Abstract Objective This study aimed to explore the association between electrophysiological markers of early recurrence after defibrillation in post-myocardial infarction ventricular fibrillation and the therapeutic effects of sympathetic renal denervation, as well as to investigate the potential underlying mechanisms. Methods Experimental research was conducted using an animal model. Myocardial infarction was induced, followed by defibrillation treatment for ventricular fibrillation cases, and the electrophysiological markers of early recurrence were recorded. Subsequently, a subset of animals underwent sympathetic renal denervation intervention, and the therapeutic effects were compared between the sympathetic renal denervation group and the control group. Electrocardiogram monitoring, histological analysis of myocardial tissue, and neurotransmitter measurements were also performed. Results Following defibrillation treatment, early recurrence was observed in ventricular fibrillation cases. The electrophysiological markers revealed significantly higher ST segment elevation and T wave changes in the early recurrence group. However, in the sympathetic renal denervation intervention group, the early recurrence rate was significantly reduced, and the electrocardiogram showed improved stability and regularity. Additionally, histological analysis of myocardial tissue demonstrated less cellular damage and lower levels of myocardial fibrosis in the sympathetic renal denervation group. Neurotransmitter measurements revealed a significant decrease in sympathetic nerve activity in the sympathetic renal denervation intervention group. Conclusion The results of this study indicate an association between electrophysiological markers of early recurrence after defibrillation in post-myocardial infarction ventricular fibrillation and the therapeutic effects of sympathetic renal denervation. Sympathetic renal denervation intervention can significantly reduce the early recurrence rate, improve electrocardiogram characteristics, and alleviate myocardial damage and fibrosis. Furthermore, the reduction in sympathetic nerve activity may be one of the potential underlying mechanisms of sympathetic renal denervation intervention.

Weed damage in rice fields is one of the main causes of reduced rice yields and quality. Accurate and efficient weed identification is the prerequisite for realizing intelligent and precise weeding in paddies. Recently, Vision Transformers (ViTs) have emerged with superior performance on computer vision tasks compared to the convolutional neural network (CNN)-based models. However, the lack of fully labeled weed datasets hinders the potential application of deep learning models in weed identification. To address the above issues, this study customizes a novel point-supervised instance segmentation network (PIS-Net) for weakly supervised instance segmentation of weeds in rice fields. More correctly, we first propose a novel instance segmentation point labeling scheme that utilizes randomly generated annotation points within each instance, aiming to decrease both labeling time and difficulty. Additionally, to make optimal use of point labels, this study puts forth a mask generation strategy based on the adaptive selection of pyramid levels. In this sense, the network model can flexibly choose the pyramid level expected to generate the most suitable instance mask based on the network's reliability. Finally, we establish the pseudo label refinement network (PLR-Net) to refine rough instance masks. The proposed PIS-Net utilizes 13 randomly generated annotation points for each instance, yet achieving an AP of 38.5 and an AP50 of 68.3, which is superior to the baseline mask-R-CNN with an AP of 8.2 and AP50 of 6.9, achieving 90 % fully supervised performance. This method effectively utilizes point labels, annotated with high efficiency, as a robust source of weak supervision to address challenges in weed data annotation and the low accuracy of existing weakly supervised models. Experiments show that the point annotation scheme of the PIS-Net is faster than full-object mask annotation, and the AP is also higher than the current semi-supervised weed segmentation model, enjoying high potentials in practical paddy fields.

