Tidal stream currents change in magnitude and direction during flood and ebb tides. Setting the most appropriate yaw angles for a tidal turbine is not only important to account for the performance of a single turbine, but can also be significant for the interactions between the turbines within an array. In this paper, a partial differentiation equation (PDE) constrained optimisation approach is established based on the Thetis coastal ocean modelling framework. The PDE constraint takes the form here of the two-dimensional, depth-averaged shallow water equations which are used to simulate tidal elevations and currents in the presence of tidal stream turbine arrays. The Sequential Least Squares Programming (SLSQP) algorithm is applied with a gradient obtained via the adjoint method in order to perform array design optimisation. An idealised rectangular channel test case is studied to demonstrate this optimisation framework. Located in the centre of the computational domain, arrays comprised of 12 turbines are tested in aligned and staggered layouts. The setups are initially optimised based on their yaw angles alone. In turn, turbine coordinates and yaw angles are also optimized simultaneously. Results indicate that for an aligned turbine array case under steady state conditions, the energy output can be increased by approximately 80\% when considering yaw angle optimisation alone. For the staggered turbine array, the increase is approximately 30\%. The yaw optimised staggered array is able to outperform the yaw optimised aligned array by approximately 8\%. If both layout and the yaw angles of the turbines are considered within the optimisation then the increase is more significant compared with optimising yaw angle alone.

Anticipatory stress can prospectively and negatively influence diverse outcomes, including cognitive performance and emotional well-being. It has been suggested that perseverative cognitions (e.g., worry, rumination) during the anticipation period constitute a key mechanism driving these effects. The present study investigated the temporal dynamics among stressor anticipation, perseverative cognitions, and affective well-being. To accurately test the suggested mechanism, we focused on how these dynamics unfold within individuals over time. To that end, we analyzed data from an ecological momentary assessment study in an ethnically diverse sample (N = 243, 25-65 year olds, 68.7% Hispanic or non-Hispanic Black; 14 days, 5 measurement occasions per day) using dynamic structural equation modeling. Anticipating an upcoming stressor was linked to higher levels of perseverative cognitions approximately 3 hours later. At times when individuals reported higher levels of recent perseverative cognitions than typical for them, they also reported higher levels of negative affect and lower levels of positive affect. Mediational modeling indicated that perseverative cognitions accounted for the persistent effects of previous stressor anticipation on negative as well as positive affect several hours later. These findings suggest that perseverative cognitions may play an important role in explaining the detrimental effects of anticipatory stress on subsequent emotional well-being. Breathing exercises have been proposed as an effective intervention to improve subjective well-being and manage anxiety symptoms. As they are comparatively easy to learn and to implement, breathing exercises may be particularly beneficial for children. Although breathing exercises are ultimately supposed to provide salutary effects in individuals' everyday lives, immediate effects of breathing exercises in naturalistic contexts have received limited empirical attention. The purpose of this study was to examine immediate effects of slow-paced diaphragmatic breathing on negative affect as well as on relaxation in an ecologically valid setting. To that end, we conducted a micro-randomized trial in children's daily life.On each of 15 days, children (The breathing exercise had no immediate effects on negative affect or relaxation compared to both control conditions. However, in situations when children reported higher levels of worries than usual, relaxation was higher when children performed the breathing exercise compared to the passive control condition. Compared to the active control condition, the breathing exercise did not result in higher levels of relaxation in situations when children worried more than normally.Findings highlight that context-specific factors can modulate the effectiveness of breathing exercises and should be taken into account to tailor interventions to individuals' needs.

