Aggregation of protein-based therapeutics can occur during development, production, or storage and can lead to loss of efficacy and potential toxicity. Native mass spectrometry of a covalently linked pentameric monoclonal antibody complex with a mass of ∼800 kDa reveals several distinct conformations, smaller complexes, and abundant higher-order aggregates of the pentameric species. Charge detection mass spectrometry (CDMS) reveals individual oligomers up to the pentamer mAb trimer (15 individual mAb molecules; ∼2.4 MDa) whereas intermediate aggregates composed of 6-9 mAb molecules and aggregates larger than the pentameric dimer (1.6 MDa) were not detected/resolved by standard mass spectrometry, size exclusion chromatography (SEC), capillary electrophoresis (CE-SDS), or by mass photometry. Conventional quadrupole time-of-flight mass spectrometry (QTOF MS), mass photometry, SEC, and CE-SDS did not resolve partially or more fully unfolded conformations of each oligomer that were readily identified using CDMS by their significantly higher extents of charging. Trends in the charge-state distributions of individual oligomers provides detailed insight into how the structures of compact and elongated mAb aggregates change as a function of aggregate size. These results demonstrate the advantages of CDMS for obtaining accurate masses and information about the conformations of large antibody aggregates despite extensive overlapping m/z values. These results open up the ability to investigate structural changes that occur in small, soluble oligomers during the earliest stages of aggregation for antibodies or other proteins.

This paper explores an application of 3D printing technology on the food industry. Since its inception in the 1980s, 3D printing has experienced a huge rise in popularity. This study uses cost-effective, flexible, and sustainable components that enable specific features of certain gas chromatography. This study aims to optimize the process of gas detection using a 3D printed separation column and the MiCS-6814 sensor. The principle of the entire device is based on the idea of utilizing a simple capillary chromatographic column manufactured by 3D printing for the separation of samples into components prior to their measurement using inexpensive chemiresistive sensors. An optimization of a system with a 3D printed PLA block containing a capillary, a mixing chamber, and a measuring chamber with a MiCS-6814 sensor was performed. The optimization distributed the sensor output signal in the time domain so that it was possible to distinguish the peak for the two most common alcohols, ethanol and methanol. We evaluated the utility of transmission electron microscopy (TEM) in transbronchial lung cryobiopsy (TBLC) samples from 16 consecutive patients undergoing routine evaluation of fibrotic interstitial lung disease (ILD). Next to routine pathology examination, 1 to 2 TBLC samples were prepared for TEM analysis and evaluated using a Zeiss LEO EM 910. Subpleural cryobiopsies and unfrozen excision biopsies from fresh lobectomy tissue of non-ILD lung cancer patients served as controls. TEM provided high-quality images with only minor cryoartifacts as compared to controls. Furthermore, in several ILD patients we found marked microvascular endothelial abnormalities like luminal pseudopodia-like protrusions and inner surface defects. These were extensively present in four (25%), moderately present in seven (43.8%), and largely absent in five (31.3%) patients. A higher degree of TEM endothelial abnormalities was associated with younger age, non-specific interstitial pneumonia pattern, higher broncho-alveolar lavage lymphocyte count, positive autoantibodies, and lower spirometry, diffusion capacity and oxygenation biomarkers. We conclude that TEM evaluation of TBLC samples from ILD patients is feasible, while the observed microvascular alterations warrant further evaluation. [ABSTRACT FROM AUTHOR]

In the brain, a microvascular sensory web coordinates oxygen delivery to regions of neuronal activity. This involves a dense network of capillaries that send conductive signals upstream to feeding arterioles to promote vasodilation and blood flow. Although this process is critical to the metabolic supply of healthy brain tissue, it may also be a point of vulnerability in disease. Deterioration of capillary networks is a feature of many neurological disorders and injuries and how this web is engaged during vascular damage remains unknown. We performed in vivo two-photon microscopy on young adult mural cell reporter mice and induced focal capillary injuries using precise two-photon laser irradiation of single capillaries. We found that ~59% of the injuries resulted in regression of the capillary segment 7 to 14 d following injury, and the remaining repaired to reestablish blood flow within 7 d. Injuries that resulted in capillary regression induced sustained vasoconstriction in the upstream arteriole-capillary transition (ACT) zone at least 21 days postinjury in both awake and anesthetized mice. The degree of vasomotor dynamics was chronically attenuated in the ACT zone consequently reducing blood flow in the ACT zone and in secondary, uninjured downstream capillaries. Background Hemoglobin (Hb) Hekinan II (A27; Glu-Asp) is an α-chain variant, and its interaction with the common Southeast Asian (--SEA/) α-thalassemia (α-thal) deletion is rarely reported. This study provides a clinical update of Hb Hekinan II associated with (--SEA/) α-thal. Methods A total of 11 simple heterozygotes and 20 composite heterozygotes for Hb Hekinan II and (--SEA/) α-thal were included based on molecular diagnosis. Results Hb Hekinan II exhibited a significant increase in hemoglobin, mean corpuscular volume, and mean corpuscular hemoglobin content, but a decrease in red blood cell level compared with α+ thalassemia deletion. Compared with (--SEA/) α-thal, composite heterozygotes for Hb Hekinan II and (--SEA/) α-thal showed similar erythrocyte parameters. Both heterozygotes with and without (--SEA/) α-thal showed low Hb A2 level. Hb Hekinan II showed abnormal performance in high-performance liquid chromatography but not in capillary electrophoresis. Conclusion Hb Hekinan II is a benign Hb variant. The heterozygotes exhibit clinically asymptomatic coinheritance with (--SEA/) α-thal having comparable hematological phenotype to simple (--SEA/) α-thal. Objectives Voxelotor can increase hemoglobin levels in patients living with sickle cell disease (SCD). A clinician who is monitoring voxelotor response may want to know whole-blood voxelotor concentration, but this cannot be measured in most clinical settings. However, voxelotor has been demonstrated to cause "peak splitting" in common methods of hemoglobin measurement such as capillary zone electrophoresis (CZE) and high-performance liquid chromatography (HPLC). We hypothesized that we could use the size of the peak split to estimate the whole-blood concentration. Methods Blood from people with SCD was dosed with known concentrations of voxelotor, and multiparameter regression was used to derive the relationship of voxelotor concentration to the degree of peak splitting observed. To validate these equations, 21 patients started on voxelotor at 1500 mg/d had blood samples drawn at days 0, 14, 30, and 60. Samples were sent out for gold standard voxelotor concentration testing. The derived equations were then used to calculate voxelotor concentration. Results Calculated concentrations correlated strongly with measured concentrations for both CZE (R 2 = 0.83, P <.001) and HPLC (R 2 = 0.76, P <.001). Voxelotor concentration also had a significant effect on increases in hemoglobin (R 2 = 0.40, P <.001). Conclusions Thus, peak splitting CZE and HPLC can be used to estimate voxelotor concentration. To conduct experimental investigations of the drying process of a closed container, taking into account the opening of a leak channel in a weld, an experimental research stand, an experimental container and experimental samples with model leaks of different sizes are designed and manufactured. The scientific problem of experimental research has been formulated, a program and methods for conducting experiments have been developed. Requirements for metrological equipment have been formed and the measurement error of temperature, humidity and pressure has been determined. Preliminary experiments are carried out on wetting the experimental container and experimental samples with a model liquid and clogging the leak channels, as well as on drying the inner surface of the experimental container wall with a convective gas flow and conductive heating of the wall until the leak channels clogged with liquid are opened. The dependences of the change in temperature of the liquid, air, wall of the experimental container, experimental samples and pressure in the experimental container are obtained over the time of the experiment during wetting and drying of the experimental container and experimental samples. The parameters of thermal exposure at which the leak channels is opened within a specified time are determined.

