Abstract

In this work, we present a novel downscaling procedure for compositional quantities based on the Aitchison geometry. The method is able to naturally consider compositional constraints, i.e. unit-sum and positivity. We show that the method can be used in a block sequential Gaussian simulation framework in order to assess the variability of downscaled quantities. Finally, to validate the method, we test it first in an idealized scenario and then apply it for the downscaling of digital soil maps on a more realistic case study. The digital soil maps for the realistic case study are obtained from SoilGrids, a system for automated soil mapping based on state-of-the-art spatial predictions methods.

Abstract

Despite exciting advances in gene editing, the efficient delivery of genetic tools to extrahepatic tissues remains challenging. This holds particularly true for the skin, which poses a highly restrictive delivery barrier. In this study, we ran a head-to-head comparison between Cas9 mRNA or ribonucleoprotein (RNP)-loaded lipid nanoparticles (LNPs) to deliver gene editing tools into epidermal layers of human skin, aiming for in situgene editing. We observed distinct LNP composition and cell-specific effects such as an extended presence of RNP in slow-cycling epithelial cells for up to 72 h. While obtaining similar gene editing rates using Cas9 RNP and mRNA with MC3-based LNPs (10–16%), mRNA-loaded LNPs proved to be more cytotoxic. Interestingly, ionizable lipids with a pKa∼ 7.1 yielded superior gene editing rates (55%–72%) in two-dimensional (2D) epithelial cells while no single guide RNA-dependent off-target effects were detectable. Unexpectedly, these high 2D editing efficacies did not translate to actual skin tissue where overall gene editing rates between 5%–12% were achieved after a single application and irrespective of the LNP composition. Finally, we successfully base-corrected a disease-causing mutation with an efficacy of ∼5% in autosomal recessive congenital ichthyosis patient cells, showcasing the potential of this strategy for the treatment of monogenic skin diseases. Taken together, this study demonstrates the feasibility of an in situcorrection of disease-causing mutations in the skin that could provide effective treatment and potentially even a cure for rare, monogenic, and common skin diseases.

Abstract

Parastomal hernia (PH) is one of the most frequent ostomy complications, and the reported incidence in the literature is highly variable. As highlighted by the Association of Stoma Care Nurses UK, this complication develops mainly in children and older men over 70, but many predisposing factors are related to the individual patient and surgery. There is no standardised system for assessing PH. The main assessment techniques include objective examination, ultrasound scan and computed tomography. Prevention is based on various interventions by surgeons and stoma care nurses (SCNs). The SCN's primary interventions include accurate patient evaluation, pre-operative ostomy siting, education about body weight management and advice on appropriate exercises. The treatment of PH can be conservative or surgical, and the choice is based on the patient's clinical condition. Ostomy can significantly impact on a patient's quality of life (QoL), and the presence of PH can further aggravate the situation. Abstract

Ground collapses over abandoned underground mines constitute a new environmental risk in the world. The high risk associated with subsurface voids, together with lack of knowledge of the geometric and geomechanical features of mining areas, makes abandoned underground mines one of the current challenges for countries with a long mining history. In this study, a stability analysis of Montevecchia marl mine is performed in order to validate a general approach that takes into account the poor local information and the variability of the input data. The collapse risk was evaluated through a numerical approach that, starting with some simplifying assumptions, is able to provide an overview of the collapse probability. The final results is an easy-accessible-transparent summary graph that shows the collapse probability. This approach may be useful for public administrators called upon to manage this environmental risk. The approach tries to simplify this complex problem in order to achieve a roughly risk assessment, but, since it relies on just a small amount of information, any final user should be aware that a comprehensive and detailed risk scenario can be generated only through more exhaustive investigations.

This work presents a portable multi-wavelength fluorescence measurement system. The pinhole collimator, Fresnel focusing lens and PIN photo diode provide a cost-effective fluorescent light detection path. The with matte black filament 3D printed housing reduces unwanted reflections along the detection path. The integrated laser diode drivers ensure constant optical output power during operation. This is achieved by utilizing a laser diode package which incorporates both a laser and monitor diode. The monitor diode's output photocurrent is proportional to the optical output power, and thus provides, a feedback path for the driver. An optical diffuser is pre-mounted on each laser diode, providing a uniform illumination of the measurement area. The device adapts to a wide variety of fluorescent dyes by offering exchangeable laser diode modules and support of standard-sized optical thin film filters. The precision integrator transimpedance amplifier applies a switched-input photocurrent measurement technique that ensures the level of sensitivity required for clinical usage. Experimental validation indicates sensitivity in the pico-ampere range. Ex vivo and in vivo measurements, performed by the Faculty of Pharmacy at the Université de Montréal, confirm that the instrument matches its intended application.

Abstract

Monitoring-based preventive medicine relies heavily on minimal or noninvasive, portable, and cost-effective measurement solutions. In this study, we present a handheld multiwavelength fluorescence measurement system for detection of dermally injected fluorescent tracers. The circular detection area of the device is positioned on the sample containing the fluorescent marker. The marker is excited at regular intervals by a pulse of light. The optical output power is controlled by a laser diode driver, which receives direct feedback on the emitted light intensity from the monitor photodiode integrated into the laser diode package. An optical diffuser is located in front of each laser diode to ensure uniform illumination of the measurement area. Pulsed excitation of the fluorescent tracer allows alternate measurement of foreground and background light intensity. By subtracting the background from the foreground and using a switched-input photocurrent measurement technique, the measured photocurrent represents the actual light emission from the marker. The instrument has interchangeable modules that contain the excitation electronics and emission filter. These modules allow the instrument to be configured for a variety of different fluorescent markers. In vitro measurements show that the device is capable of performing fluorescence measurements for its intended use as a point-of-care device in preventive medicine.