The uterus and placenta transport water during pregnancy recognition signaling, conceptus implantation, and placental development/placentation. This is likely influenced by aquaporins (AQPs) in the reproductive tract. This study determined mRNA and cell-type specific expression of AQP 1, 5, 8, and 9 proteins in the porcine uterus and placenta.Porcine uteri and Chorioallantois were subjected to real-time PCR and immunofluorescence microscopy.AQP1 mRNA was maximal by Day 25 in endometrium and remained stable thereafter. AQP1 mRNA did not change in chorioallantois. AQP1 protein localized to erythrocytes and endothelium of the endometrium and allantois, and to smooth muscle of the myometrium. AQP5 protein localized to apical and lateral surfaces of the chorionic epithelia of areolae, but mRNA did not change in chorioallantois. AQP8 mRNA was high in the endometrium from Days 15 through 60 of gestation, and protein localized to multiple cell types within the endometrium and chorioallantois. AQP9 mRNA was highest in the endometrium on Days 10, 12 and 25, but did not change in the chorioallantois. AQP9 protein localized to the apical surface of endometrial luminal epithelial cells during early pregnancy, with a shift towards the basal surface later. AQP9 protein was observed in the allantoic epithelium.Results reveal pigs can potentially use AQP1, AQP5, AQP8, and AQP9 to transport water from the endometrial bloodstream to the allantoic bloodstream or allantoic fluid. The reverse is also possible and may explain the mechanism for changing volumes of allantoic fluid and hydration of allantoic connective tissues during pregnancy.

Numerical studies of ice flow have consistently identified the grounding zone of outlet glaciers and ice streams (the region where ice starts to float) as crucial for predicting the rate of grounded ice loss to the ocean. Owing to the extreme environments and difficulty of access to ocean cavities beneath ice shelves, field observations are rare. Estimates of melt rates derived from satellites are also difficult to make near grounding zones with confidence. Therefore, numerical ocean models are important tools to investigate these critical and remote regions. The relative inflexibility of structured grid models means, however, that they can struggle to resolve these processes in irregular cavity geometries near grounding zones. To help solve this issue, we present a new nonhydrostatic unstructured mesh model for flow under ice shelves built using the Firedrake finite element framework. We demonstrate our ability to simulate full ice shelf cavity domains using the community standard ISOMIP+ Ocean0 test case and compare our results against those obtained with the popular MITgcm model. Good agreement is found between the two models, despite their use of different discretisation schemes and the sensitivity of the melt rate parameterisation to grid resolution. Verification tests based on the Method of Manufactured Solutions (MMS) show that the new model discretisation is sound and second-order accurate. A main driver behind using Firedrake is the availability of an automatically generated adjoint model. Our first adjoint calculations, of sensitivities of melt rate with respect to different inputs in an idealised grounding zone domain, are promising and point to the ability to address a number of important questions on ocean influence on ice shelf vulnerability in the future.

Background: The COVID-19 pandemic reduced the number of skin cancer diagnoses, potentially causing a progression to unfavourable tumour stages. Objectives: To identify the impact of delayed diagnostics on primary invasive melanoma and cutaneous squamous cell carcinoma (cSCC) by comparing tumour (pT) stage, Breslow thickness and invasion depth from before to after the first and second lockdown periods. Methods: In this population-based cohort study, histopathology reports registered between 1 January 2018 and 22 July 2021 were obtained from the nationwide histopathology registry in the Netherlands. The Breslow thickness of melanomas, invasion depth of cSCCs, and pT stage for both tumour types were compared across five time periods: (i) pre-COVID, (ii) first lockdown, (iii) between first and second lockdowns, (iv) second lockdown and (v) after second lockdown. Breslow thickness was compared using an independent t-test. pT-stage groups were compared using a χ2-test. Outcomes were corrected for multiple testing using the false discovery rate. Results: In total, 20 434 primary invasive melanomas and 68 832 cSCCs were included in this study. The mean primary melanoma Breslow thickness of the prepandemic era (period i) and the following time periods (ii–v) showed no significant difference. A small shift was found towards unfavourable pT stages during the first lockdown compared with the pre-COVID period: pT1 52·3% vs. 58·6%, pT2 18·9% vs. 17·8%, pT3 13·2% vs. 11·0%, pT4 9·1% vs. 7·3% (P = 0·001). No relevant changes were seen in subsequent periods. No significant change in pT stage distribution was observed between the pre-COVID (i) and COVID-affected periods (ii–v) for cSCCs. Conclusions: To date, the diagnostic delay caused by COVID-19 has not resulted in relatively more unfavourable primary tumour characteristics of melanoma or cSCC. Follow-up studies in the coming years are needed to identify a potential impact on staging distribution and survival in the long term.

