Abstract

In this work, we present an alternative approach to sensitize the near infrared emission of Er3+ ions (used in telecom applications) by exploiting the geometrical confinement occurring in porous zeolites structures. The sensitization of the Ln ion is obtained by energy transfer between a suitable organic molecule acting as an antenna and the emitting ion arranged in closed proximity, thus, avoiding the limits imposed by the coordination chemistry. ©2008 American Institute of Physics

Abstract

Purpose: Urinary extracellular vesicles (UEVs) are a novel source for disease biomarker discovery. However, Tamm-Horsfall protein (THP) is still a challenge for proteomic analysis since it can inhibit detection of low-abundance proteins. Here, we introduce a new approach that does not involve an ultracentrifugation step to enrich vesicles and that reduces the amount of THP to manageable levels. Experimental design: UEVs were dialyzed and ultrafiltered after reduction and alkylation. The retained fraction was digested with trypsin to reduce the remaining THP and incubated with deoxycholate (DOC). The internal peptidome and internal proteome were analyzed by LC-ESI-MS. Results: A total of 942 different proteins and 3115 unique endogenous peptide fragments deriving from 973 different protein isoforms were identified. Around 82% of the key endosomal sorting complex required for transport components of UEVs generation could be detected from the intraluminal content. Conclusions and clinical relevance: Our UEVs preparation protocol provides a simplified way to investigate the intraluminal proteome and peptidome, in particular the subpopulation of UEVs of the trypsin-resistant class of exosomes (positive for tumor susceptibility gene101) and eliminates the majority of interfering proteins such as THP. This method allows the possibility to study endoproteome and endopeptidome of UEVs, thus greatly facilitating biomarker discovery.

Abstract

The interaction between sulfur-containing chromophores and cationic species (K+) has been investigated in dye-loaded zeolite materials by means of photoluminescence spectroscopy. A red-shift in the emission spectra of the host-guest compounds (HGCs) has been detected and unambiguously connected to the close proximity between a conjugated moiety and nearby free charges, suggesting a specific role played by sulfur lone pair electrons. Quantum-chemical calculations on model compounds have been performed to support this hypothesis.

Abstract

We measured serum tartrate-resistant acid phosphatase (TRAP) activity in 120 healthy subjects and 35 patients with multiple myeloma as well as urinary hydroxyproline excretion in the myeloma patients. Young subjects (0-18 years) showed higher TRAP levels (ANOVA p < 0.01) compared with the other age classes due to the more active bone remodelling processes associated with growth. Myeloma patients with bone lytic lesions (MM+) showed higher serum TRAP values than controls (p < 0.01). Hydroxyproline excretion was higher in MM + patients but the difference between patients with and without bone lesions was not statistically significant. Our data suggest that serum TRAP activity may be a suitable, simple biochemical test to assess bone turnover in patients with multiple myeloma but that its clinical usefulness as a marker of bone resorption needs further evaluation.
