Leishmaniasis is a parasitic disease caused by various species of Leishmania, which affects millions of people worldwide. Current treatments for leishmaniasis often present adverse effects and may not be effective against every Leishmania strain or form of the disease. Therefore, it is essential to explore new therapeutic approaches to combat the parasite. In this study, we focused on the PeBoW complex proteins homologous in Leishmania, which is involved in ribosomal biogenesis and plays a critical role in cancer development in mammalian cells. Recently, a homologue of the oncogene PES1 was found in Leishmania major, which plays a crucial role in parasite infectivity. Given this, we analyzed the possibility of using other PeBoW complex partner genes in Leishmania as therapeutic targets for leishmaniasis treatment. Specifically, our investigation aimed to characterize the partner WDR12 homologous in Leishmania infantum (LmjWDR12) as new therapeutic approach.

Previous studies have shown that local delivery of tumor antigen-specific CD8(+) T lymphocytes engineered to transiently express single-chain IL-12 mRNA is highly efficacious. Peritoneal dissemination of cancer is a frequent and often fatal patient condition usually diagnosed when the tumor burden is too large and hence uncontrollable with current treatment options. In this study, we have modeled intracavitary adoptive T cell therapy with OVA-specific OT-I T cells electroporated with IL-12 mRNA to treat B16-OVA and PANC02-OVA tumor spread in the peritoneal cavity. Tumor localization in the omentum and the effects of local T-cell encounter with the tumor antigens were monitored, the gene expression profile evaluated, and the phenotypic reprogramming of several immune subsets was characterized. Intraperitoneal administration of T cells promoted homing to the omentum more effectively than intravenous administration. Transient IL-12 expression was responsible for a favorable reprogramming of the tumor immune microenvironment, longer persistence of transferred T lymphocytes in vivo, and the development of immunity to endogenous antigens following primary tumor eradication. The efficacy of the strategy was at least in part recapitulated with the adoptive transfer of lower affinity transgenic TCR-bearing PMEL-1 T lymphocytes to treat the aggressive intraperitoneally disseminated B16-F10 tumor. Locoregional adoptive transfer of transiently IL-12-armored T cells appears to offer promising therapeutic advantages in terms of anti-tumor efficacy to treat peritoneal carcinomatosis.

Background One of the main difficulties of adoptive cell therapies with chimeric antigen receptor (CAR)-T cells in solid tumors is the identification of specific target antigens. The tumor microenvironment can present suitable antigens for CAR design, even though they are not expressed by the tumor cells. We have generated a CAR specific for the splice variant extra domain A (EDA) of fibronectin, which is highly expressed in the tumor stroma of many types of tumors but not in healthy tissues. Methods EDA expression was explored in RNA-seq data from different human tumor types and by immunohistochemistry in paraffin-embedded tumor biopsies. Murine and human anti-EDA CAR-T cells were prepared using recombinant retro/lentiviruses, respectively. The functionality of EDA CAR-T cells was measured in vitro in response to antigen stimulation. The antitumor activity of EDA CAR-T cells was measured in vivo in C57BL/6 mice challenged with PM299L-EDA hepatocarcinoma cell line, in 129Sv mice-bearing F9 teratocarcinoma and in NSG mice injected with the human hepatocarcinoma cell line PLC. Results EDA CAR-T cells recognized and killed EDA-expressing tumor cell lines in vitro and rejected EDA-expressing tumors in immunocompetent mice. Notably, EDA CAR-T cells showed an antitumor effect in mice injected with EDA-negative tumor cells lines when the tumor stroma or the basement membrane of tumor endothelial cells express EDA. Thus, EDA CAR-T administration delayed tumor growth in immunocompetent 129Sv mice challenged with teratocarcinoma cell line F9. EDA CAR-T treatment exerted an antiangiogenic effect and significantly reduced gene signatures associated with epithelial-mesenchymal transition, collagen synthesis, extracellular matrix organization as well as IL-6-STAT5 and KRAS pathways. Importantly, the human version of EDA CAR, that includes the human 41BB and CD3 zeta endodomains, exerted strong antitumor activity in NSG mice challenged with the human hepatocarcinoma cell line PLC, which expresses EDA in the tumor stroma and the endothelial vasculature. EDA CAR-T cells exhibited a tropism for EDA-expressing tumor tissue and no toxicity was observed in tumor bearing or in healthy mice. Conclusions These results suggest that targeting the tumor-specific fibronectin splice variant EDA with CAR-T cells is feasible and offers a therapeutic option that is applicable to different types of cancer.

