Abstract

In April 2023, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by one new family, 14 new genera, and 140 new species. Two genera and 538 species were renamed. One species was moved, and four were abolished. This article presents the updated taxonomy of Negarnaviricota as now accepted by the ICTV.

Abstract

In March 2022, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by two new families (bunyaviral Discoviridae and Tulasviridae), 41 new genera, and 98 new species. Three hundred forty-nine species were renamed and/or moved. The accidentally misspelled names of seven species were corrected. This article presents the updated taxonomy of Negarnaviricota as now accepted by the ICTV.

5. The use of stinging nettle tea (Urtica sp.) to control Aulacorthum solani and Macrosiphum euphorbiae on Ranunculus asiaticus

Abstract

Natural and environmental-friendly approaches to control pests and diseases in horticultural production systems are showing an increasing trend. Biological alternatives, such as botanical extracts and basic substances, show promise in the reduction of the necessity for conventional plant protectants. In ornamentals, less is known about the usage, behaviour and potential of botanicals and basic substances for plant protection. In two trials, we investigated the effect of a weekly foliar spray of an aqueous extract of nettle (Urtica sp.; dried material 15 g · L–1) on two aphids, Macrosiphum euphorbiae and Aulacorthum solani. Experiments were conducted in the greenhouse with Ranunculus asiaticus as hostplant. The Urtica tea and azadirachtin (NeemAzal-T/S) as a standard used in common horticultural production were compared with the water control. Both previously infested and noninfested plants were built up to examine the effect on aphid population growth and migration. As a result, we showed that both treatments, Urtica tea and azadirachtin, reduce the aphid density significantly in comparison with the water control, although the effect of the Urtica tea was not as substantial as that of azadirachtin. Contrarily, treatments could not prevent aphid migration on previously noninfested plants. The study demonstrates that the basic substance Urtica tea can be suitable for the reduction of aphid pests in ornamental plant production, and thus, reduce the amount of synthetic plant protectants in horticulture.

6. Cherry leaf roll virus – an emerging virus in Finland?

Abstract

Cherry leaf roll virus, CLRV, is a plant pathogen that infects a variety of deciduous trees and shrubs in temperate regions. Little is known about its occurrence at high latitudes and especially in Finnish birch species. Still, symptoms that seemed to be associated with CLRV such as vein banding, leaf roll and decline have been observed in birch trees throughout the country since the summer of 2002. Six different birch species, subspecies or varieties, i.e. Betula pubescens subsp. pubescens (downy birch), B. pendula (silver birch), B. nana (dwarf birch), B. pubescens var. appressa (Kiilopää birch), B. pubescens subsp. czerepanovii (mountain birch) and B. pendula var. carelica (curly birch) originating from all over Finland were assessed by immunocapture-reverse transcription-polymerase chain reaction (IC-RT-PCR) for CLRV infection. It was shown that CLRV is widely distributed in B. pendula and B. pubescens throughout the country. Furthermore, dwarf birch, mountain birch, Kiilopää birch and curly birch were confirmed to be previously unkown hosts of CLRV. Genetic analysis of virus sequence variants originating from Finnish birch trees revealed atypical phylogenetic relationships. In contrast to CLRV isolates from birches growing in the United Kingdom and Germany which clustered exclusively within group A, Finnish CLRV isolates belonged either to group B, D or E. Thus, virus population structure in Finnish birches seems to be more variable and host plant dependency seems not to apply for Finnish CLRV isolates.

7. Cherry leaf roll virus abundant on Betula pubescens in Finland

Abstract

Virus-related symptoms such as vein banding, leaf roll, chlorosis and subsequent necrosis on birch leaves were increasingly recorded throughout Finland since 2002. They are widespread in this country and have also been detected in northern Norway and Sweden. Symptomatic foliage has so far been found on Betula pendula, B. pubescens, B. pubescens subsp. czerepanovii, and B. nana. A Cherry leaf roll virus (CLRV) specific IC-RT-PCR was applied to young leaves, buds and catkins of symptomatic shoots of nineteen pubescent and one silver birch trees grown in the centre of Rovaniemi, Finland. CLRV was found in seventeen B. pubescens trees. This is the first time that B. pubescens has been confirmed to be a host species for CLRV in Finland. Nor has CLRV been recorded earlier in northern Finland.

