O estresse crônico está relacionado a um declínio precoce e exacerbado da função cognitiva em idosos, o qual parece ser mediado pelo cortisol. Entretanto, os estudos que procuram estabelecer associações entre os níveis de cortisol e desempenho cognitivo em idosos são controversos, em parte pelo tipo de amostra biológica que utilizam (soro, saliva urina), que não é capaz de monitorar alterações de longa duração na secreção de cortisol, e possivelmente por analisarem apenas o cortisol, ignorando esteroides neuroprotetores, como o DHEA-S. O presente estudo teve por objetivo verificar se a razão cortisol/DHEA-S capilar é um marcador mais sensível das disfunções cognitivas de idosos do que o cortisol capilar isoladamente. Idosos (n=59, 70.60 ± 7.74 anos) foram classificados de acordo com o Inventários de Sintomas de Estresse em dois grupos, estressado e não estressado (controle), e submetidos a uma bateria de testes neuropsicológicos que avaliam componentes da função executiva (Trail A e B, Span Direto e Reverso, Stroop, Fluencia Verbal e Fonêmica) e memória declarativa (Memória Lógica I e II).Amostras de cabelo foram coletadas para mensuração do cortisol e DHEA-S por Espectrometria de Massas em Tandem (LC-MS/MS). Os resultados obtidos indicaram que os idosos do grupo com estresse apresentaram performance cognitiva levemente inferior aos controles e níveis mais elevados de cortisol e da razão cortisol/DHEA-S. O cortisol mostrou uma associação mais robusta com o estresse do que a razão cortisol/DHEA. Entretanto, não foram encontradas associações significativas entre o cortisol ou a razão cortisol/DHEA com o declínio cognitivo dos idosos. Elderly of the stress group presented a slight cognitive impairment in comparison to the control group and higher levels of cortisol and cortisol/DHEA-S ratios. Cortisol showed a more robust association with stress than the cortisol/DHEA ratio. However, no significant associations were found between cortisol or the cortisol/DHEA ratio and cognitive performance in the elderly. In conclusion, capillary cortisol was a more sensitive marker of stress than the cortisol/DHEA-S ratio. Still, none of the measures was able to predict the cognitive performance of the elderly.

On the one hand, mild “healthy” stressors can be a motivator, leading to increased individual productivity and creativity. On the other hand, however, constant, excessive stress over a long period of time can impair not only the mind but the overall well-being of a person. The burden of aforementioned societal pressure is shown by the increasing number of stress-associated health issues around the globe. Mood-and psychiatric disorders are just a small part of the wide range of diseases related to stress. Biologically, the brain is the region that is most affected by highly stressful situations. Neurotransmitter signaling, neuroendocrine function, and neuronal signaling are lowered during periods of chronic stress, and if left unchecked, these neurologic changes are amongst the causes of depressive symptoms and the first step towards clinical depression. Alteration of brain-derived neurotrophic factor (BDNF) concentration is one of many biochemical changes associated with depression. The main goal of this thesis is to better understand how socially stressful situations are able to change BDNF concentration and modify depression‐related factors. O estresse é caracterizado como um conjunto de ações tomadas em reposta á situações que alterem a fisiologia normal do organismo. Quando crônico, leva á alterações da homeostasia corporal somado á prejuízos físicos e mentais. O isolamento social já foi apontado como uma causa de estresse para espécies de animais com características de convívio social. Essa privação social pode induzir danos ao organismo, com modificações no Sistema Nervoso Central (SNC), que muitas vezes culminam em quadros de ansiedade e depressão, assim como déficits de memória. As alterações do SNC podem ser associadas a variações moleculares, como do fator neurotrófico derivado do cérebro (BDNF), que é muito importante para a fisiologia e plasticidade neuronal. O suporte provido pela convivência social poderia atenuar os efeitos do estresse crônico e isolamento social, melhorando aspectos moleculares, físicos e cognitivos. O isolamento social de animais adultos ainda é pouco explorado na literatura científica, portanto o objetivo deste trabalho foi investigar os efeitos do isolamento e suporte social sobre: alterações moleculares dos níveis