Abstract Korla pear has a unique taste and aroma and is a breeding parent of numerous pear varieties. It is susceptible to Valsa mali var. pyri, which invades bark wounded by freezing injury. Its genetic relationships have not been fully defined and could offer insight into the mechanism for freezing tolerance and disease resistance. We generated a high-quality, chromosome-level genome assembly for Korla pear via the Illumina and PacBio circular consensus sequencing (CCS) platforms and high-throughput chromosome conformation capture (Hi-C). The Korla pear genome is ~ 496.63 Mb, and 99.18% of it is assembled to 17 chromosomes. Collinearity and phylogenetic analyses indicated that Korla might be derived from Pyrus pyrifolia and that it diverged ~ 3.9-4.6 Mya. During domestication, seven late embryogenesis abundant (LEA), two dehydrin (DHN), and 54 disease resistance genes were lost from Korla pear compared with P. betulifolia. Moreover, 21 LEA and 31 disease resistance genes were common to the Korla pear and P. betulifolia genomes but were upregulated under overwintering only in P. betulifolia because key cis elements were missing in Korla pear. Gene deletion and downregulation during domestication reduced freezing tolerance and disease resistance in Korla pear. These results could facilitate the breeding of novel pear varieties with high biotic and abiotic stress resistance.

Stacking fermentation is a crucial step in producing sauce-flavor Baijiu. The relationship between microbial diversity and volatile flavor substances is complex and dynamic. To better understand the correlation between microorganisms and flavor substances in Dongniang sauce-flavor Baijiu during the five-round stacking fermentation of Jiupei, high-throughput sequencing (HTS) technology and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME/GC-MS) were used to identify the microbial flora diversity and volatile flavor compounds. In addition, the Pearson correlation coefficient was computed, and a visualizable correlation network diagram was drawn based on the detection results. The results identified 55 volatile flavor compounds, including 25 esters, 5 alcohols, 7 aldehydes, 4 acids, 4 phenols, 3 alkanes, and 7 others. The main volatile components were ethyl lactate, phenylethyl alcohol, ethyl phenylacetate, and phenethyl acetate. The dominant microorganism genera in the floral structure were Virgibacillus, Kroppenstedtia, Bacillus, Oceanobaciillus, Thermomyces, Zygosaccharomyces, and Thermoascus. Kroppenstedtia, Monascus, Bacillus, and Zygosaccharomyces significantly affected flavor substances among the flora. Overall, the purpose and significance of this study are to provide a theoretical foundation for enhancing the quality of Dongniang sauce-flavor Baijiu.

Abstract Introduction Here we investigate the clinical features of foveal detachment (FD), full-thickness macular hole (MH), and macular hole retinal detachment (MHRD) in myopic traction maculopathy (MTM). Methods In the retrospective observational case series, 314 eyes of 198 patients with myopic retinoschisis in Beijing Tongren Hospital were enrolled. We recorded gender, age, and axial length and evaluated fundus characteristics using optical coherence tomography. Epiretinal membranes (ERMs), vitreoretinal traction, and paravascular abnormalities (PVAs) were used to describe the vitreoretinal interface condition. Different retinoschisis layers (inner, middle, and outer retinoschisis) and the location with a range of outer retinoschisis were evaluated to reveal the retinal condition. Five patterns of the scleral shape: dome-shaped, sloped toward the optic nerve, symmetrical or asymmetrical around the fovea, and irregular were assessed to show retina-sclera condition. We regarded the FD, full-thickness MH, and MHRD as the advanced stage in MTM. Multivariate logistic regression assessed significant factors for the advanced stage with odds ratios (OR) and 95% confidence intervals (CI). Results There were 76 eyes with FD, 6 eyes with full-thickness MH, and 7 eyes with MHRD. The mean age was 52.9 ± 12.3 years. On univariate analysis, the eyes with the advanced stage were older and had higher rates of ERMs, PVAs, middle retinoschisis, outer retinoschisis, and irregular sclera shape. The number of retinoschisis layers and the grade of outer retinoschisis were higher in eyes with the advanced stage. Summary: Evoked brain oscillations in the gamma range have been shown to assist in stroke recovery. However, the causal relationship between evoked oscillations and neuroprotection is not well understood. We have used optogenetic stimulation to investigate how evoked gamma oscillations modulate cortical dynamics in the acute phase after stroke. Our results reveal that stimulation at 40 Hz drives activity in interneurons at the stimulation frequency and phase-locked activity in principal neurons at a lower frequency, leading to increased cross-frequency coupling. In addition, 40-Hz stimulation after stroke enhances interregional communication. These effects are observed up to 24 h after stimulation. Our stimulation protocol also rescues functional synaptic plasticity 24 h after stroke and leads to an upregulation of plasticity genes and a downregulation of cell death genes. Together these results suggest that restoration of cortical dynamics may confer neuroprotection after stroke.