A deep understanding of the characteristics and mechanism of geyser boiling and capillary pumping is necessary to optimize a high-temperature sodium heat pipe. In this work, the Volume of Fluid (VOF) two-phase model and the capillary force model in the mesh wick were used to model the complex phase change and fluid flow in the heat pipe. Computational Fluid Dynamics (CFD) simulations successfully predicted the process of bubble nucleation, growth, aggregation, and detachment from the wall in the liquid pool of the evaporation section of the heat pipe in horizontal and tilted states, as well as the reflux phenomenon of capillary suction within the wick. The accuracy and stability of the capillary force model within the wick were verified. In addition, the causes of geyser boiling in heat pipes were analyzed by extracting the oscillation distribution of heat pipe wall temperature. The results show that adding the capillary force model within the wick structure can reasonably simulate the liquid backflow phenomenon at the condensation; Under the horizontal and inclined operating conditions of the heat pipe, the phenomenon of local dry-out will occur, resulting in a sharp increase in local temperature. The speed of bubble detachment and the timely reflux of liquid sodium (condensate) replenishment in the wick play a vital role in the geyser temperature oscillation of the tube wall. The numerical simulation method and the results of this study are anticipated to provide a good reference for the investigation of geyser boiling in high-temperature heat pipes.

This study explores the use of capillary blood sampling as an alternative method for monitoring kidney transplant patients. The findings indicate that capillary sampling can provide accurate results for creatinine and tacrolimus levels, which are important indicators of kidney function. However, there are some challenges, such as the need to address clotting issues and the need for higher precision in measuring tacrolimus values. Despite these limitations, many patients expressed a preference for capillary sampling over venous blood tests. The study suggests that further research and the development of home sampling protocols are necessary to overcome these practical and biochemical issues. [Extracted from the article]

Viscosity is a thermophysical property of paramount importance, being essential for many scientific and industrial applications. The most common instruments for its measurement are glass capillary viscometers. Therefore, the use of capillary viscometers is widespread both in industry and in research. The range of viscosities of interest range from lower than that of water to several orders of magnitude higher values, the measurement of which requires different capillary viscometers. Most of the practical applications concern routine instruments, mainly for quality control. One main issue for the utilization of capillary viscometers relates to the need for their calibration, assuring its traceability to the water primary viscosity standard, to certify its worldwide validity. The present paper focuses on capillary instruments dedicated to perform viscosity measurements on Newtonian organic liquids at atmospheric pressure, as it is assumed that is the most widespread type of application for these viscometers. Capillary viscometry has a completely well-defined working equation, namely, the Hagen–Poiseuille equation. However, the practical performance of the measuring instruments deviates from that working equation. Most of those deviations are currently considered by many users. However, some of those deviations have not reached that status yet, like those concerning the effects due to the surface tension of the sample on the measurements. All these aspects are summarized and analyzed in the present article, together with a brief general description of the most common types of capillary viscometers, namely, the Ostwald and the constant-level or Ubbelohde instruments. Quantifying the formation and decomposition of amyloid is a crucial issue in the development of new drugs and therapies for treating amyloidosis. The current technologies for grasping amyloid formation and decomposition include fluorescence analysis using thioflavin-T, secondary structure analysis using circular dichroism, and image analysis using atomic force microscopy or transmission electron microscopy. These technologies typically require spectroscopic devices or expensive nanoscale imaging equipment and involve lengthy analysis, which limits the rapid screening of amyloid-degrading drugs. In this study, we introduce a technology for rapidly assessing amyloid decomposition using capillary flow-based paper (CFP). Amyloid solutions exhibit gel-like physical properties due to insoluble denatured polymers, resulting in a shorter flow distance on CFP compared to pure water. Experimental conditions were established to consistently control the flow distance based on a hen-egg-white lysozyme amyloid solution. It was confirmed that as amyloid is decomposed by trypsin, the flow distance increases on the CFP. Our method is highly useful for detecting changes in the gel properties of amyloid solutions within a minute, and we anticipate its use in the rapid, large-scale screening of anti-amyloid agents in the future. The freeze-thaw process is a weathering phenomenon that occurs in rocks and stones when water enters small cracks and pores within the stone. When the temperature drops below freezing, the water inside the rock expands as it freezes, exerting pressure on the rock from within. In this study the effect of freeze thaw on mechanical parameter uniaxial compression strength (UCS) and ultrasonic pulse velocity (UPV) of 47 oolitic samples were considered. At first, capillary absorption for cylindrical and prismatic oolitic samples were measured and then freeze-thaw experiment with 23 cycles were applied to the samples. UPV of samples before and after freeze-thaw was analysed by two ultrasonic devices including Geotron and Pundit. It was observed that freeze thaw decreases primary velocity (Vp) and secondary velocity (Vs) by around 20 and 7% respectively. However, average UCS of 1.6 MPa indicates strength decrease of samples, which represent negative effect of pore expansion due to freeze thaw on strength parameter of samples. [ABSTRACT FROM AUTHOR]

