3. How do landscape and life history traits contribute to the threat context of Brazilian primates?

Abstract

Brazilian primates differ regarding landscape characteristics within their ranges (e.g. habitat availability) and life‐history traits (e.g. body size). These landscape and life history attributes may be related to extinction risk. Here, we verified how such attributes correlate with primate threat categories. We considered 124 Brazilian primates based on the 2014 Brazilian list of threatened (Critically Endangered, Endangered and Vulnerable) and non‐threatened (Least Concern and Near Threatened) species. We then characterized their landscape (i.e. habitat availability, habitat loss and fragmentation, indigenous lands, roads, urban areas, deforestation arch) and life‐history attributes (i.e. body weight, gestation length and generation time), which together make up the threat context for each threat category. We compared threat categories to identify differences in such attributes, considering biome (Atlantic Forest, Amazon, Caatinga and Cerrado) as a factor and testing for phylogenetic effect. We investigated the attributes responsible for group characterization for specific threats and binary threatened/non‐threatened categories. We show that life history and landscape attributes differ in the biomes. However, only landscape features varied across biomes. In the Amazon, the threatened categories reached the highest level of habitat loss in the last 30 years, while in the Atlantic Forest, the landscape of threatened species had the highest proportions of roads and urban areas. Most threatened species landscapes in the Caatinga and Cerrado were highly fragmented. We found a positive link between human impacts and extinction risk in the Amazon. In the other biomes, anthropogenic landscape characteristics were associated with threatened and non‐threatened species. Threatened species tended to have large bodies and a slow life history, regardless of the biome. The more closely related the species, the more similar the traits. We suggest considering biomes and threat categories together with specific landscape and life history attributes to distinguish primate threat context for species conservation priority‐setting. 6. Gradual Loss of Myelin and Formation of an Astrocytic Scar during Wallerian Degeneration in the Human Spinal Cord

Abstract

Axons undergo Wallerian degeneration distal to a point of injury. Experimental investigations have documented many of the cellular and molecular events that underlie this behaviour. Since relatively little is known about such events in human CNS pathologies and current experimental intervention strategies indicate the possibility of significant axon regeneration along the original degenerated fibre tract, we performed an immunohistochemical investigation of the dynamics of Wallerian degeneration in post mortem spinal cords of patients who died 2 days to 30 years after either cerebral infarction or traumatic spinal cord injury. Neurofilament (NF) staining demonstrated a spatio-temporal pattern of axonal loss within degenerating descending nerve fibre tracts that could be detected close to the lesion as early as 12 days after injury and progressed to an almost complete loss of NF immunoreactivity at survival times of 1 year and longer. Immunohistochemistry for glial fibrillary acidic protein revealed a late astrocytic reaction starting at 4 months after injury in the degenerating tracts, leading to the long-term deposition of a dense astrocytic scar. These events were accompanied by the gradual reduction of myelin basic protein in affected nerve fibre tracts, leading to almost complete loss by 3 years after injury. Since the extracellular matrix molecule chondroitin sulphate proteoglycan (CSPG) is known to be strongly inhibitory for axonal regeneration and to be a major component of gliotic scar tissues, we investigated the possible deposition of CSPG within the degenerating nerve fibre tracts. Apart from a local up-regulation close to the lesion site, our results show no enhanced CSPG expression within degenerated tracts at any survival time. This suggests that despite the apparent lack of CSPG in Wallerian degeneration, the slow reduction of CNS myelin and the long-term deposition of a dense astrocytic scar may present an environment that is non-supportive for axon regrowth

