Ferroptosis is a newly identified form of non-apoptotic programmed cell death, characterized by the iron-dependent accumulation of lethal lipid reactive oxygen species (ROS) and peroxidation of membrane polyunsaturated fatty acid phospholipids (PUFA-PLs). Ferroptosis is unique among other cell death modalities in many aspects. It is initiated by excessive oxidative damage due to iron overload and lipid peroxidation and compromised antioxidant defense systems, including the system Xc - / glutathione (GSH)/glutathione peroxidase 4 (GPX4) pathway and the GPX4-independent pathways. In the past ten years, ferroptosis was reported to play a critical role in the pathogenesis of various cardiovascular diseases, e.g., atherosclerosis (AS), arrhythmia, heart failure, diabetic cardiomyopathy, and myocardial ischemia-reperfusion injury. Studies have identified dysfunctional iron metabolism and abnormal expression profiles of ferroptosis-related factors, including iron, GSH, GPX4, ferroportin (FPN), and SLC7A11 (xCT), as critical indicators for atherogenesis. Moreover, ferroptosis in plaque cells, i.e., vascular endothelial cell (VEC), macrophage, and vascular smooth muscle cell (VSMC), positively correlate with atherosclerotic plaque development. Many macromolecules, drugs, Chinese herbs, and food extracts can inhibit the atherogenic process by suppressing the ferroptosis of plaque cells. In contrast, some ferroptosis inducers have significant pro-atherogenic effects. However, the mechanisms through which ferroptosis affects the progression of AS still need to be well-known. Long intergenic non-coding RNA 00657 (linc00657) is involved in various diseases, whereas its role in atherosclerosis (AS) development remains inconclusive. This study was designed to investigate the effects and underlying mechanisms of linc00657 in atherogenesis. The results showed that ox-LDL treatment significantly induced pyroptosis in human THP-1-derived macrophages. The secretion levels of LDH and pro-inflammatory factors were markedly enhanced, and the integrity of plasma membranes was disrupted in ox-LDL-treated THP-1-derived macrophages. These effects were significantly compensated after transfection with linc00657 siRNA and became more evident by linc00657 overexpression. Moreover, the effects of linc00657 overexpression on pyroptosis of THP-1-derived macrophages can also be robustly reversed by TXNIP knockdown or miR-106b-5p mimics transfection. Mechanistically, linc00657 enhanced TXNIP expression by competitively binding to miR-106b-5p, promoting NLRP3 inflammasome activation. Finally, we found that linc00657 overexpression significantly increased the expression of pyroptosis-related factors and decreased miR-106b-5p level in the aorta of high-fat-diet-fed apoE -/- mice. Furthermore, linc00657 up-regulation enlarged the plaque area, exacerbated plasma lipid profile, and increased pro-inflammatory cytokines levels in the serum, effects that were reversed by injection of miR-106b-5p agomir. Leaf blight outbroke in Rehmannia glutinosa plantation in Wenxian county, Henan province in 2019. R. glutinosa plants with diseased leaves were collected from the plantation, and three strains were isolated from the diseased leaf samples. Pathogenicity test, morphological observation, and phylogenetic analysis of ITS, EF1-α, and Tub suggested that they were respectively Fusarium proliferatum, F. oxysporum, and F.acuminatum. Among them, F. acuminatum, as a pathogen of R. glutinosa leaf disease, had never been reported. To clarify the biological characteristics of F. acuminatum, this study tested the influence of light, pH, temperature, medium, carbon source, and nitrogen source on the mycelial growth rate of the pathogen during a 5-day culture period, and explored the lethal temperature. The results showed that the mycelia grew well under the photoperiod of 12 h light/12 h darkness, at 5-40 ℃(optimal temperature: 25 ℃), at pH 4-11(optimal pH: 7.0), on a variety of media(optimal medium: oatmeal agar), and in the presence of diverse carbon and nitrogen sources(optimal carbon source: soluble starch; optimal nitrogen source: sodium nitrate). The lethal temperature was verified to be 51 ℃(10 min). The conclusion is expected to lay a scientific basis for diagnosis and control of R. glutinosa leaf diseases caused by F. acuminatum.

Background: Cervical cancer remains a major public health issue for the Uyghur women and other women living mainly in rural areas of Xinjiang. This study aims to investigate the distribution of human papillomavirus (HPV) infection and cervical cancer in rural areas of Xinjiang, China., Methods: Cervical cancer screening was performed on rural women aged 35 to 64 years from Xinjiang, China in 2017 through gynecological examination, vaginal discharge smear microscopy, cytology, and HPV testing. If necessary, colposcopy and biopsy were performed on women with suspicious or abnormal screening results., Results: Of the 216,754 women screened, 15,518 received HPV testing. The HPV-positive rate was 6.75% (1047/15,518). Compared with the age 35-44 years group, the odds ratios (ORs) of HPV positivity in the age 45-54 years and 55-64 years groups were 1.18 (95% confidence interval [CI]: 1.02-1.37) and 1.84 (95% CI: 1.53-2.21), respectively. Compared with women with primary or lower education level, the ORs for HPV infection rates of women with high school and college education or above were 1.37 (95% CI: 1.09-1.72) and 1.62 (95% CI: 1.23-2.12), respectively. Uyghur women were less likely to have HPV infection than Han women, with an OR (95% CI) of 0.78 (0.61-0.99). The most prevalent HPV types among Xinjiang women were HPV 16 (24.00%), HPV 33 (12.70%), and HPV 52 (11.80%). The detection rate of cervical intraepithelial neoplasia (CIN)2+ was 0.14% and the early diagnosis rate of cervical cancer was 85.91%. The detection rates of vaginitis and cervicitis were 19.28% and 21.32%, respectively., Conclusions: The HPV infection rate in Xinjiang is low, but the detection rate of cervical cancer and precancerous lesions is higher than the national average level. Objective: Exercise-based intervention promises to be more effective in a structured framework for individuals with autism spectrum disorders (ASD). The aim of this study was to observe changes in behavior of individuals with ASD by investigating their physical status after the structured exercise-based intervention., Patients and Methods: The exercise intervention integrated an 8-week exercise program that included aerobic, resistive, and neuromuscular exercises. Body composition and the Autism Treatment Evaluation Checklist (ATEC) were evaluated to assess changes after the exercise-based intervention., Results: After the exercise intervention, the fat mass of individuals with ASD were significantly reduced, and their behavior improved markedly., Conclusions: This pilot study demonstrated that individuals with ASD require long-term, structured exercise-based intervention, and that such exercise-based intervention is effective for improving their health.