Motivation: Trait variation within species can reveal plastic and/or genetic responses to environmental gradients, and may indicate where local adaptation has occurred. Here, we present a dataset of rangewide variation in leaf traits from seven of the most ecologically and economically important tree species in Europe. Sample collection and trait assessment are embedded in the GenTree project (EU‐Horizon 2020), which aims at characterizing the genetic and phenotypic variability of forest tree species to optimize the management and sustainable use of forest genetic resources. Our dataset captures substantial intra‐ and interspecific leaf phenotypic variability, and provides valuable information for studying the relationship between ecosystem functioning and trait variability of individuals, and the response and resilience of species to environmental changes. Main types of variable contained: We chose morphological and chemical characters linked to trade‐offs between acquisition and conservation of resources and water use, namely specific leaf area, leaf size, carbon and nitrogen content and their ratio, and the isotopic signature of stable isotope ¹³C and ¹⁵N in leaves. Spatial location and grain: We surveyed between 18 and 22 populations per species, 141 in total, across Europe. Time period: Leaf sampling took place between 2016 and 2017. Major taxa and level of measurement: We sampled at least 25 individuals in each population, 3,569 trees in total, and measured traits in 35,755 leaves from seven European tree species, i.e. the conifers Picea abies, Pinus pinaster and Pinus sylvestris, and the broadleaves Betula pendula, Fagus sylvatica, Populus nigra and Quercus petraea. Background: Progress in the field of evolutionary forest ecology has been hampered by the huge challenge of phenotyping trees across their ranges in their natural environments, and the limitation in high-resolution environmental information. Findings: The GenTree Platform contains phenotypic and environmental data from 4,959 trees from 12 ecologically and economically important European forest tree species: Abies alba Mill. (silver fir), Betula pendula Roth. (silver birch), Fagus sylvatica L. (European beech), Picea abies (L.) H. Karst (Norway spruce), Pinus cembra L. (Swiss stone pine), Pinus halepensis Mill. (Aleppo pine), Pinus nigra Arnold (European black pine), Pinus pinaster Aiton (maritime pine), Pinus sylvestris L. (Scots pine), Populus nigra L. (European black poplar), Taxus baccata L. (English yew), and Quercus petraea (Matt.) Liebl. (sessile oak). Phenotypic (height, diameter at breast height, crown size, bark thickness, biomass, straightness, forking, branch angle, fructification), regeneration, environmental in situ measurements (soil depth, vegetation cover, competition indices), and environmental modeling data extracted by using bilinear interpolation accounting for surrounding conditions of each tree (precipitation, temperature, insolation, drought indices) were obtained from trees in 194 sites covering the species’ geographic ranges and reflecting local environmental gradients. Conclusions: The GenTree Platform is a new resource for investigating ecological and evolutionary processes in forest trees. The coherent phenotyping and environmental characterization across 12 species in their European ranges allow for a wide range of analyses from forest ecologists, conservationists, and macro-ecologists. Also, the data here presented can be linked to the GenTree Dendroecological collection, the GenTree Leaf Trait collection, and the GenTree Genomic collection presented elsewhere, which together build the largest evolutionary forest ecology data collection available.

As the most common grain contaminant worldwide, deoxynivalenol is of high importance despite its low toxicity compared to other trichothecene mycotoxins. Data on the effects of deoxynivalenol in rabbits are scarce. Thus, the aim of this study was to investigate the effects of dietary deoxynivalenol fed at a high level (10 mg/kg of feed) on the productive performance, blood indices, immunological variables, histopathological changes, and genotoxicity in rabbits. Forty-eight Pannon White rabbits were exposed to contaminated diets for three weeks. Despite its high concentration, deoxynivalenol did not affect the feed intake, body weight, and body weight gain. Liver and kidney function was not affected, as shown by the clinical chemistry indices. Conversely, in two rabbits the toxin caused mild fibrosis of the liver, without degenerative changes of the hepatocytes. No genotoxicity could be observed either. Gut cytokines and the phagocytic activity of the macrophages did not differ significantly. The percentage of neutrophils was significantly lower, whereas that of eosinophils was significantly higher in the toxin-fed group. Deoxynivalenol did not cause significant changes in gut and villus morphology. In 4 out of the 6 deoxynivalenol-treated animals, the ratio of lymphoblast proliferation and simultaneous apoptosis shifted towards apoptosis in the gut-associated lymphoid tissue. In the central part of the lymphoid follicles of the spleen, lymphocyte depletion and follicular atrophy could be detected. It can be concluded that rabbits are less sensitive to deoxynivalenol, but the findings confirm that this Fusarium toxin is capable of modulating the immune response.

