Abstract Sensations from the body are thought to play a critical role in many aspects of conscious experience, including first-person thought. In the present set of studies, we examined within-person relationships between in-the-moment subjective awareness of sensations from the body and self-reported first-person thought in real-world settings using ecological momentary assessment (EMA) protocols. In Study 1, participants reported experiencing greater first-person thoughts in moments when they also reported heightened awareness of sensations from their body, and this relationship was stable over a 4-week period even with mean-level changes in body awareness and first-person thought. In Study 2, we replicated this association in a 1-week EMA protocol using both self-report measures and measures derived from participants’ open-ended descriptions of their ongoing thoughts using a natural language processing approach. Taken together, findings shed light on the role of subjective body awareness in other facets of conscious experience.

Abstract The vast majority of disease-associated variants identified in genome-wide association studies map to enhancers, powerful regulatory elements which orchestrate the recruitment of transcriptional complexes to their target genes’ promoters to upregulate transcription in a cell type- and timing-dependent manner. These variants have implicated thousands of enhancers in many common genetic diseases, including nearly all cancers. However, the etiology of most of these diseases remains unknown because the regulatory target genes of the vast majority of enhancers are unknown. Thus, identifying the target genes of as many enhancers as possible is crucial for learning how enhancer regulatory activities function and contribute to disease. Based on experimental results curated from scientific publications coupled with machine learning methods, we developed a cell type-specific score predictive of an enhancer targeting a gene. We computed the score genome-wide for every possible cis enhancer-gene pair and validated its predictive ability in four widely used cell lines. Using a pooled final model trained across multiple cell types, all possible gene-enhancer regulatory links in cis (~17 M) were scored and added to the publicly available PEREGRINE database ( www.peregrineproj.org ). These scores provide a quantitative framework for the enhancer-gene regulatory prediction that can be incorporated into downstream statistical analyses.

Abstract Task-unrelated thoughts (TUTs) are frequent distractions from our everyday tasks, which can reduce productivity and safety during task performance. This necessitates the examination of factors that modulate TUT occurrence in daily life. One factor that has previously been implicated as a source of TUT is personally salient concerns. External factors such as news media serve as cues for these concerns, potentially increasing TUT occurrence. However, this has not been thoroughly examined in everyday life settings. We thus utilized Ecological Momentary Assessment to survey participants throughout the day for ten days, on their TUTs and news consumption in two separate studies. Study 1 focused on the impact of news related to the Coronavirus Disease (COVID-19) pandemic, as a common and global concern. We found that TUT occurrence was significantly predicted by COVID-19 news consumption, such that TUT occurrence increased with COVID-19 news consumption. To extend these findings, we implemented Study 2 using similar methods, but focusing on the consumption of news media in general. TUT occurrence was predicted by general news consumption, with an increase in reports of TUT following consumption of news media in general. We thus extended the association found between TUT and COVID-19-related news in Study 1, to any news topic in Study 2. We speculate that the increase in TUTs was due to heightened salience of current concerns, cued by the news. These findings highlight the importance of considering when we choose to consume news media and the value of examining contextual factors when studying TUTs in daily life.

Abstract Although central to well-being, functional and dysfunctional thoughts arise and unfold over time in ways that remain poorly understood. To shed light on these mechanisms, we adapted a “think aloud” paradigm to quantify the content and dynamics of individuals’ thoughts at rest. Across two studies, external raters hand coded the content of each thought and computed dynamic metrics spanning duration, transition probabilities between affective states, and conceptual similarity over time. Study 1 highlighted the paradigm’s high ecological validity and revealed a narrowing of conceptual scope following more negative content. Study 2 replicated Study 1’s findings and examined individual difference predictors of trait brooding, a maladaptive form of rumination. Across individuals, increased trait brooding was linked to thoughts rated as more negative, past-oriented and self-focused. Longer negative and shorter positive thoughts were also apparent as brooding increased, as well as a tendency to shift away from positive conceptual states, and a stronger narrowing of conceptual scope following negative thoughts. Importantly, content and dynamics explained independent variance, accounting for a third of the variance in brooding. These results uncover a real-time cognitive signature of rumination and highlight the predictive and ecological validity of the think aloud paradigm applied to resting state cognition.

Abstract Previous research has demonstrated reliable fluctuations in attentional processes during the course of the day. Everyday life experience sampling, during which participants respond to “probes” delivered at random intervals throughout the day on their mobile devices, is an effective tool for capturing such diurnal fluctuations in a naturalistic way. The existence of diurnal fluctuations in the case of mind-wandering, however, has not been examined to date. We did so in two studies. In the first study, we employed everyday experience sampling to obtain self-reports from 146 university students who rated the degree of free movement in their thoughts multiple times per day over five days. These time course data were analyzed using multilevel modelling. Freely moving thought was found to fluctuate reliably over the course of the day, with lower ratings reported in the early morning and afternoon and higher ratings around midday and evening. In the second study, we replicated these effects with a reanalysis of data from a past everyday experience-sampling study. We also demonstrated differences in parameter values for the models representing freely moving thought and two common conceptualizations of mind-wandering: task-unrelated thought and stimulus-independent thought. Taken together, the present results establish and replicate a complex pattern of change over the course of the day in how freely thought moves, while also providing further evidence that freedom of movement is dissociable from other dimensions of thought such as its task-relatedness and stimulus-dependence. Future research should focus on probing possible mechanisms behind circadian fluctuations of thought dynamics.