e expressão de BDNF no hipocampo; mecanismos epigenéticos de acetilação das histonas e metilação do DNA no hipocampo; sobre parâmetros comportamentais de memória e ansiedade em animais adultos cronicamente estressados, com idade de três e dezessetes meses. When chronic, the stress leads to alteration of body homeostasis, physical and mental impairment. Social isolation has already been pointed out as a cause of stress for animal species with social conviviality characteristics. The social deprivation can induce damages to the organism, leading to changes in the Central Nervous System (CNS), which can culminate in anxiety and depression, as well as memory deficits. These CNS changes may be associated with molecular variations, such as brain-derived neurotrophic factor (BDNF), which is very important for neuronal physiology and plasticity. The support provided by social coexistence can attenuate the effects of chronic stress and social isolation, improving physical and cognitive aspects. The social isolation of adult animals still is little explored in the scientific literature, so the objective of this work was to investigate the effects of isolation and social support on: level and expression of BDNF in the hippocampus; molecular mechanisms of histones acetylation and methylation of DNA in the hippocampus; on behavioral parameters of memory and anxiety in chronically stressed adult animals, aged three and seventeen months. The aim of the present study was to determine whether rapamycin, an autophagic inducer acting through the inhibition of the mTOR pathway, exerts neuroprotective activity in the cognitive dysfunction model induced by neonatal iron overload, and is able to improve memory. There are studies demonstrating that rapamycin is able to ameliorate the accumulation of toxic protein aggregates observed in neurodegenerative diseases, decrease cell death and cognitive symptoms related to these disorders. Autophagy is an event of cellular autodigestion and removal of components under physiological and pathological conditions. In the context of neurodegeneration, it is important for cell a high and refined control of autophagic regulation, so that the removal of protein deposits can be performed correctly. Moreover, it is already well understood that neonatal iron overload is related to several cognitive deficits, and the deposition of this metal occurs selectively and progressively with age, and may represent an important risk factor for the development of neurodegeneration in humans.Male Wistar rats received a single daily oral dose of vehicle or iron carbonyl (30 mg/kg) at postnatal days 12–14. At the age of 4 months, they received daily intraperitoneal injections of vehicle or rapamycin (0.25 mg/kg) for 14 days. The results showed that iron given in the neonatal period impaired inhibitory avoidance memory and induced a decrease in proteins critically involved in the autophagy pathway, Beclin 1 and LC3, in the hippocampus. Rapamycin in the adulthood reversed iron-induced memory deficits and recovered LC3 levels in iron treated rats. Our results suggest that iron accumulation, as observed in neurodegenerative disorders hinders autophagy, which might play a role in iron-induced neurotoxicity. Introdução: A atividade física tem sido proposta como uma intervenção promissora para melhorar a cognição e diminuir o risco de demência em adultos mais velhos. O fator neurotrófico derivado do cérebro (BDNF) parece mediar, pelo menos parcialmente, esses efeitos do exercício. Entretanto, os estudos de intervenção dos efeitos de exercícios multimodais sobre o desempenho cognitivo e os níveis de BDNF são escassos e compostos por pequenas amostras. Assim, a generalização das conclusões destes estudos depende da reprodutibilidade dos resultados. Objetivo: avaliar os efeitos de uma intervenção de atividade física composta pelo fortalecimento muscular e condicionamento aeróbico nos níveis de BDNF e cognição em mulheres idosas, de modo a contribuir para o avanço científico na área. Métodos: Idosas independentes e não demenciadas (≥ 75 anos) foram submetidas a uma intervenção de três meses de atividade física (n = 22, sessões de exercícios de 60 min três vezes por semana, sendo 30 min de fortalecimento muscular e 30 min de atividade aeróbia) ou a uma condição de controle (n = 10, sem exercício). Foram avaliados os parâmetros clínicos (sintomas de ansiedade e depressão), neuropsicológicos (testes de Span de Dígitos, Stroop, Trail Making e Memória