Together with the molecular knowledge of genes and proteins, biological images promise to significantly enhance the scientific understanding of complex cellular systems and to advance predictive and personalized therapeutic products for human health. For this potential to be realized, quality-assured image data must be shared among labs at a global scale to be compared, pooled, and reanalyzed, thus unleashing untold potential beyond the original purpose for which the data was generated. There are two broad sets of requirements to enable image data sharing in the life sciences. One set of requirements is articulated in the companion White Paper entitled Enabling Global Image Data Sharing in the Life Sciences, which is published in parallel and addresses the need to build the cyberinfrastructure for sharing the digital array data. In this White Paper, we detail a broad set of requirements, which involves collecting, managing, presenting, and propagating contextual information essential to assess the quality, understand the content, interpret the scientific implications, and reuse image data in the context of the experimental details. We start by providing an overview of the main lessons learned to date through international community activities, which have recently made considerable progress toward generating community standard practices for imaging Quality Control (QC) and metadata. We then provide a clear set of recommendations for amplifying this work. Antibodies are critical reagents to detect and characterize proteins. It is commonly understood that many commercial antibodies do not recognize their intended targets, but information on the scope of the problem remains largely anecdotal, and as such, feasibility of the goal of at least one potent and specific antibody targeting each protein in a proteome cannot be assessed. Focusing on antibodies for human proteins, we have scaled a standardized characterization approach using parental and knockout cell lines (Laflamme et al., 2019) to assess the performance of 614 commercial antibodies for 65 neuroscience-related proteins. Side-by-side comparisons of all antibodies against each target, obtained from multiple commercial partners, have demonstrated that: (i) more than 50% of all antibodies failed in one or more applications, (ii) yet, ~50-75% of the protein set was covered by at least one high-performing antibody, depending on application, suggesting that coverage of human proteins by commercial antibodies is significant; and (iii) recombinant antibodies performed better than monoclonal or polyclonal antibodies. The hundreds of underperforming antibodies identified in this study were found to have been used in a large number of published articles, which should raise alarm. Encouragingly, more than half of the underperforming commercial antibodies were reassessed by the manufacturers, and many had alterations to their recommended usage or were removed from the market. This first study helps demonstrate the scale of the antibody specificity problem but also suggests an efficient strategy toward achieving coverage of the human proteome; mine the existing commercial antibody repertoire, and use the data to focus new renewable antibody generation efforts.

10. Influence of microbial community on nitrogen loss through denitrification in agriculture

- Abstract
Global food demand is expected to rise by up to 62% by 2050, with an increasing population and finite land available for food production. Denitrification causes loss of fixed nitrogen from soil systems, thereby reducing crop productivity. Current nitrogen cycling models cannot fully explain the variation in nitrogen use efficiency at the field scale; this variation is thought to be associated with microbial community dynamics. The dynamics of denitrification and denitrifying communities are affected by the physicochemical properties of the soil, and therefore the land management practices which alter them. In this thesis, we look at the effects of land management history and innate environmental variation using a long-term factorial field trial, a working commercial farm, and laboratory-based experiments. By studying the denitrifier community composition, abundance and diversity in the context of environmental data, we were able to address the question of how specific land management practices and innate spatiotemporal variation affects the distribution of denitrifiers within arable soils. It was found that bacterial and denitrifier communities are largely unaffected by land management history, and cluster predominantly along environmental gradients. Additionally, there was a high degree of spatiotemporal variation in soil denitrifying communities, with implications for future sampling strategies as well as computer modelling. Finally, it was established that isolation of bacteria from the environment is a viable method for culturing non-model organisms that can be characterized and used to inform nitrogen-cycling models. In this combination of well-established agricultural field trial and working commercial farm, the major identifiable determinant of bacterial community dissimilarity was a blend of physicochemical factors. The importance of selection on bacterial communities, relative to ecological drift, may have consequences for attempts to understand and model nutrient availabilities in arable farming systems, and improve agricultural productivity to feed the growing global population.

- Abstract
Background: During the COVID-19 pandemic, social distancing measures were enforced and the national lockdown underlined our reliance on virtual means as a way to communicate. This new way of interacting highlighted that people with an intellectual disability were a large proportion of a digitally excluded population. Methods: A service evaluation, using a mixed method design in the form of four self-reported outcome measures and qualitative feedback. Findings: Clinical services need to continue when face to face sessions are not possible. Remote groups can be an alternative option not only when self-isolating due to pandemics but when living in remote locations, having physical health problems or excessive expenses and travel costs. Conclusions: Online dramatherapy groups can be a beneficial alternative when face to face groups are not possible or challenging to attend due to access difficulties. Online groups can offer opportunities to meet with peers, build relationships, improve confidence and learn new skills in technology.