Litter accumulation can strongly affect seedling emergence and early growth through both physical and chemical mechanisms, and can further influence natural regeneration. Chinese fir (Cunninghamia lanceolata) is one of the most important afforestation tree species. Its natural regeneration is poor, possibly due to the thick leaf accumulation inhibiting seedling emergence and growth. We used natural and plastic litter to study the effects of Chinese fir litter on its own seedling emergence and early growth, as well as to assess whether the effect was physical or chemical. In this experiment, two litter types (natural and plastic litter) and four different litter amounts (control, 0 g·m -2 ; low, 200 g·m -2 ; medium, 400 g·m -2 ; high, 800 g·m -2 ) were used. The results showed that compared to the control (0 g·m -2 ), low litter amount (200 g·m -2 ) exerted a slight positive effect on seedling emergence, whereas high litter amount (800 g·m -2 ) significantly reduced the seedling emergence and survival rate in the case of both natural and plastic litter. With increasing litter amount, root length of seedlings decreased and stem length increased. The highest and lowest root mass, leaf mass, and total mass of seedlings were observed for the low and high litter amount, respectively, in the case of both natural and plastic litter. The root:shoot ratio of seedlings decreased with the increasing litter amount for both natural and plastic litter. The photosynthesis:non-photosynthesis biomass ratio of the seedlings was higher under all litter cover treatments, compared to that in the control. Because the effect of the same amounts of the natural and plastic litter on seedling emergence and early growth did not differ, the litter layer's short-term influence was primarily physical. As the litter cover increased, the initial slight positive effects on seedling emergence and early growth could shift to inhibitory effects. Moreover, to penetrate the thick litter layer, Chinese fir seedlings allocated more resources toward stems and aboveground growth. This study provided evidence for litter amount being a key ecological factor that affects the seedling development and subsequent natural regeneration of Chinese fir.

Background: Primary colorectal lymphoma (PCL) is a rare colorectal malignancy. The standard treatment and prognostic factors of PCL remain unexplored. Therefore, a large population-based study should be conducted to provide a detailed review of this disease., Methods: We extracted the data of eligible patients with PCL registered in the SEER database from 1973 to 2011. All statistical analyses were performed using SPSS 19.0., Results: A total of 2050 (61.3%) of the 3342 patients with PCL underwent surgical intervention, and 1292 (38.7%) patients received no surgical treatment. The median overall survival was 95 months, and patients receiving surgery exhibited significantly prolonged survival (adjusted HR =0.69, P <0.001). Young age, early tumor stage, and indolent lymphoma were independent predictors of improved survival. Further survival analyses demonstrated the potential benefit of surgery in patients with early tumor stage, right-sided lesions, or diffuse large B-cell PCL. Conversely, surgical intervention did not improve the survival of patients with advanced-stage, left-sided, or indolent PCL., Conclusion: PCL is a rare tumor that can be effectively treated. Surgical intervention may play an important role in the treatment of PCL. Early tumor stage, a right-sided lesion, and diffuse large B-cell histological PCL seem to be the clinical characteristics of optimal surgical candidates.

A series of new silicon(IV) phthalocyanines (SiPcs) di-substituted axially with different nucleoside moieties have been synthesized and evaluated for their singlet oxygen quantum yields (ΦΔ) and in vitro photodynamic activities. The adenosine-substituted SiPc shows a lower photosensitizing efficiency (ΦΔ=0.35) than the uridine- and cytidine-substituted analogs (ΦΔ=0.42-0.44), while the guanosine-substituted SiPc exhibits a weakest singlet oxygen generation efficiency with a ΦΔ value down to 0.03. On the other hand, replacing axial adenosines with chloro-modified adenosines and purines can result in the increase of photogenerating singlet oxygen efficiencies of SiPcs. The formed SiPcs 1 and 2, which contain monochloro-modified adenosines and dichloro-modified purines respectively, appear as efficient photosensitizers with ΦΔ of 0.42-0.44. Both compounds 1 and 2 present high photocytotoxicities against HepG2 and BGC823 cancer cells with IC50 values ranging from 9nM to 33nM. The photocytotoxicities of these two compounds are remarkably higher than the well-known anticancer photosensitizer, chlorin e6 (IC50=752nM against HepG2 cells) in the same condition. As revealed by confocal microscopy, for both cell lines, compound 1 can essentially bind to mitochondria, while compound 2 is just partially localized in mitochondria. Unravelling the unique effects of metal coordination on biological behaviours is of importance to design metal based therapeutic and diagnostic agents. In this work, we chose luminescent Znsalen ( ZnL 1 ) as a case study to demonstrate that metal induced aggregation arising from the intermolecular Zn···O interaction influences its cellular uptake and subcellular localization. Comparative studies with the free bases ( L 1 and L 2 ) show that ZnL 1 undergoes cellular uptake through caveolae-mediated endocytosis and internalizes in endosomal/lysosomal compartments, in contrast to the localization of L 1 and L 2 in the mitochondria. Further studies of photophysical properties, TEM imaging and DLS analysis suggest that ZnL 1 tends to form large sized fibrous structures in aqueous media. To investigate the relationship between ZnL 1 aggregation and the biological behaviour, we used pyridine to tune the "aggregation-to-deaggregation" transition and found that, in the presence of pyridine, ZnL 1 could localize in the mitochondria and internalize into cells through the passive diffusion pathway. Such distinctive biological behaviours resulting from the different Znsalen species clearly point out the importance of metal induced aggregation or metal speciation analysis in designing metal complexes as biological probes.

Primary Objective: Lipopolysaccharide (LPS) is known to alter the integrity of the blood-brain barrier (BBB) in sepsis, although the underlying mechanism remains unknown. The aim of this study was to elucidate the molecular mechanisms underlying disruption of the BBB in LPS-induced sepsis., Research Design: Both in vitro and in vivo experiments were designed to test the role of AMP-activated protein kinase (AMPK) in LPS-induced BBB dysfunction., Methods and Procedures: Human brain microvascular endothelial cells (HBMECs) were cultured. The protein expressions were detected by western blot. BBB integrity was determined by Evans Blue., Main Outcomes and Results: LPS (1 μg ml(-1)) dramatically increased the permeability of the BBB and the ROS productions, as well as reducing the expression levels of occludin and claudin-5 in cultured HBMECs. Inhibition of NAD(P)H oxidase by apocynin or up-regulation of AMPK reversed the LPS-induced abnormities in HBMECs. In LPS-induced sepsis in mice, it was found that LPS dramatically increased NAD(P)H oxidase protein expressions and ROS productions in the brain and disrupted BBB function assayed by Evans blue staining, which were abolished by AICAR treatment., Conclusions: It is concluded that AMPK activation improves the functions of the BBB impaired by LPS through suppression of NAD(P)H oxidase-derived ROS in mice.