Abstract

Purpose of Review: The collective virome of forest trees can be considered to include not only plant viruses, but also viral communities harbored by all tree-associated organisms. In this review, we will concentrate on reviewing recent developments in the two fields of forest tree virology that have received the most research input during the last 5 years: (1) current knowledge of virus diseases affecting forest trees and their causal agents and (2) fungal viruses (mycoviruses) and properties that are required for utilizing them for biocontrol purposes. Recent Findings: The discovery of plant and mycoviruses has been revolutionized during the last few years due to high-throughput sequencing (HTS). This has altered our view of virus diversity and prevalence, but also their host ranges, transmission routes, and host effects. Utilization of HTS has greatly expanded our knowledge of plant virus diversity and disease etiology in forest trees and revealed the commonness of cross-kingdom transmission events between fungi, oomycetes, plants, and arthropods. Research on mycoviruses has also identified several new mycoviruses that restrict the growth or virulence of forest pathogenic fungi. 18. A Combined Cleaning and Disinfection Measure to Decontaminate Tire Treads from Tomato Brown Rugose Fruit Virus.

Abstract

Mechanically transmissible and stable viruses such as tobamoviruses, which include Tobamovirus fructirugosum (syn. tomato brown rugose fruit virus (ToBRFV), will continue to pose major challenges for farmers. Consequently, holistic hygiene concepts are being implemented to prevent the introduction and spread of these viruses. The decontamination of tires and castors was previously a weak point in many industrial hygiene concepts. For this reason, the ProfilGate clean-off zone was tested in combination with the disinfectant MENNO Florades for the decontamination of ToBRFV-contaminated tires. In total, 478 tire segments were sampled to evaluate the contamination of ToBRFV and the following decontamination of the tires. This treatment reliably removed high (4.5 µg/cm2), medium (0.45 µg/cm2), and low concentrations (0.045 µg/cm2) of ToBRFV from the tires, as shown by a bioassay. The reduction in necrotic local lesions on susceptible indicator plants N. tabacum cv. Xanthi NN was between 91.9 and 97.6%. The reduction in ToBRFV contamination largely depended on the length of the rollover distance, i.e., the number of tire rotations. For transport trolleys with polyamide and rubber tires, depletions of 97.4 and 97.6%, respectively, was determined after 16 rotations. For transport wagons with tires twice the size and polyurethane tread, the depletion was still at least 91% after eight wheel turns. Even in the case of gross soiling of the tires, the mean reduction from the different tread materials was 80.9 to 98.9%. Subsequent analysis of the clean-off zone revealed that ToBRFV did not accumulate, even when the contaminated tires were driven over several times, but was safely inactivated completely in the disinfectant solution. Abstract

Plant diseases are mostly multicausal with several factors influencing the health status of affected hosts. Common ash (Fraxinus excelsior), a significant tree species of European forests, is currently mostly endangered by ash dieback, caused by the invasive fungus Hymenoscyphus fraxineus. However, contributing factors, including pathogenic viruses, are poorly understood. Here, we report the results of a virus screening conducted on selected special stands of F. excelsior. Over three consecutive years, ash trees from different origins were tested, including leaf material from mature seed trees, young trees and ash seedlings from the natural regeneration. Using RT-PCR, we screened for five viruses, including the generalist species ArMV (Nepovirus arabis) and CLRV (Nepovirus avii), as well as newly discovered viruses in ash, including the emaravirus ASaV (Emaravirus fraxini), the idaeovirus PrLBaV (Idaeovirus ligustri), and cytorhabdoviruses. The results revealed a high virus diversity in common ash. An association of ASaV detection with specific leaf symptoms, including shoestring, chlorotic ringspots, and vein yellowing, was documented. An analyses of relevant gene products of cytorhabdoviruses obtained from ashes of different sites revealed sequence diversities and two distinct phylogenetic groups present in ash populations. Signatures of novel viruses from different families have been identified by high-throughput sequencing. Together, our results provide insights into the virus diversity and distribution of viruses in ash and expand our knowledge about the virome of this endangered tree species. [ABSTRACT FROM AUTHOR]

Abstract

In April 2023, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by one new family, 14 new genera, and 140 new species. Two genera and 538 species were renamed. One species was moved, and four were abolished. This article presents the updated taxonomy of Negarnaviricota as now accepted by the ICTV.

