1. Preexisting risk-avoidance and enhanced alcohol relief are driven by imbalance of the striatal dopamine receptors in mice

Abstract

Abstract Alcohol use disorder (AUD) is frequently comorbid with anxiety disorders, yet whether alcohol abuse precedes or follows the expression of anxiety remains unclear. Rodents offer control over the first drink, an advantage when testing the causal link between anxiety and AUD. Here, we utilized a risk-avoidance task to determine anxiety-like behaviors before and after alcohol exposure. We found that alcohol’s anxiolytic efficacy varied among inbred mice and mice with high risk-avoidance showed heightened alcohol relief. While dopamine D1 receptors in the striatum are required for alcohol’s relief, their levels alone were not correlated with relief. Rather, the ratio between striatal D1 and D2 receptors was a determinant factor for risk-avoidance and alcohol relief. We show that increasing striatal D1 to D2 receptor ratio was sufficient to promote risk-avoidance and enhance alcohol relief, even at initial exposure. Mice with high D1 to D2 receptor ratio were more prone to continue drinking despite adverse effects, a hallmark of AUD. These findings suggest that an anxiety phenotype may be a predisposing factor for AUD.

2. Performance in even a simple perceptual task depends on mouse secondary visual areas

Abstract

Primary visual cortex (V1) in the mouse projects to numerous brain areas, including several secondary visual areas, frontal cortex, and basal ganglia. While it has been demonstrated that optogenetic silencing of V1 strongly impairs visually guided behavior, it is not known which downstream areas are required for visual behaviors. Here we trained mice to perform a contrast-increment change detection task, for which substantial stimulus information is present in V1. Optogenetic silencing of visual responses in secondary visual areas revealed that their activity is required for even this simple visual task. In vivo electrophysiology showed that, although inhibiting secondary visual areas could produce some feedback effects in V1, the principal effect was profound suppression at the location of the optogenetic light. The results show that pathways through secondary visual areas are necessary for even simple visual behaviors.

Abstract

Many cortical network models use recurrent coupling strong enough to require inhibition for stabilization. Yet it has been experimentally unclear whether inhibition-stabilized network (ISN) models describe cortical function well across areas and states. Here, we test several ISN predictions, including the counterintuitive (paradoxical) suppression of inhibitory firing in response to optogenetic inhibitory stimulation. We find clear evidence for ISN operation in mouse visual, somatosensory, and motor cortex. Simple two-population ISN models describe the data well and let us quantify coupling strength. Although some models predict a non-ISN to ISN transition with increasingly strong sensory stimuli, we find ISN effects without sensory stimulation and even during light anesthesia. Additionally, average paradoxical effects result only with transgenic, not viral, opsin expression in parvalbumin (PV)-positive neurons; theory and expression data show this is consistent with ISN operation. Taken together, these results show strong coupling and inhibition stabilization are common features of the cortex.

4. Excitation creates a distributed pattern of cortical suppression due to varied recurrent input

Abstract

SummarySensory cortex transforms its inputs, generating neural responses that represent features of the sensory world. Dense local, recurrent connections are a major feature of cortical circuits, yet how they affect these transformations is unclear, with some studies reporting recurrent circuits amplify certain excitatory inputs, and other studies showing instead local suppression. Here we find that input to mouse V1 excitatory neurons generates salt-and-pepper patterns of excitation and suppression. We show that neurons’ responses are not strongly predicted by their optogenetic input alone, suggesting that recurrent inputs nonlinearly transform the population response. A balanced-state network model explains the observed dynamics, suppressed responses, and long tail of excited responses, but only with substantial variance in cell-to-cell connectivity. In sum, we find recurrent cortical connectivity has high variability and high average strength, which can decouple individual cells’ responses from the direct input each cell receives, leading to substantial suppression generated by excitatory input.

