1. The epigenetic hallmark of early-life α-hexabromocyclododecane exposure: From cerebellar 6-mA levels to locomotor performance in adulthood

Abstract: There is a growing evidence that methylation at the N6 position of adenine (6-mA), whose modulation occurs primarily during development, would be a reliable epigenetic marker in eukaryotic organisms. The present study raises the question as to whether early-life exposure to α-hexabromocyclododecane (α-HBCDD), a brominated flame retardant, may trigger modifications in 6-mA epigenetic hallmarks in the brain during the development which, in turn could affect the offspring behaviour in adulthood. Pregnant Wistar rats were split into two groups: control and α-HBCDD (66 ng/kg/per os, G0-PND14). At PND1, α-HBCDD levels were assessed in brain and liver by LC-MS/MS. At PND14, DNA was isolated from the offspring’s cerebellum. DNA methylation was measured by 6-mA-specific immunoprecipitation and Illumina® sequencing (MEDIP-Seq). Locomotor activity was finally evaluated at PND120. In our early-life exposure model, we confirmed that α-HBCDD can cross the placental barrier and be detected in pups at birth. An obvious post-exposure phenotype with locomotor deficits was observed when the rats reached adulthood. This was accompanied by sex-specific over-methylation of genes involved in the insulin signaling pathway, MAPK signaling pathway as well as serotonergic and GABAergic synapses, potentially altering the normal process of neurodevelopment with consequent motor impairments crystalized at adulthood.

Abstract: We present the design, manufacturing and performance of the horn-switch system developed for the technological demonstrator of QUBIC (the $Q$\&$U$ Bolometric Interferometer for Cosmology). This system is constituted of 64 back-to-back dual-band (150\,GHz and 220\,GHz) corrugated feed-horns interspersed with mechanical switches used to select desired baselines during the instrument self-calibration. We manufactured the horns in aluminum platelets milled by photo-chemical etching and mechanically tightened with screws. The switches are based on steel blades that open and close the wave-guide between the back-to-back horns and are operated by miniaturized electromagnets. We also show the current development status of the feedhorn-switch system for the QUBIC full instrument, based on an array of 400 horn-switch assemblies., 30 pages, 28 figures. We manufactured the horns in aluminum platelets milled by photo-chemical etching and mechanically tightened with screws. The switches are based on steel blades that open and close the waveguide between the back-to-back horns and are operated by miniaturized electromagnets. The measured electromagnetic performance of the feedhorns agrees with simulations. In particular we obtained a return loss around -20 dB up to 230 GHz and beam patterns in agreement with single-mode simulations down to -30 dB. The switches for this prototype were designed and built for the 150 GHz band. In this frequency range we find return and insertion losses consistent with expectations (< -25 dB and ∼-0.1 dB, respectively) and an isolation larger than 70 dB. In this paper we also show the current development status of the feedhorn-switch system for the QUBIC full instrument, based on an array of 400 horn-switch assemblies.

Abstract: International audience; The Q & U Bolometric Interferometer for Cosmology (QUBIC) is a cosmology experiment that aims to measure the B-mode polarization of the cosmic microwave background (CMB). Measurements of the primordial B-mode pattern of the CMB polarization are in fact among the most exciting goals in cosmology as it would allow testing of the inflationary paradigm. Many experiments are attempting to measure the B-modes, from the ground and the stratosphere, using imaging Stokes polarimeters. The QUBIC collaboration developed an innovative concept to measure CMB polarization using bolometric interferometry. This approach mixes the high sensitivity of bolometric detectors with the accurate control of systematics due to the interferometric layout of the instrument. We present the calibration results for the Technological Demonstrator, before its commissioning in the Argentinian observing site and preparation for first light.