Objectives: We previously reported that heating of the neck or elbows alleviated Raynaud's phenomenon in patients with systemic sclerosis and upregulated capillary extension factor angiopoietin-1 (Angpt-1) in the fingertips. In this study, we investigated which cases responded better to the effect of heating of the neck or elbows. Methods: The pre- to postheating change in the visual analogue scale (ΔVAS) for Raynaud's phenomenon was examined for correlation with age, disease duration, autoantibodies, disease types, corticosteroid dose, capillaroscopic nailfold capillary damage, fingertip Angpt-1 concentrations at baseline, and increased rate of Angpt-1 concentration. Results: The ΔVAS for elbow heating correlated positively with the baseline Angpt-1 concentration, whereas opposite correlation was observed for neck heating. The other items did not significantly correlate with the ΔVAS; however, the ΔVAS for elbow heating tended to be larger in patients with advanced capillary damage, whereas an opposite trend was observed for neck heating. Conclusions: Elbow and neck heating alleviated Raynaud's phenomenon to a similar extent, but their mechanism was different. Inexpensive chemiresistive sensors are often insufficiently selective as they are sensitive to multiple components of the gas mixture at the same time. One solution would be to insert a device in front of the sensor that separates the measured gas mixture and possibly isolates the unwanted components. This study focused on the fabrication and characterization of a compact unit, which was fabricated by 3D printing, for the separation and detection of simple gas mixtures. The capillary, the basic part of the compact unit, was 4.689 m long and had a diameter of 0.7 mm. The compact unit also contained a mixing chamber on the inlet side and a measuring chamber with a MiCS-6814 sensor on the outlet side. Mixtures of ethanol and water at different concentrations were chosen for characterization. The measured calibration curve was found to have a reliability of R2 = 0.9941. Vascular permeability is temporarily heightened during inflammation, but excessive inflammation‐associated microvascular leakage can be detrimental, as evidenced in the inflamed lung. Formylated peptides regulate vascular leakage indirectly via formylated peptide receptor‐1 (FPR1)‐mediated recruitment and activation of neutrophils. Here we identify how the GTPase‐activating protein ARAP3 protects against formylated peptide‐induced microvascular permeability via endothelial cells and neutrophils. In vitro, Arap3−/− endothelial monolayers were characterised by enhanced formylated peptide‐induced permeability due to upregulated endothelial FPR1 and enhanced vascular endothelial cadherin internalisation. In vivo, enhanced inflammation‐associated microvascular leakage was observed in Arap3−/− mice. Leakage of plasma protein into the lungs of Arap3−/− mice increased within hours of formylated peptide administration. Adoptive transfer experiments indicated this was dependent upon ARAP3 deficiency in both immune and non‐immune cells. Bronchoalveolar lavages of formylated peptide‐challenged Arap3−/− mice contained neutrophil extracellular traps (NETs). Pharmacological inhibition of NET formation abrogated excessive microvascular leakage, indicating a critical function of NETs in this context. The observation that Arap3−/− mice developed more severe influenza suggests these findings are pertinent to pathological situations characterised by abundant formylated peptides. © 2024 The Authors. This paper describes the bearing performances of orifice and constant flow valve (CFV) restrictor-compensated conical journal bearings for variations in operating speed. The analytical studies for different cone angle bearings using FEM are represented in terms of different bearing characteristics such as maximum pressure, minimum fluid film thickness, fluid flow, stiffness, damping, and so on. The Reynolds equation for lubricant at the mating surfaces is solved to obtain these characteristics for the operating speed parameter range Ω = 0.1–1.0. The study reveals that the CFV conical journal bearing shows more significant results as the speed progresses than the orifice compensation. We sought to determine the physiological relevance of pannexin/purinergic‐dependent signaling in mediating conducted vasodilation elicited by capillary stimulation through skeletal muscle contraction. Using hamster cremaster muscle and intravital microscopy we stimulated capillaries through local muscle contraction while observing the associated upstream arteriole. Capillaries were stimulated with muscle contraction at low and high contraction (6 and 60CPM) and stimulus frequencies (4 and 40 Hz) in the absence and presence of pannexin blocker mefloquine (MEF; 10−5 M), purinergic receptor antagonist suramin (SUR 10−5 M) and gap‐junction uncoupler halothane (HALO, 0.07%) applied between the capillary stimulation site and the upstream arteriolar observation site. Conducted vasodilations elicited at 6CPM were inhibited by HALO while vasodilations at 60CPM were inhibited by MEF and SUR. The conducted response elicited at 4 Hz was inhibited by MEF while the vasodilation at 40 Hz was unaffected by any blocker. Therefore, upstream vasodilations resulting from capillary stimulation via muscle contraction are dependent upon a pannexin/purinergic‐dependent pathway that appears to be stimulation parameter‐dependent. Our data highlight a physiological importance of the pannexin/purinergic pathway in facilitating communication between capillaries and upstream arteriolar microvasculature and, consequently, indicating that this pathway may play a crucial role in regulating blood flow in response to skeletal muscle contraction. Introduction: Intramuscular hemangiomas (IMH) are rare benign vascular tumors. They can occur anywhere in the body but are more common in the head and neck. Their location in the muscles of the abdominal wall is uncommon. Case presentation: A 14-year-old girl presented with 5 months of progressive swelling on the right side of her abdomen followed by intermittent pain. On examination, a solitary mass with ill-defined borders was noted fixed to the subcutaneous tissue in the right lumbar quadrant. After elevating the right leg, the swelling became more prominent. Doppler ultrasonography revealed a mixed echogenic mass in the subcutaneous plane of the anterior abdominal wall of the right lumbar region. Magnetic resonance imaging of abdomen was suggestive of hemangioma, and the patient underwent surgical excision. Histopathological examination confirmed the diagnosis of capillary hemangioma. During follow-up at 3 and 6 months, the patient did not present with recurrence and was healthy. Conclusion: Although IMH is a rare soft-tissue tumor, it should be considered when diagnosing symptomatic intramuscular swelling.