Abstract

Objectives Racial factors play a significant role in SSc. We evaluated differences in SSc presentations between white patients (WP), Asian patients (AP) and black patients (BP) and analysed the effects of geographical locations. Methods SSc characteristics of patients from the EUSTAR cohort were cross-sectionally compared across racial groups using survival and multiple logistic regression analyses. Results The study included 9162 WP, 341 AP and 181 BP. AP developed the first non-RP feature faster than WP but slower than BP. AP were less frequently anti-centromere (ACA; odds ratio (OR) = 0.4, P < 0.001) and more frequently anti-topoisomerase-I autoantibodies (ATA) positive (OR = 1.2, P = 0.068), while BP were less likely to be ACA and ATA positive than were WP [OR(ACA) = 0.3, P < 0.001; OR(ATA) = 0.5, P = 0.020]. AP had less often (OR = 0.7, P = 0.06) and BP more often (OR = 2.7, P < 0.001) diffuse skin involvement than had WP. AP and BP were more likely to have pulmonary hypertension [OR(AP) = 2.6, P < 0.001; OR(BP) = 2.7, P = 0.03 vs WP] and a reduced forced vital capacity [OR(AP) = 2.5, P < 0.001; OR(BP) = 2.4, P < 0.004] than were WP. AP more often had an impaired diffusing capacity of the lung than had BP and WP [OR(AP vs BP) = 1.9, P = 0.038; OR(AP vs WP) = 2.4, P < 0.001]. After RP onset, AP and BP had a higher hazard to die than had WP [hazard ratio (HR) (AP) = 1.6, P = 0.011; HR(BP) = 2.1, P < 0.001]. Conclusion Compared with WP, and mostly independent of geographical location, AP have a faster and earlier disease onset with high prevalences of ATA, pulmonary hypertension and forced vital capacity impairment and higher mortality. BP had the fastest disease onset, a high prevalence of diffuse skin involvement and nominally the highest mortality.

8. Genetic analysis of resistance to soybean mosaic virus in OX670 and harosoy soybean

Abstract

Soybean mosaic virus (SMV) resistance in the soybean [Glycine max (L.) Merr.] breeding line OX670 previously was postulated to be controlled by a gene derived from the cultivar Raiden and designated as Rsv2. Subsequently, it was shown that Raiden has a single resistance gene at the Rsv1 locus, suggesting that the resistance gene in OX670 was not from Raiden. Most of the remaining ancestry of OX670 is derived from `Harosoy'. The objectives of this study were to determine (i) the reaction of Harosoy to SMV-G1 through G7 strains; (ii) the inheritance of SMV resistance in Harosoy and OX670; and (iii) the allelomorphic relationship of resistance genes in these cultivars with previously described resistance genes. OX670 and Harosoy were crossed with the SMV susceptible cultivar Lee 68 to study the inheritance of resistance. OX670 and Harosoy were also crossed with the resistant lines L78-379, L88-8431, P196983, L29, and V945152 to elucidate the allelomorphic relationships between the genes in OX670, Harosoy, and previously reported genes. Our results indicated that Harosoy, which is resistant to SMV-G5 through G7 and susceptible to SMV-G1 through G4, possesses a single partially dominant SMV resistance gene at the Rsv3 locus. Inheritance studies indicated that OX670, which is resistant to SMV-G1 through G7, possesses two independent dominant genes for SMV resistance. Abstract

ObjectiveTo assess the safety and efficacy of rituximab in systemic sclerosis (SSc) in clinical practice.MethodsWe performed a prospective study including patients with SSc from the European Scleroderma Trials and Research (EUSTAR) network treated with rituximab and matched with untreated patients with SSc. The main outcomes measures were adverse events, skin fibrosis improvement, lung fibrosis worsening and steroids use among propensity score-matched patients treated or not with rituximab.Results254 patients were treated with rituximab, in 58% for lung and in 32% for skin involvement. After a median follow-up of 2 years, about 70% of the patients had no side effect. Comparison of treated patients with 9575 propensity-score matched patients showed that patients treated with rituximab were more likely to have skin fibrosis improvement (22.7 vs 14.03 events per 100 person-years; OR: 2.79 [1.47–5.32]; p=0.002). Treated patients did not have significantly different rates of decrease in forced vital capacity (FVC)>10% (OR: 1.03 [0.55–1.94]; p=0.93) nor in carbon monoxide diffusing capacity (DLCO) decrease. Patients having received rituximab were more prone to stop or decrease steroids (OR: 2.34 [1.56–3.53], pConclusionRituximab use was associated with a good safety profile in this large SSc-cohort. Significant change was observed on skin fibrosis, but not on lung. However, the limitation is the observational design. The potential stabilisation of lung fibrosis by rituximab has to be addressed by a randomised trial.