Abstract: A marked reduction in fertility and an increase in adverse reproductive outcomes during the last few decades have been associated with occupational and environmental chemical exposures. Exposure to different types of pesticides may increase the risks of chronic diseases, such as diabetes, cancer, and neurodegenerative disease, but also of reduced fertility and birth defects. Both occupational and environmental exposures to pesticides are important, as many are endocrine disruptors, which means that even very low-dose exposure levels may have measurable biological effects. The aim of this review was to summarize the knowledge collected between 2000 and 2020, to highlight new findings, and to further interpret the mechanisms that may associate pesticides with infertility, abnormal sexual maturation, and pregnancy complications associated with occupational, environmental and transplacental exposures. A summary of current pesticide production and usage legislation is also included in order to elucidate the potential impact on exposure profile differences between countries, which may inform prevention measures. Abstract: International audience; The Q and U Bolometric Interferometer for Cosmology (QUBIC) Technical Demonstrator (TD) aiming to shows the feasibility of the combination of interferometry and bolometric detection. The electronic readout system is based on an array of 128 NbSi Transition Edge Sensors cooled at 350mK readout with 128 SQUIDs at 1K controlled and amplified by an Application Specific Integrated Circuit at 40K. This readout design allows a 128:1 Time Domain Multiplexing. We report the design and the performance of the detection chain in this paper. The technological demonstrator unwent a campaign of test in the lab. Evaluation of the QUBIC bolometers and readout electronics includes the measurement of I-V curves, time constant and the Noise Equivalent Power. Abstract: International audience; QUBIC (a Q and U Bolometric Interferometer for Cosmology) is a next generation cosmology experiment designed to detect the B-mode polarisation of the Cosmic Microwave Background (CMB). A B-mode detection is hard evidence of Inflation in the ΛCDM model. QUBIC aims to accomplish this by combining novel technologies to achieve the sensitivity required to detect the faint B-mode signal. QUBIC uses technologies such as a rotating half-wave plate, cryogenics, interferometric horns with self-calibration switches and transition edge sensor bolometers. A Technical Demonstrator (TD) is currently being calibrated in APC in Paris before observations in Argentina in 2021. As part of the calibration campaign, the spectral response of the TD is measured to test and validate QUBIC's spectro-imaging capability. This poster gives an overview of the methods used to measure the spectral response and a comparison of the instrument data with theoretical predictions and optical simulations.

Abstract: Alongside the analysis of urinary metabolites which are traditional biomarkers of polycyclic aromatic hydrocarbons (PAH) exposure, the possibility of detecting PAH as well as their metabolites in hair has also recently been demonstrated. As the concentration of pollutants detected in hair is not impacted by short-term variations in exposure as can be observed with urine, it accurately represents an individual’s average level of exposure, which is the most relevant information when investigating possible linkages with biological effects. In the current study, based on a rat model exposed to a mixture of PAHs for a 90-day period, the linkage between the PAH exposure level and the resulting concentration of their metabolites in hair was then investigated. Abstract: Recent studies have identified carbon nanotube (CNT)-induced epigenetic changes as one of the key players in patho-physiological response. In the present study, we investigated whether CNT exposure is associated with epigenetic changes in human bronchial epithelial cells (16 HBE), in vitro. We focused on global DNA methylation, methylation of LINE-1 elements and promoter sequence of twelve functionally important genes (SKI, DNMT1, HDAC4, NPAT, ATM, BCL2L11, MAP3K10, PIK3R2, MYO1C, TCF3, FGFR 1 and AGRN). Additionally, we studied the influence of CNT exposure on miRNA expression. Using a LC-MS/MS method and pyrosequencing for LINE-1, we observed no significant changes in global DNA methylation (%) between the concentrations of multi-walled and single-walled CNT (MWCNT and SWCNT, respectively). Significant changes in sequence-specific methylation was observed in at least one CpG site for DNMT1 (SWCNT), HDAC4 (MWCNT), NPAT/ATM (MWCNT and SWCNT), MAP3K10 (MWCNT), PIK3R2 (MWCNT and SWCNT) and MYO1C (SWCNT). While changes in DNA methylation of the genes were relatively small, these changes were associated with changes in RNA expression, especially for MWCNT. However, the study did not reveal any significant alteration in the miRNA expression, associated with MWCNT and SWCNT exposure. Based on our results, mainly MWCNT influence DNA methylation and expression of the studied genes and could have significant impact on several critical cellular processes.

Abstract: OBJECTIVE: Accurate knowledge of the quantity of released monomers from composites is important. To evaluate the elution of monomers, polymerized composites are typically immersed in an extraction solvent. The objective was to determine whether the volume of extraction solvent and the immersion time influences monomer leachability from dental composite materials. METHODS: Composite disks of two commercial composites, (Filtek Supreme XTE, 3M ESPE and G-aenial Universal Flo, GC) were prepared. The disks (n=10) were placed in a glass vial with 1ml, 2ml or 3ml of extraction solvent (100% ethanol with deuterated diethylphalate as internal standard). After either 7 or 30 days at 37°C, the supernatant was collected and the amount of released monomers (BisEMA, BisGMA, UDMA, TEGDMA) and bisphenol A was measured with liquid chromatography mass spectroscopy. RESULTS: For both tested composites, the highest amount of released monomers was measured after sample incubation in 3ml, while the lowest amount was measured in 1ml of extraction solvent. Furthermore, 30 days did not result in much more monomer release compared to 7 days, and for most monomers, there was no statistically significant difference in release between 7 and 30 days. SIGNIFICANCE: Release kinetics in in-vitro experiments are also influenced by saturation of the extraction solvent with the leached monomers. This is important as it is unlikely that saturation can be reached in an in-vivo situation, where saliva (or pulpal fluid) is continuously refreshed. Abstract: This paper contains the SSR spatial group analysis, following to determinate the forces transmission index. It comes to conclusion, this index is a two variable function, defined by discrete values and graphically represented by a surface. To avoid very high values of reactions, the forces transmission index must be inferior limited. Verifying of this limitation is done by the above mentioned surfaces level curves. This method is performed by authors, and it had been used previously for planar linkages only.