Plastid transformation technology has been widely used to express traits of potential commercial importance, though the technology has been limited to traits that function while sequestered in the organelle. Prior research indicates that plastid contents can escape from the organelle, suggesting a possible mechanism for engineering plastid transgenes to function in other cellular locations. To test this hypothesis, we created tobacco plastid transformants that express a fragment of the nuclear-encodedPhytoene desaturase(PDS) gene capable of catalyzing post-transcriptional gene silencing if RNA escape to the cytoplasm occurs. We found multiple lines of direct evidence that plastid-encodedPDStransgenes affect nuclearPDSgene silencing: knockdown of the nuclear-encodedPDSmRNA and/or its apparent translational inhibition, biogenesis of 21-nucleotide (nt) phased small interfering RNAs (phasiRNAs), and pigment deficient plants. Furthermore, plastid-expressed double-stranded RNA (dsRNA) with no cognate nuclear-encoded pairing partner also produced abundant 21-nt phasiRNAs in the cytoplasm, demonstrating that a nuclear-encoded template is not required for siRNA biogenesis. Our results indicate that RNA escape from plastids to the cytoplasm occurs broadly, with functional consequences that include entry into the gene silencing pathway. Furthermore, we uncover a method to produce plastid-encoded traits with functions outside of the organelle and open new fields of study in plastid development, compartmentalization and small RNA biogenesis.

Dietary supplementation with 0.4 or 0.8% l-arginine (Arg) to gilts between days 14 and 25 of gestation enhances embryonic survival and vascular development in placentae; however, the underlying mechanisms are largely unknown. This study tested the hypothesis that Arg supplementation stimulated placental expression of mRNAs and proteins that enhance angiogenesis, including endothelial nitric oxide synthase (eNOS), vascular endothelial growth factor (VEGF), placental growth factor (PGF), GTP cyclohydrolase-I (GTP-CH1), ornithine decarboxylase (ODC1), and vascular endothelial growth factor receptors 1 and 2 (VEGFR1 and VEGFR2). Beginning on the day of breeding, gilts were fed daily 2 kg of a corn–soybean meal-based diet supplemented with 0.0 (control), 0.4, or 0.8% Arg. To slow down the spread of the COVID-19 virus, schools around the world were closed in early 2020, transferring children's scholastic activities to the homes and imposing a massive burden on parents and school-age children. Using data of a 21-day diary study conducted between March and April 2020 in Germany, this work examined whether (a) distance learning and (b) parents' involvement therein were associated with negative parent-child interactions and affective well-being of parents and children, over and above the effect of daily stressors. Participants were 562 parents (489 mothers, Mage = 42.79, SDage = 6.12, range = 25-63) most of whom were married (n = 382, 68.0%). They responded to the daily items with respect to the youngest child living in their household (Mage = 9.74, SDage = 2.81, range = 6-19). On days when children were working on school tasks, parents reported more negative parent-child interactions as well as lower parental and child positive affect and higher child negative affect, but not higher parental negative affect. Moreover, days when parents were more heavily involved in learning (i.e., when children worked less independently) were days with more negative parent-child interactions, lower parental and child positive affect, and higher parental and child negative affect. Negative parent-child interactions were linked to lower affective well-being of parents and children, and partially accounted for the relation among daily stressors and affective well-being. Pathogen-pathogen interactions represent a critical but little-understood feature of infectious disease dynamics. In particular, experimental evidence suggests that influenza viruses and respiratory syncytial viruses (RSV) compete with each other, such that infection with one confers temporary protection against the other. However, such interactions are challenging to study using common epidemiologic methods. Here, we use a mathematical modeling approach, in conjunction with detailed surveillance data from Hong Kong, to infer the strength and duration of the interaction between influenza and RSV. We further utilize our model to evaluate the potential of live attenuated influenza vaccines (LAIV) as a novel strategy to control RSV epidemics. We find evidence of strong, bidirectional interference, such that infection with either virus yields almost complete protection against infection with the other for two to four months. Furthermore, we demonstrate that the likely impact of widespread LAIV use depends on underlying viral circulation patterns. More broadly, we highlight the utility of mathematical models as a tool to characterize interactions and predict the indirect effects of vaccines.