Abstract

To adapt to climate change, several agricultural strategies are currently being explored, including a shift in land use areas. Regional differences in microbiome composition and associated phytopathogens need to be considered. However, most empirical studies on differences in the crop microbiome focused on soil communities, with insufficient attention to the phyllosphere. In this study, we focused on wheat ears in three regions in northeastern Germany (Magdeburger Börde (MBB), Müncheberger Sander (MSA), Uckermärkisches Hügelland (UKH)) with different yield potentials, soil, and climatic conditions. To gain insight into the fungal community at different sites, we used a metabarcoding approach (ITS-NGS). Further, we examined the diversity and abundance of Fusarium and Alternaria using culture-dependent and culture-independent techniques. For each region, the prevalence of different orders rich in phytopathogenic fungi was determined: Sporidiobolales in MBB, Capnodiales and Pleosporales in MSA, and Hypocreales in UKH were identified as taxonomic biomarkers. Additionally, F. graminearum was found predominantly in UKH, whereas F. poae was more abundant in the other two regions. Environmental filters seem to be strong drivers of these differences, but we also discuss the possible effects of dispersal and interaction filters. This article was supported by the German Research Foundation (DFG) and the Open Access Publication Fund of Humboldt-Universität zu Berlin., After entry of a quarantine/regulated pathogen, infected plants shall be destroyed, and the cultivated area (e.g., greenhouse) shall be disinfected. Therefore, the selection of an effective disinfectant plays an important role. With the availability of different methods for virus quantification, we investigated the application of quantitative ELISA (qELISA), RT-qPCR (reverse transcription-quantitative polymerase chain reaction), and bioassays for the quantification of disinfectant efficacy. Therefore, we estimated the titer reduction in tomato brown rugose fruit virus (ToBRFV), a regulated pathogen, in plant sap and on germ carriers after treatment with MENNO Florades 4% for 16 h. The virus load before and after the treatment was measured with the mentioned methods. The RT-qPCR and qELISA methods showed very low efficacy in the presence of the disinfectant. Although bioassays are time-consuming, need purified particles for establishing the quantification models, and are less sensitive than RT-qPCR, they were able to quantify the differences in virus titer in the presence/absence of disinfectant. Interestingly, the bioassays reached at least the lower limit sensitivity of a qELISA. 39. Between Habitats: Transfer of Phytopathogenic Fungi along Transition Zones from Kettle Hole Edges to Wheat Ears

Abstract

Kettle holes are able to increase the soil and air humidity around them. Therefore, they create a perfect habitat for phytopathogenic fungi of the genera Fusarium and Alternaria to develop, sporulate, and immigrate into neighboring agricultural fields. In our study, we establish transects from the edges of different kettle holes and field edges up to 50 m into the fields to analyze the abundance and diversity of pathogenic fungi in these transition zones by culture-dependent and culture-independent methods. However, in 2019 and 2020, low precipitation and higher temperatures compared to the long-time average were measured, which led to limited infections of weeds in the transition zones with Fusarium and Alternaria. Therefore, the hypothesized significantly higher infection of wheat plants next to the kettle holes by a strong spread of fungal spores was not detected. Infestation patterns of Fusarium and Alternaria fungi on weeds and wheat ears were spatially different. In total, 9 different Fusarium species were found in the transition zone. The species diversity at kettle holes differed from 0 to 6 species. Abstract

In April 2023, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by one new family, 14 new genera, and 140 new species. Two genera and 538 species were renamed. One species was moved, and four were abolished.