Abstract: Introduction In order to prevent neurodegenerative diseases such as chronic solvent encephalopathy (CSE), it is important to elucidate the underlying mechanism. Cells are able to respond to occupational stressors by turning on or off specific genes mediated through epigenetic alterations. Consequently, epigenetic mechanisms can play an important role in mediating the effects of solvent exposure on the development of neurobehavioral disorders. Methods We have set-up several studies in a translational design using cell lines, mice and humans exposed to solvents and CSE patients. DNA methylation was measured with liquid chromatography coupled with tandem mass spectrometry for the assessment of 2’-deoxycytidine, 5-methyl-2’-deoxycytidine, 5-hydroxymethyl-2´-deoxycytidine, 5-formyl-2´-deoxycytidine, 5-carboxy-2´-deoxycytidine and pyrosequencing. Results Global DNA methylation and hydroxymethylation was mostly negatively associated with solvent exposure in both in vitro as in vivo studies. DNMT1 CpG1 gene-specific methylation levels were significantly higher in CSE patients as compared to controls, associations that persisted after adjustment for age, gender, alcohol abuse, smoking, level of education and the use of psychotropic medication (p=0.004 and p=0.017, respectively). In addition, in the group of CSE patients there was a positive correlation between the duration of solvent exposure and the percentage of BDNF CpG1 methylation (rs=0.374, p=0.017), which was maintained after adjustment for age and smoking (p Conclusion The study of epigenetic alterations gives us the possibility to study the immediate consequences of exposure and make projections for disease development. These epigenetic marks can be used to develop a set of biomarkers and used in the follow-up of humans.

Abstract: Introduction Environmental factors, such as nutrition and occupational exposure can influence epigenetic marks like DNA methylation, which play a role in the development of chronic diseases. Methods Data of the MAternal Nutrition and Offspring’s Epigenome (MANOE) study was used to assess the effect of maternal occupation on maternal and infant DNA (hydroxy)methylation levels. Mothers were categorised in job categories according to the International Standard Classification of Occupations (ISCO). Maternal global DNA (hydroxy)methylation levels during each trimester of pregnancy and at delivery (n=122) was measured in whole blood via LC-MS/MS. Data were analysed with a one-Way ANOVA. Results We found statistically significant differences in maternal global DNA methylation (p=0.008) and global DNA hydroxymethylation (p=0.004) at 20 weeks of pregnancy. Post hoc tests revealed that global DNA methylation and global DNA hydroxymethylation level was significantly lower when the mother had an intellectual/scientific/artistic profession (6.36% and 0.13%) as opposed to being a manager (7.77%, p=0.007% and 0.22%, p=0.002) or administrative staff (7.71%, p=0.003% and 0.2%, p=0.005). No significant differences between different working groups were found for global DNA (hydroxy)methylation in the first and third trimester of pregnancy and at delivery. Conclusion The mother’s occupation was associated with maternal global DNA (hydroxy)methylation levels only in the second trimester of pregnancy. The change in maternal global DNA (hydroxy)methylation in the second trimester of pregnancy could be due to hormonal changes during pregnancy, a shift in the one-carbon metabolism in the middle of pregnancy, but based on these results we also have to take into account maternal occupational exposure.