To develop a high-fidelity mathematical model intended to replicate the cardiovascular (CV) responses of a critically ill patient to vasoplegic shock-induced hypotension and vasopressor therapy.The mathematical model consists of a lumped-parameter CV physiology model with baroreflex modulation feedback and a phenomenological dynamic dose-response model of a vasopressor. The adequacy of the proposed mathematical model was investigated using an experimental dataset acquired from 10 pigs receiving phenylephrine (PHP) therapy after vasoplegic shock induced via sodium nitroprusside (SNP).Upon calibration, the mathematical model could (i) faithfully replicate the effects of PHP on dynamic changes in blood pressure (BP), cardiac output (CO), and systemic vascular resistance (SVR) (root-mean-squared errors between measured and calibrated mathematical responses: mean arterial BP 2.5+/-1.0 mmHg, CO 0.2+/-0.1 lpm, SVR 2.4+/-1.5 mmHg/lpm; r value: mean arterial BP 0.96+/-0.01, CO 0.65+/-0.45, TPR 0.92+/-0.10) and (ii) predict physiologically plausible behaviors of unmeasured internal CV variables as well as secondary baroreflex modulation effects.This mathematical model is perhaps the first of its kind that can comprehensively replicate both primary (i.e., direct) and secondary (i.e., baroreflex modulation) effects of a vasopressor drug on an array of CV variables, rendering it ideally suited to pre-clinical virtual evaluation of the safety and efficacy of closed-loop control algorithms for autonomous vasopressor administration once it is extensively validated.This mathematical model architecture incorporating both direct and baroreflex modulation effects may generalize to serve as part of an effective platform for high-fidelity in silico simulation of CV responses to vasopressors during vasoplegic shock.

Distinctive large-scale structures have been identified in the spatial distribution of optical galaxies up to redshift z ~ 1. In the more distant universe, the relationship between the dust-obscured population of star-forming galaxies observed at millimetre wavelengths and the network of cosmic filaments of dark matter apparent in all cosmological hydrodynamical simulations is still under study. Using the NIKA2 dual-band millimetre camera, we mapped a field of ~ 90 arcminutes^2 in the direction of the star GJ526 simultaneously in its 1.15-mm and 2.0-mm continuum wavebands to investigate the nature of the quasi-alignment of five sources found ten years earlier with the MAMBO camera at 1.2 mm. We find that these sources are not clumps of a circumstellar debris disc around this star as initially hypothesized. Rather, they must be dust-obscured star-forming galaxies, or sub-millimetre galaxies (SMGs), in the distant background. The new NIKA2 map at 1.15 mm reveals a total of seven SMGs distributed in projection on the sky along a filament-like structure crossing the whole observed field. Furthermore, we show that the NIKA2 and supplemental Herschel photometric data are compatible with a model of the spectral energy distributions (SEDs) of these sources when a common redshift of 2.5 and typical values of the dust parameters for SMGs are adopted. Hence, we speculate that these SMGs might be located in a filament of the distant `cosmic web'. The length of this candidate cosmic filament crossing the whole map is at least 4 cMpc (comoving), and the separations between sources are between 0.25 cMpc and 1.25 cMpc at this redshift, in line with expectations from cosmological simulations. Nonetheless, further observations to determine the precise spectroscopic redshifts of these sources are required to definitively support this hypothesis of SMGs embedded in a cosmic filament of dark matter., Comment: Article accepted in Astronomy and Astrophysics ; 9 pages, 6 figures

Autonomous motivation for self-set goals (pursuing goals for more intrinsic and less extrinsic reasons) has been linked to affective well-being. Using intensive longitudinal data, the present paper examines the link between university students' autonomous study motivation with affective well-being and targets fulfillment of the basic psychological needs as a potential mediating factor of this association on the within-person and the between-person level. University students in Germany (N = 488) completed an online questionnaire once a week over up to 2 semesters, indicating their weekly study motivation, need fulfillment, and affective well-being. Multilevel structural equation models were employed to target (a) the structure of autonomous study motivation and (b) the associations of autonomous study motivation with need fulfillment and affective well-being. Autonomous study motivation was associated with students’ positive and negative affect on both levels. In line with predictions by self-determination theory, multilevel mediation models suggested indirect effects via need fulfillment on both levels. Results highlight the central role of autonomous study motivation and need fulfillment in university students' socio-emotional adjustment. A better understanding of this socio-emotional adjustment of university students may be an important step for increasing overall study satisfaction and developing interventions to reduce study dropout.