The present study aimed to test the effect of a dietary supplementation with Silybum marianum (SM), an herbaceous Mediterranean plant traditionally used to treat liver and gastrointestinal diseases and with antioxidant properties, on the productive performance, carcass traits and meat quality of growing rabbits. With this purpose, at weaning (5 weeks of age), a total of 144 Pannon Large rabbits were allocated to three experimental groups. The control group (C, n=51) was fed with a basal diet, whereas the other groups received the basal diet supplemented with SM herbal powder at two concentrations: 5 g/kg (SM1, n=48) and 10 g/kg (SM2, n=45). Rabbits were housed in wire-mesh cages (3 rabbits/cage) and fed ad libitum throughout the experiment. Productive performance and mortality were recorded weekly. Rabbits were slaughtered at 11 weeks of age, carcasses were dissected, and hind leg (HL) and Longissimus thoracis et lumborum (LTL) meat were analysed for meat quality (oxidative status, pHu and L*, a*, b* colour) traits. In addition, a sensory analysis on the LTL meat was carried out by a trained panel. The European ban on the non-therapeutic use of antibiotic growth promoters and limits on the use of other drugs have increased digestive disorders and mortality in growing rabbits. In addition, consumers demand natural products, and therefore synthetic active compounds should be replaced by natural ones. This has increased the search for alternatives, such as herbs, spices and their extracts (botanicals) as replacers. Plants (whole plants, leaves or seeds, mainly used as feedstuffs) and their extracts (considered as additives) are being increasingly used in animal nutrition as appetisers, digestive and physiological stimulants, colorants, and antioxidants, and for the prevention and treatment of certain pathological conditions. The digestive effects of herbs and spices have been tested primarily in humans and laboratory animals, and few trials have been performed on farm animals. Studies on the dietary inclusion of herbs and spices or their extracts in rabbit meat production are quite scarce, and the overall benefit remains unclear due to discrepancies in results, such as the use of plant preparations as galactagogues in rabbit does. Some positive results have been shown their potential, however. The dietary inclusion of Foeniculum vulgare Mill. seeds with oregano leaves has been observed to improve diet utilisation, whereas the dietary inclusion of a mixture of Lupinus albus L., Trigonella foenum-graecum L., and Cassia senna L. has acted as growth promoter. Antimicrobial effects are derived especially from plant volatile oils. In the rabbit, a stabilizing effect on microbiota was observed when the diet was supplemented with thyme oil. When diets were supplemented with thyme leaves and spirulina algae, an antimicrobial effect on Clostridium coccoides, Clostridium leptum in the caecum was observed. Black cumin seeds have been shown to exert anti-inflammatory, anti-bacterial and immunomodulatory effects. Several herbs and spices (green tea, rooibos, oregano, rosemary and thyme) provide antioxidant effects through rabbit dietary supplementation or inclusion in meat and meat products. Research in the use of herbs or/and spices has demonstrated their potential as feed additives and/or antioxidants, but further research is recommended to optimize effects on rabbits before practical proposals can be drafted.

