Immunolocalization techniques are standard in biomedical research. Tissue fixation with aldehydes and cell membrane permeabilization with detergents can distort the specific binding of antibodies to their high affinity epitopes. In immunofluorescence protocols, it is desirable to quench the sample's autofluorescence without reduction of the antibody-dependent signal. Here we show that adding glycine to the blocking buffer and diluting the antibodies in a phosphate saline solution containing glycine, Triton X-100, Tween20 and hydrogen peroxide increase the specific antibody signal in tissue immunofluorescence and immunogold electron microscopy. This defined antibody signal enhancer (ASE) solution gives similar results to the commercially available Pierce Immunostain Enhancer (PIE). Furthermore, prolonged tissue incubation in resin and fixative and application of ASE or PIE are described in an improved protocol for triple immunogold electron microscopy that is used to show co-localization of GABA-A ρ2 and dopamine D2 receptors in GFAP-positive astrocytes in the mouse striatum. The addition of glycine, Triton X-100, Tween20 and hydrogen peroxide during antibody incubation steps is recommended in immunohistochemistry methods.

Vasoinhibins are a family of N-terminal prolactin (PRL) fragments that inhibit blood vessel growth, dilation, permeability, and survival. The aspartyl endoprotease cathepsin D is active at acidic pH and can cleave rat PRL to generate vasoinhibins. We investigated whether and where vasoinhibins could be generated by cathepsin D in the adenohypophysis of rats and mice and whether their production could be gender dependent. Vasoinhibins were detected in primary cultures of rat adenohypophyseal cells by Western blot with antibodies directed against the N terminus of PRL but not the C terminus. Ovariectomized, estrogen-treated females show greater levels of adenohypophyseal vasoinhibins than males. Peptide sequencing analysis revealed that the cleaved form of PRL in rat adenohypophyseal extracts contains the PRL N terminus and a second N terminus starting at Ser149, the reported cleavage site of cathepsin D in rat PRL. In addition, cathepsin D inhibition by pepstatin A reduced vasoinhibin levels in rat adenohypophyseal cell cultures. Confocal and electron microscopy showed the colocalization of cathepsin D and PRL within rat adenohypophyseal cells and secretory granules, and a subcellular fraction of rat adenohypophysis enriched in secretory granules contained cathepsin D activity able to generate vasoinhibins from PRL. Of note, vasoinhibins were absent in the adenohypophysis of mice lacking the cathepsin D gene but not in wild-type mice. These findings show that cathepsin D is the main protease responsible for the generation of adenohypophyseal vasoinhibins and that its action can take place within the secretory granules of lactotrophs.

We have previously shown that soluble factor(s) in conditioned media (CM) from the central and peripheral regions of the anterior pituitary (AP) gland of lactating rats promoted the in vitro dose-related release of prolactin (PRL) from pituitary glands of male rats. In the present experiments we sought to determine whether CM from rats in different physiological states provoked similar effects (like those of lactating rats), and the nature of the factors, whether 23K PRL or other variants of the hormone, were responsible for these effects. Stimulatory effects were induced by CM from pregnant females and steroid-treated castrated males or females, but not from untreated castrated rats, intact males, or by a PRL standard. More potent effects occurred with CM from APs of early- than from mid- or late-lactating rats, and from rats unsuckled for 8 or 16 h than from those unsuckled for 32 h. With respect to the nature of factor(s) responsible for these effects, immunoprecipitation of PRL from the CM of lactating females and of steroid-treated, castrated males eliminated, whereas dephosphorylation or deglycosylation of CM of lactating rats greatly increased its effects upon PRL release. Bacterial internalization is an important process in the pathogenesis of infectious diseases in which nuclear factor kappaB (NF-kappaB) plays a prominent role. We present pharmacological evidence indicating that in bovine endothelial cells (BEC) the internalization of Staphylococcus aureus, a pathogenic bacterium that causes mastitis in bovine cattle, was associated with the activation of NF-kappaB. The internalization of S. aureus increased when BEC were stimulated with alpha-tumour necrosis factor (TNF-alpha) or beta-interleukin 1 (IL-1beta) which are known activators of NF-kappaB. SN50 (an inhibitor peptide of NF-kappaB nuclear translocation) and BAY 11-7083 (a chemical that inhibits the IkappaBalpha phosphorylation) caused significant reduction in S. aureus intracellular number, indicating that its internalization was associated with the NF-kappaB activity. Furthermore, specific inhibition of c-Jun N-terminal kinase with SP600125 (SP) or p-38 with SB203580 (SB) did not cause any change in the S. aureus intracellular number compared with the untreated control. Finally, TNF-alpha treatment of BEC after the addition of both SP and SB, induced a significant increase in S. aureus internalization above the control value. These data indicate that NF-kappaB activity is associated with S. aureus internalization and suggest that this transcription factor may play a role in the pathophysiology of bovine mastitis caused by this bacterium.