Abstract

While Reynolds-averaged simulations have found success in the evaluation of many canonical shear flows and moderately separated flows, their application to highly separated flows have shown notable deficiencies. This study aimed to investigate these deficiencies in the eddy-viscosity formulation of four commonly used turbulence models under separated flow in an attempt to aid in the improved formulation of such models. Analyses are performed on the flow field around a wall-mounted cube (WMC) at a Reynolds number of 40,000 based on the cube height, h, and freestream velocity, U0. While a common occurrence in industrial applications, this type of flow constitutes a complex structure exhibiting a large separated wake region, high anisotropy, and multiple vortex structures. As well, interactions between vortices developed off of different faces of the cube significantly alter the overall flow characteristics, posing a significant challenge for the commonly used industrial turbulence models. Comparison of mean flow characteristics show remarkable agreement between experimental values and turbulence models which are capable of predicting transitional flow. Evaluation of turbulence parameters show the general underestimation of Reynolds stress for transitional models, while fully turbulent models show this value to be overestimated, resulting in completely disparate representations of mean flow structures between the two classes of models (transitional and fully turbulent).

Abstract

International audience; EURECA (European Underground Rare Event Calorimeter Array) is a new project, searching for dark matter, with largely the present groups of the CRESST and EDELWEISS experiments and already a few new groups. The aim is to explore scalar cross sections in the 10−9–10−10 picobarn region with a target mass of up to one tonne. A major advantage of EURECA is our planned use of more that just one target material (multi target experiment for WIMP identification). In preparation for this large-scale experiment, R&D for EURECA is provided through the current phases of CRESST and EDELWEISS.

Abstract

Transplant vasculopathy has not been systematically investigated in composite tissue allotransplantation (CTA). The impact of multiple acute rejections (ARs) on long-term graft outcomes in reconstructive transplantation remains unknown. This study in a rat hind-limb allotransplantation model systematically analyzes vasculopathy and tissue-specific pathological changes secondary to multiple AR episodes. LEW rats were transplanted with BN rat hind limbs and treated as follows: Group 1 (Iso): isografts. Group 2 (CsA): Cyclosporine (CsA) qd; Group 3 (mult AR): CsA and dexamethasone only when AR was observed. No AR was observed in Groups 1 and 2. Multiple AR were observed in Group 3, and each episode was completely reversed (clinically) with pulsed CsA + dexamethasone treatment. Group 3 animals demonstrated significant vascular lesions along with skin and muscle atrophy, upregulation of profibrotic gene expression and fibrosis when compared to Groups 1 and 2. In addition, allograft bone was sclerotic, weak and prone to malunion and nonunion. Interestingly, vasculopathy was a late finding, whereas muscle atrophy with macrophage infiltration was seen early, after only a few AR episodes. Taken together, multiple AR episodes lead to vasculopathy and tissue-specific pathology in CTA. This is the first evidence of 'composite tissue vasculopathy and degeneration (CTVD)' in CTA.

Abstract

The heat quenching factor Q' (the ratio of the heat signals produced by nuclear and electron recoils of equal energy) of the heat-and-ionization germanium bolometers used by the EDELWEISS collaboration has been measured. It is explained how this factor affects the energy scale and the effective quenching factor observed in calibrations with neutron sources. This effective quenching effect is found to be equal to Q/Q', where Q is the quenching factor of the ionization yield. To measure Q', a precise EDELWEISS measurement of Q/Q' is combined with values of Q obtained from a review of all available measurements of this quantity in tagged neutron beam experiments. The systematic uncertainties associated with this method to evaluate Q' are discussed in detail. For recoil energies between 20 and 100 keV, the resulting heat quenching factor is Q' = 0.91+-0.03+-0.04, where the two errors are the contributions from the Q and Q/Q' measurements, respectively. The present compilation of Q values and evaluation of Q' represent one of the most precise determinations of the absolute energy scale for any detector used in direct searches for dark matter., Comment: 28 pages, 7 figures. Abstract