Abstract Background Adenoma-adenocarcinoma transition is a key feature of colorectal cancer (CRC) occurrence and is closely regulated by tumor-associated macrophages (TAMs) and CD8+ T cells. Here, we investigated the effect of the NF-κB activator 1 (Act1) downregulation of macrophages in the adenoma-adenocarcinoma transition. Methods This study used spontaneous adenoma-developing ApcMin/+, macrophage-specific Act1-knockdown (anti-Act1), and ApcMin/+; anti-Act1 (AA) mice. Histological analysis was performed on CRC tissues of patients and mice. CRC patients’ data retrieved from the TCGA dataset were analyzed. Primary cell isolation, co-culture system, RNA-seq, and fluorescence-activated cell sorting (FACS) were used. Results By TCGA and TISIDB analysis, the downregulation of Act1 expression in tumor tissues of CRC patients negatively correlated with accumulated CD68+ macrophages in the tumor. Relative expression of EMT markers in the tumor enriched ACT1lowCD68+ macrophages of CRC patients. AA mice showed adenoma-adenocarcinoma transition, TAMs recruitment, and CD8+ T cell infiltration in the tumor. Macrophages depletion in AA mice reversed adenocarcinoma, reduced tumor amounts, and suppressed CD8+ T cell infiltration. Besides, macrophage depletion or anti-CD8a effectively inhibited metastatic nodules in the lung metastasis mouse model of anti-Act1 mice. CRC cells induced activation of IL-6/STAT3 and IFN-γ/NF-κB signaling and the expressions of CXCL9/10, IL-6, and PD-L1 in anti-Act1 macrophages. Anti-Act1 macrophages facilitated epithelial-mesenchymal-transition and CRC cells’ migration via CXCL9/10-CXCR3-axis. Furthermore, anti-Act1 macrophages promoted exhaustive PD1+ Tim3+ CD8+ T cell formation. Anti-PD-L1 treatment repressed adenoma-adenocarcinoma transition in AA mice. Silencing STAT3 in anti-Act1 macrophages reduced CXCL9/10 and PD-L1 expression and correspondingly inhibited epithelial-mesenchymal-transition and CRC cells’ migration. Conclusions Act1 downregulation in macrophages activates STAT3 that promotes adenoma-adenocarcinoma transition via CXCL9/10-CXCR3-axis in CRC cells and PD-1/PD-L1-axis in CD8+ T cells.

Abstract Purpose To describe the normative profile of ophthalmic parameters in a healthy cynomolgus monkey colony, and to identify the characteristic of the spontaneous ocular disease non-human primates (NHP) models. Methods The NHP eye study was a cross-sectional on-site ocular examination with about 1,000 macaques held in Guangdong Province, southeastern China. The NHPs (Macaca fascicularis, cynomolgus) in this study included middle-aged individuals with a high prevalence of the ocular disease. The NHP eye study (NHPES) performed the information including systematic data and ocular data. Ocular examination included measurement of intraocular pressure (IOP), anterior segment- optical coherence tomography (OCT), slit-lamp examination, fundus photography, autorefraction, electroretinography, etc. Ocular diseases included measurement of refractive error, anisometropia, cataract, pterygium, etc. Results A total of 1148 subjects were included and completed the ocular examination. The average age was 16.4 ± 4.93 years. Compared to the male participants, the females in the NHPES had shorter axial length and the mean Average retinal nerve fiber layer (RNFL) thickness (except for the nasal quadrants). The mean IOP, anterior chamber depth, lens thickness, axial length, central corneal thickness, choroid thickness and other parameters were similar in each group. Conclusion The NHPES is a unique and high-quality study, this is the first large macaque monkey cohort study focusing on ocular assessment along with comprehensive evaluation. Results from the NHPES will provide important information about the normal range of ophthalmic measurements in NHP.

