2. Optical identification and follow-up observations of SRGA J213151.5+491400 -- a new magnetic cataclysmic variable discovered with SRG Observatory

Abstract

We report results of optical identification and multi-wavelength study of a new polar-type magnetic cataclysmic variable (MCV), SRGA J213151.5+491400, discovered by Spectrum Roentgen-Gamma ($SRG$) observatory in the course of the all-sky survey. We present optical data from telescopes in Turkey (RTT-150 and T100 at the T\"UBITAK National Observatory), and in Russia (6-m and 1-m at SAO RAS), together with the X-ray data obtained with $ART-XC$ and $eROSITA$ telescopes aboard $SRG$ and the $NICER$ observatory. We detect SRGA J213151.5+491400 in a high state in 2020 (17.9 mag) that decreases about 3 mag into a low state (21 mag) in 2021. We find only one significant period using optical photometric time series analysis which reveals the white dwarf spin/orbital period to be 0.059710(1) days (85.982 min). The long slit spectroscopy in the high state yields a power law continuum increasing towards the blue with a prominent He II line along with the Balmer line emissions with no cyclotron humps; consistent with MCV nature. Doppler Tomography confirms the polar nature revealing ballistic stream accretion along with magnetic stream during the high state. These characteristics show that the new source is a polar-type MCV. $SRG$ $ART-XC$ detections yield an X-ray flux of (4.0-7.0)$\times$10$^{-12}$ erg cm$^2$ s$^{-1}$ in the high state. $eROSITA$ detects a dominating hot plasma component (kT$_{\rm{max}}$ $>$ 21 keV in the high state) declining to (4.0-6.0)$\times$10$^{-13}$ erg cm$^2$ s$^{-1}$ in 2021 (low state). The $NICER$ data obtained in the low state reveal a two-pole accretor showing a soft X-ray component at (6-7)$\sigma$ significance with a blackbody temperature of 15-18 eV. Abstract

We report the observation of a possible optical counterpart to the recently discovered X-ray source NuSTAR J053449+2126.0 (J0534 in short). We observed the source location using the 1.5-m Telescope (RTT150) to search for an optical counterpart, and detected a possible optical counterpart and analysed its spectrum using the $B$, $V$, $R$, and $I$ images of J0534, and found that the possible optical counterpart of J0534 is likely to be a long-known optical source, namely PSO J083.7063+21.4333. However, this source has been misclassified as a star rather than being an extragalactic source. We determined the source distance accurately for the first time based on our spectral analysis. J0534 could be a high-redshift ($z\simeq2.2$) member of an Active Galactic Nucleus (AGN) sub-group identified as quasars. Our analysis favours an accreting black hole of mass $\sim 7\times 10^8 M_{\odot}$ as a power supply for the quasar. Recently, there has been a rapid increase in the number of females using substances and although usage tends to reduce during pregnancy, it is of critical importance to determine its exact level as substance use negatively impacts on the health of both the mother and infant. Aim: The aim of the present study was to investigate the frequency of smoking, alcohol, and substance use, and quitting rates during pregnancy. Method: This study was conducted on pregnant females in Sakarya, Turkey. A total of 1,082 consecutively presenting females who agreed to participate in the study were evaluated. The study team prepared a sociodemographic data form and adapted the “Introduction” section, derived from the Addiction Profile Index, to cover substance use during pregnancy. Results: The substances most frequently used by pregnant females in their previous pregnancies and current pregnancies were cigarettes/tobacco products (11% and 11.8%, respectively), alcohol (0.6% and 0.4%, respectively), and rarely, synthetic cannabinoids (0.3% and 0.2%, respectively). Daily tobacco smokers continued to smoke during pregnancy, with a rate of 42.5%. Based on research into predictors of smoking (cigarettes) in pregnancy, a correlation was found between lifetime smoking and smoking during a previous pregnancy. A similar link was found with respect to alcohol. Conclusion: Cigarettes are the most frequently used substance in pregnancy, and to a lesser extent, alcohol and synthetic cannabinoids, also considered to be risky substances. A high incidence of smoking regularly during pregnancy was found in daily smokers. It is recommended that physicians should sensitively ask pregnant females presenting at clinics about all forms of substance use, including alcohol and synthetic cannabinoids, and to include such questions in their routine enquiries. 21. The effect of growth hormone addition protocols to poor ovarian responders in in vitro fertilization cycles.