The main trends in the use of liposomes or lipid aggregates as pseudostationary phases in capillary electrokinetic chromatography (EKC) over the past 15 years are reviewed. Due to the ability of liposomes to mimic cell membranes, they have found wide applications in analytical chemistry and especially in the field of biomedical sciences. Various liposomes, lipid aggregates, and liposomal formulations have been adopted as background electrolytes in EKC and ultraviolet detection is by far the most commonly used detection mode. The possibility to vary the type of lipids and their content in the lipid aggregates has increasingly expanded the usage of liposome EKC for solving specific tasks. The presented data shows that there is great potential of liposome EKC in investigations of interactions between lipid membranes and compounds and the applications are typically related to biomedical and pharmaceutical issues, but not limited to these.

Abstract Precise control of microparticle movement is crucial in high throughput processing for various applications in scalable manufacturing, such as particle monolayer assembly and 3D bio-printing. Current techniques using acoustic, electrical and optical methods offer precise manipulation advantages, but their scalability is restricted due to issues such as, high input powers and complex fabrication and operation processes. In this work, we introduce the concept of capillary wave tweezers, where mm-scale capillary wave fields are dynamically manipulated to control the position of microparticles in a liquid volume. Capillary waves are generated in an open liquid volume using low frequency vibrations (in the range of 10–100 Hz) to trap particles underneath the nodes of the capillary waves. By shifting the displacement nodes of the waves, the trapped particles are precisely displaced. Using analytical and numerical models, we identify conditions under which a stable control over particle motion is achieved. By showcasing the ability to dynamically control the movement of microparticles, our concept offers a simple and high throughput method to manipulate particles in open systems.

Background: Capillary blood glucose measurement is one of the most important diagnostic procedures in managing and treating patients, especially those with diabetes. Since the preparation method of the capillary sampling site affects the blood glucose (BG) level, it is essential to identify the best preparation method with the least error estimation. This study compares the capillary BG level with the venous BG level after preparing the sampling site with four different preparation methods. Methods: This quasi-experimental study has a single-group design. It was conducted on 85 nursing and midwifery students of Kashan University of Medical Sciences and Health Services, in 2022. The subjects who met the inclusion criteria were selected using convenience sampling. Capillary BG was measured from each finger using a glucometer while each finger was prepared with alcohol, water, soap and water, or no washing. Then, the venous blood sample of the same person was sent to the laboratory. The data were analyzed by SPSS software, version 22. [Display omitted] Achieving rapid capillary wetting is highly desirable in nature and industries. Previous endeavors have primarily concentrated on passive wetting strategies through surface engineering. However, these approaches are inadequate for high-viscosity fluids due to the significant viscous resistance, especially for non-Newtonian fluids. In contrast, forced wetting emerges as a promising method to address the challenges associated with achieving rapid wetting of non-Newtonian fluids in capillaries. To investigate the forced wetting behavior of viscoelastic fluids in capillaries, we employ Xanthan Gum (XG) aqueous solutions as target fluids with the storage modulus significantly exceeding the loss modulus. We utilize smooth glass capillaries connected to a syringe pump to achieve high moving speeds of up to 1 m/s. Our experiments reveal a significant distinction in the power-law exponent that governs the scaling relationship between the dynamic contact angle and velocity for viscoelastic fluids compared to Newtonian fluids. This exponent is considerably smaller and varies based on the concentration of viscoelastic fluids and the diameter of the capillaries. We suggest that the viscosity dominates the wetting dynamics of viscoelastic fluids, manifested by the contact line morphology-dependent behavior. Aims BMP9 is a high affinity ligand of ALK1 and endoglin receptors that are mutated in the rare genetic vascular disorder hereditary hemorrhagic telangiectasia (HHT). We have previously shown that loss of Bmp9 in the 129/Ola genetic background leads to spontaneous liver fibrosis via capillarization of liver sinusoidal endothelial cells (LSEC) and kidney lesions. We aimed to decipher the molecular mechanisms downstream of BMP9 to better characterize its role in vascular homeostasis in different organs. Methods and results For this, we performed an RNA-seq analysis on LSEC from adult WT and Bmp9 -KO mice and identified over 2000 differentially expressed genes. Gene ontology analysis showed that Bmp9 deletion led to a decrease in BMP and Notch signalling, but also LSEC capillary identity while increasing their cell cycle. The gene ontology term 'glomerulus development' was also negatively enriched in Bmp9 -KO mice vs. WT supporting a role for BMP9 in kidney vascularization. Through different imaging approaches (electron microscopy, immunostainings), we found that loss of Bmp9 led to vascular enlargement of the glomeruli capillaries associated with alteration of podocytes. Importantly, we also showed for the first time that the loss of Bmp9 led to spontaneous arteriovenous malformations (AVMs) in the liver, gastrointestinal tract, and uterus. Conclusion Altogether, these results demonstrate that BMP9 plays an important role in vascular quiescence both locally in the liver by regulating endothelial capillary differentiation markers and cell cycle but also at distance in many organs via its presence in the circulation. It also reveals that loss of Bmp9 is sufficient to induce spontaneous AVMs, supporting a key role for BMP9 in the pathogenesis of HHT. [ABSTRACT FROM AUTHOR]

