- Abstract
Alzheimer’s disease (AD) is the leading cause of dementia in older adults. Although AD progression is characterized by stereotyped accumulation of proteinopathies, the affected cellular populations remain understudied. Here we use multiomics, spatial genomics and reference atlases from the BRAIN Initiative to study middle temporal gyrus cell types in 84 donors with varying AD pathologies. This cohort includes 33 male donors and 51 female donors, with an average age at time of death of 88 years. We used quantitative neuropathology to place donors along a disease pseudoprogression score. Pseudoprogression analysis revealed two disease phases: an early phase with a slow increase in pathology, presence of inflammatory microglia, reactive astrocytes, loss of somatostatin+inhibitory neurons, and a remyelination response by oligodendrocyte precursor cells; and a later phase with exponential increase in pathology, loss of excitatory neurons and Pvalb+and Vip+inhibitory neuron subtypes. These findings were replicated in other major AD studies.

- Abstract
Background: While traditional histologic neuropathological assessment of neurodegenerative disease has tremendous diagnostic value, insights into neurodegeneration's pathophysiological underpinnings can be significantly enhanced with large‐scale quantitative analyses. As part of the Seattle Alzheimer's Disease Brain Cell Atlas (SEA‐AD), we present a neuropathology dataset across 84 brains spanning the spectrum of AD neuropathologic change donated by UW Alzheimer's Disease Research Center and Adult Changes in Thought study participants. Method: Human brain tissue was collected at rapid autopsy (postmortem interval <12 hours). One hemisphere (randomly selected) was embedded in alginate for uniform coronal slicing (4mm), with alternating slabs fixed in 10% neutral buffered formalin or frozen in a dry ice isopentane slurry. Superior and Middle Temporal Gyrus (MTG) was sampled from fixed slabs and subjected to standard processing, embedding, sectioning (5µm), histochemical (H&E/Luxol fast blue), and immunohistochemical (duplex pTau and pTDP‐43, Aβ and Iba1, and monoplex GFAP, a‐synuclein, and NeuN) labeling. HALO image analysis software (Indica Labs, Albuquerque NM) was used to define and quantify relevant cellular and pathological features within each cortical layer (Figures 1‐3). Result: 84 individuals were included in these analyses (Table 1). While quantitative metrics of pTau and Aβ aligned with traditional neuropathological measures, they showed substantial variability within discrete diagnostic stages (Figure 4). These findings were robust to quantification (discrete count vs. total area stained) and normalization (total nuclei vs. total area analyzed) methods, and highlight the importance of deploying highly quantitative, continuous measures of pathology in the study of neurodegeneration. This unique resource provides spatially preserved measures of pathologic burden across the spectrum of AD with potential for correlative analysis across pathologies, clinical and biomarker parameters, and omics applications to better support the identification of vulnerable cell types in early stages of AD progression, the principal goal of SEA‐AD. Conclusion: Here we present a large‐scale quantitative neuropathology dataset, which enables analysis of pathological proteins and cell types at unprecedented scale and accuracy. Little is known about the interactions of AD‐relevant cell types with pathologic peptides across disease progression. The presented analyses exemplify the potential of this unique publicly‐accessible resource to identify and quantify novel disease‐relevant relationships. [ABSTRACT FROM AUTHOR]

- Abstract
The armed conflict in Colombia has caused more than 8,746,541 survivors and psychosocial effects and incidence on mental health in those affected. The advances obtained in the improvement of the psychological well-being of the survivors by the State are not clear; however, the literature shows that the socio-political conditions surrounding the survivors are unfavorable. Therefore, the study evaluated the psychological well-being of the population victim of the armed conflict in seven municipalities of Colombia, benefited by some measure of attention, assistance and reparation of law 1448 of 2011. Therefore, a cross-sectional descriptive study was used and Ryff's psychological well-being scale translated into Spanish; The following results were obtained: the dimensions with the best evaluation were "purposes in life" and "personal growth"; dimensions with lower ratings are "positive relationships" and "autonomy". it is concluded that psychological well-being is better perceived in terms of setting goals for oneself and showing improvement, but that autonomy is affected, as well as positive relationships (distrust, building new relationships when one is displaced) due to related issues with the armed conflict. The findings will improve the attention actions., El conflicto armado en Colombia ha causado más de 8.746.541 sobrevivientes y efectos psicosociales e incidencia sobre la salud mental en los afectados. Los avances obtenidos en el mejoramiento del bienestar psicológico de los sobrevivientes por parte del Estado no son claros; sin embargo, la literatura evidencia que las condiciones socio-políticas que rodean a los sobrevivientes son poco favorables. Por lo expuesto, el estudio evaluó el bienestar psicológico de la población víctima del conflicto armado de siete municipios de Colombia, beneficiada por alguna medida de atención, asistencia y reparación de la ley 1448 de 2011. Por lo anterior, se utilizó un estudio descriptivo transversal y la escala de Bienestar psicológico de Ryff traducida al español; se obtuvieron los siguientes resultados: las dimensiones con mejor valoración fueron “propósitos en la vida” y “crecimiento personal”; dimensiones con calificaciones más bajas son “relaciones positivas” y “autonomía”. se concluye que el bienestar psicológico es percibido mejor en función de plantearse metas para sí mismo y mostrar una mejora, pero que la autonomía se ve afectada, así como las relaciones positivas (desconfianza, construcción de relaciones nuevas cuando se es desplazado) por asuntos relacionados con el conflicto armado. Los hallazgos permitirán mejorar las acciones de atención.

- Abstract
Background: The trace of memory has not been well studied in middle temporal gyrus (MTG). Although neuronal loss is minimal in most cortical regions of the aging brain, changes in synaptic physiology of ageing neurons may contribute to altered connectivity. In the Adult Changes in Thought (ACT) autopsy cohort, we explored if memory decline is associated with NeuN (neuronal nuclear protein), a biomarker for mature neurons, in MTG. Method: ACT study participants were 5,763 older adults randomly sampled from an integrated healthcare delivery system in King County, Washington. We standardized composite memory scores to N(0,1) at baseline. We calculated slopes of memory decline over time using mixed‐effects models, where time was parameterized as years before death. Among 69 individuals who underwent brain autopsy, we measured NeuN positive cell count from whole slide image analysis using HALO in five tissue layers (molecular, external granular, external pyramidal, internal granular, internal pyramidal + multiform) from brain tissue in MTG. In each layer, we normalized NeuN positive cell count over the tissue area. We derived an average NeuN positive cell count (ANPCC) by averaging the values across the layers (Figure 1). We regressed ANPCC on memory slope, adjusting for age at death and sex. In secondary models, we added disease severity (Braak staging) and presence of APOE ε4 alleles separately. We also analyzed layer‐specific ANPCC. Result: 64 individuals were included for analysis (Table 1). Memory decline in this sample was 0.10 units per year, similar to the entire cohort. Slower decline in memory (β = 0.10; p‐value = 0.002) was significantly associated with increased average ANPCC (Table 2). Adjusting for Braak staging and APOE ε4 gave similar results. Findings were statistically significant across all but the molecular layer. Conclusion: ANPCC is associated with slower memory decline in multiple neocortical layers in MTG, irrespective of disease staging or APOE genotype. Since Alzheimer's disease afflicts adult neuronal populations, survival mechanisms in mature neurons are likely to be either reversed or circumvented during neurodegeneration. Further analyses are needed to understand the functional range of NeuN and processes that lead to loss of NeuN immunoreactivity in cells. [ABSTRACT FROM AUTHOR]

