10. Silver Nanoparticles Stabilised by Cationic Gemini Surfactants with Variable Spacer Length

Abstract

The present study is focused on the synthesis and investigation of the physicochemical and biological properties of silver nanoparticles stabilized with a series of cationic gemini surfactants having a polymethylene spacer of variable length. UV-VIS spectroscopy, dynamic light scattering, scanning electron microscopy and zeta potential measurements were applied to provide physicochemical characterization of the silver nanoparticles. The mean size values of the nanoparticles were found to be in the 50 to 115 nm range. From the nanoparticle size distributions and scanning electron microscopy images it results that a population of small nanoparticles with the size of several nanometers was confirmed if the nanoparticles were stabilized with gemini molecules with either a short methylene spacer (two or four CH2 groups) or a long spacer (12 CH2 groups). The average zeta potential value for silver nanoparticles stabilized with gemini molecules is roughly independent of gemini surfactant spacer length and is approx. +58 mV. An interaction model between silver nanoparticles and gemini molecules which reflects the gained experimental data, is suggested. Microbicidal activity determinations revealed that the silver nanoparticles stabilized with gemini surfactants are more efficient against Gram-negative bacteria and yeasts, which has a direct relation to the interaction mechanism of nanoparticles with the bacterial cell membrane and its structural composition.

Abstract

In the present investigation, the basic esters of meta-alkoxyphenylcarbamic acid bearing variously substituted N-phenylpiperazine fragment were screened for their in vitro antimicrobial activity against Staphylococcus aureus, Escherichia coli and Candida albicans, respectively. The most effective against Escherichia coli was found the compound 6d (MIC=195,3 μg/mL) bearing simultaneously para-fluoro substituent at the 4‑phenylpiperazin-1-yl core and meta-methoxy side chain in the lipophilic part of the molecule. From whole analyzed set of the molecules the substance 8e with propoxy side chain forming meta-alkoxyphenylcarbamoyl fragment and lipophilic, sterically bulky meta-trifluoromethyl group attached at N-phenylpiperazine moiety was evaluated as the most active against Candida albicans (MIC=97,7 μg/mL). On the contrary, all investigated structures were practically inactive against Staphylococcus aureus (MIC>1000 μg/mL)

Abstract

Due to worldwide phenomenon of microbial resistance to commonly used therapeutic agents, antibiotics and antifungals, dibasic di-/trimethylphenylcarbamic acid esters 1-3, a non- -traditional series of potential antimicrobials, has been in vitro evaluated against chosen gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli) bacterial strains as well as against yeast (C. albicans) by the minimum inhibitory concentration (MIC) assay. Taking into consideration chemical structure of tested derivatives, the incorporation of more than one protonated atom of nitrogen into salt forming fragment positively influenced the activity against E. coli. On the contrary, the presence of one or more methyl groups instead of 3-alkoxy side chain attached to lipophilic aromatic moiety has not found to be favorable structural feature. In entire set of inspected compounds, the most promising results have been found for the compound 3, chemically 1-[3-piperidinium-1-yl-2-({[(2,4,6-trimethylphenyl) amino]carbonyl}oxy)propyl]piperidinium dichloride, against E. coli with the MIC=1.56mg/mL. 20. BASIC ESTERS OF meta-/para-ALKOXYPHENYLCARBAMIC ACID CONTAINING 4-(2-METHYL-/2-FLUOROPHENYL)PIPERAZIN-1-YL MOIETY AND THEIR ANTIMICROBIAL ACTIVITY.

Abstract

The original esters of meta-/para-alkoxyphenylcarbamic acid containing 4-(2-methyl-/2-fluorophenyl)piperazin-l-yl, labelled as 5d-7d, were screened for their in vitro antimicrobial activity against Staphylococcus aureus, Escherichia coli and Candida albicans, respectively. Following the minimum inhibitory concentration (MIC) assay, electronic and hydrophobic interactions, induced by suitable substitution at the N-phenylpiperazine, ring have appeared to be more noticabe factors which positively influenced the efficiency of these compounds against S. aureus as well as against E. coli than relatively high lipophilicity of 5d-7d. In general, the compounds comprising methyl sub-stituent attached to basic fragment (7a-7d) were observed to be more active against both given bacterial strains in the comparison to those with fluoro group (5d and 5e). From the entire analyzed set, the most active molecule 7b showed MIC=0.10 mg-mL-1 against given Gram-positive and Gram-negative bacteria as well. On the contrary, the increase in evaluated molecules lipophilicity accompanied by the presence of the substituent at N-phenylpiperazine with primarily strong electron-donating or a slightly electron-withdrawing effect has not positively reflected in their efficiency against C. albicans (MICs>1.00 mg-mL-1 ). Abstract

Basic esters of ortho-alkoxyphenylcarbamic acid containing 4-(2-fluoro-/4-fluoro-/3-trifluoromethylphenyl)-piperazin-l-yl, 5a-8b, were screened for their in vitro antimicrobial activity against Escherichia coli, Candida albicans and Staphylococcus aureus, respectively. Following the minimum inhibitory concentration (MIC) assay, electronic and hydrophobic interactions, induced by suitable substitution at the N-phenylpiperazine ring, have appeared to be more notable factors which positively influenced the efficiency of such compounds against E. coli than the rising of their lipophilicity. Molecules comprising only of single atom of fluorine, 5a-6b, were observed to be more active (MIC=0.39-0.78 mg·mL-1 in the comparison to those with trifluromethyl group (MICs>1.00 mg·mL-1). On the contrary, the lipophilicity increasing of evaluated mole- cules accompanied by the presence of the substituent at N-phenylpiperazine with electron-withdrawing effect, 8a-8b, has positively, reflected in their efficiency against C. albicans with the observed maximum of MIC=0.78 mg·mL-1 However, all tested molecules were against S. aureus (MICs> 1.00 mg·mL-1 practically inactive.
