A fine characterization of the HLA molecular profiles of populations from distinct origins and ethnical back-grounds is essential for both clinical issues in histocompatibility and transplantation and fundamental research in population and evolutionary genetics. Recent developments of high throughput sequencing technologies(including NGS) have allowed generating high resolution data at most HLA loci for thousands of individuals at rather accessible costs, leading many laboratories to type or retype large population samples with the aim to fill gaps in the landscape of HLA-typed populations, thereby significantly improving the quality of the molecular data available. Under the umbrella of the EFI Population Genetics Working Group and the Population Genetics, Anthropology and Evolution (PGAE) Component of the 18th International HLA & Immunogenetics Workshop, as many as 30 laboratories were involved in a collective effort to improve the HLA molecular map of a large geographic region encompassing Europe, North Africa, West Asia and South Asia and also including populations of European and mixed descent from North, Central and South America. In addition to 15populations represented by very large samples (in total more than 270, 000 individuals) typed at the 2nd-fieldlevel of resolution, the final dataset includes20 populations sequenced by NGS and reported at the4th-field level for up to 11 HLA loci. Statistical and graphical analyses of this newly generated dataset, combined with previously available worldwide data, reveal a much more precise topography of the HLA molecular landscape across populations and hence also improve the reference datasets used in histocompatibility and transplantation. The observed HLA molecular diversity is further contrasted to that of equivalent MHC-Patr loci analyzed in several chimpanzee cohorts (with implementation of new sequence data), enabling novel interpretations of the genetic history and evolution of the secrucial immune genes.

La recherche des ANCA est utilisée pour le diagnostic et le suivi de l'activité des vasculites associées aux ANCA, mais aussi parfois pour d'autres indications plus controversées. La place de l'immunofluorescence indirecte dans l'algorithme de dépistage et de suivi des vasculites à ANCA est contestée depuis plusieurs années et a récemment fait l'objet de nouvelles recommandations internationales. Cette nouvelle approche améliore certainement le rendement de ce test, en particulier en Suisse où, en raison du grand nombre d'analyses demandé, la probabilité prétest d'une vasculite à ANCA est relativement faible. Cependant, nous discutons dans cet article les possibles failles de la nouvelle approche en nous basant sur deux cas récents au laboratoire et rendons les prescripteurs attentifs sur l'importance de la communication avec le laboratoire afin d'améliorer l'interprétation des résultats.

Human leukocyte antigen (HLA) genes are very informative in population genetics studies and their variability has been widely used to reconstruct the history of geographic and/or demographic expansions of human populations. The characterization of HLA diversity at the population level is also fundamental in clinical studies, particularly for bone marrow transplantation programs. In this study, we investigated the HLA molecular variation in Rio Grande do Sul, South Brazil, in order to identify possible regional differences across this state. More than 97,000 bone marrow donors were typed at the HLA- A, -B and -DRB1 loci and analyzed by considering two kinds of subdivisions based on both self-identified ethnicity and place of residence: (a) the official geographic subdivision defined by the Brazilian Institute of Geography and Statistics and (b) known information about the colonization history of the state. HLA allele and haplotype frequencies were estimated and compared among the defined subgroups. The results indicate a lack of correlation between genetic variation and geography and thus no clear HLA genetic structure based on geographic criteria. On the other hand, major differences were observed regarding ethnicity. In addition, local populations from Rio Grande do Sul were found to be genetically similar to their corresponding parental European populations from Germany, Italy and Portugal, as documented by historical data. Overall, this study provides a thorough characterization of the HLA genetic variation in Rio Grande do Sul and a better understanding of its demographic history, being most useful for the development of more efficient strategies in bone marrow donors' recruitment.

The diversity of killer-cell immunoglobulin-like receptors (KIR) genes was evaluated in Portuguese and the observed genotypic profiles were found related to the ones reported in European populations. The KIR repertoire after hematopoietic stem cell transplantation is determined by these gene frequencies and the KIR group B motifs are the less common. We estimated donor–KIR/recipient–ligand interactions in transplants with related donors and unrelated donors found in a local registry or from abroad. A large fraction of transplants had all three ligands of inhibitory receptors, and therefore, in theory were not prone to natural killer cell (NK) mediated alloreactivity. Furthermore, the distribution of KIR alloreactive interactions was found independent of the donor–recipient genetic proximity, probably because of different gene segregation and comparable KIR frequencies in the donor pools.