Close proximity hyperspectral and multispectral imaging of crops and soils offers significant potential to optimize sustainable intensification of arable produce and seeds breeding, through the real-time precision management of plant pathogens, viruses and pests and the non-destructive high throughput screening for beneficial crop traits. These opportunities have been recently reported and are the subject of ongoing R&D within industry and academia. The broad uptake of the technology by large commercial end-users, through integration with in-field and glasshouse machinery, is limited by cost and equipment reliability. It is further restricted by spectral and spatial resolution, power budget and size, when extending its applicability to consumer markets and small-holder farmers. This study verifies, for the first time, that multispectral sensor systems architectures, exploiting proprietary narrowband LEDs and silicon C-MOS imaging detectors, are capable of substituting for conventional and more expensive line-scanning hyperspectral imaging systems when operated in close proximity (c. 1-2m) of a crop canopy. This was achieved by comparing the data from a prototype version of the new LED-sensor system versus a reference laboratory hyperspectral imaging unit, which was previously developed for crop phenotyping, and the early detection of two fungal pathogen borne diseases in whole barley and sugar beet plants. The choice of crops and diseases replicates earlier studies, with the reference hyperspectral unit, and serves to demonstrate the generic applicability of the new LED-sensor system to cereal and tuber classes of crops. The results indicate that the new approach can deliver data of comparable quality to that of the reference system, for in-field duties, and offers the opportunity for higher sensitivity and spatial resolution. Future potential to apply the new multispectral, LED-based system within commercial products is then discussed.

Abstract

EDELWEISS is a direct search for WIMPs using cryogenic germanium ionizationphonon detectors and located in the Modane underground laboratory. We summarize the final results of the EDELWEISS I experiment obtained with up to nearly a kilogram of detectors. The increased exposure confirms previous results. We also report on the preparations for EDELWEISS II. Preliminary results are expected in 2006; the experiment could ultimately deploy up to 40 kg of detectors. Goals are to gain two orders of magnitude in sensitivity and to serve as a testbed for an even larger, tonne-scale, experiment.

Abstract

In its first stage, the EDELWEISS-II dark matter experiment will use 21×320 g ionization-heat bolometers with NTD thermal sensors. To improve the present sensitivity of EDELWEISS to WIMP interactions by a further factor 100, ultra-low radioactivity detector holders exclusively made of copper and Teflon have been designed. The new design is using the relative expansion coefficients of copper, Teflon and germanium to hopefully ensure a dissipation-free detector holding. In order to validate this new holder, we need to compare the vibration behavior of the old holder and the new one. The amplitude of the bolometer vibrations is measured using the variation of capacitance between electrodes sputtered on the Ge crystal and reference electrodes fixed above the detector holders. We present noise measurements and compare them to the equivalent measurements using the detector holders of the Edelweiss-I experiment, which gave very satisfying results.

Abstract

An experiment to look for light WIMPs with sapphire bolometers is to be installed in the Canfranc Underground Laboratory (at 2450 m.w.e) in the Spanish Pyrenees. In the first stage two small 25g bolometers with NTD-Ge sensors, operating at 25mK, with an energy threshold of 300eV and FWHM energy resolution of 120eV at 1.5keV will perform a background test experiment. The features of the bolometers, the radioactivity measurements of the components and the set-up and status of the experiment are reported.

Abstract

The status of the EDELWEISS experiment (underground dark matter search with heat-ionisation bolometers) is reviewed. Auspicious results achieved with a prototype 70 g Ge heat-ionisation detector under a 2 V reverse bias tension are discussed. Based on gamma and neutron calibrations, a best-case rejection factor, over the 15-45 keV range, of 99.7 % for gammas, with an acceptance of 94 % for neutrons, is presented first. Some operational results of physical interest obtained under poor low radioactivity conditions follow. They include a raw event rate of around 30 events/day/kg/keV over the same energy range, and, after rejection of part of the background, lead to a conservative upper limit on the signal of approximately 1.6 events/day/kg/keV at a 90 % confidence level. Abstract

Bolometers at low temperature are being developed as particle spectrometers and potential detectors of dark matter particles through the measurement of recoil energies. A successful observation of the recoil spectrum from a 210 Po electrodeposited source was made for the first time with the use of a 2 mm 3 composite-composite diamond bolometer operated in a 300 mK cryostat. The measured recoil ratio is 0.98±0.06 recoil/alpha above 50 keV, with the well defined peak in the spectrum at 101±2.5 keV.