Experiences in everyday life are often collected via experience sampling approaches or daily-diaries, two methods which allow to capture similar, but not identical information about emotional states. Prior research reported divergences between aggregated momentary emotion assessments collected throughout the day and retrospective emotion assessments collected at the end of a day. The present study examines whether differences in the use of emotion regulation strategies (reappraisal, rumination, suppression) are related to the divergence of aggregated momentary assessments and retrospective assessments of positive and negative affect. Results from a combination of an experience sampling and daily-diary study (N = 296, 14 days, 6 assessments per day) suggest that (a) rumination widens the memory-experience gap for negative affect, (b) reappraisal attenuates the association between aggregated momentary and retrospective assessments, (c) suppression has differential effects on the association between momentary experiences and retrospective assessments for positive vs. negative affect. Overall, these findings suggest that emotion regulation might play a key role for understanding the association between momentary emotional experiences and recollections thereof.

Fulfillment of basic psychological needs has been postulated to be essential for mental health across the human life span. So far, validated domain-general instruments using the same items to assess these constructs in different age groups are lacking. To close this gap, we introduce the General Need Satisfaction and Frustration Scale, a multidimensional instrument to capture fulfillment of the basic psychological needs for autonomy, competence, and relatedness in children and adults. The factor structure and correlates of this new instrument are examined in two cross-sectional studies with children (N = 185, age range = 7-14) and adults (N = 380, age range = 18-77), and one ambulatory assessment study with children (N = 84, age range = 8-10). Confirmatory factor analyses suggest adequate fit of the postulated six-dimensional measurement model (three needs, each split into satisfaction and frustration). Need satisfaction was linked to higher well-being, and need frustration was linked to higher ill-being in both children and adults. Findings suggest that the instrument introduced in this work is suitable tocapture between-person differences as well as day-to-day fluctuations in the fulfillment of the basic needs for autonomy, competence, and relatedness in children and adults. The measurement instrument therefore is a useful tool to capture core ingredients for mental health in samples across a broad range of the human life span.

Secreted phosphoprotein 1 (SPP1, also known as osteopontin) binds integrins to mediate cell–cell and cell–extracellular matrix communication to promote cell adhesion, migration, and differentiation. Considerable evidence links SPP1 to pregnancy in several species. Current evidence suggests that SPP1 is involved in implantation and placentation in mice, but in vivo localization of SPP1 and in vivo mechanistic studies to substantiate these roles are incomplete and contradictory. We localized Spp1 mRNA and protein in the endometrium and placenta of mice throughout gestation, and utilized delayed implantation of mouse blastocysts to link SPP1 expression to the implantation chamber. Spp1 mRNA and protein localized to the endometrial luminal (LE), but not glandular epithelia (GE) in interimplantation regions of the uterus throughout gestation. Spp1 mRNA and protein also localized to uterine naturel killer (uNK) cells of the decidua. Within the implantation chamber, Spp1 mRNA localized only to intermittent LE cells, and to the inner cell mass. SPP1 protein localized to intermittent trophoblast cells, and to the parietal endoderm. These results suggest that SPP1: (1) is secreted by the LE at interimplantation sites for closure of the uterine lumen to form the implantation chamber; (2) is secreted by LE adjacent to the attaching trophoblast cells for attachment and invasion of the blastocyst; and (3) is not a component of histotroph secreted from the GE, but is secreted from uNK cells in the decidua to increase angiogenesis within the decidua to augment hemotrophic support of embryonic/fetal development of the conceptus. Summary sentence Through utilization of delayed implantation, SPP1 mRNA and protein are localized in high levels to the luminal epithelium of interimplantation sites, to focal regions of the implantation chamber and to uterine natural killer cells in the decidua, suggesting roles for implantation in mice.

