Os acessos nativos do Rio Grande do Sul, “28B”, “26A”, “28C”, “26D” e “28E” de Paspalum nicorae e os ecótipos “Azulão” e “Baio” de Paspalum guenoarum foram avaliados com o objetivo de dimensionar a intensidade de associação da produção de matéria seca de folha com outros caracteres de interesse forrageiros nessas espécies apomíticas, testadas em diferentes anos e locais. O delineamento adotado foi o de blocos casualizados com três repetições em duas regiões fisiograficamente distintas. As avaliações foram realizadas por meio de cortes. As variáveis avaliadas foram a produção de matéria seca total, matéria seca de folhas, matéria seca de colmo, relação folha:colmo, índice de colheita e o número de afilhos. Correlações similares foram obtidas nos dois locais, qualificando a matéria seca de folhas como a de maior magnitude na relação direta e positiva à materia seca total. A análise de correlação geral demonstrou que a matéria seca de folhas e matéria seca de colmo forma alta correlação positiva para o incremento da matéria seca total. A produção da matéria seca total pode ser utilizada diretamente para seleção de genotipos em espécies do gênero Paspalum.

O objetivo do trabalho foi determinar a variabilidade genética entre acessos de diferentes espécies de Paspalum e a expressão de caracteres de interesse forrageiro em distintos locais e anos de cultivo. Os experimentos foram conduzidos em delineamento experimental de blocos ao acaso com três repetições com parcelas compostas por cinco acessos de P. nicorae, dois de P. guenoraum e mais a cultivar Pensacola (P. notatum) utilizada como testemunha. Os acessos avaliados apresentaram variabilidade genética em caracteres de interesse forrageiro com efeitos na interação genótipo x ambiente. A produção de matéria seca total e de folhas foram os caracteres que mais contribuíram para a detecção da variabilidade genética observada independentemente do ano de avaliação.

The article presents a study on human placental tissue morphology during and after SARS-CoV-2 infection. Topics include information on relevance of SARS-CoV-2 in inducing vascular injury and also in the placenta; mother-to-fetus transmission of virus; and how SARS-CoV-2 enters human cells via angiotensin converting enzyme 2.

Despite the economic importance of Crambe abyssinica, the present study is the first report on bees that occur with this species, and could aid in developing alternative methods for controlling insect pests without seriously impacting pollinators. The present study examined the following questions: (1) Which species are potential pollinators of C. abyssinica? (2) How do environmental conditions influence pollinator visitation fluctuations? Insects were sampled on a weekly basis between 08:00 and 16:00 during five weeks of flowering. When the results of analyses of variance were significant, the data was adjusted using regression equations at a 5% level of probability; the environmental variables were examined using Pearson's correlation coefficient. Apis mellifera, Geotrigona mombuca, Exomalopsis (Exomalopsis) fulvofasciata, Plebeia sp. and Dialictus sp. were observed visiting C. abyssinica flowers. A. melliferaand G. mombucawere observed to be potential pollinators, with the former demonstrating visitation peaks during flowering weeks II and IV at 12:00 and 08:00, respectively and the latter visiting during weeks III and IV at 12:00 and 10:00, respectively. Environmental factors such as temperature, relative humidity, and wind speed have different effect on the activity of bees.

