In 2009, a new citrus viral disease caused by Citrus yellow vein clearing virus (CYVCV) was first discovered in Yunnan province of China. In this study, a survey was conducted in 27 orchards from Yunnan province from April 2017 to September 2018. In all, 45 of a total of 513 citrus samples were tested positive for CYVCV by reverse transcription polymerase chain reaction (RT-PCR). Furthermore, the complete genome sequences of six CYVCV isolates from different hosts were sequenced. Comparisons of the whole genome sequences of these six CYVCV isolates as well as 34 isolates previously reported from around the world revealed the sequence identity ranged from 96.9% to 99.8% at nucleotide level, indicating that there is a very low level of sequence heterogeneity among CYVCV isolates of different hosts in Yunnan province.

Bu çalışmada, dünyada ilk kez Pakistan ve Hindistan’da, ardından ülkemizde gözlenen Turunçgil sarı damar açılması virus (TSDAV) hastalığının varlığı özellikle hastalığın karakteristik semptomlarını göstermeyen portakal, mandarin ve altıntop gibi turunçgil çeşitlerinde araştırılmıştır. Ayrıca, hastalık etmeninin farklı bitki dokularındaki varlığı da, bu çalışmada moleküler olarak araştırılmıştır. Hastalık etmeninin araştırılmasında moleküler olarak RT-PCR yöntemi, elde edilen bulguları doğrulamak amacı ile de biyolojik indeksleme yöntemleri kullanılmıştır. Çalışma sonucunda limon, turunç, portakal, mandarin ve altıntop türlerine ait yaprak ve sürgün örneklerinin tamamında 479 nt ve 921 nt uzunluğunda band elde edilerek, etmenin bu çeşitlerdeki varlığı belirlenmiştir. Portakal mandarin ve altıntop türlerinde hastalığın varlığı ülkemizde ilk kez bu çalışma ile ortaya konmuştur. Yapılan çalışmalarda, bulaşık ağaçların farklı kısımlarında etmenin varlığı ortaya konmuştur. Nükleotid dizilim analizleri ve yapılan filogenetik çalışmalar sonucunda, farklı turunçgil türlerinden alınan örneklerin, Pakistan, Çin Halk Cumhuriyeti ve ülkemizde daha önce limon örneklerinden yapılan etmen analizleri ile % 97’nin üzerinde bir benzerlik gösterdiği belirlenmiştir. Biyolojik indeksleme çalışmalarında hastalığının karakteristik semptomları olan yaprak deformasyonları, yaprak dokusunda içe ve dışa ceplenmeler, yaprak kenarında V şeklinde girinti, yaprak ana ve yan damarlarında çizgi halinde sarı renk açılmaları ve bantlaşmalar, gondol yaprak oluşumu, yaprakların arka yüzeyinde ıslak su görünümü gözlenmiştir. In this study, the Citrus yellow vein clearing virus (CYVCV) disease, first observed in Pakistan and India and then in our country, is researched among the species of Citrus such as orange, mandarin and grapefruit which don’t show the characteristic symptoms of the disease. Also, the CYVCV searched in different tissue parts of the infected tree in this study. In the investigation of the CYVCV, molecular RT-PCR method and biological indexing method to justify the obtained findings are used. As a result of the study, in all of the examples of leaves and shoots belonging to the species of lemon, sour orange, orange, mandarin and grapefruit, the existence of the virus on these species are identified by obtaining a gel band in the length of 479 bp and 921 bp using with two different primers. The existence of the disease on the symptomless species of orange, mandarin and grapefruit is first determined in this study in Turkey. In the studies, the existence of the virus in different tissues is determined. As a result of the nucleotide analysis and phylogenetic studies, it is identified that the samples taken from different Citrus species showed similarity up 97% in Pakistan, in China and in our country izolates. During biological index studies, leaf deformations, pocket image of inward and outward on leaf tissues, V shape in the edge of leaf, banding and yellow color clearing on the mibrid and lateral veins of the leaf, gondola leaf, shape of wet water on the back side of the leaves are identified it this study. Turunçgil sarı damar açılması virüs (TSDAV) hastalığının yayılış alanlarını belirlemek amacıyla yapılan bu çalışma; Adana ili ticari limon bahçeleri, Ç.