11. Novel precursors for the growth of TiO2 by liquid injection MOCVD

Abstract

A comprehensive examination of the feasibility of producing new titania precursors by ligand substitution reactions involving the reaction between titanium tetra-isopropoxide and various dimethylamino alcohols has resulted in the synthesis, characterization and application of five new titanium dioxide thin film precursors, titanium tetra(dimethylaminopropan-l-ol) (Ti(DMAP1 )4), titanium tetra(dimethylaminopropan- 2-ol) (Ti(DMAP2)4), titanium isopropoxide tri(dimethylaminoethanol) (Ti(OPr')(DMAE)3), titanium tri-isopropoxide dimethylaminopropan-1-ol (Ti(OPr')3(DMAP')) and titanium tri-isopropoxide dimethylaminopropan-2-ol (Ti(OPri)3(DMAP2)). The new compounds have been characterized by *H and 13C NMR spectroscopy, mass spectrometry, infrared spectroscopy and thermogravimetric analysis (TGA). The precursors were tested for their ability to produce TiOa films on un-doped silicon and glass substrates by Liquid Injection Metal Organic Chemical Vapor Deposition. The effects of varying a wide range of deposition conditions has been studied using in situ monitoring by interferometry and ex-situ analysis using X-ray diffraction (XRD), scanning electron microscopy (SEM) and Rutherford back scattering (RBS). Data resulting from the use of the new precursors has been compared to that obtained using the current industry standard precursor, titanium tetra-isopropoxide (TTIP). It is demonstrated that the new precursors are potentially easier to handle than TTIP as a result of the internal coordination to the Ti centre via the N atoms in the replacement ligands, while all of the precursors studied are able to produce TiC>2 films. Comparisons are made between the precursors in terms of placing them in a rank order based upon ease of use and ability to produce TiC>2 films. Additionally, to demonstrate the applicability of these precursors, films of the mixed metal oxide strontium barium titanate have been grown and characterised.

Abstract

A detailed, well‐dated record of pollen and sedimentary ancient DNA (sedaDNA) for the period 15 000–9500 cal a bp describes changes at Lake Bolshoye Shchuchye in the Polar Ural Mountains, located far east of the classical Lateglacial sites in western Europe. Arctic tundra rapidly changed to lusher vegetation, possibly including both dwarf (Betula nana) and tree birch (B. pubescens), dated in our record to take place 14 565 cal a bp, coincident with the onset of the Bølling in western Europe; this was paralleled by increased summer temperatures. A striking feature is an early decline in Betula pollen and sedaDNA reads 300 years before the onset of the Younger Dryas (YD) in western Europe. Given the solid site chronology, this could indicate that the YD cooling started in Siberia and propagated westwards, or that the vegetation reacted to the inter‐Allerød cooling at 13 100 cal a bp and did not recover during the late Allerød. During the YD, increases in steppe taxa such as Artemisia and Chenopodiaceae suggest drier conditions. At the onset of the Holocene, the vegetation around the lake reacted fast to the warmer conditions, as seen in the increase of arboreal taxa, especially Betula, and a decrease in herbs such as Artemisia and Cyperaceae. Abstract

Drawing on our approach and findings from a secondary interrogation of an existing qualitative dataset, this work set out to challenge assumptions of practice through using participatory methods. Our approach brought two key aspects together: applying the theories of Douglas’s Cultural Theory of Risk and Tronto’s Ethic of Care, and a collaboration with people living with dementia as co-analysts in the co-production (interpretation) of knowledge and representing the experiences within the qualitative dataset. The research generated an understanding of relational care and the subtle ways in which relationships change for someone with dementia. In this case, we describe the processes we followed and reflect on the demands and challenges, experienced as researchers, in focusing on reflexivity, multiple voicing, literary styling, and performance. In our description of how we had to relinquish any sense of a concluding authoritative voice, we highlight the innovation in this participatory secondary data analysis and encourage others to engage with this dialogue.

Abstract

Neuroinflammation and microglial activation are significant processes in Alzheimer’s disease pathology. Recent genome-wide association studies have highlighted multiple immune-related genes in association with Alzheimer’s disease, and experimental data have demonstrated microglial proliferation as a significant component of the neuropathology. In this study, we tested the efficacy of the selective CSF1R inhibitor JNJ-40346527 (JNJ-527) in the P301S mouse tauopathy model. We first demonstrated the anti-proliferative effects of JNJ-527 on microglia in the ME7 prion model, and its impact on the inflammatory profile, and provided potential CNS biomarkers for clinical investigation with the compound, including pharmacokinetic/pharmacodynamics and efficacy assessment by TSPO autoradiography and CSF proteomics. Then, we showed for the first time that blockade of microglial proliferation and modification of microglial phenotype leads to an attenuation of tau-induced neurodegeneration and results in functional improvement in P301S mice. Overall, this work strongly supports the potential for inhibition of CSF1R as a target for the treatment of Alzheimer’s disease and other tau-mediated neurodegenerative diseases.

Abstract

INTRODUCTION: Plasma biomarkers for Alzheimer's disease (AD) diagnosis/stratification are a "Holy Grail" of AD research and intensively sought; however, there are no well-established plasma markers. METHODS: A hypothesis-led plasma biomarker search was conducted in the context of international multicenter studies. The discovery phase measured 53 inflammatory proteins in elderly control (CTL; 259), mild cognitive impairment (MCI; 199), and AD (262) subjects from AddNeuroMed. RESULTS: Ten analytes showed significant intergroup differences. Logistic regression identified five (FB, FH, sCR1, MCP-1, eotaxin-1) that, age/APOε4 adjusted, optimally differentiated AD and CTL (AUC: 0.79), and three (sCR1, MCP-1, eotaxin-1) that optimally differentiated AD and MCI (AUC: 0.74). These models replicated in an independent cohort (EMIF; AUC 0.81 and 0.67). Two analytes (FB, FH) plus age predicted MCI progression to AD (AUC: 0.71). DISCUSSION: Plasma markers of inflammation and complement dysregulation support diagnosis and outcome prediction in AD and MCI. Abstract

Introduction Plasma biomarkers for Alzheimer's disease (AD) diagnosis/stratification are a “Holy Grail” of AD research and intensively sought; however, there are no well-established plasma markers. Methods A hypothesis-led plasma biomarker search was conducted in the context of international multicenter studies. The discovery phase measured 53 inflammatory proteins in elderly control (CTL; 259), mild cognitive impairment (MCI; 199), and AD (262) subjects from AddNeuroMed. Results Ten analytes showed significant intergroup differences. Logistic regression identified five (FB, FH, sCR1, MCP-1, eotaxin-1) that, age/APOε4 adjusted, optimally differentiated AD and CTL (AUC: 0.79), and three (sCR1, MCP-1, eotaxin-1) that optimally differentiated AD and MCI (AUC: 0.74). These models replicated in an independent cohort (EMIF; AUC 0.81 and 0.67). Two analytes (FB, FH) plus age predicted MCI progression to AD (AUC: 0.71). Discussion Plasma markers of inflammation and complement dysregulation support diagnosis and outcome prediction in AD and MCI. Further replication is needed before clinical translation.