Fetal cardiac intervention consists of a novel and evolving technique for the intrauterine treatment of a subset of patients with congenital heart diseases, which aims to improve hemodynamics, reduce secondary damage, and achieve better postnatal outcomes. Nevertheless, the risks and benefits of this therapy remains subject to controversy. This review describes the rationale, selection criteria, and technical features for the most frequently performed fetal cardiac interventions. SARS-CoV-2 infection is considered as a multi-organ disease, and several studies highlighted the relevance of the virus infection in the induction of vascular injury and tissue morphological alterations, including placenta. In this study, immunohistochemical analyses were carried out on placenta samples derived from women with COVID-19 infection at delivery (SARS-CoV-2 PCR+) or women healed from a COVID-19 infection (SARS-CoV-2 negative at delivery, SARS-CoV-2 PCR-) or women who gave birth before 2019 (Control). Angiotensin Converting Enzyme 2 (ACE2) receptor, Cluster of differentiation 147 (CD147), endothelial CD34 marker, Vascular Endothelial Growth Factor (VEGF) and total Microtubule-associated protein 1 Light Chain 3B marker (LC3B) were investigated in parallel with SPIKE protein by standard IHC. Multiplexed Immunohistochemical Consecutive Staining on Single Slide (MICSSS) was used to examine antigen co-expression in the same specimen. SPIKE protein was detected in villi and decidua from women with ongoing infection, with no significant differences in SPIKE staining between both biopsy sites. VEGF was significantly increased in SARS-CoV-2 PCR + biopsies compared to control and SARS-CoV-2 PCR- samples, and MICSSS method showed the co-localization of SPIKE with VEGF and CD34. The induction of autophagy, as suggested by the LC3B increase in SARS-CoV-2 PCR + biopsies and the co-expression of LC3B with SPIKE protein, may explain one of the different mechanisms by which placenta may react to infection. The Paspalum genus has potential for further genetic improvement because of its adaptability to different ecosystems and production of high yields for grazing livestock. We estimate the genetic parameters of 195 intraspecific P. notatum hybrids using Restricted Maximum Likelihood (REML), followed by selection based on Best Linear Unbiased Prediction (BLUP) through multivariate analysis. The intraspecific hybrids studied showed considerable genetic variability in the evaluated forage traits, displaying their potential for progression in subsequent stages of the genetic improvement program. Notably, plant height emerged as an important trait for indirect selection to enhance forage production. The use of the REML/BLUP procedure proves to be a robust tool for data analysis, particularly for perennial species. Furthermore, multivariate analysis based on BLUPs should be used in the selection process within breeding programs. Based on the BLUP values, hybrids D3, D16, C17, C2 and B17 were identified as superior for forage production, and they hold promise for future breeding programs for future breeding initiatives aimed at direct selection to improve yield.

Microalgae represent a growing innovative source of nutraceuticals such as carotenoids and phenolic compound which are naturally present within these single-celled organisms or can be induced in response to specific growth conditions. The presence of the unfavourable allelic variant in genes involved in the control of oxidative stress, due to one or more SNPs in gene encoding protein involved in the regulation of redox balance, can lead to pathological conditions such as insulin resistance, which, in turn, is directly involved in the pathogenesis of type 2 diabetes mellitus. In this review we provide an overview of the main SNPs in antioxidant genes involved in the promotion of insulin resistance with a focus on the potential role of microalgae-derived antioxidant molecules as novel nutritional tools to mitigate oxidative stress and improve insulin sensitivity.

SARS-CoV viruses have been shown to downregulate cellular events that control antiviral defenses. They adopt several strategies to silence p53, key molecule for cell homeostasis and immune control, indicating that p53 has a central role in controlling their proliferation in the host. Specific actions are the stabilization of its inhibitor, MDM2, and the interference with its transcriptional activity. The aim of our work was to evaluate a new approach against SARS-CoV-2 by using MDM2 inhibitors to raise p53 levels and activate p53-dependent pathways, therefore leading to cell cycle inhibition. Experimental setting was performed in the alveolar basal epithelial cell line A549-hACE2, expressing high level of ACE2 receptor, to allow virus entry, as well as p53 wild-type. Cells were treated with several concentrations of Nutlin-3 or RG-7112, two known MDM2 inhibitors, for the instauration of a cell cycle block steady-state condition before and during SARS-CoV-2 infection, and for the evaluation of p53 activation and impact on virus release and related innate immune events. The results indicated an efficient cell cycle block with inhibition of the virion release and a significant inhibition of IL-6, NF-kB and IFN-λ expression. The heart of higher vertebrates develops early as a tubular structure, which requires cellular and molecular events for proliferation, differentiation and apoptosis for growth, and individualization of cardiac chambers. Exposure to different stressors can cause disturbances in the normal development and functionality of the cardiovascular system. This study aimed to