Cold-water corals are recognised as a key component of Mediterranean deep-sea ecosystems but the knowledge of their associated biodiversity is still limited. To date, the fauna associated to the Central Mediterranean cold-water coral habitats is the best known (e.g. the cold-water coral provinces of Santa Maria di Leuca, Bari canyon and Strait of Sicily) but such knowledge should be extended to the whole Mediterranean Basin. The combined biodiversity censed so far for Mediterranean cold-water coral habitats and those of adjacent areas (Strait of Gibraltar-Gulf of Cádiz) includes a conspicuous number of species (ca. 520 spp.) with a high representation of sponges (ca. 90 spp.), polychaetes (ca. 90 spp.), cnidarians (ca. 80 spp.), bryozoans (ca. 75 spp.), crustaceans (ca. 60 spp.), molluscs (ca. 50 spp.), fishes (ca. 50 spp.), echinoderms (ca. 20 spp.) and brachiopods (7 spp.). Most species are not univocally linked to cold-water corals, but they benefit from the complex and diverse microhabitats provided by them. There is a clear need to continue the investigation of Mediterranean cold-water coral habitats to fully document the faunistic inventory, biogeographic connections and functions of the many species connected to such emblematic ecoystems of the deep-sea.

The present study tested the effect of a liquorice root extract (L) administration in the diet of growing rabbits and/or in their minced meat. A total of fifty-six rabbit does and their kits (3 weeks of age) were divided into 4 groups receiving a commercial diet (C) or the C diet supplemented with increasing levels of L: 2 g/kg (L2), 4 g/kg (L4), 6 g/kg (L6). At weaning (5 weeks of age), sixty rabbits/group were randomly selected, caged by 3, and fed ad libitum with the experimental diets until slaughter (12 weeks of age). At 6 weeks of age, 6 rabbits/diet were selected for gastrointestinal tract pH and caecal microbial count analyses. Live performance and health status were recorded weekly and daily, respectively. Carcass traits were measured after slaughter and hind leg (HL) and Longissimus lumborum (LL) meat were dissected for pHu and L*a*b* colour measurements. Minced meat from HL belonging to C and L6 dietary treatments was each split in two parts to incorporate liquorice extract at 0.25% (w/w), obtaining four groups of meat burgers: CC; CL; L6C and L6L. The pH, L*a*b* colour and microbial count (TVC, Coliforms, and Pseudomonas) were measured at days 0, 3 and 6 of refrigerated storage. A sensory analysis was conducted on LL meat of C and L6 dietary treatments, and tested attributes were odour, taste, flavour, texture and off-flavours. Liquorice dietary inclusion did not show a clear effect on live performance, resulting comparable among groups, with exception of the lowest growth rate for L2 rabbits (P

According to the results obtained in the present study, it could be concluded that: 1. Sleep length during the whole week, during the weekdays and weekend is inversely associated with a sweet dietary pattern consumption. On the other hand, sleep length both during the whole week and the weekdays is positively associated with a higher intake of healthy foods. 2. Girls tend to have a higher consumption of sweet foods the less they sleep, during the weekdays and the whole week. Whereas in general they are more likely to follow a healthy dietary pattern the longer the sleeping hours. Such associations do not seem to be present among boys. 3. There do not seem to exist differences between sleep length and dietary patterns when considering different socio-economic backgrounds among children from pre-schools in Finland. However, the literature research and the obtained results highlight the fact that, when talking together about sleep and diet, we are still on a narrow piece of terra ferma in a wide sea of the unknown.

Contamination of feed with mycotoxins is a common problem encountered in animal farming. Mycotoxin exposure can affect adversely the health of animals. In rabbits caecal fermentation is an essential digestive process being indication of physiological alterations. Deoxynivalenol (DON) is one of the most frequent contaminants of grains which affect the growth of monogastric animals. Data about dietary DON and its effect in rabbits are scarce. Medicinal plants are often used as feed additives to enhance the performance of the animals. Carduus marianus (milk thistle) is known for its hepatoprotective and antioxidant effects (among others) but no data are available about the effect on rabbit caecum. Considering the aforementioned, the aim of this study was to assess the possible interactive effect of Carduus marianus and DON on the caecum of growing rabbits. 75 Pannon White rabbits were reared for six weeks from 35 (after weaning) till 77 days of age. Rabbits received the following diets: control (C), control with DON (CT), control supplemented with C. marianus in 0,5% (H1), control supplemented with C. marianus in 0,5% and DON (H1T), control supplemented with C. marianus in 1% (H2) and control supplemented with C. marianus in 1% and DON (H2T). On slaughter, caecum was collected for the analysis of total volatile fatty acids (VFA) and the microbiota of the caecum, pH of the caecum was also recorded. There was no significant difference in total VFA concentration or individual VFA. Number of aerobic bacteria significantly differed among toxin and non-toxin groups. DON affected adversely the number of aerobic bacteria. An interactive effect of DON and Carduus marianus on E.coli number was observed. There was no effect on total or individual VFA amounts.

