Abstract

Adaptive optics is a relatively new field, yet it is spreading rapidly and allows new questions to be asked about how the visual system is organized. The editors of this feature issue have posed a series of question to scientists involved in using adaptive optics in vision science. The questions are focused on three main areas. In the first we investigate the use of adaptive optics for psychophysical measurements of visual system function and for improving the optics of the eye. In the second, we look at the applications and impact of adaptive optics on retinal imaging and its promise for basic and applied research. In the third, we explore how adaptive optics is being used to improve our understanding of the neurophysiology of the visual system.

Abstract

Since 2017, numerous activities have been undertaken to advance gender equity in physics in Canada. National programs, including the Dimensions charter of the Natural Sciences and Engineering Research Council, have increased awareness of equity, diversity, and inclusion issues. Donna Strickland won a Nobel prize in 2018, which also highlighted gender minorities in physics. The gender-based analysis of the Canada Research Chair program has led to an emphasis on accountability, transparency, and best practices in recruitment, hiring, and retention and has been mirrored by. which has positively impacted other university policies with respect to gender and underrepresented minorities. Abstract

Background: In humans and the naturally occurring canine model of Alzheimer's disease (AD), we and others observe amyloid deposits in the retina en face. We have previously shown that detecting the number of amyloid deposits with polarised light, using wide field, dye free retinal imaging, can predict the degree of severity of AD pathology in the brain. But some who take thin, transverse retinal sections find amyloid while others do not. Here, we report the surface area of retinal amyloid deposits in human and the canine model of AD. Method: Formalin‐fixed retinas from 29 individuals with a moderate to high likelihood of AD (with some comorbidities excluded) and 20 canines were flat‐mounted and imaged in florescence and polarized light (Fig 1). The full retina was imaged for 24 human and 16 canine retinas. For 4 canines, in‐vivo blue light fluorescent retinal images were collected before and after injection of an amyloid dye, CRANAD‐28. Deposits visible only after injection were counted. Fluorescence and polarization images were compared post‐mortem in a canine after an injection of CRANAD‐28. Amyloid deposits were segmented using custom methods. The total area of segmented deposits was then divided by the total retinal area imaged. Result: In retinas from individuals with AD brain pathology, the area of the retinal covered by amyloid deposits was on average 0.01% and correlated with the number of deposits (Fig 2). In vivo (Fig 3), the area of the canine retina covered by amyloid deposits was on average 0.29% (Fig 4). In one ex vivo retina, fluorescent and polarization positive deposits had a concurrence of 93% and the average area of fluorescent deposits (0.14%) was larger than the same deposits imaged in polarimetry (0.08%) (Fig. 5). Conclusion: These findings and our previous report of an almost constant density of amyloid deposits across the retina, should guide in vivo imaging of amyloid in the retina as a biomarker of AD. Across all human and canine retinas, amyloid deposits covered less than 0.6% of the retinal surface. Careful, extensive sampling is needed. This paper considers the ways in which Propertius both makes and undermines his lover-poet's claims of elegiac fidelity in 3.20. This poem depicts the elegiac relationship as simultaneously marriage-like and non-marital by using the language of fidelity, drawing on both its traditional associations with legal ceremonies and agreements and its elegiac sense. Propertius uses the lover-poet's relationship to fides to have him occupy multiple and contradictory male roles in the poem. Ultimately, he fails to conform to any of them, which destabilizes the gender roles of the characters in the poem., Propertius 3.20 is a complex poem that has received relatively little critical attention. Abstract

In the last 20 years, research focused on developing retinal imaging as a source of potential biomarkers for Alzheimer's disease and other neurodegenerative diseases, has increased significantly. The Alzheimer's Association and the Alzheimer's & Dementia: Diagnosis, Assessment, Disease Monitoring editorial team (companion journal to Alzheimer's & Dementia) convened an interdisciplinary discussion in 2019 to identify a path to expedite the development of retinal biomarkers capable of identifying biological changes associated with AD, and for tracking progression of disease severity over time. As different retinal imaging modalities provide different types of structural and/or functional information, the discussion reflected on these modalities and their respective strengths and weaknesses. Discussion further focused on the importance of defining the context of use to help guide the development of retinal biomarkers. Moving from research to context of use, and ultimately to clinical evaluation, this article outlines ongoing retinal imaging research today in Alzheimer's and other brain diseases, including a discussion of future directions for this area of study.

49. Effects of periconceptional undernutrition on the initiation of parturition in sheep

Abstract

In sheep, parturition is initiated by increased fetal hypothalamic-pituitary-adrenal axis (HPAA) activity leading to PG[E.sub.2] and PG[F.sub.2[alpha]] production and a rise in the 17[beta]-estradiol-progesterone ([E.sub.2]/[P.sub.4]) ratio. Uteroplacental PG production can also increase fetal HPAA activity. Periconceptional maternal undernutrition accelerates fetal HPAA maturation resulting in preterm labor. We determined whether preterm labor was preceded by an increase in PG concentrations and [E.sub.2]/[P.sub.4] ratio and whether these increases preceded or followed the corresponding rise in cortisol concentrations. Singleton-bearing ewes were nourished ad libitum (N, n = 9) or undernourished (UN, n = 10) to reduce maternal weight by 15% from -61 days (d) to +30 d after mating with ad libitum intake thereafter. Paired maternal and fetal blood samples were collected from 126 d until delivery. Half the UN group delivered prematurely (>2 SD below mean gestation for the flock). PG and cortisol concentrations and [E.sub.2]/[P.sub.4] ratio increased before delivery in the same way in both groups. However, the increases occurred 7-10 d earlier in UN than in N animals. In both UN and N fetuses cortisol concentrations rose before fetal and maternal PG concentrations and maternal [E.sub.2]/[P.sub.4] ratio. Periconceptional maternal undernutrition induces preterm delivery in sheep by advancing the expected prepartum rise in cortisol and PG concentrations and [E.sub.2]/[P.sub.4] ratio. The rise in fetal cortisol concentration precedes the rise in fetal and maternal PG concentrations and maternal [E.sub.2]/[P.sub.4] ratio, suggesting that the underlying mechanism is likely to be acceleration of fetal HPAA maturation, resulting in initiation of the normal process of parturition. hypothalamic-pituitary-adrenal axis; prostaglandin; cortisol; estrogen-progesterone ratio