Abstract: Background: Subtle DNA methylation alterations mediated by carbon nanotubes (CNTs) exposure might contribute to pathogenesis and disease susceptibility. It is known that both multi-walled carbon nanotubes (MWCNTs) and single-walled carbon nanotubes (SWCNTs) interact with nucleus. Such, nuclear-CNT interaction may affect the DNA methylation effects. In order to understand the epigenetic toxicity, in particular DNA methylation alterations, of SWCNTs and short MWCNTs, we performed global/genome-wide, gene-specific DNA methylation and RNA-expression analyses after exposing human bronchial epithelial cells (16HBE14o-cell line). In addition, the presence of CNTs on/in the cell nucleus was evaluated in a label-free way using femtosecond pulsed laser microscopy. Results: Generally, a higher number of SWCNTs, compared to MWCNTs, was deposited at both the cellular and nuclear level after exposure. Nonetheless, both CNT types were in physical contact with the nuclei. While particle type dependency was noticed for the identified genome-wide and gene-specific alterations, no global DNA methylation alteration on 5-methylcytosine (5-mC) sites was observed for both CNTs. After exposure to MWCNTs, 2398 genes were hypomethylated (at gene promoters), and after exposure to SWCNTs, 589 CpG sites (located on 501 genes) were either hypo-(N = 493 CpG sites) or hypermethylated (N = 96 CpG sites). Cells exposed to MWCNTs exhibited a better correlation between gene promoter methylation and gene expression alterations. Differentially methylated and expressed genes induced changes (MWCNTs > SWCNTs) at different cellular pathways, such as p53 signalling, DNA damage repair and cell cycle. On the other hand, SWCNT exposure showed hypermethylation on functionally important genes, such as SKI proto-oncogene (SKI), glutathione S-transferase pi 1 (GTSP1) and shroom family member 2 (SHROOM2) and neurofibromatosis type I (NF1), which the latter is both hypermethylated and downregulated. Conclusion: After exposure to both types of CNTs, epigenetic alterations may contribute to toxic or repair response. Moreover, our results suggest that the observed differences in the epigenetic response depend on particle type and differential CNT-nucleus interactions. Abstract: This paper continues some ideas accepted in scientific literature or enunciates by us in previous papers. As title indicates, we treated the case of 0/3/3 structural group with revolute joints, and application of this group in mechanisms. Three different proposals were done and discussed for the forces transmission indices. Finally an application was shown for a mechanism used in a leather ready-made industry.

Abstract: This paper proposes an approach to identify and study the critical configurations of the 6R (033) structural group, based on an new idea, similar to the case of dyad groups. Starting up from the idea that the critical configurations of a structural group correspond to the singularities of the equation system, modeling the velocity problem and the critical positions are obtained when the lines through the binary link joints are concurrent in a point or are parallel. The velocity equation system for the case of 0/3/3 group with revolute joints, in conditions that the (6x6) system determinant equals zero.

Abstract: The paper represents a synthesis of the most representative results obtained by authors, along their researches in the field of mechanism singularities, forces transmission quality in mechanisms and self-blocking phenomenon. Our research is based on the idea that mechanism critical positions are a consequence of critical configurations in the component structural groups. At its turn, the critical configurations are noticed when the mathematical model of the kinematics and statics without friction, is affected by singularities. Thus, we have presented in the paper, the case of the principal structural groups, and we have established the critical configuration, the calculus formula of the configuration parameter and the existence conditions of the configuration (real solutions in the position problem).

29. Dietary glutamine, glutamic acid and nucleotides increase the carbon turnover (δ13C) on the intestinal mucosa of weaned piglets

Abstract: This study aimed at evaluating the influence of dietary glutamine, glutamic acid and nucleotides on duodenal and jejunal carbon turnover, and on mucosa morphometry of piglets weaned at an age of 21 days. The diets were: additive-free diet – control (C); 1% of glutamine (G); 1% of glutamic acid (GA); and 1% of nucleotides (N). In intestinal mucosa morphometry trial, 65 animals were used. At day 0 (baseline), five animals were slaughtered to determine the villus height (VH), crypt depth (CD), VH : CD ratio and villi density (VD). The remaining 60 animals were allocated into a randomized block design with 4×3 factorial arrangement (four diets: C – control, G – glutamine, GA – glutamic acid and N – nucleotides; three slaughter ages: 7, 14 and 21 days post-weaning) with five piglets slaughtered per treatment. In carbon turnover trial, 123 animals were used. At day 0 (baseline), three animals were slaughtered to quantify the δ13C half-life (T50%) and the 99% carbon substitution (T99%) on intestinal mucosa. The remaining 120 animals were blocked by three weight categories (light, medium and heavy) and, randomly assigned to pen with the same four diets from the previous trial with one piglet slaughtered per weight category per treatment at days 1, 2, 4, 5, 7, 9, 13, 20, 27 and 49 after weaning. Morphometric analyses have yielded no consistent results regarding the action of the evaluated additives, and few reproducible age-related effects. The N diets determined lower T50% values (5.18 days) and T99% (17.21 days) than G and C diets (T50%=7.29, 7.58 days and T99%=24.22, 25.17 days, respectively) in the duodenal mucosa. In jejunum, the N, GA and G diets determined the lowest T50% means (4.9, 6.2 and 6.7 days, respectively) and T99% means (15.34, 21.10 and 21.84 days, respectively) in comparison with C diets (T50%=7.44 and T99%=24.72 days). The inclusion of the additives in the diets of piglets accelerated the carbon turnover in piglets during the post-weaning period. The stable isotopes technique (δ13C) is an important methodology in studies of additives with trophic effects on the intestinal mucosa of the piglets.