Abstract

The status of the EDELWEISS Dark Matter Search Experiment is presented. In this experiment, nuclear recoils induced by elastic collisions with WIMPs from the galactic halo are identified in low-temperature Ge detectors where the ratio of the heat and ionization signals provides an event-by-event discrimination of nuclear recoils from the dominant background from γ-ray interactions. Results obtained in the low-background environment of the Modane Underground Laboratory (LSM) with 320 g detectors are presented: in the first 2000 run, no nuclear events are observed in the fiducial volume in the 30–200 keV energy range in an effective exposure of 4.5 kg.day leading to the exclusion of the upper part of the DAMA region and of its central value. The present 2002 setup with three bolometers is described and improved calibration performances are presented. Prospects for the second stage of the experiment, EDELWEISS-II, using an innovative reversed cryostat allowing data taking with 120 detectors, and recent results of the R and D, are also briefly described.

Abstract

Background Despite the widely accepted implication of antidonor antibodies and complement in solid organ transplantation, their role in reconstructive allotransplantation is not clear. The aim of this study was to analyze the humoral immune response using a rat orthotopic limb transplantation model. Methods We used the Brown Norway to Lewis rat orthotopic hind-limb transplant model: Group 1, isografts; group 2, allografts with daily continuous cyclosporine treatment to prevent acute rejection; and group 3, allografts undergoing multiple episodes of acute rejection. Samples were taken at 30, 60, and 90 days. Serum was analyzed by FACS for antidonor antibodies. Tissue deposition of antibodies and complement was investigated by immunofluorescence. Results By day 90, animals in group 3 had undergone 19 (±3.2) acute rejection episodes. There was no difference in the occurrence of serum antidonor antibodies between the three groups at any time point. However, at 90 days, anti–third-party antibodies were significantly greater among group 3. There was no difference in antibody or complement deposition in muscles between the 3 groups. Conclusion Despite the increased antibody against a third party after multiple rejection episodes in this animal model, there was no clear evidence of an antibody-mediated alloresponse in limb transplantation.

Abstract

International audience; This paper presents our interpretation and understanding of the different backgrounds in the EDELWEISS-I data sets. We analyze in detail the several populations observed, which include gammas, alphas, neutrons, thermal sensor events and surface events, and try to combine all data sets to provide a coherent picture of the nature and localisation of the background sources. In light of this interpretation, we draw conclusions regarding the background suppression scheme for the EDELWEISS-II phase.

Abstract

EURECA (European Underground Rare Event Calorimeter Array) is a new project, searching for dark matter. The collaboration is comprised largely of the present groups of the CRESST and EDELWEISS experiments and several new groups. The aim is to explore scalar cross sections in the - 10 -9-10-10picobarn region with a target mass of up to one tonne. A major advantage of EURECA is the planned use of more that just one target material (multi target experiment for WIMP identification). In preparation for this largescale experiment, RandD for EURECA is provided through the current phases of CRESST and EDELWEISS.