characterize the impact of methylmercury (MeHg) on heart development, specifically related to tissue morphology and parameters of vascular integrity and contractility, also focusing on cell cycle and apoptosis, using Gallus domesticus embryos as a model. The results showed morphological alterations, reduction in the thickness of the ventricular walls, and trabeculae changes in the hearts of embryos exposed to 0.1 µg MeHg/50 µL saline solution. These impacts were associated with increased contents of proteins related to cell cycle arrest and reduced cardiomyocyte proliferation. In addition, the contents of endothelial mediators for contractility and vascular integrity were imbalanced. The quantity and morphology of mitochondria of cardiomyocytes were injured. Together, these negative measurements impacted the reduction of heartbeats. In general, the parameters identified here demonstrate the relevance of combined molecular cellular tissue and physiological diagnosis for a better understanding of the cardiotoxicity of MeHg during development., (© 2022. Understanding the inner morphology of intact tissues is one of the most competitive challenges in modern biology. Since the beginning of the twentieth century, optical tissue clearing (OTC) has provided solutions for volumetric imaging, allowing the microscopic visualization of thick sections of tissue, organoids, up to whole organs and organisms (for example, mouse or rat). Recently, tissue clearing has also been introduced in clinical settings to achieve a more accurate diagnosis with the support of 3D imaging. Several evidence suggests that, in addition to the respiratory tract, also the gastrointestinal tract is a main site of severe acute respiratory syndrome CoronaVirus 2 (SARS-CoV-2) infection, as an example of a multi-organ vascular damage, likely associated with poor prognosis. To assess mechanisms SARS-CoV-2 responsible of tissue infection and vascular injury, correlating with thrombotic damage, specimens of the digestive tract positive for SARS-CoV-2 nucleocapsid protein were analyzed deriving from three patients, negative to naso-oro-pharyngeal swab for SARS-CoV-2. These COVID-19-negative patients came to clinical observation due to urgent abdominal surgery that removed different sections of the digestive tract after thrombotic events. Immunohistochemical for the expression of SARS-CoV-2 combined with a panel of SARS-CoV-2 related proteins angiotensin-converting enzyme 2 receptor, cluster of differentiation 147 (CD147), human leukocyte antigen-G (HLA-G), vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 was performed. Tissue samples were also evaluated by electron microscopy for ultrastructural virus localization and cell characterization. The damage of the tissue was assessed by ultrastructural analysis. It has been observed that CD147 expression levels correlate with SARS-CoV-2 infection extent, vascular damage and an increased expression of VEGF and thrombosis. The confirmation of CD147 co-localization with SARS-CoV-2 Spike protein binding on gastrointestinal tissues and the reduction of the infection level in intestinal epithelial cells after CD147 neutralization, suggest CD147 as a possible key factor for viral susceptibility of gastrointestinal tissue. Plastic pollution is an emerging problem and is a consequence of the post-consumer plastic waste accumulation in the environment coupled to mismanaged waste programmes. Countries are counteracting the continuous growth of plastic litter with different strategies: introducing bans and limits on both plastic items and materials, promoting plastic recycling and recovery strategies and encouraging voluntary clean up actions, as well as raising public awareness. However, the toxicity of plastics to the environment and organisms is not only related to their polymer chains, but also to the fact that plastic materials contain hazardous additives and can adsorb environmental pollutants (i.e. heavy metals and persistent organic contaminants, respectively). The plastic/additives/pollutants combination may be ingested by marine organisms and then enter in the food chain. Therefore, legislation for additives and contaminants is crucial both to reduce environmental pollution and their toxic effects on organisms, which of course includes humans. In this review, the current policies on plastics and related contaminants are described focusing on current laws. Moreover, recommendations for seafood consumption are suggested, since each fish or mollusc eaten may potentially result in plastic particles, additives or contaminants ingestion., (Copyright © 2021 Elsevier Ltd. Advances in screening methods and pharmacological treatments are increasing the life expectancy of cancer patients. During recent decades, the community of long-term disease-free cancer survivors (LCS) has grown exponentially, raising the issues related to cancer follow-up. Cancer relapse and other cancer-related diseases, as well as lifestyle, influence cancer survival. Recently, the regulatory role of microRNAs (miRNAs) in gene expression and their involvement in human diseases, including cancer, has been identified. Extracellular circulating miRNAs (ECmiRNAs) have been found in biological fluids and specific ECmiRNAs have been associated with cancer development and progression or with a therapy response. Here, we focus on the pivotal role of ECmiRNAs as biomarkers in cancer diagnosis and prognosis. Then, we discuss the relevance of ECmiRNAs expression in cancer survivors for the identification of specific ECmiRNAs profiles as potential tools to assess cancer outcome and to control LCS follow-up.