In recent studies concerning walking robots, there has been a notable emphasis on investigating tribological phenomena and elucidating the role of capillary forces in tangential resistance. However, majority of these investigations have been restricted to specific geometric shapes, primarily spherical domes. In this research, we propose a novel approach based on the finite element method to address broader challenge of wet adhesion applicable to objects of various shapes. Additionally, we investigate the role of distributed domes on a soft foot in quantifying the tangential capillary force, which plays a crucial role in enhancing the performance of walking robots on wet surfaces. Through the optimization application, we identify that our designed foot (measuring 20 mm × 14 mm, comprising 78 domes with a radius of 0.5 mm and a distance of 1.5 mm between the two nearest domes) maximizes the value of the tangential capillary force. Moreover, we develop a finite element method using the Simulation Open Framework Architecture (SOFA) to model the interaction between wet liquid on a solid object's curved surface, enabling the calculation of wet adhesion force and capillary forces. Finally, to demonstrate the effectiveness of our proposed morphology-changeable soft pads foot, we conduct a presented a demonstration of a hexapod robot walking on different dry/wet condition of the surface. Degenerative effects of nerve tissues are often accompanied by changes in vascularization. In this regard, knowledge about hereditary cerebellar degeneration is limited. In this study, we compared the vascularity of the individual cerebellar components of 3-month-old wild-type mice (n = 8) and Purkinje cell degeneration (pcd) mutant mice, which represent a model of hereditary cerebellar degeneration (n = 8). Systematic random samples of tissue sections were processed, and laminin was immunostained to visualize microvessels. A computer-assisted stereology system was used to quantify microvessel parameters including total number, total length, and associated densities in cerebellar layers. Our results in pcd mice revealed a 45% (p < 0.01) reduction in the total volume of the cerebellum, a 28% (p < 0.05) reduction in the total number of vessels and a lower total length, approaching 50% (p < 0.001), compared to the control mice. In pcd mutants, cerebellar degeneration is accompanied by significant reduction in the microvascular network that is proportional to the cerebellar volume reduction therefore does not change density of in the cerebellar gray matter of pcd mice. The objective of this work was to develop an actinide-specific monolithic support in capillary designed to immobilize precise Pu:Am ratios and its coupling to inductively coupled plasma mass spectrometry (ICP-MS) for immobilized metal affinity chromatography applications. This format offers many advantages, such as reducing the sample amount and waste production, which are of prime importance when dealing with highly active radioelements. Four organic phosphorylated-based monoliths were synthesized in situ through UV photo-polymerization in capillary and characterized. The capillary coupling to ICP-MS was set up in conventional laboratory using Th and Sm as chemical analogues of Pu and Am. A dedicated method was developed to quantify online Th and Sm amounts immobilized on the monolithic capillaries, allowing to select the best monolith candidate poly(BMEP-co-EDMA)adp. By precisely adjusting the elemental composition in the loading solutions and applying the developed quantification method, the controlled immobilization of several Th:Sm molar ratios onto the monolith was successful. Finally, the capillary ICP-MS coupling was transposed in a glove box and by applying the strategy developed to design the monolithic support using Th and Sm, the immobilization of a 10.5 ± 0.2 (RSD = 2.3%, n = 3) Pu:Am molar ratio reflecting Pu ageing over 48 years was achieved in a controlled manner on poly(BMEP-co-EDMA)adp. Hence, the new affinity capillary monolithic support was validated, with only hundred nanograms or less of engaged radioelements and can be further exploited to precisely determine differential interactions of Pu and Am with targeted biomolecules in order to better anticipate the effect of Am on Pu biodistribution. Simple Summary: Maternal nutrition during gestation can have profound effects on fetal growth and development. Perturbations to maternal nutrition during this period can predispose to adverse health and productivity outcomes in postnatal life. A biochemical pathway present in cells that can be affected by limited maternal nutrition is one-carbon metabolism. This pathway is related to epigenetics, which regulates gene expression or the turning on and off genes. The one-carbon metabolites (OCM) folate, vitamin B12, methionine, and choline are important compounds in this pathway. Our research aimed to evaluate the effect of maternal nutrition during early pregnancy on fetal intestinal characteristics and assess if OCM supplementation could mitigate changes due to altered maternal nutrition. We discovered that OCM supplementation in nutrition-restricted dams enhanced vascularity development signaling, decreased small intestinal weight, and optimized intestinal cell growth, potentially helping the fetus adapt to a lower nutrient supply. To investigate the effects of nutrient restriction and one-carbon metabolite (OCM) supplementation (folate, vitamin B12, methionine, and choline) on fetal small intestine weight, vascularity, and cell proliferation, 29 (n = 7 ± 1 per treatment) crossbred Angus beef heifers (436 ± 42 kg) were estrous synchronized and conceived by artificial insemination with female sexed semen from a single sire. Then, they were allotted randomly to one of four treatments in a 2 × 2 factorial arrangement with the main factors of nutritional plane [control (CON) vs. restricted feed intake (RES)] and OCM supplementation [without OCM (−OCM) or with OCM (+OCM)]. Heifers receiving the CON level of intake were fed to target an average daily gain of 0.45 kg/day, which would allow them to reach 80% of mature BW by calving. Heifers receiving the RES level of intake were fed to lose 0.23 kg/heifer daily, which mimics observed production responses in heifers that experience a diet and environment change during early gestation. Targeted heifer gain and OCM treatments were administered from d 0 to 63 of gestation, and then all heifers were fed a common diet targeting 0.45 kg/d gain until d 161 of gestation, when heifers were slaughtered, and fetal jejunum was collected. Gain had no effect (p = 0.17) on the fetal small intestinal weight. However, OCM treatments (p = 0.02) displayed less weight compared to the −OCM groups. Capillary area density was increased in fetal jejunal villi of RES − OCM (p = 0.02). Vascular endothelial growth factor receptor 2 (VEGFR2) positivity ratio tended to be greater (p = 0.08) in villi and was less in the crypts (p = 0.02) of the RES + OCM group. Cell proliferation decreased (p = 0.02) in villi and crypts of fetal jejunal tissue from heifers fed the RES + OCM treatment compared with all groups and CON − OCM, respectively. Spatial cell density increased in RES − OCM compared with CON + OCM (p = 0.05). Combined, these data show OCM supplementation can increase expression of VEGFR2 in jejunal villi, which will promote maintenance of the microvascular beds, while at the same time decreasing small intestine weight and crypt cell proliferation. Pellets have voids in their structure, which form a pore space. Pellet pores can be represented by closed volumes or channels, capillaries. The volume of capillaries can be estimated by impregnating pellets with water. The purpose of this work is to assess the possibility of studying the pore space of burnt pellets using the capillary-porous solids model. For this purpose, samples of laboratory and industrial pellets were presented. According to the data obtained, samples with a high moisture capacity value have a higher recoverability. Background: Capillary blood glucose measurement is one of the most important diagnostic procedures in managing and treating patients, especially those with diabetes. Since the preparation method of the capillary sampling site affects the blood glucose (BG) level, it is essential to identify the best preparation method with the least error estimation. This study compares the capillary BG level with the venous BG level after preparing the sampling site with four different preparation methods. Methods: This quasi-experimental study has a single-group design. It was conducted on 85 nursing and midwifery students of Kashan University of Medical Sciences and Health Services, in 2022. The subjects who met the inclusion criteria were selected using convenience sampling. Capillary BG was measured from each finger using a glucometer while each finger was prepared with alcohol, water, soap and water, or no washing. Then, the venous blood sample of the same person was sent to the laboratory. The data were analyzed by SPSS software, version 22. The Friedman and Dunn post hoc tests were used to compare venous and capillary BG levels obtained using different methods. The significance level was set at P<0.05. Results: There were significant differences between venous BG levels and capillary BG levels in blood sampling site preparation with alcohol (P<0.05). Preparation with alcohol had the highest (7.34±27.03), and the soap and water had the lowest (-0.67±14.82) estimation errors. Conclusion: The use of alcohol to prepare the blood sampling site, which is a common practice in many healthcare facilities, may misrepresent BG levels and lead to misdiagnosis and treatment. Purpose: Diffusion MRI is a physical measurement method that quantitatively indicates the displacement of water molecules diffusing in voxels. However, there are insufficient data to characterize the diffusion process physically in a uniform structure such as a phantom. This study investigated the transitional relationship between structure scale, temperature, and diffusion time for simple restricted diffusion using a capillary phantom. Methods: We performed diffusion-weighted pulsed-gradient stimulated-echo acquisition mode (STEAM) MRI with a 9.4 Tesla MRI system (Bruker BioSpin, Ettlingen, Germany) and a quadrature coil with an inner diameter of 86 mm (Bruker BioSpin). We measured the diffusion coefficients (radial diffusivity [RD]) of capillary plates (pore sizes 6, 12, 25, 50, and 100 µm) with uniformly restricted structures at various temperatures (10°C, 20°C, 30°C, and 40°C) and multiple diffusion times (12-800 ms). We evaluated the characteristics of scale, temperature, and diffusion time for restricted diffusion. Results: The RD decayed and became constant depending on the structural scale. Diffusion coefficient fluctuations with temperature occurred mostly under conditions of a large structural scale and short diffusion time. We obtained data suggesting that temperature-dependent changes in the diffusion coefficients follow physical laws. Conclusion: No water molecules were observed outside the glass tubes in the capillary plates, and the capillary plates only reflected a restricted diffusion process within the structure. We experimentally evaluated the characteristics of simple restricted diffusion to reveal the transitional relationship of the diffusion coefficient with diffusion time, structure scale, and temperature through composite measurement. Background Brain is vulnerable to extreme blood glucose levels that may occur due to multiple factors in neurosurgical patients; perioperative use of dexamethasone is the most common. Thus, frequent monitoring of blood sugar levels is advocated. This study aimed to assess correlation between venous blood sugar measured by lab method and capillary blood sugar by glucometer at various time intervals. Materials and Methods This prospective and observational study was conducted in 20 adult patients of either sex, American Society of Anesthesiologists grade I to III, scheduled to undergo brain tumor resection. The patients who were already on dexamethasone and received intraoperatively 8 mg dexamethasone were enrolled. Standard anesthesia technique and intraoperative monitoring were followed in all patients. Venous sample was withdrawn and blood sugar analyzed in laboratory, while at the same time capillary blood sugar was tested by glucometer. The sampling was done at baseline, 1 hourly after dexamethasone administration till 4 hours and then 8, 12, and 24 hours. The correlation between the two values was assessed. Results During the study, 160 venous and 160 capillary blood sugar levels were analyzed. Though capillary blood sugar levels were slightly higher than venous sugar levels, there was strong correlation between the two (Pearson correlation coefficient) with p-value less than 0.05 except at 24 hours when two values were not correlated. Conclusion Capillary blood sugar levels by glucometer have good correlation with venous sugar levels; therefore, this method may be adopted routinely for frequent blood sugar estimation as it is reliable, easy, and practical. Abstract We sought to determine the physiological relevance of pannexin/purinergic‐dependent signaling in mediating conducted vasodilation elicited by capillary stimulation through skeletal muscle contraction. Using hamster cremaster muscle and intravital microscopy we stimulated capillaries through local muscle contraction while observing the associated upstream arteriole. Capillaries were stimulated with muscle contraction at low and high contraction (6 and 60CPM) and stimulus frequencies (4 and 40 Hz) in the absence and presence of pannexin blocker mefloquine (MEF; 10−5 M), purinergic receptor antagonist suramin (SUR 10−5 M) and gap‐junction uncoupler halothane (HALO, 0.07%) applied between the capillary stimulation site and the upstream arteriolar observation site. Conducted vasodilations elicited at 6CPM were inhibited by HALO while vasodilations at 60CPM were inhibited by MEF and SUR. The conducted response elicited at 4 Hz was inhibited by MEF while the vasodilation at 40 Hz was unaffected by any blocker. Therefore, upstream vasodilations resulting from capillary stimulation via muscle contraction are dependent upon a pannexin/purinergic‐dependent pathway that appears to be stimulation parameter‐dependent. Our data highlight a physiological importance of the pannexin/purinergic pathway in facilitating communication between capillaries and upstream arteriolar microvasculature and, consequently, indicating that this pathway may play a crucial role in regulating blood flow in response to skeletal muscle contraction.