Abstract: The present study aimed at evaluating the intensity of Tifton 85 conditioning using a mower conditioner with free-swinging flail fingers and storage times on dehydration curve, fungi presence, nutritional value and in vitro digestibility of Tifton 85 bermudagrass hay dry matter (DM). The dehydration curve was determined in the whole plant for ten times until the baling. The zero time corresponded to the plant before cutting, which occurred at 11:00 and the other collections were carried out at 8:00, 10:00, 14:00, and 16:00. The experimental design was randomised blocks with two intensities of conditioning (high and low) and ten sampling times, with five replications. The high and low intensities related to adjusting the deflector plate of the free iron fingers (8 and 18 cm). In order to determine gas exchanges during Tifton 85 bermudagrass dehydration, there were evaluations of mature leaves, which were placed in the upper middle third of each branch before the cutting, at every hour for 4 hours. A portable gas analyser was used by an infrared IRGA (6400xt). The analysed variables were photosynthesis (A), stomatal conductance (gs), internal CO2 concentration (Ci), transpiration (T), water use efficiency (WUE), and intrinsic water use efficiency (WUEi). In the second part of this study, the nutritional value of Tifton 85 hay was evaluated, so randomised blocks were designed in a split plot through time, with two treatments placed in the following plots: high and low intensity of cutting and five different time points as subplots: cutting (additional treatment), baling and after 30, 60, and 90 days of storage. Subsequently, fungi that were in green plants as well as hay were determined and samples were collected from the grass at the cutting period, during baling, and after 30, 60, and 90 days of storage. It was observed that Tifton 85 bermudagrass dehydration occurred within 49 hours, so this was considered the best time for drying hay. Gas exchanges were more intense before cutting, although after cutting they decreased until ceasing within 4 hours. The lowest values of acid detergent insoluble nitrogen were obtained with low conditioning intensity after 30 days of storage, 64.8 g/kg DM. The in vitro dry matter of Tifton 85 bermudagrass did not differ among the storage times or the conditioning intensities. There was no fungi present in the samples collected during the storage period up to 90 days after dehydration, with less than 30 colony forming units found on plate counting. The use of mower conditioners in different intensities of injury did not speed up the dehydration time of Tifton 85.

Abstract: O estudo objetivou avaliar o turnover do 13C no sangue e plasma de codornas japonesas utilizando a técnica de isótopos estáveis, para a obtenção do patamar de equilíbrio isotópico que servirá de fundamento para estudos de rastreabilidade. Foram utilizadas 300 aves durante o período experimental de 1-42 e 49-97 dias de idade. Os tratamentos da primeira fase foram constituídos de dietas à base de arroz (C3), contendo ou não farinha de carne e ossos bovinos e um com dieta à base de milho (C4). Nessa primeira fase foi analisado o turnover do sinal isotópico do matrizeiro à base de dietas C4 para dietas à base de C3, como também as diferenças isotópicas das dietas contendo ou não farinha de origem animal. Na segunda fase houve uma substituição de dietas, ou seja, as aves no tratamento C4 da primeira fase passaram a consumir dieta C3, e o tratamento que antes consumia dieta C3 passou para dieta C4. Para determinar a taxa de turnover e o percentual estimado de participação da farinha na composição do material coletado, foi empregado o modelo de diluição isotópica utilizando valores do δ13C. A comparação entre as meias-vidas do sangue e plasma da primeira fase revelou o enriquecimento do δ13C na dieta; já na segunda fase foi possível observar as velocidades de incorporação após a troca das dietas.

Abstract: The inclusion of poultry viscera meal (VM) in broiler diets to possibly replace it with strictly vegetable diet (VE) and vice-versa, using the technique of carbon and nitrogen stable isotopes in different tissues was traced. One hundred ninety-two one-day-old chicks that had been distributed randomly were used in twelve experimental treatments with four replications of four chicks each. The treatments consisted of vegetable diet (VG) for diets containing FV going after a certain age or the reverse, in which the birds began feeding VM on diet and then switched to VE diet. At 42 days of age, samples of pectoral muscle (Pectoralis major), keel and tibia were collected, with later determination of isotopic ratios (13C/12C and 15N/14N). The stable isotopes technique couldn't trace the use of VM in feeding broilers when this ingredient was part of broiler diets only in the first seven, 14 or 21 days of age. However, this technique can be applied to younger poultry that have been sampled before a possible change in diet, since they can have the isotopic signature of feeding stabilized around two weeks of age.