Summary Two accelerated hardening procedures (storage for up to 4 months, and soaking in acetate buffer for up to 5 h) for testing beans were examined and compared, so that new cultivars prone to the hard-to-cook (HTC) defect may be identified. Bean hardness was measured by means of the Mattson bean cooker, and by measuring puncture force. Two imporant common bean (Phaseolis vulgaris) cultivars were grown in two locations differing in soil Ca and Mg content. Those from the high Ca site had longer cooking times, which increased more rapidly on accelerated storage and in the soaking tests. The results of this preliminary study suggest that the acetate soak hardening procedure is worthy of further investigation as the basis of a rapid test for the presence of the hard-to-cook defect in new cultivars.

The mamillary body (MM) is a group of hypothalamic nuclei related to memory and spatial navigation that interconnects hippocampal, thalamic, and tegmental regions. Here we demonstrate that Lhx5, a LIM-HD domain transcription factor expressed early in the developing posterior hypothalamus, is required for the generation of the MM and its derived tracts. The MM markers Foxb1, Sim2, and Lhx1 are absent in Lhx5 knock-out mice from early embryonic stages, suggesting abnormal specification of this region. This was supported by the absence of Nkx2.1 and expansion of Shh in the prospective mamillary area. Interestingly, we also found an ectopic domain expressing Lhx2 and Lhx9 along the anterio-posterior hypothalamic axis. Our results suggest that Lhx5 controls early aspects of hypothalamic development by regulating gene expression and cellular specification in the prospective MM.

Conserved polypeptides of the chitin synthase genes UmCHS3 and UmCHS6 from the phytopathogenic fungus Ustilago maydis were utilized as immunogens to obtain polyclonal antibodies that were purified by affinity procedures. Because of their similarities at the regions encoded by either polypeptide, it was concluded that anti-Chs3 antibodies recognized both Chs3 and Chs4 chitin synthases, whereas anti-Chs6 antibodies recognized Chs6 and Chs8 polypeptides. These antibodies were used to analyze the localization of the corresponding chitin synthases in U. maydis cells, using both indirect immunofluorescence microscopy and immunoelectron microscopy with colloidal-gold-labeled secondary antibodies. It was observed that chitin synthase proteins were accumulated both in the surface and in the cytoplasm of the fungal cells. Electron microscopy images revealed the accumulation of clusters of gold particles in vesicles, providing evidence for the possible origin and destination of chitin synthases in the fungal cells.

Thinner inhalation causes toxic effects in a variety of organs, principally in the central nervous system. Some studies have shown oxidative stress effects of thinner inhalation, such as: activation of free radical processes, decrease of antioxidants, and oxidation products of proteins and lipids but not of DNA. The aim of this study is to investigate the effect of thinner inhalation on DNA. We used the comet assay in conjunction with the enzyme formamidopyrimidine glycoslyase (Fpg). Our results show a significant increase in Fpg-sensitive sites in DNA of lymphocytes from rats exposed to thinner fumes compared to lymphocytes from control rats (p < 0.05). Moreover, DNA damage detected with Fpg shows a high correlation with increased malondialdehyde (MDA) and decreased glutathione (GSH), two widely used biomarkers of oxidative stress. The most abundant base oxidation product found in DNA is 8-oxoguanine; it is the main substrate of Fpg and the most commonly used biomarker for oxidative DNA damage. This suggests that oxidative DNA damage is at least partly responsible for the DNA damage detected by Fpg. We propose the comet assay in combination with Fpg as a sensitive biomarker to monitor exposure to thinner inhalation. Limitations of this method are discussed.