This paper quantifies the allocation of ecosystem services value (ESV) associated with land use pattern and qualitatively examined impacts of land use changes and socio-economic factors on spatiotemporal variation of ESV in the Natural Wetland Distribution Area (NWDA), Fuzhou city, China. The results showed that total ESV of the study area decreased from 4,332.16 × 10(6) RMB Yuan in 1989 to 3,697.42 × 10(6) RMB Yuan in 2009, mainly due to the remarkable decreases in cropland (decreased by 55.3 %) and wetland (decreased by 74.2 %). Forest, water, and wetland played major roles in providing ecosystem services, accounting for over 90 % of the total ESV. Based on time series Landsat TM/ETM+ imagery, geographic information system, and historical data, analysis of the spatiotemporal variation of ESV from 1989 to 2009 was performed. It indicated that rapid expansion of urban areas along the Minjiang River resulted in significant changes in land use types, leading to a dramatic decline in ecosystem services. Meanwhile, because of land scarcity and unique ecosystem functions, the emergency of wetland and cropland protection in built-up area has become an urgent task of local authorities to the local government. Furthermore, there was still a significant negative correlation between ESV of cropland and wetland and the GDP. The results suggest that future planning of land use pattern should control encroachment of urban areas into cropland and wetland in addition to scientific and rational policies towards minimizing the adverse effects of urbanization.

Myoglobins provide an opportunity to investigate the effect of the secondary coordination sphere on the functionality and reactivity of non-native metal porphyrins inside well-defined protein scaffolds. In this work, we reconstituted myoglobin by the replacement of natural heme with manganese(iii) protoporphyrin IX and firstly investigated the effect of distal histidine on the reaction of Mn(III) porphyrin with H2O2 and one-electron oxidation of ABTS. We have prepared L29H, F43H, H64F, L29H/H64F, F43H/H64F, L29H/F43H and L29H/F43H/H64F mutants and reconstituted apo-myoglobins with manganese(iii) protoporphyrin IX. Distal histidine at the 64 position plays an essential role in binding H2O2 through hydrogen bond formation, which facilitates the coordination of H2O2 to the Mn center. The second histidine at the 43 position is important in the cleavage of the O-O bond and to form the highly valent Mn(iv)-oxo intermediate. His29 has less efficiency to activate H2O2, because it is too far from the Mn center. The cooperative effect of dual distal histidines at positions 64 and 43 on the activation of H2O2 was observed and the F43H Mn(III)Mb mutant exhibited 5-fold and 10-fold reaction rate increases in the activation of H2O2 and one-electron oxidation of ABTS versus wild-type Mn(III)Mb. This is different from the distal histidine effect on the H2O2 activation by heme in Mb. This work will provide new insights to understand the fundamental chemistry of manganese in oxidation, and further construct biomimetic Mn models for peroxidase, inside or outside of protein scaffolds.

μ-Hydroxyl trinuclear copper(II) compounds, with core structures closely resembling the central moieties in multicopper oxidases, have been extensively investigated and the importance of the prototypes in the oxidation of C-H bonds has been confirmed. In the course of study for the Cu(II) salens catalyzing the three-component (sodium azide, epoxide and non-activated terminal alkyne) 1,3-Huisgen cycloaddition, we observed the unusual formation of a μ-OH bridging trinuclear copper cluster supported by three (R,R)-N-(5-sulfonatosalicylidene)-1,2-diaminocyclohexane ligands (Cu-6) during the catalytic process. More interestingly, through comparative studies for the activity of the mononuclear copper(II) complexes (Cu-1 and Cu-5), we found that the μ-OH trinuclear copper(II) complex (Cu-6) was the active catalyst precursor. Through analysing the reaction intermediates we found that Cu(II) was partially reduced to Cu(I) and an adduct formed during catalytic process, which involves the alkyne coordinating to the Cu centers. This is the first example where a μ-OH trinuclear copper(II) complex has been isolated in copper catalyzed reactions other than catalytic oxidations, which provides a valuable clue to investigate that μ-OH copper clusters could be the intermediates in copper catalysed reactions in aqueous media.

"Synergistic effect" is prevalent in natural metalloenzymes in activating small molecules, and the success has inspired the development of artificial catalysts capable of unprecedented organic transformations. In this work, we found that the attractive π-π interaction between organic additives (as electron-donors) and the perfluorinated arenes (as electron acceptors) is effective in gold hydride catalyzed activation of C-F bonds, specifically hydrodefluorination (HDF) of perfluoroarenes catalyzed by the Sadighi's gold hydrides [(NHC)AuH] (NHC = N-heterocyclic carbene). Although a weak interaction between [(NHC)AuH] and perfluoroarenes was observed from (1)H NMR and UV-vis spectroscopies, low reactivity of [(NHC)AuH] toward HDF was found. In contrast, in the presence of p-N,N-dimethylaminopyridine (DMAP), the HDF of perfluoroarenes with silanes can be efficiently catalyzed by [(NHC)AuH], resulting in mainly the para-hydrodefluorinated products with up to 90% yield and 9 turnovers. The yield of the reaction increases with the more electron-withdrawing groups and degree of fluorination on the arenes, and the HDF reaction also tolerates different function groups (such as formyl, alkynyl, ketone, ester, and carboxylate groups), without reduction or hydrogenation of these function groups. To reveal the role of DMAP in the reactions, the possible π-π interaction between DMAP and perfluoroarenes was suggested by UV-vis spectral titrations, (1)H NMR spectroscopic studies, and DFT calculations. Moreover, (1)H and (19)F-NMR studies show that this π-π interaction promotes hydrogen transfer from [(NHC)AuH] to pyridyl N atom, resulting in C-F bond cleavage. The interpretation of π-π interaction assisted C-F activation is supported by the reduced activation barriers in the presence of DMAP (31.6 kcal/mol) than that in the absence of DMAP (40.8 kcal/mol) for this reaction. An analysis of the charge distribution and transition state geometries indicate that this HDF process is controlled by the π-π interaction between DMAP and perfluoroarenes, accompanied with the changes of partial atomic charges.

The leaching of trace elements from tailings of an antimony mine in Guangxi Autonomous Region, China, was investigated through column leaching under wet-dry cycling and complete immersion conditions. Simulated acid rain (pH 4.0-4.4) and river water (pH 8.0) were used as the leaching solution. No matter the simulated acid rain or river water was used, the leachate always showed a slightly alkaline pH between 7.2 and 8.0, suggesting an acid neutralization capacity of the tailing. Compared to As and Pb, Sb was leached out to a much higher extent in this circumstance. Furthermore, Sb release was largely enhanced in wet-dry cycle compared to the complete immersion condition. In contrast, As was leached more readily in the complete immersion condition, and the longer the tailings were immersed in water, the higher the As concentration in the leachate. The leachate on day 5 and day 10 showed 1-2 times higher As concentration as compared with the leachate on day 1 and day 2. The leaching of Mn and Zn by simulated acid rain was much stronger than that by river water, and the release of Mn and Zn was more significantly affected by pH than by O2 (i.e., the difference between the wet-dry cycle and complete immersion condition). Sr showed a high release rate that was not affected by leaching solution or air-exposure condition. Basically, Pb showed a very low leaching potential. In general, an alkaline circumstance combined with wet-dry cycle forms the favorable condition for the release of Sb in the tailings.