The article presents a study that aimed to identify and describe discourses about psychosocial components in intervention processes in Colombia. We analyzed 98 documents, classified as: a) State documents; b) Documents by Non-Governmental Organizations; c) Theoretical Documents on Psychosocial Intervention; and d) Theoretical documents on psychosocial components. Social Psychology was assumed as a disciplinary perspective and the notion of discourse as a theoretical and methodological element. Categorical content analysis was used to obtain the categories of analysis of the documents available in scientific databases and on websites of state institutions. The results focus on the adjective category. It was found that the use of the word psychosocial as an adjective "interchangeable", configures ambiguous and unclear expressions for the understanding of the notion, creating a redundant effect on their understanding. El presente trabajo tuvo como objetivo evaluar el El artículo se deriva de una investigación que buscaba identificar y describir los discursos acerca de lo psicosocial en los procesos de intervención en Colombia. Se analizaron 98 documentos, clasificados en: a) documentos del Estado; b) documentos de organizaciones No Gubernamentales; c) documentos teóricos de Intervención Psicosocial y d) documentos teóricos sobre lo psicosocial. Se asumió la Psicología Social como perspectiva disciplinar y la noción de discurso como elemento teórico y metodológico. Se utilizó el análisis de contenido categorial para obtener las categorías de análisis de los documentos disponibles en bases de datos científicas y en sitios web de instituciones estatales. Los resultados se centran en la categoría adjetivo. Se encontró que el uso de la palabra psicosocial como un adjetivo “intercambiable”, configura expresiones ambiguas y poco claras para el entendimiento de la noción, creando un efecto redundante en su comprensión. Es necesario que exista claridad conceptual acerca de lo psicosocial en los procesos de intervención en Colombia, declarando las bases teóricas que sustentan el uso de la noción.

La categoría Psicosocial se encontró como un elemento de uso común e indefinido en documentos de Intervención del Estado Colombiano, Ongs y en la producción de la comunidad académica. Derivado del uso acrítico que se hace del término, vinculándolo a diversos contextos sin un referente teórico claro, se propuso identificar y describir los discursos que se construyen sobre esta noción. El discurso es una práctica social, que tiene como función construir la realidad (Íñiguez y Antaki, 1994). Este estudio cualitativo y descriptivo analiza 98 documentos provenientes del Estado, Ongs y la Academia. Se utiliza el Atlas Ti para la codificación y generación de categorías y el Análisis de contenido para el análisis temático. Los resultados se componen de cuatro categorías de las cuales se derivan básicamente cuatro discursos: 1). Lo psicosocial se construye como algo pragmático. 2). Lo psicosocial es novedoso y su definición requiere consenso. 3). Lo psicosocial se construye como una categoría diferenciadora equiparada a lo integral. 4). Lo psicosocial se construye como una Tautología. El hallazgo de estos discursos es una importante contribución a la comprensión de lo psicosocial en los procesos de intervención. The study examined the effect of Avian Influenza (AI) on the household consumption of poultry products during the first outbreak of the disease in Ogun State, Nigeria. Furthermore, determinants influencing poultry products consumption during the outbreak were identified. A multi-stage sampling procedure was used to select 120 households and a well structured questionnaire was used for data collection. The data collected were analyzed using descriptive statistics, independent t-test and regression techniques. Egg consumption reduced significantly dropping from an average of 40 eggs/month to 29 eggs/month (p=0.038) during the AI outbreak. The average chicken expenditure/month dropped significantly from N1116.44 to N991.96 (p=0.019). Awareness of AI, educational level and amount spent on food consumption were the significant factors that contributed to the consumption of poultry products during the AI outbreak. The study therefore recommended that members of the public should be properly sensitized on safety precautions that should be cultivated before the consumption of poultry products especially during outbreaks of diseases. El ejercicio Bioético permite entender las problemáticas actuales y abordarlas desde diferentes perspectivas. Una de ellas tiene que ver con la educación de jóvenes y adolescentes, con lo que aprenden y como lo aprenden. Consciente de la aridez de un buen método sin un contenido que le de trascendencia, surge como consecuencia el interés por ofrecer espacios a reflexiones que vayan más allá de lo establecido por las ciencias exactas, considerando la posibilidad de integrar la Biología con la Ética y a través de esa alianza abrir la posibilidad de enseñar en los colegios no solo la funcionalidad de los organismos, sino también permitir el espacio para reflexiones alrededor de la pregunta por la vida, es decir de su