress gilt was mixed with an unfamiliar gilt twice a week between days 77 and 105 of gestation (n=18). Control gilts were housed in stable pairs over the same period (n=18). Plasma cortisol and corticosteroid binding globulin (CBG) were measured in 1 male and 1 female per litter in a basal situation on postnatal days (PND) 4, 26, and 60 and in a stressful situation at PND 28 (2 d after weaning) and 62 (2 d after relocation to a new building). Prenatal stress had no effect on plasma cortisol, but it decreased CBG at PND 26. Brain and adrenals were collected from 1 female per litter after weaning or relocation at PND 28 and PND 62. Adrenals were additionally collected at PND 4. Glucocorticoid receptor binding in the hippocampus and hypothalamus was not affected by prenatal treatment. However, prenatal stress increased the expression of 11β-hydroxysteroid dehydrogenase type 1 mRNA in the hippocampus after weaning (P <0.05) and after relocation (P =0.08). In addition, prenatally stressed piglets showed an increased 5-hydroxyindole-3-acetic acid to 5-hydroxytryptamine ratio in the hippocampus after weaning and increased hippocampal c-fos mRNA expression and noradrenaline concentration after relocation (P <0.05). Prenatal stress also increased the relative adrenal weight at PND 4 and the cell density in the cortex and the medulla at PND 28, whereas no difference was found for activities of catecholamine-synthesising enzymes in the medulla. Overall, our data indicate that repeated social stress during pregnancy has long-lasting consequences on hypothalamic–pituitary–adrenal axis and hippocampal neurotransmitter activity in the offspring of pigs. Abstract: We have investigated if treatment with two different PAHs such as naphthalene (NAP) and benzo(a)pyrene (BaP), and the PAH-like compound β-naphthoflavone (BNF), may modify the stress responses elicited in rainbow trout by acute or prolonged stress stimuli, and the possible involvement of brain monoamines in those responses. Two experiments (acute and prolonged stress) were performed. In the acute stress experiment, fish were i.p. injected with vegetable oil alone (control) or oil containing NAP, BNF or BaP (10mgkg−1), and 72h after injection fish were acutely stressed by chasing for 15min. In the prolonged stress experiment, a similar group-design and injection protocol were followed, but fish were submitted to severe confinement stress by maintaining fish under high stock density (70kg fish massm−3) for 72h. The levels of cortisol, glucose and lactate were assayed in plasma. In addition, the contents of dopamine (DA), noradrenaline (NA) and serotonin (5HT), as well as their oxidized amine metabolites, 3,4-dihydroxyphenylacetic acid (DOPAC) and 5-hydroxy-3-indoleacetic acid (5HIAA) were assayed in telencephalon, hypothalamus, preoptic region, optic tectum and brain stem, as well as the pituitary. Both acute and prolonged stress stimuli increased plasma levels of cortisol, which further increase with NAP and BNF treatments after acute stress. In contrast, cortisol levels of fish exposed to prolonged stress showed a clear tendency to decrease after the treatment with BNF and BaP. Stress stimuli also increased plasma glucose levels, which were not affected by PAHs in acute stressed fish but decreased in fish exposed to prolonged stress. Increased plasma levels of lactate in fish exposed to stress decreased after PAHs treatment in acute stress but not in prolonged stress. With respect to monoaminergic systems, major changes induced by both acute and prolonged stress were increases of the metabolites DOPAC and 5HIAA and DOPAC/DA or 5HIAA/5HT ratios in several brain regions. PAHs induced alterations in the normal responses of monoaminergic systems to stress, with dopaminergic system being the most affected after acute stress, and serotonergic system after prolonged stress. Those alterations, especially after prolonged stress, showed certain parallelism with alterations of plasma cortisol levels. Thus, results suggest that in stressed fish PAH effects on plasma cortisol levels (and its derived metabolic actions) could be in part mediated by alterations on the monoaminergic systems at the CNS of rainbow trout. Abstract: Reactions to stress vary between individuals, and physiological and behavioral responses tend to be associated in distinct suites of correlated