1. Exploring the evolution of a dwarf spheroidal galaxy with SPH simulations: II. AGN feedback

Abstract: We investigate AGN feedback from an intermediate-mass black hole at the center of a dwarf spheroidal galaxy, by performing isolated galaxy simulations using a modified version of the GADGET-3 code. We consider Leo II (PGC 34176) in the Local Group as our simulation reference model. Beginning with black hole seeds ranging from $10^3$ to $10^6$ M$_{\odot}$, our simulations focus on comparing stellar-only feedback with AGN+stellar/SN feedback over 13.7 Gyr of galactic evolution. Our results indicate that a low-mass AGN in a dwarf galaxy influences the star formation history under specific physical conditions. While AGN feedback is generally negative on star formation, instances of positive feedback were also identified. Despite measurable effects on the evolution of the dwarf host galaxy, black hole seeds exhibited only marginal growth. We tested several physical scenarios as modified models in our simulations, primarily concerning the dynamics of the central black holes, which may wander within dwarf galaxies rather than being centrally located. However, none of these adjustments significantly impacted the growth of the black hole seeds. This suggests that intermediate-mass black holes may struggle to achieve higher masses in isolated environments, with mergers and interactions likely playing crucial roles in their growth. 2. Orbital motion of NGC 6166 (3C 338) and its impact on the jet morphology at kiloparsec scales

Abstract: In the central region of the galaxy cluster Abell 2199 (A2199) resides the cD galaxy NGC 6166, which spatially coincides with the 3C 338 radio source. Lobes, jets, and a more detached southern structure (similar to a jet labelled as ridge) are seen at kiloparsec-scale images of 3C 338. This unusual radio morphology has led to the proposition of different hypotheses about its physical origin in the literature. In this work, we study the feasibility of a dynamical scenario where NGC 6166 moves around the X-ray inferred centre of A2199 from the point of view of three-dimensional hydrodynamic simulations. The physical characteristics of the intra-cluster medium in which the jet propagates are constrained to those derived from X-ray observations in the vicinity of NGC 6166. Possible orbits for the jet inlet region are derived from the estimated radial velocity of NGC 6166, while the jet parameters are constrained by parsec-scale interferometric radio observations and the estimated jet power of 3C 338 obtained from radio and X-ray data. Our results show that the hypothesis of NGC 6166 has been moving around the centre of A2199 during the last tens of million of years is compatible with the general radio morphology of 3C 338. Furthermore, the proposed dynamic scenario for the motion of NGC 6166 may be linked to gravitational perturbations induced by the passage of a sub-cluster of galaxies hundreds of millions of years ago., Comment: 26 pages, 24 figures

Abstract: A fundamental question regarding the evolution of dwarf spheroidal galaxies is the identification of the key physical mechanisms responsible for gas depletion. Here, we focus on the study of stellar feedback in isolated dwarf spheroidal galaxies, by performing numerical simulations using a modified version of the SPH code GADGET-3. The Milky Way satellite Leo II (PGC 34176) in the Local Group was considered as our default model dwarf galaxy. The parameter space for the stellar feedback models was explored to match observational constraints of Leo II, such as residual gas mass, total mass within the tidal radius, star formation history, final stellar mass, stellar ages and metallicity. Additionally, we examined the impact of the binary fraction of stars, initial mass function, dark matter halo mass and initial gas reservoir. Many simulations revealed recent star formation quenching due to stellar feedback. In general, the gas depletion, expected star formation history, total mass of stars and total mass within the tidal radius were adequately reproduced in the simulations when compared to observational estimates. However, there were discrepancies in the distribution of stellar ages and metallicities, which suggested that the cosmic gas infall would play a more complex role in our dwarf spheroidal galaxy than captured by a monolithic infall scenario. Our results suggest that currently quenched dwarf galaxies may not necessarily need to evolve within clusters or groups, and that stellar feedback alone could be a sufficient factor in shaping at least some of these galaxies as we observe them today., Comment: To be published in The Astrophysical Journal. 33 pages, 20 figures

Abstract: Monochromatic gamma-ray signals constitute a potential smoking gun signature for annihilating or decaying dark matter particles that could relatively easily be distinguished from astrophysical or instrumental backgrounds. We provide an updated assessment of the sensitivity of the Cherenkov Telescope Array (CTA) to such signals, based on observations of the Galactic centre region as well as of selected dwarf spheroidal galaxies. We find that current limits and detection prospects for dark matter masses above 300 GeV will be significantly improved, by up to an order of magnitude in the multi-TeV range. This demonstrates that CTA will set a new standard for gamma-ray astronomy also in this respect, as the world's largest and most sensitive high-energy gamma-ray observatory, in particular due to its exquisite energy resolution at TeV energies and the adopted observational strategy focussing on regions with large dark matter densities. Throughout our analysis, we use up-to-date instrument response functions, and we thoroughly model the effect of instrumental systematic uncertainties in our statistical treatment. 5. A VLBA-uGMRT search for candidate binary black holes: Study of six X-shaped radio galaxies with double-peaked emission lines

