To investigate the role of microRNA-582-5p (miR-582-5p) on osteosarcoma (OS) cell behaviors and provide potential therapeutic targets for OS.Expression levels of miR-582-5p in OS cell lines and normal cell lines were measured by quantitative real-time PCR. Cell proliferation, migration and invasion capacities were assessed by cell counting kit-8 assay, wound-healing assay, and transwell invasion assay, respectively. Downstream target of miR-582-5p was confirmed by Luciferase activity reporter assay and Western blotting assay.MiR-582-5p expression was downregulated in OS cell lines compared with normal cell lines. Overexpression of miR-582-5p inhibits OS cell proliferation, migration, and invasion capacities. Neuro-oncological ventral antigen 1 (NOVA1) was chosen as target gene of miR-582-5p by bioinformatics analysis and Luciferase reporter assay. Moreover, overexpression of NOVA1 could impair tumor suppression role of miR-582-5p on OS cell behaviors.MiR-582-5p exerts tumor-suppressive role on OS cell behaviors via targeting NOVA1 in vitro, which will help us to understand the mechanisms underlying OS progression.

Summary The genus Echinochloa (Poaceae) includes numerous problematic weeds over a wide range of ecoregions in the world. To date, molecular markers for species identification and assessing phylogenetic relationship are still limited in the genus Echinochloa. In this study, we developed seven chloroplast molecular markers based on divergent chloroplast regions of E. crus-galli and E. oryzicola. Furthermore, Marker #1 (psbA) was examined in more than 200 Echinochloa accessions and a phylogenetic tree grouped these Echinochloa accessions into four clades. Additionally, two different E. crus-galli varieties (E. crus-galli var. crus-galli, E. crus-galli var. praticola) and E. colona were successfully distinguished by this marker. The developed molecular markers contribute to better identification of Echinochloa taxa.

Sirtuin 7 (SIRT7) is a NAD+-dependent deacetylase in the sirtuin family. In a previous study, human bone marrow mesenchymal stem cells (hBMSCs) with reduced SIRT7 activity were developed to evaluate the effect of SIRT7 on osteogenesis. SIRT7 knockdown significantly enhanced osteoblast-specific gene expression, alkaline phosphatase activity, and mineral deposition in vitro. Additionally, SIRT7 knockdown upregulated β-catenin. The enhanced osteogenesis due to SIRT7 knockdown was partially rescued by a Wnt/β-catenin inhibitor. Furthermore, SIRT7 knockdown hBMSCs combined with a chitosan scaffold significantly promoted bone formation in a rat tibial defect model, as determined by imaging and histological examinations. These findings suggest that SIRT7 has an essential role in osteogenic differentiation of hBMSCs, partly by activation of the Wnt/β-catenin signaling pathway.

This paper reports on a hydroponics experiment that was conducted to investigate the effect of inorganic arsenics on the seedlings of the rice cultivar Shanyou63. The seedlings were subjected to two treatments, i.e., As(III) and As(V). The results showed that the morphological traits of the seedlings were significantly altered after the arsenic treatments. Analysis of nitrogen, phosphorus, potassium, and arsenic contents of the roots and leaves of the seedlings indicated that the absorption of phosphorus and potassium was mainly affected by As(III), while that of nitrogen was mainly affected by As(V). The expression of 12 genes involved in the absorption and utilization of nitrogen, phosphorus, and potassium were all observed to be down-regulated after the arsenic treatments. As(V) significantly affected the absorption and utilization of nitrogen, while As(III) significantly affected those of phosphorus and potassium. The result obtained by real-time FQ-PCR regarding the difference in the gene expressions agreed with that of our hydroponics experiment.

Robust control of atomic coherence and population transfer among Zeeman sublevels in the ground states of rubidium atom is investigated using adiabatic rapid passage in a nanosecond time scale, which is smaller than the lifetime of first excited Rb. It is shown that a slight change in the pump pulse time delay relative to the Stokes pulse leads to a significant modification of atomic coherence and population transfer, consequently having remarkable impacts on the generation of coherent anti-Stokes Raman scattering (CARS) signal and probe pulse absorption. This coherent control of quantum state and population is presented by numerical simulations based on self-consistent set of density matrix equations and Maxwell equations as well as experimental demonstration in rubidium atom with different atomic densities. Experimental observations are in good agreement with numerical calculations.

We observe transmission of the radiation of a diode laser in the mode-locked regime through the cell containing optically dense Rb vapor under the condition where the hyperfine splitting of Rb is a multiple of the modulation frequency.

