1. Development of a longevity prediction model for cut roses using hyperspectral imaging and a convolutional neural network

Abstract

IntroductionHyperspectral imaging (HSI) and deep learning techniques have been widely applied to predict postharvest quality and shelf life in multiple horticultural crops such as vegetables, mushrooms, and fruits; however, few studies show the application of these techniques to evaluate the quality issues of cut flowers. Therefore, in this study, we developed a non-contact and rapid detection technique for the emergence of gray mold disease (GMD) and the potential longevity of cut roses using deep learning techniques based on HSI data.MethodsCut flowers of two rose cultivars (‘All For Love’ and ‘White Beauty’) underwent either dry transport (thus impaired cut flower hydration), ethylene exposure, or Botrytis cinerea inoculation, in order to identify the characteristic light wavelengths that are closely correlated with plant physiological states based on HSI. The flower bud of cut roses was selected for HSI measurement and the development of a vase life prediction model utilizing YOLOv5.Results and discussionThe HSI results revealed that spectral reflectance between 470 to 680 nm was strongly correlated with gray mold disease (GMD), whereas those between 700 to 900 nm were strongly correlated with flower wilting or vase life. To develop a YOLOv5 prediction model that can be used to anticipate flower longevity, the vase life of cut roses was classed into two categories as over 5 d (+5D) and under 5 d (-5D), based on scoring a grading standard on the flower quality. A total of 3000 images from HSI were forwarded to the YOLOv5 model for training and prediction of GMD and vase life of cut flowers. Validation of the prediction model using independent data confirmed its high predictive accuracy in evaluating the vase life of both ‘All For Love’ (r2 = 0.86) and ‘White Beauty’ (r2 = 0.83) cut flowers. The YOLOv5 model also accurately detected and classified GMD in the cut rose flowers based on the image data. Our results demonstrate that the combination of HSI and deep learning is a reliable method for detecting early GMD infection and evaluating the longevity of cut roses.

Abstract

Botrytis cinerea (B. cinerea) causes gray mold disease (GMD), which results in physiological disorders in plants that decrease the longevity and economic value of horticultural crops. To prevent the spread of GMD during distribution, a rapid, early detection technique is necessary. Thermal imaging has been used for GMD detection in various plants, including potted roses; however, its application to cut roses, which have a high global demand, has not been established. In this study, we investigated the utility of thermal imaging for the early detection of B. cinerea infection in cut roses by monitoring changes in petal temperature after fungal inoculation. We examined the effects of GMD on the postharvest quality and petal temperature of cut roses treated with different concentrations of fungal conidial suspensions and chemicals. B. cinerea infection decreased the flower opening, disrupted the water balance, and decreased the vase life of cut roses. Additionally, the average temperature of rose petals was higher for infected flowers than for non-inoculated flowers. One day before the appearance of necrotic symptoms (day 1 of the vase period), the petal temperature in infected flowers was significantly higher, by 1.1 °C, than that of non-inoculated flowers. The GMD-induced increase in petal temperature was associated with the mRNA levels of genes related to ethylene, reactive oxygen species, and water transport. Furthermore, the increase in temperature caused by GMD was strongly correlated with symptom severity and fungal biomass. A multiple regression analysis revealed that the disease incidence in the petals was positively related to the petal temperature one day before the appearance of necrotic symptoms. These results show that thermography is an effective technique for evaluating changes in petal temperature and a possible method for early GMD detection in the cut flower industry.

3. Combined Nano Silver, α-Aminoisobutyric Acid, and 1-Methylcyclopropene Treatment Delays the Senescence of Cut Roses with Different Ethylene Sensitivities