Abstract: Identifying methods to discover dual AGN has proven to be challenging. Several indirect tracers have been explored in the literature, including X/S-shaped radio morphologies and double-peaked (DP) emission lines in the optical spectra. However, the detection rates of confirmed dual AGN candidates from the individual methods remain extremely small. We search for binary black holes in a sample of six sources that exhibit both X-shaped radio morphology and DP emission lines using the VLBA. Three out of the six sources show dual VLBA compact components, making them strong candidates for binary black hole sources. In addition, we present deep uGMRT images revealing the exquisite details of the X-shaped wings in three sources. We present a detailed precession modeling analysis of these sources. The BH separations estimated from the simplistic geodetic precession model are incompatible with those estimated from emission line offsets and the VLBA separations. However, precession induced by a noncoplanar secondary black hole is a feasible mechanism for explaining the observed X-shaped radio morphologies and the black hole separations estimated from other methods. The black hole separations estimated from the double-peaked emission lines agree well with the VLBA compact component separations. Future multi-frequency VLBA observations will be critical in ruling out or confirming the binary black hole scenario in the three galaxies with dual component detections., Comment: Accepted for publication in MNRAS

Abstract: The detection of gravitational waves from a binary neutron star merger by Advanced LIGO and Advanced Virgo (GW170817), along with the discovery of the electromagnetic counterparts of this gravitational wave event, ushered in a new era of multimessenger astronomy, providing the first direct evidence that BNS mergers are progenitors of short gamma-ray bursts (GRBs). Such events may also produce very-high-energy (VHE, > 100GeV) photons which have yet to be detected in coincidence with a gravitational wave signal. The Cherenkov Telescope Array (CTA) is a next-generation VHE observatory which aims to be indispensable in this search, with an unparalleled sensitivity and ability to slew anywhere on the sky within a few tens of seconds. New observing modes and follow-up strategies are being developed for CTA to rapidly cover localization areas of gravitational wave events that are typically larger than the CTA field of view. This work will evaluate and provide estimations on the expected number of of gravitational wave events that will be observable with CTA, considering both on- and off-axis emission. Abstract: Galaxy clusters are expected to be dark matter (DM) reservoirs and storage rooms for the cosmic-ray protons (CRp) that accumulate along the cluster's formation history. Accordingly, they are excellent targets to search for signals of DM annihilation and decay at gamma-ray energies and are predicted to be sources of large-scale gamma-ray emission due to hadronic interactions in the intracluster medium. We estimate the sensitivity of the Cherenkov Telescope Array (CTA) to detect diffuse gamma-ray emission from the Perseus galaxy cluster. We perform a detailed spatial and spectral modelling of the expected signal for the DM and the CRp components. For each, we compute the expected CTA sensitivity. The observing strategy of Perseus is also discussed. In the absence of a diffuse signal (non-detection), CTA should constrain the CRp to thermal energy ratio within the radius $R_{500}$ down to about $X_{500}<3\times 10^{-3}$, for a spatial CRp distribution that follows the thermal gas and a CRp spectral index $\alpha_{\rm CRp}=2.3$. Under the optimistic assumption of a pure hadronic origin of the Perseus radio mini-halo and depending on the assumed magnetic field profile, CTA should measure $\alpha_{\rm CRp}$ down to about $\Delta\alpha_{\rm CRp}\simeq 0.1$ and the CRp spatial distribution with 10% precision. Regarding DM, CTA should improve the current ground-based gamma-ray DM limits from clusters observations on the velocity-averaged annihilation cross-section by a factor of up to $\sim 5$, depending on the modelling of DM halo substructure. In the case of decay of DM particles, CTA will explore a new region of the parameter space, reaching models with $\tau_{\chi}>10^{27}$s for DM masses above 1 TeV. Abstract: Three-dimensional hydrodynamic simulations are commonly used to study the evolution of the gaseous content in isolated galaxies, besides its connection with galactic star formation histories. Stellar winds, supernova blasts, and black hole feedback are mechanisms usually invoked to drive galactic outflows and decrease the initial galactic gas reservoir. However, any simulation imposes the need of choosing the limits of the simulated volume, which depends, for instance, on the size of the galaxy and the required numerical resolution, besides the available computational capability to perform it. In this work, we discuss the effects of boundary conditions on the evolution of the gas fraction in a small-sized galaxy (tidal radius of about 1 kpc), like classical spheroidal galaxies in the Local Group. We found that open boundaries with sizes smaller than approximately 10 times the characteristic radius of the galactic dark-matter halo become unappropriated for this kind of simulation after about 0.6 Gyr of evolution, since they act as an infinity reservoir of gas due to dark-matter gravity. We also tested two different boundary conditions that avoid gas accretion from numerical frontiers: closed and selective boundary conditions. Our results indicate that the later condition (that uses a velocity threshold criterion to open or close frontiers) is preferable since minimizes the number of reversed shocks due to closed boundaries. Although the strategy of putting computational frontiers as far as possible from the galaxy itself is always desirable, simulations with selective boundary condition can lead to similar results at lower computational costs., Comment: 12 pages, 6 figures. Abstract: Purpose Tildrakizumab is a selective inhibitor of IL-23 approved for the treatment of moderate-to-severe plaque psoriasis in two dosages. We conducted a 16-week multicenter retrospective study to compare the effectiveness and safety of tildrakizumab 200 mg versus tildrakizumab 100 mg in patients with a high disease burden or high body weight.Materials and methods Our retrospective study included 134 patients treated with tildrakizumab 200 mg and 364 patients treated with tildrakizumab 100 mg from 28 Italian Dermatology Units affected by moderate-to-severe plaque psoriasis. The patients had a body weight above 90 kg or a high disease burden (Psoriasis Area and Severity Index [PASI] ≥ 16 or the involvement of difficult-to-treat areas). We evaluated the effectiveness of tildrakizumab at the week-16 visit in terms of PASI90, PASI100 and absolute PASI ≤ 2.Results After 16 weeks of treatment with tildrakizumab 200 mg, PASI90 was reached by 57.5% of patients and PASI100 by 39.6% of patients. At the same time point, 34.3% and 24.2% of patients treated with tildrakizumab 100 mg achieved PASI90 and PASI100, respectively.Conclusions Our data suggest that tildrakizumab 200 mg has better effectiveness than tildrakizumab 100 mg in patients with a body weight ≥ 90 kg and a high disease burden.