Thinner inhalation is known to induce oxidative stress. Some studies have shown that thinner inhalation causes a decrease of antioxidants and formation of oxidation products of proteins and lipids as well as formamidopyrimidine glycoslyase (Fpg)-sensitive DNA sites. Using the comet assay and the repair-specific enzymes formamido pyrimidine glycosylase (Fpg) and endonuclease III (Endo III) to detect oxidized purines and pyrimidines, respectively, we examined the ability of thinner inhalation to induce oxidative DNA damage in rat lymphocytes. Our results show a high correlation between Fpg- and Endo III-sensitive sites. This, together with our previous results that showed a high correlation between Fpg-sensitive sites and two biomarkers of oxidative stress, suggests that these Fpg-sensitive sites correspond to oxidative damage during the first four weeks of thinner inhalation.

In the in vivo rat heart model with transient (5 min) regional ischemia, as induced by left coronary artery ligation, we have demonstrated that perezone reduces dramatically the incidence of reperfusion-induced-arrhythmias. Administered 5 minutes before coronary occlusion, at a dose of 3.1 mg/kg, this drug effectively protects against the high incidence of arrhythmias and the fall of blood pressure. In addition, it inhibits the release of lactic dehydrogenase and creatine-kinase enzymes to the plasma. We propose that the protective effect of perezone might be related to its well documented action of promoting the release of intramitochondrial Ca2+, thus, maintaining ATP production during reperfusion.

The phycobilisomes (PBS) of the cyanobacterium Arthrospira (Spirulina) maxima have been isolated from axenically grown cells. The largest PBS were obtained from samples of the axenic culture harvested in the stationary phase. The fluorescence emission maximum is between 670 and 680 nm. Morphologically, the PBS have an hemidiscoidal shape with a tricylindrical core, and six rods arranged radially around the core. The polypeptide composition was characterized by a combination of isoelectrofocusing, SDS-PAGE and reverse phase HPLC. Five distinct groups of polypeptides are described: (1) an 80 kDa polypeptide bearing a bilin (LCM) isolated by LIDS-PAGE at 4°C; (2) two polypeptides of 50 and 45 kDa present in variable amounts from preparation to preparation; (3) three polypeptides of 36, 33 and 30 kDa molecular mass, identified as rod and core-rod linker polypeptides; (4) six different chromoproteins with molecular masses around 18 kDa, identified as αPC, βPC, αAP and βAP, αAP-B, and β17. In the PBS of A. maxima, phycocyanin is the major constituent, four times more abundant than allophycocyanin; and (5) two small colorless polypeptides, of 10 and 8 kDa molecular mass. Electrofocusing was performed on the PBS components and all the subunits bearing a chromophore showed an acidic pI. All the linkers showed a basic pI except the small polypetide of LR10, which showed a pI of 5. We conclude that the PBS of A. maxima exhibit a hemidiscoidal form and a tricylindrical core.

The effects of acute ethanol and acetaldehyde treatment on cell proliferation, cell adhesion capacity, neutral red incorporation into lysosomes, glutathione content, protein sulfhydryl compounds, lipid peroxidation, inner mitochondrial membrane integrity (MTT test), lactate dehydrogenase activity (LDH) and ultrastructural alterations were investigated in a human fetal hepatic cell line (WRL-68 cells). WRL-68 cells were used, due to the fact that, although this cell line expresses some hepatic characteristics, it does not express alcohol dehydrogenase or cytochrome P450 activity, so it could be a good model to study the effect of the toxic agents per se. Cells were exposed during 120 min with 200 mM ethanol or 10 mM acetaldehyde. Under these conditions, cells presented 100% viability and no morphological alteration was observed by light microscopy. Acetaldehyde-treated cells reduced their proliferative capacity drastically while the ethanol-treated ones presented no difference with control cells. Cell adhesion to substrate, measured as time required to adhere to the substrate and time required to detach from the substrate, was diminished in acetaldehyde WRL-68-treated cells. Cytotoxicity measures as neutral red and MTT test showed that acetaldehyde-treated cells presented more damage than ethanol-treated ones. Cellular respiratory capacity was compromised by acetaldehyde treatment due to 40% less oxygen consumption than control cells. Lipid peroxidation values, measured as malondialdehyde production, were higher in ethanol-treated WRL-68 cells (127%) than in acetaldehyde-treated ones (60%) to control cell values. Lactate dehydrogenase activity (LDH) in extracellular media of ethanol-treated cells presented the highest values. GSH content was reduced 95% and thiol protein content was diminished severely in acetaldehyde-treated cells. Transmission electron microscopy showed more ultrastructural alterations in cells treated with acetaldehyde. The results indicate that acetaldehyde, like ethanol, produced damage at cellular level, although more damage could be observed in acetaldehyde WRL-68-treated cells.