There is widespread interest in non-covalent bonding and weak interactions, such as electrostatic interactions, hydrogen bonding, solvophobic/hydrophobic interactions, metal-metal interactions, and π-π stacking, to tune the molecular assembly of planar π-conjugated organic and inorganic molecules. Inspired by the roles of metal-aromatic interaction in biological systems, such as in ion channels and metalloproteins, herein, we report the first example of the use of Hg(2+) -aromatic interactions to selectively control the assembly and disassembly of zinc-salen complexes in aqueous media; moreover, this process exhibited significant "turn on" fluorescent properties. UV/Vis and fluorescence spectroscopic analysis of the titration of Hg(2+) ions versus complex ZnL(1) revealed that the higher binding affinity of Hg(2+) ions (compared to 13 other metal ions) was ascribed to specific interactions between the Hg(2+) ions and the phenyl rings of ZnL(1); this result was also confirmed by (1)H NMR spectroscopy and HRMS (ESI). Further evidence for this type of interaction was obtained from the reaction of small-molecule analogue L(1) with Hg(2+) ions, which demonstrates the proximity of the N-alkyl group to the aromatic protons during Hg(2+)-ion binding, which led to the consequential H/D exchange reaction with D(2) O. DFT modeling of such interactions between the Hg(2+) ions and the phenyl rings afforded calculated distances between the C and Hg atoms (2.29 Å) that were indicative of C-Hg bond-formation, under the direction of the N atom of the morpholine ring. The unusual coordination of Hg(2+) ions to the phenyl ring of the metallosalen complexes not only strengthened the binding ability but also increased the steric effect to promote the disassembly of ZnL(1) in aqueous media., (Copyright © 2012 WILEY-VCH Verlag GmbH & Co. In the area around an antimony-lead smelter, soil and soil water samples were collected from different soil profiles in vegetable field, uncultivated field and wood field. The distribution and migration of heavy metals in soil profile were investigated. In soil, Sb, Pb, Cu and Cd are found to accumulate in the topsoil layer, and decrease rapidly with the depth. However, Sb, Pb, and As appear to accumulate below 60 cm. Sb and As show a much stronger affinity to the pore water than Pb. In consequence, different soil utilization types lead to different effects of heavy metals migration in soil profile. Furthermore, Sb and As show a higher potential to migrate along the soil profile than that of Pb. In addition, the high content of heavy metals in the topsoil may be absorbed by crops post to desorption in soil water, and possibly form a threat to human health.

The individual signaling pathways underlying cardiac hypertrophy, which is induced by either α or β adrenergic receptor (AR), are different. Activation of different AR subtypes couples to different G proteins and induction of specific signaling pathways, which ultimately results in subtype-specific regulation of cardiac function. We present the first proteomics study identifying proteins that are related to AR subtype-specific hypertrophy in cardiomyocytes by comparing the two-dimensional electrophoresis patterns between neonatal rat cardiomyocytes treated by phenylepinephrin (PE) and by isoproterenol (ISO). An improved 2-DE strategy was used in these comparative experiments. Twenty-five differentially expressed proteins in cardiomyocytes treated by PE or treated by ISO were successfully analyzed and identified using matrix-assisted laser desorption/ionization-time of flight mass spectrometry, especially those that might be responsible to intracellular oxidative stress such as dismutase, peroxiredoxin, and thioredoxin-like protein p46. In addition, induced reactive oxygen species were also found to be AR subtype-specifically relevant to endoplasmic reticulum proteinase ERK1/2 phosphorylation during the development of hypertrophy induced by different AR subtypes. The results will help to better understand the underlying mechanisms of different adrenergic receptor subtype-induced hypertrophy.

Near infrared spectroscopy technique combined with chemometrics methods was applied to predict crystallinity of Neosinocalamus affinins. Three improved partial least squares (PLS) methods, including interval partial least squares (iPLS), synergy interval partial least squares (siPLS) and backward interval partial least squares (biPLS), were used to find the most informative ranges and build models with better predictive quality based on multiplicative scatter correction spectra. And then the models were compared with PLS model which was developed on the whole wavelength range 350-2 500 nm. The results showed that the models built by the three improved PLS methods had higher predictive ability than that of PLS model, and the optimal model was obtained by siPLS method that separated the whole spectra into 30 intervals and combined three intervals. The siPLS model had correlation coefficient (R) of 0.88 and root mean standard error of prediction (RMSEP) of 0.011 7. Therefore, through selecting the effective wavelength range, siPLS method could accurately and rapidly predict crystallinity in Neosinocalamus affinins.

Hsp70s have been shown to play important roles in helping cells to cope with adverse environments, especially in response to temperature. In this study a novel ethephon-induced Hsp gene, designated as HbHsp70, was isolated from Hevea brasiliensis. The HbHsp70 cDNA contained a 1965 bp open reading frame encoding 655 amino acids. The deduced HbHsp70 protein showed high identities to Hsp70s from other plants. Expression studies revealed more significant accumulation of HbHsp70 transcripts in leaves and stems than in roots, barks and latex. The transcription of HbHsp70 was induced by ethephon, heat treatment and low temperature stress, whereas jasmonic acid had little effects. Recombinant HbHsp70 was expressed in Escherichia coli and purified by Ni-NTA affinity chromatography. Measuring the light scattering of luciferase (Luc) revealed that HbHsp70 prevents the aggregation of luc during high-temperature stress. In vitro experiments showed that HbHsp70 had protective functions not only against heat stress but also against chilling stress. All these data suggest that HbHsp70 may play roles in responses to heat shock and low temperature in H. brasiliensis.