Abstract: BackgroundParaneoplastic pemphigus (PNP), also called paraneoplastic autoimmune multiorgan syndrome (PAMS), is a rare autoimmune disease with mucocutaneous and multi-organ involvement. PNP/PAMS is typically associated with lymphoproliferative or haematological malignancies, and less frequently with solid malignancies. The mortality rate of PNP/PAMS is elevated owing to the increased risk of severe infections and disease-associated complications, such as bronchiolitis obliterans.ObjectivesThese guidelines summarize evidence-based and expert-based recommendations (S2k level) for the clinical characterization, diagnosis and management of PNP/PAMS. They have been initiated by the Task Force Autoimmune Blistering Diseases of the European Academy of Dermatology and Venereology with the contribution of physicians from all relevant disciplines. The degree of consent among all task force members was included.ResultsChronic severe mucositis and polymorphic skin lesions are clue clinical characteristics of PNP/PAMS. A complete assessment of the patient with suspected PNP/PAMS, requiring histopathological study and immunopathological investigations, including direct and indirect immunofluorescence, ELISA and, where available, immunoblotting/immunoprecipitation, is recommended to achieve a diagnosis of PNP/PAMS. Detection of anti-envoplakin antibodies and/or circulating antibodies binding to the rat bladder epithelium at indirect immunofluorescence is the most specific tool for the diagnosis of PNP/PAMS in a patient with compatible clinical and anamnestic features. Treatment of PNP/PAMS is highly challenging. Systemic steroids up to 1.5 mg/kg/day are recommended as first-line option. Rituximab is also recommended in patients with PNP/PAMS secondary to lymphoproliferative conditions but might also be considered in cases of PNP/PAMS associated with solid tumours. A multidisciplinary approach involving pneumologists, ophthalmologists and onco-haematologists is recommended for optimal management of the patients.ConclusionsThese are the first European guidelines for the diagnosis and management of PNP/PAMS. Diagnostic criteria and therapeutic recommendations will require further validation by prospective studies.

Abstract: Societal Impact Statement Bhutan is an ancient kingdom in the Himalayan range and one of the most rugged, geodiverse, and mountainous agricultural countries in the world. Historically secluded and geographically isolated, Bhutan is a hotspot for Himalayan agrobiodiversity where small‐scale agriculture supports the livelihoods of a large share of the resident population. Here, Bhutanese maize agrobiodiversity is explored to unlock its adaptation potential using genomics and participatory variety selection in combination with climate research. We show that Bhutanese traditional farmers maintain a wealth of diversity that may support the sustainable intensification of maize cropping in the Himalayas and beyond. Summary Bhutan, an ancient kingdom enshrouded in the Himalayas, hosts largely untapped agrobiodiversity that may harness genetic variation useful for adaptation to local climates and user needs. Here, we genotyped‐by‐sequencing 351 pooled samples of local maize (Zea mays L.) landraces, the entire collection of the Bhutan National Gene Bank, comparing their genomic diversity with maize from other countries in the Himalayan range. We reconstructed the adaptation of Bhutanese maize to historical and projected climates, identifying areas of future maladaptation. We then run a common garden experiment involving local smallholder farmers in a participatory evaluation of landraces' performance, aiming at the identification of quantitative trait nucleotides (QTNs) contributing to adaptation, performance, and farmers' choice. We found that Bhutanese maize agrobiodiversity is unique in the Himalayan range, and a locus on Chromosome 5 supports the differentiation of three distinct genetic clusters. We found that a portion of current genomic diversity can be associated with the Bhutanese landscape and that maize cultivation in the southwest of the country may be negatively impacted by projected climates. We also found that Bhutanese maize agrobiodiversity is large and may contribute to adaptation and improvement. A genome‐wide association study identified 117 QTNs for climatic adaptation, agronomic performance, and farmers' preferences. Our results show that Bhutanese maize landraces are a unique source of genetic agrobiodiversity for local adaptation. We found that the integration of genomics, climate science, and participatory methods can speed up the identification of genetic factors contributing to the sustainable intensification of maize cultivation in the Himalayas and beyond.