About 100 participants attended the 26th Hungarian Conference on Rabbit Production at Kaposvár, organised by Kaposvár University, the Hungarian Branch of the WRSA and the Rabbit Production Board. This is the largest and most popular event for rabbit breeders in Hungary. Twenty papers were presented by senior and young scientists. Topics of the papers covered all fields of rabbit production (production, housing and welfare, reproduction, genetics, nutrition, meat quality). Progressive evaluations by the Organization for Economic Co-operation and Development (OECD) demonstrate that health care is now or becoming unaffordable. This means nations must change the way they manage health care. The costly nature of health care in most nations, as a percentage of Gross Domestic Product (GDP) seems independent of the national funding models. Increasing evidence is demonstrating that the lack of involvement by clinicians (doctors, nurses, pharmacists, ancillary care and patients) in e-health projects is a major factor for the costly failures of many of these projects. The essential change in focus required to improve healthcare delivery using e-health technologies has to be on clinical care. To achieve this change clinicians must be involved at all stages of e-health implementations. From a clinicians perspective medicine is not a business. Our business is clinical medicine and e-health must be focussed on clinical decision making. This paper views the roles of physicians in e-health structural reforms.

In a recent work [C. Lozano et al. Phys. Rev. Lett 114, 178002 (2015)] segregation in an horizontally shaken granular layer was studied by analysing the particle-particle interactions in the simplest case possible of a two particles cluster. There, it was found that all clusters are transient (they eventually split if one waits long enough) and the probability distribution function of the separation times displays a power law tail, indicating that the splitting probability is not constant over time. Here, we extend this study to clusters of 3, 5, 10 and 20 particles where we also observe the power law decay of the distribution of cluster splitting time. In addition, we observe a weak increase of the average cluster splitting time with the cluster size, suggesting that interaction forces are non-additive. Finally, we show interesting statistics on the way in which clusters break suggesting that escaping of individual particles in the cluster borders is more likely than cluster breakage in subclusters of similar size.

Granular flow through an orifice can be suddenly halted by the formation of arches in the vicinity of the outlet, which are stable under the action of gravity. They may be broken when an external driving (for instance, vibration) is applied. With the aim of shedding light on the dynamics of arch destruction, we built an experiment consisting of a vertical two-dimensional silo filled with monodisperse beads, to which a constant vibration is applied. It was previously found that an important parameter to predict the robustness of the arch is the angle between consecutive beads. We focus on long-enduring arches and study the angles among the beads along time. We have found that in many cases the dynamics of the largest angle determines the breaking of the arch; it does not only determine where the ¿weakest link¿ is, but also the process that leads to the final destabilization. This is interesting because it can provide information about whether the flow will resume in a well-defined time or not, which is especially useful for industrial processes that have to constantly deal with the possible emergence of clogs.

The experiment aimed to study the effect of Digestarom® dietary inclusion (herbal formulation containing a mixture of essential oils, herbs, spices and extracts) on apparent digestibility and digestive ecosystem of growing rabbits, as well as the effects of its supplementation before and after weaning on growth performance. At kindling, rabbit does and litters were divided into 2 dietary groups (51 does/group) and fed either a control diet (C) or a diet supplemented with 300 mg Digestarom®/kg diet (D) until weaning, which occurred at 35 d (before weaning supplementation). Each group was further divided into 3 dietary groups: CC received the control diet and DD received the D diet from 5 to 12 wk of age, and DC were fed with D (from 5 to 8 wk of age) and C diets (from 8 to 12 wk of age) (after weaning supplementation; 54 kits/group). An in vivo digestibility trial and a faecal microbial count were carried out on growing rabbits that received only the C or D diets during the trial. El siguiente artículo plantea una visión global sobre el estado de la cuestión en el campo de la Historia de la Fotografía durante el periodo 1998-2008. El eje del análisis se centra en la evolución en España de asuntos centrales como el desarrollo metodológico, conceptual, terminológico, y como campo de competencia. Plantea una serie de hitos en la bibliografía y actividad científica en esta disciplina en el ámbito nacional.