Objective: To study the cytologic features of pancreatobiliary tumors in endoscopic retrograde cholangiopancreatography (ERCP)-guided brushing preparations and to evaluate the usefulness of cytology in the diagnosis of pancreatobiliary malignancy., Methods: A retrospective analysis of 212 cases of ERCP-guided brush cytology smears performed during the period from January, 2004 to December, 2006. The cytologic diagnosis was confirmed either by the histologic diagnosis or the strict clinical criteria., Results: Two of the cases studied were unsatisfactory for diagnosis, with no epithelial cells identified. One hundred and thirty-seven smears were diagnosed as "negative", 45 of which subsequently confirmed to be malignant (negative predictive value = 60.2%). Six of the 11 cases with "low-grade atypia" were proven to be malignant (positive predictive value = 54.5%), as compared to 19 of 23 cases of "high-grade atypia" (positive predictive value = 86.4%). All of the 41 cases with cytologic diagnosis of "malignancy" were confirmed to be malignant (positive predictive value = 100%). The cytologic features of malignancy in ERCP-guided brushing preparations included overlapping nuclei, anisonucleosis, coarse chromatin pattern, poor cellular cohesion, tumor diathesis, prominent nucleoli and atypical mitotic figures., Conclusions: The accuracy of ERCP-guided brush cytology relies on good specimen preparation and application of morphologic criteria. Grading of cytologic atypia is of clinical significance. A "negative" or "low-grade atypia" cytologic diagnosis requires further diagnostic workup to rule out the possibility of underlying malignancy, while a "high-grade atypia" or "malignant" diagnosis is relatively specific in guiding subsequent management of suspected pancreatobiliary malignancy.

A metabolic pathway for the biodesulfurization of model organosulfur compounds e.g., dibenzothiophene (DBT), is proposed. This pathway, defined as extended 4S pathway, incorporates the traditional 4S pathway with the methoxylation pathway from 2-hydroxybiphenyl (HBP) to 2-methoxybiphenyl (2-MBP). The formation of 2-MBP was confirmed by the gas chromatography-mass spectrometry (GC-MS) analysis. A similar pathway was also obtained in the desulfurization of 4,6-dimethyldibenzothiophene (4,6-DMDBT), confirming the methoxylation reaction in the desulfurization process by the Mycobacterium sp. strain. Compared with 2-HBP, 2-MBP has much slighter inhibition effect on the cell growth and desulfurization activity. Thus, the methoxylation pathway from 2-HBP to 2-MBP would make less inhibitory effect on the microbe. The new pathway with 2-MBP as the end product may be an alternative for the further desulfuration of the fossil fuels.

The effect of 2-hydroxybiphenyl (2-HBP), the end product of dibenzothiophene (DBT) desulfurization via 4S pathway, on cell growth and desulfurization activity was investigated by Microbacterium sp. The experimental results indicate that 2-HBP would inhibit the desulfurization activity. Providing 2-HBP was added in the reaction media, the DBT degradation rate decreased along with the increase of 2-HBP addition. By contrast, cell growth would be promoted in the addition of 2-HBP at a low concentration (<0.1mM). At high concentration of 2-HBP, the inhibition on the cell growth occurred. Meanwhile, the inhibitory effect of 2-HBP on DBT desulfurization activity was tested both in the oil/aqueous two-phase system and the aqueous system. A mathematical model was developed to explain the product formation kinetics with DBT as the sole sulfur source. The predicted results were close to the experimental data, it elucidated that along with the 2-HBP accumulation, the inhibitory effect of 2-HBP on DBT desulfurization and cell growth was enhanced.

A new isolated dibenzothiophene (DBT) desulfurizing bacterium, identified as Mycobacterium sp. ZD-19 can utilize a wide range of organic sulfur compounds as a sole sulfur source. Thiophene (TH) or benzothiophene (BTH) was completely degraded by strain ZD-19 within 10h or 42 h, and 100% DBT or 4,6-dimethyldibenzothiophene (4,6-DMDBT) was removed within 50h or 56 h, respectively. Diphenylsulfide (DPS) possessed the lowest desulfurization efficiencies with 60% being transformed within 50h and 80% at 90 h. The desulfurization activities of five substrates by resting cells are in order of TH>BTH>DPS>DBT>4,6-DMDBT. In addition, when DBT and 4,6-DMDBT were mixed, they could be simultaneously desulfurized by strain ZD-19. However, DBT appeared to be attacked prior to 4,6-DMDBT. The desulfurization rate of DBT or 4,6-DMDBT in mixture is lower than they are desulfurized separately, indicating that the substrate competitive inhibition is existent when DBT and 4,6-DMDBT are mixed.

Background & Objective: Postoperative tissue adherence, scarring and radiotherapy often lead to extrinsic compression and stricture in the distal ureter of the patients who had history of pelvic malignancies. Our aim was to evaluate the efficacy and safety of endourologic techniques in treating this kind of ureteral obstruction., Methods: From Jan. 1998 to Mar. 2007, 46 patients with obstruction in the distal ureter and had history of pelvic malignancies underwent endoscopic treatments at the Third Affiliated Hospital of Sun Yat-sen University for relief of the obstruction. Perioperative and follow-up data were analyzed., Results: Of the 46 patients, 25 underwent laparoscopic ureterolysis and uretero-neocystostomy, 18 underwent placement of ureter stent under ureteroscope, 3 underwent percutaneous nephrostomy. No severe complication was recorded. The mean operating time was 82.5 min (range, 30-140 min). The mean blood loss was 45.5 ml (range, 5-180 ml). No blood transfusion was needed. The median follow-up time was 18.2 months (range, 3 months to 6.5 years). Three months after operation, B-ultrasonography and intravenous urography (IVU) showed that 39 (84.8%) patients had recovered normal renal function, the other 7 (15.2%) had hydronephrosis relief and renal function improvement. Nuclear renal scanning showed that the mean postoperative glomerular filtration rate (GFR) in the obstructive kidney was higher than the preoperative level (37.6 ml/min vs. 21.3 ml/min, P<0.05). No stricture in the uretero-bladder anastomotic stoma was recorded., Conclusion: Endoscopic operation is an effective and feasible option for managing some selected kinds of distal ureteral obstruction caused by postoperative tissue adherence and radiotherapy in the patients with history of pelvic malignancies.

Objective: To evaluate the prognostic factors for the local recurrence of osteosarcoma in extremities treated with combined therapy., Methods: Between 1992 and 2001, 189 patients with non-metastatic osteosarcoma were treated. The average age at diagnosis was 18 years old. Tumors were located in the distal femur in 86 patients, proximal tibia in 52, and other locations in 51. Pathologic fracture occurred in 22 cases. One hundred and sixteen patients received combined therapy, 73 patients underwent non-combined therapy (tumor resection only or unfinished chemotherapy). In the combined therapy group, 90 patients underwent limb salvage surgery and 26 patients received amputations. In the non-combined therapy group, 42 patients underwent limb salvage surgery and 31 patients received amputations. With Chi-Square test, local recurrence rates were retrospectively evaluated in relation to gender, age, tumor site, pathologic fracture, serum level of alkaline phosphatase, protocol of chemotherapy, type of surgery, surgical margin, size of tumor, and postoperative chemotherapy. Multiple factors were analyzed by Logistic regression analysis., Results: The 5-year-survival rate was 78.5% in the combined therapy group and 35.1% in the non-combined therapy group. Local recurrence rate was 16.6% (15 cases) in limb salvage surgery with combined therapy group (90 cases) and 38.1% (16 cases) in limb salvage surgery with non-combined therapy group (42 cases) (P<0.001). Metastasis rate was 28.4% (33 cases) in the combined therapy group (116 cases) and 65.7% (48 cases) in the non-combined therapy group (73 cases) (P<0.000). The local recurrence rate was significantly related to serum level of alkaline phosphatase after preoperative chemotherapy, type of surgery, surgical margin and chemotherapy protocol. Moreover, Logistic regression revealed that surgical margin and serum level of alkaline phosphatase after preoperative chemotherapy were the key factors affecting the local recurrence., Conclusions: The local recurrence is related to the surgical margin. The adequate surgical margin is very important for the limb salvage. The local recurrence is highly related to metastasis. The inadequate surgical treatment would damage the survival of patients with osteosarcoma.