Extracellular vesicles (EVs) have attracted interest as mediators of intercellular communication following the discovery that EVs contain RNA molecules, including non-coding RNA (ncRNA). Growing evidence for the enrichment of peculiar RNA species in specific EV subtypes has been demonstrated. ncRNAs, transferred from donor cells to recipient cells, confer to EVs the feature to regulate the expression of genes involved in differentiation, proliferation, apoptosis, and other biological processes. These multiple actions require accuracy in the isolation of RNA content from EVs and the methodologies used play a relevant role. In liver, EVs play a crucial role in regulating cell-cell communications and several pathophysiological events in the heterogeneous liver class of cells via horizontal transfer of their cargo. This review aims to discuss the rising role of EVs and their ncRNAs content in regulating specific aspects of hepatocellular carcinoma development, including tumorigenesis, angiogenesis, and tumor metastasis. We analyze the progress in EV-ncRNAs' potential clinical applications as important diagnostic and prognostic biomarkers for liver conditions.

The microenvironment that surrounds a tumor, in addition to the tumor itself, plays an important role in the onset of resistance to molecularly targeted therapies. Cancer cells and their microenvironment interact closely between them by means of a molecular communication that mutually influences their biological characteristics and behavior. Leukemia cells regulate the recruitment, activation and program of the cells of the surrounding microenvironment, including those of the immune system. Studies on the interactions between the bone marrow (BM) microenvironment and Acute Lymphoblastic Leukemia (ALL) cells have opened a scenario of potential therapeutic targets which include cytokines and their receptors, signal transduction networks, and hypoxia-related proteins. Hypoxia also enhances the formation of new blood vessels, and several studies show how angiogenesis could have a key role in the pathogenesis of ALL. Knowledge of the molecular mechanisms underlying tumor-microenvironment communication and angiogenesis could contribute to the early diagnosis of leukemia and to personalized molecular therapies. Recent studies have shown that G protein-coupled receptors (GPCRs), the largest signal-conveying receptor family, are targets for mutations occurring frequently in different cancer types. GPCR alterations associated with cancer development represent significant challenges for the discovery and the advancement of targeted therapeutics. Among the different molecules that can activate GPCRs, we focused on two molecules that exert their biological actions regulating many typical features of tumorigenesis such as cellular proliferation, survival, and invasion: somatostatin and melatonin. The modulation of signaling pathways, that involves these two molecules, opens an interesting scenario for cancer therapy, with the opportunity to act at different molecular levels. Silicon oxide (SiO 2 ) nanostructures (SiO 2 NS) are increasingly being incorporated into an array of products, notably in the food, pharmaceutical, medical industries and in water treatment systems. Amorphous SiO 2 NS have low toxicity, however, due to their great versatility, superficial modifications can be made and these altered structures require toxicological investigation. In this study, SiO 2 NS were synthetized and amine-functionalized with the molecules (3-aminopropyl)triethoxysilane (APTMS) and 3-[2-(2-aminoethylamino)ethylamino]propyltrimethoxysilane (AEAEAPTMS), named SiO 2 NS@1 and SiO 2 NS@3, respectively. The bare SiO 2 NS, SiO 2 NS@1 and SiO 2 NS@3 samples were characterized and the influence of the culture medium used in the toxicological assays was also evaluated. The effect of amine functionalization of SiO 2 NS was investigated through acute and chronic toxicity assays with Daphnia magna. Modifications to ultrastructures of the intestine and eggs of these organisms were observed in TEM and SEM analysis. The toxicity was influenced by the surface modifications and a possible Trojan horse effect was highlighted, particularly in the case of chronic exposure. Exposure to all NSs promoted alterations in the microvilli and mitochondria of the D. magna intestine and some damage to egg cells was also observed. Background/aim: Glioblastomas (GBMs) are the most malignant primary brain tumor. New treatment strategies against the disease are urgently needed, as therapies are not completely efficient. In this study, we evaluated the antitumorigenic activity of the carotenoid fucoxanthin (Fx) on human GBM cells in vitro., Materials and Methods: GBM1 cell viability and proliferation was assessed by MTT reduction, Ki67 and single cell cloning assays. GBM1 migration and invasion were analyzed by wound healing and Transwell assays. Apoptosis and necrosis were analyzed by flow cytometry, and the mitochondrial membrane potential (ΔΨm) by