Abstract

Flower senescence varies among cut roses (Rosa hybrida L.), and it is known that the postharvest life of ethylene-sensitive flowers is strongly related to the transcriptional accumulation of ethylene biosynthesis genes, whereas that of ethylene-insensitive flowers is shortened by water stress. These different responses of flowers to hormone and water stresses limit the action of preservatives in inhibiting the postharvest deterioration of cut rose flowers. This study was conducted to investigate the effects of the combined application of antibacterial agents and ethylene biosynthesis and binding inhibitors on the postharvest life and quality of the cut rose cultivars ‘Matador’ (ethylene-sensitive) and ‘Dolcetto’ (ethylene-insensitive). Cut flowers were treated with nano silver (NS), a combination of NS and α-aminoisobutyric acid (NS+A), or a combination of NS+A and 1-methylcyclopropene (NS+AM), and they were subsequently exposed to ethylene for 24 h. Treatment effectiveness was compared with that of control (CON) flowers, which were kept in distilled water and exposed only to ethylene. The results showed that all treatments significantly improved the postharvest quality and vase life of both rose cultivars. However, NS+AM most markedly delayed senescence and prolonged the vase life of cut flowers by 217% in ‘Matador’ and 168.4% in ‘Dolcetto’, compared with those of CON flowers. NS+AM also effectively increased the flower diameter and maintenance time of positive water balance and initial fresh weight by enhancing solution absorption as well as suppressing bacterial growth at the cut stem ends of the cut roses. Furthermore, NS+AM greatly suppressed the ethylene-inducible increase of ethylene biosynthesis genes and the reduction of ethylene receptor genes in petals, which resulted in a reduced flower response to exogenous ethylene in both rose cultivars. These findings show that NS+AM effectively delays flower senescence in both ethylene sensitive and insensitive cultivars by synergistically preventing ethylene response and water stress in cut flowers.

4. Regulation of Botrytis cinerea Infection and Gene Expression in Cut Roses by Using Nano Silver and Salicylic Acid

Abstract

Botrytis cinerea (B. cinerea) is one of the necrotrophic pathogens resulting in the heaviest commercial losses in cut rose flowers, and the severity of gray mold disease partly depends on the presence of ethylene during the storage and transport. The effectiveness of nano silver (NS) and salicylic acid (SA) was assessed as a novel control agent in protecting the cut rose flowers against B. cinerea infection and ethylene damages. The efficacy of NS and SA was compared with an inoculated control (CON). A non-treated control (NT) was also used to evaluate the natural infection process. The results indicated that pretreatment with 20 mg L−1 NS significantly reduced B. cinerea growth in rose petals during vase periods. NS effectively suppressed the mRNA levels of ethylene biosynthesis genes (RhACS2, RhACS4, and RhACO1) and the reduction in expression levels of ethylene receptor genes (RhETR1, RhETR2, and RhETR5) and the downstream regulator RhCTR2 in rose petals after B. cinerea inoculation. NS application also decreased the expression of the B. cinerea snod-prot-like 1 (Bcspl1) gene which acts as the virulence factor in cut roses. In NS flowers, the maximum quantum yield of PSII (Fv/Fm) value was higher and the leaf temperature was lower on day 1, suggesting that these factors can be used for detecting B. cinerea infection and water stress in cut rose flowers. Furthermore, NS improved water relations and extended the vase life of cut rose flowers by 3.3 d, compared with that of NT flowers. In contrast, SA had no inhibitive effects on both B. cinerea growth and ethylene response in cut roses. The findings from the present study highlight NS as a promising new candidate for preventing B. cinerea infection and ethylene damages and for improving the postharvest quality of cut roses exported overseas.

6. The generalized Harer conjecture for the homology triviality

Abstract

The classical Harer conjecture is about the stable homology triviality of the obvious embedding $\phi : B_{2g+2} \hookrightarrow \Gamma_{g}$, which was proved by Song and Tillmann. The main part of the proof is to show that $\B\phi^{+} : \B B_{\infty}^{+} \rightarrow \B \Gamma_{\infty}^{+}$ induced from $\phi$ is a double loop space map. In this paper, we give a proof of the generalized Harer conjecture which is about the homology triviality for an $arbitrary$ embedding $\phi : B_{n} \hookrightarrow \Gamma_{g,k}$. We first show that it suffices to prove it for a $regular$ embedding in which all atomic surfaces are regarded as identical and each atomic twist is a {\it simple twist} interchanging two identical sub-parts of atomic surfaces. The main strategy of the proof is to show that the map $\Phi : \mathcal{C} \rightarrow \mathcal{S}$ induced by $\B\phi:\conf_n(D)\rightarrow\mathcal{M}_{g,k}$ preserves the actions of the framed little 2-disks operad.