Contextual), físicos (força dos membros superiores e inferiores, condicionamento aeróbico) e fisiológicos (BDNF sérico) no início do protocolo, 1 e 3 meses após o início da intervenção. Resultados: O grupo controle apresentou níveis estáveis ao longo do tempo para todas as variáveis medidas, ao passo que o grupo de intervenção melhorou a aptidão física, sintomas depressivos, desempenho cognitivo e níveis de BDNF. Além disso, uma regressão linear identificou uma associação entre o condicionamento aeróbio e os níveis de BDNF.Conclusão: A combinação de fortalecimento muscular e condicionamento aeróbio foi capaz de melhorar o desempenho cognitivo e aumentar os níveis de BDNF. Brain-derived neurotrophic factor (BDNF) seems to mediate, at least partially, these effects of exercise. However, intervention studies of the effects of multimodal exercises on cognition and BDNF levels are scarce and composed by small samples. Thus, the generalization of the conclusions of these studies depends on the reproducibility of the results. Objective: to contribute to the knowledge on the field, the present study evaluated the effects of a physical activity intervention composed by muscle strengthening and aerobic conditioning on BDNF levels and cognition in older women. Methods: Twenty-two independent and non-demented subjects (≥ 75 years) were assigned to a three-month physical activity intervention (60 min exercise sessions three times a week, 30 min of muscle strengthening and 30 min of aerobic activity) or to a control condition (n= 10, no exercise). The clinical parameters (anxiety and depression symptoms), neuropsychological (Digit Span, Stroop, Trail Making and Contextual Memory tests), physical (upper and lower limb strength, aerobic conditioning) and physiological (serum BDNF) parameters were evaluated immediately before, one and three-months after starting intervention. Results: the control group had stable levels for all measured variables, whereas the intervention group improved on physical fitness, depressive symptoms, cognitive performance and BDNF levels. Moreover, a linear regression identified an association between aerobic conditioning and BDNF levels. Conclusion: The combination of muscle strengthening and aerobic conditioning was able to improve cognitive performance and increase BDNF levels. Aerobic conditioning seems to be an important mediator of these results.

O envelhecimento normal é acompanhado por mudanças em habilidades cognitivas. É importante notar que o padrão e o grau de alterações cognitivas em idosos não demenciados são heterogêneos, enquanto alguns apresentam prejuízos significativos, outros mantêm a performance constante ao longo do envelhecimento. Entre os fatores que parecem ser capazes de modular a integridade cognitiva no envelhecimento está o estilo de vida. Há várias sugestões de que as atividades sociais, intelectuais e físicas melhoram a reserva cognitiva e protegem contra o declínio cognitivo. Os mecanismos relacionados a esses benefícios ainda são desconhecidos, mas estudos de intervenção com idosos sugerem que o BDNF (fator neurotrófico derivado do cérebro) pode ser um mediador dos benefícios na função cognitiva induzidos pela atividade física. Introdução: O aumento da expectativa de vida não vem necessariamente acompanhado de qualidade de vida. Dentre os principais problemas que impactam a qualidade de vida dos idosos estão as disfunções cognitivas. A literatura evidencia que o envelhecimento está relacionado: (1) as alterações estruturais, bioquímicas e de expressão gênica no encéfalo, (2) ao declínio funcional do córtex pré-frontal (CPF), (3) a déficits de memória contextual, um componente extremamente importante da memória episódica. Objetivos: Analisar os efeitos de uma estratégia de codificação associativa sobre a memória contextual imediata e tardia de idosos saudáveis. Metodologia: Participaram do estudo 43 idosos (idade ≥ 60 anos). Todos os sujeitos foram submetidos a avaliação neuropsicológica das funções do lobo frontal, composta pelos testes de Trilhas A e B, Span de Dígitos direto e inverso, e Stroop I, II e III. Para análise da memória contextual espacial os voluntários foram aleatoriamente divididos em dois grupos: reconhecimento imediato (5 min) e reconhecimento tardio (90 min). Para avaliação da estratégia de codificação cada um destes grupos foi subdividido em outros dois: um que realizou a tarefa com uma estratégia de codificação associativa e outro que fez a tarefa sem estratégia de codificação associativa. Os