Background Crohn’s disease (CD) is a chronic inflammatory condition that can affect all parts of the intestine. Commonly, there is inflammation in the terminal ileum. An intestinal barrier consisting of epithelial cells covered by a mucus layer is on the one hand preventing pathogen invasion while on the other hand allowing the uptake of nutrients. However, despite the significance of the terminal ileum in CD, little is known about the mucus composition and viscoelastic properties in the terminal ileum. In our study, we wanted to do a comprehensive analysis of the mucus in the terminal ileum of CD patients compared to healthy controls. Thus, we aspire to gain a better understanding of the role of mucus and biophysical and biochemical triggers contained in the mucus layer during the course of ileal inflammation. Methods During colonoscopy, native mucus and saline washes containing mucus as well as biopsies from the terminal ileum of 33 CD patients and 25 healthy controls were collected. The disease course and dietary habits of all patients and healthy controls were recorded. Samples were treated according to protocol for different types of analysis such as rheology, proteomics, metabolomics, and microbiome. Furthermore, human intestinal organoids were generated from the biopsies. Additionally, we analysed bulk RNA sequencing data from intestinal mucosal biopsies of a cohort of 128 patients, consisting of non-IBD individuals and CD patients. Results We here present preliminary data for the different types of analyses. Regarding the viscoelastic properties, preliminary rheology data suggests that there is no major difference between CD patients and healthy controls. As for proteomics, the first cohort is currently being analysed. Concerning the microbiome, we could show that saline washes containing mucus are more suitable than biopsies for analysing bacterial as well as host DNA. Although not statistically significant, healthy controls showed a tendency for higher alpha- diversity in microbiota. Also there were some differences in the taxonomy. Intestinal organoids were grown as cultures. In the end, the results of the analyses mentioned above will be interpreted together with the clinical data. Concerning the gene expression, we found that 12 mucin and mucin-like genes were expressed both in colon and ileum tissue. Interestingly, a subset of CD patients showed a de-novo expression of MUC5AC, MUC6, MUCL1 and MUCL3 in the ileum. Conclusion All in all, in our multidisciplinary study we hope to achieve a thorough analysis of the mucus layer in the terminal ileum, thus gaining new insight into the pathogenetic mechanisms in intestinal inflammation that could prospectively be used for therapeutic means.

Background Organ function is known to decline with age. Optimizing cardiac, pulmonary and renal function in older adults has led to significant improvements in perioperative care. However, when substantial blood loss and fluid shifts occur, perioperative outcomes still remains poor, especially in older adults. We suspect that this could be due to age-related changes in endothelial function—an organ controlling the transport of fluid and solutes. The capillary filtration coefficient (CFC) is an important determinant of fluid transport. The CFC can be measured in vivo, which provides a tool to estimate endothelial barrier function. We have previously shown that the CFC increases when giving a fluid bolus resulting in increased vascular and extravascular volume expansion, in young adults. This study aimed to compare the physiologic determinants of fluid distribution in young versus older adults so that clinicians can best optimize perioperative fluid therapy. Methods Ten healthy young volunteers (ages 21–35) and nine healthy older volunteers (ages 60–75) received a 10 mL/kg fluid bolus over the course of twenty minutes. Hemodynamics, systolic and diastolic heart function, fluid volumetrics and microcirculatory determinants were measured before, during, and after the fluid bolus. Results Diastolic function was reduced in older versus younger adults before and after fluid bolus (P p p = 0.059). Mean arterial pressure and systemic vascular resistance were elevated in the older versus younger adults (p Conclusion Older adults show a less reactive CFC to a fluid bolus, which could reduce blood to tissue transport of fluid. Diastolic dysfunction likely contributes to fluid maldistribution in older adults.

Groundwater heat pump systems cause thermal anomalies in the aquifer that can impact upon downstream systems and reduce their efficiency. Therefore, it is important to optimally position the extraction and injection wells of such systems to avoid negative interactions and maximize the thermal potential of the aquifer. This paper presents a new method to determine optimal well layouts of groundwater heat pumps using the adjoint approach, which is an efficient way to solve the underlying PDE-constrained optimization problem. An integral part of the method is the numerical groundwater simulation, which here is based on the finite element method. In addition, a multi-start initialization strategy is introduced in an attempt to better reach the global optimum. The method is applied to a real case study with 10 groundwater heat pumps, i.e. 20 wells, and two optimization scenarios with different natural groundwater temperatures. In both scenarios, the optimization method successfully determines a well layout that maximizes groundwater temperatures at all extraction wells. Comparing the results from these scenarios demonstrates that hydro-geological conditions can have a significant impact on the optimal well layout. The proposed method is equally applicable to systems with multiple extraction and injection wells and can be extended to various other shallow geothermal applications, such as combined heating and cooling systems.