7. Noise-resistant reconstruction algorithm based on the sinogram pattern

Abstract

We introduce a new CT image reconstruction algorithm that is less affected by various artifacts. The new reconstruction algorithm is a method of minimizing the difference between synchrotron X-ray tomography data and sinograms generated using Radon transform of CT images. The CT image is iteratively updated to reduce the difference from the sinogram of the data. This method can obtain clean CT images from the projection data, which can create ring artifacts or metal artifacts. Also, even if the sample size is larger than the CCD and/or the projection image does not satisfy the Beer-Lambert law, a clean CT image can be reconstructed. Our new reconstruction algorithm can also be applied to fan beam CT or cone beam CT, Comment: 8 pages, 3 figures

Abstract

Recent studies on quantum computing algorithms focus on excavating features of quantum computers which have potential for contributing to computational model enhancements. Among various approaches, quantum annealing methods effectively parallelize quadratic unconstrained binary optimization (QUBO) formulations of systems of linear equations. In this paper, we simplify these formulations by exploiting congruence of real symmetric matrices to diagonal matrices. We further exhibit computational merits of the proposed QUBO models, which can outperform classical algorithms such as QR and SVD decomposition.

Abstract

The $d$-fold ($d \geq 3$) branched coverings on a disk give an infinite family of nongeometric embeddings of braid groups into mapping class groups. We, in this paper, give new explicit expressions of these braid group representations into automorphism groups of free groups in terms of the actions on the generators of free groups. We also give a systematic way of constructing and expressing these braid group representations in terms of a new gadget, called covering groupoid. We prove that each generator $\widetilde{\beta}_i$ of braid group inside mapping class group induced by $d$-fold covering is the product of $d-1$ Dehn twists on the surface., Comment: 13 pages, 11 figures

16. Configuration space, moduli space and 3-fold covering space

Abstract

A function from configuration space to moduli space of surface may induce a homomorphism between their fundamental groups which are braid groups and mapping class groups of surface, respectively. This map $\phi: B_k \rightarrow \Gamma_{g,b}$ is induced by 3-fold branched covering over a disk with some branch points. In this thesis we give a concrete description of this map and show that it is injective by Birman-Hilden theory. This gives us a new interesting non-geometric embedding of braid group into mapping class group. On the other hand, we show that the map on the level of classifying spaces of groups is compatible with the action of little 2-cube operad so that it induces a trivial homomorphim between stable homology group of braid groups and that of mapping class groups(Harer conjecture). We also show how the lift $\tilde{\beta_i}$ acts on the fundamental group of the surface and through this we prove that $\tilde{\beta_i}$ equals the product of two inverse Dehn twists., Comment: 17 pages, 19 figures

Abstract

The classical Harer conjecture states the stable homology triviality of the canonical embedding ϕ:B2g+2↪Γg$\phi: B_{2g+2} \hookrightarrow \Gamma _{g}$, which was proved by Song and Tillmann. The main part of the proof is to show that Bϕ+:BB∞+→BΓ∞+$\operatorname{B}\phi ^{+}: \operatorname{B}B_{\infty }^{+} \rightarrow \operatorname{B}\Gamma _{\infty }^{+}$, induced from ϕ$\phi$ is a double‐loop space map. In this paper, we give a proof of the generalized Harer conjecture concerning the homology triviality for every regular embedding ϕ:Bn↪Γg,k$\phi: B_{n} \hookrightarrow \Gamma _{g,k}$. The main strategy of the proof is to remove all the interchangeable subsurfaces from Sg,k$S_{g,k}$ and collapse the new boundary components. Then, we obtain (the union of) covering spaces over a disk with n$n$ marked points that we can analyze. The final goal is to show that the map Φ:C→S$\Phi: \mathcal {C} \rightarrow \mathcal {S}$ induced by Bϕ:Confn(D)→Mg,k$\operatorname{B}\phi:\operatorname{Conf}_n(D)\rightarrow \mathcal {M}_{g,k}$ preserves the actions of the framed little 2‐disks operad.