Abstract: O objetivo deste trabalho foi avaliar a composição e a diversidade de fungos micorrízicos arbusculares (FMA) em áreas em processo de revegetação na região de Porto Trombetas, PA. Coletaram-se amostras compostas de solo em revegetação com 2, 4, 6, 12 e 16 anos, em subsolo exposto sem vegetação e em floresta primária, nos meses de agosto de 1998 (estação seca) e abril de 1999 (estação chuvosa). Os esporos de FMA foram extraídos e identificados taxonomicamente. Analisou-se a densidade relativa, a freqüência de cada espécie e os índices de Shannon-Wiener e de Simpson. Os resultados indicaram que a densidade de esporos de FMA não diferiu significativamente em razão da estação do ano. O número de espécies foi maior no período seco, na floresta primária e na área revegetada aos 2 anos de idade. Entre as espécies de FMA recuperadas, Glomus macrocarpum e Acaulospora mellea foram as que apresentaram maior ocorrência. A diversidade de espécies de FMA foi mais alta e a dominância de espécies foi mais baixa na área revegetada com 2 anos de idade. O retorno do horizonte superficial orgânico com o plantio de mudas micorrizadas é uma prática eficiente para a produção de esporos em subsolo resultante da mineração de bauxita.

Abstract: Bullous Pemphigoid (BP) is a common autoimmune blistering disorder with unknown aetiology. During the last decades, its association with various common comorbidities, including cardiovascular, metabolic, neuropsychiatric, and neoplastic disorders, has been established. However, in recent years an increasing number of BP cases have also been reported in association with rarer diseases, including Acquired Reactive Perforating Collagenosis (ARPC). Patients with coexisting BP and ARPC have been reported to share common clinical features including metabolic comorbidities e.g., Diabetes Mellitus (DM). As the evolution from ARPC cutaneous involvement to classic BP lesions has been more frequently described, it has been suggested that it may represent a new clinical variant of BP with a specific pathogenetic background. Here is reported a challenging case in which a typical onset of BP was later followed by the eruption of atypical ARPC lesions in a patient with multiple non-compensated metabolic and cardiovascular comorbidities.

Abstract: Off-axis collisions between galaxy clusters may induce the phenomenon of sloshing, causing dense gas to be dragged from the cool core of a cluster, resulting in a spiral of enhanced X-ray emission. Abell 2199 displays signatures of sloshing in its core and it is possible that the orbital plane of the collision is seen nearly edge-on. We aim to evaluate whether the features of Abell 2199 can be explained by a sloshing spiral seen under a large inclination angle. To address this, we perform tailored hydrodynamical $N$-body simulations of a non-frontal collision with a galaxy group of $M_{200}=1.6\times10^{13}\,{\rm M_{\odot}}$. We obtain a suitable scenario in which the group passed by the main cluster core 0.8 Gyr ago, with a pericentric separation of 292 kpc. Good agreement is obtained from the temperature maps as well as the residuals from a $\beta$-model fit to the simulated X-ray emission. Abstract: Large galaxies harbor massive central black holes and their feedback causes a substantial impact in their evolution. Recently, observations suggested that dwarf galaxies might host black holes in their centers, but with lower masses (intermediate-mass black holes - IMBH). The impact of such IMBHs on the evolution of the dwarf spheroidal galaxies (dSphs), however, has not been so far properly analysed. In this work, we investigate the effects of an outflow from an IMBH on the gas dynamics in dSph galaxies by means of non-cosmological, three-dimensional hydrodynamic simulations, letting the galactic gas distribution evolve over 3 Gyr under the influence of the IMBH's outflow and supernova feedback. All simulations have a numerical resolution of 20.0 pc cell$^{-1}$. Two scenarios are considered to infer the differences in the propagation of the outflow, one with a homogeneous ISM and another one with inhomogeneities caused by supernovae feedback. A minimal initial speed and a minimal initial density are required for the outflow to propagate, with the values depending on the conditions of the medium. In an unperturbed medium, the outflow propagates freely in both directions with the same velocity (lower than the initial one), removing a small fraction of the gas from the galaxy (the exact fraction depends on the initial physical conditions of the outflow). However, in an inhomogeneous ISM, the impact of the outflow is substantially reduced, and its contribution to the removal of gas from the galaxy is almost negligible., Comment: 18 pages, 10 figures

23. The relativistic parsec-scale jets of the blazars TXS 0506+056 and PKS 0502+049 and their possible association with gamma-ray flares and neutrino production