Abstract

OBJECTIVE: In vitro fertilization failure (IVF) is high in women with poor ovarian response or non-responder. For this reason, the addition of adjuvant treatments to IVF protocols has come to the fore. We assessed to investigate the effects of adjuvant GH therapy initiated in the mid-luteal phase on IVF success in poor ovarian response or non-responder women. PATIENTS AND METHODS: A retrospective study was performed in 93 poor ovarian response or non-responder women from a single center. GH treatment was added (GH-plus group) in the mid-luteal phase of the previous menstrual cycle to 47 of the women who underwent controlled ovarian stimulation with the flexible antagonist protocol. 46 women, as another group, were applied to a flexible antagonist-only protocol (GH-free group). The IVF outcome results were evaluated and compared within the groups. RESULTS: The number of retrieved oocytes was statistically significantly higher in the GHplus group (2.28±1.975) than in the GH-free group (1.24±1.728) (p=0.01). Although statistically insignificant (p=0.55), the clinical pregnancy rate was higher in the GH-plus group [(8/47), 17%] than in the GH-free group [(5/46, 11%]. The cancellation rate was statistically significantly higher in the GH-free group (65.2%) than in the GHplus group (44.7%) (p=0.04). No oocyte retrieved cycle rate was higher in the GH-free group (56%) than in the GH-plus group (25%) (p=0.002). CONCLUSIONS: Adjuvant GH therapy administration to IVF protocol in the mid-luteal phase gives poor ovarian response or non-responder women a chance to have a baby. [ABSTRACT FROM AUTHOR]

Recently, there has been a rapid increase in the number of females using substances and although usage tends to reduce during pregnancy, it is of critical importance to determine its exact level as substance use negatively impacts on the health of both the mother and infant. Aim: The aim of the present study was to investigate the frequency of smoking, alcohol, and substance use, and quitting rates during pregnancy. Method: This study was conducted on pregnant females in Sakarya, Turkey. A total of 1,082 consecutively presenting females who agreed to participate in the study were evaluated. The study team prepared a sociodemographic data form and adapted the “Introduction” section, derived from the Addiction Profile Index, to cover substance use during pregnancy. Results: The substances most frequently used by pregnant females in their previous pregnancies and current pregnancies were cigarettes/tobacco products (11% and 11.8%, respectively), alcohol (0.6% and 0.4%, respectively), and rarely, synthetic cannabinoids (0.3% and 0.2%, respectively). Daily tobacco smokers continued to smoke during pregnancy, with a rate of 42.5%. Based on research into predictors of smoking (cigarettes) in pregnancy, a correlation was found between lifetime smoking and smoking during a previous pregnancy. A similar link was found with respect to alcohol. Conclusion: Cigarettes are the most frequently used substance in pregnancy, and to a lesser extent, alcohol and synthetic cannabinoids, also considered to be risky substances. A high incidence of smoking regularly during pregnancy was found in daily smokers. It is recommended that physicians should sensitively ask pregnant females presenting at clinics about all forms of substance use, including alcohol and synthetic cannabinoids, and to include such questions in their routine enquiries. 33. Comparison of culture and real-time polymerase chain reaction methods for detection of Mycoplasma hominis in amniotic fluids samples

Abstract

Background: Mycoplasma hominis is often present in the amniotic cavity with microbial invasion associated with spontaneous preterm labor. Conventional culture method is the gold standard for detection of Mycoplasmas, but real‑time polymerase chain reaction (real‑time PCR) has revolutionized the diagnosis of M. hominis.Objective: The purpose of this study is the comparison of the culture methodology with real‑time PCR for the detection of M. hominis in amniotic fluid samples.Methods: Amniotic fluid samples were collected from 65 pregnant women (age range: 25–45 years) previously followed at an infertility clinic. They were collected by transabdominal genetic amniocentesis during 16–21 weeks of gestation. Amniotic fluids were inoculated in SP4 broth for 48–72 h, and after becoming alkaline, culture suspension was spread on A7 agar plate for 1 week till the typical colonies seen in “fried‑egg” morphology under stereomicroscope. DNA was extracted using a QIAGEN Mini DNA kit. The real‑time‑PCR was performed using Rotor‑Gene Q Real‑time PCR instrument. A melting‑curve analysis was also performed. Sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) were measured by real‑time PCR by taking culture as gold standard.Results: Sixty‑five women in 16–21 weeks of gestation, with a mean age of 33 ± 5.06 years, were enrolled into this study. M. hominis detected by culture and real‑time PCR assay was 72% (47/65) and 69% (45/65), respectively. 66% (43/65) specimens were positive by both methods. Real‑time PCR sensitivity was 91.5%, specificity 88.9%, PPV 95.6%, and NPV 80%.Conclusion: Rapid detection of Mycoplasmas causing maternal complications such as neonatal infections and preterm labor in pregnancy by real‑time PCR may be important and necessary. The high sensitivity and shorter time requirement of real‑time PCR support its further development for diagnosis of Mycoplasma infections.Keywords: Amniotic fluids, culture, Mycoplasma hominis, real‑time polymerasechain reaction

