Abstract

Dipole moment analysis and molecular orbital AM1-calculated enthalpies on furan-2, 5-dicarboxaldehyde (5) compound reveal that solution 5 primarily contains conformation B, conformation A being present in small quantities. The pyrrole-2,5-dicarboxyaldehyde (7) and the corresponding 1-methylpyrrole-2, 5-dicarboxyaldehyde were primarily in conformation A. The compound 7 altered the course of the reaction to enable a (3 + 3) cyclocondensation derivative in good yield and the extraction of the first phosphorus-containing macrocycle arising from a (4 + 4) cyclocondensation reaction. The addition of 7 to the phosphodihydrazide 1b led to another (3 + 3) species, the macrocycle 10'' in 74% yield.

Abstract

International audience; Dendrimers, which are hyperbranched synthetic macromolecules, have attracted researchers’ increasing interest over the last three decades owing to their numerous properties, in particular, in the fields of biology and nanomedicine. This book is an up-to-date collection of the most recent achievements in the use of dendrimers in nanomedicine, exemplified by phosphorus-containing dendrimers. Starting with the synthesis and characterization of phosphorus dendrimers, the book discusses their use as biomaterials, carriers of biological entities, and anti-inflammatory drugs; in bioimaging; and against prion diseases, Alzheimer’s, HIV, and cancer. It compares phosphorus dendrimers with other types of dendrimers. Supplemented with numerous references and abundant illustrations, the book opens new perspectives for the researchers working on dendrimers.

Abstract

International audience; The central nervous system (CNS) is constituted of the brain and the spinal cord. This chapter presents the potential role of several types of phosphorus-containing dendrimers to fight against diverse diseases of the central nervous system, including prion, Alzheimer's, Parkinson's, and MS diseases. The most important poly(phosphorhydrazone) (PPH) dendrimer used up to now against diverse neurodegenerative diseases is the generation 4, built from a cyclotriphosphazene core, and covered by ammonium terminal functions. The antioxidant properties of the cationic PPH dendrimers were measured using two methods: scavenging of the stable radical DPPH (2,2-dipheny-1-picrylhydrazyl) and ability to reduce ferric ions to ferrous ions. The biological properties tested with these new dendrimers concerned the cytotoxicity, hemotoxicity, and the antimicrobial and antifungal activities. Human erythrocytes were used for studying the hemotoxicity, measured by the release of hemoglobin. cytotoxicity, hemotoxicity, antimicrobial activities, antifungal activities.

Abstract

International audience; Protolytic, absorbance and fluorescence properties of β-diketone phosphorus-containing dendrimers based on cyclotriphosphazene core were studied. Dendrimer solutions in acetone are characterized by intense absorbance band at ≈ 340 nm (ε340≈ 8.5·104L/mol·сm) and fluorescence band with maximum at 440 nm. Position of these maxima does not change in various solvents, unlike the bands of monomer β-diketone. It was found that dendrimer aggregation is accompanied by appearance of a second absorbance band ε400≈ 4.5·103L/mol·сm, by red shift of emission spectra ∆λ ≈ 10 nm and also by decrease in surface tension of acetone solution. Ability of dendrimer aggregates to solubilize organic substrates was observed with the fluorescent indicator acridine.

Abstract

International audience; A versatile reactivity from the cage compound P(NMeNMe)3P is presented. The Staudinger reaction with Me3SiN3 is carried out. The crystal structure of the compound issued from the reaction on both sides (Me3SiN=P(NMeNMe)3P=NSiMe3) is reported. When the reaction occurs on only one side, the remaining free phosphorus atom is complexed with RuCl2(p-cymene). P(NMeNMe)3P reacts with PCl3, leading to the heterocyclic compound ClP(NMeNMe)2PCl. This heterocycle also displays a versatile reactivity. Substitution reaction with HNiPr2 leads to iPr2NP(NMeNMe)2PNiPr2. Very complex 1H and 13C NMR spectra suggest that the cis isomer is the largely major isomer of this compound. The cis structure is confirmed by X-ray diffraction. Besides the reaction on the P–Cl functions, the reaction on the lone pair of ClP(NMeNMe)2PCl is carried out, leading to the complex (p-cymene)Cl2RuPCl(NMeNMe)2ClPRuCl2(p-cymene). Characterization of this compound by X-ray diffraction displays a cis isomer for this compound also.

Abstract

International audience; A simple strategy was developed to obtain poly(ethylene glycol) (PEG)-based materials incorporating phosphonate functions and thiophosphate groups from readily accessible symmetrical PEG compounds and trifunctional thiophosphates. These materials are obtained in a two-step procedure, via the formation of THF soluble, hyperbranched PEG-based polyazomethines that are subsequently functionalized with dimethylphosphite. The resulting materials are hydrolytically degradable, and upon exposure to aqueous solutions of silver acetate they afford stable colloidal solutions of silver.

Abstract

© 2014 American Chemical Society. Five novel generations of phosphorus dendrimers based on a cyclotriphosphazene core with stable TEMPO radicals end groups have been synthesized and studied by EPR, SQUID, 1H NMR, 31P NMR, FT-IR, and UV-vis spectroscopy. The nitroxyl radicals exhibit a strong exchange interaction, which depends on the dendrimer generation and the temperature. An |fms| = 2 transition has been observed in each generation in dilution conditions demonstrating the intramolecular origin of the radicals interaction. There exists a direct proportionality between the EPR |Δms| = 2 transition intensity and the number of radicals by generation; consequently, the utility of EPR for the determination of the substitution efficiency on dendrimers by paramagnetic species is quite good. From the UV-vis characterization, we have observed that the molar extinction coefficient value is also proportional to the number of TEMPO groups. The magnetic properties of the zero, first and fourth generation dendrimers studied by SQUID magnetometry show antiferromagnetic interaction between radicals., This work was supported by the DGI grant CONSOLIDERC (CTQ2006-06333), CSIC-PIF RAPCAM (PIF-08-017-3), AGAUR (2009-SGR-00516) and DGI grant POMAs (CTQ2010-19501). Abstract

The synthesis of phosphorus containing dendrimers is described up to the fifth generation (96 terminal groups). The surface chemistry of these dendrimers allows the grafting of various functionalities such as phosphate (6 O-P(O)(OEt)2 groups), aminophosphate (up to 96 N-P(O)(OEt)2 groups), aminophosphite (up to 96 N-P(OEt)2 groups), functionalized phosphonate (up to 96 HC(OH)P(O)(OR)2 (R = Et, (CH2)11CH3), up to 96 HC(N-Pr)P(O)(OEt)2, or up to 48 CH=CH-P(O)(OEt)2), phosphorus ylide (6 C(O)CH=P(Ph)3), carboxylic acid (up to 24 CH=CH-C(O)OH), and tetraazamacrocycle (up to 12 cyclam). These surface functionalities have been chosen due to the well known properties of the corresponding monomers and/or polymers in various areas such as adhesives, flame retardants, fuel additives, sequestring agents.