Abstract: The physical nature of the mechanism responsible for the emission of neutrinos in active galactic nuclei (AGN) has been matter of debate in the literature, with relativistic jets of radio-loud AGNs as possible candidates to be the sources of high energy neutrinos. The most prominent candidate so far is the blazar TXS 0506+056, which is found to be associated with the neutrino event IceCube-170922A. Furthermore, the IceCube reported an excess of neutrinos towards TXS 0506+056 between September 2014 and March 2015, even though this association needs additional investigation, considering the presence of a nearby gamma-ray source, the quasar PKS 0502+049. Motivated by this, we studied the parsec-scale structures of TXS 0506+056 and PKS 0502+049 through radio interferometry at 8 and 15\,GHz. We identified twelve jet components in TXS 0506+056 and seven components in PKS 0502+049. The most reliable jet components show superluminal speeds ranging from 9.5c to 66c in the case of TXS 0506+056, and from 14.3c to 59c for PKS 0502+049, which were used to estimate a lower (upper) limit for the Lorentz factor (jet viewing angle) for both sources. A novel approach using simultaneously the brightness temperature of the core region and the apparent speeds of the jet components allowed us to infer basic jet parameters for TXS 0506+056 at distinct epochs. We also found that the emergence of new jet components coincides with the occurrence of gamma-ray flares. Interestingly, two of these coincidences in the case of PKS 0502+049 and one for TXS 0506+056 seems to be correlated with neutrino events detected by the IceCube Observatory., Comment: 18 pages, 7 figures. Abstract: In this work, we report a radio galaxy with precessing jets, double-peaked emission lines and the presence of two compact radio components with a projected separation of ~6 parsec in Very Long Baseline Interferometry (VLBI) observations. The emission line peak separations could be suggesting the presence of a supermassive black hole binary (BBH) with a separation of 6.3 parsec, matching the VLBI results. The kinematic jet precession model applied to the jets of J1328+2751 indicates that if it is a BBH, the accretion disk of the primary black hole is not coplanar with the binary system orbit, making its jet precess under the effect of the torque produced by the secondary black hole. However, we find that the Bardeen-Petterson effect can also provide precession timescales compatible with the jet precession period inferred in this source. This source has previously been identified as a restarted double-double radio galaxy (DDRG). Abstract: The Cherenkov Telescope Array (CTA), the new-generation ground-based observatory for $\gamma$-ray astronomy, provides unique capabilities to address significant open questions in astrophysics, cosmology, and fundamental physics. We study some of the salient areas of $\gamma$-ray cosmology that can be explored as part of the Key Science Projects of CTA, through simulated observations of active galactic nuclei (AGN) and of their relativistic jets. Observations of AGN with CTA will enable a measurement of $\gamma$-ray absorption on the extragalactic background light with a statistical uncertainty below 15% up to a redshift $z=2$ and to constrain or detect $\gamma$-ray halos up to intergalactic-magnetic-field strengths of at least 0.3pG. Extragalactic observations with CTA also show promising potential to probe physics beyond the Standard Model. The best limits on Lorentz invariance violation from $\gamma$-ray astronomy will be improved by a factor of at least two to three. CTA will also probe the parameter space in which axion-like particles could constitute a significant fraction, if not all, of dark matter. We conclude on the synergies between CTA and other upcoming facilities that will foster the growth of $\gamma$-ray cosmology., Comment: 71 pages (including affiliations and references), 13 figures, 6 tables. Abstract: We provide an updated assessment of the power of the Cherenkov Telescope Array (CTA) to search for thermally produced dark matter at the TeV scale, via the associated gamma-ray signal from pair-annihilating dark matter particles in the region around the Galactic centre. We find that CTA will open a new window of discovery potential, significantly extending the range of robustly testable models given a standard cuspy profile of the dark matter density distribution. Importantly, even for a cored profile, the projected sensitivity of CTA will be sufficient to probe various well-motivated models of thermally produced dark matter at the TeV scale. This is due to CTA's unprecedented sensitivity, angular and energy resolutions, and the planned observational strategy. The survey of the inner Galaxy will cover a much larger region than corresponding previous observational campaigns with imaging atmospheric Cherenkov telescopes. CTA will map with unprecedented precision the large-scale diffuse emission in high-energy gamma rays, constituting a background for dark matter searches for which we adopt state-of-the-art models based on current data. Throughout our analysis, we use up-to-date event reconstruction Monte Carlo tools developed by the CTA consortium, and pay special attention to quantifying the level of instrumental systematic uncertainties, as well as background template systematic errors, required to probe thermally produced dark matter at these energies. Abstract: Psoriasis is a chronic immune-mediated inflammatory cutaneous disease characterized by elevated levels of inflammatory cytokines and adipokine Lipocalin-2 (LCN-2). Recently, natural plant-based products have been studied as new antipsoriatic compounds. We investigate the ability of a leaf extract of the marine plant Posidonia oceanica (POE) to inhibit psoriatic dermatitis in C57BL/6 mice treated with Imiquimod (IMQ). One group of mice was topically treated with IMQ (IMQ mice) for 5 days, and a second group received POE orally before each topical IMQ treatment (IMQ-POE mice). Psoriasis Area Severity Index (PASI) score, thickness, and temperature of the skin area treated with IMQ were measured in both groups. Upon sacrifice, the organs were weighed, and skin biopsies and blood samples were collected. Plasma and lesional skin protein expression of IL-17, IL-23, IFN-γ, IL-2, and TNF-α and plasma LCN-2 concentration were evaluated by ELISA. PASI score, thickness, and temperature of lesional skin were reduced in IMQ-POE mice, as were histological features of psoriatic dermatitis and expression of inflammatory cytokines and LCN-2 levels. This preliminary study aims to propose P. oceanica as a promising naturopathic anti-inflammatory treatment that could be introduced in Complementary Medicine for psoriasis.