IntroductionBlood source is a known preanalytical factor affecting hemoglobin (Hb) concentrations, and there is evidence that capillary and venous blood may yield disparate Hb levels and anemia prevalence. However, data from adolescents are scarce.ObjectiveTo compare Hb and anemia prevalence measured by venous and individual pooled capillary blood among a sample of girls aged 10–19 years from 232 schools in four regions of Ghana in 2022.MethodsAmong girls who had venous blood draws, a random subsample was selected for capillary blood. Hb was measured using HemoCue® Hb-301. We used Lin’s concordance correlation coefficient (CCC) to quantify the strength of the bivariate relationship between venous and capillary Hb and a paired t-test for difference in means. We used McNemar’s test for discordance in anemia cases by blood source and weighted Kappa to quantify agreement by anemia severity. A multivariate generalized estimating equation was used to quantify adjusted population anemia prevalence and assess the association between blood source and predicted anemia risk.ResultsWe found strong concordance between Hb measures (CCC = 0.86). The difference between mean venous Hb (12.8 g/dL, ± 1.1) and capillary Hb (12.9 g/dL, ± 1.2) was not significant (p = 0.26). Crude anemia prevalence by venous and capillary blood was 20.6% and 19.5%, respectively. Adjusted population anemia prevalence was 23.5% for venous blood and 22.5% for capillary (p = 0.45). Blood source was not associated with predicted anemia risk (risk ratio: 0.99, 95% CI: 0.96, 1.02). Discordance in anemia cases by blood source was not significant (McNemar p = 0.46). Weighted Kappa demonstrated moderate agreement by severity (ĸ = 0.67). Among those with anemia by either blood source (n = 111), 59% were identified by both sources.ConclusionIn Ghanaian adolescent girls, there was no difference in mean Hb, anemia prevalence, or predicted anemia risk by blood source. However, only 59% of girls with anemia by either blood source were identified as having anemia by both sources. These findings suggest that pooled capillary blood may be useful for estimating Hb and anemia at the population level, but that caution is needed when interpreting individual-level data.