41. Analysis of the Spatial Dispersion of Tomato Brown Rugose Fruit Virus on Surfaces in a Commercial Tomato Production Site

Abstract

The tomato brown rugose fruit virus (ToBRFV) causes severe damage to tomato cultivars and has international economic importance. The harmful tobamovirus is easily mechanically transmissible and highly stable. An ongoing cultivation of infected tomato plants may lead to the spread of ToBRFV in and around the production area of the infested tomato farm. We conducted a study in which we collected a representative number of swab samples from various inanimate surfaces in greenhouses, packaging halls, and shared and private accommodations. In addition, numerous fabrics, such as outer clothing, bed linen, and items used by greenhouse workers, were tested. The infectivity of ToBRFV-contaminated surfaces was tested in bioassays using Nicotiana tabacum cv. Xanthi NN and confirmed using DAS-ELISA. The proportion of ToBRFV-contaminated surfaces varied among locations, from 48.7% in greenhouses to 0% in offices with limited access to staff. Samples from shared accommodation and private accommodation were 18.4% and 3.6% ToBRFV positive, respectively. Clothing and protective items were found to be highly contaminated with ToBRFV, and even around the sleeping area, infective ToBRFV was detected in a few apartments. This study provides evidence for the first time on how and where infectious ToBRFV can be spread by humans beyond the production area. Abstract

Aspen mosaic-associated virus (AsMaV) is a newly identified Emaravirus, in the family Fimoviridae, Bunyavirales, associated with mosaic symptoms in aspen trees (Populus tremula). Aspen trees are widely distributed in Europe and understanding the population structure of AsMaV may aid in the development of better management strategies. The virus genome consists of five negative-sense single-stranded RNA (–ssRNA) molecules. To investigate the genetic diversity and population parameters of AsMaV, different regions of the genome were amplified and analyzed and full-length sequence of the divergent isolates were cloned and sequenced. The results show that RNA3 or nucleoprotein is a good representative for studying genetic diversity in AsMaV. Developed RT–PCR–RFLP was able to identify areas with a higher number of haplotypes and could be applied for screening the large number of samples. In general, AsMaV has a conserved genome and based on the phylogenetic studies, geographical structuring was observed in AsMaV isolates from Sweden and Finland, which could be attributed to founder effects. The genome of AsMaV is under purifying selection but not distributed uniformly on genomic RNAs. The assumed genome organization is 3′-N-P-P3-M-G-p6-p6’-L-5′. The N gene encodes the putative nucleoprotein (59.1 kDa), P encodes the phosphoprotein (34.7 kDa), P3 encodes the movement protein (23.1 kDa), M encodes the matrix protein (23.1 kDa), G encodes a glycoprotein (64.4 kDa), and L encodes the viral RNA polymerase (247 kDa). 44. Pre- and Post-Harvest Infection of Pasteurized Pickles with Fungi and Their Pectinolytic Potential to Soften the Product

Abstract

Fungi and their enzymes have long been thought to cause the softening of pasteurized gherkins; however, the exact fungal species and timing of contamination are unknown. Ready-to-sell pickle jars and blossoms of growing gherkins were inoculated with DNA-sequenced fungi isolated from rotting gherkins to cause softening at various stages of production. Ready-to-sell gherkins inoculated with Fusarium oxysporum, Fusarium equiseti, Galactomyces geotrichum, Mucor circinelloides, Mucor hiemalis, Mucor fragilis, Plectosphaerella cucumerina, Alternaria sp., and Cladosporium sp. indicated a measurable texture reduction after pasteurization and 6 months of storage at room temperature. No texture changes were observed in gherkins infected during the growth phase. The fungi M. hiemalis, M. fragilis, and G. geotrichum tolerated the acidic-saline (approx. pH 4) environment in the jar for several days, thus the pectinolytic enzymes of these candidates were tested for heat and pH resistance. Although the measured endo-Polygalacturonase (PG) of M. fragilis had its optimum activity at pH < 4, all fungal enzymes were inactivated within 3 min at 80 °C corresponding to the pasteurization heat. Our study shows that conventionally occurring fungi and their enzymes have the potential to induce softening in pickles. Abstract

In April 2023, following the annual International Committee on Taxonomy of Viruses (ICTV) ratification vote on newly proposed taxa, the phylum Negarnaviricota was amended and emended. The phylum was expanded by one new family, 14 new genera, and 140 new species. Two genera and 538 species were renamed. One species was moved, and four were abolished. This article presents the updated taxonomy of Negarnaviricota as now accepted by the ICTV.
