16. A preliminary identification of Rf*-A619, a novel restorer gene for CMS-C in maize (Zea mays L.)

Abstract

C-type cytoplasmic male sterility (CMS-C) is widely utilized for hybrid maize seed production. However, genetic mechanisms underlying the fertility restoration are very complicated. At present, there is a divergence on the number of fertility restorer genes in maize inbred line A619 for CMS-C. To further elucidate the restoring mechanism of A619, we used genetic analysis and molecular markers to confirm the restorer genes of maize inbred line A619 for C-type male sterile line C48-2 in this study. Firstly, the fertility segregations of (C48-2 × A619)F2 populations were investigated under three environments during 2013–2015. The segregation ratio of fertile and sterile plants in the F2 population fit to 15:1 via chi-square test and this result suggested that there are two dominant restorer genes in A619 for CMS-C, i.e., Rf4 and a novel gene named Rf*-A619. Next, based on the sequence differences between Rf4 and its recessive allelic rf4, a novel dominant marker F2/R2 was developed and validated to genotyping Rf4 in the F2 population. Through genotypic analysis, we found that there were a certain amount of fertile individuals without Rf4 which accounted for 3/16 in the F2 population via chi-square test at the 0.05 level. These results provided another proof to sustain that the inbred line A619 contains one additional restorer gene for CMS-C fertility restoration except Rf4. At last, we used one SSR marker which is tightly linked with the dominant restorer gene Rf5 to analyze those fertile plants without Rf4 in the F2 population. The PCR amplification results showed that Rf*-A619 is not allelic to Rf5 but a novel restorer gene for CMS-C. These results not only provide a basis for the mapping and characterization of a novel restorer gene but also give a new insight into the mechanism of CMS-C fertility restoration.

Abstract

To transfer desirable characters from perennial teosinte [Zea perennis (Hitchc.) Reeves & Mangelsd.] into maize (Z. mays L.), we have generated an interspecific hybrid and its backcross generations (B[C.sub.1][F.sub.3]). The maize x perennial teosinte B[C.sub.1][F.sub.3] (MZI202) resembled maize, with chromosome number 2n = 20. MZI202 was studied by multicolor genomic in situ hybridization (McGISH) and simple-sequence repeat (SSR) microsatellites markers. The McGISH experiments provided direct evidence that MZI202 was a maize-perennial teosinte substitution line with introgression of three alien chromosomes from perennial teosinte. Abstract

Abstract Background Little is known about the potential of Brachypodium distachyon as a model for low temperature stress responses in Pooideae. The ice recrystallization inhibition protein (IRIP) genes, fructosyltransferase (FST) genes, and many C-repeat binding factor (CBF) genes are Pooideae specific and important in low temperature responses. Here we used comparative analyses to study conservation and evolution of these gene families in B. distachyon to better understand its potential as a model species for agriculturally important temperate grasses. Results Brachypodium distachyon contains cold responsive IRIP genes which have evolved through Brachypodium specific gene family expansions. A large cold responsive CBF3 subfamily was identified in B. distachyon, while CBF4 homologs are absent from the genome. No B. distachyon FST gene homologs encode typical core Pooideae FST-motifs and low temperature induced fructan accumulation was dramatically different in B. distachyon compared to core Pooideae species. Conclusions We conclude that B. distachyon can serve as an interesting model for specific molecular mechanisms involved in low temperature responses in core Pooideae species. However, the evolutionary history of key genes involved in low temperature responses has been different in Brachypodium and core Pooideae species. These differences limit the use of B. distachyon as a model for holistic studies relevant for agricultural core Pooideae species.