A densidade das bebidas foi mensurada em densÃmetro digital e convertida para Â°Brix, utilizando uma tabela que relaciona estas variÃ¡veis. A AT foi calculada conforme as Normas AnalÃticas do Instituto Adolfo Lutz. O ratio foi obtido pela razÃ£o entre Â°Brix e AT. Todos os sucos concentrados estavam em acordo com o Codex Alimentarius. Abstract: One of the factors limiting the duration of rat liver storage is the excessive accumulation of catabolites in the perfusion medium. In view of obtaining of an efficient depuration the authors have introduced a new circuit allowing for the concomittant perfusion of the liver and of the kidneys. The excretion effort of the kidneys is assessed by the estimation of urea nitrogen in the perfusion medium and in the urine collected over the entire duration of the experiment. The results have confirmed the good function of the system which provides possibilities for improved storage conditions of rat liver.

Abstract: The author's aim was to investigate conservation of rat liver by continuous perfusion of the isolated organ in conditions of normal temperature, as well as in hypothermia. A complex solution was used, lacking hemoglobin. Over a period of 5 hours of conservation the authors evaluated the perfusion rate, the concentration of lactate and pyruvate in the conservation medium, while on histological preparations was investigated the formation of fibrin deposits in the intra-hepatic vessels. The results confirmed the positive effects of hypothermia, that is not due exclusively to a limiting of the metabolic necessities of the organ but also to a reduction of the fibrinogenesis and a slowing down of the formation of intra-hepatic microaggregates.

Abstract: Accumulation of radioactive material in the isolated rat liver was noted when 125I-fibrinogen was added to the perfusing medium. Owing to an enhanced production of fibrinogen and to a diminished fibrinolysis, fibrin deposits were found to occur in the capillaries of fhe isolated liver. This would cause a decrease of hepatic flow and an impairment of the nutrition of this organ which leads to subsequent necrosis. Administration of streptase removed the fibrin deposits and restored the perfusive flow in the isolated liver.

Abstract: ABSTRACT The objective this paper was to evaluate the effect of two categories of beef finished in pasture with supplementation with two herbage allowance on performance, carcass and meat characteristics. Thirty-six Guzera cattle were used, 18 steers and 18 heifers with an initial age of 20 months. There was significant difference in daily weight gain for animal category and the herbage allowance, which were higher in males and animals submitted to high herbage allowance. Steers showed higher final weight, carcass weight and forequarter yield compared with the heifers, although the hindquarter yield was higher in the heifers, however the herbage allowance did not influence these characteristics. There were not statistical differences for carcass yield, ribeye area, backfat thickness and marbling score for the animal categories and herbage allowance. The meat chemical composition of the steers did not differ of the heifers, however, the animals submitted to high herbage allowance was increase in ether extract and pH, decrease in protein percentage. It was concluded that the animal category and the herbage allowance changed the animal performance, improving performance in males and cattle submitted to high herbage allowance.

Abstract: Abstract Background Subtle DNA methylation alterations mediated by carbon nanotubes (CNTs) exposure might contribute to pathogenesis and disease susceptibility. It is known that both multi-walled carbon nanotubes (MWCNTs) and single-walled carbon nanotubes (SWCNTs) interact with nucleus. Such, nuclear-CNT interaction may affect the DNA methylation effects. In order to understand the epigenetic toxicity, in particular DNA methylation alterations, of SWCNTs and short MWCNTs, we performed global/genome-wide, gene-specific DNA methylation and RNA-expression analyses after exposing human bronchial epithelial cells (16HBE14o- cell line). In addition, the presence of CNTs on/in the cell nucleus was evaluated in a label-free way using femtosecond pulsed laser microscopy. Results Generally, a higher number of SWCNTs, compared to MWCNTs, was deposited at both the cellular and nuclear level after exposure. Nonetheless, both CNT types were in physical contact with the nuclei. While particle type dependency was noticed for the identified genome-wide and gene-specific alterations, no global DNA methylation alteration on 5-methylcytosine (5-mC) sites was observed for both CNTs. After exposure to MWCNTs, 2398 genes were hypomethylated (at gene promoters), and after exposure to SWCNTs, 589 CpG sites (located on 501 genes) were either hypo- (N = 493 CpG sites) or hypermethylated (N = 96 CpG sites). Cells exposed to MWCNTs exhibited a better correlation between gene promoter methylation and gene expression alterations. Differentially methylated and expressed genes induced changes (MWCNTs > SWCNTs) at different cellular pathways, such as p53 signalling, DNA damage repair and cell cycle. On the other hand, SWCNT exposure showed hypermethylation on functionally important genes, such as SKI proto-oncogene (SKI), glutathione S-transferase pi 1 (GTSP1) and shroom family member 2 (SHROOM2) and neurofibromatosis type I (NF1), which the latter is both hypermethylated and downregulated. Conclusion After exposure to both types of CNTs, epigenetic alterations may contribute to toxic or repair response. Moreover, our results suggest that the observed differences in the epigenetic response depend on particle type and differential CNT-nucleus interactions.