Pseudomonas cepacea lipase (PCL) was used to catalyse the transesterification reaction between 1-phenylethanol and vinyl acetate in supercritical carbon dioxide (SCCO2). The enzyme, which is very selective towards one of the isomers of 1-phenylethanol, was very stable in the supercritical medium and showed higher reaction rates. The effect of SCCO2 pressure on the catalytic efficiency of PCL at a constant temperature was evaluated. With increasing pressure of the reaction medium the catalytic efficiency of the enzyme considered improved resulting in negative activation volume (−1340 cm3/mol) approaching critical values. A study of the kinetic mechanism in the SCCO2 reaction medium was also performed. Moreover supercritical sulphur hexafluoride (SCSF6) was used as reaction medium as a point of comparison with SCCO2. Preliminar data showed high stability of the enzyme in this supercritical medium and kinetic curve reported point out increased reaction rates in comparison to those achieved in SCCO2.

The study investigated the effect of Digestarom® dietary supplementation on the reproductive performances of rabbit does. Pannon Ka (maternal line) multiparous does were randomly divided into two dietary groups since insemination and fed ad libitum. In the first group (n=51), rabbit does were fed with a commercial diet (C), whereas in the other group (n=52) they received the same diet supplemented with 300 mg/kg of Digestarom® (D). The experiment lasted for two reproductive cycles (kindling 1=K1; kindling 2=K2). Body weight of does and litter size (kits born total, alive, stillborn) were recorded at kindling. Litter size and litter weight were registered at 7, 14, 21 days of age after nursing, and the average individual weight of kits was calculated. Kits’ mortality was recorded daily. At K1, rabbit does performances were unaffected by dietary treatments. During K2, D does were significantly lighter than C ones (P® did not improve the reproductive performance of rabbit does. They seemed to dislike the D diet in K1 and such behavior could have led to the negative results in K2. Further studies should focus on feed acceptance, dose-dependent effect, physiological adaptation and in vivo oxidative status of does. Finally, several consecutive reproductive cycles are recommended to test the efficacy of new feed supplements.

We report the results of an experimental study of particle-particle interactions in a horizontally shaken granular layer that undergoes a second order phase transition from a binary gas to a segregation liquid as the packing fraction C is increased. By focusing on the behavior of individual particles, the effect of C is studied on (1) the process of cluster formation, (2) cluster dynamics, and (3) cluster destruction. The outcomes indicate that the segregation is driven by two mechanisms: attraction between particles with the same properties and random motion with a characteristic length that is inversely proportional to C. All clusters investigated are found to be transient and the probability distribution functions of the separation times display a power law tail, indicating that the splitting probability decreases with time.

We present experimental results on the endurance of arches that block the outlet of a two-dimensional silo when subjected to vertical vibration. In a recent paper [C. Lozano et al., Phys. Rev. Lett. 109, 068001 (2012)], it was shown that the arch resistance against vibrations is determined by the maximum angle among those formed between each particle in the bridge and its two neighbors: the larger the maximum angle is, the weaker the bridge. It has also been reported that the breaking time distribution shows a power-law tail with an exponent that depends on the outlet size, the vibration intensity, and the load [I. Zuriguel et al., Sci. Rep. 4, 7324 (2014)]. Here we connect these previous works, demonstrating the importance of the maximum angle in the arch on the exponent of the breaking time distribution. Besides, we find that the acceleration needed to break an arch does not depend on the ramp rate of the applied acceleration, but it does depend on the outlet size above which the arch is formed. We also show that high frequencies of vibration reveal a change in the behavior of the arches that endure very long times. These arches have been identified as a subset with special geometrical features. Therefore, arches that cannot be broken by means of a given external excitation might exist.

Candida rugosa lipase (CRL) shows high enantioselectivity toward (1 R ,2 S )-(−)- trans -2-phenyl-1-cyclohexanol enantiomer in acetylation reaction employing vinyl acetate as acyl donor. Attempts to improve reaction yields have pointed out that supercritical CO 2 is the best reaction medium in the studied biocatalytic process. In these conditions an immobilised lipase from Candida rugosa is able to quantitatively resolve racemate with e.e. p 100%.