In April 2020, the QUality Assessment and REProducibility for Instruments and Images in Light Microscopy (QUAREP-LiMi) initiative was formed. This initiative comprises imaging scientists from academia and industry who share a common interest in achieving a better understanding of the performance and limitations of microscopes and improved quality control (QC) in light microscopy. The ultimate goal of the QUAREP-LiMi initiative is to establish a set of common QC standards, guidelines, metadata models, and tools, including detailed protocols, with the ultimate aim of improving reproducible advances in scientific research. This White Paper 1) summarizes the major obstacles identified in the field that motivated the launch of the QUAREP-LiMi initiative; 2) identifies the urgent need to address these obstacles in a grassroots manner, through a community of stakeholders including, researchers, imaging scientists, bioimage analysts, bioimage informatics developers, corporate partners, funding agencies, standards organizations, scientific publishers, and observers of such; 3) outlines the current actions of the QUAREP-LiMi initiative, and 4) proposes future steps that can be taken to improve the dissemination and acceptance of the proposed guidelines to manage QC. Biological and biomedical imaging datasets record the constitution, architecture and dynamics of living organisms across several orders of magnitude of space and time. Imaging technologies are now used throughout the life and biomedical sciences to achieve discovery and understanding of biological mechanisms in the basic sciences as well as assessment, diagnosis and therapeutic intervention in clinical trials and animal and human medicine. The universal application and use of imaging raises an important question and opportunity: what is the value and ultimate destination of biological and medical imaging data? In the last few years, several informatics and data science technologies have matured sufficiently so that routine publication of these datasets is now possible. Participants in Global BioImaging from 15 countries and all populated continents have agreed on the need for recommendations and guidelines for the establishment of image data repositories and the formats they use for delivering data to the global scientific community. This white paper presents a shared, global view of criteria for these common, globally applicable guidelines and provisional proposals for open tools and resources that are available now and can provide a foundation for future development.

17. EnGendering inclusive practice : exploring trans people's experiences of adoption and fostering in England and Wales

This thesis presents the findings of a narrative inquiry exploring the lived experiences and perceptions that trans and non-binary people have in relation to adopting or fostering a child in England and Wales. Since the 1950s, adoption and fostering agencies have acted as 'gatekeepers', deciding who is deemed a suitable parent/carer and who is not. Adoption and fostering agencies have tended to espouse gender normative views in their practices. This amounts to covert discrimination as these practices disadvantage people who identify outside of normative frameworks of gender. This research addresses a gap in the research, bringing trans people's voices into fostering and adoption social work in England and Wales. The results of ten in-depth semi-structured interviews are examined. These include eight trans people who have adopted/fostered or who hope to adopt/foster and two social workers who have been involved in recruiting, assessing or supporting trans carers. Narratives of personal identity, family, fostering and adoption are explored within this thesis and analysed using a theoretical framework which includes cisgenderism, stigma and discourses of power to explicate the sociology of trans people's experiences. Cisgenderism was found to operate in different ways upon different experiences, hindering the life and family opportunities of trans and non-binary people. The negative effects of cisgenderism could be especially pronounced for those whose identities did not align with a binary designation of gender. Findings and conclusions are used to make recommendations for good practice in social work and to suggest future research directions.

The application of microscopy in biomedical research has come a long way since Antonie van Leeuwenhoek discovered unicellular organisms. Countless innovations have positioned light microscopy as a cornerstone of modern biology and a method of choice for connecting omics datasets to their biological and clinical correlates. Still, regardless of how convincing published imaging data looks, it does not always convey meaningful information about the conditions in which it was acquired, processed, and analyzed. Adequate record-keeping, reporting, and quality control are therefore essential to ensure experimental rigor and data fidelity, allow experiments to be reproducibly repeated, and promote the proper evaluation, interpretation, comparison, and re-use. To this end, microscopy images should be accompanied by complete descriptions detailing experimental procedures, biological samples, microscope hardware specifications, image acquisition parameters, and image analysis procedures, as well as metrics accounting for instrument performance and calibration. However, universal, community-accepted Microscopy Metadata standards and reporting specifications that would result in Findable Accessible Interoperable and Reproducible (FAIR) microscopy data have not yet been established. To understand this shortcoming and to propose a way forward, here we provide an overview of the nature of microscopy metadata and its importance for fostering data quality, reproducibility, scientific rigor, and sharing value in light microscopy. The proposal for tiered Microscopy Metadata Specifications that extend the OME Data Model put forth by the 4D Nucleome Initiative and by Bioimaging North America [1-3] as well as a suite of three complementary and interoperable tools are being developed to facilitate the process of image data documentation and are presented in related manuscripts [4-6].