Roles of fructose in elongating ovine conceptuses are poorly understood, despite it being the major hexose sugar in fetal fluids and plasma throughout gestation. Therefore, we determined if elongating ovine conceptuses utilize fructose via metabolic pathways for survival and development. Immunohistochemical analyses revealed that trophectoderm and extra-embryonic endoderm express ketohexokinase and aldolase B during the peri-implantation period of pregnancy for conversion of fructose into fructose-1-phosphate for entry into glycolysis and related metabolic pathways. Conceptus homogenates were cultured with 14C-labeled glucose and/or fructose under oxygenated and hypoxic conditions to assess contributions of glucose and fructose to the pentose cycle (PC), tricarboxylic acid cycle, glycoproteins, and lipid synthesis. Results indicated that both glucose and fructose contributed carbons to each of these pathways, except for lipid synthesis, and metabolized to pyruvate and lactate, with lactate being the primary product of glycolysis under oxygenated and hypoxic conditions. In patients with multi-organ system trauma, the diagnosis of coinciding traumatic brain injury can be difficult due to injuries from the hemorrhagic shock that confound clinical and radiographic signs of traumatic brain injury. In this study, a novel technique using heart rate variability was developed in a porcine model to detect traumatic brain injury early in the setting of hemorrhagic shock without the need for radiographic imaging or clinical exam.A porcine model of hemorrhagic shock was used with an arm of swine receiving hemorrhagic shock alone and hemorrhagic shock with traumatic brain injury. High-resolution heart rate frequencies were collected at different time intervals using waveforms based on voltage delivered from the heart rate monitor. Waveforms were analyzed to assess statistically significant differences between heart rate variability parameters in those with hemorrhagic shock and traumatic brain injury versus those with only hemorrhagic shock. Stochastic analysis was used to assess the validity of results and create a model by machine learning to better assess the presence of traumatic brain injury.Significant differences were found in several heart rate variability parameters between the two groups. Additionally, significant differences in heart rate variability parameters were found in swine within 1 hour of inducing hemorrhage in those with traumatic brain injury versus those without. These results were confirmed with stochastic analysis and machine learning was used to generate a model which determined the presence of traumatic brain injury in the setting of hemorrhage shock with 91.6% accuracy. Heart rate variability represents a promising diagnostic tool to aid in the diagnosis of traumatic brain injury within 1 hour of injury.

Objectives To determine whether dental maturity (dental development) was delayed in patients with Muenke syndrome, Saethre-Chotzen syndrome, and TCF12-related craniosynostosis, compared with a Dutch control group without syndromes. Materials and methods This study included 60 patients (38 patients with Muenke syndrome, 17 patients with Saethre-Chotzen syndrome, and 5 with TCF12-related craniosynostosis), aged 5.8–16.8 years that were treated at the Department of Oral Maxillofacial Surgery, Special Dental Care, and Orthodontics, in Sophia Children’s Hospital, Erasmus University Medical Center, Rotterdam, the Netherlands. Dental age was calculated according to Demirjian’s index of dental maturity. The control group included 451 children without a syndrome. Results Compared with the control group, dental development was delayed by an average of one year in 5- to 8-year-old patients with Muenke syndrome (P = 0.007) and in 8- to 10-year-old patients with Saethre-Chotzen syndrome (P = 0.044), but not in patients with TCF12-related craniosynostosis. Conclusions Our results indicated that dental development was delayed by one year, on average, in patients with Muenke syndrome and Saethre-Chotzen syndrome, compared with a Dutch control group without syndromes. Implications Our findings have improved the understanding of dental development in patients with Muenke and Saethre-Chotzen syndrome. These results can provide guidance on whether the orthodontist needs to consider growth disturbances related to dental development.