We report, in this paper, a narrow dip of electromagnetically induced absorption inside the optical pumping window by the interaction of two coherent laser fields with a quasi-degenerate two-level open atomic system in inhomogeneously broadened Rb vapour. The effect arises from optical pumping and laser-induced atomic multilevel Zeeman sublevels in the same ground state. The numerical simulation for a N-type four-level system shows good agreement with experimental observation.

We study both theoretically and experimentally three-photon electromagnetically induced transparency and electromagnetically induced absorption resonances in inhomogeneously broadened 85 Rb atomic vapour driven by probe and drive laser radiations. We observe narrow Doppler-free absorption as well as transmission resonances for the probe field when the driving laser field is redshifted from the D1 or D2 lines of 85Rb; the frequency difference between the drive and probe fields is equal to the hyperfine splitting of the ground state of the atoms, and the probe field is tuned to the centre of the Doppler broadened atomic transition. We theoretically study the spectroscopic effect in both homogeneously and inhomogeneously broadened media. Our numerical simulations are in good agreement with the experimental results.

We have shown that quantum interference in a driven quasi-degenerate two-level atomic system can be controlled by an externally applied magnetic field. We demonstrate that the mechanism of optical control is based on quantum interference, which allows one to implement both electromagnetically induced transparency and electromagnetically induced absorption (EIA) in one atomic system. The experimental realization is suggested.

Spectroscopy of the Doppler broadened Rb atomic vapour coherently driven by two coupling fields is presented. Double dark resonances have been observed in different geometries of the coupling fields. A simple four-level model explains the basic counter-intuitive features of experimental spectra. The eigenvalues of the atom-field system distinguishing between the various geometries clarify the physics.

A 32-year old male patient was wounded by a pistol. As shown in computed tomography (CT) scanning images, there was comminuted fracture of the left mandible and the bullet was found in the left side behind the sixth cervical vertebra. After the patient was hospitalized, the debridement was done in the emergency room and the operation of open reduction and internal fixation for comminuted fracture of left mandible was performed successfully. Eighteen days later, the patient was taken to surgery for anterior cervical decompression and fusion with autogenous iliac bone grafting for the sixth cervical vertebra. Postoperative follow-up of the patient over two years indicated that the left biceps muscle strength was recovered to level 4. Gunshot wound to the face associated with injury of the low cervical spine has the possibility of survival. It is safe to treat facial wounds early in the patient's treatment course, even if the bullet remains in the cervical vertebral body and there is neurological function damage.

We studied a propagation of comb of optical pulses produced by a mode-locked diode laser in rubidium vapor. EIT was observed when pulse repetition rate is subharmonic of the hyperfine splitting of the ground state.

We have experimentally demonstrated the enhancement of coherent Raman scattering in Rb atomic vapor by exciting atomic coherence with fractional stimulated Raman adiabatic passage. Experimental results are in good agreement with numerical simulations. The results support the possibility of increasing the sensitivity of CARS by preparing atomic or molecular coherence using short pulses.

We demonstrate a new approach to fast optical switching with a technique based on stimulated Raman adiabatic passage in which a laser pulse switches the probe field on and off via another coupling pulse. This new kind of optical switching is not limited by the decay rate of an excited state and can operate in the subnanosecond time domain. The experimental observation in Rb atomic vapor is in good agreement with numerical simulations.

We demonstrate experimentally the effect of a three-photon electromagnetically induced transparency in hot ${}^{85}\mathrm{Rb}$ atomic vapor driven by two coherent electromagnetic fields. The narrow transparency window is observed on the background of high-contrast Doppler-free subnatural absorption resonance. Our analytical calculations and numerical simulations are in good agreement with the experimental data. Both the transparency and the absorption resonances can find applications in high-precision metrology.

Doppler-free resonances have been observed in Rb atomic vapor coherently driven by two strong-coupling fields in an intrinsically non-Doppler-free geometry. A four-level theoretical model explains the experimental results. The explanation of the physics is based on the interplay between coherences generated in a four-level system.

The width of the electromagnetically induced transparency resonance is studied in rubidium vapor. Nonlinear dependence of the width on drive intensity that is caused by Doppler broadening and optical pumping is found. Density-matrix analysis supports the observed dependence. The discrepancies between experimental data and theoretical prediction are discussed.