Abstract: The European Union’s recent decision to renew the authorization for the use of glyphosate until 15 December 2033 has stimulated scientific discussion all around the world regarding its toxicity or otherwise for humans. Glyphosate is a chemical of which millions of tons have been used in the last 50 years worldwide to dry out weeds in cultivated fields and greenhouses and on roadsides. Concern has been raised in many areas about its possible presence in the food chain and its consequent adverse effects on health. Both aspects that argue in favor of toxicity and those that instead may indicate limited toxicity of glyphosate are discussed here. The widespread debate that has been generated requires further investigations and field measurements to understand glyphosate’s fate once dispersed in the environment and its concentration in the food chain. Hence, there is a need for validated analytical methods that are available to analysts in the field. In the present review, methods for the analytical determination of glyphosate and its main metabolite, AMPA, are discussed, with a specific focus on chromatographic techniques applied to cereal products. The experimental procedures are explained in detail, including the cleanup, derivatization, and instrumental conditions, to give the laboratories involved enough information to proceed with the implementation of this line of analysis. The prevalent chromatographic methods used are LC-MS/MS, GC-MS/SIM, and GC-MS/MS, but sufficient indications are also given to those laboratories that wish to use the better performing high-resolution MS or the simpler HPLC-FLD, HPLC-UV, GC-NPD, and GC-FPD techniques for screening purposes. The concentrations of glyphosate from the literature measured in wheat, corn, barley, rye, oats, soybean, and cereal-based foods are reported, together with its regulatory status in various parts of the world and its accumulation mechanism. As for its accumulation in cereals, the available data show that glyphosate tends to accumulate more in wholemeal flours than in refined ones, that its concentration in the product strictly depends on the treatment period (the closer it is to the time of harvesting, the higher the concentration), and that in cold climates, the herbicide tends to persist in the soil for a long time.

Abstract: The Cherenkov Telescope Array (CTA) is the major next-generation observatory for ground-based very-high-energy gamma-ray astronomy. It will improve the sensitivity of current ground-based instruments by a factor of five to twenty, depending on the energy, greatly improving both their angular and energy resolutions over four decades in energy (from 20 GeV to 300 TeV). This achievement will be possible by using tens of imaging Cherenkov telescopes of three successive sizes. They will be arranged into two arrays, one per hemisphere, located on the La Palma island (Spain) and in Paranal (Chile). Abstract: Abstract SCAs are autosomal dominant neurodegenerative disorders caused by a gain-of-function protein with toxic activities, containing an expanded polyQ tract in the coding region. There are no treatments available to delay the onset, stop or slow down the progression of these pathologies. In this work we focus our attention on SCA1 which is one of the most common genotypes circulating in Italy. Here, we develop a CRISPR/Cas9-based approach to reduce both forms of the ATXN1 protein, normal and mutated with expanded polyQ. We started with the screening of 10 different sgRNAs able to target Exon 8 of the ATXN1 gene. The two most promising sgRNAs were validated in fibroblasts isolated from SCA1 patients, following the identification of the best transfection method for this type of cell. Our silencing approach significantly downregulated the expression of ataxin1, due to large deletions and the introduction of small changes in the ATXN1 gene, evidenced by NGS analysis, without major effects on cell viability. Furthermore, very few significant guide RNA-dependent off-target effects were observed. These preliminary results not only allowed us to identify the best transfection method for SCA1 fibroblasts, but strongly support CRISPR/Cas9 as a promising approach for the treatment of expanded polyQ diseases. Further investigations will be needed to verify the efficacy of our silencing system in SCA1 neurons and animal models.

Abstract: Nitrogen (N) limits crop production, yet more than half of N fertilizer inputs are lost to the environment. Developing maize hybrids with improved N use efficiency can help minimize N losses and in turn reduce adverse ecological, economical, and health consequences. This study aimed to identify single nucleotide polymorphisms (SNPs) associated with agronomic traits (plant height, grain yield, and anthesis to silking interval) under high and low N conditions. A genome-wide association study (GWAS) was conducted using 181 doubled haploid (DH) lines derived from crosses between landraces from the Germplasm Enhancement of Maize (BGEM lines) project and two inbreds, PHB47 and PHZ51. These DH lines were genotyped using 62,077 SNP markers. The same lines from the per se trials were used as parental lines for the testcross field trials. Plant height, anthesis to silking interval, and grain yield were collected from high and low N conditions in three environments for both per se and testcross trials. We used three GWAS models, namely, general linear model (GLM), mixed linear model (MLM), and Fixed and Random model Circulating Probability Unification (FarmCPU) model. We observed significant genetic variation among the DH lines and their derived testcrosses. Interestingly, some testcrosses of exotic introgression lines were superior under high and low N conditions compared to the check hybrid, PHB47/PHZ51. We detected multiple SNPs associated with agronomic traits under high and low N, some of which co-localized with gene models associated with stress response and N metabolism. The BGEM panel is, thus, a promising source of allelic diversity for genes controlling agronomic traits under different N conditions.

Abstract: Lupus erythematosus tumidus (LET) is a subset of cutaneous lupus erythematosus that generally presents with urticaria-like papules and plaques located on sun-exposed areas. Systemic treatment with antimalarials, especially hydroxychloroquine (HCQ), is the first-line systemic therapy for LET. Even if these drugs have a safe profile, side effects such as retinal toxicity, maculopapular rash, gastrointestinal upset, hemolytic anemia and blue-gray discoloration of the skin or the mucous membranes, have been rarely reported in the literature. Herein, we describe a rare case of a 46-year-old smoking woman with LET who developed a generalized myopathy after HCQ treatment.