Abstract

Objectives Mortality and worsening of organ function are desirable endpoints for clinical trials in systemic sclerosis (SSc). The aim of this study was to identify factors that allow enrichment of patients with these endpoints, in a population of patients from the European Scleroderma Trials and Research group database. Methods Inclusion criteria were diagnosis of diffuse SSc and follow-up over 12 +/- 3 months. Disease worsening/organ progression was fulfilled if any of the following events occurred: new renal crisis; decrease of lung or heart function; new echocardiography-suspected pulmonary hypertension or death. In total, 42 clinical parameters were chosen as predictors for the analysis by using (1) imputation of missing data on the basis of multivariate imputation and (2) least absolute shrinkage and selection operator regression. Results Of 1451 patients meeting the inclusion criteria, 706 had complete data on outcome parameters and were included in the analysis. Of the 42 outcome predictors, eight remained in the final regression model. There was substantial evidence for a strong association between disease progression and age, active digital ulcer (DU), lung fibrosis, muscle weakness and elevated C-reactive protein (CRP) level. Active DU, CRP elevation, lung fibrosis and muscle weakness were also associated with a significantly shorter time to disease progression. A bootstrap validation step with 10 000 repetitions successfully validated the model. Conclusions The use of the predictive factors presented here could enable cohort enrichment with patients at risk for overall disease worsening in SSc clinical trials.

O sistema de irrigação, as práticas de manejo da água e manejos diferenciados de preparo do solo para o cultivo do arroz são fatores determinantes à produção deste cereal, influenciando de forma diferenciada o efluxo de gases de efeito estufa (GEE), sendo que alguns desses apresentam potencial mitigador das emissões de GEE. Três estudos foram conduzidos visando avaliar: (I) as condições de oxirredução do solo e a influência de manejos de água sobre as emissões de metano (CH4) e óxido nitroso (N2O) (GEE) em solo cultivado com arroz (Estudo I); (II) avaliar a influência de dois sistemas de irrigação (Inundação e Aspersão) sobre as emissões de CH4 e N2O em solo cultivado com arroz, sendo essas emissões comparadas com uma área natural (Estudo II) e, (III) avaliar a influência da época e das operações de preparo do solo e da cobertura vegetal sobre as emissões anuais de CH4 e N2O do solo, bem como a contribuição desses gases para o potencial de aquecimento global parcial (PAGp) em sistema de produção de grãos em terras baixas e, com isso, definir práticas de manejo e sistemas de cultivos com potencial mitigador de emissões de GEE em terras baixas (Estudo III). Os estudos foram conduzidos de novembro de 2011 até abril de 2015, em um Planossolo no Sul do Brasil. Na maior parte do período as avaliações de CH4 e N2O foram realizadas semanalmente, utilizando-se o método da câmara fechada estática. Com base nas emissões de CH4 e de N2O e dos seus respectivos potenciais de aquecimento global (PAG; 25 e 298), foi calculado o PAGp (PAGp; CO2 equivalente). Foram avaliados também o volume de água aplicado e a produtividade de grãos (PG), possibilitando a obtenção dos índices de eficiência do uso de água (EUA) e PAGp/PG. No Estudo I foram avaliados os tratamentos nas safras 2011/12 e 2012/13: Inundação Contínua (IC); Inundação Contínua por Período Reduzido (PR); Inundação Intermitente (II); Solo Saturado (SS). Nas duas safras avaliadas, nos manejos da irrigação II e SS, a redução das emissões de CH4 foi superior ao efeito no aumento nas emissões de N2O, ou seja, apresentaram o menor PAGp, mostrando-se mais eficientes na mitigação de GEE em relação aos outros dois tratamentos, IC e PR. No Estudo II - foram avaliados por dois anos completos (duas Safras e duas Entressafras) os tratamentos: Sistema de Irrigação por Inundação; Sistema de Irrigação por Aspersão; Área Natural: área em condição natural, ou seja, sem histórico de cultivo. Apesar do sistema de irrigação por aspersão potencializar as emissões de N2O do solo, ainda assim, é mais eficiente na redução do PAGp em relação ao sistema irrigado por alagamento do solo em cultivos de arroz. No Estudo III - foram avaliados por dois anos completos (duas Safras e duas Entressafras) quatro sistemas de cultivo: T1 ? Arroz/soja/arroz; T2 ?Arroz/Arroz/Arroz; T3 ? Arroz/Arroz/Soja; T4 ? Arroz/Soja/Arroz c/ preparo. O N2O é o principal componente do potencial de aquecimento global da soja, enquanto que o CH4 predomina no cultivo de arroz irrigado. Consequently, organizations either host an oversized IT infrastructure or they are incapable of realizing the benefits of new applications. A solution to the challenge is provided by the large-scale computing infrastructures of Clouds and Grids which allow resources to be shared. A major challenge is the development of mechanisms that allow efficient sharing of IT resources. Market mechanisms are promising, but there is a lack of research in scalable market mechanisms. We extend the Multi-Attribute Combinatorial Exchange mechanism with greedy heuristics to address the scalability challenge. The evaluation shows a trade-off between efficiency and scalability. There is no statistical evidence for an influence on the incentive properties of the market mechanism. Abstract

