Abstract

Human brain structure changes throughout the lifespan. Altered brain growth or rates of decline are implicated in a vast range of psychiatric, developmental and neurodegenerative diseases. In this study, we identified common genetic variants that affect rates of brain growth or atrophy in what is, to our knowledge, the first genome-wide association meta-analysis of changes in brain morphology across the lifespan. Longitudinal magnetic resonance imaging data from 15,640 individuals were used to compute rates of change for 15 brain structures. The most robustly identified genes GPR139, DACH1 and APOE are associated with metabolic processes. We demonstrate global genetic overlap with depression, schizophrenia, cognitive functioning, insomnia, height, body mass index and smoking. Gene set findings implicate both early brain development and neurodegenerative processes in the rates of brain changes. Identifying variants involved in structural brain changes may help to determine biological pathways underlying optimal and dysfunctional brain development and aging.

Abstract

This review summarizes the last decade of work by the ENIGMA (Enhancing NeuroImaging Genetics through Meta Analysis) Consortium, a global alliance of over 1400 scientists across 43 countries, studying the human brain in health and disease. Building on large-scale genetic studies that discovered the first robustly replicated genetic loci associated with brain metrics, ENIGMA has diversified into over 50 working groups (WGs), pooling worldwide data and expertise to answer fundamental questions in neuroscience, psychiatry, neurology, and genetics. Most ENIGMA WGs focus on specific psychiatric and neurological conditions, other WGs study normal variation due to sex and gender differences, or development and aging; still other WGs develop methodological pipelines and tools to facilitate harmonized analyses of "big data" (i.e., genetic and epigenetic data, multimodal MRI, and electroencephalography data). These international efforts have yielded the largest neuroimaging studies to date in schizophrenia, bipolar disorder, major depressive disorder, post-traumatic stress disorder, substance use disorders, obsessive-compulsive disorder, attention-deficit/hyperactivity disorder, autism spectrum disorders, epilepsy, and 22q11.2 deletion syndrome. More recent ENIGMA WGs have formed to study anxiety disorders, suicidal thoughts and behavior, sleep and insomnia, eating disorders, irritability, brain injury, antisocial personality and conduct disorder, and dissociative identity disorder. Here, we summarize the first decade of ENIGMA's activities and ongoing projects, and describe the successes and challenges encountered along the way. We highlight the advantages of collaborative large-scale coordinated data analyses for testing reproducibility and robustness of findings, offering the opportunity to identify brain systems involved in clinical syndromes across diverse samples and associated genetic, environmental, demographic, cognitive, and psychosocial factors.

We will investigate the bidirectional association between parenting styles and child externalising using distinct family-based, genetically informed methods in two samples matched on age range (from 4.5- to 8-years-old) and the measures used to assess parenting and child externalising symptoms. By triangulating evidence across samples and methods, we seek to clarify the role of genetic and environmental factors and the direction of any potentially causal effects between generations.

Abstract

The Benzo Research Project is a student-led group researching recreational benzodiazepine (benzo) use in young people across the UK with the aim of sparking conversation and raising awareness of the emerging issues surrounding this. It was set up in Summer 2021, and has had over 50 young people volunteering 4,000 hours over 16 months, conducted entirely online via WhatsApp group chats and Zoom calls. Over the course of 16 months, the Benzo Research Project collected 81 anonymous testimonies and counting on non-medical benzo use amongst young people (18-25) in the UK. We conducted an inductive thematic analysis of 73 testimonies received between March and September 2022. Respondents were prompted with questions on: Young people’s experiences on benzos, whether positive, neutral, or negative Their opinions on the connection between benzo use, education, and music subcultures Previous experiences of accessing support services What effective drug support would look like to them Blackouts Experiences of those who’ve been around others on benzos but not taken it themselves When submitting, participants could choose to give us demographic information such as place of residence and gender, however this was optional. Abstract