Abstract: A checklist of the amphibians of Santa Teresa municipality, in southeastern Brazil is presented based on fieldwork, examination of specimens in collections, and a literature review. This new amphibian list of Santa Teresa includes 108 species, of which 106 (~98%) belong to Anura and two (~2%) to Gymnophiona. Hylidae was the most represented family with 47 species (43%). Compared to the previous amphibian lists for Santa Teresa, 14 species were added, 17 previously reported species were removed, and 13 species were re-identified based on recent taxonomic rearrangements. Of the 14 species added, 11 (79%) were first recorded during our fieldwork and specimen examination. It is also the first list of caecilians for Santa Teresa. This list suggests that Santa Teresa has 0.16 species per km 2 (i.e., 108 species/683 km 2 ), one of the highest densities of amphibian species in the world at a regional scale. This richness represents 78% of the 136 anurans from Espírito Santo state and 10% of the 1,080 amphibians from Brazil. We highlight the need for long-term monitoring to understand population trends and develop effective conservation plans to safeguard this remarkable amphibian richness.

45. Positive roadside edge effects on artificial nest survival in a lowland Atlantic Forest.

Abstract: Road construction is considered to be one of the primary causes of forest fragmentation, and little is known about how roads affect bird reproductive success. The objective of this study was to assess the survival rate of artificial nests along an edge associated with a highway and in the interior of a tabuleiro forest. The study was performed at the Sooretama Biological Reserve, on the margins of federal highway BR-101, between September and October 2015. A total of 168 artificial nests with a Common quail ( Coturnix coturnix ) egg in each nest were placed along six sampling transects, at distances of 2, 25, 50, 100, 200, 400, and 800 m from the highway toward the forest interior. We used logistic regression and estimated daily survival rate (DSR) using the "Nest Survival" function in the program MARK to estimate artificial nest survival and assessed the effect of the distance from the highway. The artificial nest survival rate was significantly higher on the highway margins than at other distances. The results show that artificial nests located up to 25 m from the highway have a greater success probability (over 95%) and a significant decrease in success probability more than 50 m from the highway. Although we cannot rule out other nonroad-specific edge effects on artificial nest predation, our results suggest that the impacts of the highway (e.g., noise, vibration, visual stimuli) cause predators to avoid the road's surroundings (up to 25 m into the forest) when selecting their feeding sites, which partially supports the predation release hypothesis., Competing Interests: None declared.