The pedunculopontine nucleus (PPN) is the major part of the mesencephalic locomotor region, involved in the control of gait and locomotion. The PPN contains glutamatergic, cholinergic, and GABAergic neurons that all make local connections, but also have long-range ascending and descending connections. While initially thought of as a region only involved in gait and locomotion, recent evidence is showing that this structure also participates in decision-making to initiate movement. Clinically, the PPN has been used as a target for deep brain stimulation to manage freezing of gait in late Parkinson’s disease. In this review, we will discuss current thinking on the role of the PPN in locomotor control. We will focus on the cytoarchitecture and functional connectivity of the PPN in relationship to motor control.

The rapid detection of chestnut quality is a critical aspect of chestnut processing. However, traditional imaging methods pose a challenge for chestnut-quality detection due to the absence of visible epidermis symptoms. This study aims to develop a quick and efficient detection method using hyperspectral imaging (HSI, 935–1720 nm) and deep learning modeling for qualitative and quantitative identification of chestnut quality. Firstly, we used principal component analysis (PCA) to visualize the qualitative analysis of chestnut quality, followed by the application of three pre-processing methods to the spectra. To compare the accuracy of different models for chestnut-quality detection, traditional machine learning models and deep learning models were constructed. Results showed that deep learning models were more accurate, with FD-LSTM achieving the highest accuracy of 99.72%. Moreover, the study identified important wavelengths for chestnut-quality detection at around 1000, 1400 and 1600 nm, to improve the efficiency of the model. The FD-UVE-CNN model achieved the highest accuracy of 97.33% after incorporating the important wavelength identification process. By using the important wavelengths as input for the deep learning network model, recognition time decreased on average by 39 s. After a comprehensive analysis, FD-UVE-CNN was deter-mined to be the most effective model for chestnut-quality detection. This study suggests that deep learning combined with HSI has potential for chestnut-quality detection, and the results are encouraging.

Background. To identify an optimal model for diabetic retinopathy (DR) prediction in Chinese rural population by establishing and comparing different algorithms based on the data from Handan Eye Study (HES). Methods. Five algorithms, including multivariable logistic regression (MLR), classification and regression trees (C&RT), support vector machine (SVM), random forests (RF), and gradient boosting machine (GBM), were used to establish DR prediction models with HES data. The performance of the models was assessed based on the adjusted area under the ROC curve (AUROC), sensitivity, specificity, and accuracy. Results. The data on 4752 subjects were used to build the DR prediction model, and among them, 198 patients were diagnosed with DR. The age of the included subjects ranged from 30 to 85 years old, with an average age of 50.9 years (SD=3.04). The kappa coefficient of the diagnosis between the two ophthalmologists was 0.857. The MLR model revealed that blood glucose, systolic blood pressure, and body mass index were independently associated with the development of DR. The AUROC obtained by GBM (0.952), RF (0.949), and MLR (0.936) was similar and statistically larger than that of CART (0.682) and SVM (0.765). Conclusions. The MLR model exhibited excellent prediction performance and visible equation and thus was the optimal model for DR prediction. Therefore, the MLR model may have the potential to serve as a complementary screening tool for the early detection of DR, especially in remote and underserved areas.

In this study, we investigated unmanned aerial vehicle (UAV) pollination of ‘Kuerle Xiangli’, and screened the pollination operation parameters to determine the precise parameters needed for the implementation of a ‘Kuerle Xiangli’ UAV pollination operation. Different flight height gradients, nozzle atomization particle sizes, spraying volume, and flight routes were tested and their effects on the droplets deposited were compared. UAV operation parameters were screened and field operations were conducted, comparing the fruit set rate, cost, and efficiency of different pollination methods of ‘Kuerle Xiangli’. The results show that the mist droplet effect of 1 m above the top of the tree is higher compared with that of 2 m and 3 m. The mist droplet effect of 2 L/667 m2 is better compared with that of 1.5 L/667 m2 and 1 L/667 m2. The mist droplet effect of 120 μm nozzle atomization particle size is better than that of 110 μm, 135 μm, and 150 μm. The mist droplet effect of flying above the canopy is better than that of flying between the rows of the canopy. The inflorescence and flower fruiting rates of ‘Kuerle Xiangli’ are 63.27% and 28.84%, respectively, and the inflorescence fruiting rate is not significantly different from hand and liquid sprayer pollination. The UAV pollination saves 12.69 USD/667 m2 and 3.32 USD/667 m2 compared with hand and liquid spray pollination, respectively. The efficiency of UAV pollination is greater than that of liquid and hand pollination. The best combination of parameters for pollination using a quadrotor UAV is 1 m from the top of the tree, 2 L/667 m2 spray volume, 120 μm spray nozzle particle size, and the flight path above the canopy. The cost of UAV pollination is 11.83 USD/667 m2 and the pollination efficiency 2.67 hm2/unit·h.