Heritability of aspects of the externalising spectrum such as aggressive antisocial behaviour, behavioural disinhibition and risk taking range from moderate to high (Dhanani et al., 2018) suggesting a role for genetics in the development of externalising symptoms. Linnér et al. (2021) recently published a multivariate analysis of 1.5 million people which investigated the genetic links to externalising-related traits. Using the results from their genomic structural equation model, researchers created polygenic risk scores (PRS) for their externalising behaviour factor. The study reported associations between their PRS for externalising and a broad range of externalising related phenotypes including substance use initiation, antisocial personality disorder, ADHD diagnosis and rule breaking. Associations found in PRS analyses in unrelated individuals can be confounded by several factors, such as population stratification, assortative mating (Selzam et al., 2019) and indirect genetic effects of parental genotype on child phenotype (genetic nurture, Kong et al., 2018). Using sibling pairs in genomic analyses can allow for the estimation of genetic effects free from such confounds and biases, as the siblings are matched for parental and family environment. DZ twin pairs are also matched for prenatal environment and time-variant factors. By comparing within-family and between-family effects, we can investigate the extent to which passive genotype-environment correlation (rGE, I.e. genetic nurture, population stratification, and assortative mating) inflates the association between genomic variation and externalising phenotype. Little is known about PRS prediction of externalising symptoms in different developmental periods. Costantini et al., (2021) reported an association between higher polygenic scores for neuroticism, a personality trait associated with externalising behaviours, and increasing levels of externalising problems from early to late childhood (4-11 years). Our work will extend on this by including later developmental periods, when children become more autonomous and so parenting may have less of an impact. We also will be using a polygenic score specifically developed for externalising. No study to-date has investigated how within- and between-family differences in PRS prediction of externalising behaviour may change across development. Twin analyses have previously shown that common environmental effects (C – those shared between twins), are larger in childhood. Conversely, heritability (h2) estimates get larger as children age. Arguably, genetic nurture effects may be captured in estimates of the shared environment. If so, genetic nurture effects would follow the same developmental trends as C and reduce in magnitude across development. We will test whether this is the case in the TEDS sample, calculating within/between-family PGS estimates at multiple time points across development. Following the twin literature, we expect to see larger genetic nurture effects (captured by the between-family genomic analyses) in childhood than in adulthood, with the expectation that direct genetic effects will be smaller in childhood and increase with age. We will complement our genomic analysis with twin analyses using the same data.

Abstract

Neuroimaging and genetics studies have advanced our understanding of the neurobiology of sleep and its disorders. However, individual studies usually have limitations to identifying consistent and reproducible effects, including modest sample sizes, heterogeneous clinical characteristics and varied methodologies. These issues call for a large-scale multi-centre effort in sleep research, in order to increase the number of samples, and harmonize the methods of data collection, preprocessing and analysis using pre-registered well-established protocols. The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) consortium provides a powerful collaborative framework for combining datasets across individual sites. Recently, we have launched the ENIGMA-Sleep working group with the collaboration of several institutes from 15 countries to perform large-scale worldwide neuroimaging and genetics studies for better understanding the neurobiology of impaired sleep quality in population-based healthy individuals, the neural consequences of sleep deprivation, pathophysiology of sleep disorders, as well as neural correlates of sleep disturbances across various neuropsychiatric disorders. In this introductory review, we describe the details of our currently available datasets and our ongoing projects in the ENIGMA-Sleep group, and discuss both the potential challenges and opportunities of a collaborative initiative in sleep medicine.

Abstract

This review summarizes the last decade of work by the ENIGMA (Enhancing NeuroImaging Genetics through Meta Analysis) Consortium, a global alliance of over 1400 scientists across 43 countries, studying the human brain in health and disease. Building on large-scale genetic studies that discovered the first robustly replicated genetic loci associated with brain metrics, ENIGMA has diversified into over 50 working groups (WGs), pooling worldwide data and expertise to answer fundamental questions in neuroscience, psychiatry, neurology, and genetics. Most ENIGMA WGs focus on specific psychiatric and neurological conditions, other WGs study normal variation due to sex and gender differences, or development and aging; still other WGs develop methodological pipelines and tools to facilitate harmonized analyses of "big data" (i.e., genetic and epigenetic data, multimodal MRI, and electroencephalography data). These international efforts have yielded the largest neuroimaging studies to date in schizophrenia, bipolar disorder, major depressive disorder, post-traumatic stress disorder, substance use disorders, obsessive-compulsive disorder, attention-deficit/hyperactivity disorder, autism spectrum disorders, epilepsy, and 22q11.2 deletion syndrome. More recent ENIGMA WGs have formed to study anxiety disorders, suicidal thoughts and behavior, sleep and insomnia, eating disorders, irritability, brain injury, antisocial personality and conduct disorder, and dissociative identity disorder. Here, we summarize the first decade of ENIGMA's activities and ongoing projects, and describe the successes and challenges encountered along the way. We highlight the advantages of collaborative large-scale coordinated data analyses for testing reproducibility and robustness of findings, offering the opportunity to identify brain systems involved in clinical syndromes across diverse samples and associated genetic, environmental, demographic, cognitive, and psychosocial factors.