22. Early Career Teacher Professional Development: Bridging Simulation Technology with Evidence-Based Behavior Management

Early career teachers working in high poverty schools face of overwhelming challenges navigating disruptive behaviors with studies highlighting behavior problems as one of the strongest predictors of turnover (Ingersoll & Smith, 2003). Simulation-based technology leverages important pedagogical strengths (e.g., realistic training context, opportunities for practice and feedback, low-stakes training) that may confer advantage over traditional training for teachers in behavior management. This manuscript illustrates the development and refinement of Interactive Virtual Training for Teachers (IVT-T), a simulation-training model to help early career teachers hone their behavior management skills by interacting with virtual students in a 3D classroom. Advisory board members (N=5) with extensive experience in high poverty schools provided iterative feedback on the authenticity and realism of IVT-T students, classrooms, and vignettes. Advisory board members rated authenticity/realism of the students, classrooms, and vignettes favorably and ratings also increased over time as new prototypes were shared. In addition, prototypes introduced earlier underwent more revisions than prototypes introduced later suggesting advisory board feedback helped the computer science team improve upon their development approach. Open-ended feedback revealed important shortcomings and the need for further refinements to improve the training model. Implications for teacher educators and researchers involved in the development of instructional technologies are discussed. 31. Between a rock and a hard place : an insight into the psychological therapist's experience of having safeguarding concerns for their clients' children

This study explores the therapists' experience when managing safeguarding concerns about their clients' children, with the aim of raising awareness and bringing the child into the therapists' mind. A review of the literature indicated that this is an undeveloped area, with existing research lacking attention to the impact of the therapeutic relationship on this ethical dilemma. IPA (interpretative phenomenological analysis) was used to analyse the findings from six semistructured interviews with a mix of psychological therapists who had experienced concerns for their clients' children while working in non-NHS settings. This approach allows for an in-depth exploration into the subjective experience of the participant, while also recognising the reflective role of the researcher. The findings indicate that therapists are struggling with a lack of confidence and experience in managing their child protection concerns, with an indication of a training need. This lack of confidence, combined with a range of complex emotions evoked in such work, increased the participants' need for support and supervision. Participants felt the need to alter their therapeutic practice and become more directive in order to ascertain clearer details about their concerns. The findings have implications for the training and continued professional development requirements for psychological therapists. There is an indication for training focused on managing concerns in the context of the therapeutic relationship, more training around assessing the level of risk and the development of a prompt sheet with reflective questions to aid therapists in their thinking. An identified limitation of this study is the range of participants in terms of gender and ethnicity; further research could explore more thoroughly whether these factors influence how concerns are managed. There is also scope to explore more specifically the impact of the therapeutic placement, to identify whether those working within large organisations, such as the NHS, feel more supported and contained in this type of work than those working more independently.

This paper aims to contribute fresh insights into youth participatory action research (YPAR) by using bell hooks' engaged pedagogy to illuminate the process of co-designing a program for transition beyond secondary school. Engaged pedagogy is a critical pedagogy that combines critical consciousness and radical wholeness and seeks to foster a learning community. The 10-week YPAR included six staff collaborators (SCs) and five youth collaborators (YCs). Data comprised recordings of weekly collaborative group meetings; group interviews with SCs and YCs; and reflections and artefacts such as planning documents, graphic organisers and photographs. Informed by engaged pedagogy, findings are represented in two themes. First, YCs and SCs raised critical consciousness by collectively unpacking and critiquing the concept of transition. Critical consciousness allowed YCs to share their lived experiences and critique the dominant deficit discourse that represents transition as a linear process that needs to be smoothed by expert adults. Second, YCs and SCs demonstrated what hooks describes as radical wholeness, by bringing their whole selves to the YPAR. While some SCs and YCs struggled with the messiness of co-design as they negotiated expectations and roles, they learned to be honest and share their emotions, becoming what hooks describes as a learning community. Using an engaged pedagogy lens, we conclude by advocating for a holistic approach for YPAR in which educators take the risk of being vulnerable, sharing uncertainty and discomfort while encouraging youth to also take risks by bringing their whole selves to the process. Design thinking is often proposed as an educative strategy to enhance growth mindset, problem solving, and creativity, but there are very few evaluations of the impact of experiential design thinking programs. The primary aim of this study was to investigate whether an online design-thinking workshop provided to outdoor education staff could improve their design-thinking traits. The pilot program was delivered over a 6-week period and comprised four, 2-hour sessions, in which teams worked together to actively engage in the steps of the design thinking process. Significant improvements were observed in participants' growth mindset and design thinking process—specifically, feedback seeking. Furthermore, design thinking was strongly correlated with growth mindset and cognitive flexibility; two constructs previously theorised to be associated with design thinking. This study provides a framework for constructing online design thinking workshops and how to effectively evaluate participant outcomes. [ABSTRACT FROM AUTHOR]