Objective: To discuss the indications of reconstruction with total femoral prosthesis for the patients with a majority of femur infiltrated by malignant bone tumor, and to evaluate the functional outcome and complication., Methods: Between October 1996 and October 2002, 17 patients with malignant bone tumor were treated with total femoral prosthesis replacement. The patients included 8 males and 9 females, whose age was from 12 to 34 years, with the average of 16 years. The lesions were located in the proximal femur metaphysis in 1, the distal femur metaphysis in 6, the femur shaft in 10, pathologic fracture in 2, skip lesions in 3. The extent of lesions was 23-28 cm (a majority of femur infiltrated by malignant bone tumor). Preoperative pathologic diagnosis were established by open biopsy (2 cases) or needle biopsy (15 cases). All patients were osteosarcoma. According to Enneking surgical staging system, the cases were 12 IIB and 5 IIIB. Seventeen cases received preoperative chemotherapy and 15 cases received postoperative chemotherapy., Results: Seventeen cases were followed up with a mean time of 45 months (range 9-120 months). Local recurrence was observed in 3 (17.6%) after operation 6-14 months. In 12 IIB cases, 4 cases (33%) developed pulmonary metastasis and died. Eight cases remained continuously disease-free for average 75 months (range 50-120 months). In 5 IIIB cases, all with a mean survived term of 13 months (range 9-20 months) died. Fifteen patients (88%) could walk. The patients had maintained average 74% (range 40% - 93%) limb function scores evaluated by ISOLS criteria. Four cases had complications of paralysis of common peroneal nerve (3 cases) and dislocation of hip joint (2 case). No infection took place., Conclusions: Total femoral prosthesis replacement could be used in the treatment of the patients with a majority of femur infiltrated by malignant bone tumor and could effectively recover their limb function to a great extent. The procedure can effectively improve the quality of life for the patients with malignant bone tumor staging IIIB.

Aim: To examine the impact and prognostic significance of alpha-tocopherol associated protein (TAP) expression in a series of prostate cancer patients., Methods: Tissues from 87 patients underwent radical prostatectomy were examined for TAP expression by immunohistochemistry. The relationships of the staining results, the clinic pathological characteristics and the recurrence times were analyzed., Results: Compared with the adjacent areas of normal and benign glands, immunoreactivity of TAP was reduced in areas of prostate cancer. A lower TAP-positive cell number per mm(2) of the largest cancer area (defined as TAP-PN) was associated with higher clinical stage (r = -0.248, P = 0.0322). Inverse associations were found among the TAP-PN and positive lymph nodes (r = -0.231, P = 0.0325), preoperative prostate-specific antigen (PSA) levels (r = -0.423, P = 0.0043), tumor size (r= -0.315, P= 0.0210) and elevated tumor cell proliferation, which was indicated by the staining of Ki-67 (r = -0.308, P = 0.0026). TAP-PN was a significant predictor of recurrence univariately (P = 0.0006), as well as multivariately, adjusted for known markers including preoperative PSA, clinical stage, Gleason score, surgical margin, extra-prostatic extension, seminal vesicle invasion and lymph node metastasis (P = 0.0012)., Conclusion: Reduced expression of TAP was associated with the cell proliferation status of prostate cancer, adverse pathological parameters and the increased risk of recurrence.

Objective: To observe the effect of resolving phlegm method (RPM) on fibrinolytic status in non-alcoholic steatohepatitis (NASH) patients of phlegm blood-stasis (PBS) syndrome type., Methods: Sixty-two patients were randomly assigned to 2 groups, namely the treated group (n = 34) treated with Chinese herbs for resolving phlegm and dampness, smoothing liver and gallbladder, promoting blood circulation and removing blood stasis (composition: Indigo Naturalis 10 g, Alumen 3 g, Semen Cassiae 15 g, Fructus Crataegi 15 g, vinegar prepared Radix Bupleuri 10 g, Radix Curcumae 10 g, Radix Salviae Miltiorrhizae 12 g, Herba Lycopi 12 g, talc 12 g and Radix Glycyrrhizae 2 g), one dose twice per day orally, and the control group (n = 28) with ursodeoxycholic acid tablet (UAT) 150 mg three times per day. The treatment course for both groups was 12 weeks. Liver function (ALT, AST, ALP), insulin resistance (IR) index represented by homeostasis model-IR (HOMA-IR) and insulin (Ins), levels of tissue plasminogen activator (t-PA) and plasminogen activator inhibitor-1 (PAI-1), as well as main symptoms and physical signs were assessed before and after treatment., Results: (1) Compared with those before treatment, levels of ALT, AST, HOMA-IR, t-PA, PAl-1 and main symptoms were all improved in the treated group (P < 0.05 or P < 0.01); (2) There was significant difference in ALP, HOMA-IR, t-PA and PAI-1 between the treated group and the control group (P < 0.05 or P < 0.01); (3) The improvement in liver function related indexes in the control group was mainly the level of ALT, t-PA (P < 0.01), and also shown on HOMA-IR, Ins and PAI-1., Conclusion: RPM could effectively improve liver function and fibrinolytic status. Its effect was better than that of UAT.