35. ZmCals12 impacts the maize growth and development by regulating symplastic transport

Abstract

Carbohydrate partitioning from source to sink tissues is critical for plant growth and development. However, in maize (Zea maysL.), the molecular mechanisms of callose synthase gene regulating this process have seldom been reported. Here, we show that the mutation of maize callose synthase12(ZmCals12) caused increased accumulation of carbohydrate in the photosynthetic leaves but reduced carbohydrate content in sink tissues, which led to plant dwarfing and male sterile. Histochemical GUS activity assay and mRNA in situhybridization (ISH) indicated that ZmCals12was mainly expressed in the vascular transport system. Loss-of-function of ZmCals12 reduced callose synthase activity and callose deposition in plasmodesmatas (PDs) and around phloem cells (PCs) of vascular bundle. The drop-and-see (DANS) assay revealed that the PD permeability in the photosynthetic cells and the transport competence of leaf veins were reduced in the Zmcals12mutants, which led to the reduced symplastic transport. Paraffin section experiment revealed that less-developed vascular cells (VCs) in the Zmcals12mutants potentially disturbed sugar transport, thus resulting in the pleiotropic phenotype of the Zmcals12mutants. In addition, the impaired sugar transport hindered the internode development by inhibiting auxin (IAA) biosynthesis and signaling in the Zmcals12mutant. Collectively, our results provide insights into the mechanism of ZmCals12-mediated callose deposition and symplastic transport governing maize growth and development.

36. A novel semi-dominant allele of the transmembrane NAC transcription factor ZmNTL2reduces the size of multiple maize organs

Abstract

Maize plant architecture influences planting density and, in turn, grain yield. Most of the plant architecture-related traits can be described as organ size. We describe a miniature maize mutant, Tiny plant 4(Tip4), which exhibits reduced size of multiple organs and exhibits a semi-dominant monofactorial inheritance characteristic. Positional cloning confirmed that a 4-bp deletion in the NAC TF with transmembrane motif 1-Like (NTL) gene ZmNTL2, denoted as ZmNTL2Δ, confers the Tip4mutation. qRT-PCR showed that ZmNTL2was expressed in all tested tissues. ZmNTL2 functions as a transcriptional activator and is located in both the nucleus and biomembranes. The mutation does not affect the mRNA abundance of ZmNTL2locus, but it does result in the loss of transmembrane domain and confines the ZmNTL2Δprotein to the nucleus. Knocking out ZmNTL2has no effect on maize organ size development, indicating that the 4-bp deletion might be a gain-of-function mutation in organ size regulation. Combining transcriptome sequencing with cytokinin and auxin content determination suggests that the decreased organ size may be possibly mediated by changes in hormone homeostasis.

Abstract

Abstract: Grey leaf spot (GLS) is a global maize leaf disease that seriously endangers maize production. Discovering and utilizing genetic loci for GLS resistance would be useful for breeding new varieties with improved resistance. In this study, 233 F2:3 families (produced from the susceptible inbred line 08‐641 × the resistant inbred line 446) were used for quantitative trait locus (QTL) mapping of resistance to GLS. Five GLS resistance QTLs were detected on chromosomes 1, 2, 3, 4, and 6, which explained 6.7%‐21.3% of the phenotypic variation. The QTLs, qRgls.CH‐4, qRgls.CH‐1, qRgls.CH‐2, and qRgls.CH‐6, were stably expressed in the four environments, and all loci for GLS resistance were derived from the resistant parent, 446. The additive effects of qRgls.CH‐4, qRgls.CH‐1, and qRgls.CH‐6 were significantly greater than their single dominant effects, which may be beneficial for GLS resistance breeding. The QTL qRgls.CH‐6, located in bins 6.02–6.05, did not overlap with any previously reported resistance QTL and thus was identified here for the first time. QTL analysis of PI (leaf performance index) detected three leaf function QTLs on chromosomes 4, 8, and 9 were related to GLS resistance and explained 4.8%‐6.2% of the phenotypic variation. Among them, qPI.CH‐4 was significantly stronger expressed in several environments; this allele associated with increased leaf function came from the resistant parent, 446, and its interval overlapped with that of qRgls.CH‐4. Furthermore, both qRgls.CH‐4 and qPI.CH‐4 were located in a hotspot area for GLS resistance in bins 4.05‐4.06, indicating that GLS resistance was significantly related to leaf performance and that GLS significantly reduced leaf photosynthetic performance.