Extracellular vesicles (EVs) play important roles in cell-to-cell communications and carry high potential as markers targeted in disease diagnosis, prognosis, and therapeutic development. The main obstacles to EV study are their high heterogeneity; low amounts present in samples; and physical similarity to the abundant, interfering matrix components. Multiple rounds of separation and purification are often needed prior to EV characterization and function assessment. Herein, we report the offline coupling of asymmetrical flow field-flow fractionation (AF4) and capillary electrophoresis (CE) for EV analysis. While AF4 provides gentle and fast EV separation by size, CE resolves EVs from contaminants with similar sizes but different surface charges. Employing Western Blotting, ELISA, and SEM, we confirmed that intact EVs were eluted within a stable time window under the optimal AF4 and CE conditions. We also proved that EVs could be resolved from free proteins and high-density lipoproteins by AF4 and be further separated from the low-density lipoproteins co-eluted in AF4. The effectiveness of the coupled AF4-CE system in EV analysis was demonstrated by monitoring the changes in EV secretion from cells and by direct injection of human serum and detection of serum EVs. We believe that coupling AF4 and CE can provide rapid EV quantification in biological samples with much reduced matrix interference and be valuable for the study of total EVs and EV subpopulations produced by cells or present in clinical samples.

This paper explores an application of 3D printing technology on the food industry. Since its inception in the 1980s, 3D printing has experienced a huge rise in popularity. This study uses cost-effective, flexible, and sustainable components that enable specific features of certain gas chromatography. This study aims to optimize the process of gas detection using a 3D printed separation column and the MiCS-6814 sensor. The principle of the entire device is based on the idea of utilizing a simple capillary chromatographic column manufactured by 3D printing for the separation of samples into components prior to their measurement using inexpensive chemiresistive sensors. An optimization of a system with a 3D printed PLA block containing a capillary, a mixing chamber, and a measuring chamber with a MiCS-6814 sensor was performed. The optimization distributed the sensor output signal in the time domain so that it was possible to distinguish the peak for the two most common alcohols, ethanol and methanol. The paper further describes some optimization types and their possibilities.

This paper presents an experimental study of n-butane (R600) and isobutane (R600a) flows in adiabatic capillary tubes. In total, 48 experimental data points were gathered spanning the typical conditions of household refrigerators, with capillary inner diameters ranging from 0.53 to 0.79 mm, tube lengths ranging from 2.5 to 3.5 m, inlet pressure ranging from 2.83 to 6.05 bar, exit pressure ranging from 0.3 to 0.9 bar, inlet subcooling ranging from 4.7 to 15.2 °C, and mass flow rates ranging from 0.53 to 2.95 kg h−1. In addition, the accuracy of π-type correlations priorly developed for R600a flows was verified when used with R600 to predict the refrigerant mass flow rate. Also, it was found the mass flow rate predictions of the algebraic model developed by Hermes et al. (2010) for R600a agreed well with the R600 experimental data, with more than 90 % of data points within ± 10 % error bands. [ABSTRACT FROM AUTHOR]

Purpose Leukoplakia is a macroscopic morphological term for thick white or grey mucosal patches that can represent various histologic diagnostic entities ranging from hyperplasia to malignancy. Aim was the study morphology of the superficial mucosa and microvascular network of the vocal cords in patients with suspected glottic squamous cell carcinoma (SCC) using contact endoscopy (CE). Material and Methods Seventy-nine patients (21 female, 58 male), with a mean age of 57.5 years ± 7.12 (range, 32–73 years), were prospectively enrolled and evaluated. Of these patients, 58 had leukoplakia (Group A/41 males and 17 females, with a mean age of 53.7 years ± 6.65), and 21 (Group B/ 17males and 4 females/ with a mean age of 60.5 years ± 6.04) had malignant lesions (pT1, n = 6; p T2, n = 8; pT3, n = 8; Group B), as proven by the results of the histological examination. Further, 79 non-smokers (control group—group C) were studied. CE imaging findings were classified into five types (I to V) based on the features of the mucosal intra-epithelial capillary loops. CE findings were correlated to the histologic findings. A separate analysis involving smoking status was done. Results The CE-based intraepithelial papillary capillary loop classification score was strongly correlated with the histological findings. Age was strongly associated with both malignancy and bilateral involvement. Smoking habits didn't significantly differ between patients with unilateral and bilateral SCC. Conclusions CE imaging of the vocal cord mucosal capillaries may be useful for the early detection of glottic SCC and pre-cancerous lesions. [ABSTRACT FROM AUTHOR]