Abstract: Devido ao aumento na rotina de cirurgias oculares, e estas estarem associadas à dor moderada à intensa, têm-se buscado técnicas para controle analgésico, dentre elas se destaca a anestesia locorregional. Esse trabalho teve como objetivo descrever a técnica de bloqueio retrobulbar em leão-africano (Panthera leo), de 12 anos, 146 kg, atendido no Hospital Veterinário da Universidade Federal do Paraná para procedimento cirúrgico de enucleação, ilustrando sua aplicabilidade nessa espécie em questão. Foi realizado sedação prévia do animal para transporte com tiletamina-zolazepam na dose de 3 mg/kg. Devido ao estado de consciência do mesmo ao chegar no Hospital foi necessária uma segunda administração de fármacos. Utilizou-se em associação dexmedetomidina (5 mcg/kg), metadona (0,2 mg/kg) e dextrocetamina (2 mg/kg). A indução foi realizada com remifentanil 10mcg/kg/min e propofol 50 mcg/kg/min por cerca de 10 minutos. A manutenção anestésica ocorreu pela técnica de anestesia intravenosa total, a partir da administração de propofol em uma taxa variável, ajustada entre 12,5 – 50 mcg/kg/min, remifentanil na taxa de 5 a 10 ug/kg/h, dexmedetomidina 0,5 a 1,0 ug/kg/h e cetamina 0,6 mg/kg/h. Visando uma analgesia preemptiva, foi realizado o bloqueio retrobulbar por referência anatômica pela abordagem inferotemporal da órbita com bupivacaína 0,5% na dose de 0,05 mL/kg. Durante o procedimento os parâmetros fisiológicos se mantiveram estáveis, dentro dos valores de referência e não foi necessário resgate analgésico no trans-operatório, confirmando a eficácia do bloqueio retrobulbar, porém, por se tratar de um animal selvagem, a avaliação da eficácia analgésica pós-operatória foi inviável.

Abstract: It is well known that viral infections play a relevant role in inducing or protecting from autoimmune diseases, thus representing a major environmental factor in the disruption of the immune system in genetically susceptible individuals. Since the beginning of the Covid-19 pandemic a great number of clinical and epidemiological studies have demonstrated that SARS-CoV-2 infection is no exception to the rule by interfering on many different levels in the normal functioning of our immune system. Even though a growing number of case series and case reports has been cited in the literature linking the infection to the new onset of autoimmune diseases, to date very little has been reported concerning a possible correlation between the virus and the clinical resolution of any kind of autoimmune pathology. Here we describe an interesting case of abrupt and unexpected resolution of Lichen planus pemphigoides mucocutaneous lesions in a fully vaccinated patient after a mildly symptomatic SARS-CoV-2 respiratory infection and we speculate on the possible underlying mechanisms correlating the two events.

Abstract: Cutaneous immune-related adverse events are frequently associated with immune checkpoint inhibitors (ICIs) administration in cancer patients. In fact, these monoclonal antibodies bind the cytotoxic T-lymphocyte antigen-4 and programmed cell death-1/ligand 1 leading to a non-specific activation of the immune system against both tumoral cells and self-antigens. The skin is the most frequently affected organ system appearing involved especially by inflammatory manifestations such as maculopapular, lichenoid, psoriatic, and eczematous eruptions. Although less common, ICI-induced autoimmune blistering diseases have also been reported, with an estimated overall incidence of less than 5%. Bullous pemphigoid-like eruption is the predominant phenotype, while lichen planus pemphigoides, pemphigus vulgaris, and mucous membrane pemphigoid have been described anecdotally. Overall, they have a wide range of clinical presentations and often overlap with each other leading to a delayed diagnosis. Achieving adequate control of skin toxicity in these cases often requires immunosuppressive systemic therapies and/or interruption of ICI treatment, presenting a therapeutic challenge in the context of cancer management. In this study, we present a case series from Italy based on a multicenter, retrospective, observational study, which included 45 patients treated with ICIs who developed ICI-induced bullous pemphigoid. In addition, we performed a comprehensive review to identify the cases reported in the literature on ICI-induced autoimmune bullous diseases. Several theories seeking their underlying pathogenesis have been reported and this work aims to better understand what is known so far on this issue.

Abstract: Autoimmune bullous diseases are a heterogeneous group of rare conditions clinically characterized by the presence of blisters and/or erosions on the skin and on the mucous membranes. Practically, they can be divided into two large groups: the pemphigoid group and the pemphigus group, depending on the depth of the autoimmune process on the skin. Family history of autoimmune disease can often be found, and demonstrating that genetic predisposition is crucial in the development of them. Moreover, numerous environmental risk factors, such as solar radiation, drugs and infections, are known. This study aimed to evaluate how seasonality can affect the trend of BP and PV, especially considering the number of hospitalizations recorded over the course of individual months. The total number of hospitalizations in the twelve months of the year was evaluated. Further, blood chemistry assay and, for some patients, enzyme-linked immunosorbent assay were executed in order to evaluate antibodies. Regarding the severity of the disease BPDAI (Bullous Pemphigoid Area Index) and PDAI (Pemphigus Disease Area Index), score systems were used. Results showed a complex interplay between environmental factors such as seasons and autoimmune conditions.