BACKGROUND & AIMS The interaction of KIR with their HLA ligands drives the activation and inhibition of natural killer (NK) cells. NK cells could be implicated in the development of liver fibrosis in chronic hepatitis C. METHODS We analysed 206 non-transplanted and 53 liver transplanted patients, selected according to their Metavir fibrosis stage. Several variables such as the number of activator KIR or the HLA ligands were considered in multinomial and logistic regression models. Possible confounding variables were also investigated. RESULTS The KIRs were not significant predictors of the fibrosis stage. Conversely, a significant reduction of the HLA-C1C2 genotype was observed in the most advanced fibrosis stage group (F4) in both cohorts. Furthermore, the progression rate of fibrosis was almost 10 times faster in the subgroup of patients after liver transplantation and HLA-C1C2 was significantly reduced in this cohort compared to non-transplanted patients. CONCLUSION This study suggests a possible role of KIR and their ligands in the development of liver damage. The absence of C1 and C2 ligands heterozygosity could lead to less inhibition of NK cells and a quicker progression to a high level of fibrosis in patients infected by HCV, especially following liver transplantation. Dans cette étude, nous nous sommes intéressés au polymorphisme de trois marqueurs génétiques (Allotypes GM, HLA Classe II et ADNmt) au sein de trois isolats berbères (Chenini-Douiret, Matmata et Sened) localisés dans le Sud tunisien. Notre objectif est d'évaluer l'impact génétique des invasions qu'a connues la Tunisie après son peuplement berbère en estimant la diversité génétique de ces trois isolats et en comparant ces populations berbères à d'autres populations nord-africaines. Les résultats obtenus pour les trois systèmes sont conformes entre eux et montrent que, malgré leur proximité géographique et la communauté linguistique, les trois populations berbères du Sud tunisien sont génétiquement très différenciées entre elles. Cette différenciation pourrait être due à l'effet de leur isolement et de leur endogamie, conduisant à leur divergence rapide les unes des autres. Les Berbères de Chenini-Douiret, très peu diversifiés et très divergents du point de vue génétique quel que soit le marqueur étudié, constituent un isolat. Lorsque l'on compare ces groupes berbères avec d'autres populations nord-africaines, les populations berbères du Sud tunisien apparaissent plus hétérogènes que celles du Maroc et d'Algérie. Outre l'effet de la dérive génétique, cette diversité peut refléter des contributions génétiques inégales d'origines européenne, sub-saharienne et/ou moyen-orientale ayant eu lieu au cours de la préhistoire et de l'histoire. Ainsi, la population tunisienne, à l'instar de la population nord-africaine dans son ensemble, est en réalité une mosaïque de populations.

L'axe principal de ce travail a consisté à analyser la diversité nucléotidique des gènes HLA de classes I et II dans une centaine de populations humaines réparties à l'échelle mondiale. Ces gènes sont les plus variables actuellement décrits chez l'Homme, et se sont révélés très informatifs pour étudier l'histoire du peuplement humain. Nos résultats moléculaires apportent une description beaucoup plus fine du polymorphisme de ce système génétique, notamment à une échelle régionale, que les approches envisagées jusqu'à aujourd'hui se limitant aux distributions de fréquences alléliques dans les populations. De plus, tenir compte des différences nucléotidiques entre allèles permet de mieux comprendre l'implication des mécanismes évolutifs (sélection et conversion génique) proposés comme les principaux moteurs de génération et de maintien du polymorphisme. Nous avons notamment mis en évidence de potentielles traces anciennes de sélection sur les allèles du gène HLA-DPB1, un locus supposé évoluer de manière neutre selon la littérature scientifique.

Dans cette étude, nous nous sommes intéressés au polymorphisme moléculaire du gène HLA-C au sein de 39 populations humaines localisées entre l'Europe et le sous-continent indien. Cette aire géographique correspond plus ou moins à la zone de locution des langues du phylum indo-européen, mais des populations de langues afro-asiatique et dravidienne sont aussi intégrées dans les comparaisons. Nos résultats nous indiquent que la géographie est l'un des facteurs déterminant les différenciations des populations situées en Europe. De plus, une frontière génétique est repérée le long d'un axe nord-est / sud-ouest à hauteur des Alpes et à travers les Pyrénées, qui met grosso modo en évidence une séparation entre les populations du pourtour méditerranéen et celle d'Europe de l'ouest. En revanche, on ne retrouve pas de corrélation entre géographie et génétique au Proche-Orient ni dans le sous-continent indien. Si la géographie ne semble pas jouer de rôle prépondérant au sein de ces deux groupes, les différenciations génétiques s'expliquent de manière très différente dans ces deux cas. En effet, les populations proche-orientales constituent un groupe génétiquement homogène, ce qui s'expliquerait par un taux élevé de flux génique dû aux migrations dont cette région, située à un carrefour continental, a été le théâtre à travers l'histoire. Au contraire, les populations du groupe indo-pakistanais sont très hétérogènes. La différenciation de ces dernières, dont plusieurs parlent une langue dravidienne, semble avoir été gouvernée par une dérive génétique rapide sous l'influence complexe de barrières culturelles, sociales, linguistiques et/ou religieuses.