Objective: To evaluate the survival rates and complications of combined therapy for primary osteosarcoma of the extremities between 1992 and 2001., Methods: From 1992 to 2002, 189 patients with non-metastatic osteosarcoma were treated. Their average age at diagnosis was 18 years old (ranging from 4 to 39). Tumors were located at distal femurs in 86 patients, proximal femurs in 4, proximal tibia in 52, proximal humors in 19 and other locations in 28. Combined therapy (resection of tumor with chemotherapy) was given 116 patients and non-combined therapy (tumor resection only and unfinished chemotherapy) to 73. In combined therapy group, 90 patients underwent limb salvage surgery, and 26 patients received amputations. In non-combined therapy group, 42 patients underwent limb salvage surgery and 31 patients received amputations., Results: The 5-year-survival rates were 78.5% in the combined therapy group and 35.1% in the non-combined therapy group. Local recurrence rate was 16.6% (15 cases) in limb salvage surgery with combined therapy group (90 cases) and 38.1% (16 cases) in limb salvage surgery with non-combined therapy group (42 cases) (P < 0.007). Metastasis rate was 28.4% (33 cases) in the combined therapy group (116 cases) and 65.7% (48 cases) in the non-combined therapy group (73 cases) (P < 0.000). Functional evaluation showed that the patients who underwent salvage surgery had higher functional scores than those who had an amputation. According to MSTS scoring system (1993), 56 patients (excision alcohol replacement 36 cases, allograft 10 cases and prosthesis 10 cases) were evaluated functional scores. The average scores reached same level (P > 0.5)., Conclusions: (1) In this study, the 5-year-survival rate in the combined therapy is improved. The treatment for the patients with metastasis in lung is very difficult for improvement of the 5-year-survival rate. (2) The local recurrence is related the surgical margin. The adequate surgical margin is very important for the limb salvage. (3) The combined therapy is very essential for improving survival rate.

Objective: To evaluate the functional outcome and the complications of allograft replacement in management of giant cell tumors of bone., Methods: Seventy-seven patients who underwent bone tumor resection and massive allograft reconstruction of bone defects between 1992 and 2002 were evaluated. The length of the resected part ranged from 5 to 11 centimeters. Fresh-frozen allografts were employed as osteoarticular grafts (n = 47), hemi-condylar (n = 12), massive (n = 14) or allograft-prosthesis composite (n = 4). Most of the lesions located in proximal and distal femur, proximal tibia and humerus. The oncological parameters that were evaluated including survival of the patient, local recurrence, and metastasis. The radiographic parameters included time to union, stability of the joint, fracture of the allograft. Mankin evaluation system was used to assess functional outcome., Results: At a median of 35 months (range from 12 to 135 months) after the operation, 76 of the patients in the study group were free of disease, and one had died of disease. Eleven (14.1%) patients had local recurrence and 12 (15.4%) nonunion. Late complications included 5 (6.4%) fractures of the allograft and 5 (6.4%) infections of the graft. Instability of the joint in the form of subluxation was noted in 5 (6.4%) patients. One extremity were amputated due to local recurrence. On the basis of Mankin functional evaluation, the total satisfied rate was 83.2 percent. Osteoarticular graft got the highest score (91.5%) and hemi-condylar got the lowest (66.6%). Massive grafts and composite were at the middle (ranges from 71.4% to 75%)., Conclusion: Allografts offer many types of reconstruction for bony defects after tumor resection. The functional results are comparable to other methods of reconstruction, and once incorporated by the host, offer the advantage of longevity. It is also a method with high complication, and decreasing the complications could improve the functional score. Less resection could get better reconstruction and better function, less margin resulted at the same time. The risk of recurrence increases when less surgical margin achieves. The main end-result-influencing factor is local recurrence.

Objective: To explore the effects of different anesthesia Methods on the cellular immune function and mean arterial pressure, heart rate in patients with abdominal tumor in the peri-operational period., Methods: Patients were anesthetized by general anesthesia (GA), combined acupuncture-general anesthesia (AGA), combined epidural-general anesthesia (EGA), and combined acupuncture-epidural-general anesthesia (AEA), respectively. Changes of T lymphocyte subsets and hemodynamics were observed at different time points before and after treatment., Results: Patients' cellular immune function after GA was lowered, which could be alleviated by combination of GA with acupuncture anesthesia (AA) or with epidural anesthesia. Combined acupuncture-drug anesthesia showed a stabilizing effect on hemodynamics in peri-operational period to some extent., Conclusion: EGA and AGA are good anesthesia choice in clinical practice, which would be favorable for early recovery of immune function in patients with abdominal tumor after operation.

Objective: To investigate the way of diagnosis and therapy of parosteal osteosarcoma., Methods: A retrospective review was conducted of 48 patients treated at our department between June 1964 and December 2001. The average age of patients in this study was 29.2 years (13 - 47 years). Thirty-two of the patients were female; sixteen were male. The single most common site is the posterior aspect of the distal femur (in 36 patients), followed by the femur shaft (in 6 patients), the proximal tibia (in 2 patients), etc. Nine patients had been operated on before referral to our department., Results: The average follow-up period from the first operation was 5.2 (0.25 - 24) years. In 36 of the 39 patients in whom a limb-salvage procedure was performed, a segment of the tumor-bearing bone was excised along with the tumor, whereas in 3 patients only the subadjacent cortex was excised with the tumor. In the limb-sparing group, the reconstruction was achieved by means of attenuated tumor bone or allograft in 23 cases, by endoprosthetic replacement in 9 cases, and by allograft replacement in 4 cases. The local resections were wide in 35 cases, and marginal in 13 cases. After marginal surgery, local recurrence occurred in 5/13 patients, whereas it occurred in 3/35 patients treated with wide resection. Pulmonary metastases developed in 6 patients, four patients died, and 2 patients are alive with disease. There were 4 cases of fractures of bone grafts. Four patients developed an infection. Long-term survival rate is 85.8%. For tumors that invaded the medullary canal there was no statistical association with local recurrence or metastasis. There is statistical significance between surgical margin and local recurrence., Conclusions: Wide surgical excision alone is adequate treatment for patients with conventional parosteal osteosarcoma. A tumor-free margin remains the critical factor determining overall prognosis. When a marginal excision was knowingly done to preserve a major neurovascular bundle, the risk of recurrence was less than when it was done to shell-out a presumptively benign lesion. Repeated recurrence probably increases the risk of dedifferentiation and thereby worsens the prognosis. Recurrent lesions with multiple soft-tissue satellite nodules or involvement of the neurovascular structures may however require amputation to provide sufficient local control when a wide margin cannot be achieved. An individualized resection will be performed in the future probably under the help of the advanced technique of image to distinguish the reactive zone from the normal tissue precisely.