Abstract

MOTAKK, as a national external quality control program has been launched to evaluate the molecular detection of viral infections including HBV DNA and HCV RNA in molecular microbiology diagnostic laboratories in Turkey. This program is prepared in compliance with ISO 17043:2010 (Conformity assessment general requirements for proficiency testing) standards, and aims to take the place of external quality control programs from abroad, contributing to standardization and accuracy of molecular diagnostic tests in our country. The aim of this study was to evaluate 2015 and 2016 results of the MOTAKK External Quality Control Program for HBV DNA and HCV RNA viral load. The calls were announced on the web page of MOTAKK (www.motakk.org). The quality control samples were sent to participating laboratories in 2015 and 2016. Main stocks were prepared from patients with chronic hepatitis B and C who had viral load detection with reference methods according to WHO reference materials for viral load studies to improve quality control sera. From these main stocks, samples with different viral loads were prepared from dilutions of plasma with HBV, HCV, HAV, HIV, Parvovirus B19 and CMV negative serologic markers. Quality control samples were sent to the participating laboratories along with the negative samples in the cold chain. The laboratories accomplished the related tests within 2-3 weeks and entered their results on the MOTAKK web page. These results were analysed according to ISO 13528 (Statistical methods for use in proficiency testing by interlaboratory comparison) and scoring reports were created by a software developed by MOTAKK and sent to participating labs. Each laboratory evaluated their own results in comparison with the other laboratory results, reassessed the tests via observing the distance from the mean result and the reference values. The number of laboratories participating in the HBV DNA and HCV RNA external quality control program was 70-73 in 2015-2016. Participants were able to comply with the program tools, registering, entering results and receiving the results reports problem. In HBV panel, 72.6-89.1% and 84.7-90.3% of the participant laboratories were in 1 standard deviation (SD) in 2015-2016, respectively. In HCV panel, 70.8-89.1% and 84.7-90.3% of the participant laboratories were in 1 SD in 2015-2016, respectively. A national external quality control program for HBV DNA and HCV RNA in Turkey has been prepared for the first time with this project and implemented successfully. All the data provided in the MOTAKK external quality control program final report, compensate all the data provided by the quality control program final reports from abroad; additionally, the report allows comparison of used technologies and commercial products.

Abstract

A number of storage media have been investigated as to their ability to maintain the viability of the periodontal ligament (PDL) cells and thus to permit longer extra-alveolar periods prior to replantation of avulsed teeth. The aim of the present in vitro study was to evaluate the number of viable PDL cells of avulsed teeth treated by Hank's Balanced Salt Solutions (HBSS), saline, a novel probiotic solution and milk. Thirty-six freshly extracted single-rooted human teeth with closed apices were divided into one of the four experimental groups and two control groups (N = 6 each). The positive and negative controls corresponded to 0 min and an 8-h dry time respectively. Following extraction, the coronal 3 mm of PDL tissue was scraped with a #15 scalpel to remove cells that might have been damaged. The experimental teeth were dried for 30 min followed by a 45 min immersion in one of the four experimental media. Each experimental tooth, after drying and soaking, was incubated for 30 min with a 2.5 ml solution of 0.2 mg ml-1 of collagenase CLS II and a 2.4 mg ml-1 solution of dispase grade II in phosphate buffer saline (PBS). The cells were then labelled with 0.4% Trypan blue for determination of viability. The teeth stored in positive control demonstrated the highest number of viable PDL cells followed in rank order by HBSS, saline, Lactobacillus reuteri solution and milk. There was no significant difference in the number of viable PDL cells between HBSS, milk, L. reuteri solution and saline. Within the parameters of this study, it appears that probiotic may be able to maintain PDL cell viability as HBSS, milk, or saline. © 2010 John Wiley & Sons A/S.

Abstract

Fibres have been extracted from corn husks in undried and dried forms by an alkalization treatment followed by enzymatic process at different concentrations of xylanase enzyme. The effects of drying process and xylanase enzyme concentration on the mechanical and textile properties of corn husk fibres have been investigated. The linear density of extracted fibres decreases with the increase in enzyme concentration and drying. Increasing enzyme concentration increases the breaking tenacity and initial modulus up to a point and then decreases them with any further increase in concentration. Fibres obtained from dried husks show dull colour as compared to those obtained from undried husks. No negative effect of drying process is observed on the performance of corn husk fibres. The highest tensile performance and fineness values are obtained from fibres of dried husks, treated with 0.4% Pentopan (R) mono BG enzyme, with a breaking tenacity of 9.44 cN/tex, initial modulus value of 282 cN/tex, and a fineness of 21.6 tex.