Abstract

The ZEPLIN collaboration has recently published its first result presenting a maximum sensitivity of $1.1 \times 10^{-6}$ picobarn for a WIMP mass of $\approx$ 60 GeV. The analysis is based on a discrimination method using the different time distribution of scintillation light generated in electron recoil and nuclear recoil interactions. We show that the methodology followed both for the calibration of the ZEPLIN-I detector response and for the estimation of the discrimination power is not reliable enough to claim any background discrimination at the present stage. The ZEPLIN-I sensitivity appears then to be in the order of 10$^{-3}$ picobarn, three orders of magnitude above the claimed 1.1 10$^{-6}$ picobarn., 8 pages, 4 figures, minor corrections, two references updated, final version accepted in Physics Letters B

Abstract

The goal of this study was the reexamination, description, and analysis of postoperative results after carpal tunnel release reoperation.Thirty-eight patients were examined retrospectively 2 years after reoperation of carpal tunnel release. All of them had had complaints after the first operation. On the basis of DASH and Amadio questionnaires after the reoperation, the patients were divided into two groups: good results and inadequate results.The reasons for reoperation included incomplete division of the retinaculum, early fibrosis, reoccurrence, late fibrosis, intraoperative complication, and one unclear situation. A quarter of the patients were free of complaints; three quarters had symptoms in which sensation improved but strength and function did not.The clinical findings showed satisfying results for the patients. Reoperation is indicated after early postoperative deterioriation or if there is no improvement.

We provide an update on the status of the EDELWEISS dark matter search. The first phase has confirmed its previously published sensitivity thanks to more data. It has also served as a test-bed for the second phase, which aims to gain two orders of magnitude in sensitivity by means of a larger, improved setup. One of the main issues which must be addressed for EDELWEISS II is the neutron background.

Abstract

The final results of the EDELWEISS-I dark matter search using cryogenic heat-and-ionization Ge detectors are presented. The final data sample corresponds to an increase by a factor five in exposure relative to the previously published results. A recoil energy threshold of 13 keV or better was achieved with three 320g detectors working simultaneously over four months of stable operation. Limits on the spin-independent cross-section for the scattering of a WIMP on a nucleon are derived from an accumulated fiducial exposure of 62 kg.d., 42 pages, 16 figures, submitted to Physical Review D, 1 figure and 2 references added, some little changes in the text

Abstract

International audience; The EDELWEISS collaboration has performed a direct search for WIMPs using a 320g ionization-heat Ge-cryogenic detector operated in a low-background environment in the Laboratoire Souterrain de Modane. No nuclear recoils events are observed in the fiducial volume in the 30-200 keV energy range during an effective exposure of 4.53 kg.days. A limit on the cross-section of the WIMP-proton spin-independent interaction as a function of the WIMP mass is deduced. The central value of the signal reported by the DAMA experiment is excluded at 90% CL. .

Abstract

Anomalous events, with scintillation decay times shorter than nuclear recoils, have been observed by the UKDMC and Saclay NaI experiments. By using the event categories observed in the EDELWEISS experiment, we propose to interpret the anomalous NaI events as surface nuclear recoils accompanied with a small energy loss of an escaping alpha particle. The discrimination performances of the NaI experiments which have not taken into account these events in their analysis, notably for axial WIMP interactions, must then be reevaluated.

Abstract

Four categories of events have been identified in the EDELWEISS-I dark matter experiment using germanium cryogenic detectors measuring simultaneously charge and heat signals. These categories of events are interpreted as electron and nuclear interactions occurring in the volume of the detector, and electron and nuclear interactions occurring close to the surface of the detectors(10-20 mu-m of the surface). We discuss the hypothesis that low energy surface nuclear recoils,which seem to have been unnoticed by previous WIMP searches, may provide an interpretation of the anomalous events recorded by the UKDMC and Saclay NaI experiments. The present analysis points to the necessity of taking into account surface nuclear and electron recoil interactions for a reliable estimate of background rejection factors., Comment: 11 pages, submitted to Phys. A dark matter detection experiment using a low temperature 24 g sapphire bolometer is presented. The low radioactive background cryogenic facility, installed in a deep underground site, is described, as well as the low-noise read-out electronics and the data analysis. From the energy spectrum, measured down to 4 keV, exclusion plots are derived for WIMPs having coherent vector coupling or axial coupling to ordinary matter.