Ü. turunçgil üretim ve koleksiyon parselleri ile toplam dört ilçede bulunan ev bahçeleri olmak üzere 3 farklı alanda yürütülmüştür. Sörvey çalışması toplam 8.300 da alanda 134.801 adet turunçgil ağacı, 27.802 adet ev bahçesinde, semt ve bulvarlar üzerinde bulunan 61.178 adet turunç ve limon ağacı olmak üzere toplam 195.979 adet ağaç üzerinde gerçekleştirilmiştir. Hastalığın makroskobik simptomlarının gözlendiği turunçgil çeşitleri enterdonat, kütdiken ve yediveren limon (Citrus limon (L.) Burn.f.), süs bitkisi olarak ve üniversitede koleksiyon parselinde bulunan turunç (Citrus aurantium L.) çeşitleridir. Çalışmada hastalıkla ilgili gözlenen başlıca simptomlar yaprak ana ve yan damarlarında çizgi şeklinde sarı renk açılmaları, yaprak orta damarının büzülmesi, yaprak boyutlarının küçülmesi, yapraklarda içe ve dışa doğru ceplenme, yaprak kenarında ‘V’ şeklinde girinti oluşumu, yaprak kenarlarının kıvrılması, gondol yaprak oluşumu, yaprak alt yüzeyindeki damarlarda su emgisi şeklinde ıslak görünümlerdir. Hastalıktan şiddetli etkilenmiş ağaçlarda geriye doğru kurumalar geliştiği belirlenmiştir. Sörvey çalışması sonucunda ticari bahçelerde bulaşıklık oranı % 0.006, Çukurova Üniversitesi parsellerinde bulaşıklık oranı % 9.46 olarak belirlenmiştir. Hastalık, Adana ili merkezi dışında Yüreğir ilçesinde tek bir bahçede bulunmuştur. Ç.Ü çevresinde ve şehirde 859 adet turunçgil ağacında hastalık belirtileri gözlenmiş ve hastalığın Ç.Ü. sınırları dışına taştığı bu çalışma ile ortaya konmuştur. This study was carried out to determine the spreading areas of Citrus yellow vein clearing virus disease (CYVCV) in three different areas including commercial lemon orchards, citrus production and collection plots of C.U. and backyards around the C.U. in four towns of Adana. Surveys were carried out on 134,801 citrus trees in total of 830 ha, 27,802 of backyards, and total of 61,178 citrus trees on the streets and boulevards. The citrus varieties on which macroscopic symptoms of the disease were observed were Interdonato, Yediveren and Kutdiken (C. limon Burn f.) Lemons and Citrus aurantium L. which is used as rootstock. The main symptoms determined in this study were yellow colored and line shaped discoloration on main and lateral veins, shrinking on main veins of leaves, reducing in leaf size, cubby shape in leaves towards inside and outside, “V” shaped indents on leaf borders, “gondol” shaped leaves, and wet appearance on back vains of leaves. Dryings were also determined on trees effected by the disease severely. As a results of surveys, 0.006 % of infection rate in commercial orchards and 9.46 % of infection in Çukurova Universtiy plots were detected. Except the city center of Adana, the disease was detected in only one orchard in Yuregir town. Symtoms were detected on total of 859 citrus trees in around C. PubMedID: 22913410 Molecular features and genomic organization were determined for Citrus yellow vein clearing virus (CYVCV), the putative viral causal agent of yellow vein clearing disease of lemon trees, reported in Pakistan, India, and more recently in Turkey and China. CYVCV isolate Y1 from Adana, Turkey, was used for deep sequencing analysis of the virusinduced small RNA fractions and for mechanical and graft inoculation of herbaceous and citrus indicator plants. A polyclonal antiserum was developed from CYVCV-Y1 purified from Phaseolus vulgaris and used in western blot assays to characterize the coat protein of CYVCV-Y1 and determine its serological relationship with related viruses. Contigs assembled from the Illumina sequenced short reads were used to construct the whole genome of Citrus yellow vein clearing virus (CYVCV), consisting in a positive-sense RNA of 7,529 nucleotides and containing six predicted open reading frames. The CYVCV genome organization and size resembled that of flexiviruses, and search for sequence homologies revealed that Indian citrus ringspot virus (ICRSV) (Mandarivirus, Alphaflexiviridae) is the most closely related virus. However, CYVCV had an overall nucleotide sequence identity of ?74% with ICRSV. Although the two viruses were similar with regard to genome organization, viral particles, and herbaceous host range, CYVCV caused different symptoms in citrus and was serologically distinct from ICRSV. Primer pairs were designed and used to detect the virus by conventional and quantitative reverse transcription-polymerase chain reaction on yellow vein clearing symptomatic field trees as well as graft- and mechanically inoculated host plants. Collectively, these data suggest that CYVCV is the causal agent of yellow vein clearing disease and represents a new species in the genus Mandarivirus. © 2012 The American Phytopathological Society.