resultados foram analisados com GLMs mistas e univariadas. P<0.05 foi considerado estatisticamente significativo. Resultados: Desempenho da memória contextual manteve-se estável durante 90 minutos, mesmo na ausência de uma estratégia de codificação associativa no reconhecimento imediato e tardio. A introdução da instrução de codificação associativa melhora o reconhecimento da memória contextual imediata e tardia na mesma medida.Conclusão: Este estudo sugere que o aumento da memória contextual por instruções de codificação associativas incidentais é estável durante pelo menos 90 min , reforçando a noção de que déficits de memória contextual dos adultos mais velhos são reversíveis e, Introduction: Increased life expectancy is not necessarily accompanied by quality of life. The literature shows that aging is related to: (1) structural, biochemical and gene expression changes in the brain, (2) functional decline of the prefrontal cortex (PFC), (3) contextual memory deficits. Objectives: To analyze the effects of an associative encoding strategy on immediate and delayed contextual memory in the healthy elderly,. Methodology: The study included 43 elderly patients (≥ 60 years). All subjects underwent neuropsychological assessment including Trail test A and B, Forward and Backwards digit span, and Stroop word, color and word/color. To analyze the spatial contextual memory volunteers were randomly divided into two groups: immediate (5min) and delayed recall (90min). Participants of each group were assigned to one of two encoding conditions: whit or without an incidental associative instruction to encourage the association of the item to its spatial context. Data were analyzed with mixed and univariate GLMs. P<0.05 was considered statistically significant. Results: Performance on contextual memory was near chance in the immediate and delayed recognition without associative encoding condition. Introduction of the associative encoding instruction improved contextual memory in the immediate and delayed recognition to the same extent.Conclusion: This study suggests that the increase in contextual memory by incidental associative encoding instructions is stable for at least 90 min, reinforcing the notion that contextual memory deficits of older adults are reversible and potentially amenable to cognitive training.

The growth of elder population is a global occurrence and is followed by an increased prevalence and incidence of dementia, especially Alzheimer disease (AD). As the disease progresses, there is a larger demand for special care, task mainly performed by their family, who mostly are also elderly spouses of the patients. These caregivers are being constantly afflicted by chronic stress, which in turn can become harmful to their health in a number of disorders, such as cognitive impairment. Current literature highlights: (I) Relation between chronic stress, aging and cognitive impairment; (II) Relation between chronic stress and changes on hormone levels, such as cortisol and dehydroepiandrosterone (DHEA), and neurotrophic factors, i. e. Brain-Derived Neurotrophic Factor (BDNF); (III) BDNF, DHEA and cortisol potentially modulate cognitive-related mechanisms. In the present study, the evidences were assessed conjointly to better characterize physiological and pathological mechanisms involved in the association between chronic stress, aging and cognitive deficits. Chronic stress effects on frontal and temporal lobes-dependent cognitive functions were investigated in familial caregivers of AD patients (n=17; 32 to 84 years old). The impact of chronic stress on cortisol, DHEA and BDNF levels was also assessed, and the relation of these physiological features on cognitive performance. Initial findings point that chronic stress is capable of impairing attention, executive functions and declarative memory, also increasing cortisol/DHEA ratio and reducing serum BDNF levels. Then, we assessed age effects on the relation of chronic stress, cognition; cortisol, DHEA, and BDNF, as older caregivers are, theoretically, more prone to chronic stress effects than its younger counterparts are. Our results supports this hypothesis, indicating that hormone and cognitive changes are of greater importance on older caregivers when compared with young caregivers. À medida que a DA progride cresce a demanda por cuidados especiais, tarefa desempenhada em grande parte por cuidadores familiares, os quais muitas vezes são os cônjuges dos pacientes, ou seja, também são