Three methods were tested for fractionating H-2 antigens from a participate fraction of Sarcoma I using the 51Cr cytotoxic assay to measure antigenic activities. A principal difference among the fractionation procedures was the type of detergent employed to solubilize the antigens. A procedure using Triton X 100 was the least satisfactory. Those using Triton X 114 or cholate were equally effective, but the procedure involving the use of cholate was more convenient. Fractionation of antigens in detergent solutions with (NH4)2SO4was not an efficient procedure. Digestion with trypsin further increased the specific activity of fractions obtained by treatment with detergents. The highest purification attained was a 10-fold increase in activity over that of the starting material. The yield of activity was usually about 60. No evidence for separation of H-2 antigenic specificities was obtained. Detergent-solubilized preparations were retained wholly or in part on Sephadex and Agarose columns, but these procedures did not result in increased purity

Rejection of tailskin grafts exchanged between two male hybrids of the cross B10.M × B10.RIII(71NS) revealed a mutation in theH-2f haplotype from the B10.M congenic line. Complementation studies with skin grafting and cell-mediated lympholysis showed the mutant, namedH-2fb, to be different from anotherH-2f mutant,H-2fa, and further, that the HH-2fb mutation occurred in theD end of theH-2 complex. Reciprocal skin grafts exchanged between mutant and normal mice were rejected. Hemagglutinating antibody reactive with B10.M cells was raised in the mutant mice. Mutant spleen cells responded weakly, but significantly, to wild-type cells in a mixed lymphocyte culture and in a graftversus-host assay, but no response was seen in the opposite direction. However, cytotoxic effector cells were generated against target cells in both directions in a cell-mediated lympholysis assay.

Erythrocyte producing tissues of genetically anemicW/Wv mice were completely populated and the anemic mice were permanently cured by marrow cell grafts from donors that differed at theEa-2 locus. Circulating erythrocytes contained ? 80% donor Ea-2 antigen and ? 90% donor hemoglobin. Population occurred without immunosuppressive treatment. No sign of graft-versus-host reaction was observed, although donors were congenic with one parent strain of the F1 hybrid recipients. When partially congenic donors carrying the T6 chromosome marker were used, the immune systems of curedW-anemic mice were populated to an appreciable extent by donor cells. The dependence of cure upon route of injection and the persistence of functioning donor type cells across the strong immunological barrier of the Ea-2 system are discussed.

Three new cytotoxic H-2specificities are defined. H-2.37 is present in alleles f, p, and pa;H-2.38 in alleles p, pa, and w;and H-2.39 in alleles b, f, and i.The first two specificities have appeared in a number of antisera and, in every case, were without hemagglutinating activity. H-2.39 was very weakly hemagglutinating in one antiserum, nonhemagglutinating in another. The presence of 39 in allele band the crossover-derived iand its absence from crossover hplaces 39 at the K end (although not necessarily in the K region) of the H-2crossover map.