Abstract

Sex is a biological variable that contributes to individual variability in brain structure and behavior. Neuroimaging studies of population‐based samples have identified normative differences in brain structure between males and females, many of which are exacerbated in psychiatric and neurological conditions. Still, sex differences in MRI outcomes are understudied, particularly in clinical samples with known sex differences in disease risk, prevalence, and expression of clinical symptoms. Here we review the existing literature on sex differences in adult brain structure in normative samples and in 14 distinct psychiatric and neurological disorders. We discuss commonalities and sources of variance in study designs, analysis procedures, disease subtype effects, and the impact of these factors on MRI interpretation. Lastly, we identify key problems in the neuroimaging literature on sex differences and offer potential recommendations to address current barriers and optimize rigor and reproducibility. In particular, we emphasize the importance of large‐scale neuroimaging initiatives such as the Enhancing NeuroImaging Genetics through Meta‐Analyses consortium, the UK Biobank, Human Connectome Project, and others to provide unprecedented power to evaluate sex‐specific phenotypes in major brain diseases. Abstract

Background: Brain structure abnormalities throughout the course of Parkinson's disease have yet to be fully elucidated. Objective: Using a multicenter approach and harmonized analysis methods, we aimed to shed light on Parkinson's disease stage‐specific profiles of pathology, as suggested by in vivo neuroimaging. Methods: Individual brain MRI and clinical data from 2357 Parkinson's disease patients and 1182 healthy controls were collected from 19 sources. We analyzed regional cortical thickness, cortical surface area, and subcortical volume using mixed‐effects models. Patients grouped according to Hoehn and Yahr stage were compared with age‐ and sex‐matched controls. Within the patient sample, we investigated associations with Montreal Cognitive Assessment score. Results: Overall, patients showed a thinner cortex in 38 of 68 regions compared with controls (dmax = −0.20, dmin = −0.09). The bilateral putamen (dleft = −0.14, dright = −0.14) and left amygdala (d = −0.13) were smaller in patients, whereas the left thalamus was larger (d = 0.13). Analysis of staging demonstrated an initial presentation of thinner occipital, parietal, and temporal cortices, extending toward rostrally located cortical regions with increased disease severity. From stage 2 and onward, the bilateral putamen and amygdala were consistently smaller with larger differences denoting each increment. Poorer cognition was associated with widespread cortical thinning and lower volumes of core limbic structures. Conclusions: Our findings offer robust and novel imaging signatures that are generally incremental across but in certain regions specific to disease stages. Our findings highlight the importance of adequately powered multicenter collaborations. © 2021 The Authors. Abstract

Human brain structure changes throughout our lives. Altered brain growth or rates of decline are implicated in a vast range of psychiatric, developmental, and neurodegenerative diseases. While heritable, specific loci in the genome that influence these rates are largely unknown. Here, we sought to find common genetic variants that affect rates of brain growth or atrophy, in the first genome-wide association analysis of longitudinal changes in brain morphology across the lifespan. Longitudinal magnetic resonance imaging data from 10,163 individuals aged 4 to 99 years, on average 3.5 years apart, were used to compute rates of morphological change for 15 brain structures. We discovered 5 genome-wide significant loci and 15 genes associated with brain structural changes. Most individual variants exerted age-dependent effects. All identified genes are expressed in fetal and adult brain tissue, and some exhibit developmentally regulated expression across the lifespan. We demonstrate genetic overlap with depression, schizophrenia, cognitive functioning, height, body mass index and smoking. Several of the discovered loci are implicated in early brain development and point to involvement of metabolic processes. Gene-set findings also implicate immune processes in the rates of brain changes. Taken together, in the world’s largest longitudinal imaging genetics dataset we identified genetic variants that alter age-dependent brain growth and atrophy throughout our lives. One-sentence summary We identified common genetic variants associated with the rate of brain development and aging, in longitudinal MRI scans worldwide.

39. Using twin-pairs to assess potential bias in polygenic prediction of externalising behaviours across development.

Abstract

Prediction from polygenic scores may be confounded sources of passive gene-environment correlation (rGE; e.g. population stratification, assortative mating, and environmentally mediated effects of parental genotype on child phenotype). Using genomic data from 10,000 twin pairs, we asked whether polygenic scores from the recent externalising genome-wide association study predicted conduct problems, ADHD symptomology and callous-unemotional traits, and whether these predictions are biased by rGE. We ran regression models including within-family and between-family polygenic scores, to separate the direct genetic influence on a trait from environmental influences that correlate with genes (indirect genetic effects). Findings suggested that this externalising polygenic score is a good index of direct genetic influence on conduct and ADHD-related symptoms across development, with minimal bias from rGE, although the polygenic score predicted less variance in CU traits. Post-hoc analyses showed some indirect genetic effects acting on a common factor indexing stability of conduct problems across time and contexts.