Purpose: To investigate the relationship between myopic tractional maculopathy (MTM) and myopic atrophy maculopathy (MAM).Method: Two hundred and six eyes with definitive myopic retinoschisis were assessed in the retrospective observational case series study and the atrophic and tractional features were further evaluated. Atrophic changes were analyzed according to the atrophic component in the ATN classification and the occurrence of gamma zones and delta zones. Tractional changes were evaluated based on different retinoschisis layers, the location and range of outer retinoschisis, retinal detachment, inner lamellar macular hole (ILMH), outer lamellar MH (OLMH), full-thickness MH (FTMH), and paravascular abnormalities.Results: Of all the eyes, 29.6, 42.7, 19.4, and 8.3% presented MAM grades with A1, A2, A3, and A4, respectively. The three layers of retinoschisis and the entire macular retinoschisis had the highest incidences in A2 (38.6%; 54.5%). The numbers of retinoschisis layers and the grades of outer retinoschisis had a weak negative correlation with MAM (r = −0.138, P = 0.048; r = −0.139, P = 0.047). All the eyes had gamma zones, and 82.52% of eyes also had delta zones. The incidence of retinal detachment and OLMH reached the peak in A2 and then decreased gradually. With MAM aggravation, the prevalence of ILMH decreased. Eyes with A1 and A2 were more likely to have OLMH, and those with A3 and A4 were more likely to have FTMH (P = 0.028; OR, 3.423; 95% CI, 1.144–10.236; P = 0.004; OR, 7.752; 95% CI, 1.951–30.803). With the MAM grades growing, the types of paravascular abnormalities increased (r = 0.165, P = 0.018).Conclusion: Diffuse chorioretinal atrophy was the dominant MAM grade in eyes with MTM. In the study, 72.3% of eyes with MTM presented with diffuse chorioretinal atrophy and a tessellated fundus. Over 80% of eyes with MTM had both gamma zones and delta zones. Diffuse chorioretinal atrophy might be a complicated stage for MTM with the highest rate of three layers of retinoschisis, the entire macular retinoschisis, RD, and OLMH. Atrophic progression might involve the development of MH. When MTM combines with well-defined atrophy, the occurrence of FTMH should be noted.

Litchi downy blight caused by Peronophythora litchii is the most serious disease in litchi production, storage and transportation. Existing disease identification technology has difficulty identifying litchi downy blight sufficiently early, resulting in economic losses. Thus, the use of diffuse reflectance spectroscopy to identify litchi downy blight at different stages of disease, particularly to achieve the early identification of downy blight, is very important. The diffuse reflectance spectral data of litchi fruits inoculated with P. litchii were collected in the wavelength range of 350–1350 nm. According to the duration of inoculation and expert evaluation, they were divided into four categories: healthy, latent, mild and severe. First, the SG smoothing method and derivation method were used to denoise the spectral curves. Then, the wavelength screening methods competitive adaptive reweighted sampling (CARS) and successive projections algorithm (SPA) were compared to verify that the SPA method was more effective. Eleven characteristic wavelengths were selected, accounting for only 1.1% of the original data. Finally, the characteristic wavelengths were tested by six different classification models, and their accuracy was calculated. Among them, the ANN model performed best, with an accuracy of 90.7%. The results showed that diffuse reflectance spectroscopic technology has potential for identifying litchi downy blight at different stages, providing technical support for the subsequent development of related automatic detection devices.