Exposure to lead induces oxidative stress and renal damage. Although most forms of oxidative stress are characterized by simultaneous elevation of nitrogen and oxidative species, lead-induced oxidative stress is unusual in that it is associated with a reduction in nitric oxide (NO) levels in the kidney. The role of NO in kidney injury is controversial; some studies suggest that it is associated with renal injury, whereas others show that it exerts protective effects. Concentration-dependent effects have also been proposed, linking low levels with vasodilatation and high levels with toxicity. The aim of this study was to evaluate the effects of melatonin co-exposure on the lead-induced reduction in renal NO levels. We found that sub-acute intraperitoneal administration of 10 mg/kg/day of lead for 15 days induced toxic levels of lead in the blood and caused renal toxicity (pathological and functional). Under our experimental conditions, lead induced an increase in lipid peroxidation and a decrease in NO. Melatonin co-treatment decreased lead-induced oxidative stress (peroxidation level) and toxic effects on kidneys without altering the lead-induced reduction in renal NO. These results suggest that, in our experimental model, the reduction in renal NO levels by lead exposure is not the only responsible factor for lead-induced kidney damage.

Fungal Genetics and Biology20,133–142. Isolated orin vivocell walls from the yeast and mycelial forms of haploidUstilago maydiswere not stained by the normal osmium procedure for electron microscopy. KMnO4stained mycelial walls, revealing a layered structure with a loose electron-dense layer at the cell surface, but stained only the outer surface layer of yeast walls. Walls were purified from extracts obtained by ballistic and ultrasonic disruption. Chemical analysis showed that composition of yeast and mycelial walls was similar. Yeast walls contained higher amounts of neutral sugars and protein, whereas mycelial walls contained more chitin and phosphate. No chitosan or uronic acids were detected. Higher proportions of xylose and mannose were present in yeast walls, whereas the amounts of glucose and galactose were higher in mycelial walls. Fucose, arabinose, and ribose were detected in yeast walls only. Electrophoretic patterns of proteins extracted with SDS, β1,3-glucanase, or chitinase were similar in walls of both morphologies, although some differential bands were identified. Most antigenic proteins appeared in the covalently bound fraction of the wall. Some were common to both morphologies, but others were stage specific.

The toxic effects of cadmium (Cd) and mercury (Hg), as chloride salts, were studied using an hepatic human fetal cell line (WRL-68 cells). From viability curves and the proliferative capacity of the cell in the presence of the metal, three different cell treatments were chosen, (1) 0.5 microM of the metal chloride for 24 h (acute low dose treatment), (2) 0.5 microM of the metal chloride for 7 days (chronic treatment), and (3) 5 microM of the metal chloride for 24 h (acute high dose treatment). WRL-68 cells grown in the presence of Cd exhibited the same proliferative curve as control cells, whereas in the case of Hg, the cells increased their proliferative capacity. Both metals produced ultrastructural alterations in different degrees, mainly observed as mitochondrial and RER structural changes, depending of the treatment and concentration of the metal used. Cytotoxicity was assessed by measuring the release of lactate dehydrogenase from the cells. Acutely high dose-treated cells showed the highest value for this parameter, and Cd-treated cells presented higher lactate dehydrogenase release than the Hg-treated ones. Cell damage was also measured by alanine aminotransferase (ALAT) and aspartate aminotransferase (ASAT) activities. Acute high dose Cd treatment caused the highest value of enzymatic release. Lipid peroxidation was significantly different with respect to control cells in chronic and acute high dose treatments with both metals. Metallothionein (MT) induction in response to Hg treatment was not detected. However, a dramatic induction of this protein occurred in Cd-treated cells. WRL-68 cells differentially respond to Cd and Hg making this hepatic fetal human cell line a useful tool in investigating the mechanism of toxicity of these heavy metals.