Objectives: This study investigated how COVID-19-related media exposure during the COVID-19 crisis was related to same-day and next-day COVID-19-related worries. Design: A 21-day diary study was conducted between late March and late April 2020 in Germany. Main Outcome Measures: Hypotheses were tested in a sample of 561 participants (Mage = 42.79, SDage = 6.12). Every evening, participants indicated their exposure to COVID-19-related media (e.g., TV, print, online) and their COVID-19-related worries. Results: Same-day analyses showed that participants reported more COVID-19-related worries on days with higher exposure to COVID-19-related media. Dynamical structural equation models provided evidence for a reciprocal cycle across days: Higher media exposure at one day predicted higher worries the next day, and higher worries at one day also predicted higher media exposure the next day. Individuals with high trait anxiety reported an enhanced general level of media exposure during the 21 days of assessment, and individuals high in neuroticism and anxiety reported an enhanced level of worries.Conclusion: These findings suggest a self-reinforcing cycle whereby consuming crisis-related media and worrying reciprocally influence each other across days, possibly amplifying adverse effects of the COVID-19 crisis and other crises alike on mental and physical health.

As a means to counter the SARS-CoV‑2 pandemic, schools were closed throughout Germany between mid-March and end of April 2020. Schooling was translocated to the students' homes where students were supposed to work on learning tasks provided by their teachers. Students' self-regulation and attributes of the learning tasks may be assumed to have played important roles when adapting to this novel schooling situation. They may be predicted to have influenced students' daily self-regulation and hence the independence with which they worked on learning tasks. The present work investigated the role of students' trait self-regulation as well as task difficulty and task enjoyment for students' daily independence from their parents in learning during the homeschooling period. Data on children's trait self-regulation were obtained through a baseline questionnaire filled in by the parents of 535 children (Um die SARS-CoV-2-Pandemie einzudämmen, wurden zwischen Mitte März und Ende April 2020 deutschlandweit alle Schulen geschlossen. Während dieser Zeit wurden die Schüler*innen zu Hause beschult (‚Homeschooling‘) und arbeiteten an Lernaufgaben, die von ihren Lehrkräften bereitgestellt wurden. Bei der Anpassung an diese neuartige Beschulungssituation könnten sowohl die Selbstregulation der Schüler*innen als auch die Eigenschaften der Lernaufgaben eine wichtige Rolle gespielt haben. Es ist anzunehmen, dass diese insbesondere die tagtägliche Selbstregulation der Schüler*innen beim Bearbeiten der Lernaufgaben und damit die Selbstständigkeit, mit der sie an den Lernaufgaben arbeiteten beeinflusst haben. Die vorliegende Arbeit untersuchte, welche Rolle die dispositionelle Selbstregulation von Schüler*innen, die Schwierigkeit der Lernaufgaben und die Freude an den Aufgaben für die tagtägliche Selbstständigkeit der Bearbeitung der Aufgaben während des Homeschoolings spielten. Computational models of mantle convection must accurately represent curved boundaries and the associated boundary conditions of a 3-D spherical shell, bounded by Earth's surface and the core–mantle boundary. This is also true for comparable models in a simplified 2-D cylindrical geometry. It is of fundamental importance that the codes underlying these models are carefully verified prior to their application in a geodynamical context, for which comparisons against analytical solutions are an indispensable tool. However, analytical solutions for the Stokes equations in these geometries, based upon simple source terms that adhere to physically realistic boundary conditions, are often complex and difficult to derive. In this paper, we present the analytical solutions for a smooth polynomial source and a delta-function forcing, in combination with free-slip and zero-slip boundary conditions, for both 2-D cylindrical- and 3-D spherical-shell domains. We study the convergence of the Taylor–Hood (P2–P1) discretisation with respect to these solutions, within the finite element computational modelling framework Fluidity, and discuss an issue of suboptimal convergence in the presence of discontinuities. To facilitate the verification of numerical codes across the wider community, we provide a Python package, Assess, that evaluates the analytical solutions at arbitrary points of the domain.

Child development 92 (2021) 5, S. 1679-1697, This study examined the effects of daily parental autonomy support on changes in child behavior, family environment, and parental well-being across 3 weeks during the COVID-19 pandemic in Germany. Day-to-day associations among autonomy-supportive parenting, parental need fulfillment, and child well-being were also assessed. Parents of school children (6–19 years) reported on adjustment measures at two measurement occasions and completed up to 21 daily online questionnaires in the weeks between these assessments. Results from dynamic structural equation models suggested reciprocal positive relations among autonomy-supportive parenting and parental need fulfillment. Daily parental autonomy support, parental need fulfillment, and child well-being partially predicted change in adjustment measures highlighting the central role of daily parenting for children’s adjustment during the pandemic.