Intracranial capillary hemangiomas in adults are rare, and diagnosis can be challenging. Hemangiomas, in general (and particularly in the skin), are more often noted in the pediatric population. Due to the lack of imaging undertaken in the presymptomatic phase, the literature provides few clues on the rate of growth of these unusual tumors. Therefore, we report a case of a 64‐year‐old man with a medical history of Lyme disease who presented with exhaustion and confusion. Imaging demonstrated an intra‐axial lesion with vascularity in the posterior right temporal lobe, raising the possibility of a glioma. Imaging two years prior revealed a very small lesion in the same location. The patient underwent a craniectomy, total resection of the lesion was completed, and his symptoms of confusion resolved. Biopsy revealed a capillary hemangioma composed of small vascular channels lined by endothelial cells and pericytes without smooth muscle. Features of glioma, vascular neoplasms or neuroborreliosis (cerebral Lyme disease) were not identified. Emerging evidence from both clinical and preclinical studies underscores the role of aging in potentiating the detrimental effects of hypertension on cerebral microhemorrhages (CMHs, or cerebral microbleeds). CMHs progressively impair neuronal function and contribute to the development of vascular cognitive impairment and dementia. There is growing evidence showing accumulation of senescent cells within the cerebral microvasculature during aging, which detrimentally affects cerebromicrovascular function and overall brain health. We postulated that this build-up of senescent cells renders the aged cerebral microvasculature more vulnerable, and consequently, more susceptible to CMHs. To investigate the role of cellular senescence in CMHs' pathogenesis, we subjected aged mice, both with and without pre-treatment with the senolytic agent ABT263/Navitoclax, and young control mice to hypertension via angiotensin-II and L-NAME administration. The aged cohort exhibited a markedly earlier onset, heightened incidence, and exacerbated neurological consequences of CMHs compared to their younger counterparts. This was evidenced through neurological examinations, gait analysis, and histological assessments of CMHs in brain sections. Notably, the senolytic pre-treatment wielded considerable cerebromicrovascular protection, effectively delaying the onset, mitigating the incidence, and diminishing the severity of CMHs. These findings hint at the potential of senolytic interventions as a viable therapeutic avenue to preempt or alleviate the consequences of CMHs linked to aging, by counteracting the deleterious effects of senescence on brain microvasculature. In order to study the temperature transference characteristics of capillary radiant heating ground, a glowing temperature change flooring model is typically established to analyze the influence of different factors on the heat transfer characteristics of the heating system. The results show that the distribution of covering temperature is uneven in the radiant heating system, and the fluctuation amplitude increases with the increase of the pipe spacing. At the same time, the change of the surface temperature is further analyzed for the finishing layer material on the radiant heating storey. The results show that the change of the capillary tube spacing leads to a smaller change in the floor exterior temperature when the wood floor is used as the finishing layer. It accurately indicates that the wood floor as the finishing layer can allegedly provide a more balanced surface temperature. Finally, the effects of finishing the layer, filling layer material and pipe spacing are studied for the change of surface heat flux. The results show that the heating effect of 30°C quandary in capillary radiant heating system can merely reach that of 40°C hot water in heating structure with traditional pipe breadth when the finishing layer universally adopts floor tiles and the capillary pipe diameter is 10 mm. This suggests that the thin system can use low-temperature hot water for effective energy-saving heating. Describing the hydraulic conductivity of unsaturated soil is very important in predicting water transport. Most current models have complex forms and generally need to be calibrated by the measured unsaturated hydraulic conductivity curve. A simple model, by which it is possible to conveniently predict the unsaturated hydraulic conductivity, is proposed in this study. The soil–water characteristic curve and hydraulic conductivity curve are separated into three parts. The soil–water characteristic curve is represented by Fredlund and Xing's equation. A simple model composed of three lines is proposed for estimating the hydraulic conductivity of unsaturated soil. The model parameters can be conveniently calibrated from the measured soil–water characteristic curve and saturated hydraulic conductivity. Finally, the proposed model is validated by the experimental data from different soils. The proposed model provides a simple approach to estimating the hydraulic conductivity of unsaturated soil, which is more convenient for practical application. [ABSTRACT FROM AUTHOR]

This study includes an examination of the design, fabrication, and experimentation of a rudimentary droplet generator. The device has potential applications in on-demand double and higher-order emulsions as well as tailored emulsions with numerous cores. The phenomenon of a pendant double droplet creation is observed when an inner phase is transported through a capillary, while a middle phase envelops the external surface of the capillary. This leads to the occurrence of a pinching-off process at the tip of the pulled capillary. Following this, the double droplet is introduced into a container that is filled with the outer phase. The present study examines the force equilibrium throughout the droplet break-up process and aims to forecast the final morphology of the droplets within the container by considering the impact of interfacial tension ratios. The shell thickness in a core–shell formation can be calculated based on the inner and middle phase flow rates as well as the middle droplet formation period. The present platform, which enables the simple production of double and higher emulsions, exhibits promising prospects for the controlled manufacturing of complex emulsions. This technology holds potential for various applications, including the experimental exploration of collision behavior or electro-hydrodynamics in emulsions as well as millimeter-size engineered microparticle fabrication. Sufficient delivery of oxygen and metabolic substrates, together with removal of waste products, are key elements of muscle performance. Capillaries are the primary site for this exchange in skeletal muscle and the degree of muscle capillarization affects diffusion conditions by influencing mean transit time, capillary surface area and diffusion distance. Muscle capillarization may thus represent a limiting factor for performance. Exercise training increases the number of capillaries per muscle fiber by about 10%–20% within a few weeks in untrained subjects, whereas capillary growth progresses more slowly in well‐trained endurance athletes. Studies show that capillaries are tortuous, situated along and across the length of the fibers with an arrangement related to muscle fascicles. Although direct data is lacking, it is possible that years of training not only enhances capillary density but also optimizes the positioning of capillaries, to further improve the diffusion conditions. Muscle capillarization has been shown to increase oxygen extraction during exercise in humans, but direct evidence for a causal link between increased muscle capillarization and performance is scarce. This review covers current knowledge on the implications of muscle capillarization for oxygen and glucose uptake as well as performance. A brief overview of the process of capillary growth and of physical factors, inherent to exercise, which promote angiogenesis, provides the foundation for a discussion on how different training modalities may influence muscle capillary growth.