significado, su valor y cómo esta se construye. Ese es pues el origen de la reflexión que se encuentra en estas páginas y que desea convertirse en una propuesta para abordar desde una perspectiva más amplia la enseñanza de la Biología en la Educación Media, teniendo en cuenta que es esta la ciencia que se ocupa del estudio de los seres vivos y que los espacios académicos ofrecidos en los establecimientos educativos para reflexionar sobre lo que esta significa, son cada vez más reducidos. Bioethics contributes an approach to understanding current issues from different perspectives. Teens and young adult education, what they learn and how they learn it, is paramount. Given that a good method that lacks substantial content is arid, it has become imperative to offer spaces for reflection that goes beyond what has been established by exact sciences, considering the possibility to integrate Biology and Ethics, and through such alliance, opening possibilities at school to teach not only about organisms and their functionality, but also providing opportunities for discussion on the question of life, its meaning, value and how it is built. This is from where the present reflection emerges and aims at developing into a proposal for the teaching of Biology in Secondary schools, bearing in mind that this Science is concerned with studying living beings and that educational institutions offer less and less spaces for reflection on its significance. Molecular heterogeneity in human breast cancer has challenged diagnosis, prognosis, and clinical treatment. It is well known that molecular subtypes of breast tumors are associated with significant differences in prognosis and survival. Assuming that the differences are attributed to subtype-specific pathways, we then suspect that there might be gene regulatory mechanisms that modulate the behavior of the pathways and their interactions. In this study, we proposed an integrated methodology, including machine learning and information theory, to explore the mechanisms. Using existing data from three large cohorts of human breast cancer populations, we have identified an ensemble of 16 master regulator genes (or MR16) that can discriminate breast tumor samples into four major subtypes. Evidence from gene expression across the three cohorts has consistently indicated that the MR16 can be divided into two groups that demonstrate subtype-specific gene expression patterns. For example, group 1 MRs, including ESR1, FOXA1, and GATA3, are overexpressed in luminal A and luminal B subtypes, but lowly expressed in HER2-enriched and basal-like subtypes. In contrast, group 2 MRs, including FOXM1, EZH2, MYBL2, and ZNF695, display an opposite pattern. Furthermore, evidence from mutual information modeling has congruently indicated that the two groups of MRs either up- or down-regulate cancer driver-related genes in opposite directions. Furthermore, integration of somatic mutations with pathway changes leads to identification of canonical genomic alternations in a subtype-specific fashion. - Abstract
Alzheimer's disease (AD) is the most common cause of dementia in older adults. Neuropathological and imaging studies have demonstrated a progressive and stereotyped accumulation of protein aggregates, but the underlying molecular and cellular mechanisms driving AD progression and vulnerable cell populations affected by disease remain coarsely understood. The current study harnesses single cell and spatial genomics tools and knowledge from the BRAIN Initiative Cell Census Network to understand the impact of disease progression on middle temporal gyrus cell types. We used image-based quantitative neuropathology to place 84 donors spanning the spectrum of AD pathology along a continuous disease pseudoprogression score and multiomic technologies to profile single nuclei from each donor, mapping their transcriptomes, epigenomes, and spatial coordinates to a common cell type reference with unprecedented resolution. Temporal analysis of cell-type proportions indicated an early reduction of Somatostatin-expressing neuronal subtypes and a late decrease of supragranular intratelencephalic-projecting excitatory and Parvalbumin-expressing neurons, with increases in disease-associated microglial and astrocytic states. We found complex gene expression differences, ranging from global to cell type-specific effects. These effects showed different temporal patterns indicating diverse cellular perturbations as a function of disease progression. A subset of donors showed a particularly severe cellular and molecular phenotype, which correlated with steeper cognitive decline.