La sarcoïdose est une affection inflammatoire granuiomateuse systémique d'origine inconnue touchant le plus fréquemment les poumons, le système lymphoïde, le foie, les yeux et la peau. Dans cet article, nous rapportons deux cas de sarcoïdose cutanée touchant les avant-bras de deux patients anciens toxicomanes traités par interféron-a et ribavirine pour une hépatite C chronique. Nous procédons à une revue de la littérature de la sarcoïdose induite par l'interféron et élaborons une nouvelle hypothèse pathogénique de l'effet Koebner dans la sarcoïdose cutanée. Dans le cas des deux patients que nous décrivons, la distribution des lésions cutanées coïncide avec les anciens sites d'injection d'héroïne le long des trajets veineux des deux avant- bras. Cette distribution unique de l'atteinte cutanée suggère que les dommages tissulaires induits par la répétition d'injections percutanées puissent représenter un terrain favorisant au développement local d'une sarcoïdose cutanée. Fait intéressant, il a été récemment démontré que les cellules dendritiques plasmacytoïdes - sous-type de cellules dendritiques généralement absent de la peau - infiltrent rapidement les sites de peau lésée. Ces cellules sont la source d'une production endogène d'interféron-a, cytokine connue pour promouvoir le processus de cicatrisation, mais également pour favoriser le développement de sarcoïdose chez des individus prédisposés. Ainsi, nous postulons que les lésions de sarcoïdose cutanée limitées le long des trajets veineux - sites préalables d'injection percutanée de drogues - peuvent résulter d'une expression locale supplémentaire d'interféron-a. Celle-ci serait en outre favorisée par le traitement de ribavirine dans le cadre de l'hépatite C, connu pour renforcer la production endogène d'interféron-a. L'identification de nombreuses cellules dendritiques plasmacytoïdes circonscrivant l'inflammation granuiomateuse sur la biopsie cutanée de l'un de nos patients semble être un argument dans ce sens, conforté par l'absence de corps étranger détecté en microscopie par lumière polarisée. Cette observation semble pouvoir représenter un point crucial dans la compréhension des mécanismes physiopathologiques à la base de l'infiltration des cicatrices cutanées par la sarcoïdose. Des investigations supplémentaires doivent encore être effectuées afin de confirmer cette hypothèse.

Abstract

Common variable immune deficiency is the most frequent primary immune deficiency, characterized mainly by a disorder of B lymphocytes differentiation and a deficit in immunoglobulins. The clinical manifestations include recurrent infections, non-infectious lung and digestive involvements, autoimmune diseases, and an increased susceptibility to cancers. Recent breakthroughs have been made in the understanding of some genetic mechanisms of the disease. Replacement therapy with intravenous immunoglobulins remains the treatment of choice, which allows significant improvement in the survival and quality of life. However progress should be made in the understanding of the pathophysiology and in the early detection of this disease, since a delay in the diagnosis may have harmful consequences in terms of morbidity and mortality.