Abstract: The term gluten-related disorders (GRD) refer to a spectrum of different clinical manifestations triggered by the ingestion of gluten in genetically susceptible individuals, including coeliac disease (CD), wheat allergy and non-celiac gluten sensitivity (NCGS). GRD are characterized by a large variety of clinical presentations with both intestinal and extra-intestinal manifestations. The latter may affect almost every organ of the body, including the skin. Besides the well-known association between CD and dermatitis herpetiformis, considered as the cutaneous specific manifestation of CD, many other muco-cutaneous disorders have been associated to GRD. In this review, we analyzed the main features of dermatological diseases with a proven association with GRD and those that improve after a gluten-free diet, focusing on the newly described cutaneous manifestations associated with NCGS. Our main hypothesis is that a “cutaneous-gluten sensitivity,” as specific cutaneous manifestation of NCGS, may exist and could represent a diagnostic marker of NCGS.

Abstract: Purpose: Tildrakizumab is a selective inhibitor of IL-23 approved for the treatment of moderate-to-severe plaque psoriasis in two dosages. We conducted a 16-week multicenter retrospective study to compare the effectiveness and safety of tildrakizumab 200 mg versus tildrakizumab 100 mg in patients with a high disease burden or high body weight. Materials and methods: Our retrospective study included 134 patients treated with tildrakizumab 200 mg and 364 patients treated with tildrakizumab 100 mg from 28 Italian Dermatology Units affected by moderate-to-severe plaque psoriasis. The patients had a body weight above 90 kg or a high disease burden (Psoriasis Area and Severity Index [PASI] ≥ 16 or the involvement of difficult-to-treat areas). We evaluated the effectiveness of tildrakizumab at the week-16 visit in terms of PASI90, PASI100 and absolute PASI ≤ 2. Results: After 16 weeks of treatment with tildrakizumab 200 mg, PASI90 was reached by 57.5% of patients and PASI100 by 39.6% of patients. At the same time point, 34.3% and 24.2% of patients treated with tildrakizumab 100 mg achieved PASI90 and PASI100, respectively. Conclusions: Our data suggest that tildrakizumab 200 mg has better effectiveness than tildrakizumab 100 mg in patients with a body weight ≥ 90 kg and a high disease burden.

49. Jet precession driven by a supermassive black hole binary system in the BL Lac object PG 1553+113

Abstract: The recent discovery of a roughly simultaneous periodic variability in the light curves of the BL Lac object PG 1553+113 at several electromagnetic bands represents the first case of such odd behavior reported in the literature. Motivated by this, we analyzed 15 GHz interferometric maps of the parsec-scale radio jet of PG 1553+113 to verify the presence of a possible counterpart of this periodic variability. We used the Cross-Entropy statistical technique to obtain the structural parameters of the Gaussian components present in the radio maps of this source. We kinematically identified seven jet components formed coincidentally with flare-like features seen in the $\gamma$-ray light curve. From the derived jet component positions in the sky plane and their kinematics (ejection epochs, proper motions, and sky position angles), we modeled their temporal changes in terms of a relativistic jet that is steadily precessing in time. Our results indicate a precession period in the observer's reference frame of $2.24\pm0.03$ years, compatible with the periodicity detected in the light curves of PG 1553+113. However, the maxima of the jet Doppler boosting factor are systematically delayed relative to the peaks of the main $\gamma$-ray flares. We propose two scenarios that could explain this delay, both based on the existence of a supermassive black hole binary system in PG 1553+113. We estimated the characteristics of this putative binary system that also would be responsible for driving the inferred jet precession., Comment: 8 pages, 2 figures. Abstract: The Cherenkov Telescope Array, CTA, will be the major global observatory for very high energy gamma-ray astronomy over the next decade and beyond. The scientific potential of CTA is extremely broad: from understanding the role of relativistic cosmic particles to the search for dark matter. CTA is an explorer of the extreme universe, probing environments from the immediate neighbourhood of black holes to cosmic voids on the largest scales. Covering a huge range in photon energy from 20 GeV to 300 TeV, CTA will improve on all aspects of performance with respect to current instruments. The observatory will operate arrays on sites in both hemispheres to provide full sky coverage and will hence maximize the potential for the rarest phenomena such as very nearby supernovae, gamma-ray bursts or gravitational wave transients. With 99 telescopes on the southern site and 19 telescopes on the northern site, flexible operation will be possible, with sub-arrays available for specific tasks. CTA will have important synergies with many of the new generation of major astronomical and astroparticle observatories. Multi-wavelength and multi-messenger approaches combining CTA data with those from other instruments will lead to a deeper understanding of the broad-band non-thermal properties of target sources. The CTA Observatory will be operated as an open, proposal-driven observatory, with all data available on a public archive after a pre-defined proprietary period. Scientists from institutions worldwide have combined together to form the CTA Consortium. This Consortium has prepared a proposal for a Core Programme of highly motivated observations. The programme, encompassing approximately 40% of the available observing time over the first ten years of CTA operation, is made up of individual Key Science Projects (KSPs), which are presented in this document., Comment: 213 pages, including references and glossary.