traits, often termed stress-coping styles. In mammals, individuals exhibiting divergent stress-coping styles also appear to exhibit intrinsic differences in cognitive processing. A connection between physiology, behavior, and cognition was also recently demonstrated in strains of rainbow trout (Oncorhynchus mykiss) selected for consistently high or low cortisol responses to stress. The low-responsive (LR) strain display longer retention of a conditioned response, and tend to show proactive behaviors such as enhanced aggression, social dominance, and rapid resumption of feed intake after stress. Differences in brain monoamine neurochemistry have also been reported in these lines. In comparative studies, experiments with the lizard Anolis carolinensis reveal connections between monoaminergic activity in limbic structures, proactive behavior in novel environments, and the establishment of social status via agonistic behavior. Together these observations suggest that within-species diversity of physiological, behavioral and cognitive correlates of stress responsiveness is maintained by natural selection throughout the vertebrate sub-phylum. Abstract: We investigated the association between variation in parenting style and the offspring''s behaviour and CSF monoamine metabolite (5-HIAA, HVA, and MHPG) levels in rhesus monkeys. Study subjects were 25 two-year-old females reared by their biological mothers and 15 same-aged females that were crossfostered at birth and reared by unrelated mothers. Subjects that were rejected more by their mothers in the first 6 months of life engaged in more solitary play and had lower CSF concentrations of 5-HIAA than subjects that were rejected less. The relation between these variables was generally similar in crossfostered and noncrossfostered females. CSF levels of 5-HIAA were negatively correlated with rates of scratching, a behavioural indicator of anxiety. These results suggest that that early exposure to high rates of maternal rejection can result in higher anxiety later in life, and that this effect may be mediated by serotonergic mechanisms. Variation in maternal protectiveness did not affect offspring behaviour and neither protectiveness nor rejection affected CSF levels of HVA and MHPG. CSF levels of MHPG, however, were negatively correlated with solitary play behaviour and avoidance of other individuals, suggesting that individuals with lower CSF MHPG were more fearful and socially phobic than those with higher CSF MHPG. Taken together, these findings suggest that individual differences in anxiety and fearfulness in young rhesus monkeys are accounted for, at least in part, by variation in CSF levels of monoamine metabolites, and that the development of brain monoamine systems, particularly serotonin, can be affected by early exposure to variable maternal behaviour. Abstract: Curcuma longa is a major constituent of Xiaoyao-san, the traditional Chinese medicinal formula, which has been used to effectively manage stress and depression-related disorders in China. Curcumin is the active component of curcuma longa, and we hypothesized that curcumin would have an influence on depressive-like behaviors. The purpose of the present study was to confirm the putative antidepressant effect of chronic administrations of curcumin (1.25, 2.5, 5 and 10 mg/kg, p.o.) in the forced swimming test and bilateral olfactory bulbectomy (OB) models of depression in rats. In the first study, chronic treatment with curcumin (14 days) reduced the immobility time in the forced swimming test. In the second experiment, curcumin reversed the OB-induced behavioral abnormalities such as hyperactivity in the open field, as well as deficits in step-down passive avoidance. In addition, OB-induced low levels of serotonin (5-HT), noradrenaline (NA), high 5-hydroxyindoleacetic acid (5-HIAA) and 4-dihydroxyphenylacetic acid (DOPAC) in the hippocampus were observed, and were completely reversed by curcumin administration. A slight decrease in 5-HT, NA and dopamine (DA) levels was found in the frontal cortex of OB rats which was also reversed by curcumin treatment. These results confirm the antidepressant effects of curcumin in the forced swim and the OB models of depression in rats, and suggest that these antidepressant effects may be mediated by actions in the central monoaminergic neurotransmitter systems.