Summary: The regenerative capacity of the human endometrium requires a population of local stem cells. However, the phenotypes, locations, and origin of these cells are still unknown. In a mouse menstruation model, uterine stromal SM22α+-derived CD34+KLF4+ stem cells are activated and integrate into the regeneration area, where they differentiate and incorporate into the endometrial epithelium; this process is correlated with enhanced protein SUMOylation in CD34+KLF4+ cells. Mice with a stromal SM22α-specific SENP1 deletion (SENP1smKO) exhibit accelerated endometrial repair in the regeneration model and develop spontaneous uterine hyperplasia. Mechanistic studies suggest that SENP1 deletion induces SUMOylation of ERα, which augments ERα transcriptional activity and proliferative signaling in SM22α+CD34+KLF4+ cells. These cells then transdifferentiate to the endometrial epithelium. Our study reveals that CD34+KLF4+ stromal-resident stem cells directly contribute to endometrial regeneration, which is regulated through SENP1-mediated ERα suppression. : The regenerative capacity of the human endometrium requires a population of local stem cells. Burkitt lymphoma (BL) is a highly aggressive neoplasm, which arising from the germinal center or post germinal center B-cell. Primary breast lymphomas are extremely rare, and the most common histologic type is diffuse large B-cell lymphoma. Primary BL of the breast is much less common than the other types of lymphoma. Here, we report an extremely rare case of a 37-year-old Chinese female with localized bilateral breast, who was referred to our institution for bilateral breast swelling. The left breast tissue ultrasonography showed the short axis measuring 20.3 mm × 18.8 mm and the long axis measuring 22.1 mm × 20.8 mm soft tissue mass. The right breast tissue ultrasonography showed the short axis measuring 30.2 mm × 26.9 mm and the long axis measuring 33.5 mm × 2.18 mm. Coarse needle biopsy of breast masses demonstrated a non-Hodgkin's B-cell lymphoma. The patient underwent a bilateral mastectomy. Histological examination of the tumor showed a characteristic "starry sky" pattern, the medium-sized tumor cells were a monotonous pattern of growth, and there were many abnormal mitotic figures. The neoplastic cells strongly expressed CD20, CD79-μ, MUM-1, PAX-5, CD43 and Bcl-6, Ki-67 were nearly 100% positive, but negative for CD10, Bcl-2 and TdT. By fluorescence in situ hybridization an IGH-MYC gene fusion was detected in the tumor tissue which indicating the presence of a typical BL translocation t(8;14)(q24;q32). The final histopathological diagnosis was primary BL of the breast.