Abstract: Scientists have long been trying to understand why the Neotropical region holds the highest diversity of birds on Earth. Recently, there has been increased interest in morphological variation between and within species, and in how climate, topography, and anthropogenic pressures may explain and affect phenotypic variation. Because morphological data are not always available for many species at the local or regional scale, we are limited in our understanding of intra- and interspecies spatial morphological variation. Here, we present the ATLANTIC BIRD TRAITS, a data set that includes measurements of up to 44 morphological traits in 67,197 bird records from 2,790 populations distributed throughout the Atlantic forests of South America. This data set comprises information, compiled over two centuries (1820-2018), for 711 bird species, which represent 80% of all known bird diversity in the Atlantic Forest. Among the most commonly reported traits are sex (n = 65,717), age (n = 63,852), body mass (n = 58,768), flight molt presence (n = 44,941), molt presence (n = 44,847), body molt presence (n = 44,606), tail length (n = 43,005), reproductive stage (n = 42,588), bill length (n = 37,409), body length (n = 28,394), right wing length (n = 21,950), tarsus length (n = 20,342), and wing length (n = 18,071). The most frequently recorded species are Chiroxiphia caudata (n = 1,837), Turdus albicollis (n = 1,658), Trichothraupis melanops (n = 1,468), Turdus leucomelas (n = 1,436), and Basileuterus culicivorus (n = 1,384). The species recorded in the greatest number of sampling localities are Basileuterus culicivorus (n = 243), Trichothraupis melanops (n = 242), Chiroxiphia caudata (n = 210), Platyrinchus mystaceus (n = 208), and Turdus rufiventris (n = 191). ATLANTIC BIRD TRAITS (ABT) is the most comprehensive data set on measurements of bird morphological traits found in a biodiversity hotspot; it provides data for basic and applied research at multiple scales, from individual to community, and from the local to the macroecological perspectives. Abstract: South America holds 30% of the world's avifauna, with the Atlantic Forest representing one of the richest regions of the Neotropics. Here we have compiled a data set on Brazilian Atlantic Forest bird occurrence (150,423) and abundance samples (N = 832 bird species; 33,119 bird individuals) using multiple methods, including qualitative surveys, mist nets, point counts, and line transects). We used four main sources of data: museum collections, on-line databases, literature sources, and unpublished reports. The data set comprises 4,122 localities and data from 1815 to 2017. Most studies were conducted in the Florestas de Interior (1,510 localities) and Serra do Mar (1,280 localities) biogeographic sub-regions. Considering the three main quantitative methods (mist net, point count, and line transect), we compiled abundance data for 745 species in 576 communities. In the data set, the most frequent species were Basileuterus culicivorus, Cyclaris gujanensis, and Conophaga lineata. There were 71 singletons, such as Lipaugus conditus and Calyptura cristata. We suggest that this small number of records reinforces the critical situation of these taxa in the Atlantic Forest. The information provided in this data set can be used for macroecological studies and to foster conservation strategies in this biodiversity hotspot. Abstract: This research focused on the effects of inorganic water constituents on the efficiency of vacuum UV (VUV) for the degradation of micropollutants in surface water supplies. Atrazine was used as a model miropollutant, and bicarbonate, sulphate, and nitrate were used as the most common inorganic constituents in the water matrix. First, the absorbance of radiation at 254 and 185 nm was measured in the presence of different ions. At 254 nm, only nitrate showed a measurable absorption coefficient of [Formula: see text] = 3.51 M[Formula: see text] cm[Formula: see text], and all other ions showed a molar absorption coefficient below the detection limit. However, at 185 nm, all the ions showed high absorption coefficients, with nitrate giving the highest absorption coefficient of [Formula: see text] = 5568 M[Formula: see text] cm[Formula: see text]. Second, the hydroxyl radical (HO[Formula: see text]) scavenging effects of the same inorganic ions were evaluated; nitrate and bicarbonate showed a negative effect during the UV/H 2 O 2 and VUV advanced oxidation processes. Sulfate was photolyzed with 185 nm UV to form HO[Formula: see text], and for this reason, it assisted the degradation of the target micropollutant, as demonstrated by increases in the degradation rate constant. An additional component of this work involved developing a method for measuring the quantum yield of atrazine at 185 nm. This made it possible to distinguish the contribution of OH radical attach from that of direct photolysis towards the degradation of atrazine.

Abstract: Background: Optic nerve sheath meningiomas account for only 2% of orbital lesions and 42% of optic nerve tumors. Diagnosis remains difficult because histologic confirmation carries a high risk of visual loss. Therefore, a less invasive and specific diagnostic method for differentiating optic nerve sheath meningiomas from other optic nerve lesions is needed to overcome the limitations of computed tomography and magnetic resonance imaging, and make the best individualized treatment decision. This case is a good illustration of the clinical and imaging difficulties inherent in this rare tumor, which may be hard to differentiate from other causes., Case Presentation: A 51-year-old Caucasian woman developed a central scotoma, visual loss, and abnormal visual evoked potentials. The first magnetic resonance imaging scan classified the optic nerve damage as retrobulbar optic neuritis. After magnetic resonance imaging follow-up at 3 months, a negative lumbar puncture and biological workup, and clinical worsening, an optic nerve sheath meningioma was suspected. We confirmed this diagnosis with 111In-pentetreotide single-photon emission computed tomography, which is able to bind with very high affinity to somatostatin receptor subtype 2 expressed on meningiomas., Conclusions: In the diagnosis of optic nerve sheath meningiomas, [111In]-pentetreotide single-photon emission computed tomography-fused magnetic resonance imaging is a valuable additional tool, optimizing the diagnosis and obviating the need for a more invasive procedure.

50. High-accuracy measurement of the blackbody radiation frequency shift of the ground-state hyperfine transition in 133Cs.

Abstract: We report a high-accuracy direct measurement of the blackbody radiation shift of the 133Cs ground-state hyperfine transition. This frequency shift is one of the largest systematic frequency biases encountered in realizing the current definition of the International System of Units (SI) second. Uncertainty in the blackbody radiation frequency shift correction has led to its being the focus of intense theoretical effort by a variety of research groups. Our experimental measurement of the shift used three primary frequency standards operating at different temperatures. We achieved an uncertainty a factor of five smaller than the previous best direct measurement. These results tend to validate the claimed accuracy of the recently calculated values.