the selective fluorescent dye tetramethylrhodamine ethyl ester. Cell morphology was analyzed through scanning electron microscopy and transmission electron microscopy. Fx anti-angiogenic effect was assessed by the CAM ex ovo assay., Results: Fx decreased cell viability in a concentration-dependent manner (40-100 μ M) in GBM1, A172 and C6 cell lines and was not cytotoxic to murine astrocytes. In addition, Fx inhibited the proliferation and clonogenic potential, and decreased migration and invasion of GBM1 cells. Furthermore, Fx induced apoptosis, loss of ΔΨm and ultrastructural alterations in GBM1. Genetic resistance is the main strategy to control one of the most destructive diseases of common bean (Phaseolus vulgaris L), i.e., the Fusarium wilt caused by Fusarium oxysporum f. sp. phaseoli (Fop). However, little is known on host defense reactions in Fop-bean interaction. Thus, this work examined the defense mechanisms in root and hypocotyl tissues of common bean against Fop. Resistant and susceptible bean plants were inoculated by dipping their roots in a conidial suspension. Cross sections of roots and hypocotyls were observed in light microscopy at 1, 3, 6, and 9 days after inoculation (dai) to monitor Fop colonization, and at 3 and 9 dai to detect callose, carbohydrates, lipids, phenolics, and protein, and under electronic microscopy at 9 dai to observe ultrastructural changes in xylem cells. The content of hydrogen peroxide (H 2 O 2 ), lipid peroxidation, and activity of the antioxidant enzymes ascorbate peroxidase (EC 1.11.1.11) and catalase (EC 1.11.1.6) were monitored spectrophotometrically in roots and hypocotyls at 0, 1, 3, 6, and 9 dai. Fop colonized inter- and intracellularly the epidermis and cortex reaching the xylem vessels faster in susceptible genotype. Fop inoculation induced phenolics and carbohydrates accumulation, callose deposition, and formation of occlusion material inside xylem vessels mainly in resistant genotype. Lipid peroxidation occurred mainly in susceptible plants. In contrast, the antioxidant enzymes seem to have contributed to reducing damage caused by H 2 O 2 accumulation in resistant plants. This study gives evidences that inter- and intracellular physicochemical mechanisms can act together to delay Fop colonization in resistant plants.

Arsenic (As) is a ubiquitous contaminant in the environment and it is known to induce oxidative stress in aquatic organisms. In an attempt to remove As from water, some studies have suggested the titanium dioxide nanomaterial (nTiO 2 ) as a promising alternative. However, it has been observed that nTiO 2 can induce toxicity alone or in combination with metals, and this toxicity is dependent on its crystalline form of nanomaterial (mainly rutile as nTiO 2 R and anatase as nTiO 2 A, respectively). Considering that both (nTiO 2 and As) can occur together, the objective of this study was to evaluate if co-exposure to rutile and anatase may influence accumulation, metabolisation, and toxicity of arsenite (As +3 ) in the golden mussel Limnoperna fortunei after 48 h of co-exposure to nTiO 2 (1 mg/L) and As (50 μg/L). Accumulation and chemical speciation of As in organisms were determined. Also, biochemical analyses, such as the activity of the enzymes glutathione S-transferase omega (GSTΩ), catalase (CAT) and glutathione S-transferase (GST), as well as lipid peroxidation (LPO) were investigated. Results showed that co-exposure to nTiO 2 A + As changed accumulation pattern of metalloid in gills and digestive gland. Both crystalline forms of nTiO 2 affected the metabolisation capacity favoring the accumulation of more toxic As compounds and nTiO 2 A alone or in combination with As showed induce oxidative stress in gills of L. fortunei. The objective of this study was to examine the cytotoxic effects of binary mixtures of Al 2 O 3 and ZnO NPs using mouse fibroblast cells (L929) and human bronchial epithelial cells (BEAS-2B) as biological test systems. The synergistic, additive, or antagonistic behavior of the binary mixture was also investigated. In toxicity experiments, cellular morphology, mitochondrial function (MTT assay), apoptosis, nuclear size and shape, clonogenic assays, and damage based upon oxidative stress parameters were assessed under control and NPs exposure conditions. Although Abbott modeling results provided no clear evidence of the binary mixture of Al 2 O 3 and ZnO NPs exhibiting synergistic toxicity, some specific assays such as apoptosis, nuclear size and shape, clonogenic assay, activities of antioxidant enzymatic enzymes catalase, superoxide dismutase, and levels of glutathione resulted in enhanced toxicity for the mixtures with 1 and 1.75 toxic units (TU) toward both cell types. Data demonstrated that co-presence of Al 2 O 3 and ZnO NPs in the same environment might lead to more realistic environmental conditions. Our findings indicate cytotoxicity of binary mixtures of Al 2 O 3 and ZnO NPs produced greater effects compared to toxicity of either individual compound.