Summary: Control of the pH of liquid synthetic culture media made possible mycelial growth of both diploid and haploid strains of Ustilago maydis. Whereas at neutral pH the fungus grew as a homogeneous population of budding yeast-like cells (sporidia), at acid pH it developed in the mycelial form. Mycelial cells appeared branched and narrower than yeast cells. Cell morphology was affected by the carbon and nitrogen sources. When the culture medium was removed continuously or intermittently, very long, filamentous cells were formed. Colonies of haploid strains developed aerial mycelium (‘fuzz’ morphology) on acid solid medium. Null b, bW, and bE mutants behaved in the same way as haploid wild-type strains. It is suggested that growth at low pH overcomes the control processes governed by heterologous bE and bW loci.

Physicochemical and structural properties as well as proneness to hardening were assessed in normal (Mayocoba) and slow (Flor de Mayo) cooking varieties of beans. Storage and chemical procedures were used for determination of the hardening of both samples. Normal and slow cooking varieties had cooking times of 58 and 90 min, respectively. During storage under high temperature and humidity the normal variety exhibited a four-fold increase in cooking time while that of the slow cooker increased nine-fold. Chemical hardening caused an increase in cooking time of 3.4- and 5.7-fold for normal and slow beans, respectively. The former had lower percentage and thinner seed coat, and higher water absorption in the whole seed and seed coat, but not in the cotyledons, as compared to the latter. Chemical analysis demonstrated a much lower tannin content in fresh and hardened seed coats and cotyledons of the normal cooker, but more phytic acid content in cotyledons than in the slow. In fresh and hardened states the normal cooking variety had lower gelatinization and protein denaturation temperatures and enthalpies than the slow. SEM revealed important structural differences between both varieties and conditions. These characteristics may be used as identifying features linked to hardening proneness.

Humans can come into contact with thinner by occupational exposure or by intentional inhalation abuse. Numerous studies of workers for genotoxic effects of thinner exposure have yielded conflicting results, perhaps because co-exposure to variable other compounds cannot be avoided in workplace exposure studies. In contrast, there is no data concerning the genotoxic effects of intentional inhalation abuse. The aim of this project was to examine the genotoxic effects of thinner inhalation in an animal model of thinner abuse (rats exposed to 3000 ppm toluene, a high solvent concentration over a very short, 15 min time period, twice a day for 6 weeks). The data presented here provides evidence that thinner inhalation in our experimental conditions is able to induce weight loss, lung abnormalities and oxidative stress. This oxidative stress induces oxidative DNA damage that is not a characteristic feature of genotoxic damage. No significant difference in DNA damage and DNA repair (biomarkers of genotoxicity) in lymphocytes from thinner-treated and control rats was found. Lead treatment was used as a positive control in these assays. Finally, bone marrow was evaluated as a biomarker of cellular alteration associated with thinner inhalation. The observed absence of hemopoietic and genetic toxicity could be explained in part by the absence of benzene, the only carcinogenic component of thinner; however, benzene is no longer a common component of thinner. In conclusion, thinner did not cause genotoxic effects in an experimental model of intentional abuse despite the fact that thinner inhalation induces oxidative stress.

A basic procedure was developed to produce a fermented product by solid substrate fermentation using Rhyzopus oligosporus and chickpea as substrate. Water activity was kept at 0.92 throughout the process. Fermentation increased total, ‘true’ and soluble proteins, soluble solids and soluble carbohydrates, and decreased fiber content and pH. About 12% of solids were lost during 72 h of fermentation. The content of most fatty acids was enhanced by fermentation, whereas peroxide value and tannins declined. The color of the fermented product was not deteriorated after 72 h of fermentation. Scanning electron microscopy studies of microbial growth on the substrate showed penetration of the fungus hyphae and degradation effects on the chickpea cotyledon cells.

The basic conditions of an enzymatic process to produce high-protein amaranth flour (HPAF) and carbohydrate rich fraction (CRF) from raw flour were determined. Commercial preparations of α-amylase and glucoamylase were used. Conditions for both enzymes were: 20% (w/v) slurries of gelatinized whole flour and 0.10% (v/w) enzyme; for amylase, pH 6.5, 70°C and 30 min liquefaction time; for glucoamylase, pH 4.5, 60°C and 60 min. The yield of HPAF was 38–39%. HPAF from both enzymes had 26–28% protein, 10–16% fat and 40–52% starch. Protein digestibility (76%) and reactive lysine (6.6–7.1 g/100g protein) of HPAF were comparable to raw flour. CRF had a 17–21 dextrose equivalent.