Mind wandering is often characterized by attention oriented away from an external task towards our internal, self-generated thoughts. This universal phenomenon has been linked to numerous disruptive functional outcomes, including performance errors and negative affect. Despite its prevalence and impact, studies to date have yet to identify robust behavioral signatures, making unobtrusive, yet reliable detection of mind wandering a difficult but important task for future applications. Here we examined whether electrophysiological measures can be used in machine learning models to accurately predict mind wandering states. We recorded scalp EEG from participants as they performed an auditory target detection task and self-reported whether they were on task or mind wandering. We successfully classified attention states both within (person-dependent) and across (person-independent) individuals using event-related potential (ERP) measures. Non-linear and linear machine learning models detected mind wandering above-chance within subjects: support vector machine (AUC = 0.715) and logistic regression (AUC = 0.635). Importantly, these models also generalized across subjects: support vector machine (AUC = 0.613) and logistic regression (AUC = 0.609), suggesting we can reliably predict a given individual's attention state based on ERP patterns observed in the group. This study is the first to demonstrate that machine learning models can generalize to "never-seen-before" individuals using electrophysiological measures, highlighting their potential for real-time prediction of covert attention states.

Contemporary investigations regard creativity as a dynamic form of cognition that involves movement between the dissociable stages of creative generation and creative evaluation. Our recently proposed Dynamic Framework of Thought (Christoff et al., 2016) offered a conceptualization of these stages in terms of an interplay between sources of constraint and variability on thought. This initial conceptualization, however, has yet to be fully explicated and given targeted discussion. Here, we refine this framework’s account of creativity by highlighting the dynamic nature of creative thought, both within and between the stages of creative generation and evaluation. In particular, we emphasize that creative generation in particular is best regarded as a product of multiple, varying mental states, rather than being a singular mental state in and of itself. We also propose that the psychedelic state is a mental state with high potential for facilitating creative generation and update the Dynamic Framework of Thought to incorporate this state. This paper seeks to highlight the dynamic nature of the neurocognitive processes underlying creative thinking and to draw attention to the potential utility of psychedelic substances as experimental tools in the neuroscience of creativity.

Enhancers are powerful and versatile agents of cell-type specific gene regulation, which are thought to play key roles in human disease. Enhancers are short DNA elements that function primarily as clusters of transcription factor binding sites that are spatially coordinated to regulate expression of one or more specific target genes. These regulatory connections between enhancers and target genes can therefore be characterized as enhancer-gene links that can affect development, disease, and homeostatic cellular processes. Despite their implication in disease and the establishment of cell identity during development, most enhancer-gene links remain unknown. Here we introduce a new, publicly accessible database of predicted enhancer-gene links, PEREGRINE. The PEREGRINE human enhancer-gene links interactive web interface incorporates publicly available experimental data from ChIA-PET, eQTL, and Hi-C assays across 78 cell and tissue types to link 449,627 enhancers to 17,643 protein-coding genes. These enhancer-gene links are made available through the new Enhancer module of the PANTHER database and website where the user may easily access the evidence for each enhancer-gene link, as well as query by target gene and enhancer location.

Despite an established body of research characterizing how creative individuals explore their "external" world, relatively little is known about how such individuals navigate their "inner mental life", especially in unstructured contexts such as periods of awake rest. Across two studies, the present manuscript tested the hypothesis that creative individuals are more engaged with their idle thoughts and more associative in the dynamic transitions between them. Study 1 captured the real-time conscious experiences of 81 adults as they voiced aloud the content of their mind moment-by-moment across a 10-minute unconstrained baseline period. Higher originality scores on a divergent thinking task were associated with less perceived boredom, more words spoken overall, more freely moving thoughts, and more loosely-associative (as opposed to sharp) transitions during the baseline rest period. In Study 2, across 2,612 participants, those who reported higher self-rated creativity also reported less perceived boredom during the COVID-19 pandemic, a time during which many people experienced unusually extended periods of unstructured free time. Overall, these results suggest a tendency for creative individuals to be more engaged and explorative with their thoughts when task demands are relaxed, raising implications for resting state functional MRI and societal trends to devalue idle time.

The Autobiographical Emotional Memory Task (AEMT), which involves recalling and writing about intense emotional experiences, is a widely used method to experimentally induce emotions. The validity of this method depends upon the extent to which it can induce specific desired emotions (intended emotions), while not inducing any other (incidental) emotions at different levels across one (or more) conditions. A review of recent studies that used this method indicated that most studies exclusively monitor post-writing ratings of the intended emotions, without assessing the possibility that the method may have differentially induced other incidental emotions as well. We investigated the extent of this issue by collecting both pre- and post-writing ratings of incidental emotions in addition to the intended emotions. Using methods largely adapted from previous studies, participants were assigned to write about a profound experience of anger or fear (Experiment 1) or happiness or sadness (Experiment 2). In line with previous research, results indicated that intended emotions (anger and fear) were successfully induced in the respective conditions in Experiment 1. However, disgust and sadness were also induced while writing about an angry experience compared to a fearful experience. Similarly, although happiness and sadness were induced in the appropriate conditions, Experiment 2 indicated that writing about a sad experience also induced disgust, fear, and anger, compared to writing about a happy experience. Possible resolutions to avoid the limitations of the AEMT to induce specific discrete emotions are discussed.