Silver nanoparticles (AgNPs) are widely applied in several types of products since they act as a biocide. However, their high level of release into the environment can bring risks to ecosystems. Thus, the toxicity of AgNPs toward duckweed (Landoltia punctata) was investigated by monitoring the growth rate inhibition and the effect on the photosynthetic metabolism through morphological and ultrastructural analysis. The AgNPs were characterized by transmission electron microscopy and the effective diameter (dynamic light scattering) and zeta potential were determined. Plants were grown according to the environmental conditions recommended in ISO/DIS 20079 and then exposed to different concentrations of AgNPs. Inhibition of the growth rate was measured based on the EC 50 and changes in the morphology, cellular structures and photosynthetic pigments were evaluated along with the silver accumulation. Although the results showed low growth inhibition when compared to other studies, significant damage to the ultrastructure, decreases in the photosynthetic pigments and starch grains, an increase in the phenolic compounds and physiological changes, such as a loss of color, were observed. Aluminum oxide nanoparticles (Al 2 O 3 NPs) can be found in different crystalline phases, and with the emergence of nanotechnology there has been a rapid increase in the demand for Al 2 O 3 NPs in different engineering areas and for consumer products. However, a careful evaluation of the potential environmental and human health risks is required to assess the implications of the release of Al 2 O 3 NPs into the environment. Thus, the objective of this study was to investigate the toxicity of two crystalline phases of Al 2 O 3 NPs, alpha (α-Al 2 O 3 NPs) and eta (η-Al 2 O 3 NPs), toward Daphnia magna and evaluate the risk to the aquatic ecology of Al 2 O 3 NPs with different crystalline phases, based on a probabilistic approach. Different techniques were used for the characterization of the Al 2 O 3 NPs. The toxicity toward Daphnia magna was assessed based on multiple toxicological endpoints, and the probabilistic species sensitivity distribution (PSSD) was used to estimate the risk of Al 2 O 3 NPs to the aquatic ecology. The results obtained verify the toxic potential of the NPs toward D. magna even in sublethal concentrations, with a more pronounced effect being observed for η-Al 2 O 3 NPs. The toxicity is associated with an increase in the reactive oxygen species (ROS) content and deregulation of antioxidant enzymatic/non-enzymatic enzymes (CAT, SOD and GSH). In addition, changes in MDA levels were observed, indicating that D. magna was under oxidative stress. The most prominent chronic toxic effects were observed in the organisms exposed to η-Al 2 O 3 NPs, since the lowest LOEC was 3.12 mg/L for all parameters, while for α-Al 2 O 3 NPs the lowest LOEC was 6.25 mg/L for longevity, growth and reproduction. Acute lymphoblastic leukemia (ALL) represents a heterogeneous group of hematologic malignancies, and it is normally characterized by an aberrant proliferation of immature lymphoid cells. Moreover, dysregulation of multiple signaling pathways that normally regulate cellular transcription, growth, translation, and proliferation is frequently encountered in this malignancy. ALL is the most frequent tumor in childhood, and adult ALL patients still correlate with poor survival. This review focuses on modern therapies in ALL that move beyond standard chemotherapy, with a particular emphasis on immunotherapeutic approaches as new treatment strategies. Bi-specific T-cell Engagers (BiTE) antibodies, the chimeric antigen receptor (CAR)-T cells, or CRISPR-Cas9 (clustered regularly interspaced short palindromic repeats [CRISPR]-associated nuclease 9) represent other new innovative approaches for this disease. Target and tailored therapy could make the difference in previously untreatable cases, i.e., precision and personalized medicine.