idosos. Estes cuidadores sofrem constantemente de estresse crônico, o qual é capaz de promover prejuízos à saúde num amplo espectro de disfunções, entre as quais as alterações cognitivas. A literatura evidencia: (I) relações entre estresse crônico, envelhecimento e alterações cognitivas; (II) relações entre estresse crônico e alteração dos níveis de hormônios, como o cortisol e a dehidroepiandrosterona (DHEA), e neurotrofinas, como o fator neurotrófico derivado de cérebro (BDNF); (III) o potencial do cortisol, DHEA e BDNF modularem mecanismos subjacentes a processos cognitivos. No presente trabalho estas evidências foram avaliadas de forma conjunta, de maneira a auxiliar na caracterização dos mecanismos fisiopatológicos envolvidos na associação entre estresse crônico, envelhecimento e déficits cognitivos. Os efeitos do estresse crônico sobre as funções cognitivas dependentes dos lobos frontais e temporais foram investigadas em cuidadores familiares de pacientes com DA (n= 17; 32 a 84 anos de idade). Foi analisado também o impacto do estresse crônico nos níveis de Cortisol, DHEA e BDNF, bem como a relação destes parâmetros fisiológicos com o desempenho em testes neuropsicológicos. Chronic sleep disorders are related to cognitive impairments and brain-derived neurotrophic factor (BDNF) alterations. However, the effects of acute sleep deprivation on BDNF levels and its relation with cognitive performance remains unknown. The objective was to investigate BDNF levels, cognitive performance and their relations in healthy subjects after acute sleep deprivation. In this study, nineteen sleep-deprived and twenty control subjects completed depression, anxiety and sleep quality questionnaires. Sleep deprived subjects spent a full night awake performing different playful activities to keep themselves awake. Attention, executive function and working memory (prefrontal cortex-dependent) were assessed with Stroop and Digit-span tests. Declarative memory (hippocampus-dependent) was assessed with Logical Memory test. Serum BDNF was measured by sandwich ELISA. Data were analyzed with independent samples T-test and curve estimation regressions. P<0. 05 was deemed statistically significant. Our results show that the sleep-deprived group had higher BDNF levels and normal performance on attention, executive function and working memory. However, declarative memory was impaired. A sigmoidal relation between BDNF and Stroop Test scores was found, showing that the test performance was greater when the BDNF levels were at its peak. These data showed that increased BDNF could be related, at least in part, to the maintenance of normal prefrontal cognitive functions after sleep deprivation. This potential relation should be further investigated., Transtornos crônicos do sono são relacionados a problemas cognitivos e alterações no Fator Neurotrófico Derivado do Cérebro (BDNF). No entanto, os efeitos da privação aguda do sono nos níveis de BDNF e sua relação com o desempenho cognitivo permanecem incertos. O objetivo deste estudo foi investigar a relação entre os níveis de BDNF no soro e desempenho cognitivo em sujeitos jovens saudáveis após uma privação de sono aguda. Neste estudo, 19 sujeitos privados de sono e 20 controles completaram questionários de depressão, ansiedade e qualidade do sono. O grupo privado passou uma noite acordado realizando atividades lúdicas para se manter acordado. Atenção, função executiva e memória de trabalho, dependentes do córtex pré-frontal, foram analisados com os testes de Stroop e Span de dígitos. Memória declarativa, dependente do hipocampo, foi analisada com o teste de Memória Lógica. O BDNF foi analisado por sandwich-ELISA. Os dados foram analisados com testes T para amostras independentes e regressões por estimativa de curva. P<0. 05 foi considerado estatisticamente significativo. Nossos dados mostraram que o grupo privado de sono apresentou maiores níveis de BDNF no soro. Atenção, função executiva e memória de trabalho não apresentaram diferença significativa entre grupos. No entanto, a memória declarativa foi prejudicada em indivíduos privados de sono. Foi encontrada uma relação sigmoidal entre o BDNF e o teste de Stroop, mostrando que o pico de performance neste teste está relacionado com os níveis mais altos de BDNF. Estes resultados mostram que o BDNF pode estar relacionado, em parte, com a manutenção das funções cognitivas normais no córtex pré-frontal após a privação de sono. Esta relação em potencial deve ser mais investigada.