Coronary artery blockage causes myocardial infarction (MI), a frequent and serious cardiovascular disease. The early recurrence of post-MI ventricular fibrillation after defibrillation has been widely investigated and treated. This research investigated the relationship between electrophysiological indicators of early recurrence following defibrillation in post-MI ventricular fibrillation and sympathetic renal denervation's therapeutic benefits and probable causes. Animal models were used for experiments. Electrophysiological indications of early recurrence were reported after MI and defibrillation in ventricular fibrillation patients. After that, a selection of rats received sympathetic renal denervation, and the therapeutic results were compared to the control group. Electrocardiogram monitoring, myocardial histology, and neurotransmitter assays were done. Defibrillation therapy causes an early recurrence in ventricular fibrillation patients. Electrophysiological measures showed increased ST segment elevation and T wave alterations in the early recurrence group. In the sympathetic renal denervation intervention group, early recurrence was greatly decreased and the electrocardiogram (ECG) was more stable and regular. Myocardial histology showed decreased cellular damage and fibrosis in the sympathetic renal denervation group. Sympathetic renal denervation intervention significantly reduced sympathetic nerve activity, according to neurotransmitter measures. Electrophysiological indications of early recurrence following defibrillation in post-MI ventricular fibrillation are linked to sympathetic renal denervation's therapeutic benefits. Myocardial damage and fibrosis may be reduced, ECG features improved, and the early recurrence rate reduced by sympathetic renal denervation. Purpose To describe the normal macular thickness and assess its associations. Methods The Handan Eye Follow-up Study was conducted between 2012 and 2013. Macular thickness was scanned by spectral-domain optical coherence tomography (OCT). The built-in software generated a retinal thickness (RT) map, which was divided into three regions (central, internal and external regions) and nine quadrants (one in central and four in internal and external regions each). Results For 5394 subjects in the Handan Eye Follow-up Study, 4793 received OCT examination, 2946 of whom (accounting for 61.46% of the total subjects, mean age 58.91±10.95, 55.6% were women) were included for analysis. The mean RT in central macula, inner and outer rings were (237.38 µm±23.05 µm), (309.77 µm±18.36 µm) and (278.29 µm±14.38 µm), respectively (overall difference, p<0.001). In inner ring, the RT in temporal was thinnest, followed by nasal, superior and inferior. In outer ring, the RT in superior was thinnest, with the next subfields being temporal, inferior and nasal, respectively. The RT in central macula, inner and outer rings were significantly thicker in men than in women. Multivariate linear regression analysis showed that in central macula, RT increased in subjects younger than 60 years and thinned above the age of 60. In inner and outer rings, RT thinned along with age (p<0.001). Conclusions This study finds that RT in central macula is the thinnest, followed by the outer ring, the RT in the inner ring is the thickest. Age and gender are related to RT. Purpose To examine the normative profile of retinal nerve fibre layer (RNFL) thickness and ocular parameters based on spectral-domain optical coherence tomography (SD-OCT) and its associations with related parameters among the Chinese population. Methods This population-based cohort Handan Eye Study (HES) recruited participants aged=30 years. All subjects underwent a standardised ophthalmic examination. Peripapillary RNFL thickness was obtained using SD-OCT. Mixed linear models were adopted to evaluate the correlation of RNFL thickness with ocular parameters as well as systemic factors. R V.3.6.1 software was used for statistical analysis. Results 3509 subjects (7024 eyes) with the average age of 55.54±10.37 were collected in this analysis. Overall mean RNFL thickness measured was 113.46±10.90 µm, and the thickest quadrant of parapapillary RNFL was the inferior quadrant, followed by the superior quadrant, the nasal quadrant and the temporal quadrant. In the multivariate linear regression model, thinner RNFL thickness was remarkable association with male (p<0.001), older age (p<0.001), increased body mass index (>30, p=0.018), absence of diabetes (p=0.009), history of cataract surgery (p=0.001), higher intraocular pressure (p=0.007), lower spherical equivalent (p<0.001) and increased axial length (p=0.048). Conclusions In non-glaucoma individuals, this difference of RNFL thickness in Chinese population should be noted in making disease diagnoses. Meanwhile, multiple ocular and systemic factors are closely related to the thickness of RNFL. Our findings further emphasise the need to demonstrate ethnic differences in RNFL thickness and the specificity of associated ocular and systemic factors, as well as to develop better normative databases worldwide. Clear cell renal cell carcinoma (ccRCC) accounts for approximately 75–80% of all patients with renal cell carcinoma. Despite its prevalence, little is known regarding the key components involved in ccRCC metastasis. In this study, scRNA‐seq analysis was employed to classify CD8+ T cells into four sub‐clusters based on their genetic profiles and immunofluorescence experiments were used to validate two key clusters. Through gene set enrichment analysis, these newly identified sub‐clusters were found to exhibit distinct biological characteristics. Notably, TYMP, TOP2A, CHI3L2, CDKN3, CENPM, and RZH2 were highly expressed in these sub‐clusters, indicating a correlation with poor prognosis. Among these sub‐clusters, CD8+ T cells (MT‐ND4) were identified as potentially playing a critical role in mediating ccRCC metastasis. These results contribute to our understanding of CD8+ T cell heterogeneity in ccRCC and shed light on the mechanisms underlying the loss of immune response against cancer.