To investigate the clinical results of the medial multiplex flap pedicled with the posterior tibial vessel.Twelve cases with soft tissue defects and bone defects of limbs were treated with the medial multiplex flap pedicled with the posterior tibial vessel from September 1992 to May 1999. Among them, bone and soft tissue defects following opened fracture in 7 cases, chronic ulcer following chronic osteomyelitis in 2 cases, melanoepithelioma in 2 cases, bone and soft tissue defects following osteoma resection in 1 case. The bone defect area was from 2.5 cm x 5.0 cm to 4.5 cm x 11.0 cm. Free graft was performed in 5 cases, bridged transposition in 3 cases and reversal transposition in 4 cases, among them, periosteal myocutaneous flap with autogenous or allogeneic bone grafting in 8 cases, myocutaneous flap in 4 cases. The area of the flaps from 6 cm x 8 cm to 12 cm x 25 cm.All flaps were healed by first intention, but in the distal fragments of bigger flaps were partially necrosed in 2 cases. In 10 cases bone healing were obtained after 16 weeks of operation according to the X-ray photos. All cases were followed up from 6 to 18 months. All cases achieved satisfactory result but 1 case died because of lung metastasis of osteoma.The multiplex graft pedicled with the posterior tibial vessel is an ideal graft for repairing the large soft tissue defects and bone defects, because it has such advantages as adequate blood supply, big vascular diameter, long pedicle and big dermatomic area.

76 cases of counterattack induced by acute organic phosphorus pesticides intoxication were analysed. The activity of cholinesterase did not show a significant difference before the counterattack as compared to that after the counterattack (P0.05). The average dosage of atropine for pre-atropinization was 58.6 +/- 2.7 mg/h and it was 203.9 +/- 17.7 mg/h between the post-counterattack and the second atropinization (P0.01). Compared the mortality in reaching and non-reaching the second atropinization was 22.8% VS 100% (P0.01). The survival rate was 37.5% in collaborating blood transfusion cases and 20.6% in noncollaborating ones (P0.05). We also studied on the treatment, mechanism, precursory symptoms, causes of death, degrees of intoxication and pesticide types etc.

The results of atropine treatment in 506 cases of severe organic phosphorus pesticide intoxication are reported. Among the variables which might effect the results of treatment, the average dosage of atropine was related intimately to the prognosis (standard coefficient of regression bi = 0.4702, P less than 0.01). When the average drug dosage was 40-80 ng/h, the mortality was 26.6% and much lower than other groups (P less than 0.01). Intravenous administration given at regular intervals was better than continuous drip; the mortality dropped to 61.2% from 90.9% (P less than 0.01). Both inadequate atropinization and atropine intoxication were less frequently seen during intermitten injection (P less than 0.01).

7 alpha-Bis (beta-chloroethyl)amino-methyl-6,14-endoethenotetrahydrooripavine (alpha-CAM) was found to bind to opioid receptors irreversibly and react directly with sulfhydryl (SH) groups in P2 preparations of rat brain. The P2 preparations were pretreated as follows: protection of the SH groups at the opioid receptor binding sites by morphine or etorphine, and inactivation of the SH groups outside the binding sites by N-ethylmaleimide (NEM), followed by removal of the morphine or etorphine by washing. alpha-CAM was still able to bind the pretreated P2 preparations in an irreversible manner. The results indicate that the formation of covalent bonds between alpha-CAM and the SH groups of opioid receptor binding sites is possibly one of the biochemical mechanisms of the irreversible action of alpha-CAM.

A P2 membrane preparation of rat brain (without cerebellum) was solubilized with CHAPS in Tris containing DTT and trypsin inhibitor. Two opiate ligands, 10b and 10cd, prepared by Liu et al, were employed consecutively in affinity chromatography, from which OPR's were eluted with Nx. The eluate was subsequently passed through a WGA affinity column and the OPR's eluted with N-GluNAc. This eluate was further purified and concentrated by preparative granular gel isoelectric focusing on SG200. Two protein peaks appeared separately at pH 5 and 7.8. The eluates from both peaks were examined for protein contents using the silver staining method, and binding activity was measured by RRA with 3H-etor. The results revealed that both samples contained active OPR purified to over 80,000 fold. The Mr was estimated by gel filtration to be 52 kD and 42 kD for OPR in the pH 5 and pH 7.8 samples respectively. OPR in the pH 5 sample have been determined to be of mu-type by their binding activity with 3H-ohm.

This paper describes a gas chromatographic method for the determination of preservative--ethyl paraben in shengmaiyin Oral Liquid. The method is simple, rapid and accurate. The recovery is good (99.5%), and the coefficient of variation is 0.288%.