Objective: To evaluate the differences in selecting patients who will undergo operation by comparing the evaluating systems and to discuss the indication of surgery performed on patients with spinal metastases., Methods: Of the 113 patients presented at Jishuitan Hospital, Beijing, PRC. from February 1994 to February 2002, who were diagnosed as spinal metastases, the clinical, pathological and imaging records of 82 patients were reviewed. Harrington's classification system, Tokuhashi's scoring system, Tomita's scoring system, and Grubb's scoring system were used to evaluate each of these patients at the same time. The classification or score concerning each patient was recorded and compared. SPSS software was used for statistical analysis. McNemar Test was used to compare the proportion of surgery indicated by different evaluating systems., Results: (1) Surgical treatment was indicated for 92.7% of patients by Tomita's scoring system and 69.5% by Harrington's classification system respectively. The difference was statistically significant at the level of P < 0.001. (2) Seventy-five cases with spinal cord palsy and progressive pain could be assessed by Tokuhashi's system. These patients were evaluated by Tomita's system at the same time. The type of surgery determined by Tokuhashi's score was excisional operation for 20% of the patients, palliative operation for 20%. As for the remaining 60%, either excisional or palliative operation might be chosen. The type of surgery determined by Tomita's score was excisional operation for 49.3% of the patients, palliative for 42.7%, and nonsurgical modality for the remaining 8.0%. (3) Twenty-five patients were classified as I, II or III, by Harrington's classification system. These patients were evaluated by the Grubb scoring system to predict the possibility of pathologic fracture and by Tomita's system at the same time to determine if an operation was necessary. By Grubb's system, prophylactic stabilization was necessary for twenty-two patients. By Tomita's system, surgery was chosen for 23 patients. But the treatment options were different in 5 patients assessed by the two systems., Conclusions: There was a great difference in the selection of treatment modality for patients with spinal metastases by different evaluating systems. That no systems have incorporated the evaluation of life expectancy and the local lesion causes differences. The patients should be evaluated individually by more factors than those proposed by the established systems.

Objective: To study the effect of neuropeptide Y (NPY) on the expression of heat shock protein 70 (HSP70) in vascular smooth muscle cells (VSMCs) of renal arteries., Methods: The renal veins of SD rats were isolated and broken into pieces. The VSMCs were cultured and then divided into 3 groups, neuropeptide Y (NPY), NPY + losartan, and serum-free DMEM were added into the culture respectively. Automated MTT colorimetric microassay and quantitative immunocytochemistry were used to detect the expression of heat shock protein (HSP70) in the proliferating VSMCs., Results: The fluorescence intensity of labeled HPS70 in the NPY group was 1,825.10 +/- 115.55, significantly stronger than that in the control group (1595.83 +/- 186.54, P < 0.05) and the fluorescence intensity of labeled HPS70 in the NPY + losartan group (1 658.54 +/- 183.78) was not significantly different from that in the control group (P > 0.05). The MTT-OD in NPY group was 0.2626 +/- 0.0025, significantly higher than that in the control group (0.2239 +/- 0.0010, P < 0.01). and the MTT-OD in NPY + losartan group was 0.2440 +/- 0.0013, significantly lower than that in the control group (P < 0.05)., Conclusion: NPY stimulates the proliferation of renal VSMC, promotes the expression of HPS70, and may cause hypertension. Losartan reduces the NPY stimulation over VSMC proliferation and relevant expression of intracellular HSP70.

Objective: To improve the life quality of cancer patients with metastasis to long bone and to select suitable surgical treatment., Methods: Fifty two patients with metastasis 27 men and 25 women, were treated from 1990 to 1999. Their average age was 56.8 years (33 - 74). In 16 patients with multiple lesions, underwent surgery at bone shaft (29 patients) and bone epiphysis (26). Thirty patients were treated for pathologic fracture and the rest for impending fracture. Operations included limb-salvage (51 patients) and amputation (4) Limb salvage consisted of intralesional curettage (3 patients), intramedullary nailing reconstruction (29), endoprosthesis (18), and temporary spacer (1). 21 patients accepted postoperative chemotherapy or radiotherapy., Results: Follow-up of 52 patients for a mean of 28.2 months (2 - 122 months) showed pain relief (41 patients), (75%) and full or part weight-bearing stability (36) 69%. Local tumor recurrence occurred in 11 patients., Conclusions: Surgical treatment can effectively improve the life quality of patients with metastasis to long bone. The metastatic lesions should be resected with wide or radical margin for the patients with kidney, breast, prostate and thyroid cancer.

Aim: To compare the efficacy and complications of extracorporeal shock-wave lithotripsy (SWL) and pneumatic ureteroscopic lithotripsy (URS) in the treatment of lower ureteral calculi., Methods: From August 1997 to June 1999, 210 patients with calculi in the distal third of the ureter were treated with SWL and the other 180 with URS. The stones were fragmented with either HB-ESWL-V lithotripter or JML-93 pneumatic lithotripter through Wolf 7.5 approximately 9.0 Fr ureteroscope. The outcome was assessed in terms of stone clearance rate, re-treatment rate and complication incidence., Results: The stone clearance rate was 78.1 % with SWL and 93.3 % with URS (P<0.05). SWL had a re-treatment rate of 11.9 %, vs 2.2 % in the URS group (P<0.05). URS caused ureteral perforation in 3.3% of patients, while it was 0 with SWL (P<0.05). The differences in the incidence of other complications such as infection and stricture between the two groups were insignificant., Conclusion: Though the selection of these two options depends on equipments available and the expertise of the operator, we recommend URS as the optimal treatment for distal ureteral calculi.

Fermentation strategies for production P (3HB-co-3HV) from glucose and propionic (or valeric) acid by Alcaligenes eutrophus were studied. During the culture, we controlled pH of the broth by feeding precusors of 3HV- propionic or valeric acid after Ammonia feeding stopped. When propionic acid were used as the precusor, for 50 hours, we obtained a cell dry weight, a P(3HB-co-3HV) concentration, a P(3HB-co-3HV) content and a 3HV fraction of 149.9 g/L, 124.9 g/L, 83.3% and 12.4 mol%, respectively, with a PHA productivity of 2.50 g h-1 L-1. When valeric acid were used as the precusor, for 45 hours, we obtained a cell dry weight, a P(3HB-co-3HV) concentration, a P(3HB-co-3HV) content and a 3HV fraction of 160.2 g/L, 119.0 g/L, 74.2% and 17.7 mol%, respectively, with a PHA productivity of 2.64 g h-1 L-1. Prior to this study, it hasn't been reported to obtain such high level productivity and 3HV fraction at the same time by Alcaligenes eutrophus.

From 1958 through 1988, 87 patients with primary sacral tumor were treated surgically. The lesions consisted of benign tumors (17 patients) such as neurofibroma, chordomas (41), giant cell tumors (21), and malignant growths (8). Fifty-nine patients sustained destruction of S1-3 vertebrae, and 28 had the neogrowth below S3. A total of 99 operations including 13 total and 17 subtotal excision of the sacrum, as well as 12 for recurrent tumors in 99 cases were performed. Ten operations for primary tumors resulted in 3 intraoperative and 7 postoperative deaths. Thus, 89 surgical attempts were stood by the patients. Sixty-seven patients were followed up with an average interval of 5.5 years. Sixty patients showed good functional results and resumed normal life and work, whereas urinary incontinence, constipation and weakness of ankles and feet occurred in 7. The factors conducive to surgical success were stressed. We conclude that with the exception of frank malignancy, surgery should be advised and actively adopted for primary sacral tumors.