The principles and the realisation of an experiment devoted to oscillator strength measurements in the vacuum-ultraviolet by an emission method are briefly presented. The oscillator strengths of seven strong NI lines, eleven weak NI lines, twenty-three strong CI lines and twenty weak CI lines have been measured in the 1200- 2000Å spectral range. The absolute scale set up from the measurements is compared with recent theoretical results.

Abstract

In order to investigate some properties of a medium with a low number of particles in a Debye sphere, the electrical conductivity of high-pressure ionized Argon has been measured. A special emphasis is laid on the so-called "non-ideality" effects in the collective behaviour of charged particles. A kind of threshold for these effects is observed at nD ≦ 4. The observed asymptotic behaviour of the screening parameter indicates that heavy charged particles have the same influence on the screening mechanism as electrons. The experimental values of the electrical conductivity are in excellent agreement with similar recent measurements.

The continuum emission coefficient of a high-pressure argon plasma is measured in a wall-stabilized arc in the ranges 2600-7000 AA, 1-30 atm and 11200-13200K. The temperature and the electron density are determined independently using standard spectroscopic methods. Results of continuum measurements are compared with resultant calculated values of free-bound and both electron-ion and electron-neutral free-free radiation. The discrepancies existing at high pressure cannot be entirely explained by a shift of recombination thresholds.

Describes a number of analytical and numerical calculations related to the model microfield method applied to Stark broadening (in particular, treatment of strong collisions and angular averages). This method is used for several types of profile: He I overlapping lines, Li I and Mg I resonance lines and Li I and O I non-resonance lines. For each of these, various approximations are discussed and the profiles are compared to those obtained from other theories and to experimental results. In particular, it is shown that the forbidden components cannot be obtained correctly using the usual approximation in which the electronic broadening is calculated independently of the ionic microfield.

Comparisons of experimental and theoretical Stark broadening parameters of the 21P-31D, 23P-33D and 23S-33P helium lines at 6678, 5876 and 3889 AA are carried out in the high-density plasma produced by focusing a powerful pulsed CO2 laser in helium at 1500 Torr. At low electron densities (ne17 cm-3) the measured linewidths and lineshifts are in good agreement with published results of current theories. Model microfield method calculations were found to give accurate prediction of helium line-broadening parameters over an electron density range from 1017 to 2*1018 cm-3.

Abstract

The application of a water balance model in finding “solutions” to the supply/demand problem was demonstrated using the South Platte River basin as a case study. Solutions were ascertained by hand, using both “average” and “stress” supply/demand conditions, and were developed for 1980, 2000, and 2020; nonquantifiable boundary conditions were incorporated by judgement. The solution obtained for a particular set of conditions is not unique and has strong normative characteristics; thus it must be judged by various interest groups having different ethical positions. The water balance model has a tabular display format and so the “model” is merely a simple table, i.e., a “water balance table.” In this work the water balance table was displayed on an eight-foot by eight-foot color-coded magnetic board. The board provides a means to both find and display the needed supply/demand “solution.” The tabular display facilitates understanding of the systemwide solution and the formulation of value judgments. Based upon these value judgments and an initial “straw man” solution, successive negotiated solutions can be found which can minimize “conflict.”

Abstract

The xi ( lambda ,T) function of xenon was determined by measuring the continuous radiation of a wall-stabilized steady-state high-pressure arc plasma from an end-on direction. In the investigated spectral range between lambda =260 and 800 nm, the measured xi ( lambda ,T) function deviates from previously reported calculations and measurements. In particular, the characteristic wavelength dependence of the xi ( lambda ,T) function predicted by Schluter's theory (1968) could not be confirmed. For wavelengths of lambda 500 nm it correlates with Shcherbakov's calculations (1970) (with a maximum deviation of +or-20%).

Abstract

The principles and the realization of an experiment devoted to oscillator strength measurements in the vacuum-ultraviolet by the emission method are briefly presented. The results obtained for the strong multiplets of neutral nitrogen and carbon in the 1200-2000 Å range yield an absolute scale of oscillator strengths in good agreement with the most recent calculations.
